1. Detection of bromochloro alkanes in indoor dust using a novel CP-Seeker data integration tool

Abstract

Bromochloro alkanes (BCAs) have been manufactured for use as flame retardants for decades and preliminary environmental risk screening suggests they are likely to behave similarly to polychlorinated alkanes (PCAs), subclasses of which are restricted as Stockholm Convention Persistent Organic Pollutants (POPs). BCAs have rarely been studied in the environment, though some evidence suggests they may migrate from treated-consumer materials into indoor dust, resulting in human exposure via inadvertent ingestion. In this study, BCA-C14 mixture standards were synthesized and used to validate an analytical method. This method relies on chloride-enhanced liquid chromatography-electrospray ionization-Orbitrap-high resolution mass spectrometry (LC-ESI-Orbitrap-HRMS) and a novel CP-Seeker integration software package for homologue detection and integration. Dust sample preparation via ultrasonic extraction, acidified silica clean-up and fractionation on neutral silica cartridges was found to be suitable for BCAs, with absolute recovery of individual homologues averaging 66 to 78% and coefficients of variation $\le$10% in replicated spiking experiments (n=3). In addition, a total of 59 indoor dust samples from six countries including Australia (n=10), Belgium (n=10), Colombia (n=10), Japan (n=10), Thailand (n=10) and the United States of America (n=9) were analysed for BCAs. BCAs were detected in seven samples from the USA, with carbon chain lengths of C8, C10, C12, C14, C16, C18, C24 to C28, C30 and C31 observed overall, though not detected in samples from any other countries. Bromination of detected homologues in the indoor dust samples ranged from Br1-4 as well as Br7, while chlorine numbers ranged from Cl2-11. BCA-C18 were the most frequently detected, observed in each of the USA samples, while the most prevalent halogenation degrees were homologues of Br2 and Cl4-5. Broad estimations of BCA concentrations in the dust samples indicated that levels may approach those of other flame retardants in at least some instances. These findings suggest that development of quantification strategies and further investigation of environmental occurrence and health implications are needed.

International audience; IntroductionMetabolomics data analysis is a complex and multistep process, which is constantly evolving with the development of new analytical technologies, mathematical methods, bioinformatics tools and databases. The Workflow4Metabolomics (W4M) infrastructure[1] provides tools in a single online web interface (through Galaxy[1]). During the last two years, W4M has evolved with new upgrades for LC-MS, LC-MSMS, GC-MS and NMR pipelines, including preprocessing, quality control, statistical analysis and annotation tools. W4M also proposes new community resources promoting open science in metabolomics.Technological and methodological innovationW4M major updates include: Specific Galaxy interactive tools with NMRPro[2] for NMR spectra visualization and Xseeker for visualization and annotation of LC-MSMS data (In collaboration with CHOPIN ANR project[3]); An improved MSMS pipeline; New annotation tools suite allowing connexion with PeakForest[4], a new spectral data manager infrastructure for laboratories; New functionalities regarding mixed model computation for repeated measure designs, and improvements in annotation of complex mixture bidimensional NMR spectra. New training resources through the Galaxy Training Network (GTN) are also proposed, and Training Infrastructure as a Service (TIass) is available through the new usegalaxy.fr host.Results and impactW4M’s improvements increase raw data input management and LC/GC-MS(MS) workflow efficiency. We highlight how current advances, along with community training as through the yearly international school Workflow4Experimenters, contribute to open data analysis practices worldwide. Workflow4Metabolomics[1] is a collaborative portal dedicated to metabolomics data processing, analysis and annotation for the Metabolomics community. In the latest version of W4M, the core team proposes new upgrades for LC-MS, GC-MS and NMR pipelines, including new preprocessing steps, as well as enhancement of statistical analysis and annotation tools. W4M aims to promote open science in Metabolomics and facilitate knowledge dissemination by providing community resources.

Abstract

Skeletal stem cells have been isolated from various tissues, including periosteum and bone marrow, where they exhibit key functions in bone biology and hematopoiesis, respectively. The role of periosteal skeletal stem cells in bone regeneration and healing has been extensively studied, but their ability to contribute to the bone marrow stroma is still under debate. In the present study, we characterized a whole bone transplantation model that mimics the initial bone marrow necrosis and fatty infiltration seen after injury. Using this model and a lineage tracing approach, we observed the migration of periosteal skeletal stem cells into the bone marrow after transplantation. Once in the bone marrow, periosteal skeletal stem cells are phenotypically and functionally reprogrammed into bone marrow mesenchymal stem cells that express high levels of hematopoietic stem cell niche factors such as Cxcl12 and Kitl. In addition, using in-vitro and in-vivo approaches, we found that periosteal skeletal stem cells are more resistant to acute stress than bone marrow mesenchymal stem cells. These results highlight the plasticity of periosteal skeletal stem cells and their potential role in bone marrow regeneration after bone marrow injury.
