Background: The genus Haematopinus contains 21 species of blood-sucking lice, parasitizing both even-toed ungulates (pigs, cattle, buffalo, antelopes, camels and deer) and odd-toed ungulates (horses, donkeys and zebras). The mitochondrial genomes of the domestic pig louse, Haematopinus suis, and the wild pig louse, Haematopinus apri, have been sequenced recently; both lice have fragmented mitochondrial genomes with 37 genes on nine minichromosomes. To understand whether the composition of mitochondrial minichromosomes and the gene content and gene arrangement of each minichromosome are stable within the genus, we sequenced the mitochondrial genome of the horse louse, Haematopinus asini. Methods: We used a PCR-based strategy to amplify four mitochondrial minichromosomes in near full-length, and then amplify the entire coding regions of all of the nine mitochondrial minichromosomes of the horse louse. These amplicons were sequenced with an Illumina Hiseq platform. Results: We identified all of the 37 mitochondrial genes typical of bilateral animals in the horse louse, Haematopinus asini; these genes are on nine circular minichromosomes. Each minichromosome is 3.5–5.0 kb in size and consists of a coding region and a non-coding region except R-nad4L-rrnS-C minichromosome, which contains two coding regions and two non-coding regions. Six of the nine minichromosomes of the horse louse have their counterparts in the pig lice with the same gene content and gene arrangement. However, the gene content and arrangement of the other three minichromosomes of the horse louse, including R-nad4L-rrnS-C, are different from that of the other three minichromosomes of the pig lice. Conclusions: Comparison between the horse louse and the pig lice revealed variation in the composition of mitochondrial minichromosomes within the genus Haematopinus, which can be accounted for by gene translocation events between minichromosomes. The current study indicates that inter-minichromosome recombination plays a major role in generating the variation in the composition of mitochondrial minichromosomes and provides novel insights into the evolution of fragmented mitochondrial genomes in the blood-sucking lice. We conducted a molecular phylogenetic analysis based on mitochondrial cytochrome oxidase subunit I and nuclear 28S rRNA gene sequences of species of Japanese elmids (23 species from 12 genera) and examined the hind-wings of 24 species in order to determine the incidence of hind-wing degeneration among species and the presence of dimorphic species with respect to hindwing degeneration. Based on the molecular phylogenetic analysis, we determined that the previously separated winged and wingless species, Stenelmis vulgaris and S. miyamotoi, and Leptelmis gracilis and L. parallela, are two forms of the same species. Of the 24 species whose hind wings were studied, we found apterous (3 species of Zaitzeviaria), brachypterous (2 species of each of Optioservus and Paramacronychus) and dimorphic species (2 species as above) in separate clades of the phylogeny. These were the smallest or medium-sized species. Dimorphic species occurred in mid- to downstream areas and used reeds and wood as substrates. The percentage of species with hind-wing degeneration (wingless or dimorphic) was high among the species (29%) studied compared to the perceived percentage for temperate beetles (< 10%). Thus, we found that the degeneration of hind wings has occurred repeatedly in these elmid species. However, we identified only ambiguous habitat and life history correlates of hind-wing degeneration, and the adaptive significance of hind-wing degeneration in these species of elmids remains unclear. The phylogenetic relationships among the Japanese members of the genus Eubrianax (Coleoptera: Psephenidae) were examined using the mitochondrial cytochrome oxidase subunit I ( COI) gene and nuclear 28S rRNA gene sequences. Based on the molecular phylogeny as well as morphological features, the species status of Eubrianax brunneicornisNakane, 1952 was proposed. The phylogenetic analyses recovered monophyly of the previously proposed pellucidus species group with four Japanese species, whereas a single Japanese species of the granicollis group was included in the lineage of the ramicornis group with five Japanese species. The divergence times of the species were estimated by dating the phylogenetic tree against the fossil record and a molecular clock based on the COI gene. Mating frequency has important implications for patterns of sexual selection and sexual conflict and hence for issues such as speciation and the maintenance of genetic diversity. Knowledge of natural mating patterns can also lead to more effective control of pest tephritid species, in which suppression programmes, such as the sterile insect technique (SIT) are employed. Multiple mating by females may compromise the success of SIT. We investigated the level of polyandry and sperm utilization in a Brisbane field population of the tropical fruit fly, Bactrocera cacuminata (Hering), using seven polymorphic microsatellite loci. The offspring of 22 wild-caught gravid females were genotyped to determine the number of males siring each brood and paternity skew, using the programsgerud andscare. Our data showed that 22.7% of females produced offspring sired by at least two males. The mean number of mates per female was 1.72. Paternal contributions of double-sired broods were skewed with the most successful male having sired between 76.9% and 87.5% of the offspring. These results have implications for SIT, because the level of remating we have identified would indicate that wild females could mate with one or more resident fertile males. [ABSTRACT FROM AUTHOR]

Bactrocera cacuminata (Hering) is a nonpest member of the Bactrocera dorsalis complex offering a platform to check hypotheses extrapolated from the more studied pest species. Six polymorphic microsatellite loci were isolated from enriched genomic libraries constructed using a biotin/streptavidin capture protocol. Allele number varied between three and nine; the expected heterozygosity ranged between 0.29 and 0.81. No significant deviations from Hardy–Weinberg equilibrium or linkage disequilibrium were found.
