Rabies is a zoonotic infectious disease that causes at least 59 000 human deaths worldwide annually, with 95% of the cases occurring in the developing countries of Asia and Africa. There are two Lyssavirus rabies (RABV) variants circulating in South Africa, notably the canid and mongoose RABV biotypes. The canid RABV biotype is maintained in the domestic dog and two wild carnivore species, the black-backed jackal (Canis mesomelas) and the bat-eared fox (Otocyon megalotis). The yellow mongoose, a member of the Herpestidae family, is a reservoir and vector species for the mongoose RABV biotype. Rabies trends showed an increase in rabiespositive cases in aardwolves between 2011 and 2016 surpassing the bat-eared fox as the most rabies-affected wild carnivore in the Northern Cape Province of South Africa. The aim of the study was to establish the genetic relationships amongst rabies viruses recovered from both the aardwolves and bat-eared foxes. A partial region of the glycoprotein gene and the variable G-L intergenic region of the viral genome were analysed using nucleotide sequences generated from PCR amplicons. The rabies viruses recovered from the aardwolves between the year 2015 and 2017 were 100% nucleotide sequence identical, suggesting a single or common source and possible evidence for a host shift. Furthermore, the phylogenetic reconstruction demonstrated that the rabies viruses obtained from the two wild carnivore species from the Northern Cape Province clustered independently of each other with 96% nucleotide sequence identity, suggesting that the aardwolf may be able to maintain the canid RABV variant in this geographical area. This article submits that the 1963 Charter of the Organisation of African Unity provided a legal foundation for continental economic integration. This was followed by successive declarations and legal agreements such as the Lagos Plan of Action, the Treaty Establishing the African Economic Community and the Constitutive Act of the African Union, all of which became more explicit. While the Organisation of African Unity of 1963 and the African Union of 2002 respectively pronounced the direction of the continental organisations, the Treaty Establishing the African Economic Community was then established to translate the vision into actionable activities towards a continental economic community through the establishment of the Regional Economic Communities that would ultimately be the foundation blocks for the integration of the continent. The article shows that all the Regional Economic Communities have a strong legal appeal to achieve African unity and the desire to integrate African economies. Consequently, the African Union has to date attained a continental Free Trade Area and is still to attain a continental customs union and the common market using the same legal architecture, these being stages in developing an African Economic Community. The article also demonstrates that the various Regional Economic Communities owe their existence to the Charter of the Organisation of African Unity or the Lagos Plan of Action or the Abuja Treaty, all under the umbrella of the Constitutive Act of the African Union. Rabies is a neglected zoonotic disease that has no effective treatment after onset of illness. However the disease can be prevented effectively by prompt administration of post exposure prophylaxis which includes administration of passive immunizing antibodies (Rabies Immune Globulin, RIG). Currently, human RIG suffers from many restrictions including limited availability, batch-to batch inconsistencies and potential for contamination with blood-borne pathogens. Anti-rabies monoclonal antibodies (mAbs) have been identified as a promising alternative to RIG. Here, we applied a plant-based transient expression system to achieve rapid, high level production and efficacy of the two highly potent anti-rabies mAbs E559 and 62-71-3. Expression levels of up to 490 mg/kg of recombinant mAbs were obtained in Nicotiana benthamiana glycosylation mutants by using a viral based transient expression system. The plant-made E559 and 62-71-3, carrying human-type fucose-free N-glycans, assembled properly and were structurally sound as determined by mass spectrometry and calorimetric density measurements. Both mAbs efficiently neutralised diverse rabies virus variants in vitro. Importantly, E559 and 62-71-3 exhibited enhanced protection against rabies virus compared to human RIG in a hamster model post-exposure challenge trial. Collectively, our results provide the basis for the development of a multi-mAb based alternative to RIG.

Rabies post-exposure prophylaxis (PEP) currently comprises administration of rabies vaccine together with rabies immunoglobulin (RIG) of either equine or human origin. In the developing world, RIG preparations are expensive, often in short supply, and of variable efficacy. Therefore, we are seeking to develop a monoclonal antibody cocktail to replace RIG. Here, we describe the cloning, engineering and production in plants of a candidate monoclonal antibody (E559) for inclusion in such a cocktail. The murine constant domains of E559 were replaced with human IgG1κ constant domains and the resulting chimeric mouse-human genes were cloned into plant expression vectors for stable nuclear transformation of Nicotiana tabacum. The plant-expressed, chimeric antibody was purified and biochemically characterized, was demonstrated to neutralize rabies virus in a fluorescent antibody virus neutralization assay, and conferred protection in a hamster challenge model., (© The Author 2014. Canine rabies is enzootic throughout Sub-Saharan Africa, including the Republic of South Africa. Historically, in South Africa the coastal provinces of KwaZulu-Natal and Eastern Cape were most affected. Alarmingly, outbreaks of canine rabies have been increasingly reported in the past decade from sites where it has previously been under control. From January 2010 to December 2011, 53 animal rabies cases were confirmed; these were mostly in domestic dogs from southern Johannesburg, which was previously considered to be rabies free. In addition, one case was confirmed in a 26-month old girl who had been scratched by a pet puppy during this period. The introduction of rabies into Gauteng Province was investigated through genetic analysis of rabies positive samples confirmed during the outbreak period. In addition, the nucleotide sequences of incidental cases reported in the province for the past ten years were also included in the analysis. It was found that the recent canine rabies outbreak in the Gauteng Province came from the introduction of the rabies virus from KwaZulu-Natal, with subsequent local spread in the susceptible domestic dog population of southern Johannesburg. The vulnerability of the province was also highlighted through multiple, dead-end introductions in the past ten years. This is the first report of a rabies outbreak in the greater Johannesburg area with evidence of local transmission in the domestic dog population.

Isolations of Mokola virus (MOKV) are rare, but in South Africa and Zimbabwe this genotype 3 lyssavirus variant has been occasionally found in domestic mammals (cats and a dog) with a total of 17 virus isolates (South Africa 10, Zimbabwe 7) having been recovered during the past 30 years. We report the identification of a MOKV isolate involved in a human contact in Grahamstown (Eastern Cape, South Africa) and a genetic comparison with previously characterized isolates. This reported MOKV case was in a previously immunized cat. While the continual recovery of MOKV isolates in domestic cats is speculative of the existence of a reservoir host species among bats or rodents, the lack of protection with currently used vaccines is discussed and the need for biologicals with a wider spectrum of protection against this lyssavirus variant is highlighted.

We recently identified 2 Mokola viruses from domestic mammals (a dog and a cat) in South Africa. These cases occurred 8 years after the last reported case of infection with this virus. Our findings emphasize the endemicity of rabies-related lyssaviruses in South Africa and the need to better understand the epidemiology of Mokola viruses.

Heartwater, the tick-borne disease caused by the rickettsia Cowdria ruminantium has historically been confined to the southern and western lowvelds of Zimbabwe. Since 1986, however, cases of heartwater have been diagnosed with increasing frequency in the central and eastern regions of the previously heartwater-free highveld plateau. During the same period, collections of the two major tick vectors of heartwater in Zimbabwe, Amblyomma hebraeum and Amblyomma variegatum, were made for the first time in these areas, suggesting that spread of these ticks was responsible for the changed distribution of the disease. The factors associated with this spread have not been determined, but increased cattle and wildlife movement and reduced intensity of dipping undoubtedly play important roles. Currently, the distribution of heartwater and its vectors in the highveld is still largely restricted to the central and eastern regions. The northern regions of the highveld appear to be predominantly uninfected, though it is likely that, eventually, heartwater will spread further with considerable impact on livestock production in Zimbabwe.
