The exact cause of the development of anxiety disorders (AD) in present time has not been fully established and is a subject of debate in many countries. Interest in studying the mechanisms of action of proteins of S100 group, in particular, neurospecific protein S100b, is caused by its participation in processes of integrative activity of brain/neuron and development of diseases of nervous system. The functions of S100 proteins determine their influence on synaptic plasticity and participation in the regulation of stress-realizing and stress-limiting systems, the imbalance of which (primarily, the insufficiency of the GABA-ergic system) is the neurobiological basis of the majority of anxiety-depressive pathologies. Preparations regulating the activity of S100 protein have a distinct clinical anti-anxiety effect and additionally contribute to the restoration of neuronal plasticity processes.

Relevance. After smallpox eradication, in conditions of population immunity to orthopoxviruses absence, Monkeypox virus became most significant orthopoxvirus, pathogenic for humans. Therefore the generalization of data on the areas of infection outbreaks, human diseases and methods of prevention and treatment of monkey pox is important task. Aim. To characterize the problem of monkeypox in the world based on an analysis of foreign scientific publications over the past 20 years. Materials and methods. The work used publications presented in the main international medical information databases PubMed, Web of Science, Embase, etc. To analyze the publications, the analytical epidemiological method was used. Results and discussion. Monkeypox virus, obtained and identified in 1958, by genetic and phenotypic differences divides on two clades: West-African with lethality 3.6% and Central-African (Congo Basin) with lethality 10%. Monkeypox virus transmission to men happens in two ways, either from animal-to-human or human-to-human. Monkey pox is endemic only on African continent, but In 2003 year the first outbreak, numbering 47 confirmed cases, was occurred in non-endemic country – USA and the largest monkeypox outbreak began in Nigeria in September 2017 year and continue to the present. Comparison of the genome sequences of strains, isolated from patients in non-endemic countries, showed, that it genetically close to West-african strains, belong to II clades and were descended from a common ancestor. Many cases of disease in humans in the current outbreak have been traced to sexual transmission especially among men, who identify ourselves as gay or bisexual. The basis method for identification of agent in present time is PCR-RT targeting on the tumor necrosis factor (TNF) receptor gene. Usually monkeypox of human is mild, self-limiting disease. The symptoms of monkeypox are varied and non-specific. One of the most frequently observed clinical symptoms is lymphadenopathy. Most patients recover during some weeks. However, specific antiviral treatment – tecovirimat (S-246) and brincidofovir (CMX-001) – may be used for seriously ill or immunocompromised individuals. For prophylactic disease in present time are use vaccines JYNNEOSTM, ACAM2000R and Aventis Pasteur (APSV). Conclusion. General vaccination against monkeypox don't develop accordingly to modern recommendations WHO. Ring vaccination is recommended to conduct for suppression of spread virus in nidus of infection among population. Timely international coordination is needed to prevent the global spread of a disease with epidemic potential.

Inflammatory processes accompanied by damage to the cell bodies of neurons are combined into the group of neuroinflammation. At the molecular, cellular and tissue levels, neuroinflammation serves as a sanogenetic response to a variety of injuries, including post-traumatic conditions and neurodegeneration. However, inflammatory changes in long-lived cells such as neurons inevitably trigger a range of adverse effects. As a result, prognosis often depends on the severity of neuroinflammation. In this work, we review the spectrum of biomarkers involved in two key mechanisms of neuroinflammation: immune-mediated and mechanical injury, that have a potential clinical application: cytokines, neurospecific proteins and their proteolysis products, markers of oxidative stress, matrix metalloproteinases and endocrine parameters. Over the past decades, a significant pool of data on neuroinflammation biomarkers has been accumulated and continues to grow, but no consensus has been reached on the actual gradation of their clinical significance. Applied branches of medicine require further systematization of information on biomarkers of neuroinflammation for effective prognostic decisions.

After the abolition of the mandatory smallpox vaccination, the humanity lost the immunity not only to smallpox, but also to infections caused by pathogens of this family (Orthopoxvirus): monkeypox, cowpox, buffalo pox, camelpox. Since the camelpox and African gerbil viruses are the closest to the variola virus (genomic homology is 97%) in phylogenetic and genetic terms, it cannot be ruled out that a mutation in a small fragment of the genome of one of these viruses will lead to the replacement of a relatively safe virus with an epidemically dangerous pathogen. The purpose of is article is to summarize materials on the study of camelpox virus. The sources for this research is scientific articles and other English-language literature available via the Internet. The research method is an analysis of scientific sources on camelpox from the general to the specific. The authors considered the epizootic danger of the virus, its virulence for humans, phylogenetic relationship with other orthopoxviruses, means of specific prevention and treatment of camel pox in camels. The discussion and the results. The causative agent of camelpox causes a nodular-pastular rash on the skin and mucous membranes in Camelus dromedaries and Camelus bactrianus. The disease is contagious, and its epizootics lead to significant economic damage. From December 2008 to May 2009, several laboratory-confirmed cases of camelpox in humans were reported in India, Somalia and eastern Sudan. Nowadays for the identification of the camelpox virus, a RT-PCR test system with primers for the C18L gene is usually offered, which detects only this virus. The established host range of the virus is limited to one animal - the camel. To treat sick camels, chemotherapy drugs are used: cidofovir and tocoverimate (ST-246). For immunoprophylaxis, live and inactivated vaccines are used. The conclusion. Camelpox virus poses a risk to humans in regions where people raise camels and are in close contact with them. The immunodeficient populations of people may serve as an additional «window» for the penetration of this virus into human society. The genetic variability of the virus and the plasticity of its genome make it possible to obtain virus strains with altered properties. Synthetic biology methods create a risk, through small substitutions in the genome of the virus, of turning it into an epidemic danger for humans. Constant monitoring of this disease is necessary, since there is a danger of the transmission of camelpox from Kazakhstan to areas bordering the Russian Federation.

Aim. We sought to assess the effects of exogenous Hsp70 (single subcutaneous low- and high-dose injections) on organ structure and functions in adult mice.Materials and methods. We randomized CD1 90-day-old male mice (n = 30) to three groups (10 mice per group). We injected the animals with single subcutaneous saline solution for Group 1 (control), low dose (500 μg/kg) of recombinant human Hsp70 (HspA1A) for Group 2, and high dose (5000 μg/kg) of the Hsp70 for Group 3. We examined the behavior of the mice on Day 3 after the injections (distance traveled, velocity, and bowel movement number). We lethalized the mice on Day 5 with further histological study and morphometrics of cerebral cortex, thymus, spleen, and liver. The statistics included one-factor ANOVA test with post hoc Tukey test.Results. All study groups exhibited no significant difference of behavioral parameters. Some liver sinusoids were wider in control group and Hsp70 500 μg/kg group comparing to Hsp70 5000 μg/kg group. We obtained also data for morphometrics: no difference was found for the number of neurons in ganglionic cerebral cortex, the lymphocytic cellularity difference between thymic cortex and medulla, the number of lymphocytes in white splenic pulp, and the number of hepatocyte nuclei in the liver. Red splenic pulp exhibited 1774,5 ± 24,8, 1623,0 ± 26,7, 1553,6 ± 47,0 macrophages for control, low-dose and high-dose groups, respectively (р < 0,0001). Tukey test showed a significant difference between control group and each of Hsp70 groups 500 μg/kg (р = 0,012) and 5000 μg/kg (р < 0,0001).Conclusion. Our study revealed no negative impact of subcutaneous Hsp70 administration at low and high doses on organ structure and functions in mice.

The ability of mesenchymal stromal cells (MSCs) to migrate into tissue defects and stimulate regeneration makes them a valuable resource for cell therapy. However, in many cases, in vitro cultivation and the influence of the pathological microenvironment in the patient’s body reduce the viability and therapeutic efficacy of MSCs, so their regenerative potential needs to be strengthened. Preconditioning with hormones, cytokines, various chemical or physical factors, cultivation in a three-dimensional environment or at a reduced oxygen content improves the ability of MSCs to colonize damaged tissue, survive in it, and produce regulatory molecules for regeneration. The same goals can be achieved by genetic modification of MSCs. In addition, with the help of transfected MSCs, it is possible to deliver genes necessary for the treatment of hereditary or oncological diseases into the tissue. Finally, an alternative to avoid a decrease in the therapeutic potential of subsequently transplanted MSCs during cultivation can be stimulation of the migration of endogenous patient cells from tissue niches through the systemic circulation to the area of damage. The development of these approaches opens the way to increasing the efficiency of using MSCs in regenerative medicine.

The contribution to the development of histology of the Imperial Moscow University professors — P.P. Einbrodt (Jr.), I.M.Sokolov, J.A. Borzenkova is analyzed. The authors examined the original documents on the history of the Physiological Institute (cabinet) at the Department of Physiology, healthy person (or rather — histological Cabinet department of histology, embryology and comparative anatomy) of the Medical Faculty of IMU.

Acute cerebral circulation disorder is one of the most discussed issues in modern intensive care and neurology, as it is a severe condition, leading to disability or death of the patient, in the absence of immediate medical care. This review discusses general and specific biological markers of stroke, genetic markers of stroke, and current data on their diagnostic significance. The main mechanisms of brain tissue cell death in stroke, such as apoptosis, necrosis, ferroptosis, parthanatosis, sarmoptosis, autolysis, autophagy, oncosis, excitotoxic death are analyzed; the morphological features of the observed processes and their structural manifestations are reviewed. For each type of cell death in nervous tissue, the most frequently detected molecular markers are discussed: specific kinases, Toll-like receptors in the case of apoptosis; serine-threonine protein kinases, components of the polyubiquitin system detected in necrosis; transferrin 1 receptors, typical for ferroptosis; poly(ADP-ribose)-polymerase, whose activity increases in parthanatosis; slow Wallerian degeneration protein that accumulates during sarmoptosis; and other biomarkers characteristic of both individual types of nerve cell death and general pathological processes affecting the brain.

Aim. To study the age-related dynamics of structural and functional interactions of striatal neurons in the implementation of acts of motor behaviour in rats of both sexes.Materials and methods. The study was carried out on 36 Wistar rats of both sexes aged 2, 7 and 16 months (n = 6 per group). In animals of all groups, locomotor activity was determined using a Laboras device (Metris, the Netherlands) for15 minutes, after which the brain was sampled to determine the number and size of neurons in the striatum. The median and interquartile range of the index of motor activity and the number of neurons were determined, and to study the relationship between these indicators, a correlation and regression analysis was performed with the construction of linear and polynomial trends, and the coefficient of determination R2 was calculated.Results. The size of neurons did not change significantly with age in the rats of both sexes. The number of neurons differed statistically in the rats of different sexes in all age groups. In male rats, the maximum number of neurons was noted at the age of 7 months with a decrease to 16 months. In female rats, the maximum number of neurons was recorded at the age of 2 months with a further decrease to 7 and 16 months. According to the regression analysis, a linear strong relationship (R2 =0.80 for males, R2 = 0.79 for females) was established between the number of neurons in the striatum and motor activity in 2-month-old animals. At the age of 7 and 16 months the relationship is non-linear.Conclusion. The number of neurons in the striatum is subject to sex and age dynamics, while their size remains unchanged from 2 to 16 months. For animals of both sexes, a decrease in the role of the striatum in providing motor activity in the process of growing up was noted. This relationship reaches its maximum in 2-month-old rats and then decreases.

Introduction. The outbreaks of the Zaire ebolavirus (ZE) disease (ZED) that have arisen in the last decade determine the need to study the infection pathogenesis, the formation of specific immunity forming as well as the development of effective preventive and therapeutic means. All stages of fight against the ZED spread require the experimental infection in sensitive laboratory animals, which are rhesus monkeys in case of this disease . The aim of the study is to evaluate the rhesus monkey cellular immunity following the ZE experimental infection by the means of flow cytometry (cytofluorimetry). Material and methods. Male rhesus monkeys were intramuscularly infected by the dose of 15 LD50 (dose of the pathogen that causes 50% mortality of infected animals) of the ZE, the Zaire strain (ZEBOV). Levels of 18 peripheral blood lymphocyte populations of the animals before the ZE experimental infection and at the terminal stage of the disease were assessed using flow cytometry. Results and discussion. The certain changes in the levels of the lymphocyte populations were observed following infection, indicating simultaneous activation and suppression of the immune system during ZED. The increase in content was observed for T-lymphocytes, T-helper and cytotoxic T-lymphocytes expressing the corresponding markers of early activation. The decrease was recorded for T-lymphocytes and double-positive T-lymphocytes expressing corresponding markers of late activation, as well as natural killer cells expressing CD8 (p 0.05). Conclusion. For the first time in the Russian Federation, the rhesus monkey cellular immunity before and after the ZE experimental infection was assessed using flow cytometry.

Objective of the study was the development of experimental ELISA tests and lateral flow immunoassays for detection of staphylococcal enterotoxins, A and B types.Materials and methods. Hybridomas, producing monoclonal antibodies against staphylococcal enterotoxins A and B from State Collection of the Affiliated Branch of the “48th Central Research Institute” of the Ministry of Defense of the Russian Federation, BALB/c mice and staphylococcal enterotoxins A and B were used in the research. Hybridoma cells were incubated in culture flasks and in the peritoneal cavity of BALB/c mice. Monoclonal antibodies were isolated from ascitic fluids through precipitation with saturated ammonium sulfate subsequently purified using ion-exchange chromatography. Obtained preparations of monoclonal antibodies were used for the construction of ELISA tests and immune-chromatographic reagent panels for the detection of staphylococcal enterotoxins A and B. Specific components of ELISA tests were lyophilized in protective media.Results and discussion. ELISA tests and lateral flow immunoassays which allow for detecting staphylococcal enterotoxins A and B at concentrations of 0.5 ng/ml and higher, including in food samples, have been constructed.

Kidney diseases are an important medical problem. Kidney injuries are accompanied by oxidative stress, cell death, capillary destruction, inflammation and fibrosis. Mesenchymal stromal cells (MSCs) have a complex effect on the regeneration by producing various regulatory molecules, including those inside extracellular vesicles, and therefore are considered as a promising therapeutic resource for cell therapy of kidney diseases. Their renoprotective effect has been shown in different experimental models, but the results of the clinical trials are ambiguous. Clinical use of MSCs is complicated by their low survival rate in the injured kidney, potential immunogenicity, tumorogenicity and fibrogenicity. Cell-free therapy with the secretory products of MSCs such as conditioned environments or extracellular vesicles is a promising direction for using their regenerative potential. However, introduction of MSCs and their secretory products into medical practice requires further research into the mechanisms of their proregenerative action, improvement of cultivation protocols, and more clinical trials.

Introduction. Docetaxel is widely used for cancer treatment. Actual issue for newly developed polymer-based Docetaxel formulations is applicability of current quality requirements. The technology of polymeric forms, including gamma-sterilization, are differ from actual Docetaxel injections technology. In this study were used PLGA-based particles with Docetaxel. The main attention was directed to studying qualitative and quantitative content of related impurities in polymeric forms during long-term storage and gamma-sterilization in comparison with pharmacopoeial requirements for Docetaxel injections.Aim. Studying of possible patterns of Docetaxel-related impurities formation between intact and gamma-irradiated batches.Materials and methods. Objects of study – lyophilizates of Docetaxel polymeric forms was previously obtained by the authors. Qualitative and quantitative analysis was performed by high pressure liquid chromatography.Results and discussion. Docetaxel-related impurities was determined and evaluated in the intact and gamma-treated batches. It was cleared, that related substances in the gamma-treated batches was different in comparison with non-treated but after vary storage periods.Conclusion. Docetaxel-loaded polymeric drugs shown proper stability during long term storage. The influence of gamma-treatment to Docetaxelrelated impurities content was found. Gamma sterilization can be promising method for novel drugs, but it needs individual study in the each case.

Introduction. Antiseptic Dorogov's stimulator 3rd fraction (ASD-3) is a multicomponent substance, which is obtained by the thermal processing of cattle-origin tissues, which contains more than 120 substances, including biologically active substances. Among the components of ASD-3, necessary for quantitative analysis, pyrrole, phenol, cresol and its derivatives, and indole are distinguished as components. Despite the fact that antiseptic Dorogov's stimulator 3rd fraction was discovered more than half a century ago, there is no information about the pharmacokinetics of the drug components to date. Phenol is one of the preferred substances for research because that is a substance applied in the pharmaceutical technology for the manufacture of medicines.Aim. Study of pharmacokinetic parameters of phenol as a component of a drug containing an antiseptic Dorogov's stimulator 3rd fraction in a dose of 0.5 g/kg, with a single use in a blood serum of 84 female rats weighing 100–120 g.Materials and methods. In the blood serum of the 84 female Wistar rats weighing 100–120 g a study of a phenol pharmacokinetics as a component of ASD-3 drug in a dose of 0.5 g/kg was performed by a single skin application. The first group of animals was intraperitoneally injected with an aqueous emulsion containing 5% ASD-3, while the second one received zinc paste containing 5% ASD-3 on a skin surface. After the application of the emulsion or paste, the animals were euthanized after 0.25; 0.5; 1; 1.5; 2; 6 and 8 hours with further collection of blood from the abdominal aorta into centrifuge tubes. The study was carried out by high performance liquid chromatography.Results and discussion. The elimination rate constant for intraperitoneal drug emulsion injection is 0,0968 h-1, and when the drug paste is applied, 0.0581 h-1. As a result, the half-life of phenol in the first case is 7.2 hours, and in the second case – 11.9 hours. The relative bioavailability of the phenol from the paste containing 5% of the ASD-3 is 14.38% to the 5% emulsion injected into the rats body intraperitoneally.Conclusion. Comparison of the phenol concentration in the blood serum of experimental animals in two ways of application showed that the skin application of a drug paste containing 5% of ASD-3 provides a 10 times lower content of phenol in the blood serum compared to the intraperitoneal application of an emulsion with 5% ASD-3. 15 minutes after the skin application, the components of the drug start to be detected in the blood serum, reaching its maximum concentration in 1.5–2 hours, subsequently gradually decreasing.

Background. Anthropogenic human activity contributes to the inclusion of zinc compounds in the biological cycle in areas where there is pollution with this metal. This leads to the accumulation of the substance in organs and tissues, which can have a neurotoxic effect on the body. Aim. To evaluate the toxic effect of zinc succinate on the structure and function of the cerebral cortex of laboratory rats. Materials and methods. The study was performed on 12 Wistar male rats aged 2 months. The animals were divided into 2 groups: control and experimental, 6 rats each. Within 30 days, the animals of the experimental group were intragastrically injected with a solution containing zinc succinate in a dose of 100 mg/kg. The rats from group 1 served as intact controls. Fluorescent analysis of coenzyme activity of the brain was performed, for which excitation was used at wavelengths of 365 nm and 450 nm, which corresponds to the reduced form of nicotinamide adenine dinucleotide (NADH) and flavin adenine dinucleotide (FAD); morphological analysis of the motor cortex of the cerebral hemispheres.Results. Objective. Optimization of vaccine virus cultivation in the suspended cell culture BHK-21 for infectious activity increment of virus-containing suspension as the base material for smallpox vaccine preparations. Materials and methods. We used suspended culture line of the cells BHK-21 of 72-hour age and nutrient medium of the MEM type in accordance with the guidelines on preparation in our studies. For challenging of the cells, vaccine virus (strain B-51) was used. The virus was adapted through three consequent passages on horion-allantois shell of developing chicken embryos of commercial dermovaccine series 449а at the premises of the Federal State Budgetary Institution “the 48th Central Research Institute” of the Ministry of Defense of the Russian Federation. Information on its genetic features is absent. Cultivation and precipitation of infected cells BHK-21 was carried out in bioreactor with priming volume of 1 liter at (36.5±0.5) °C and aeration with air mixture with varying content of CO2. Results and conclusions. Gas massexchange intensity was enhanced alongside simultaneous maintaining of sparing hydrodynamic conditions for mixing suspended cell cultures in bioreactor. Two-fold increase (up to (4.48±0.63)·109 cell/l) in suspended BHK-21 cell culture concentration at the end of reproduction cycle was achieved. Concentration of the vaccine virus was 3–5 times raised, from (8.1±0.3) lg PFU (plaque forming unit)/ml up to the level of infectious activity – (8.8±0.3) lg PFU/ml. Specific multiplicity of cell infection in recalculation per a cell was 1–5 PFU/cell and by virus yield – 20–100 PFU/cell. Enhanced infectious activity of the virus in concentrated suspension of infected BHK-21 cells substantiates the perspectives of the proposed method for improvement of vaccine virus accumulation phase in the development of anti-smallpox preparations based on cell cultures.

The purpose of the study was to examine the effect of the intraocular lens (IOL) position in the probability of occurrence of the phenomenon of light reflection from its surface and to justify the method of its reduction from the standpoint of the optics laws and eye anatomical parameters. Methods. Based on the laws of physical optics, the reflection of light from the surfaces of the IOL was calculated. The phenomenon of light reflection from the IOL was analyzed, in particular, influence such factors as the lens position in the eye and pupil diameter on its occurrence. Results. It was found that with increasing of distance from the iris to posterior chamber IOL, the angle of appearance of the phenomenon of light reflection from the lens decreases significantly, with the most significant change (angle narrowing from 80° to 19°, i.e., 4.21 times) with increasing of the distance from 1 to 3 mm, which is quite achievable at the present stage of intraocular correction. It was shown that the pupil diameter is less influences by appearance of the phenomenon of light reflection from the lens: it is significant only at a minimum distance from the iris to the IOL (the position of the standard posterior chamber IOL) and insignificant when the IOL is removed from the iris. The obtained results demonstrate the optimality of the method for preventing the appearance of the phenomenon of light reflection from the IOL by removing the intraocular lens from the iris at a distance corresponding to the position of posterior capsule of native lens. Conclusions. In this study, on the basis of the laws of physical optics and mathematical calculations, the leading role of such factor as the IOL position in the eye in appearance and intensity of the phenomenon of light reflection from the lens is substantiated for the first time. The proposed method for reducing the appearance of the phenomenon of light reflection from the IOL, which manifests itself in the form of glare in the eye, visible to others, on the basis of increasing the distance from the iris to the IOL is the optimal solution to the important problem of improving the quality of life of patients with pseudophakia.

The article presents the results of bioassays investigation by the specialists of the Center for Special Laboratory Diagnostics of Particularly Dangerous and Exotic Infectious Diseases, isolated during 2002–2015, obtained from deceased people suspected for rabies virus infection. Outlined are the main challenges of working with the received samples of biological material from dead people and ways of handling these issues. Put forward are the pressing problems of epizootic and epidemiological bias that require comprehensive solutions on the part of healthcare authorities and the veterinary service of the Russian Federation. Given are the results of the laboratory-diagnostic activities of the Center for Special Laboratory Diagnostics of Particularly Dangerous and Exotic Infectious Diseases, which indicate that the applied methods of isolation and identification of the rabies pathogen allow for efficient and in-time diagnosis of the disease.

Biological and physical-chemical properties of the pseudotuberculosis agent cultures were shown to be diverse in the process of submerged cultivation at various temperatures. The microbe cultivation at the increased temperature (37°C) was characterized by fast development and death of the population. Decelerated lag phase and extended exponential phase were observed under the conditions of cultivation at low temperature. The culture grown at 27 °C was found to get an intermediate position between those grown at 10 and 37 °C.

Temperate tularemia bacteriophage was for the first time isolated from the organs of guinea-pig infected with live tularemia vaccine strain N 15 of RIEH line. Negative colonies of bacteriophage were up to 0.2 mm in diameter with incomplete lysis zone at the periphery. In view of the results of electronic microscopy bacteriophage represented filamentous carpuscules. Bacteriophage lyzed bacteria of three subtypes of tularemia etiological agent and bacteria of the main species of legionellosis etiological agents. The simple use of bacteriophage allows to recommend new tularemia bacteriophage GAL for practical application.

Analysis of VNTRs was used for typing of V. cholerae strains deposited in microbial culture collection of 48 Central Research Institute. The most informative loci and flanking primers were chosen, VNTR profiles were compiled, and allele variability analysis of the strains was carried out. Laboratory series of test system were manufactured . The developed test system can be used for epidemiological monitoring and investigations, as well as for passportization of collection strains.

The influence of artificial immunodeficiency state upon brucellosis course has been studied experimentally. Trials on laboratory animals demonstrated faster and more intensive progression of infectious process as well as increased frequency of the fatal cases of the disease caused by brucellosis agents.
