In adults, affective touch leads to widespread activation of cortical areas including posterior Superior Temporal Sulcus (pSTS) and Inferior Frontal Gyrus (IFG). Using functional Near Infrared Spectroscopy (fNIRS), we asked whether similar areas are activated in 5-month-old infants, by comparing affective to non-affective touch. We contrasted a human touch stroke to strokes performed with a cold metallic spoon. The hypothesis that adult-like activation of cortical areas would be seen only in response to the human touch stroke was not confirmed. Similar patterns of activation were seen in both conditions. We conclude that either the posterior STS and IFG have not yet developed selective responses to affective touch, or that additional social cues are needed to be able to identify this type of touch. Density matrix exponentiation (DME) is a general technique for using a quantum state ρ to enact the quantum operation e^{-iρθ} on a target system. It was first proposed in the context of quantum machine learning, but has since been shown to have broad applications in quantum metrology and computation. No experimental demonstration of DME has been performed thus far due to its demanding circuit depths and the need to efficiently generate multiple identical copies of ρ during the finite lifetime of the target system. In this work, we describe the first demonstration of the DME algorithm, which we accomplish using a superconducting quantum processor. Our demonstration relies on a 99.7% fidelity controlled-phase gate implemented using two tunable superconducting transmon qubits. We achieve a fidelity surpassing 90% at circuit depths exceeding 70 when comparing the output of the circuit executed on our quantum processor to a simulation assuming perfect operations and measurements.

Abstract Introduction Accumulation of β‐amyloid is a pathological hallmark of Alzheimer's disease (AD). β‐Amyloid activates the plasma contact system leading to kallikrein‐mediated cleavage of intact high‐molecular‐weight kininogen (HKi) to cleaved high‐molecular‐weight kininogen (HKc). Increased HKi cleavage is observed in plasma of AD patients and mouse models by Western blot. For potential diagnostic purposes, a more quantitative method that can measure HKc levels in plasma with high sensitivity and specificity is needed. Methods HKi/c, HKi, and HKc monoclonal antibodies were screened from hybridomas using direct ELISA with a fluorescent substrate. Results We generated monoclonal antibodies recognizing HKi or HKc specifically and developed sandwich ELISAs that can quantitatively detect HKi and HKc levels in human. These new assays show that decreased HKi and increased HKc levels in AD plasma correlate with dementia and neuritic plaque scores. Discussion High levels of plasma HKc could be used as an innovative biomarker for AD.

A quantum random access memory (qRAM) is considered an essential computing unit to enable polynomial speedups in quantum information processing. Proposed implementations include the use of neutral atoms and superconducting circuits to construct a binary tree but these systems still require demonstrations of the elementary components. Here, we propose a photonic-integrated-circuit (PIC) architecture integrated with solid-state memories as a viable platform for constructing a qRAM. We also present an alternative scheme based on quantum teleportation and extend it to the context of quantum networks. Both implementations realize the two key qRAM operations, (1) quantum state transfer and (2) quantum routing, with already demonstrated components: electro-optic modulators, a Mach-Zehnder interferometer (MZI) network, and nanocavities coupled to artificial atoms for spin-based memory writing and retrieval. Our approaches furthermore benefit from built-in error detection based on photon heralding. Detailed theoretical analysis of the qRAM efficiency and query fidelity shows that our proposal presents viable near-term designs for a general qRAM.

Brain and nervous system development in human infants during the first 1000 days (conception to two years of age) is critical, and compromised development during this time (such as from under nutrition or poverty) can have life-long effects on physical growth and cognitive function. Cortical mapping of cognitive function during infancy is poorly understood in resource-poor settings due to the lack of transportable and low-cost neuroimaging methods. Having established a signature cortical response to social versus non-social visual and auditory stimuli in infants from 4 to 6 months of age in the UK, here we apply this functional Near Infrared Spectroscopy (fNIRS) paradigm to investigate social responses in infants from the first postnatal days to the second year of life in two contrasting environments: rural Gambian and urban UK. Results reveal robust, localized, socially selective brain responses from 9 to 24 months of life to both the visual and auditory stimuli. In contrast at 0–2 months of age infants exhibit non-social auditory selectivity, an effect that persists until 4–8 months when we observe a transition to greater social stimulus selectivity. These findings reveal a robust developmental curve of cortical specialisation over the first two years of life. Abstract Global cerebral blood flow (gCBF) and cerebrovascular reactivity to hypercapnia (CVRCO2) are modulated by gonadal hormone activity, while insulin‐like growth factor 1 facilitates exercise‐mediated cerebral angiogenesis in adults. Whether critical periods of heightened hormonal and neural development during puberty represent an opportunity to further enhance gCBF and CVRCO2 is currently unknown. Therefore, we used duplex ultrasound to assess gCBF and CVRCO2 in n = 128 adolescents characterised as endurance‐exercise trained (males: n = 30, females: n = 36) or untrained (males: n = 29, females: n = 33). Participants were further categorised as pre‐ (males: n = 35, females: n = 33) or post‐ (males: n = 24, females: n = 36) peak height velocity (PHV) to determine pubertal or ‘maturity’ status. Three‐factor ANOVA was used to identify main and interaction effects of maturity status, biological sex and training status on gCBF and CVRCO2. Data are reported as group means (SD). Pre‐PHV youth demonstrated elevated gCBF and slower CVRCO2 mean response times than post‐PHV counterparts (both: P ≤ 0.001). gCBF was only elevated in post‐PHV trained males when compared to untrained counterparts (634 (43) vs. 578 (46) ml min−1; P = 0.007). However, CVRCO2 mean response time was faster in pre‐ (72 (20) vs. 95 (29) s; P ≤ 0.001), but not post‐PHV (P = 0.721) trained youth when compared to untrained counterparts. Cardiorespiratory fitness was associated with gCBF in post‐PHV youth (r2 = 0.19; P ≤ 0.001) and CVRCO2 mean response time in pre‐PHV youth (r2 = 0.13; P = 0.014). Higher cardiorespiratory fitness during adolescence can elevate gCBF while exercise training during childhood primes the development of cerebrovascular function, highlighting the importance of exercise training during the early stages of life in shaping the cerebrovascular phenotype.

Abstract The sudden mortality of African elephants (Loxodonta africana) in Botswana and Zimbabwe in 2020 provoked considerable public interest and speculation. Poaching and malicious poisoning were excluded early on in the investigation. Other potential causes included environmental intoxication, infectious diseases, and increased habitat stress due to ongoing drought. Here we show evidence of the mortalities in Zimbabwe as fatal septicaemia associated with Bisgaard taxon 45, an unnamed close relative of Pasteurella multocida. We analyse elephant carcasses and environmental samples, and fail to find evidence of cyanobacterial or other intoxication. Post-mortem and histological findings suggest a bacterial septicaemia similar to haemorrhagic septicaemia caused by P. multocida. Biochemical tests and 16S rDNA analysis of six samples and genomic analysis of one sample confirm the presence of Bisgaard taxon 45. The genome sequence contains many of the canonical P. multocida virulence factors associated with a range of human and animal diseases, including the pmHAS gene for hyaluronidase associated with bovine haemorrhagic septicaemia. Our results demonstrate that Bisgaard taxon 45 is associated with a generalised, lethal infection and that African elephants are susceptible to opportunistically pathogenic Pasteurella species. This represents an important conservation concern for elephants in the largest remaining metapopulation of this endangered species.

Background: Algorithms to automatically adjust atrioventricular (AV) and interventricular (VV) intervals in cardiac resynchronization therapy (CRT) devices are common, but their clinical efficacy is unknown. Objective: The purpose of this study was to evaluate automatic CRT algorithms in patients with heart failure for the reduction of mortality, heart failure hospitalizations, and clinical improvement. Methods: We performed a systematic review and meta-analysis of randomized controlled trials (RCTs) in patients with CRT using automatic algorithms that change AV and VV intervals dynamically without manual input, on a beat-to-beat basis. We performed a subgroup analysis including intracardiac electrogram-based (EGM) algorithms and contractility-based algorithms. Results: Nine RCTs with 8531 participants were included, of whom 4275 (50.1%) were randomized to automatic algorithm. Seven of the 9 trials used EGM-based algorithms, and 2 used contractility sensors. There was no difference in all-cause mortality (10.3% vs 11.3%; odds ratio [OR] 0.90; 95% confidence interval [CI] 0.71–1.03; P = .13; I2 = 0%) or heart failure hospitalizations (15.0% vs 16.1%; OR 0.924; 95% CI 0.81–1.04; P = .194; I2 = 0%) between the automatic algorithm group and the control group. Study-defined clinical improvement was also not significantly different between groups (66.6% vs 63.3%; risk ratio 1.01; 95% CI 0.95–1.06; P = .82; I2 = 50%). In the contractility-based subgroup, there was a trend toward greater clinical improvement with the use of the automatic algorithm (75% vs 68.3%; OR 1.45; 95% CI 0.97–2.18; P = .07; I2 = 40%), which did not reach statistical significance. The overall risk of bias was low. Conclusion: Automatic algorithms that change AV or VV intervals did not improve mortality, heart failure hospitalizations, or cardiovascular symptoms in patients with heart failure and CRT.

Introduction In Australia, only 22% of male and 8% of female adolescents meet the muscle-strengthening physical activity guidelines, and few school-based interventions support participation in resistance training (RT). After promising findings from our effectiveness trial, we conducted a state-wide dissemination of the ‘Resistance Training for Teens’ (RT4T) intervention from 2015 to 2020. Despite high estimated reach, we found considerable variability in programme delivery and teachers reported numerous barriers to implementation. Supporting schools when they first adopt evidence-based programmes may strengthen programme fidelity, sustainability, and by extension, programme impact. However, the most effective implementation support model for RT4T is unclear.Objective To compare the effects of three implementation support models on the reach (primary outcome), dose delivered, fidelity, sustainability, impact and cost of RT4T.Methods and analysis We will conduct a hybrid type III implementation–effectiveness trial involving grade 9 and 10 (aged 14–16 years) students from 90 secondary schools in New South Wales (NSW), Australia. Schools will be recruited across one cohort in 2023, stratified by school type, socioeconomic status and location, and randomised in a 1:1:1 ratio to receive one of the following levels of implementation support: (1) ‘low’ (training and resources), (2) ‘moderate’ (training and resources+external support) or ‘high’ (training and resources+external support+equipment). Training includes a teacher workshop related to RT4T programme content (theory and practical sessions) and the related resources. Additional support will be provided by trained project officers from five local health districts. Equipment will consist of a pack of semiportable RT equipment (ie, weighted bars, dumbbells, resistance bands and inverted pull up bar stands) valued at ~$A1000 per school. Study outcomes will be assessed at baseline (T0), 6 months (T1) and 18 months (T2). A range of quantitative (teacher logs, observations and teacher surveys) and qualitative (semistructured interviews with teachers) methods will be used to assess primary (reach) and secondary outcomes (dose delivered, fidelity, sustainability, impact and cost of RT4T). BackgroundStereotactic arrhythmia radioablation (STAR) is a potential new therapy for patients with refractory ventricular tachycardia (VT). The arrhythmogenic substrate (target) is synthesized from clinical and electro-anatomical information. This study was designed to evaluate the baseline interobserver variability in target delineation for STAR.MethodsDelineation software designed for research purposes was used. The study was split into three phases. Firstly, electrophysiologists delineated a well-defined structure in three patients (spinal canal). Secondly, observers delineated the VT-target in three patients based on case descriptions. To evaluate baseline performance, a basic workflow approach was used, no advanced techniques were allowed. Thirdly, observers delineated three predefined segments from the 17-segment model. Interobserver variability was evaluated by assessing volumes, variation in distance to the median volume expressed by the root-mean-square of the standard deviation (RMS-SD) over the target volume, and the Dice-coefficient.ResultsTen electrophysiologists completed the study. For the first phase interobserver variability was low as indicated by low variation in distance to the median volume (RMS-SD range: 0.02–0.02 cm) and high Dice-coefficients (mean: 0.97 ± 0.01). In the second phase distance to the median volume was large (RMS-SD range: 0.52–1.02 cm) and the Dice-coefficients low (mean: 0.40 ± 0.15). In the third phase, similar results were observed (RMS-SD range: 0.51–1.55 cm, Dice-coefficient mean: 0.31 ± 0.21).ConclusionsInterobserver variability is high for manual delineation of the VT-target and ventricular segments. This evaluation of the baseline observer variation shows that there is a need for methods and tools to improve variability and allows for future comparison of interventions aiming to reduce observer variation, for STAR but possibly also for catheter ablation.

Dietary flavanols are known for disease preventative properties but are often poorly absorbed. Gut microbiome flavanol metabolites are more bioavailable and may exert protective activities. Using metabolite mixtures extracted from the urine of rats supplemented with flavanols and treated with or without antibiotics, we investigated their effects on INS-1 832/13 β-cell glucose stimulated insulin secretion (GSIS) capacity. We measured insulin secretion under non-stimulatory (low) and stimulatory (high) glucose levels, insulin secretion fold induction, and total insulin content. We conducted treatment-level comparisons, individual-level dose responses, and a responder vs. non-responder predictive analysis of metabolite composition. While the first two analyses did not elucidate treatment effects, metabolites from 9 of the 28 animals demonstrated significant dose responses, regardless of treatment. Differentiation of responders vs. non-responder revealed that levels of native flavanols and valerolactones approached significance for predicting enhanced GSIS, regardless of treatment. Although treatment-level patterns were not discernable, we conclude that the high inter-individual variability shows that metabolite bioactivity on GSIS capacity is less related to flavanol supplementation or antibiotic treatment and may be more associated with the unique microbiome or metabolome of each animal. These findings suggest flavanol metabolite activities are individualized and point to the need for personalized nutrition practices.

This cross-sectional study investigated how stretch-shortening cycle (SSC) function and kinetic variables differed between young female gymnasts of varying competitive levels. Drop jump (DJ) force–time profiles were examined in 118 female gymnasts, sub-divided by competitive level (n = 21 recreational, n = 41 regional and n = 50 elite). DJ force–time data were analyzed to calculate performance and kinetic variables. Participants’ SSC function was categorized as poor, moderate, or good, depending on the presence of an impact peak and spring-like behavior. A high proportion of gymnasts across each group were categorized as having “good” or “moderate” SSC function (i.e., >94.8%), with a trend of increasingly better SSC function observed with competitive level. Significant differences in reactive strength index, contact time, time of landing peak force, relative propulsive peak force, impulse, and ratio of braking: propulsive impulse were found between the elite and recreational group (p < 0.05). While SSC function was generally good to moderate, elite gymnasts had a more desirable kinetic jump-landing strategy than recreational level gymnasts. Drop jump kinetic variables appear to distinguish between elite and recreational gymnasts but not between regional standard gymnasts. Practitioners should consider the kinetic profile of gymnasts when benchmarking and setting training objectives.

The aim of this review was to describe and summarize the scientific literature on programming parameters related to jump or plyometric training in male and female soccer players of different ages and fitness levels. A literature search was conducted in the electronic databases PubMed, Web of Science and Scopus using keywords related to the main topic of this study (e.g., “ballistic” and “plyometric”). According to the PICOS framework, the population for the review was restricted to soccer players, involved in jump or plyometric training. Among 7556 identified studies, 90 were eligible for inclusion. Only 12 studies were found for females. Most studies (n = 52) were conducted with youth male players. Moreover, only 35 studies determined the effectiveness of a given jump training programming factor. We have constrained syneruptive pressure-temperature-time (P-T-t) paths of mafic magmas using a combination of short-timescale cooling and decompression chronometers. Recent work has shown that the thermal histories of crystals in the last few seconds to hours of eruption can be constrained using concentration gradients of MgO inside olivine-hosted melt inclusions, produced in response to syneruptive cooling and crystallization of olivine on the inclusion walls. We have applied this technique to the study of melt inclusions erupted by arc and ocean island volcanoes, including the 1974 subplinian eruption of Fuego volcano; the 1977 fire-fountain eruption of Seguam volcano; and three eruptions of Kilauea volcano (episode 1 of the 1959 Kilauea Iki fire-fountain eruption, the 1500 CE vigorous fire-fountain eruption, and the 1650 CE subplinian eruption). Of the eruptions studied so far, melt inclusions from the 1959 Kilauea Iki eruption record the highest syneruptive cooling rates (3–11°C/s) and the shortest cooling durations (4–19 s), while inclusions from the 1974 Fuego eruption record the slowest cooling rates (0.1–1.7°C/s) and longest cooling durations (21–368 s). The high cooling rates inferred for the Kilauea Iki and Seguam fire fountain eruptions are consistent with air quenching over tens of seconds during and after fragmentation and eruption. Melt inclusions sampled from the interiors of small (∼6 cm diameter) volcanic bombs at Fuego are found to have cooled more slowly on average than inclusions sampled from ash (with particle diameters < 2 mm) during the same eruption, as expected based on conductive cooling models. We find evidence for a systematic relationship between cooling rates and decompression rates of magmas, in which rapidly ascending gas-bearing magmas experience slower cooling during ascent and eruption than slowly ascending magmas. Our magma P-T-t constraints for the Kilauea Iki eruption are in broad agreement with isentropic models that show that the dominant driver of cooling in the conduit is adiabatic expansion of a vapor phase; however, at Fuego and Seguam, our results suggest a significant role for latent heat production and/or open-system degassing (both of which violate assumptions required for isentropic ascent). We thereby caution against the application of isentropic conduit models to magmas containing relatively high initial water concentrations (e.g., arc magmas containing ∼4 wt% water). We note that several processes that have been inferred to occur in volcanic conduits such as magma stalling, magma mingling, open- and closed-system degassing, vapor fluxing, and vapor accumulation (in foam layers or as slugs of gas) are associated with different implied vapor volume fractions during syneruptive ascent. Given the sensitivity of magma P-T-t paths to vapor volume fraction, the syneruptive thermometer presented here may be a means of identifying these processes during the seconds to hours preceding the eruption of mafic magmas.

This study aimed to: (1) examine differences in physical performance across birth-quartiles and maturity-status, and (2) determine the relationships among relative age, maturation and physical performance in young male soccer players. The sample included 199 males aged between 8.1 and 18.9 years, from two professional soccer academies in the English Football League. Data were collected for height, weight, self-reported biological parent heights, 30 m sprint time and countermovement jump (CMJ) height. Relative age was conveyed as a decimal, while maturity status was determined as the percentage of predicted adult height (PAH). There were no significant differences in any measure between birth quartiles, however early maturers outperformed on-time and later maturers in most performance measures. Pearson-product-moment correlations revealed that maturation was inversely associated with 30 m sprint time in U12 to U16 (r = −0.370–0.738; p < 0.05), but only positively associated with CMJ performance in U12 (r = 0.497; p < 0.05). In contrast, relative age was unrelated to sprint performance and only significantly associated with superior CMJ performance in U16. This study indicates that maturity has a greater association with sprint performance than relative age in English male academy soccer players. Practitioners should monitor and assess biological maturation in young soccer players to attempt to control for the influence on physical performance, and avoid biasing selection on absolute performance rather than identifying the most talented player.

The purpose of this article is to provide an overview of the growth, maturation and resistance training-related changes in muscle-tendon and neuromuscular mechanisms in youth, and the subsequent effect on performance. Sprinting, jumping, kicking, and throwing are common movements in sport that have been shown to develop naturally with age, with improvements in performance being attributed to growth and maturity-related changes in neuromuscular mechanisms. These changes include moderate to very large increases in muscle physiological cross-sectional area (CSA), muscle volume and thickness, tendon CSA and stiffness, fascicle length, muscle activation, pre-activation, stretch reflex control accompanied by large reductions in electro-mechanical delay and co-contraction. Furthermore, a limited number of training studies examining neuromuscular changes following four to 20 weeks of resistance training have reported trivial to moderate differences in tendon stiffness, muscle CSA, muscle thickness, and motor unit activation accompanied by reductions in electromechanical delay (EMD) in pre-pubertal children. However, the interaction of maturity- and training-related neuromuscular adaptions remains unclear. An understanding of how different neuromuscular mechanisms adapt in response to growth, maturation and training is important in order to optimise training responsiveness in youth populations. Additionally, the impact that these muscle-tendon and neuromuscular changes have on force producing capabilities underpinning performance is unclear.

Leading global physical activity guidelines advocate that young children need to engage in activities that strengthen musculoskeletal tissues and improve movement skill competency. The purpose of this study was to examine the effects of delivering strength and conditioning as part of the physical education curriculum on athletic motor skill competencies (AMSC), physical performance, and psychosocial factors. Forty-six school children aged 11–14 were included in the study, and sub-divided firstly by sex and then into intervention and control groups. Intervention groups received nine lessons of strength and conditioning based activities over a six-week period, while the control groups continued with traditional physical education curricula. The resistance training skills battery (RTSB) and tuck jump assessment (TJA) assessed AMSC. Standing long jump distance assessed lower limb strength, and online surveys examined motivation, physical self-efficacy and self-esteem. Male and female intervention groups significantly improved RTSB (p > 0.05) whereas no changes were observed in the control groups. No changes were observed in the intervention groups TJA and only trivial and small non-significant changes in standing long jump performance. Significant increases in motivation of the male intervention group occurred. Strength and conditioning integrated in physical education can improve AMSC in short-term interventions.

The purpose of this study was to examine the effects of a neuromuscular training (NMT) program on lower-extremity biomechanics in youth female netball athletes. The hypothesis was that significant improvements would be found in landing biomechanics of the lower-extremities, commonly associated with anterior cruciate ligament (ACL) injury, following NMT. Twenty-three athletes (age = 12.2 ± 0.9 years; height = 1.63 ± 0.08 m; mass = 51.8 ± 8.5 kg) completed two testing sessions separated by 7-weeks and were randomly assigned to either a experimental or control group. Thirteen athletes underwent 6-weeks of NMT, while the remaining 10 served as controls and continued their regular netball training. Three-dimensional lower-extremity kinematics and vertical ground reaction force (VGRF) were measured during two landing tasks, a drop vertical jump and a double leg broad jump with a single leg landing. The experimental group significantly increased bilateral knee marker distance during the bilateral landing task at maximum knee-flexion range of motion. Knee internal rotation angle during the unilateral landing task at maximum knee flexion-extension range of motion was significantly reduced (p ≤ 0.05, g > 1.00). The experimental group showed large, significant decreases in peak vertical ground reaction force in both landing tasks (p ≤ 0.05, g > −1.30). Control participants did not demonstrate any significant pre-to-post-test changes in response to the 6-week study period. Results of the study affirm the hypothesis that a 6-week NMT program can enhance landing biomechanics associated with ACL injury in 11–13 year old female netball athletes.
