Amyloids are proteinaceous aggregates related to the so-called conformational diseases, such as Alzheimer's and prion diseases. The cytotoxicity of amyloids may be related to the interaction of the amiloidogenic peptides or proteins with the cell membrane. In order to gain information on the physico-chemical effects of amyloids on membranes, we have studied the interaction of the human prion amyloidogenic fragment PrP 185-206 with negatively charged model membranes. The results show that the peptide causes the destabilization of the membrane, making it permeable to potassium ions and to charged organic compounds. This effect correlates with the interaction of the peptide with the membrane, causing a variation in the magnitude of the electrostatic surface and dipole membrane potentials. This effect on the electrostatic properties of the membranes may help explaining the observed permeability: a neutralization of the surface negative charge and a decrease of the inside-positive dipole potential would facilitate the translocation of positive ions. The structural analysis of the peptide in the presence of model membranes reveals that it adopts a predominantly unordered structure without any signs of amyloid formation. The results may be relevant in relation to the recently described cell toxic capacity of the peptide.

Circularly polarized chlorophyll luminescence (CPL) was recently shown to be an effective tool for the study of chiral macroaggregate formation of the light-harvesting chlorophyll a/b pigment-protein complexes (LHCIIs) in isolated chloroplasts. The CPL measuring system was modified to study green leaves. Spectral and intensity features of pea leaf CPL signals suggested that CPL indeed detected chiral macroaggregates. The signals were found to depend on the excitation vs emission optical alignment, as well as on the side (adaxial or abaxial) of the leaf. Illumination of attached leaves with either low intensity or strong photoinhibitory light did not affect the chiral macroaggregation status. In contrast, the induction of drought stress in detached leaves led to full disruption of the chiral macroaggregates. The disruption developed gradually during slow dehydration, whereas under fast, heat-stimulated dehydration it was manifested in cooperative diminution of both CPL and photochemical capacity. Simultaneous exposure of the leaves to photoinhibitory illumination and fast dehydration resulted in negative CPL signals. The results demonstrate the potency of CPL as a non-destructive tool for structural studies of LHCII in intact leaves under varying environmental conditions.

Effects of crambin on the phospholipase activity, lipid peroxidation and structure of mitochondrial membranes have been investigated. Crambin has been shown to inhibit lipid peroxidation and phospholipase activity induced by Ca2+ and freezing-thawing of mitochondria. As shown by ESR studies, these effects are based on the ability of crambin to cause changes in mitochondrial membrane structure.

Induction of lipid peroxidation is shown to alter the structure of mitochondrial membrane and to activate endogenous phospholipases A2, C, D, and lysophospholipase A. Fe(2+)-, cumene hydroperoxide- and ultraviolet-induced lipid peroxidation results in the activation of mitochondrial phospholipases.

Interaction between cytotoxin Vc5 from cobra venom and model membranes from phosphatidylcholine and phosphatidyl acid was investigated by means of 1H-NMR, ESR spin probes and microcalorimetric methods. Cytotoxic was shown to induce in the membrane formation of non-bilayer structures, to increase the membrane permeability and to produce the intermembrane exchange with lipids.

By the methods of EPR spinal probes, energy migration of triplet excitation and NMR spectroscopy, the structural changes on hydrocarbon region of membranes, the changes in dynamic state of water of lipid hydrate jacket, the intermembrane lipid material exchange and fusion of membranes induced by cytotoxins of cobra venom have been studied. The sequence of events preceded the membrane fusion is suggested. The probability of membrane fusion has been shown not to be determined by fusogenic agent structure only, but much it depends on lipid composition of membranes.

The interaction of cytotoxin Vc1 and Vc5 from Central Asian cobra and melittin from the bee venom with multilayer liposomes prepared from dimiristoylphosphatidylcholine with an addition of phosphatidic acid have been studied by the method of differential scanning calorimetry. Incorporation of Vc1, Vc5 and melittin into the lipid resulted in pronounced changes in the thermodynamic properties of the lipid. Polypeptides studied induced lateral phase separation in the lipid. Interaction between molecules of the toxins and the lipid resulted in the formation of a new lipid phase characterized by a higher melting temperature and lower phase transition enthalpy.

A rate of CA2+ accumulation and efficiency of Ca2+-pump were decreased in the fractions of skeletal muscle sarcoplasmic reticulum isolated from rabbits with thyrotoxicosis. Temperature of phase transition in membranes of sarcoplasmic reticulum was changed by 4-5 degrees towards low temperatures. Under conditions of thyrotoxicosis fatty acid composition of total lipids from the sarcoplasmic reticulum fractions was altered as follows: amount of palmitic acid was increased 1.4-fold but oleic and arachidonic acids were decreased 21.8% and 40.5%, respectively. Modification of Ca2+-transport properties in sarcoplasmic reticulum caused by long-term administration of thyroid hormones into animals appears to alter the membrane protein-lipid composition.

The processes of membrane aggregation, permeability and fusion induced by cytotoxins from Central Asian cobra venom were investigated by studying optical density of liposome samples, permeability of liposome membranes for ferricyanide anions and exchange of lipid material between the membranes of adjacent liposomes. Cytotoxins Vc5 and Vc1 were found to induce aggregation of PC + CL and PC + PS liposomes. Cytotoxin Vc5 increased also the permeability of the liposomes for K3[Fe(CN)6] and enhanced their fusion. Cytotoxin Vc1 increased membrane permeability and enhanced fusion of PC + CL samples only. The changes in membrane permeability and fusion were found to occur within a single value of cytotoxin concentrations. The fusogenic properties of the cytotoxins studied are supposed to be due to the ability to dehydrate membrane surface and to destabilize the lipid bilayer structure. Fusion probability is largely defined by the phospholipid composition of the membranes. A model of interaction of cytotoxins with cardiolipin-containing membranes is offered.

Interaction between organophosphorous insecticide GA-41 and liposomes formed from lecithin and multilamellar dispersion from lecithin and cardiolipin mixture (molar ratio is 7:3) has been studied by 1H- and 31P-NMR spectroscopy. The results obtained showed that while interacting with model membranes the GA-41 molecules intercalate into the range of hydrocarbon chains of phospholipids. At certain concentration of GA-41 its molecules induce the formation of some defects on bilayer structure, leading to permeability for Pr3+ ions.

Interaction between cytotoxin of the Central Asia cobra venom and dimiristoylphosphatidylcholine bilayer depending on its phase state was studied by ESR with spin label. A conclusion can be drawn that the efficiency of cytotoxin effect on the membranes depends on their phase state. Cytotoxin molecules are incorporated into myophile region of the bilayer, only if the latter is in the liquid crystal state. The interaction between cytotoxins and lipids of the bilayer in a gel state is in the main conditioned by electrostatic forces.

Liposomes and proteoliposomes obtained from rat brain were used; structural changes induced by Vc5 cytotoxin (CT) from Central Asia cobra venom have been studied by the EPR method using spin probes (5-, 10-, or 12-doxylstearic acid). The addition of CT to liposome samples, containing spin probes resulted in the appearance of a new EPR signal in the initial spectrum (samples without CT), typical of probes with strongly retarded mobility. The presence of hydrophobic interaction between the CT molecules and spin labelled fat acids permits the assumption that CT molecules in liposomes trap both lipid probes and phospholipids localized in the reach of action of hydrophobic forces. CT may be supposed to induce formation in membranes of liposomes with domain structures. As a result of hydrophobic interaction with CT molecules both the phospholipid and lipid probe mobility in the domain is substantially less than that in liposome regions free of CT molecules. Due to this, a new signal appears in the initial EPR spectrum of the spin probes. An analysis of the dependence of the probe order parameter value on CT concentration in samples has suggested that CT act uniformly along the membrane lipid profile with a certain CT concentration range. At high concentrations CT molecules cannot penetrate the lipid region deep enough, due to mutual electrostatic repulsion and steric factors at membrane surface. As a result, structural changes involve regions adjacent to the membrane surface only.
