Abstract With regard to deep mining in metal mines, an investigation into the failure mode of deep fractured rock masses and their corresponding acoustic emission signal characteristics is conducted via uniaxial compression tests. Subsequently, a fractal damage renormalization group mechanical model is developed to explain the behavior of those fractured rock masses. Employing the bonded block model (BBM) numerical simulation method, fracture process in synthetic rock samples is analyzed, thereby validating the efficacy of the mechanical model. The numerical simulations highlight the critical role of fractures expansion in underlying the deterioration of rock mass strength. As the peak load decreases, the fracture fractal dimension increases, leading to a significant 14.2% reduction in compressive strength accompanied by an approximate 8.7% rise in average fracture fractal dimension. A comparative analysis of tetrahedral and voronoi block synthetic rock samples reveals the tetrahedral block samples exhibit a superior ability to depict the fracture behavior of fractured rock masses. Specifically, they offer a more accurate simulation of acoustic emission characteristics and failure modes. Furthermore, variations in the fracture fractal dimension with respect to the hole defect’s position are observed, with the maximum value occurring along the vertical axis of the hole defect. This observation underscores the potential utility of visually monitoring deep rock fracture dynamics as an effective mean for quantitatively evaluating fracture damage and strength degradation in deep rock formations.

ABSTRACT: In this study, milk fat globule membrane (MFGM) ingredients enriched in polar lipids were prepared using membrane filtration, including microfiltration, diafiltration, and ultrafiltration from butter serum powder. Polar lipids (phospholipids, sterols, and gangliosides) in prepared MFGM ingredients were analyzed by 31P nuclear magnetic resonance spectroscopy, GC-MS, and ultra-performance liquid chromatography (UPLC)-MS/MS, respectively. The lipolysis degree and microstructure of MFGM ingredient and soybean lecithin (SL) emulsions during in vitro digestion were also analyzed. Microfiltration showed higher concentration efficiency than ultrafiltration, which increased by 2.16% and 2.73% in phospholipids, respectively. Moreover, diafiltration concentrated more polar lipids (6.39% of phospholipids) than microfiltration. Milk fat globule membrane ingredients had high levels of sphingomyelin (1.27%–1.36%) and ratio of GD3 to GM3 is 9.25- to 9.88-fold. The different lipolysis behaviors between MFGM ingredient emulsions and SL emulsions were correlated with their different polar lipid compositions. Phospholipids from both MFGM ingredients and SL could help maintain the initial structure during the gastric digestion. These results could provide a scientific basis for developing high-polar-lipids food, particularly infant formulas and special functional foods.

Abstract Background Postoperative pneumonia is one of the common complications after video-assisted thoracoscopic surgery. There is no related study on the effect of lung isolation with different airway devices on postoperative pneumonia. Therefore, in this study, the propensity score matching method was used to retrospectively explore the effects of different lung isolation methods on postoperative pneumonia in patients undergoing video-assisted thoracoscopic surgery. Methods This is A single-center, retrospective, propensity score-matched study. The information of patients who underwent VATS in Weifang People 's Hospital from January 2020 to January 2021 was retrospectively included. The patients were divided into three groups according to the airway device used in thoracoscopic surgery: laryngeal mask combined with bronchial blocker group (LM + BB group), tracheal tube combined with bronchial blocker group (TT + BB group) and double-lumen endobronchial tube group (DLT group). The main outcome was the incidence of pneumonia within 7 days after surgery; the secondary outcome were hospitalization time and hospitalization expenses. Patients in the three groups were matched using propensity score matching (PSM) analysis. Results After propensity score matching analysis, there was no significant difference in the incidence of postoperative pneumonia and hospitalization time among the three groups (P > 0.05), but there was significant difference in hospitalization expenses among the three groups (P

The maturation of brain microvascular endothelial cells leads to the formation of a tightly sealed monolayer, known as the blood–brain barrier (BBB). The BBB damage is associated with the pathogenesis of age-related neurodegenerative diseases including vascular cognitive impairment and Alzheimer’s disease. Growing knowledge in the field of epigenetics can enhance the understanding of molecular profile of the BBB and has great potential for the development of novel therapeutic strategies or targets to repair a disrupted BBB. Histone deacetylases (HDACs) inhibitors are epigenetic regulators that can induce acetylation of histones and induce open chromatin conformation, promoting gene expression by enhancing the binding of DNA with transcription factors. We investigated how HDAC inhibition influences the barrier integrity using immortalized human endothelial cells (HCMEC/D3) and the human induced pluripotent stem cell (iPSC)-derived brain vascular endothelial cells. The endothelial cells were treated with or without a novel compound named W2A-16. W2A-16 not only activates Wnt/β-catenin signaling but also functions as a class I HDAC inhibitor. We demonstrated that the administration with W2A-16 sustained barrier properties of the monolayer of endothelial cells, as evidenced by increased trans-endothelial electrical resistance (TEER). The BBB-related genes and protein expression were also increased compared with non-treated controls. Analysis of transcript profiles through RNA-sequencing in hCMEC/D3 cells indicated that W2A-16 potentially enhances BBB integrity by influencing genes associated with the regulation of the extracellular microenvironment. These findings collectively propose that the HDAC inhibition by W2A-16 plays a facilitating role in the formation of the BBB. Pharmacological approaches to inhibit HDAC may be a potential therapeutic strategy to boost and/or restore BBB integrity.

Abstract Background Abnormal blinking pattern is associated with ocular surface diseases. However, blink is difficult to analyze due to the rapid movement of eyelids. Deep learning machine (DLM) has been proposed as an optional tool for blinking analysis, but its clinical practicability still needs to be proven. Therefore, the study aims to compare the DLM-assisted Keratograph 5M (K5M) as a novel method with the currently available Lipiview in the clinic and assess whether blinking parameters can be applied in the diagnosis of dry eye disease (DED). Methods Thirty-five DED participants and 35 normal subjects were recruited in this cross-sectional study. DED questionnaire and ocular surface signs were evaluated. Blinking parameters including number of blinks, number of incomplete blinking (IB), and IB rate were collected from the blinking videos recorded by the K5M and Lipiview. Blinking parameters were individually collected from the DLM analyzed K5M videos and Lipiview generated results. The agreement and consistency of blinking parameters were compared between the two devices. The association of blinking parameters to DED symptoms and signs were evaluated via heatmap. Results In total, 140 eyes of 70 participants were included in this study. Lipiview presented a higher number of IB and IB rate than those from DLM-assisted K5M (P ≤ 0.006). DLM-assisted K5M captured significant differences in number of blinks, number of IB and IB rate between DED and normal subjects (P ≤ 0.035). In all three parameters, DLM-assisted K5M also showed a better consistency in repeated measurements than Lipiview with higher intraclass correlation coefficients (number of blinks: 0.841 versus 0.665; number of IB: 0.750 versus 0.564; IB rate: 0.633 versus 0.589). More correlations between blinking parameters and DED symptoms and signs were found by DLM-assisted K5M. Moreover, the receiver operating characteristic analysis showed the number of IB from K5M exhibiting the highest area under curve of 0.773. Conclusions DLM-assisted K5M is a useful tool to analyze blinking videos and detect abnormal blinking patterns, especially in distinguishing DED patients from normal subjects. Large sample investigations are therefore warranted to assess its clinical utility before implementation.

Abstract Norovirus (NoV) and hepatitis A virus (HAV) pose a considerable health risk worldwide. In recent years, many cases of virus infection caused by virus‐contaminated strawberries have occurred worldwide. This study applied a critical control point system to analyze the main hazards during the production and marketing of strawberries imported into China and explore the key control points in the whole process. To further evaluate the risks in the supply chain, the established quantitative microbial risk assessment (QMRA) was used to determine the probability that residents would be infected with viruses after consuming imported strawberries. It was found that the risk of virus contamination from imported strawberries was low, and the virus contamination mainly results from water resources and personnel. This research helps the regulatory authorities formulate strategies to ensure the long‐term microbial safety of imported strawberries. In addition, the methods may prove useful in evaluating the risks of other agricultural produce.

Twig blight is a highly destructive disease primarily caused by Pestalotiopsis versicolor. It affects bayberry (Myrica rubra) cultivation and production. However, the molecular and regulatory defence responses to twig blight remain unclear. We inoculated a susceptible cultivar bayberry variety with P. versicolor XJ27 and carefully profiled the disease development. The bayberry disease index increased sharply following inoculation. We conducted integrative transcriptomic and metabolomic analyses to track key genes and secondary metabolomic changes in susceptible bayberry leaves. Herein, we identified 9588 differentially expressed genes (DEGs) and 748 differentially accumulated metabolites (DAMs). Of these, seven structural genes and seven metabolites (including L-phenylalanine, coniferin, p-coumaric acid, cinnamaldehyde, eleutheroside B, ferulaldehyde, and isoeugenol) related to lignin biosynthesis markedly increased following pathogen infection. These observations suggest that lignin biosynthesis may facilitate bayberry defence responses to twig blight. Quantitative real-time PCR analysis further confirmed the higher expression levels of pivotal lignin synthesis genes in infected leaves. In addition, higher foliar lignin levels and increased xylem formation in infected leaves provided evidence that lignin synthesis was enhanced during bayberry response to twig blight. These findings not only provide effective management strategies for bayberry twig blight but also promote the breeding of high-quality resistant varieties.

In this study, Senescence Accelerated Mice (SAMP8) were supplemented with exogenous DHA milk, endogenous DHA milk, normal milk, or 0.9 % saline solution. Enzyme-linked immunosorbent assay (ELISA), gas chromatography (GC), ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI MS/MS), and Morris water maze were used to characterize the effects of diet on oxidative stress and cognition in SAMP8 mice. Supplementation endogenous DHA milk or exogenous DHA milk can enhance the antioxidant capacity of mice organs. Endogenous DHA milk increased the superoxide dismutase (SOD) activity of mice brain and serum than normal milk and 0.9 % saline solution (P ≤ 0.05), as well as increased SOD activity of mice liver and glutathione peroxidase (GSH-Px) activity of mice brain than normal milk (P ≤ 0.05). Exogenous DHA milk increased SOD activity of mice brain than normal milk and 0.9 % saline solution, as well as increased SOD activity of mice serum than 0.9 % saline solution (P ≤ 0.05). Several polar lipid relative content, such as 18:0/18:2 PS, 17:0 Ceramide, and 20:4 LPC in mice brain was affected by dietary supplementation with DHA-containing milk. Lipid oxidation metabolites in mice brain were not affected by DHA-containing milk. Endogenous DHA milk increased the number of platform location crossing times of mice in the Morris water maze test, compared with Exogenous DHA milk, normal milk, and 0.9 % saline solution (P ≤ 0.05).

Zooplankton grazers, like copepods, can feed on toxic microalgae and live normally. We hypothesize that gut microbial communities (GMCs) may contribute to the detoxification of the host by changing their compositions and recruiting more beneficial bacteria. Here, we measured the physiological responses of two copepod species (Acartia sp. and Paracalanus sp.) fed with toxic (Alexandrium tamarense) and non-toxic (Alexandrium andersonii) dinoflagellates, respectively. Both copepods maintained consistently high survival rates but slightly reduced ingestion rates when feeding upon the toxic dinoflagellate (when compared to the non-toxic one), suggesting a compensatory mechanism. The compositional variation of copepod GMCs, at the amplicon sequence variant (ASV) level, was mostly significantly different among copepod host species (R = 0.83, by ANOSIM test), while diet type played minor but significant roles. Under the toxic diet, Acartia sp. enriched only five ASVs while Paracalanus sp. recruited a wide range of taxa (38 ASVs) mostly belonging to Alphaproteobacteria (e.g., Rhodobacteraceae) and Gammaproteobacteria (e.g., Alteromonadaceae). In contrast, when clustering GMCs by predicted functions, diet type was the key regulating factor, suggesting the functional convergence of copepod GMCs in response to algal toxins. This can be explained by the fact that most of the enriched bacteria under the toxic diet have similar functions on detoxification and maintaining the host homeostasis. This study deepens our understanding of the roles of GMC in the detoxification and adaptation mechanisms of copepods during harmful algal blooms.

It is crucial to assess the nodal modulation for tides in high-latitude coast areas within the context of global warming. In this paper, five stations (Maloy, Rorvik, Andenes, Vardo, and Honningsvag) along the Norwegian coast are selected to analyze the nodal modulation using the S_TIDE toolbox, which is developed from the enhanced harmonic analysis method. Three criterions are proposed to determine the optimal number of independent points (IPs), a parameter in S_TIDE toolbox, and the decision steps are elaborated in detail. The optimal number of IPs is evaluated by comparing the primary and the hindcasts tidal amplitudes. The amplitudes of 18.61-year cycle and 4.42-year cycle show noticeable temporal and spatial variations, which can be attributed to the changes of sea levels, local topography, and the active and robust mesoscale activity in the Norwegian Sea. Moreover, the temporal and spatial variations in nodal modulation are quantitatively demonstrated at the Rorvik and Vardo stations, highlighting the importance of nodal modulation in assessing tides over interdecadal periods.

Abstract Mounting evidence highlights the crucial role of aging in the pathogenesis of Alzheimer's disease (AD). We have previously explored human apoE-targeted replacement mice across different ages and identified distinct molecular pathways driven by aging. However, the specific contribution of different brain cell types to the gene modules underlying these pathways remained elusive. To bridge this knowledge gap, we employed a computational deconvolution approach to examine cell-type-specific gene expression profiles in major brain cell types, including astrocytes (AS), microglia (MG), oligodendroglia (OG), neurons (NEU), and vascular cells (VC). Our findings revealed that immune module genes were predominantly expressed in MG, OG, and VC. The lipid metabolism module genes were primarily expressed in AS, MG, and OG. The mitochondria module genes showed prominent expression in VC, and the synapse module genes were primarily expressed in NEU and VC. Furthermore, we identified intra- and inter-cell-type interactions among these module genes and validated their aging-associated expression changes using published single cell studies. Our study dissected bulk brain transcriptomics data at the cellular level, providing a closer examination of the cell-type contributions to the molecular pathways driven by aging.

ABSTRACT: In this study, we aimed to detect the physicochemical properties of distilled products (residue and distillate) obtained from anhydrous milk fat (AMF) and its dry fractionation products (liquid and solid fractions at 25°C [25 L and 25 S]). The results showed that the saturated fatty acids and low- and medium molecular–weight triglycerides were easily accumulated in the distillate, and the percentage of unsaturated fatty acid and high molecular–weight triglycerides in the residue were higher, and these components in 25 S and 25 L were influenced more significantly than those in the AMF. In addition, the distillate had larger melting ranges in comparison with the distilled substrate, while the melting ranges of residue was smaller. The triglycerides were presented as the mixture crystal forms (α, βʹ, and β crystal) in 25 S, AMF, and their distilling products, and it was transformed gradually to a single form as the increasing of distilling temperature. Moreover, the accumulated pattern of triglycerides was double chain length in 25 S, AMF, and their distilling products. These results provide a new approach to obtain the milk fat fractions with different properties, and the findings of this study enrich the theoretical basis of milk fat separation in practical production.

The study of submesoscale ageostrophic motion is crucial for enhancing our comprehension of ocean dynamics. This paper employs global sea surface velocity reanalysis data and mixed layer depth data to examine the factors influencing submesoscale ageostrophic energy in the Kuroshio region as well as the energy transition between ageostrophic and geostrophic energy. The findings indicate that submesoscale ageostrophic kinetic energy in the Kuroshio region peaks during winter and spring. Mixed layer depth and geostrophic strain significantly boost ageostrophic kinetic energy, especially in strong current area. Analysis of kinetic energy spectral density reveals how energy distribution and transition scale vary across strong and slow current zones during different seasons, highlighting that submesoscale kinetic energy is susceptible to seasonal variations. In summer and autumn, the transition scale of kinetic energy is generally larger compared to those in spring and winter. Submesoscale ageostrophic motion predominantly gains kinetic energy from the release of available potential energy (APE) and horizontal shear production (HSP) while losing a small portion of its kinetic energy through vertical shear production (VSP) in the Kuroshio.

Background: The metabolic-status-related mechanisms underlying the deterioration of the lung function in obese asthma have not been completely elucidated. Objective: This study aimed to investigate the basal metabolic rate (BMR) in patients with obese asthma, its association with the lung function, and its mediating role in the impact of obesity on the lung function. Methods: A 12-month prospective cohort study (n = 598) was conducted in a real-world setting, comparing clinical, body composition, BMR, and lung function data between patients with obese (n = 282) and non-obese (n = 316) asthma. Path model mediation analyses for the BMR and skeletal muscle mass (SMM) were conducted. We also explored the effects of the BMR on the long-term lung function in patients with asthma. Results: Patients with obese asthma exhibited greater airway obstruction, with lower FEV1 (1.99 vs. 2.29 L), FVC (3.02 vs. 3.33 L), and FEV1/FVC (65.5 vs. 68.2%) values compared to patients with non-obese asthma. The patients with obese asthma also had higher BMRs (1284.27 vs. 1210.08 kcal/d) and SMM (23.53 vs. 22.10 kg). Both the BMR and SMM mediated the relationship between obesity and the lung function spirometers (FEV1, %FEV1, FVC, %FVC, and FEV1/FVC). A higher BMR or SMM was associated with better long-term lung function. Conclusions: Our study highlights the significance of the BMR and SMM in mediating the relationship between obesity and spirometry in patients with asthma, and in determining the long-term lung function. Interventions for obese asthma should focus not only on reducing adiposity but also on maintaining a high BMR.

This paper addresses the critical need to determine the stable operating limit of modular multilevel converter-based high voltage direct current (MMC-HVDC) systems, particularly concerning the integration of extensive renewable energy sources. To achieve this, the steady-state mathematical model and state-space model of bundled hydropower and photovoltaic integration through MMC-HVDC systems are established. A novel methodology considering steady-state and small-signal stability constraints is proposed to compute the stable operating region of the system. The quantitative assessment reveals that diminishing AC system short-circuit capacities amplify restrictions from small-signal stability constraints, thereby reducing the system's stable operating region. Eigenvalue and participation factor analyses shed light on the pivotal factors affecting small-signal stability in weak AC systems. To expand the system's stable operating region, a supplementary frequency damping control strategy is proposed. The theoretical analysis and calculation results are validated by building a simulation model for the bundled hydropower and photovoltaic integration through MMC-HVDC systems in PSCAD/EMTDC.

AbstractOver the past decades, intensive reclamation has caused significant land degradation, threatening both ecological and food security. The Chinese government has taken a series of novel Conservation Tillage (CT) measures according to regional climate and soil conditions to increase agriculture’s multifunctional and resilience. Previous studies have reported CT in northeast China can improve soil organic carbon and reduce ammonia emissions in northeast China. However, whether the regionally adapted CT can avoid its inherent limitations in seed emergence and crop growth is still lacking comprehensive understanding and investigation. By using high-resolution and time-series Unmanned Aerial Vehicle (UAV) observations, our study investigated and evaluated the performance of different advanced and regional adapted CT treatments in crop emergence and growth compared with conventional tillage during the whole growing season. Based on the field measurements, we found that although the residues can impact the seed emergence, there was no significant difference between CT treatments and conventional tillage, with some CT treatments can even raise the emergence rate. We also found that the high residues covered CT treatments obstructed the maize grown in the early growing season including the V3 and V6 period, while the crop growth in these treatments caught up with the conventional tillage in the following growing periods. Our study suggested that well-adjusted CT treatments in Northeast China have the potential to maintain crop production accompanied by enhancing the ecosystem’s multifunctionality. The results from this study provided observational evidence of crop growth impact from CT, which will have great implications in convincing the farmers and beneficial to the promotion of conservation tillage.

Abstract Background Lignin is a major restriction factor for the industrial production of biomass resources, such as pulp and bioenergy. Eucalyptus is one of the most important sources of pulp and bioenergy. After polyploidization, the lignin content of forest trees is generally reduced, which is considered a beneficial genetic improvement. However, the differences in the lignin content between triploid and diploid Eucalyptus and the underlying regulatory mechanism are still unclear. Results We conducted a comprehensive analysis at the phenotypic, transcriptional and metabolite levels between Eucalyptus urophylla triploids and diploids to reveal the effects of polyploidization on the lignin content and lignin metabolic pathway. The results showed that the lignin content of Eucalyptus urophylla triploid stems was significantly lower than that of diploids. Lignin-related metabolites were differentially accumulated between triploids and diploids, among which coniferaldehyde, p-coumaryl alcohol, sinapaldehyde and coniferyl alcohol had significant positive correlations with lignin content, indicating that they might be primarily contributing metabolites. Most lignin biosynthetic genes were significantly downregulated, among which 11 genes were significantly positively correlated with the lignin content and above metabolites. Furthermore, we constructed a co-expression network between lignin biosynthetic genes and transcription factors based on weighted gene co-expression network analysis. The network identified some putative orthologues of secondary cell wall (SCW)-related transcription factors, among which MYB52, MYB42, NAC076, and LBD15 were significantly downregulated in Eucalyptus urophylla triploids. In addition, potential important transcription factors, including HSL1, BEE3, HHO 3, and NAC046, also had high degrees of connectivity and high edge weights with lignin biosynthetic genes, indicating that they might also be involved in the variation of lignin accumulation between triploid and diploid Eucalyptus urophylla. Conclusions The results demonstrated that some lignin-related metabolites, lignin biosynthetic genes and transcription factors in Eucalyptus urophylla triploids may be relatively sensitive in response to the polyploidization effect, significantly changing their expression levels, which ultimately correlated with the varied lignin content. The analysis of the underlying formation mechanism could provide beneficial information for the development and utilization of polyploid biomass resources, which will be also valuable for genetic improvement in other bioenergy plants.

Endometritis (inflammation of the endometrial lining) is one of the most devastating reproductive diseases in dairy cattle, resulting in substantial production loss and causing more than $650 million in lost revenue annually in the USA. We hypothesize that alternative polyadenylation (APA) sites serve as decisive sensors for endometrium health and disease in dairy cows. Endometrial cells collected from 18 cows with purulent vaginal discharge scored 0 to 2 were used for APA profiling with our whole transcriptome termini site sequencing (WTTS-seq) method. Overall, pathogens trigger hosts to use more differentially expressed APA (DE-APA), more intronic DE-APA, more DE-APA sites per gene and more DE-genes associated with inflammation. Host CD59 molecule (CD59), Fc fragment of IgG receptor IIa (FCGR2A), lymphocyte antigen 75 (LY75) and plasminogen (PLG) may serve as initial contacts or combats with pathogens on cell surface, followed by activation of nuclear receptor subfamily 1 group H member 4 (NR1H4) to regulate AXL receptor tyrosine kinase (AXL), FGR proto-oncogene, Src family tyrosine kinase (FGR), HCK proto-oncogene, Src family tyrosine kinase (HCK) and integrin subunit beta 2 (ITGB2) for anti-inflammation. This study is the first to show significance of cilium pathways in endometrium health and animal reproduction. MIR21 and MIR30A would be perfect antagonistic biomarkers for diagnosis of either inflammation or anti-inflammation. These novel findings will set precedent for future genomic studies to aid the dairy industry develop new strategies to reduce endometritis incidence and improve fertility.

Lacticaseibacillus rhamnosus is applied as a probiotic to alleviate various metabolic, gastrointestinal, and psychological symptoms and diseases, and its probiotic effectiveness is strain-specific. In this study, we obtained 21 strains of Ls. rhamnosus, and their genomes were sequenced. We defined the pan- and core-genomes of Ls. rhamnosus. Phenotypes such as the assimilation of carbohydrates and antibiotic resistance were experimentally characterized and associated with genome annotations. Nine strains were selected and tested for growth rates, tolerance to acidity/alkalinity and bile acids, the production of short-chain fatty acids, and competition with pathogenic microbes. Strains WL11 and WL17 were targeted as potential probiotics and were applied in mouse model tests for the alleviation of chronic fatigue syndrome (CFS) and irritable bowel syndrome (IBS). The results showed that WL11 and WL17 effectively alleviated slow body weight gain, anxiety, poor memory, and cognitive impairment in CFS mouse models. They also reduced the expression of pro-inflammatory factors, such as TNF-α and IL-6, and alleviated intestinal peristalsis, visceral hypersensitivity, and anxiety-like behavior in IBS mouse models. This study reports new Ls. rhamnosus strain resources and their effect on alleviation of both IBS and CFS symptoms with mouse models; the probiotic functions of those strains in human patients remain to be further tested.

We aimed to obtain the optimal formula for human milk fat substitute (HMFS) through a combination of software and an evaluation model and further verify its practicability through an animal experiment. The results showed that a total of 33 fatty acid (FA) and 63 triglyceride (TAG) molecular species were detected in vegetable oils. Palmitic acid, oleic acid, linoleic acid, 18:1/16:0/18:1, 18:2/16:0/18:2, 18:1/18:1/18:1 and 18:1/18:2/18:1, were the main molecular species among the FAs and TAGs in the vegetable oils. Based on the HMFS evaluation model, the optimal mixed vegetable oil formula was blended with 21.3% palm oil, 2.8% linseed oil, 2.6% soybean oil, 29.9% rapeseed oil and 43.4% maize oil, with the highest score of 83.146. Moreover, there was no difference in the weight, blood routine indices or calcium and magnesium concentrations in the feces of the mice between the homemade mixed vegetable oil (HMVO) group and the commercial mixed vegetable oil (CMVO) group, while nervonic acid (C24:1) and octanoic acid (C8:0) were absorbed easily in the HMVO group. Therefore, these results demonstrate that the mixing of the different vegetable oils was feasible via a combination of computer software and an evaluation model and provided a new way to produce HMFS.

The study aims to analyze the composition of the gut microbiota in Chinese individuals using metagenomic sequencing technology, with a particular focus on the abundance of Akkermansia muciniphila (Akk). To improve the efficiency of Akk isolation and identification accuracy, modifications were made to the enrichment culture medium and 16S rRNA universal primers. Additionally, potential growth-promoting factors that stimulate Akk growth were explored through in vitro screening. The research results revealed that the abundance of Akk in Chinese fecal samples ranged from 0.004% to 0.4%. During optimization, a type of animal protein peptide significantly enhanced the enrichment efficiency of Akk, resulting in the isolation of three Akk strains from 14 fecal samples. Furthermore, 17 different growth-promoting factors were compared, and four factors, including galactose, sialic acid, lactose, and chitosan, were identified as significantly promoting Akk growth. Through orthogonal experiments, the optimal ratio of these four growth-promoting factors was determined to be 1:1:2:1. After adding 1.25% of this growth-promoting factor combination to the standard culture medium, Akk was cultivated at 37° for 36 h, achieving an OD600nm value of 1.169, thus realizing efficient proliferation and optimized cultivation of Akk. This study provides important clues for a deeper understanding of the gut microbiota composition in Chinese individuals, while also offering effective methods for the isolation and cultivation of Akk, laying the groundwork for its functional and application research in the human body.

Leaves are crucial photosynthetic plant organs. The development of poplar leaves has spatio-temporal specificity and it is of great significance to study the single-cell transcription atlas of leaves to reveal the temporal regulation of gene expression in different cell types. Here, single-cell RNA sequencing was performed on 17,768 tender leaf and 5846 functional leaf cells of Poplar 84K to construct a transcriptome atlas and developmental trajectory. The results showed that there were five and six cell types in tender and functional leaves, respectively. According to a pseudo-time trajectory analysis and the clustering of expressed genes into different cell types, the development of tender and functional leaves was divided into two temporal stages. Tender leaf epidermal cells developed earliest and were enriched with genes related to cell division and growth, indicating that tender leaves were in the stage of cell expansion and functional differentiation. Functional leaf palisade mesophyll cells were enriched with genes related to photosynthesis and carbon metabolism and cell types performing different functions tended to mature, indicating that functional leaves were in the stage of leaf development and the initial formation of photosynthesis. Our in-depth analysis of the transcriptional regulation at the single-cell level during leaf development provides an important basis for studying the mechanisms involved in cell differentiation and leaf development in poplar as well as other plants.

Abstract The alcohol extracts of Chinese bayberry (Myrica rubra) branches (MRBE) are rich in flavonoids which have a variety of medicinal benefits, but their effects on human HepG2 were unknown. In this study, the effects of MRBE on HepG2 cell growth and its potential for inhibiting cancer were explored. The results displayed that MRBE inhibited HepG2 proliferation both by arresting cells in S phase and promoting apoptosis. Quantitative reverse‐transcription PCR (qRT‐PCR), western blotting, and immunofluorescence showed that MRBE induced S‐phase arrest by upregulating p21, which in turn downregulated cyclin and cyclin‐dependent kinase messenger RNA (mRNA) and protein. Apoptosis was induced by blocking the expression of BCL‐2 and suppression of the Raf/ERK1 signaling pathways. These results indicated that MRBE may have the potential for treatment of human liver cancer, highlighting novel approaches for developing new pharmacological tools for the treatment of this deadly type cancer. Meanwhile, it provides a new direction for the medicinal added values of Chinese bayberry, which helped to broaden the diversified development of its industry chain.

The asphalt-concrete pavement is an essential component of the bridge deck. Its stress characteristics are different from the general pavement structure. This research employs finite element technology (ANSYS) to model a three-span bridge and pavement layer in order to ascertain the stress distribution and provide a deeper knowledge of the mechanical properties of the pavement layer on concrete bridge decks. The effects of varying the thickness and stiffness of the deck pavement on the stress distribution in its different layers were investigated. Stress absorption is increased as the stiffness of the deck layers increases, and with increases in pavement thickness, stress on pavement layers decreases. The response of deck pavement under moving load, both under full bonding and sliding conditions, was simulated to determine stress distribution. By comparing the response under the full bonding and sliding conditions, it was found that stress induced on layers during sliding is high as compared to the full bonding condition; pavement layers will be prone to failure if sliding occurs between layers.

Abstract Background To evaluate the accuracy of different preoperative calculation methods of osteotomy size in ankylosing spondylitis with thoracolumbar or lumbar kyphosis and analyze its clinical significance. Methods Twenty-two cases of AS patients with thoracolumbar or lumbar kyphosis, from January 2015 to December 2018, who underwent one-level SPO surgery in our hospital, were retrospectively reviewed. The sagittal parameters were measured at pre-operation and last follow up using Surgimap software, and theoretical values of sagittal parameters were calculated according to pre-operative PI. The osteotomy angles of different methods were measured using Surgimap software. Paired t test was used to for the statistical analysis. Results The mean follow-up time of all patients was 30.00 $$\pm$$ ± 3.56 months. The osteotomy sites were located at T12 in 3 cases, L1 in 6 cases, L2 in 9 cases, and L3 in 4 cases. Compared to pre-operative sagittal parameters, post-operative PT, SS, LL, and SVA were significantly improved (P $$$$ > 0.05). Conclusion For AS patients with thoracolumbar or lumbar kyphosis, the SFA method, FBI method, and Surgimap method can be used to predict the osteotomy angle precisely, however, considering the rationality of parameter settings and the operability, SFA method is relatively more suitable for such population.

Fruit development and softening play pivotal roles in determining fruit quality and post-harvest shelf life in Chinese bayberry (Myrica rubra). However, the specific role of beta (β)-galactosidase, particularly β-galactosidase of M. rubra (MrBGAL), in facilitating fruit softening remains unclear. In this study, we aimed to address this gap by investigating the involvement of MrBGALs genes in fruit softening. We identified all 15 MrBGALs and conducted a comprehensive analysis, including phylogenetic relationships, gene structure, protein motifs, co-linearity, and expression patterns. Using phylogenetic analysis, we classified all MrBGALs into five distinct groups. Additionally, cis-element prediction and comparative genome analysis provided insightful clues about the functionality of MrBGALs. Transcriptome data revealed unique expression patterns of MrBGALs throughout various fruit development stages. These findings introduce valuable candidate genes that can contribute to unraveling the functions and molecular mechanisms governing fruit development and softening in Chinese bayberry.

Significant temporal and spatial variability in soil moisture (SM) is observed during the warm season in China and its surrounding regions. Because of the existence of two different evapotranspiration regimes, i.e., soil moisture-limited and energy-limited, averaging the land–atmosphere (L–A) coupling strength for all soil wetness scenarios may result in the loss of coupling signals. This study examines the daytime-only L–A interactions under different soil moisture conditions, by using two-legged metrics in the warm season from May to September 1981–2020, partitioning the interactions between SM and latent heat flux (SM–LH, the land leg) from the interactions between latent heat flux and the lifting condensation level (LH–LCL, the atmospheric leg). The statistical results reveal large regional differences in warm-season daytime L–A feedback in China and its surrounding areas. As the soil becomes wetter, the positive SM–LH coupling strength increases in arid regions (e.g., northwest China, Hetao, and the Great Indian Desert) and the positive feedback shifts to the negative one in semi-arid/semi-humid regions (northeast and northern China). The negative LH–LCL coupling is most pronounced in wet soil months in arid regions, while the opposite is true for the Tibetan Plateau. In terms of intraseasonal variation, the large variability of SM in north China, the Tibetan Plateau, and India due to the influence of the summer monsoon leads to the sign change in the land segment coupling index, comparing pre- and post-monsoon periods. To further examine the impact of SM anomalies on L–A coupling and to explore evapotranspiration regimes in the North China Plain, four sets of sensitivity experiments with different soil moisture levels over a period of 10 years were conducted. Under relatively dry soil conditions, evapotranspiration is dominated by the soil moisture-limited regime with positive L–A coupling, regardless of external moisture inflow. The critical soil moisture value separating a soil moisture-limited and an energy-limited regime lies between 0.24 m3/m3 and 0.29 m3/m3. Stronger positive feedback under negative soil moisture anomalies may increase the risk of drought in the North China Plain.

In the current context of global urbanization and climate change, balancing ecological protection and economic development is a particular challenge in the optimal allocation of regional land use. Here, we propose a research framework for the optimal allocation of land use that considers the regional ecological security pattern (ESP) and allocates space for land-use activities to areas with low ecological risk. Taking Baicheng, China as our study area, ecological sources were first identified by integrating their ecological importance and landscape connectivity, and ecological corridors and functional zones were extracted using the minimum cumulative resistance difference and circuit theory. The ecological source areas were then taken as limiting factors, and four future scenarios were established for 2030 using the parcel-level land-use simulator (PLUS) model. The ecological corridors and functional zones served as areas having restricted ecological conditions, and the four future scenarios were coupled into the corresponding functional zones to optimize the land-use structure in 2030. The results indicate that under the coupled ESP–PLUS scenario, the spatial distribution and structure of land use in Baicheng balance the needs of ecological source area protection and economic development, resulting in greater sustainability. By 2030, the cultivated land area will steadily increase, but attention will also be given to the protection of ecological land (e.g., woodland and marshland), aligning with current policy planning demands. An analysis of the landscape indices for each future scenario found all scenarios to be effective in reducing negative changes in landscape patterns. These findings provide a novel perspective for the rational allocation of future land resources and the optimization of land-use structures.

Twig blight disease is the primary disease that affects the production of Myrica rubra in China. It was reported that exogenous brassinolide (BL) can improve disease resistance in plants. Here, we examined the effects of exogenous BL on disease resistance, chlorophyll contents, antioxidant enzyme activity, ROS accumulation, and key gene expression of M. rubra to analyze the mechanism of BR-induced resistance of twig blight disease in M. rubra. The results demonstrated that 2.0 mg·L−1 of BL could significantly lessen the severity of twig blight disease in M. rubra. Exogenous BL increased the contents of chlorophyll a, chlorophyll b, carotenoids, and total chlorophyll. Moreover, exogenous BL also significantly enhanced the activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), and decreased malondialdehyde (MDA) content and reactive oxygen species (ROS) accumulation in leaves, such as H2O2 and O2·−. Additionally, exogenous BL dramatically up-regulated the expression of pathogenesis-related (PR) genes such as MrPR1, MrPR2, and MrPR10, as well as important genes such as MrBAK1, MrBRI1, and MrBZR1 involved in brassinosteroid (BR) signaling pathway. The transcriptome analysis revealed that a total of 730 common differentially expressed genes (DEGs) under BL treatment were found, and these DEGs were primarily enriched in four Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Based on these findings, nine important candidate genes related to the resistance of twig blight disease under BL treatment were further identified. In this study, we elucidated the effects of exogenous BL on enhancing the resistance of M. rubra to twig blight disease and preliminary analyzed the potential mechanism of resistance induction, which will provide a crucial foundation for the management and prevention of twig blight disease in M. rubra.

Purpose: To evaluate the effects of age and gender on meibomian gland (MG) parameters and the associations among MG parameters in aged people using a deep-learning based artificial intelligence (AI).Methods: A total of 119 subjects aged ≥60 were enrolled. Subjects completed an ocular surface disease index (OSDI) questionnaire, received ocular surface examinations including Meibography images captured by Keratograph 5M, diagnosis of meibomian gland dysfunction (MGD) and assessment of lid margin and meibum. Images were analyzed using an AI system to evaluate the MG area, density, number, height, width and tortuosity.Results: The mean age of the subjects was 71.61 ± 7.36 years. The prevalence of severe MGD and meibomian gland loss (MGL) increased with age, as well as the lid margin abnormities. Gender differences of MG morphological parameters were most significant in subjects less than 70 years old. The MG morphological parameters detected by AI system had strong relationship with the traditional manual evaluation of MGL and lid margin parameters. Lid margin abnormities were significantly correlated with MG height and MGL. OSDI was related to MGL, MG area, MG height, plugging and lipid extrusion test (LET). Male subjects, especially the ones who smoke or drink, had severe lid margin abnormities, and significantly decreased MG number, height, and area than the females.Conclusion: The AI system is a reliable and high-efficient method for evaluating MG morphology and function. MG morphological abnormities developed with age and were worse in the aging males, and smoking and drinking were risk factors.

IntroductionGlycosylation is one of the essential post-translational modifications that influences the function of milk proteins.MethodsIn the present study, 998 proteins and 764 glycosylated sites from 402 glycoproteins were identified in human milk by TMT labeling proteomics. Compared to human milk proteins, the glycoproteins were mainly enriched in cell adhesion, proteolysis, and defense/immune process.ResultsThe abundance of 353 glycosylated sites and their 179 parent proteins was quantified. After normalization to their parent protein’s abundance, 78 glycosylated sites in 56 glycoproteins and 10 glycosylated sites in 10 glycoproteins were significantly higher in colostrum and mature milk, respectively. These changed glycoproteins were mainly related to host defense. Intriguingly, one glycosylated site (Asp144) in IgA and two glycosylated sites (Asp38 and Asp1079) in tenascin are significantly upregulated even though their protein abundance was downregulated during lactation.DiscussionThis study helps us figure out the critical glycosylated sites in proteins that might influence their biological function in an unbiased way.

Abstract Background Peste des petits ruminants (PPR) is a serious disease that affects goats, sheep and other small ruminants. As one of the earliest and most serious countries, PPR has seriously threatened India's animal husbandry economy. Results In this study, the spatiotemporal characteristics of the PPR in India outbreaks were analyzed. Between 2010 and 2018, the epidemic in India broke out all over the country in a cluster distribution. Epidemic clusters in northern and southern India are at higher risk, and the outbreak time of PPR has significant seasonality. The results of the analysis of the development and transmission of PPR under the natural infection conditions showed that the PPR outbreak in India reached a peak within 15 days. Finally, the quantitative risk analysis results based on scenario tree show showed that the average probability of infecting PPRV in live sheep exported from India was 1.45 × 10–4. Conclusions This study analyzed the prevalence of PPR in India. The analysis of transmission dynamics on the development of the epidemic provides a reference for the prevention and control of the epidemic. At the same time, it provides risk analysis and suggestions on trade measures for the trading countries of India.

Abstract Wind speed prediction has an important impact on the planning, economic operation and safe maintenance of wind power systems. However, the high volatility and intermittency of wind speed make it difficult to predict accurately. To improve the prediction accuracy, we developed a hybrid multistep wind speed prediction model named EWP‐CS‐RELM. In this model, a secondary decomposition technique of ensemble empirical mode decomposition (EEMD) and wavelet packet transform (WPT) is used, and it is called the EWP decomposition technique. This decomposition technique can achieve an adaptive processing of the data and accurately extract the characteristic components of the signal, avoiding the occurrence of pattern overlap and reducing the mutual interference between components. At the same time, the high and low‐frequency parts of the complex signal (component) can be decomposed into different frequency bands, and the corresponding frequency band can be selected adaptively to match the signal spectrum. The subsequence obtained after EWP decomposition is then predicted using a regularised extreme learning machine (RELM) optimised by the cuckoo search (CS) algorithm with strong global search ability to obtain the results. The hybrid prediction model is validated using four seasons of wind speed data from two wind farms in Shandong, China, and compared with seven other prediction models. Simulation results illustrate that the EWP‐CS‐RELM model outperforms the other seven models with the smallest statistical errors.

This research evaluated permeability and hydraulic conductivity of Open Graded Friction Course (OGFC) in multiple directions. Using Computational Fluid Dynamics (CFD), Ansys Fluent was utilized to simulate the laminar flow model for twelve cubic specimens of OGFC with different geometry and various porosities. Reynolds number (Re), permeability (K), and hydraulic conductivity (k) were computed for each simulation horizontally and vertically. According to the study, the horizontal permeability measurements were greater than the vertical measurements. Moreover, the permeability of OGFC is governed by the percentage of the air voids, aggregate particle size, and the thickness of the specimens. As the diameter and percentage of air voids increased, the permeability increased. Specimens with large size of aggregates and larger thicknesses have the highest values of permeability, while specimens with the small size of aggregates have the lowest values of permeability. In addition, the velocity of the flow inside the voids decreased as the diameter and percentage of air voids increased, while the pressure increased, according to the study.

Background: Ischemia reperfusion (I/R) play an imperative role in the expansion of cardiovascular disease. Sinomenine (SM) has been exhibited to possess antioxidant, anticancer, anti-inflammatory, antiviral and anticarcinogenic properties. The aim of the study was scrutinized the cardioprotective effect of SM against I/R injury in rat. Methods: Rat were randomly divided into normal control (NC), I/R control and I/R + SM (5, 10 and 20 mg/kg), respectively. Ventricular arrhythmias, body weight and heart weight were estimated. Antioxidant, inflammatory cytokines, inflammatory mediators and plasmin system indicator were accessed. Results: Pre-treated SM group rats exhibited the reduction in the duration and incidence of ventricular fibrillation, ventricular ectopic beat (VEB) and ventricular tachycardia along with suppression of arrhythmia score during the ischemia (30 and 120 min). Stress response in plant is regulated by a large number of genes co-operating in diverse networks that serve multiple adaptive process. To understand how gene regulatory networks (GRNs) modulating abiotic stress responses, we compare the GRNs underlying drought and cold stresses using samples collected at 4 or 6 h intervals within 48 h in Chinese bayberry (Myrica rubra). We detected 7,583 and 8,840 differentially expressed genes (DEGs) under drought and cold stress respectively, which might be responsive to environmental stresses. Drought- and cold-responsive GRNs, which have been built according to the timing of transcription under both abiotic stresses, have a conserved trans-regulator and a common regulatory network. In both GRNs, basic helix-loop-helix family transcription factor (bHLH) serve as central nodes. MrbHLHp10 transcripts exhibited continuous increase in the two abiotic stresses and acts upstream regulator of ASCORBATE PEROXIDASE (APX) gene. To examine the potential biological functions of MrbHLH10, we generated a transgenic Arabidopsis plant that constitutively overexpresses the MrbHLH10 gene. Compared to wild-type (WT) plants, overexpressing transgenic Arabidopsis plants maintained higher APX activity and biomass accumulation under drought and cold stress. Consistently, RNAi plants had elevated susceptibility to both stresses. Taken together, these results suggested that MrbHLH10 mitigates abiotic stresses through the modulation of ROS scavenging.

The NAC transcription factor family is a large plant gene family, participating in plant growth and development, secondary metabolite synthesis, biotic and abiotic stresses responses, and hormone signaling. Eucommia ulmoides is a widely planted economic tree species in China that can produce trans-polyisoprene: Eucommia rubber (Eu-rubber). However, genome-wide identification of the NAC gene family has not been reported in E. ulmoides. In this study, 71 NAC proteins were identified based on genomic database of E. ulmoides. Phylogenetic analysis showed that the EuNAC proteins were distributed in 17 subgroups based on homology with NAC proteins in Arabidopsis, including the E. ulmoides-specific subgroup Eu_NAC. Gene structure analysis suggested that the number of exons varied from 1 to 7, and multitudinous EuNAC genes contained two or three exons. Chromosomal location analysis revealed that the EuNAC genes were unevenly distributed on 16 chromosomes. Three pairs of genes of tandem duplicates genes and 12 segmental duplications were detected, which indicated that segmental duplications may provide the primary driving force of expansion of EuNAC. Prediction of cis-regulatory elements indicated that the EuNAC genes were involved in development, light response, stress response and hormone response. For the gene expression analysis, the expression levels of EuNAC genes in various tissues were quite different. To explore the effect of EuNAC genes on Eu-rubber biosynthesis, a co-expression regulatory network between Eu-rubber biosynthesis genes and EuNAC genes was constructed, which indicated that six EuNAC genes may play an important role in the regulation of Eu-rubber biosynthesis. In addition, this six EuNAC genes expression profiles in E. ulmoides different tissues were consistent with the trend in Eu-rubber content. Quantitative real-time PCR analysis showed that EuNAC genes were responsive to different hormone treatment. These results will provide a useful reference for further studies addressing the functional characteristics of the NAC genes and its potential role in Eu-rubber biosynthesis.

Camel milk has unique compositional, functional and therapeutic properties compared to cow's milk and also contains many protective proteins with anti-cancer, anti-diabetic and anti-bacterial properties. In this experiment, fresh camel milk was heat-treated at different temperatures and times, and the changes in Millard reaction products were analyzed. Meanwhile, headspace-gas chromatography-ion migration spectrometry (HS-GC-IMS), electronic nose and electronic tongue were used to analyze the changes of volatile components in camel milk after different heat treatments. The results showed that the Maillard reaction was more severe with the increase of heat treatment, and the contents of furosine and 5-hydroxymethylfurfural increased significantly when the heat treatment temperature was higher than 120°C. HS-GC-IMS results showed that the contents of aldehydes and ketones increased obviously with the increase of heat treatment degree. The study clarifies the effects of different heat treatment degrees on Maillard reaction degree and flavor of camel milk, which has practical production guidance significance for the research and industrialization of liquid camel milk products.

Ulcerative colitis (UC) is a non-specific inflammatory bowel illness characterized by intestinal mucosal barrier degradation, inflammation, oxidative damage, and gut microbiota imbalances. Rosa roxburghii Tratt Fruit extract (RRTE) was extracted from Rosa roxburghii Tratt fruit, exhibiting an excellent prevention effect against UC; RRTE could prevent the damage of DSS-induced human normal colonic epithelial (NCM 460) cells, especially in cell viability and morphology, and oxidative damage. Additionally, in UC mice, RRTE could limit the intestinal mucosal barrier by increasing the expression of intestinal tight junction proteins and mucin, reducing inflammation and oxidative damage in colon tissue. More importantly, RRTE can increase the abundance of beneficial bacteria to regulate gut microbiota such as Ruminococcus, Turicibacter, and Parabacteroides, and reduce the abundance of harmful bacteria such as Staphylococcus and Shigella. Furthermore, transcriptomics of colonic mucosal findings point out that the beneficial effect of RRTE on UC could be attributed to the modulation of inflammatory responses such as the IL-17 and TNF signaling pathways. The qPCR results confirm that RRTE did involve the regulation of several genes in the IL-17 signaling pathway. In conclusion, RRTE could prevent DSS-induced damage both in vitro and in vivo.

As a subtropical and tropical tree, bayberry (Myrica rubra) is an important fruit tree grown commercially in southern China. Interestingly, our studies found that the fruit quality of bayberry with accompanying ryegrass was significantly improved, but its mechanism remains unclear. The aim of this study was to explore the mechanism of accompanying ryegrass on the beneficial effect of the fruit quality of bayberry by measuring the vegetative growth parameters, fruit parameters with economic impact, physical and chemical properties of rhizosphere soil, microbial community structure, and metabolites of the bayberry with/without ryegrass. Notably, the results revealed a significant difference between bayberry trees with and without accompanying ryegrass in fruit quality parameters, soil physical and chemical properties, microbial community structure, and metabolites. Compared with the control without accompanying ryegrass, the planting of ryegrass increased the titratable sugar, vitamin C, and titratable flavonoid contents of bayberry fruits by 2.26%, 28.45%, and 25.00%, respectively, and decreased the titratable acid contents by 9.04%. Furthermore, based on 16S and ITS amplicon sequencing of soil microflora, the accompanying ryegrass caused a 12.47% increment in Acidobacteriota while a 30.04% reduction in Actinobacteria was recorded, respectively, when compared with the bayberry trees without ryegrass. Redundancy discriminant analysis of microbial communities and soil properties indicated that the main variables of the bacterial community included available nitrogen, available phosphorus, exchangeable aluminum, and available kalium, while the main variables of the fungal community included exchangeable aluminum, available phosphorus, available kalium, and pH. In addition, the change in microbial community structure was justified by the high correlation analysis between microorganisms and secondary metabolites. Indeed, GC-MS metabolomics analysis showed that planting ryegrass caused a 3.83%–144.36% increase in 19 metabolites such as 1,3-Dipentyl-heptabarbital and carbonic acid 1, respectively, and a 23.78%–51.79% reduction of 5 metabolites compared to the bayberry trees without the accompanying ryegrass. Overall, the results revealed the significant change caused by the planting of ryegrass in the physical and chemical properties, microbiota, and secondary metabolites of the bayberry rhizosphere soils, which provides a new insight for the ecological improvement of bayberry.

This study investigated the effects of five lipases on enzyme-modified cheese (EMC) flavor development. Results showed that lipase 30SD contained high hydrolytic activity for short, medium, and long-chain fatty acids within 24 h incubation time, and the highest content of them among different times could reach 47.24, 475.90, 1 563.92 mg/100 g fat, respectively. Lipase DF15 and MER showed moderate capacity to hydrolyze volatile fatty acids, while lipase F3G had a stronger ability to produce long-chain fatty acids. Twenty-seven new volatiles were formed during lipolysis, most of them were acids and esters. Principal component analysis results showed that EMC produced by lipase 30SD for 18 h was similar to the commercial product with a pungent, rancid, and cheddar flavor. EMCs produced by lipase DF15 were significantly distinguished from other products by their high content of ethyl heptanoate, ethyl nonanoate, and ethyl tridecanoate. The findings might be useful for the researchers who focus on lipolysis or EMC products.

The interaction between shale and various fluids is crucial as it modifies pore structures, which govern the effective development of shale gas and the geological storage of carbon dioxide in shale formations. In this study, samples from the Longmaxi Formation shale in Sichuan Basin of China were exposed to different fluids, including 6 MPa CO2, 12 MPa CO2, 6 MPa CO2+brine, and 12 MPa CO2+brine, at 45 °C for 100 days. Various methods, including X-ray diffraction (XRD), X-ray fluorescence (XRF), field-emission scanning electron microscopy (FESEM), and the low-pressure gas adsorption (N2) test, were adopted to evaluate chemical and structural changes during the exposure process. After being treated with supercritical CO2+brine and subcritical CO2+brine, the shale underwent significant changes in its major element composition. The content of Ca, Al, and K in shale saturated with supercritical CO2+brine decreased from 13.00% to 10.34%, from 3.65% to 3.36%, and from 1.56% to 1.37%, respectively. Meanwhile, the content of Si and Na in the same shale increased slightly after saturation. The amount of quartz and dolomite increased, while the levels of clay and calcite slightly decreased. The surface of the shale sample became rougher and small bumps and cracks appeared after saturation with different fluids, as shown by the FESEM analysis results. Furthermore, the changes in both the total pore volume and pore size followed a similar pattern to the alterations in the specific surface areas. The highest level of variation occurred with the shale that was saturated with 12 MPa of CO2, indicating that gas pressure and CO2 phase state have a significant influence on the shale’s pore structure. In addition, the distribution of pore sizes showed a bias towards larger sizes across all diameters; this suggests that the reaction resulted in a decrease in the number of micropores. This also highlights that the impact of varying fluid saturation was primarily focused on micropores and macropores. The results of this study provided experimental evidence to further test the mechanisms and permeability of geological storage of CO2 in organic-rich self-sourced shale.

Temporal and spatial variations in land use/land cover (LULC) and their driving factors are direct reflections of regional natural and anthropogenic impacts. To explore the pathways for green upgrading development in ecologically fragile areas, this study focused on Baicheng, located in the northern agropastoral transition zone, China. Based on the topographic map of 1954 and Landsat remote sensing images taken from 1976 to 2020, the spatial distribution of LULC data for the study area in 1954, 1976, 1988, 2000, 2010, and 2020 was obtained. The temporal and spatial characteristics of LULC changes and their driving factors under the combined influence of human activities and climate were analyzed using dynamic degree, flowchart, spatial analysis, and principal component analysis. The results indicate that (1) the dominant LULC type in Baicheng is cropland. By 2020, dry land accounted for over 41% of the total area, while the area of saline–alkaline land increased the most, and grassland decreased most drastically. (2) The dynamic degree of different LULC types ranked from highest to lowest as follows: paddy field > unused land and other types > woodland > saline–alkaline land. (3) LULC Changes in Baicheng were mainly influenced by human activities and economic development, especially regional gross domestic product and the sown area of crops. These research findings can provide a scientific basis for formulating sustainable development and protection strategies to ensure regional green upgrading development.

The pore structure of uranium-bearing sandstone is one of the critical factors that affect the uranium leaching performance. In this article, uranium-bearing sandstone from the Yili Basin, Xinjiang, China, was taken as the research object. The fractal characteristics of the pore structure of the uranium-bearing sandstone were studied using mercury intrusion experiments and fractal theory, and the fractal dimension of the uranium-bearing sandstone was calculated. In addition, the effect of the fractal characteristics of the pore structure of the uranium-bearing sandstone on the uranium leaching kinetics was studied. Then, the kinetics was analyzed using a shrinking nuclear model, and it was determined that the rate of uranium leaching is mainly controlled by the diffusion reaction, and the dissolution rate constant (K) is linearly related to the pore specific surface fractal dimension (DS) and the pore volume fractal dimension (DV). Eventually, fractal kinetic models for predicting the in-situ leaching kinetics were established using the unreacted shrinking core model, and the linear relationship between the fractal dimension of the sample's pore structure and the dissolution rate during the leaching was fitted.

The Mongolian Plateau is an arid and semi-arid region with grassland as its main vegetation. It has a fragile ecosystem and is a sensitive area for global warming. The study is based on MODIS NDVI data and growth season meteorological data from 2000 to 2018, this study examined the spatial and temporal variation characteristics of grassland vegetation on the Mongolian Plateau during the growing season using trend analysis, partial correlation analysis, and residual analysis, and it explores the dual response of NDVI changes to climate and human activities. The study’s findings demonstrated that the growing season average NDVI of grassland vegetation on the plateau gradually increased from southwest to northeast during the growing season; the growing season average NDVI demonstrated a significant overall increase of 0.023/10a (p < 0.05) from 2000 to 2018, with an increase rate of 0.030/10a in Inner Mongolia and 0.019/10a in Mongolia; the area showing a significant increase in NDVI during the growing season accounted for 91.36% of the entire study area. In Mongolian Plateau grasslands during the growing season of 2000–2018, precipitation and downward surface shortwave radiation grew significantly at rates of 34.83mm/10a and 0.57 W/m2/10a, respectively, while average air temperature decreased slightly at a rate of −0.018°C/10a. Changes in meteorological factors of grassland vegetation varied by region as well, with Inner Mongolia seeing higher rates of precipitation, lower rates of average air temperature, and lower rates of downward surface shortwave radiation than Mongolia. On the Mongolian Plateau, the NDVI of grassland vegetation in the growing season showed a significant positive correlation with precipitation (0.31) and a significant negative correlation with average air temperature (−0.09) and downward surface shortwave radiation (−0.19), indicating that increased in NDVI was driven by an increase in precipitation paired with a decrease in air temperature and a decrease in surface shortwave radiation. The overall increase in NDVI caused by human activity in the grasslands of the Mongolian Plateau was primarily positive, with around 18.37% of the region being beneficial. Climate change and human activity both affect NDVI variations in Mongolian Plateau grasslands, which are spatially heterogeneous. Moderate ecological engineering and agricultural production activities are crucial for vegetation recovery. This work is crucial to further understanding surface–atmosphere interactions in arid and semi-arid regions in the context of global climate change.

Chinese bayberry (Myrica rubra) is an important tree in South China, with its fruit being of nutritional and high economic value. In this study, early ripening (ZJ), medium ripening (BQ) and late ripening (DK) varieties were used as test materials. Young leaves of ZJ, BQ and DK in the floral bud morphological differentiation periods were selected for transcriptome sequencing to excavate earliness related genes. A total of 4,538 differentially expressed genes were detected. Based on clustering analysis and comparisons with genes reportedly related to flowering in Arabidopsis thaliana, 25 homologous genes were identified. Of these, one gene named MrSPL4 was determined, with its expression down-regulated in DK but up-regulated in ZJ and BQ. MrSPL4 contained SBP domain and the target site of miR156, and its total and CDS length were 1,664 bp and 555 bp respectively. The overexpression vector of MrSPL4 (35S::35S::MrSPL4-pCambia2301-KY) was further constructed and successfully transfected into tobacco to obtain MrSPL4-positive plants. Based on the results of qRT-PCR, the relative expression of MrSPL4 was up regulated by 3,862.0-5,938.4 times. Additionally, the height of MrSPL4-positive plants was also significantly higher than that of wild-type (WT), with the bud stage occurring 12 days earlier. Altogether, this study identified an important gene -MrSPL4 in Chinese bayberry, which enhanced growth and flowering, which provided important theoretical basis for early-mature breeding of Chinese bayberry.

Objective: Keluoxin capsule (KLXC) has been widely used in diabetic kidney disease (DKD), but its efficacy and safety have not yet been clarified. A systematic review and meta-analysis were performed to assess the efficacy and safety of KLXC for DKD.Methods: The randomized control trials (RCTs) included KLXC searched from seven major English and Chinese databases up until 3 June 2022. The methodological quality and risk of bias were assessed by version 2 of the Cochrane risk-of-bias tool (RoB 2) for the RCTs from the Cochrane Handbook. The analyses were conducted by RevMan 5.4 and Stata 17.0.Results: A total of 20 trials with 1,500 participants were identified. The meta-analysis showed that KLXC combined with Western medicine was superior to the use of Western medicine alone for DKD which included improvements in the estimated glomerular filtration rate (eGFR) [MD = 3.04, 95% CI (0.30, 5.78), p = 0.03], reduction in microalbuminuria (mALB) [MD = −25.83, 95% CI (−41.20, −10.47), p = 0.001], urinary albumin excretion rate (UAER) [SMD = −0.97, 95% CI (−1.50, −0.44), p = 0.0003], 24-h urine protein (24hUpro) [SMD = −1.31, 95% CI (−1.82, −0.80), p < 0.00001], serum creatinine (Scr) [MD = −11.39, 95% CI (−18.76, −4.02), p = 0.002], blood urea nitrogen (BUN) [MD = −1.28, 95% CI (−1.67, −0.88), p < 0.00001], fasting blood glucose (FBG) [MD = −0.51, 95% CI (−0.90, −0.11), p = 0.01], total cholesterol (TC) [MD = −1.04, 95% CI (−1.40, −0.68), p < 0.00001], triglycerides (TG) [MD = −0.36, 95% CI (−0.50, −0.23), p < 0.00001], and low-density lipoprotein cholesterol (LDL) [MD = −0.39, 95% CI (−0.71, −0.07), p = 0.02]. Results showed no statistically significant difference in glycated hemoglobin (HbA1c) (p = 0.14) or adverse events (p = 0.81) between the two groups.Conclusion: The combination of KLXC and Western medicine had a positive effect on DKD. However, due to the high clinical heterogeneity and low quality of included studies, further standardized, large-scale, rigorously designed RCTs for DKD in the definitive stage are still necessary to achieve more accurate results.Systematic Review Registration:https://inplasy.com/inplasy-2021-11-0067/, identifier INPLASY 2021110067.

Ehrlichia chaffeensis, the causative agent of human monocytic ehrlichiosis (HME), is a Gram-negative obligatory intracellular bacterium, which infects and multiplies in human monocytes and macrophages. Host immune cells produce reactive oxygen species (ROS) to eliminate E. chaffeensis upon infection. E. chaffeensis global transcriptional regulator CtrA activates the expression of GshA and GshB to synthesize glutathione (GSH), the most potent natural antioxidant, upon oxidative stress to combat ROS damage. However, the mechanisms exploited by E. chaffeensis to utilize GSH are still unknown. Here, we found that in E. chaffeensis CtrA activated the expression of glutathione S-transferase (GST) upon oxidative stress, and E. chaffeensis GST utilizes GSH to eliminate ROS and confers the oxidative stress resistance to E. chaffeensis. We found that CtrA bound to the promoter regions of 211 genes, including gst, in E. chaffeensis using chromatin immunoprecipitation coupled to deep sequencing (ChIP-seq). Recombinant E. chaffeensis CtrA directly bound to the gst promoter region determined with electrophoretic mobility shift assay (EMSA), and activated the gst expression determined with reporter assay. Recombinant GST showed GSH conjugation activity towards its typical substrate 2,4-dinitrochlorobenzene (CDNB) in vitro and peptide nucleic acid (PNA) transfection of E. chaffeensis, which can knock down the gst transcription level, reduced bacterial survival upon oxidative stress. Our results demonstrate that E. chaffeensis CtrA regulates GSH utilization, which plays a critical role in resistance to oxidative stress, and aid in the development of new therapeutics for HME.
