ABSTRACT Royal jelly renowned for its robust nutritional, functional, and biological properties, is a pivotal product derived from honeybees. The purpose of this investigation was to assess the theory that in ovo injection of freeze‐dried royal jelly (FDRJ) solutions at varying concentrations can influence the hatchability, blood properties and hatching characteristics of day‐old chicks. A total of 480 eggs (54.81 ± 0.187 g) were allocated into four experimental groups: negative control (NC), without injection, positive control (PC), administered with regular saline, a low FDRJ dose group (9 mg/egg), and a high FDRJ dose group (18 mg/egg). The in ovo injections were administered on Day 18 of incubation, and the experiment was subsequently continued until the incubation period concluded at 21 days. Results revealed that the lower FDRJ dose (9 mg/egg) significantly improved hatchability percentages compared to other treatments. Conversely, the higher FDRJ dose (18 mg/egg) and control groups (NC and PC) resulted in significantly higher chick yield percentages than the lower FDRJ and PC groups. The NC group showed the supreme yolk sac (YS) percentage, whereas the yolk‐free body mass (YFBM) percentages displayed an inverse trend. Furthermore, the in ovo FDRJ injection did not affect haematological values or the relative organ weight of day‐old chicks. In conclusion, in ovo FDRJ injection demonstrated beneficial effects on hatchability and chick weight, as evidenced by the studied parameters. [ABSTRACT FROM AUTHOR]

The complex pathogenesis of and the implication of a vast number of components in the inflammasome-activation pathway prompted us to search for compounds that have a wide therapeutic index and act at the level of multiple cellular targets. Although CRID3 blocks NLRP3 with high specificity in the laboratory, clinical trials of the compound reported weaker potency.Methods: We used NSC328382, a P2X7R antagonist, as an adjunctive therapy and generated a strategy to potentiate the effects of CRID3 in rats with DSS-induced colitis.Results: NSC328382/CRID3 combined therapy exhibited a significantly increased efficacy compared with either of the monotherapies. NSC328382/CRID3 was more efficient in 1) attenuating colon shortening and disease activity; 2) improving goblet cell density and both the macroscopic and microscopic scenario of the injured colon; 3) improving the antioxidant defense mechanisms of the inflamed colon against oxidative stress; and 4) mitigating the inflammation state by downregulating the proinflammatory cytokines. Pyroptotic cell death was also conspicuously restrained. Additionally, NSC328382 interrupted the MyD88/NF-κB axis. Moreover, NSC328382/CRID3 exhibited the ability to alter Th1/Th2 dominance.Conclusion: The clinical application of NSC328382/CRID3 may result in the generation of a novel approach for the treatment of IBDs.Keywords: NSC328382, CRID3, P2X7R/NLRP3, MyD88/NF-κB, Colitis

16. Effect of Adding the Antimicrobial L-Carnitine to Growing Rabbits' Drinking Water on Growth Efficiency, Hematological, Biochemical, and Carcass Aspects.

The current study was designed to assess the impact of L-carnitine (LC) supplementation in the drinking water of growing Alexandria-line rabbits on performance and physiological parameters. Two hundred eighty-eight 35-day-old rabbits were divided into four groups of twenty-four replicates each (seventy-two rabbits/treatment). The treatment groups were a control group without LC and three groups receiving 0.5, 1, and 1.5 g/L LC in the drinking water intermittently. The results showed that the group receiving 0.5 g LC/L exhibited significant improvements in final body weight, body weight gain, feed conversion ratio, and performance index compared to the other groups. The feed intake remained unaffected except for the 1.5 g LC/L group, which had significantly decreased intake. Hematological parameters improved in all supplemented groups. Compared with those in the control group, the 0.5 g LC/L group showed significant increases in serum total protein and high-density lipoprotein, along with decreased cholesterol and low-density lipoprotein. Compared to other supplemented groups, this group also demonstrated superior carcass traits (carcass, dressing, giblets, and percentage of nonedible parts). In conclusion, intermittent supplementation of LC in the drinking water, particularly at 0.5 g/L twice a week, positively influenced the productivity, hematology, serum lipid profile, and carcass traits of Alexandria-line growing rabbits at 84 days of age. So far c.1331+1G>A (IVS4+1G>A); NM_001038603.3, variant in deafness, has only reported previously in one Pakistani family in 2008 and it is reported for the first time in Iran and second time in the world. The case is a 21-year-old Iranian woman who has NSHL referred for genetic consultation, and her parents had a consanguineous marriage. To study the responsible genes for the mentioned disorder, whole exome sequencing (WES) was performed for the case. The result of WES analysis revealed a transition at the splice donor variant site of the MARVELD2 gene. 25. Impact of increasing dietary oil concentrations with a constant energy level on the tolerance of broiler chickens to a high ambient temperature

Broiler males (n= 140) were used in a straight-run experimental design and distributed randomly among four treatment groups with seven replicates per treatment and five broilers per replicate. During 21-42 d old, the chickens were fed iso-caloric and iso-nitrogenous diets containing four levels of dietary vegetable oils (DVO), of 2.7, 4, 6 and 8%. During d 25-27, 31-33, and 38-40 of age, broilers were exposed to heat stress for 4 h a day (1000-1400 h) at 34 ºC, 70-75 % relative humidity. Feeding an 8% DVO diet significantly increased body weight gain compared to the other DVO levels. The feed conversion ratio, protein conversion ratio, metabolizable energy conversion ratio and European production index were significantly enhanced due to feeding an 8% DVO diet compared to a diet containing 6% DVO. Feeding 8% DVO significantly increased the meat protein and lipid percentages, compared to the control group (2.7 % DVO), but decreased the plasma low-density lipoprotein, very-low-density lipoprotein and lymphocytes. Feeding 8% DVO significantly increased the mean cell volume and mean cell hemoglobin, and bursa weight and percentage compared to the control. In addition, 6 and 8% DVO significantly increased the plasma total antioxidant capacity compared to the control group, but decreased the malondialdehyde. Thus, broilers fed a diet containing 8% DVO have an increased tolerance to heat stress, as evidenced by increasing the productive performance, meat quality, blood hematological and biochemical traits, antioxidants and immunity.

This work aimed at investigating the potential as a growth enhancer of different dietary concentrations of turmeric (Curcuma longa Linn.) as an alternative to oxytetracyline and antibiotics and as comparable to mannan oligosaccharide for broiler chicks. A total of 252 Hubbard broiler chicks at one day of age were distributed randomly in a straight run experimental design among six treatments, each replicated seven times, with six unsexed chicks per replicate. The basal diet was administered without supplements (control group) or supplemented with turmeric at 0.5, 1, and 2 g/kg diet, or with mannan oligosacride (MOS) at 1 g/kg feed or with oxytetracycline (OTC) at 50 mg/kg feed. Growth performance, carcass characteristics, meat quality traits, blood biochemical constituents, antioxidant status and red blood cell (RBCs) were investigated. Turmeric supplementation at 1 g/kg feed significantly improved feed conversion ratio (FCR) and European production index compared to the control group and MOS groups. The results indicated that turmeric can be used at 1 kg/t feed as a phytogenic feed additive as an alternative to OTC or MOS without negative effects on the productive and economic traits of broilers. There were no differences from using OTC and MOS, while there was an increase in the European production efficiency index and the broilers’ health status.

The past, current and planned future developments of X-ray imagers in the Photon-Science Detector Group at DESY-Hamburg is presented. the X-ray imagers are custom developed and tailored to the different X-ray sources in Hamburg, including the storage ring PETRA III/IV; the VUV-soft X-ray free electron laser FLASH, and the European Free-Electron Laser. Each source puts different requirements on the X-ray detectors, which is described in detail, together with the technical solutions implemented.

- Abstract
The European X-ray Free Electron Laser (EuXFEL) began its user operation five years ago, opening and offering new research possibilities. The facility delivers high brilliance, ultra-short, spatially coherent X-ray pulses with a high repetition rate to six instruments (FXE, SPB/SFX, MID, HED, SCS and SQS) by means of three different beamlines (SASE 1, SASE 2 and SASE 3). One of the first detectors used for early-stage experiments was the Adaptive Gain Integrating Pixel Detector (AGIPD), custom designed to meet the challenging needs of scientific instruments. The AGIPD is a megahertz-rate integrating hybrid megapixel camera with a per-pixel adaptive gain amplification, allowing the integration of up to 104 of 12 keV photons per pixel in its low gain stage. Currently, three scientific instruments, namely SPB/SFX, MID and HED employ the AGIPD systems, the latter mentioned using a prototype, half-megapixel camera with an upgraded version of readout ASICs. The AGIPDs at EuXFEL are successfully used for experimental techniques like serial femtosecond crystallography, MHz single particle imaging, MHz X-ray photon correlation spectroscopy or MHz diffraction of materials under high pressures in a diamond anvil cell. Since September 2017, the AGIPD is continuously used and has become an established detector technology, with further advancements and developments planned. Delivering quality experimental data requires reliable and reproducible detector characterisation and calibration that have to be performed regularly with a continuous improvement of correction methods in close collaboration with scientific instruments. This work summarises five years of experience operating the AGIPD detectors at the EuXFEL scientific instruments. It gives an overview of scientific capabilities and examples of successful studies performed with AGIPD detectors. Moreover, challenges concerning detector calibration and characterisation are presented.

Aim of study: To evaluate the use of compost and natural rocks as partial replacement of mineral fertilizers in ‘Flame’ seedless grape vineyards. Area of study: The present work was conducted during three successive seasons (2016, 2017 and 2018), being the first season a preliminary trial on 4-yr old grapes cultivated in two different soil types (sandy and clay) at two different locations, Egypt (Abo Galeb, Giza governorate; EL-Mahala, Gharbia governorate). Material and methods: Treatments were applied as natural raw materials at 20, 40, 60, 80 and 100% out of recommended mineral NPK rate. The mineral fertilizers used were ammonium sulfate (21.6% N), calcium super phosphate (15.5% P2O5) and potassium sulfate (48% K2O). The natural rocks used were phosphate rock (22.0% P2O5) and Feldspar (10.12% K2O). Yield and fruit characteristics and leaf mineral content were determined. Main results: Using compost in combination with natural rocks enriched with NPK mobilization bacteria and mineral NPK enhanced leaf nutrients content and gave the highest yield and cluster weight. This mix also improved berries physical and chemical characteristics. There was an increase in soluble solids content (SSC), SSC/acid ratio, and anthocyanin content, associated with a reduction in nitrate content of the berry juice. The most pronounced effect was related to using 60% mineral fertilization + 40% organic and natural rocks in both vineyard locations. Research highlights: We can reduce the recommended doses of mineral NPK by about 40%, reducing then the soil pollution.

We report the first record of intersexuality in little tunny Euthynnus alletteratus caught in the Mediterranean Sea. The little tunny were sampled in traps from La Azohía (Murcia) south-eastern Spanish Mediterranean coast and Tarifa (Cádiz) between April and June, during 2003 to 2012. This document is the final report of the second year of the short-term contract of the Small Tuna Year Program by ICCAT. In 2018, the Small Tuna Species Group decided to prioritize Little tunny (LTA) (Euthynnus alletteratus) Atlantic bonito (BON) (Sarda sarda) and Wahoo (WAH) (Acanthocybium solandri), based on their economic importance and the deficiency of knowledge of their biology. The objectives of the contract for three species were: i) Collect biological samples for estimating growth parameters, assessing the maturity and stock structure analysis (populations genetics), and ii) Conclude the analysis of the stock structure for at least one of the three species and provide preliminary results for the remaining. The obtained samples for growth, maturity and stock structure analysis was almost completed for Little tunny and Atlantic bonito, whereas for Wahoo the samples are scarce. This study assesses the stock structure of Atlantic bonito (Sarda sarda) using the mitochondrial control region as a genetic marker. About 615 individuals distributed in seven locations were analyzed. Two of the locations were in the Mediterranean Sea (MD/BIL95) (Spain and Tunis), three in the northeast Atlantic (AT-NE/BIL94B) (Portugal, Tunis, Morocco and Mauritania), and one in the southeast Atlantic (AT-SE/BIL97) (Côte d’Ivoire). All these samples were obtained thanks to the participation of all authors in two Small Tuna Research Programs funded by ICCAT. The analysis of the genetic variability of the sequence of mitochondrial control regions depicts a clear heterogeneity among locations. The shared genetic pool that comprises the locations within the Mediterranean (Spain and Tunis), including also a sample from the northeast Atlantic (Portugal), is clearly different from the African locations (Senegal and Côte d’Ivoire). Moreover, these two African locations are also genetically differentiated between them. Morocco and Mauritania locations seems to be located in an intermediate situation between these two groups of locations. These results can be used to infer a management policy by ICCAT on the fisheries of this species

This study intended to assess the properties of in-ovo administration of Melittin (MLT) on hatchability, chick yield, hematology, immunological indices and relative organs weight of Alexandria chickens at hatch. A total of 600 eggs with an average weight of (45.12 g), were gathered and split into five groups: a non-injected group or negative control (NC), a saline injection group or positive control (PC), and three concentrations of MLT (5, 10 and 15 µg of MLT per egg, respectively). On day 18 of incubation, eggs from the injection groups were injected into the amniotic fluid from the large end with the in-ovo injection solutions (0.2 ml per egg). Results indicated that 10 µg MLT/egg positively affected the weight and yield of chicks. In addition, our findings indicated that the in-ovo administration with 10 or 15 µg MLT/egg was superior in most of the immunological indicators (spleen and bursa relative weights, immunoglobulins IgG and IgM, T cells and B cells). In conclusion, in order to improve the immune efficiency (early immune acceleration) of Alexandria chicks, which may contribute to offering a significant boost to their future performance, this study suggests injecting eggs with 5 or 10 µg MLT/egg. The present study aimed to investigate the effect of early access to feed and water post-hatch on broiler chicks' performance. One hundred and twenty chicks were transferred from the hatchery to the rearing house and randomly divided into two groups. The first group: chicks were immediately access to feed and water (F-time 0). The second group: was held without feed and water for 24 h (F-time 24). Then, feed and water were provided ad-libitum, for both groups until 35 days of age. Results indicated that F-time 0 increased body weight and body weight gain throughout the experimental period. It increased feed intake during all experimental periods except from (22–28 days). Additionally, the F-time 0 enhanced the European production efficiency factor index. The F-time 24, increased red blood cells (RBCs) count, hemoglobin (HGB), and packed cell volume (PCV) percentage after 24 h. However, the F-time 0 had a higher RBCs count, HGB, and PCV at 35 days of age. F-time 24 increased total plasma protein, albumin, cholesterol, and triglycerides, after 24 h. In conclusion, early access to feed and water post-hatch enhances broiler chicks' performance and productivity and increases producers' revenue. [ABSTRACT FROM AUTHOR]

HMO is genetically heterogeneous, localized on at least three chromosomal loci including 8q24.1 (EXT1), 11p11-p13 (EXT2), and 19p (EXT3). The median age of diagnosis is 3 years; almost all affected individuals are diagnosed by age 12. The risk for malignant degeneration to osteochondrosarcoma increases with age, although the lifetime risk of malignant degeneration is low (~1%).Methods and results: This study was performed on an Iranian family with nine affected individuals from three consecutive generations. Here, the proband was an affected woman who received genetic counseling prior to pregnancy. All exons of the three genes were examined in the proband using polymerase chain reaction and sequencing methods (the last member of this family is a male with severe deformities and lesions, especially around his large joints). Exon 4 of EXT1 (c.1235 G>A) was changed in affected individuals. This mutation alters tryptophan to a premature stop codon on amino acid position 412 (p.Trp412x).Conclusion: The outcome of this study has extended the genotypic spectrum of Iranian patients with HMO, revealing a way for improving detection and genetic counseling in carriers.
