This study aimed to examine the impact of olive leaf extract (OL-E) and olive leaf extract coating chitosan (OLCh) on Bacillus cereus and its toxin genes, as well as, the sensory qualities of refrigerated beef burgers. The primary phenolic components of olive leaf extract were oleeuropein and protocatechuic acid, identified by HPLC-DAD analysis. The concentrations of OL-E and OL-Ch that were in vitro effective against B. cereus were 62.5 mg/mL and 125 mg/mL, respectively. In burgers treated with OL-E and OL-Ch, viable B. cereus dramatically decreased (p < 0.05) over time during cold storage. The count decreased to 2 log CFU/g in the OL-E group and 1.3 log CFU/g in the OL-Ch group after thirteen days of preservation. The entFM gene expression levels on day thirteen were 0.09 in the OL-Ch group and 0.2 in the OL-E group. Conversely, the cytK gene expression levels in the OL-E and OL-Ch groups were 0.09 and 0.15, respectively. Both supplements could control physicochemical characteristics (pH, volatile basic nitrogen (TVBN), and thiobarbituric acid (TBA)) to be 6.12±1.55, 16.1±2.98, and 0.869±0.28, respectively, in the OL-E group during a storage period. Moreover, they were 5.79, 14.3±2.79, and 0.816±0.31 in the OL-Ch group. They also did not appear to affect the average sensory score of burgers. Gumboro, or infectious bursal disease (IBD), represents a significant threat to poultry worldwide due to its genetic and pathogenic variations. Viruses can mutate and produce new variants, which can have an impact on the epidemiological situation. The study will determine and assess the genetics, protein sequence, and pathogenicity of IBDV present in broiler flocks in different locations within Ismalia and Dakahlia Governorates, Egypt, in 2022. A total of 150 bursa samples were collected from 15 broiler flocks exhibiting symptoms indicative of IBDV infection. The VP2 (viral protein2) gene was partially detected and then subjected to complete gene sequencing in order to summarize the key genetic mutations found and histopathological examination. The results demonstrated that 12 out of 15 farms were infected with IBDV. Genetic analysis revealed that 10 of the 12 identified strains belonged to genogroup 3, which was similar to previous Egyptian strains. The remaining two strains were of genus 1. The VP2 gene's hypervariable region displayed amino acid alterations at several positions. Classic IBDV strains (genogroup 2) showed similar amino acid profiles. Amino acid alterations were identified in the VP2 gene, indicating that classic IBDV strains may cause damage to the bursa of Fabricius, leading to immunosuppression. In Egypt, vvIBDV (very virulent infectious bursal disease virus) strains have led to recurring outbreaks and high chick mortality rates. These findings are anticipated to contribute to the latest features of IBDV in the examined regions. Therefore, it is recommended that IBDV immunization programs in Egypt, where live attenuated virus vaccinations are common, be reevaluated. Antimicrobial resistance is considered a major threat facing humanity. It doesn’t only affect public health, but also causes great losses in the dairy industry. Mastitis is a major threat to the dairy industry. The aim of this study was to monitor the antimicrobial resistance of Escherichia coli (E. coli) collected from raw milk of both healthy and mastitis-infected cows and buffaloes in Egypt. In total, 450 milk samples were collected and examined in the period from 2018 to 2021. The samples were collected from healthy cows and buffaloes (30, 58), suffering from clinical mastitis (139, 223), respectively. E. coli was isolated from 33 mastitis milk samples (9.1%) and from 3 (3.4%) normal milk samples. The antibiotic susceptibility testing was performed using the disc diffusion method (Kirby-Bauer method). E. coli isolated from mastitis milk samples showed resistance The Extended Spectrum b-Lactamases test (ESBL) performed on the E. coli isolates showed positive results in 9% of mastitis milk samples, but no results in normal milk samples. Out of 36 E. coli isolates, 34 possessed the ampC gene, but blaTEM and blaSHV were detected in 5 isolates with percentages of 94%, 1.4%, and 1.4%, respectively, while BlaIPM and Sul1 were found in one isolate (2.7%). In dairy industry, bovine mastitis is the most prevalent disease, which reduces milk production and causes economic losses. This study was conducted to estimate the prevalence of Mycoplasma bovis and some bacteria causing mastitis in dairy farms and partial sequencing of 16SrRNA target genes and Quinolones Resistance Determining Regions (QRDRs) (gyrA and parC) in M. bovis isolates. 370 milk samples were obtained from farms located in villages in Fayoum governorate, Egypt. The examined milk samples (8,91%) were positive for the California mastitis test (CMT). Multiplex RTPCR was used for the recognition of microorganisms causing mastitis (Staphylococcus (S.) aureus, Streptococcus species (spp.), Escherichia (E.) coli, and Mycoplasma (M.) bovis) from mastitic milk. The results revealed that E. coli was the most predominant (84.8%) followed by S. aureus (81.8%) while M. bovis was the lowest one (51.5%). Mixed infection with two or more mastitic bacterial agents was also identified. All 33 examined mastitic milk samples were diagnosed with mixed infection with E. coli, S. aureus, Streptococcus spp. and M. bovis (36.36%), E. coli and S. aureus (21.21%), and rephrase E. coli, M. bovis, and Streptococcus spp. (6.06%). The sequence analysis of M. bovis 16SrRNA genes illustrated a high similarity of examined isolates to strains previously deposited in the GenBank recovered from the same locality. The gyrA amino acids showed no substitution but showed 100% similarity with M. bovis isolates worldwide. However, the amino acid sequence of parC, showed substitution at positions 2 (Gln to Arg) (CAG >>CGT), 75 (Ile to Ser) (ATT>>AGC), and 79 (Asn to Asp) (AAC>>GAT). Sequence results can lead to the creation of appropriate treatment and control measures for M. bovis, while multiplex RT-PCR, can be exploited as a standard diagnostic method for major mastitis pathogens. Avian pox is a disease that has devastating impacts on both poultry and wild bird species. Avian pox is caused by various strains of avipoxviruses (APV). Nevertheless, the virus has been detected in pigeons and chickens that are raised in backyard areas, leading to substantial financial damage for small-scale producers. There is a lack of comprehensive information regarding the transmission of APV among birds in the backyards and residential areas. Hence, the present investigation closely monitored and observed APV in domesticated birds residing in backyard areas, with the aim of impeding the transmission of the virus to nearby poultry farms. In 2023, a total of fifty backyard flocks were surveyed for the presence of avian pox disease. Sixteen backyards (14 pigeons and 2 chickens) exhibited warty nodular lesions on their heads and nonfeathered body parts. APV was confirmed in nodular lesions by polymerase chain reaction (PCR) amplification and genetic sequencing. All samples from the lesions showed successful amplification of the p4b locus (core protein p4b). Four confirmed samples were tested for pathogenicity on the chicken embryo chorioallantoic membrane (CAM). Histopathological examination revealed ballooning degeneration and numerous intracytoplasmic inclusion bodies (Bollinger bodies) in the ectoderm of the infected CAM. Phylogenetic analysis revealed that the strains clustered into main clade A, with 11 in subclade A2 and 5 in subclade A1. Amino acid identity showed 100% similarity between the vaccine (fowlpox/VSVRI/Egypt) and some detected strains (PP537574 and PP537575). In addition, the PP537576.1 to PP537580.1 and PP537582.1 to PP537585.1 had 2-point mutations compared to the fowlpox/VSVRI/Egypt vaccine. The overall finding of low biosecurity levels in the investigated backyard birds emphasizes the significance of establishing sanitary measures and control vectors to reduce virus transmission routes and disease severity. In conclusion, it is necessary to emphasize the tracking of APV in backyard birds. Concurrently, we advised enhancing hygiene protocols, vector management, and subsequent vaccination to restrict the occurrence of APV outbreaks and prevent their transmission to neighboring poultry farms. Coagulase-positive (CPS) and coagulase-negative (CNS) staphylococci cause staphylococcal food poisoning. Recently, CPS and CNS have received increasing attention due to their potential role in the dissemination of antibiotic resistance markers. The present study aimed to evaluate CPS and CNS species distribution and their antibiotic resistance profile isolated from chicken and beef meat. Fifty fresh, uncooked chicken parts and 50 beef meat cuts (local n=27; imported n=23) were used. One hundred staphylococcal isolates belonging to 11 species were isolated and identified from chicken (n=50) and beef (n=50) raw meat samples. Staphylococcus hyicus (26/100), lugdunensis (18/100), aureus (15/100) and epidermidis (14/100) were dominant. S. aureus was 100% resistant to penicillin and sulfamethoxazole/trimethoprim. Vancomycin-resistant S. aureus showed intermediate resistance (51%), which might indicate the dissemination of vancomycin resistance in the community and imply food safety hazards. The percentage of resistance to β-lactams was variable, with the highest resistance being to penicillin (94%) and lowest to ampicillin-sulbactam (22%). Antimicrobial resistance was mainly against penicillin (94%), clindamycin (90%) and sulfamethoxazole/trimethoprim (82%). The results indicate that chicken and beef raw meat are an important source of antibiotic-resistant CPS and CNS.
