Novel nano-heterojunctions with excellent antibacterial properties are regarded as effective alternatives to antibiotics for bacterial disinfection. Herein, we have developed a universal strategy to synthesize a series of urchin-shaped Prussian blue analogues@Prussian blue (PBA@PB) nano-heterojunctions via a facile co-precipitation route for efficient bacterial disinfection, for the first time, which could overcome the disadvantages of singe PBA or PB. The optimized PBA@PB nano-heterojunctions showed remarkable bactericidal efficiency ability against multi drug-resistant (MDR) Escherichia coli (E.coli) and methicillin-resistant Staphylococcus aureus (MRSA), (99.4% and 98.8% bactericidal ratios, respectively), attributed to their outstanding photothermal and catalytic performance. It should be emphasized that the as-prepared nano-heterojunctions also showed amazing bacterial elimination efficiency in sewage. The antibacterial mechanism was also investigated using a variety of spectroscopic and microscopic techniques. It can be attributed to the formation of unique nano-heterojunctions between PB and PBA, which improves the absorption of NIR light and facilitates the separation of generated electron-hole pairs. Therefore, the urchin-shaped PBA@PB nano-heterojunctions may be promising candidates for rapidly bacterial elimination in environmental remedy and wastewater treatment.

This work presents a novel laser continuous powder bed fusion (L-CPBF) process technique, tailored for the additive manufacturing of intricate components with annular hollow structures, emphasizing both precision and efficiency. Utilizing this advanced L-CPBF method, Ti-6.5Al-2Zr-Mo-V (TA15) titanium alloy samples were successfully fabricated, and their microstructures and properties were investigated. The optimized process parameter window for L-CPBF processing of TA15 was determined through Taguchi experiments, ranging from 100 to 125 J/mm3, with the relative density of the samples achieved an outstanding 99.95%. Microstructural evolution under varying volumetric energy densities (VED) was revealed through characterization methods such as SEM and EBSD. The formation of acicular α′-martensites was observed during initial the cooling phase, with thermal cycling temperature playing a pivotal role in the development of martensites of varying sizes. Furthermore, through process optimization, remarkable mechanical properties, with a tensile strength of 1200.5 ± 98.5 MPa, a yield strength of 1109 ± 102 MPa, and an elongation of 7.05 ± 1.35% were achieved. These mechanical properties are comparable to those of TA15 alloys prepared by traditional reciprocating scraper powder spreading L-PBF. However, our approach boasts a substantial increase in manufacturing efficiency, surpassing the traditional method by over 45%. Collectively, this study offers invaluable theoretical and experimental insights, paving the way for accelerated additive manufacturing of hollow annular components in demanding sectors such as aerospace and beyond.

Abstract Background Wolfram syndrome (WS) is a rare autosomal recessive multisystem neurodegenerative disease characterized by non-autoimmune insulin-dependent diabetes mellitus, optic atrophy, sensorineural deafness, and diabetes as the main features. Owing to clinical phenotypic heterogeneity, the misdiagnosis rate is high. However, early accurate diagnosis and comprehensive management are key to improving quality of life and prolonging life. Results Eleven patients from seven WS pedigrees with 10 mutation sites (c.1314_1317delCTTT, c.C529T, c.C529A, c.G2105A, c.C1885T, c.1859_1860del, c.G2020A, c.C529A, c.G2105A, and c.G1393C) in the WFS1 gene were included. We conducted further expert department analysis to clarify the diagnosis and analyze the correlation between genes and phenotypes. Conclusions The genotypes of these patients were closely associated with their phenotypes. The clinical data of the patients were analyzed to provide a basis for the diagnosis and clinical management of the disease.

Ultrasonic-assisted Ti6Al4V Directed Energy Deposition (DED) additive manufacturing technology can improve the problem of uneven microstructure caused by laser heating and sudden cooling of the molten pool. In this paper, the numerical analysis and experimental verification methods were adopted. The influencing factors, such as the cavitations’ effect, sound flow enhancement effect, and sound flow thermal effect related to the ultrasonic assistance in the molten pool, were analyzed. After equating the energy of the ultrasound, the model of additive manufacturing was introduced in the form of a heat source. The temperature gradient changes during the solidification process of the molten pool with the addition of ultrasound assistance and the effect of ultrasonic vibration during the manufacturing process on its deposited state and microstructure of solution-aged formed parts were studied. The results showed that when the wire feeding rate is 5 mm/s and the laser scanning speed is 5 mm/s, the optimal laser power is 1000 W~1100 W, corresponding to the optimal ultrasonic amplitude of 120 μm. Then, by comparing the temperature field with the same amplitude of 0 μm (i.e., no ultrasonic vibration) and the microstructure of the formed parts, it was verified that ultrasonic vibration facilitates fluid flow in the molten pool, which could lead to a more uniform temperature distribution. This optimized approach not only enhances the understanding of the process but also contributes significantly to the advancement of related research endeavors.

Using finite element simulation and single-variable experimental methods, this study analyzes the variations in the microstructure and hardness of a 304 stainless steel wire during Joule heating fused filament fabrication. The effects of current intensity, printing speed, and roller pressure on the macroscopic morphology, microstructure, and microhardness of a single-layer single-channel formation were investigated. The results indicate that when the current intensity is 400 A, the printing speed is 1000 mm/min, and the roller pressure is 0.3 N, the surface of the single-layer single-channel formation is smooth and exhibits optimal forming characteristics with a width-to-height ratio of 3.23, a dilution rate of 51.61%, and an average microhardness of 238.17 HV. As the current intensity increases, the microstructure in the fusion zone initially decreases in size and then increases; similarly, with the increase in printing speed, the microstructure in the fusion zone first decreases and then increases; as the roller pressure increases, the microstructure in the fusion zone initially increases in size and then decreases. The microhardness initially increases and then decreases with the increase in process parameters, resulting in uneven hardness distribution due to the variations in microstructure size. The optimal combination of process parameters achieves a balance between heat input, cooling rate, and growth rate, thereby achieving grain refinement and hardness improvement, ultimately enhancing the mechanical properties of the material.

Abstract Silicon monoxide (SiO) (silicon [Si] mixed with silicon dioxide [SiO2])/graphite (Gr) composite material is one of the most commercially promising anode materials for the next generation of high‐energy‐density lithium‐ion batteries. The major bottleneck for SiO/Gr composite anode is the poor cyclability arising from the stress/strain behaviors due to the mismatch between two heterogenous materials during the lithiation/delithiation process. To date, a meticulous and quantitative understanding of the highly nonlinear coupling behaviors of such materials is still lacking. Herein, an electro–chemo–mechanics‐coupled detailed model containing particle geometries is established. The underlying mechanism of the regulation between SiO and Gr components during electrochemical cycling is quantitatively revealed. We discover that increasing the SiO weight percentage (wt%) reduces the utilization efficiency of the active materials at the same 1 C rate charging and enhances the hindering effects of stress‐driven flux on diffusion. In addition, the mechanical constraint demonstrates a balanced effect on the overall performance of cells and the local behaviors of particles. This study provides new insights into the fundamental interactions between SiO and Gr materials and advances the investigation methodology for the design and evaluation of next‐generation high‐energy‐density batteries.

Abstract Background and objective As a common chronic disease, diabetes is called the “second killer” among modern diseases. Currently, there is no medical cure for diabetes. We can only rely on medication for auxiliary treatment. However, many diabetic patients still die each year. In addition, a considerable number of people do not pay attention to their physical health or opt out of treatment due to lack of money, which eventually leads to various complications. Therefore, diagnosing diabetes at an early stage and intervening early is necessary; thus, developing an early detection method for diabetes is essential. Methods In this study, a diabetes prediction model based on Boruta feature selection and ensemble learning is proposed. The model contains the use of Boruta feature selection, the extraction of salient features from datasets, the use of the K-Means++ algorithm for unsupervised clustering of data and stacking of an ensemble learning method for classification. It has been validated on a diabetes dataset. Results The experiments were performed on the PIMA Indian diabetes dataset. The model was evaluated by accuracy, precision and F1 index. The obtained results show that the accuracy rate of the model reaches 98% and achieves good results. Conclusion Compared with other diabetes prediction models, this model achieved better results, and the obtained results indicate that this model is superior to other models in diabetes prediction and has better performance.

To explore the application effects of the model of Fenlong “145” on newly planted sugarcane and to analyze its physiological and ecological bases, Guiliu 05136 was used as the experimental material. Conventional tillage (CK) and the model of Fenlong “145” (FL145) were set in this study. Soil properties, and agronomic characters, photosynthetic characteristics, yield and quality of newly planted sugarcane were measured under field conditions, and economic benefits were analyzed. The results were as follows: (1) Compared with CK, at 0-20 and 20-40 cm depth, the soil bulk density in FL145 were significantly decreased by 1.25%-5.98%, while the soil porosity and the content of soil water were significantly increased by 1.08%-4.77% and 1.78%-8.23%, respectively. (2) FL145 could promote the growth of roots and seedling emergence of newly planted sugarcane. The agronomic characters in FL145 showed well, and the plant height were significantly increased by 2.20%-7.86%. (3) The leaf area per plant, the chlorophyll content, the net photosynthetic rate, stomatal conductance, and intercellular CO2 concentration were significantly increased by 15.88%, 1.41%, 6.84%, 18.67%, 10.06%, respectively, photosynthetic capacity was enhanced, and the dry matter accumulation per plant in FL145 was significantly increased by 9.26%. (4) At harvest, the number of millable canes, stem length and stem diameter of newly planted sugarcane in FL145 were significantly increased. The theoretical yield and actual yield in FL145 significantly were increased by 5.07% and 5.11%, respectively, while the sucrose content in cane juice and the Brix were significantly increased by 1.61% and 1.50%, respectively, and the content of reducing sugar was significantly decreased by 12.50%. (5) Compared with CK, the total production cost of newly planted sugarcane in FL145 was reduced by 378 yuan·hm-2, while the total output value and the profit were increased by 1 934.4 and 2 312.4 yuan·hm-2, respectively. Therefore, FL145 has the effect of reducing cost and increasing efficiency in the production of newly planted sugarcane by reducing the investment in soil preparation and ditching and chemical weeding. Meanwhile, FL145 creates better soil conditions, and has a positive effect on the root growth, thereby promoting photosynthetic capacity and the growth and development of plant, and has a positive effect on the accumulation of dry matter and sugar, resulting in the increase of the quality and yield of newly planted sugarcane.

In this study, a solution to the issue of a large heat-affected zone in Wire Arc Additive Manufacturing is presented by employing the Joule Heat Additive Manufacturing method to create a single layer and single channel with a reduced heat-affected zone. The microstructure of the single layer and single channel is thoroughly investigated using various detection methods, including optical microscopy (OM), scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), and electron backscatter diffraction (EBSD). The results reveal that the heat-affected zone formed by the Joule Heat Additive Manufacturing method is smaller than that produced by the Wire Arc Additive Manufacturing method. Additionally, the grains in the single layer and single channel progress from planar to columnar, then equiaxed, and finally back to columnar from the fusion line to the top of the wire. The element content and distribution are relatively uniform. The microstructure of the single layer consists of austenite and a small amount of ferrite, with austenite accounting for 99.71% of the content. The grain size in the middle of the wire is mainly around 10 μm, with the smallest angle grain boundaries within 10°. The distribution of local grain orientation differences in the three regions is found to be largely consistent. The analysis of the microstructure of the single layer and single channel serves as a valuable reference for understanding the behavior of single-channel multi-layers in future studies.

Sugarcane yield in China is low because of the shallow A-horizon soil layer, or as it is commonly called by farmers, the “plow soil layer”, as well as low soil organic matter and fertilizer utilization efficiency. Fenlong-ridging deep tillage (FT), also called vertical rotary tillage, and amendment with biochar have been shown to improve soil quality and crop yield. In this study, field trials were conducted with newly planted and ratoon sugarcane to evaluate the effectiveness of FT, together with amendment with biochar and nitrogen fertilization, to improve sugarcane yield. The treatments were conventional tillage with chemical fertilizer without biochar (CT-CF, which was the control of this experiment), FT with chemical fertilizer without biochar (FT-CF), conventional tillage with chemical fertilizer mixed with biochar (CT-CFB), and FT with chemical fertilizer mixed with biochar (FT-CFB). FT-CFB treatment presented higher soil porosity, as well as higher contents of available N, P, K, total N, and organic matter, and lower soil bulk density. Similarly, results showed that FT-CFB presented higher sugarcane root fresh and dry weights, higher germination percentage, higher tiller number, and higher yield with statistically significant differences among treatments for both newly planted and ratoon sugarcane plants. Significant interactions between biochar and FT were observed for these crop traits. The interactions of FT and amendment with biochar improved the soil’s physical and chemical properties and increased the available nutrients, resulting in improved root growth and sugarcane yield. The statistical results of the present study imply that Fenlong-ridging deep tillage combined with chemical fertilizer mixed with biochar (FT-CFB) application is a new promising farm management practice for improving the soil’s physical and chemical properties and root growth, increasing total yield in China’s sugarcane belt area.

Background and objectiveInsulin-like growth factor-1 (IGF-1) regulates cardiomyocyte survival, hypertrophy, and ageing. We aimed to investigate the potential correlation between IGF-1 and interventricular septal (IVS) thickening.MethodsMedical record data were obtained from patients hospitalized between May 1, 2012 and April 30, 2022. All patients underwent echocardiography and had laboratory data on plasma IGF-1. We analyzed the relationship between IGF-1 levels and IVS thickening based on logistic regression models.ResultsPropensity score matching at 1:4 ratio was performed for 180 patients with IVS thickening and 1,964 patients without IVS thickening. Finally, 180 (case group) and 623 (control group) patients were enrolled. Of the total 803 patients, the mean age was 59 years, and 59.7% were male. In multivariate-adjusted models that adjusted for propensity scores, the risk of IVS thickening increased with increasing IGF-1 levels; specifically, the risk of IVS thickening increased per 1 ng/mL [odds ratio (OR) 1.003; 95% confidence interval (CI): 1.002–1.004; P < 0.001], per 5 ng/mL (OR, 1.016; 95% CI, 1.010–1.022; P < 0.001), and per 10 ng/mL(OR, 1.032; 95% CI, 1.019–1.045; P < 0.001) increase in IGF-1 levels. When the IGF-1 levels were expressed as a categorical variable, the increased levels of IGF-1 led to an increased risk of IVS thickening; specifically, the OR of IVS thickening for T3 >152.00 ng/mL was 2.020 (95% CI: 1.310–3.115, P < 0.01) compared with T1 164.00 ng/mL, 45–60 years when IGF-1 value > 140.34 ng/mL and ≥ 60 years when IGF-1 value >108.20 ng/mL. In female patients aged 45–60 years, the risk of IVS thickening increased when the IGF-1 levels were >207.45 ng/mL. However, IGF-1 was not significantly correlated with IVS thickening in female patients aged 18–45 and ≥60 years. Sensitivity analysis by excluding those with acromegaly did not change the relationship between IGF-1 and the risk of IVS thickening.ConclusionThe plasma IGF-1 levels were related to the risk of IVS thickening irrespective of blood pressure.

Abstract Background The angiotensin-converting enzyme 2 (ACE2)/angiotensin 1–7 (Ang-(1–7)) axis has been shown to protect against the age-associated decline in skeletal muscle function. Here, we investigated the protective effects of ACE2 in mitigating the age-associated decline of skeletal muscle function and to identify the potential underlying molecular mechanisms. Methods We measured the expression levels of Ang-(1–7) in C57BL/6J mice of different ages and correlated these levels with measures of skeletal muscle function. We also investigated the expression of myocyte enhancer factor 2 A (MEF2A) in ACE2 knockout (ACE2KO) mice and its relationship with muscle function. We then treated aged ACE2KO mice for four weeks with Ang-(1–7) and characterized the levels of MEF2A and skeletal muscle function before and after treatment. We assessed the impact of Ang-(1–7) on the growth and differentiation of C2C12 cells in vitro and assessed changes in expression of the glucose transporter type 4 (Glut4). Results Aged mice showed reduced skeletal muscle function and levels of Ang-(1–7) expression in comparison to young and middle-aged mice. In ACE2KO mice, skeletal muscle function and MEF2A protein expression were significantly lower than in age-matched wild-type (WT) mice. After one month of Ang-(1–7) treatment, skeletal muscle function in the aged ACE2KO mice improved, while MEF2A protein expression was similar to that in the untreated group. In C2C12 cells, Ang-(1–7) was shown to promote along with the upregulated expression of Glut4. Conclusions The ACE2/ Ang-(1–7) axis has a protective function in skeletal muscle and administration of exogenous Ang-(1–7) can delay the age-related decline in the function of skeletal muscle.

The protoplast experimental system eis an effective tool for functional genomics and cell fusion breeding. However, the physiological and molecular mechanisms of protoplast response to enzymolysis are not clear, which has become a major obstacle to protoplast regeneration. Here, we used physiological, cytological, proteomics and gene expression analysis to compare the young leaves of sugarcane and enzymolized protoplasts. After enzymatic digestion, we obtained protoplasts with viability of > 90%. Meanwhile, the content of malondialdehyde, an oxidation product, increased in the protoplasts following enzymolysis, and the activity of antioxidant enzymes, such as peroxidase (POD), catalase (CAT), acid peroxidase (APX), and O2-, significantly decreased. Cytologic analysis results showed that, post enzymolysis, the cell membranes were perforated to different degrees, the nuclear activity was weakened, the nucleolus structure was not obvious, and the microtubules depolymerized and formed several short rod-like structures in protoplasts. In this study, a proteomics approaches was used to identify proteins of protoplasts in response to the enzymatic digestion process. GO, KEGG, and KOG enrichment analyses revealed that the abundant proteins were mainly involved in bioenergetic metabolism, cellular processes, osmotic stress, and redox homeostasis of protoplasts, which allow for protein biosynthesis or degradation. RT-qPCR analysis revealed that the expression of osmotic stress resistance genes, such as DREB, WRKY, MAPK4, and NAC, was upregulated, while that of key regeneration genes, such as CyclinD3, CyclinA, CyclinB, Cdc2, PSK, CESA, and GAUT, was significantly downregulated in the protoplasts. Hierarchical clustering and identification of redox proteins and oxidation products showed that these proteins were involved in dynamic networks in response to oxidative stress after enzymolysis. Our findings can facilitate the development of a standard system to produce regenerated protoplasts using molecular markers and antibody detection of enzymolysis.

Background: Phelan–McDermid syndrome (PMS), caused by deletions at 22q13.3 and pathogenic variants in the SHANK3 gene, is a rare developmental disorder characterized by hypotonia, developmental delay (DD), intellectual disability (ID), autism spectrum disorder (ASD), dysmorphic features, absence of or delayed language, and other features.Methods: Conventional karyotyping, chromosomal microarray analysis (CMA), and whole exome sequencing (WES) have been used to detect genetic defects causing PMS. We summarized the genetic and clinical findings from prenatal to postnatal stages of detected cases of PMS and mapped potential candidate haploinsufficient genes for deletions of 22q13. This study aimed to summarize the laboratory findings, genetic defects, and genotype–phenotype correlations for Chinese patients with PMS.Results: Seven prenatal cases and fourteen postnatal cases were diagnosed with PMS in our center. Thirteen cases had a deletion ranging in size from 69 to 9.06 Mb at 22q13.2-q13.33, and five cases had a pathogenic variant or an intragenic deletion in the SHANK3 gene. Three familial cases with a parental carrier of a balanced translocation were noted. A review of the literature noted another case series of 29 cases and a report of five cases of PMS in China. Genotype–phenotype correlations confirmed haploinsufficiency of the SHANK3 gene for PMS and suggested other candidate haploinsufficient genes TNFRSFI3C and NFAM1 genes for immunological features and TCF20, SULT4A1, PARVB, SCO2, and UPK3A genes for intellectual impairment and behavioral abnormality, neurological features, macrocephaly/hypotonia, oculopathy, and renal adysplasia, respectively.Conclusion: Indications for prenatal diagnosis of PMS are not specific, and approximately 85% prenatally diagnosed PMS elected termination of pregnancies after genetic counseling. For postnatal cases, 62.5% were caused by a deletion at 22q13 and 37.5% were caused by a pathogenic variant or an intragenic deletion in the SHANK3 gene. Approximately 6.7% of cases with a deletion were familial, and almost all pathogenic variants were de novo. Combined karyotype, CMA, and WES should be performed to increase the diagnostic yield. The identification of other candidate haploinsufficient genes in deletions of 22q13.2-q13.33 could relate to more severe dysmorphic features, neurologic defects, and immune deficiency. These results provided evidence for diagnostic interpretation, genetic counseling, and clinical management for the Chinese cases of PMS.

Fenlong-ridging (FL) is a new type of conservation tillage. In many crops, FL increases crop yield and quality; however, the cytology and molecular mechanisms of crops under FL is not completely understood. This study investigated soil physical and chemical properties under FL and conventional tillage (CK) during 2018–2019 (plant cane) and 2019–2022 (first stubble), and analyzed the agronomic trait, physiology, leaf anatomical structure, and gene expression related to photosynthesis between FL and CK of sugarcane (Guitang 42). Soil bulk density significantly increased, and soil porosity, water storage, and content of available nitrogen and phosphorus under FL were significantly higher than those under CK. Plant height, stem diameter, single stem weight, effective stem number and yield significantly increased under FL compared to under CK. Sugar content significantly increased in plant cane under FL. Chlorophyll content and the photosynthetic rate increased, with significantly higher activity of photosynthetic enzymes including NADP-malate dehydrogenase (NADP-MDH), phosphoenolpyruvate carboxylase (PEPC), and ribulose-1,5-bisphosphate carboxylase (RuBPC) under FL compared to CK. Fenlong-ridging cytology results showed that the mesophyll cells were large and arranged well, the Kranz anatomy was noticeable, and there were a high number of large chloroplasts in mesophyll cell and in the vascular bundle sheath. Furthermore, the bundle sheath in FL was larger than that in CK. Transcriptomics results showed that 19,357 differentially genes (DEGs) were up-regulated and 28,349 DEGs were down-regulated in sugarcane leaves under FL vs. CK. The Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis revealed that abundant DEGs were enriched in photosynthesis, photosynthesis-antenna protein, carotenoid biosynthesis, and other pathways associated with photosynthesis. Most expression was up-regulated, thus, facilitating photosynthesis regulation. Quantitative real-time polymerase chain reaction analysis revealed the up-regulation of genes related to photosynthesis (PsaH and PsbS) under FL. Overall, this study provides insights into the role of FL in increased sugarcane yield by integrating physiology, cytology, and proteomics analysis. These findings could be used to further improve its application and promotion.

To ensure the high purity and biological activity of the adenovirus vector to be used for clinical applications, a stable and linearly scalable preparation method is highly imperative. During the adenovirus-harvesting process, the Triton X-100-based lysis method possesses the advantages of higher efficiency as well as easier linearization and amplification. Most Triton X-100 can be removed from the adenovirus sample by chromatographic purification. However, there is no report that a small amount of residual Triton X-100, present in adenovirus sample, can affect the particle integrity, infectivity, and structure of adenoviruses. Here, we found that although residual Triton X-100 affected the short-term stability, purity, infectivity, and structure of adenoviruses at 37°C, it did not hamper these properties of adenoviruses at 4°C. This study suggests that although the Triton X-100-based lysis method is a simple, efficient, and easy-to-scale process for lysing host cells to release the adenovirus, the storage conditions of adenovirus products must be taken into consideration.

This study developed an experimental system based on Joule heat of sliding-pressure additive manufacturing (SP-JHAM), and Joule heat was used for the first time to accomplish high-quality single-layer printing. The roller wire substrate is short-circuited, and Joule heat is generated to melt the wire when the current passes through. Through the self-lapping experimental platform, single-factor experiments were designed to study the effects of power supply current, electrode pressure, contact length on the surface morphology and cross-section geometric characteristics of the single-pass printing layer. Through the Taguchi method, the effect of various factors was analyzed, the optimal process parameters were obtained, and the quality was detected. The results show that with the current increase, the aspect ratio and dilution rate of a printing layer increase within a given range of process parameters. In addition, with the increase in pressure and contact length, the aspect ratio and dilution ratio decrease. Pressure has the greatest effect on the aspect ratio and dilution ratio, followed by current and contact length. When a current of 260 A, a pressure of 0.60 N and a contact length of 1.3 mm are applied, a single track with a good appearance, whose surface roughness Ra is 3.896 μm, can be printed. Additionally, the wire and the substrate are completely metallurgically bonded with this condition. There are also no defects such as air holes and cracks. This study verified the feasibility of SP-JHAM as a new additive manufacturing strategy with high quality and low cost, and provided a reference for developing additive manufacturing technology based on Joule heat.

Background. Melanomas, the most common human malignancy, are primarily diagnosed visually, beginning with an initial clinical screening and followed potentially by dermoscopic analysis, a biopsy, and histopathological examination. We aimed to systematically review the performance and quality of machine learning-based methods in distinguishing melanoma and benign nevus in the relevant literature. Method. Four databases (Web of Science, PubMed, Embase, and the Cochrane library) were searched to retrieve the relevant studies published until March 26, 2022. The Predictive model Deviation Risk Assessment tool (PROBAST) was used to assess the deviation risk of opposing law. Result. This systematic review included thirty researches with 114007 subjects and 71 machine learning models. The convolutional neural network was the main machine learning method. The pooled sensitivity was 85% (95% CI 82–87%), the specificity was 86% (82–88%), and the C-index was 0.87 (0.84–0.90). Conclusion. The findings of our study showed that ML algorithms had high sensitivity and specificity for distinguishing between melanoma and benign nevi. This suggests that state-of-the-art ML-based algorithms for distinguishing melanoma from benign nevi may be ready for clinical use. However, a large proportion of the earlier published studies had methodological flaws, such as lack of external validation and lack of clinician comparisons. The results of these studies should be interpreted with caution.

Lodging is one of the major constraints in attaining high yield in crop production. Major factors associated with stalk lodging involve morphological traits and anatomical features along with the chemical composition of the stem. However, little relevant research has been carried out in sorghum, particularly on the anatomical aspects. In this study, with a high-throughput procedure newly developed by our research group, the nine parameters related to stem regions and vascular bundles were generated in 58 sorghum germplasm accessions grown in two successive seasons. Correlation analysis and principal component analysis were conducted to investigate the relationship between anatomical aspects and stalk mechanical traits (breaking force, stalk strength and lodging index). It was found that most vascular parameters were positively associated with breaking force and lodging index with the correlation coefficient r varying from −0.46 to 0.64, whereas stalk strength was only associated with rind area with the r = 0.38. The germplasm resources can be divided into two contrasting categories (classes I with 23 accessions and II with 30 accessions). Compared to class II, the class I was characterized by a larger number (+40.7%) and bigger vascular bundle (+30%), thicker stem (+19.6%) and thicker rind (+36.0%) but shorter internode (plant) (−91.0%). This study provides the methodology and information for the studies of the stem anatomical parameters in crops and facilitates the selective breeding of sorghum.

Fairy rings are a unique ecological phenomenon caused by the growth of the fungal mycelium in the soil. Fairy rings formed by Leucocalocybe mongolica (LM) are generally distributed in the Mongolian Plateau, where they promote plant growth without fertilization and alleviate fertilizer use. We previously investigated the soil factors regulating growth promotion in a fairy ring ecosystem; however, the aspects of the plant (Leymus chinensis, LC) that promote growth have not been explored. Therefore, the present study investigated the endophyte diversity and metabolome of LC in an LM fairy ring ecosystem. We analyzed the leaf and root samples of LC from the DARK (FR) and OUT (CK) zones. The fairy rings significantly improved the fungal diversity of roots and leaves and the bacterial diversity of leaves in the FR zone. Ralstonia was the dominant bacteria detected in the LC leaves. In addition, Marasmius, another fairy ring fungal genus, was also detected with a high abundance in the roots of the FR zone. Furthermore, widely targeted metabolome analysis combined with KEGG annotation identified 1011 novel metabolites from the leaves and roots of LC and seven pathways significantly regulated by the fairy ring in the FR zone. The fairy ring ecosystem significantly downregulated the flavonoid metabolism in the leaves and roots of LC. The correlation analysis found Ralstonia is a potential regulatory factor of flavonoid biosynthesis in LC. In addition, salicylic acid and jasmonic acid were found upregulated in the leaves, probably related to Marasmius enrichment. Thus, the study details plant factors associated with enhanced growth in an LM fairy ring ecosystem. These findings lay a theoretical foundation for developing the fairy ring ecosystem in an agricultural system.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused the coronavirus disease 2019 (COVID-19) and has become the world's most pressing public health threat. Although not as common as respiratory symptoms, a substantial proportion of patients with COVID-19 presented the gastrointestinal symptoms. ACE2, as the receptor of SARS-CoV and SARS-CoV-2, is highly expressed in the epithelia of the epithelium cells in lung and intestine. In addition, ACE2 is essential for the innate immunity, amino acid transportation and the homeostasis of intestinal microecology. The composition of gut microbiota in COVID-19 patients was altered and concordant with inflammatory, which may explain the gastrointestinal symptoms in patients. Here we reviewed and discussed the evolving role for ACE2 and gut microbiota in SARS-CoV-2 infection which might provide innovative approaches to targeting ACE2 and gut microbiota for the COVID-19 therapy.

Sugarcane somatic cell hybridization can break through the barrier of genetic incompatibility between distantly related species in traditional breeding. However, the molecular mechanisms of sugarcane protoplast regeneration and the conditions for protoplast preparation remain largely unknown. In this study, young sugarcane (ROC22) leaves were enzymatically digested, and the viability of protoplasts reached more than 90% after enzymatic digestion (Enzymatic combination: 2% cellulase + 0.5% pectinase + 0.1% dissociative enzyme + 0.3% hemicellulase, pH = 5.8). Transcriptome sequencing was performed on young sugarcane leaves and protoplasts after enzymatic digestion to analyze the differences in gene expression in somatic cells before and after enzymatic digestion. A total of 117,411 unigenes and 43,460 differentially expressed genes were obtained, of which 21,123 were up-regulated and 22,337 down-regulated. The GO terms for the 43,460 differentially expressed genes (DEGs) were classified into three main categories: biological process, cellular component and molecular function, which related to developmental process, growth, cell proliferation, transcription regulator activity, signal transducer activity, antioxidant activity, oxidative stress, kinase activity, cell cycle, cell differentiation, plant hormone signal transduction, and so on. After enzymatic digestion of young sugarcane leaves, the expressions of GAUT, CESA, PSK, CyclinB, CyclinA, CyclinD3 and cdc2 genes associated with plant regeneration were significantly down-regulated to 65%, 47%, 2%, 18.60%, 21.32%, 52% and 45% of young leaves, respectively. After enzymatic digestion, Aux/IAA expression was up-regulated compared with young leaves, and Aux/IAA expression was 3.53 times higher than that of young leaves. Compared with young leaves, these key genes were significantly changed after enzymatic digestion. These results indicate that the process of somatic enzymatic digestion process may affect the regeneration of heterozygous cells to a certain extent.

Deep-learning-based, cross-age face recognition has improved significantly in recent years. However, when using the discriminative method, it is still challenging to extract robust age-invariant features that can reduce the interference caused by age. In this paper, we propose a novel, effective, attention-based feature decomposition model, the age-invariant features extraction network, which can learn more discriminative feature representations and reduce the disturbance caused by aging. Our method uses an efficient channel attention block-based feature decomposition module to extract age-independent identity features from facial representations. Our end-to-end framework learns the age-invariant features directly, which is more convenient and can greatly reduce training complexity compared with existing multi-stage training methods. In addition, we propose a direct sum loss function to reduce the interference of age-related features. Our method achieves a comparable and stable performance. Experimental results demonstrate superior performance on four benchmarked datasets over the state-of-the-art. We obtain the relative improvements of 0.06%, 0.2%, and 2.2% on the cross-age datasets CACD-VS, AgeDB, and CALFW, respectively, and a relative 0.03% improvement on a general dataset LFW.

Fenlong-ridging (FL) is a recently proposed conservation tillage technology which has dramatic differences to traditional ones. Previous studies have demonstrated in many crops that FL has yield-increasing effects without additional inputs. However, little is known about the role that microbes play in mediating the growth-promoting effects of FL, which restricts its further application and improvement. Here, we characterized variation in the soil and root microbial diversity of sugarcane (GT44) under FL and traditional turn-over plough tillage (CK) by conducting 16S rRNA and ITS metabarcoding surveys. We also measured several phenotypic traits to determine sugarcane yields and analyzed the chemical properties of soil. We found that: (i) plant height (PH) and total biomass weight (TW) of sugarcane plants were 9.1% and 21.7% greater under FL than those under CK, indicating\increased biomass yield of the sugarcane in FL operation; (ii) contents of organic matter, total nitrogen, available phosphorus, and available potassium were lower in soil under FL than those under CK, which indicates the utilization of soil nutrients was greater in FL soil; (iii) FL promoted the activity of endophytic microbes in the roots, and these diverse microbial taxa might have an effect on sugarcane yield and soil chemical properties; and (iv) Sphingomonas, Rhizobium, and Paraburkholderia and Talaromyces, Didymella, and Fusarium were the top three most abundant genera of bacteria and fungi, respectively, in soil and root samples. In addition, strains from Rhizobium and Talaromyces were isolated to verify the results of the metabarcoding survey. Overall, our study provides new insights into the role of microbes in mediating the growth-promoting effects of FL. These findings could be used to further improve applications of this novel conservation tillage technology.

In recent years, scholars have proposed a metal wire forming method based on the Joule heat principle in order to improve the accuracy of additive manufacturing and reduce energy consumption and cost, but it is still in the theoretical stage. In this paper, a mathematical model of resistance additive manufacturing was established using finite element software, and the temperature variation of the melting process under different currents was analyzed. A suitable current range was preliminarily selected, and an experimental system was built. Through experimental study of the current and wire feeding speed, the influences of different process parameters on the forming appearance of the coating were analyzed. The results showed that the forming appearance was the best for Ti-6Al-4V titanium alloy wire with a diameter of 0.8 mm, when the current was 160 A, the voltage was 10 V, the wire feeding speed was 2.4 m/min, the workbench moving speed was 5 mm/s, and the gas flow rate was 0.7 m3/h. Finally, the process parameters were used for continuous single-channel multilayer printing, verified the feasibility of the process at the experimental level and provided reference data for the subsequent development of this technology.

Drug delivery is essential for effective therapy. Implantable controlled drug delivery systems (ICDDS) have become a research focus due to the associated advantages of continuous, long-duration, sustained delivery and the reduced side effects associated with this form of drug release. ICDDS can be designed to release drugs in accordance with different demands. Zero-order drug release, which involves a theoretically linear relationship between the cumulative amount of released drug and the amount of time taken for the release, is an important drug release rule for the control of chronic diseases. However, the release process is influenced by various parameters and zero-order drug release is therefore difficult to achieve. In this study, to achieve zero-order drug release, an implantable controlled drug delivery system with micro-hole (ICDDSM) was designed and fabricated with degradable polymer. Based on micro electromechanical systems (MEMS) technology, ICDDSM matrixes with circular and honeycomb structures were fabricated using the hot-press shaping method at a temperature of 75 °C, 30 N of force which were maintained for 150 s. A micro-hole with a diameter 100 μ m was formed using a femto-second laser. 5-fluorouracil was loaded into the ICDDSM and in-vitro experiments were conducted in 37 °C normal saline solution. The experimental results showed that the 20 mg of loaded 5-fluorouracil was released in 720 h with the relationship between the cumulative amount of released drug and the drug release time tending predominantly toward linearity. Zero-order drug release was thus achieved. This study proposed a new degradable ICDDS structure to achieve zero-order drug release, and the hot-press shaping process proved feasible for the efficient fabrication of the lower-cost polymer structure.

This paper presents a systematic numerical investigation of the transient transport phenomenon during the pileup of molten metal droplets on the substrate. The physical mechanisms of the pileup process, including the bulk liquid, capillarity effects at the liquid-solid interface, heat transfer, and solidification, are identified and quantified numerically. The droplet diameter is 100 μm, and the impact velocities are 1–3 m/s. These conditions correspond to Re = O(100), We = O(1). The initial substrate temperature is 350 K. The initial droplet temperature of aluminum alloy molten droplets is 960 K. The numerical models are validated with experiments. The comparison between numerical simulations and experimental findings shows a good agreement. The effects of impacting velocity and relative distances between two successive molten droplets on the end-shapes of impact regime are examined. This investigation is essential to implement effective process control in metal microdroplet deposition manufacture.
