Abstract

In the anuran family Dendrobatidae, aposematic species obtain their toxic or unpalatable alkaloids from dietary sources, a process known as sequestering. To understand how toxicity evolved in this family, it is paramount to elucidate the pathways of alkaloid processing (absorption, metabolism, and sequestering). Here, we used an exploratory skin gene expression experiment in which captive-bred dendrobatids were fed alkaloids. Most of these experiments were performed with Dendrobates tinctorius, but some trials were performed with D. auratus, D. leucomelas and Allobates femoralis to explore whether other dendrobatids would show similar patterns of gene expression. We found a consistent pattern of up-regulation of genes related to muscle and mitochondrial processes, probably due to the lack of mutations related to alkaloid resistance in these species. Considering conserved pathways of drug metabolism in vertebrates, we hypothesize alkaloid degradation is a physiological mechanism of resistance, which was evidenced by a strong upregulation of the immune system in D. tinctorius, and of complement C2 across the four species sampled. Probably related to this strong immune response, we found several skin keratins downregulated, which might be linked to a reduction of the cornified layer of the epidermis. Although not conclusive, our results offer candidate genes and testable hypotheses to elucidate alkaloid processing in poison frogs.

Abstract

The interaction between a viral capsid and its genome governs crucial steps in the life cycle of a virus, such as assembly and genome uncoating. Tuning cargo–capsid interactions is also essential for successful design and cargo delivery in engineered viral systems. Here we investigate the interplay between cargo and capsid for the picorna-like Triatoma virus using a combined native mass spectrometry and atomic force microscopy approach. We propose a topology and assembly model in which heterotrimeric pentons that consist of five copies of structural proteins VP1, VP2 and VP3 are the free principal units of assembly. The interpenton contacts are established primarily by VP2. The dual role of the genome is first to stabilize the densely packed virion and, on an increase in pH, second to trigger uncoating by relaxing the stabilizing interactions with the capsid. Uncoating occurs through a labile intermediate state of the virion that reversibly disassembles into pentons with the concomitant release of protein VP4. Fil: Snijder, J.. 24. Phylogeny and classification of fejervaryan frogs (Anura: Dicroglossidae).

Abstract

Systematics and classification of Asian frogs of the genus Fejervarya and related genera (family Dicroglossidae; hereafter referred to as fejervaryan frogs) have been the subject of intensive debates in the past few years. We complement previous phylogenetic studies with analyses of concatenated sequences from 14 nuclear loci and mitochondrial gene fragments, totaling 12,752 nucleotides for 46 species representing all major lineages and relevant outgroups. We find strong support for two major clades within Fejervarya: a South Asian clade and a Southeast Asian clade. Previously, South Asian species have been hypothesized to constitute a separate genus, Zakerana (currently considered a junior synonym of Fejervarya), and also include species previously described as members of the genus Minervarya. Although parsimony and species tree analyses found support for the monophyly of Fejervarya as currently understood, partitioned Bayesian inference and unpartitioned Maximum Likelihood analyses of concatenated sequences recovered Southeast Asian species as a clade sister to the genus Sphaerotheca, albeit with low nodal support. We discuss the advantages and disadvantages of alternative classification schemes in light of previously proposed criteria for naming supraspecific taxa. The current single-genus taxonomy would impart desirable economy of nomenclatural change, and morphological diagnosability. However, other taxon naming criteria such as support for monophyly, temporal framework for diversification, and biogeographic regionalism would support a contrasting two-genus alternative. Because new species of Fejervarya are increasingly being discovered and described, and because a single-genus classification (Fejervarya) will remain controversial, given ambiguous support for its inferred monophyly, we propose recognizing two genera: Southeast Asian Fejervarya, and South Asian Minervarya. This classification results in two genera whose monophyly is strongly supported, respectively, and unlikely to be challenged by future analyses. Abstract

Abstract: The postembryonic development of amphibians has been characterized as divided into three predominant periods, hereafter named primary developmental stages: premetamorphosis (PreM), prometamorphosis (ProM), metamorphic climax (Meta), and completion of metamorphosis (PostM), largely based on examination of anuran development. Here, we categorized the postembryonic development of larvae of a poisonous fire salamander (Salamandra salamandra) by integrating morphology and gene expression (transcriptomic) data. Morphological analysis revealed three distinct clusters suggestive of PreM, ProM, and Meta, which were confirmed in parallel by microarray‐derived gene expression analysis. In total, 3,510 probes targeted transcripts differentially expressed between the clusters we identified. Genes upregulated in PreM related to organogenesis, and those upregulated in Meta underlie structural proteins and related to development of anatomical structures and pigmentation. Biosynthesis pathways of pigments (pteridines and melanin) were upregulated during late ProM and Meta. Gas chromatographic analysis of alkaloids indicated the onset of steroidal alkaloid biosynthesis at ProM. When comparing gene expression in the fire salamander to that in other amphibians—three anurans, Xenopus laevis, X. tropicalis, and Michrohyla fissipes, and one caudate, Ambystoma mexicanum— we identified genes with conserved expression patterns involved in basic metamorphic processes such as skin restructuring and tail fin resorption. Our results support that primary stages of postembryonic development in caudates are homologous to those of anurans, and offer a baseline for the study of the evolution of developmental modes. 29. Diversity and distribution of deep mitochondrial lineages of the common frog, Rana temporaria, in northern Spain.

Abstract

We provide an update of the geographical distribution and phylogenetic relationships of mitochondrial haplotype groups of the European common frog, Rana temporaria, one of the most widespread amphibians on Earth. Our data set of 378 newly determined DNA sequences of the cytochrome b gene is combined with 640 previously published sequences, mainly filling a sampling gap in the Spanish Basque country. A haplotype network based on 331 bp provides evidence for a previously unknown, deeply divergent haplogroup in this region, which in the eastern part of its range occurs sympatrically with a widespread group of haplotypes from the Pyrenean mountains, and to the West contacts a haplogroup typically for the central Cantabrian mountains. In contrast, samples from the isolated Montseny massif in the Spanish region of Catalonia are not differentiated from Pyrenean populations. Samples from Scotland cluster in a widespread haplogroup with samples from northern France, western Germany, and Ireland. Our data furthermore suggest that previous records of haplotype sharing of British R. temporaria populations with those from northwestern Spain are probably erroneous. A phylogenetic analysis based on 4,413 bp of mitochondrial DNA confirms the strong divergence of the newly discovered Basque haplogroup and indicates that it might be the sister group of a clade containing all other R. temporaria haplogroups. Abstract

Future progress in materials for tissue engineering and 3D cell cultures applications requires control of two key physical properties: nanoscale mechanical properties and mass transport. These requirements remain uncontrolled partly due to a lack of physical parameters and quantitative measurements. Using chitosan scaffolds as a model system in close-to-physiological conditions and a combination of experimental techniques and theory, we link structure with local nanomechanical properties. Additionally we introduce a parameter, the free volume, to predict variations in transport properties. By fabricating nanocomposites with single walled carbon nanotubes (SWNTs) we are able to test our approach: incorporation of acid-treated, soluble, ∼80 nm SWNTs in a chitosan matrix leads to a 2 fold increase in mean local elastic modulus and a decrease of 3% of the free volume available for oxygen diffusion. Inclusion of hydrophobic, ∼800 nm SWNTs leads to a 100 fold increase of elastic modulus and doubles the voids percentage available for the transport of glucose. [ABSTRACT FROM AUTHOR]

Abstract

Background: The impact of biologics on the risk of postoperative complications (PC) in inflammatory bowel disease (IBD) is still an ongoing debate. This lack of evidence is more relevant for ustekinumab and vedolizumab. Aims: To evaluate the impact of biologics on the risk of PC. Methods: A retrospective study was performed in 37 centres. Patients treated with biologics within 12 weeks before surgery were considered “exposed”. The impact of the exposure on the risk of 30-day PC and the risk of infections was assessed by logistic regression and propensity score-matched analysis. Results: A total of 1535 surgeries were performed on 1370 patients. Of them, 711 surgeries were conducted in the exposed cohort (584 anti-TNF, 58 vedolizumab and 69 ustekinumab). In the multivariate analysis, male gender (OR: 1.5, 95% CI: 1.2–2.0), urgent surgery (OR: 1.6, 95% CI: 1.2–2.2), laparotomy approach (OR: 1.5, 95% CI: 1.1–1.9) and severe anaemia (OR: 1.8, 95% CI: 1.3–2.6) had higher risk of PC, while academic hospitals had significantly lower risk. Exposure to biologics (either anti-TNF, vedolizumab or ustekinumab) did not increase the risk of PC (OR: 1.2, 95% CI: 0.97–1.58), although it could be a risk factor for postoperative infections (OR 1.5, 95% CI: 1.03–2.27). Conclusions: Preoperative administration of biologics does not seem to be a risk factor for overall PC, although it may be so for postoperative infections.

Abstract

The interaction between a viral capsid and its genome governs crucial steps in the life cycle of a virus, such as assembly and genome uncoating. Tuning cargo-capsid interactions is also essential for successful design and cargo delivery in engineered viral systems. Here we investigate the interplay between cargo and capsid for the picorna-like Triatoma virus using a combined native mass spectrometry and atomic force microscopy approach. We propose a topology and assembly model in which heterotrimeric pentons that consist of five copies of structural proteins VP1, VP2 and VP3 are the free principal units of assembly. The interpenton contacts are established primarily by VP2. The dual role of the genome is first to stabilize the densely packed virion and, second, on an increase in pH to trigger uncoating by relaxing the stabilizing interactions with the capsid. Uncoating occurs through a labile intermediate state of the virion that reversibly disassembles into pentons with the concomitant release of protein VP4.
