1. The underground argon project: procurement and purification of argon for dark matter searches and beyond

Abstract

The existence of dark matter in the universe is inferred from abundant astrophysical and cosmological observations. The Global Argon Dark Matter Collaboration (GADMC) aims to perform the searches for dark matter in the form of weakly interacting massive particles (WIMPs), whose collisions with argon nuclei would produce nuclear recoils with tens of keV energy. Argon has been considered an excellent medium for the direct detection of WIMPs as argon-based scintillation detectors can make use of pulse shape discrimination (PSD) to separate WIMP-induced nuclear recoil signals from electron recoil backgrounds with extremely high efficiency. However, argon-based direct dark matter searches must confront the presence of intrinsic 39Ar as the predominant source of electron recoil backgrounds (it is a beta-emitter with an endpoint energy of 565 keV and half-life of 269 years). Even with PSD, the 39Ar activity in atmospheric argon (AAr), mainly produced and maintained by cosmic ray-induced nuclear reactions, limits the ultimate size of argon-based detectors and restricts their ability to probe very-low-energy events. The discovery of argon from deep underground wells with significantly less 39Ar than that in AAr was an important step in the development of direct dark matter detection experiments using argon as the active target. Thanks to pioneering research and successful R&D, in 2012, the first 160 kg batch of underground argon (UAr) was extracted from a CO2 well in Cortez, Colorado. The DarkSide-50 experiment at the Gran Sasso National Laboratory (LNGS) in Italy, the first liquid argon detector ever operated with a UAr target, demonstrated a ∼ 1,400 suppression of the 39Ar activity with respect to the atmospheric argon. An even larger suppression is expected for 42Ar (another intrinsic beta-emitter with the 42K daughter isotope, also a beta-emitter) as its production is expected mainly in the upper atmosphere. Following the results of DarkSide-50, the GADMC initiated the UAr project for extraction from underground and cryogenic purification of 100 t of argon to be used as a target in the next-generation experiment DarkSide-20k. This paper contains a description of the Urania Plant in Cortez, Colorado, where UAr is extracted; the Aria Plant in Sardinia, Italy, an industrial-scale plant comprising a 350-m state-of-the-art cryogenic isotopic distillation column, designed for further purification of the extracted argon and further reduction of the isotopic abundance of 39Ar; and DArT, a facility for UAr radiopurity qualification at the Canfranc Underground Laboratory (LSC), Spain. Moreover, the high radiopurity of UAr leads to other possible applications, for instance, for those neutrinoless double-beta decay experiments using argon as shielding material or, more generally, for all those activities on argon-based detectors in high-energy physics or nuclear physics, which will be briefly discussed.

Abstract

Abstract The Aria cryogenic distillation plant, located in Sardinia, Italy, is a key component of the DarkSide-20k experimental program for WIMP dark matter searches at the INFN Laboratori Nazionali del Gran Sasso, Italy. Aria is designed to purify the argon, extracted from underground wells in Colorado, USA, and used as the DarkSide-20k target material, to detector-grade quality. In this paper, we report the first measurement of argon isotopic separation by distillation with the 26 m tall Aria prototype. We discuss the measurement of the operating parameters of the column and the observation of the simultaneous separation of the three stable argon isotopes: $${}^{36}\hbox {Ar}$$ 36 Ar , $${}^{38}\textrm{Ar}$$ 38 Ar , and $${}^{40}\textrm{Ar}$$ 40 Ar . We also provide a detailed comparison of the experimental results with commercial process simulation software. This measurement of isotopic separation of argon is a significant achievement for the project, building on the success of the initial demonstration of isotopic separation of nitrogen using the same equipment in 2019.

Abstract

Abstract Aria is a plant hosting a $${350}\,\hbox {m}$$ 350 m cryogenic isotopic distillation column, the tallest ever built, which is being installed in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Aria is one of the pillars of the argon dark-matter search experimental program, lead by the Global Argon Dark Matter Collaboration. It was designed to reduce the isotopic abundance of $${^{39}\hbox {Ar}}$$ 39 Ar in argon extracted from underground sources, called Underground Argon (UAr), which is used for dark-matter searches. Indeed, $${^{39}\hbox {Ar}}$$ 39 Ar is a $$\beta $$ β -emitter of cosmogenic origin, whose activity poses background and pile-up concerns in the detectors. In this paper, we discuss the requirements, design, construction, tests, and projected performance of the plant for the isotopic cryogenic distillation of argon. We also present the successful results of the isotopic cryogenic distillation of nitrogen with a prototype plant.

Abstract

Abstract Proportional electroluminescence (EL) in noble gases is used in two-phase detectors for dark matter searches to record (in the gas phase) the ionization signal induced by particle scattering in the liquid phase. The “standard” EL mechanism is considered to be due to noble gas excimer emission in the vacuum ultraviolet (VUV). In addition, there are two alternative mechanisms, producing light in the visible and near infrared (NIR) ranges. The first is due to bremsstrahlung of electrons scattered on neutral atoms (“neutral bremsstrahlung”, NBrS). The second, responsible for electron avalanche scintillation in the NIR at higher electric fields, is due to transitions between excited atomic states. In this work, we have for the first time demonstrated two alternative techniques of the optical readout of two-phase argon detectors, in the visible and NIR range, using a silicon photomultiplier matrix and electroluminescence due to either neutral bremsstrahlung or avalanche scintillation. The amplitude yield and position resolution were measured for these readout techniques, which allowed to assess the detection threshold for electron and nuclear recoils in two-phase argon detectors for dark matter searches. To the best of our knowledge, this is the first practical application of the NBrS effect in detection science.

Abstract

In this paper, an innovative approach based on an artificial neural network (ANN) load forecasting model to improve the distribution system state estimation accuracy is proposed. High-quality pseudomeasurements are produced by a neural model fed with both exogenous and historical load information and applied in a realistic measurement scenario. Aggregated active and reactive powers of small or medium enterprises and residential loads are simultaneously predicted by a one-step ahead forecast. The correlation between the forecasted real and reactive power errors is duly kept into account in the definition of the estimator together with the uncertainty of the overall measurement chain. The beneficial effects of the ANN-based pseudomeasurements on the quality of the state estimation are demonstrated by simulations carried out on a small medium-voltage distribution grid.

Abstract

The detection of small exoplanets with the radial velocity (RV) technique is limited by various poorly known noise sources of instrumental and stellar origin. As a consequence, current detection techniques often fail to provide reliable estimates of the significance levels of detection tests (p-values). We designed an RV detection procedure that provides reliable p-value estimates while accounting for the various noise sources. The method can incorporate ancillary information about the noise (e.g., stellar activity indicators) and specific data- or context-driven data (e.g., instrumental measurements, simulations of stellar variability) . The detection part of the procedure uses a detection test that is applied to a standardized periodogram. Standardization allows an autocalibration of the noise sources with partially unknown statistics. The estimation of the p-value of the test output is based on dedicated Monte Carlo simulations that allow handling unknown parameters. The procedure is versatile in the sense that the specific pair (periodogram and test) is chosen by the user. We demonstrate by extensive numerical experiments on synthetic and real RV data from the Sun and aCenB that the proposed method reliably allows estimating the p-values. The method also provides a way to evaluate the dependence of the estimated p-values that are attributed to a reported detection on modeling errors. It is a critical point for RV planet detection at low signal-to-noise ratio to evaluate this dependence. The python algorithms are available on GitHub. Accurate estimation of p-values when unknown parameters are involved is an important but only recently addressed question in the field of RV detection. Abstract

Delrez, L., et al., Exoplanets transiting bright nearby stars are key objects for advancing our knowledge of planetary formation and evolution. The wealth of photons from the host star gives detailed access to the atmospheric, interior and orbital properties of the planetary companions. ν Lupi (HD 136352) is a naked-eye (V = 5.78) Sun-like star that was discovered to host three low-mass planets with orbital periods of 11.6, 27.6 and 107.6 d via radial-velocity monitoring. The two inner planets (b and c) were recently found to transit, prompting a photometric follow-up by the brand new Characterising Exoplanets Satellite (CHEOPS). Here, we report that the outer planet d is also transiting, and measure its radius and mass to be 2.56 ± 0.09 R and 8.82 ± 0.94 M, respectively. With its bright Sun-like star, long period and mild irradiation (~5.7 times the irradiation of Earth), ν Lupi d unlocks a completely new region in the parameter space of exoplanets amenable to detailed characterization. We refine the properties of all three planets: planet b probably has a rocky mostly dry composition, while planets c and d seem to have retained small hydrogen–helium envelopes and a possibly large water fraction. This diversity of planetary compositions makes the ν Lupi system an excellent laboratory for testing formation and evolution models of low-mass planets., The MOC activities have been supported by ESA contract 4000124370. Abstract

Morris, B. M. et al., [Context] 55 Cnc e is a transiting super-Earth (radius 1.88 R⊕ and mass 8 M⊕) orbiting a G8V host star on a 17-h orbit. Spitzer observations of the planet's phase curve at 4.5 μm revealed a time-varying occultation depth, and MOST optical observations are consistent with a time-varying phase curve amplitude and phase offset of maximum light. Both broadband and high-resolution spectroscopic analyses are consistent with either a high mean molecular weight atmosphere or no atmosphere for planet e. A long-term photometric monitoring campaign on an independent optical telescope is needed to probe the variability in this system. [Aims] We seek to measure the phase variations of 55 Cnc e with a broadband optical filter with the 30 cm effective aperture space telescope CHEOPS and explore how the precision photometry narrows down the range of possible scenarios. [Methods] We observed 55 Cnc for 1.6 orbital phases in March of 2020. We designed a phase curve detrending toolkit for CHEOPS photometry which allowed us to study the underlying flux variations in the 55 Cnc system. [Results] We detected a phase variation with a full-amplitude of 72 ± 7 ppm, but did not detect a significant secondary eclipse of the planet. The shape of the phase variation resembles that of a piecewise-Lambertian; however, the non-detection of the planetary secondary eclipse, and the large amplitude of the variations exclude reflection from the planetary surface as a possible origin of the observed phase variations. They are also likely incompatible with magnetospheric interactions between the star and planet, but may imply that circumplanetary or circumstellar material modulate the flux of the system. [Conclusions] This year, further precision photometry of 55 Cnc from CHEOPS will measure variations in the phase curve amplitude and shape over time., Centre of Competence in Research (NCCR) supported by the Swiss National Science Foundation (SNSF). Abstract

M. J. Hooton et al., [Context] The light curves of tidally locked hot Jupiters transiting fast-rotating, early-type stars are a rich source of information about both the planet and star, with full-phase coverage enabling a detailed atmospheric characterisation of the planet. Although it is possible to determine the true spin–orbit angle Ψ – a notoriously difficult parameter to measure – from any transit asymmetry resulting from gravity darkening induced by the stellar rotation, the correlations that exist between the transit parameters have led to large disagreements in published values of Ψ for some systems., [Aims] We aimed to study these phenomena in the light curves of the ultra-hot Jupiter MASCARA-1 b, which is characteristically similar to well-studied contemporaries such as KELT-9 b and WASP-33 b., [Methods] We obtained optical CHaracterising ExOPlanet Satellite (CHEOPS) transit and occultation light curves of MASCARA-1 b, and analysed them jointly with a Spitzer/IRAC 4.5 μm full-phase curve to model the asymmetric transits, occultations, and phase-dependent flux modulation. For the latter, we employed a novel physics-driven approach to jointly fit the phase modulation by generating a single 2D temperature map and integrating it over the two bandpasses as a function of phase to account for the differing planet–star flux contrasts. The reflected light component was modelled using the general ab initio solution for a semi-infinite atmosphere., [Results] When fitting the CHEOPS and Spitzer transits together, the degeneracies are greatly diminished and return results consistent with previously published Doppler tomography. Placing priors informed by the tomography achieves even better precision, allowing a determination of Ψ = 72.1−2.4+2.5 deg. From the occultations and phase variations, we derived dayside and nightside temperatures of 3062−68+66 K and 1720 ± 330 K, respectively.Our retrieval suggests that the dayside emission spectrum closely follows that of a blackbody. As the CHEOPS occultation is too deep to be attributed to blackbody flux alone, we could separately derive geometric albedo Ag = 0.171−0.068+0.066 and spherical albedo As = 0.266−0.100+0.097 from the CHEOPS data, and Bond albedoAB = 0.057−0.101+0.083 from the Spitzer phase curve.Although small, the Ag and As indicate that MASCARA-1 b is more reflective than most other ultra-hot Jupiters, where H− absorption is expected to dominate., [Conclusions] Where possible, priors informed by Doppler tomography should be used when fitting transits of fast-rotating stars, though multi-colour photometry may also unlock an accurate measurement of Ψ. Abstract

Large liquid argon detectors offer one of the best avenues for the detection of galactic weakly interacting massive particles (WIMPs) via their scattering on atomic nuclei. The liquid argon target allows exquisite discrimination between nuclear and electron recoil signals via pulse-shape discrimination of the scintillation signals. Atmospheric argon (AAr), however, has a naturally occurring radioactive isotope, $^{39}$Ar, a $\beta$ emitter of cosmogenic origin. For large detectors, the atmospheric $^{39}$Ar activity poses pile-up concerns. The use of argon extracted from underground wells, deprived of $^{39}$Ar, is key to the physics potential of these experiments. The DarkSide-20k dark matter search experiment will operate a dual-phase time projection chamber with 50 tonnes of radio-pure underground argon (UAr), that was shown to be depleted of $^{39}$Ar with respect to AAr by a factor larger than 1400. Assessing the $^{39}$Ar content of the UAr during extraction is crucial for the success of DarkSide-20k, as well as for future experiments of the Global Argon Dark Matter Collaboration (GADMC). This will be carried out by the DArT in ArDM experiment, a small chamber made with extremely radio-pure materials that will be placed at the centre of the ArDM detector, in the Canfranc Underground Laboratory (LSC) in Spain. The ArDM LAr volume acts as an active veto for background radioactivity, mostly $\gamma$-rays from the ArDM detector materials and the surrounding rock. Abstract

In photometry, the short-timescale stellar variability ("flicker"), such as that caused by granulation and oscillations, can reach amplitudes comparable to the transit depth of Earth-sized planets and is correlated over the typical transit timescales. It can introduce systematic errors on the inferred planetary parameters when a small number of transits are observed. The objective of this paper is to characterize the statistical properties of this noise and quantify its impact on the inferred transit parameters. We used the extensive solar observations obtained with SoHO/VIRGO to characterize flicker noise. We simulated realistic transits across the solar disk using SDO/HMI data and used these to obtain transit light curves, which we used to estimate the errors made on the transit parameters. We make these light curves publicly available. To extend the study to a wider parameter range, we derived the properties of flicker noise using Kepler observations and studied their dependence on stellar parameters. Finally, we predicted the limiting stellar apparent magnitude for which the properties of the flicker noise can be extracted using high-precision CHEOPS and PLATO observations. Stellar granulation is a stochastic colored noise, and is stationary with respect to the stellar magnetic cycle. Both the flicker correlation timescales and amplitudes increase with the stellar mass and radius. If these correlations are not taken into account when fitting for the parameters of transiting exoplanets, this can bias the inferred parameters. In particular, we find errors of up to 10$\%$ on $R_p/R_s$ for an Earth-sized planet orbiting a Sun-like star. For F and G stars, flicker will significantly affect the inferred parameters of transits observed with CHEOPS and PLATO. Abstract

Biology as an integral part of science learning continues to be maximized by integrating technology into its learning design. The definite manifestation of this integration process is to achieve meaningfulness, activeness, and effectiveness of the learning carried out. The flipped classroom (FC) is gaining popularity as a pedagogical model that uses the important benefits of technology in its learning environment. FC is proven to be an effective teaching mode that empowers students' 21st-century skills, stimulates students' self-learning, and improves academic performance. The potential of the FC is interesting to explore to what extent biology welcomes the FC. The aimed of this systematic literature review is to review and analyze articles published by Scopus indexed journals. We used the keywords "Flipped classroom and biology" in the search menu in the Scopus database and succeeded in finding 72 articles. There were 21 articles that met the criteria for further analysis. The inclusion and exclusion model used is PRISMA. The findings obtained are the trend of FC publications in biology learning over the last ten years. The approaches used in this research include quantitative, mixed method, case study and qualitative. Key words that are often used by writers are student, human, biology. The biology keyword has a branch, namely active learning. There are 9 countries of origin of authors who publish articles. The United States is a country that is active in publishing. Articles published on each continent are not evenly distributed. The dominant articles are published in collaboration with one country and rarely are articles published by one author. We have succeeded in formulating a picture of the existence of the FC in biology learning over the last ten years. These findings can be a consideration or reference for researchers to explore and study more deeply research related to the FC in biology learning based on their respective urgency.

Opinion discourse that contained in a rubric of mass media can become an alternative media of bahasa Indonesia learning in schools, especially in opinion discourse material . One of the elements which need to be researched in opinion discourse is cohesion aspect. Cohesive subject of discourse can be determined by grammatical and lexical aspect. Grammatical cohesion aspect is characterized by the conjunction, ellipsis, substitution, and the reference. Lexical cohesion among other aspects of the reiteration and collocation. In order to get maximum results, these studies only focuses on one of grammatical cohesion aspect, i.e. conjunction aspect. Based on the analysis of the data that has been carried out, from 16 opinion discourse, there is 48 paragraph of samples with 95 pairs of adjacent obtained the result that 10 pairs were using grammatical cohesion in the form of the conjunction is around 10.5% and 85 pairs of adjacent sentences that not use the cohesion in the form of the conjunction is about 89,4%. However, it doesn't mean that the adjecent pair is not cohesive in grammatical basis, considering there were still 3 other factors grammatical cohesion forming beside the conjunction, i.e., reference, substitution and ellipsis.

Abstract

The electronic-module (e-module) is one of the teaching materials needed so that the learning process can take place optimally. E-module owned by UM biology students generally do not use the latest technology. Augmented Reality (AR) is a form of technology where there is a process of integrating the virtual world with the real world. Not a few students experience difficulties in understanding the flower structure material. Student scientific literacy is still far from what is expected. The research conducted is Research and Development with the Lee and Owens Development Model. Data collection was carried out during October-December 2022. The technique of analyzing quantitative descriptive data. The subjects in this study were 37 biology students class of 2021 without any differences in gender, age and study program. The results of the effectiveness of the AR electronic-module based on n-gain calculations obtained a score of 0.7 where on that score the AR e-module was categorized as quite effective in increasing scientific literacy. It is hoped that in the future there will be teaching materials and media that can improve some of the competencies needed by the latest generation.

Abstract

The main goal of this paper is to analyze the behavior of an electric power network in the presence of harmonic distortion, when capacitors are installed. The work starts from a case study, where failure and malfunctioning of an industrial plant are described with the help of experimental measurements. The theoretical and mathematical details of the phenomena involved are analyzed by suitable computer simulations. The study is performed by considering some important electrical quantities, evaluated according to the definitions proposed by the standard IEEE 1459. The results put in evidence that using some typical quantities, such as the power factor, could lead to ambiguous conclusions in evaluating the actual "quality" of the loads, and this can assume special importance in virtue of the economic relevance of such quantities

Abstract

Genista monosperma Lam. can be propagated by herbaceous cuttings, but seasonal fluctuations in rooting ability can be observed. These appear to be correlated with the ratio of two main endogenous phenols, 3-hydroxymandelic acid and luteolin-7-O-glucoside, accumulating in the tissues, the first acting as an inhibitor and the second as a promoter of adventive rhizogenesis. Their biological activity has been tested and evaluated on G. monosperma in vitro micropropagated explants. The inhibitory effect on rooting of 3-hydroxymandelic acid accumulation in cutting basal tissues can be lessened by a soaking treatment with distilled water, applied to the basal part of the cuttings before planting in rooting benches.

The study aims to improve the student skills in writing The Javanese Script and learning activities through group discussion assisted by letter cards in the 6th-grade students of SD 1 Prambatan Kidul Kudus Even Semester Academic Year 2018/2019. This study is a Classroom Action Research (CAR) conducted in two cycles using The Javanese Script cards as the learning media and group discussion as the learning method. Research data were collected using nontest techniques (classroom observation, unstructured interviews, and documentation collection) and test techniques (individual written tests). Quantitative data were the scores of The Javanese Script writing skill were analyzed descriptively and qualitative data were the result of learning activities observations that converted into scores and categorized classically. The Pre-cycle stage test score category consisted of poor (62.16%) and fair scores (37.84%), with students had reached learning mastery standard by 37.84%. The Cycle I stage test score category consisted of poor (27.03%), fair (56.76%), and good scores (16.22%), with students had reached learning mastery standard by 72.97%. The Cycle II stage test score category consisted of fair (48.65%), good (40.54%), and very good scores (10.81%), with students had reached learning mastery standard by 100%. The classical learning activities observations are classified as good, both in Cycle I and Cycle II were 70.27% and 71.89%, respectively. Thus, the method of group discussion assisted by letter cards is proven to improve the student skills in writing The Javanese Script and learning activities.

Kondisi struktur pelat tongkang dan banyak volume muatan, akan mempengaruhi jumlah tegangan yang mampu ditahan oleh suatu struktur. Analisa kekuatan deck dengan variasi pembebanan bertujuan untuk mengetahui tegangan maksimum yang berada di titik tertentu pada deck kapal, pada studi kasus ini adalah kapal tongkang Nania dengan adanya variasi metode pembebanan. Metode penelitian dimulai dengan pembebanan batu bara di atas tongkang dan membentuk gunungan yang jumlahnya bervariasi serta disusun secara memanjang kapal. Pemodelan elemen hingga hanya dilakukan pada daerah midship dengan ukuran model adalah panjang 18,28 meter, lebar 27,45 meter, dan tinggi 5,5 meter, yang disesuaikan dengan studi kasus di lapangan. Variasi pembenanan dilakukan untuk mendapatkan nilai tegangan maksimum, antara lain: variasi ke-1 adalah midship kapal dibebani 1 gunungan yaitu ketika tongkang membawa 4 gunungan batu bara keseluruhan sepanjang kapal, dengan tiap gunungan menyerupai prisma. Variasi ke-2 adalah 2 gunungan pada midship jika tongkang mengangkut 5 gunungan batu bara secara keseluruhan dengan bentuk gunungan adalah limas. Dengan menggunakan software elemen hingga didapatkan hasil tegangan maksimum berada pada area replating pada kedua variasi yang memiliki tegangan maksimum sebesar 394,39 MPa dan 369,97 MPa. Kedalaman deformasi dari kedua tipe pembebanan yaitu 27,783 mm dan 28,765 mm yang masih dibawah toleransi deformasi maksimum (36 mm). Nilai safety factor berdasarkan tegangan ultimate sertifikat material kapal Nania ialah 1,303 dan 1,389. Nilai rata-rata safety factor dari keseluruhan percobaan yaitu 1,346, diatas angka standar SF =1.

Abstract

Teacher-centered learning has no ability in empowering students’ science process skills and interpersonal intelligence. This study aimed at analyzing the effect of guided inquiry on students’ Science Process Skills (SPS) and Interpersonal Intelligence (II). This quasi-experiment used nonequivalent control group design. This study involved control and experimental classes of eleventh graders in biology class in which the material learnt was respiratory system. The SPS data was collected using test instrument, whereas the II data was gained using observation sheet. The analysis results informed that the students who joint the guided inquiry class possessed the better competencies, both in their SPS (p = 0.04) and II (p = 0.001). Therefore, the guided inquiry significantly contributes toward the improvement of students’ SPS and II. The findings of this research can be the considerable information to creat the learning which acommodates students’ SPS and II.

ABSTRACT: The study examines fat and water content, and the level of pesticide residues in rabbit’s meat which were fed cabbage leaves waste. The materials used in the study were 12 New Zealand White rabbits raised from 1.5 to 3 months of age. Those rabbits were divided into 3 different groups (large, medium and small) based on their initial body weight. The laboratory test shows there were several types of pesticide residues in the cabbage leaves and rabbit’s meat namely endosulfan, profenofos and chlorpyrifos. The lowest pesticide residues were found at small rabbit’s meat groups. In contrast, the fat content as well as the water content was found high at large rabbit’s meat groups. Abstract

The use of Solid Oxide Fuel Cell (SOFC) has created various interest in many parties, due to its capability to convert gases into electricity. The main requirement of SOFC cell components is to be produced as thin as possible to minimize the losses of electrical resistance, as well as able to support internal and external loads. This paper discusses the procedure of making a thin electrolyte layer, as well as a porous thin layer cathode using atmospheric spraying technique. The procedure of spraying was in room temperature with the process of sintering at temperature of 13500 C held for 3 hours. The SOFC characterization of electrolyte and cathode microstructure was determined by using the SEM, FESEM, XRD and impedance spectroscopy, to measure the impedance of SOFC cells. The results show that the thickness of thin layer electrolyte and porous cathode obtained of about 20 µm and 4 µm, respectively. Also the SOFC cell impedance was measured of 2.3726 x 106 Ω at room temperature. The finding also demonstrated that although the materials (anode, cathode and electrolyte) possess different coefficient thermal expansion, there was no evidence of flaking layers which seen the materials remain intact. Thus, the atmospheric spraying method can offer an alternative method to manufacturing of SOFC thin layer electrolyte and cathode. [Key words: SOFC; spraying method; electrolyte; cathode]

Objectives: Herein, we investigate the relationships between the COVID-19 pandemic and overcoming the virus, and its effects on body composition parameters in young adults from Slovakia., Methods: We assessed 773 adults aged 18 to 30 years in pandemic-status sub-groups. Individual lifestyles and overcoming the COVID-19 effects were evaluated by a detailed questionnaire, and body composition parameters were analyzed using the InBody 770 bioimpedance analyzer., Results: Statistically significant lower values were observed in the male group during the pandemic for the following parameters; proteins and minerals, fat free mass (FFM), skeletal muscle mass (SMM), lean body mass (LBM) and its values in the right and left arm and trunk, total body water (TBW) and its values in the right and left arm and trunk, body cell mass (BCM), basal metabolic rate (BMR) and phase angle (PA). The regression analysis confirmed the negative pandemic effect and the negative impact of COVID-19 on men in the following parameters: proteins (p = .027 for pandemic and p = .005 for COVID-19), FFM (p = .023 for pandemic and p = .005 for COVID-19), LBM (p = .022 for pandemic and p = .004 for COVID-19), SMM (p = .028 for pandemic and p = .005 for COVID-19), TBW (p = .020 for pandemic and p = .004 for COVID-19), BMR (p = .024 for pandemic and p = .005 for COVID-19) and PA (p = .009 for pandemic). Physical activity was a further significant predictor in men for all the above parameters., Conclusions: We observed significantly lower body composition parameters associated with fat free mass in young adult men during the pandemic than before it. Objectives: This study investigates the relationships between the COVID-19 pandemic, lifestyle factors, and their impact on bone mineral density in the radius forearm bone and the total bone mineral content in young adults from Slovakia., Methods: We assessed 773 Slovak young adults aged 18 to 30 years, divided into subgroups on their pandemic status. Bone mineral density (BMD) was analyzed by the QUS device (Sunlight MiniOmni™), and bone mineral content (BMC) and fat mass (FM) were measured by InBody 770 bioimpedance analyzer. Finally, linear regression analysis tested the associations., Results: Statistically significant lower speed of sound (SOS) along the length of the forearm radius bone and Z-score values was determined in participants during the COVID-19 pandemic than before it, and statistically significant lower BMC values were observed in the male group during COVID-19 than beforehand. Regression analysis confirmed the negative pandemic effect in the following indices: SOS (p < .001 for women and p = .035 for men), Z-score (p < .001 for women and p = .003 for men), and BMC (p = .024 for men). Vitamin D was a further significant SOS predictor in women at p = .029, but this association was not detected in men. In contrast, the significant male BMC predictors were pandemic presence (p = .028), physical activity (p = .028), and fat mass percentage (p = .001)., Conclusions: Significant COVID-19 pandemic effects on bone tissue were determined on bone mass density in the radius forearm bone and the total bone mineral content. Abstract

Background: Sarcopenic obesity is a common condition in the elderly associated with excessive adiposity and low muscle mass and strength., Aims: This study aims to establish a method for detecting bioelectrical characteristics in individuals with sarcopenic obesity through specific Bioelectrical Impedance Vector Analysis (specific BIVA), while considering the characteristics of individuals with healthy, sarcopenic, and obese conditions., Methods: The sample was composed by 915 Italian adults over 50 years of age (men: 74.6 ± 8.8 y; women:76.3 ± 8.8 y) living in Sardinia (Italy). A dataset of 1590 US adults aged 21 - 49 years retrieved from the 2003 - 2004 National Health and Nutrition Examination Survey was also considered in a final step of the study. Anthropometric (stature, weight, waist, arm, and calf circumferences) and whole-body bioelectrical variables were taken. In the Italian sample, bioelectrical impedance was applied to estimate the relative content of fat mass and skeletal muscle mass. Groups with healthy body composition (NS-NO), or consistent with sarcopenia (S), sarcopenic obesity (S-O), and obesity (O) were defined based on the cut-offs suggested by European expert guidelines (EWGSOP2 and ESPEN-EASO). Specific BIVA was applied to compare groups and to identify the area for sarcopenic obesity within young-adults tolerance ellipses. The position of the specific vector of US individuals with S-O, selected on the basis of DXA measurements, was also considered., Results: In both sexes of the Italian sample, the bioelectrical characteristics of the four groups were different (p < 0.001). The differences were mainly related to vector length, indicative of higher fat mass, which was longer in the O and S-O groups, and phase angle, a proxy of intracellular/extracellular water and muscle mass, lower in the sarcopenic groups. Bioelectrical vectors of the S-O group fell in the right quadrant, outside of the 95 % tolerance ellipses of young adults. The mean vector of the US sample with S-O fell in the same area. Within the S-O area, women had similar bioelectrical values, while men showed phase angle variability, which was related to the severity of the condition., Conclusions: Specific BIVA detects body composition peculiarities of individuals with sarcopenic obesity, thus allowing their diagnosis when associated with low handgrip strength values., (Copyright © 2024 The Author(s). Abstract

The citrate levels in the bone start to decrease after death. Therefore, it may be possible to estimate the post-mortem interval if the citrate content decreases at regular and predictable intervals. To verify this, it was used twelve fresh porcine tibiae and femora, fifteen recently buried porcine bones, two samples of human bones curated from the anatomical teaching collection from Bournemouth University, UK (never been buried) and fifteen samples of archaeological human bones from Saint Augustine the Less, Bristol, UK. The results obtained in this study align together with the data available in the literature, with higher citrate content in the fresh porcine bones and lower in the archaeological samples, however, a statistical significance was not found. Moreover, the formula used to estimate the time since death largely overestimated the known PMI. Further research should be done with a larger sample and a new formula to estimate the post-mortem interval is required.

Abstract

The signal transducer and activator of transcription 3 (STAT3) protein is a master regulator of most key hallmarks and enablers of cancer, including cell proliferation and the response to DNA damage. G-Quadruplex (G4) structures are four-stranded noncanonical DNA structures enriched at telomeres and oncogenes' promoters. In cancer cells, stabilization of G4 DNAs leads to replication stress and DNA damage accumulation and is therefore considered a promising target for oncotherapy. Here, we designed and synthesized novel quinazoline-based compounds that simultaneously and selectively affect these two well-recognized cancer targets, G4 DNA structures and the STAT3 protein. Using a combination of in vitro assays, NMR, and molecular dynamics simulations, we show that these small, uncharged compounds not only bind to the STAT3 protein but also stabilize G4 structures. In human cultured cells, the compounds inhibit phosphorylation-dependent activation of STAT3 without affecting the antiapoptotic factor STAT1 and cause increased formation of G4 structures, as revealed by the use of a G4 DNA-specific antibody. As a result, treated cells show slower DNA replication, DNA damage checkpoint activation, and an increased apoptotic rate. Importantly, cancer cells are more sensitive to these molecules compared to noncancerous cell lines. This is the first report of a promising class of compounds that not only targets the DNA damage cancer response machinery but also simultaneously inhibits the STAT3-induced cancer cell proliferation, demonstrating a novel approach in cancer therapy.

Here, by using a genetically encoded fluorescent sensor that includes a spectroscopic reference (an element insensitive to Cl - and pH), we show that ratiometric imaging is strongly affected by the optical properties of the brain. We have designed a method that fully corrects for this source of error. Parallel measurements of [Cl - ] i and pH i at the single-cell level in the mouse cortex showed the in vivo presence of the widely discussed developmental fall in [Cl - ] i and the role of the K-Cl cotransporter KCC2 in this process. Abstract

The development of fluorescent indicators represented a revolution for life sciences. Genetically encoded and synthetic fluorophores with sensing abilities allowed the visualization of biologically relevant species with high spatial and temporal resolution. Synthetic dyes are of particular interest thanks to their high tunability and the wide range of measureable analytes. However, these molecules suffer several limitations related to small molecule behavior (poor solubility, difficulties in targeting, often no ratiometric imaging allowed). In this work we introduce the development of dendrimer-based sensors and present a procedure for pH measurement in vitro, in living cells and in vivo. We choose dendrimers as ideal platform for our sensors for their many desirable properties (monodispersity, tunable properties, multivalency) that made them a widely used scaffold for several biomedical devices. The conjugation of fluorescent pH indicators to the dendrimer scaffold led to an enhancement of their sensing performances. In particular dendrimers exhibit reduced cell leakage, improved intracellular targeting and allow ratiometric measurements. These novel sensors were successfully employed to measure pH in living HeLa cells and in vivo in mouse brain.

Abstract

Dendrimers have been described as one of the most tunable and therefore potentially applicable nanoparticles both for diagnostics and therapy. Recently, in order to realize drug delivery agents, most of the effort has been dedicated to the development of dendrimers that could internalize into the cells and target specific intracellular compartments in vitro and in vivo. Here, we describe cell internalization properties and diffusion of G4 and G4-C12 modified PAMAM dendrimers in primary neuronal cultures and in the CNS of live animals. Confocal imaging on primary neurons reveals that dendrimers are able to cross the cell membrane and reach intracellular localization following endocytosis. Moreover, functionalization of PAMAMs has a dramatic effect on their ability to diffuse in the CNS tissue in vivo and penetrate living neurons as shown by intraparenchymal or intraventricular injections. 100 nM G4-C12 PAMAM dendrimer already induces dramatic apoptotic cell death of neurons in vitro. On the contrary, G4 PAMAM does not induce apoptotic cell death of neural cells in the sub-micromolar range of concentration and induces low microglia activation in brain tissue after a week. Our detailed description of dendrimer distribution patterns in the CNS will facilitate the design of tailored nanomaterials in light of future clinical applications.

Abstract

Background: A very high prevalence (22.3%) of gestational diabetes mellitus (GDM) was recently reported following our study on a large group of Sardinian women. In order to explain such a high prevalence we sought to characterise our obstetric population through the analysis of risk factors and their association with the development of GDM., Methods: The prevalence of risk factors and their association with the development of GDM were evaluated in 1103 pregnancies (247 GDM and 856 control women). The association of risk factors with GDM was calculated according to logistic regression. Sensitivity and specificity of risk assessment strategy were also calculated., Results: None of the risk factors evaluated showed an elevated frequency in our population. The high risk patients were 231 (20.9%). Factors with a stronger association with GDM development were obesity (OR 3.7, 95% CI 2.08-6.8), prior GDM (OR 3.1, 95% CI 1.69-5.69), and family history of Type 2 diabetes (OR 2.6, 95% CI 1.81-3.86). Only patients over 35 years of age were more represented in the GDM group (38.2% vs 22.6% in the non-GDM cases, P < 0.001). Type 2 diabetes in second-degree relatives was equally represented in GDM and non-GDM subjects, while prior poor obstetrical outcomes mostly characterized non-GDM women (17.5% vs 10.6%, P < 0.001). The "average risk" assessment better characterized non-GDM patients (76.8% vs 57.8%, P < 0.001). The logistic regression analysis confirmed that Type 2 diabetes in second-degree relatives, prior poor obstetrical outcomes and the "average risk" definition did not predict the development of GDM., Conclusion: Such a high prevalence of GDM in our population does not seem to be related to the abnormal presence of some known risk factors, and appears in contrast with the prevalence of Type 2 diabetes in Sardinia. Further studies are needed to explain the cause such a high prevalence of GDM in Sardinia. The "average risk" definition is not adequate to predict GDM in our population.

Abstract

Background: We previously reported a high prevalence (22.3%) of gestational diabetes mellitus (GDM) in a large group of Sardinian women, in contrast with the prevalence of Type 2 diabetes. Sardinia has an unusual distribution of haplotypes and genotypes, with the highest population frequency of HLA DR3 in the world, and after Finland, the highest prevalence of Type 1 diabetes and Autoimmune-related Diseases. In this study we preliminarily tested the prevalence of serological markers of Type 1 diabetes in a group of Sardinian GDM patients., Methods: We determined glutamic decarboxylase antibodies (anti-GAD65), protein tyrosine phosphatase ICA 512 (IA2) antibodies (anti-IA2), and IAA in 62 GDM patients, and in 56 controls with matching age, gestational age and parity., Results: We found a high prevalence and very unusual distribution of antibodies in GDM patients (38.8%), the anti-IA2 being the most frequent antibody. Out of all our GDM patients, 38.8% (24 of 62) were positive for at least one antibody. Anti-IA2 was present in 29.0 % (18 out of 62) vs. 7.1% (4 out of 56) in the controls (P < 0.001). IAA was present in 14.5% (9 out of 62) of our GDM patients, and absent in the control subjects (P < 0.001). Anti-GAD65 was also present in GDM patients, with a prevalence of 3.2% (2 out of 62) while it was absent in the control group (P = NS). Pre-gestational weight was significantly lower (57.78 +/- 9.8 vs 65.9 +/- 17.3 P = 0.04) in auto-antibodies- positive GDM patients., Conclusion: These results are in contrast with the very low prevalence of all antibodies reported in Italy. If confirmed, they could indicate that a large proportion of GDM patients in Sardinia have an autoimmune origin, in accordance with the high prevalence of Type 1 diabetes.
