El propósito de la presente investigación, ha sido el de realizar un análisis estratégico del sector de educación superior universitario peruano, utilizando para ello, el conocido modelo de las cinco fuerzas competitivas de Michael Porter. El estudio ha seguido un enfoque cualitativo, con diseño de estudio de caso. Las técnicas e instrumentos de recolección de datos usados, han respondido a la naturaleza de las fuerzas competitivas analizadas (entrevista y observación documental). El análisis de los datos, ha seguido los lineamientos establecidos por el modelo en cuestión, habiendo demostrado los resultados, no solo la utilidad práctica del modelo como una eficaz herramienta para el análisis estratégico de las organizaciones, sino, además, ha apoyado los postulados teóricos expuestos por la literatura especializada en administración estratégica.

Chitin and its derivative chitosan (Q) are abundant structural elements in nature. Q has modulatory and anti-inflammatory effects and also regulates the expression of adhesion molecules. The interaction between cells expressing the αEβ7 integrin and E-cadherin facilitates tolerogenic signal transmission and localization of lymphocytes at the frontline for interaction with luminal antigens. In this study we evaluated the ability of orally administered Q to stimulate E-cadherin and CD103 expression in vitro and in vivo. Our findings show that Q promoted epithelial cell migration, accelerated wound healing and increased E-cadherin expression in IEC-18 cells and isolated intestinal epithelial cells (IECs) after Q feeding. The upregulation of E-cadherin was dependent on TLR4 and IFNAR signaling, triggering CD103 expression in lymphocytes. One of the challenges for producing active chitinase formulations relies on the gap between the laboratory tests and the biological scenarios where the enzyme will perform its function. In this work, we have employed different Langmuir monolayer arrays to evaluate the interfacial behavior of a recently purified recombinant chitinase, Chi18-5. We have demonstrated that two conformations exist for the chitinase at pH values close to its pI, showing very distinct structural properties at the air/aqueous interface. Enzyme activity was assessed by implementing different kinetic approaches and using a chitosan-1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) mixed film as organized substrate model membrane. Combining these strategies, we demonstrated that better catalytic efficiencies can be obtained for Chi18-5 at pH 5. Moreover, the chitinase activity at the air/aqueous interface can be tuned by introducing in situ pH modifications over the surrounding milieu. We also studied the changes in the topography at the mesoscale level using Brewster Angle Microscopy (BAM). We found that Chi18-5 segregated onto the chitosan domains of the membrane, showing differences in homogeneity depending on the pH imposed. Alternatively, pure Chi18-5 was tested for immobilization onto a hydrophilic activated solid support using the Langmuir-Blodgett technique. Chitinase chi18-5 is an enzyme able to hydrolyze chitin and chitosan producing chitooligosaccharides (COS) of potential technological interest. chi18-5 is produced naturally by the fungus Trichoderma atroviride. It belongs to the glycosyl hydrolase (GH) family 18 of the Carbohydrate Active Enzyme (CAZy) database and it has 83% identity compared to the well-characterized chi42 of Trichoderma harzianum. Several efforts have been made to characterize the biochemical activity of the enzyme and its structure. Here, we studied the biophysical properties of recombinant chi18-5. In order to gain insight into its structure and stability, we studied thermal denaturation by Circular Dichroism (CD), Intrinsic Fluorescence (FL), and attenuated total reflection Fourier transform infrared spectroscopy (ATR-FT-IR) at several pH between 3 and 8. We observed that the conformation of chi18-5 changes near its pI, and the transitions as a function of the temperature involved an increment in β-sheet secondary structure at the expenses of ⍺-helix. We also performed amide hydrogen exchange dynamics in selected conditions. At pH ≤ 6, the proportion of fast exchanging residues are larger than at pH ≥ 6. Our results suggest that at pH below pI, chi18-5 is in a less compact structure which may have influence in the interaction with substrate and enzyme activity. The Maier-Saupe-Zwanzig model for the nematic phase transitions in liquid crystals is investigated in a diamond hierarchical lattice. The model takes into account a parameter to describe the biaxiality of the microscopic units. Also, a suitably chosen external field is added to the Hamiltonian to allow the determination of critical parameters associated with the nematic phase transitions. Using the transfer-matrix technique, the free energy and its derivatives are obtained in terms of recursion relations between successive generations of the hierarchical lattice. In addition, a real-space renormalization-group approach is developed to obtain the critical parameters of the same model system. Results of both methods are in excellent agreement. There are indications of two continuous phase transitions. One of them corresponds to a uniaxial-isotropic transition, in the class of universality of the three-state Potts model on the diamond hierarchical lattice. The transition between the biaxial and the uniaxial phases is in the universality class of the Ising model on the same lattice.

We studied the conformational changes of the fatty acid-binding protein ReP1-NCXSQ in the interface of anionic lipid membranes. ReP1-NCXSQ is an acidic protein that regulates the activity of the Na + /Ca 2+ exchanger in squid axon. The structure is a flattened barrel composed of two orthogonal β-sheets delimiting an inner cavity and a domain of two α-helix segments arranged as a hairpin. FTIR and CD spectroscopy showed that the interactions with several anionic lipids in the form of small unilamellar vesicles (SUVs) induced an increase in the proportion of helix secondary structure. Lower amount or no increase in α-helix was observed upon the interaction with anionic lipids in the form of large unilamellar vesicles (LUVs). The exception was 1,2-dimyristoyl-sn-glycero-3-phosphoglycerol (DMPG) that was equally efficien to to induce the conformational change both in SUVs and in LUVs. In solution, the infrared spectra of ReP1-NCXSQ at temperatures above the unfolding displayed a band at 1617 cm -1 characteristic of aggregated strands. This band was not observed when the protein interacted with DMPG, indicating inhibition of aggregation in the interface. Similarly to the observed in L-BABP, another member of the fatty acid binding proteins, a conformational change in ReP1-NCXSQ was coupled to the gel to liquid-crystalline lipid phase transition.

We perform statistical mechanics calculations to analyze the global phase diagram of a fully connected version of a Maier-Saupe-Zwanzig lattice model with the inclusion of couplings to an elastic strain field. We point out the presence of uniaxial and biaxial nematic structures, depending on temperature T and on the applied stress σ. Under uniaxial extensive tension, applied stress favors uniaxial orientation, and we obtain a first-order boundary along which there is a coexistence of two uniaxial paranematic phases, and which ends at a simple critical point. Under uniaxial compressive tension, stress favors biaxial orientation; for small values of the coupling parameters, the first-order boundary ends at a tricritical point, beyond which there is a continuous transition between a paranematic and a biaxially ordered structure. For some representative choices of the model parameters, we obtain a number of analytic results, including the location of critical and tricritical points and the line of stability of the biaxial phase.

Objective: To test the relationship between patient experience, as measured by the Hospital Consumer Assessment of Healthcare Providers and Systems (HCAHPS), and actual quality of care, as measured by 30-day readmission rates., Data Sources: Both HCAHPS data and outcome data reported to the Centers of Medicare & Medicaid Services (CMS)., Study Design: This secondary, nationwide (N = 4060), hospital-level study focused only on acute care hospitals. HCAHPS question 22 "Would you recommend this hospital to your friends and family?" was used to determine level of overall satisfaction, and 30-day readmission rates, as reported to the CMS, was used as a proxy for actual quality of care., Principal Findings: A statistically significant relationship was found between patient experience and actual quality outcomes., Conclusion: The results of this study reinforce the inclusion of patient experience in Medicare's Value Based Purchasing program as a matter of good public policy.

GumK is a membrane-associated glucuronosyltransferase of Xanthomonas campestris that is involved in xanthan gum biosynthesis. GumK belongs to the inverting GT-B superfamily and catalyzes the transfer of a glucuronic acid (GlcA) residue from uridine diphosphate (UDP)-GlcA (UDP-GlcA) to a lipid-PP-trisaccharide embedded in the membrane of the bacteria. The structure of GumK was previously described in its apo- and UDP-bound forms, with no significant conformational differences being observed. Here, we study the behavior of GumK toward its donor substrate UDP-GlcA. Turbidity measurements revealed that the interaction of GumK with UDP-GlcA produces aggregation of protein molecules under specific conditions. Moreover, limited proteolysis assays demonstrated protection of enzymatic digestion when UDP-GlcA is present, and this protection is promoted by substrate binding. Circular dichroism spectroscopy also revealed changes in the GumK tertiary structure after UDP-GlcA addition. According to the obtained emission fluorescence results, we suggest the possibility of exposure of hydrophobic residues upon UDP-GlcA binding. We present in silico-built models of GumK complexed with UDP-GlcA as well as its analogs UDP-glucose and UDP-galacturonic acid. Through molecular dynamics simulations, we also show that a relative movement between the domains appears to be specific and to be triggered by UDP-GlcA. The results presented here strongly suggest that GumK undergoes a conformational change upon donor substrate binding, likely bringing the two Rossmann fold domains closer together and triggering a change in the N-terminal domain, with consequent generation of the acceptor substrate binding site., (© The Author 2016. We introduce shape variations in a liquid-crystalline system by considering an elementary Maier-Saupe lattice model for a mixture of uniaxial and biaxial molecules. Shape variables are treated in the annealed (thermalized) limit. We analyze the thermodynamic properties of this system in terms of temperature T, concentration c of intrinsically biaxial molecules, and a parameter Δ associated with the degree of biaxiality of the molecules. At the mean-field level, we use standard techniques of statistical mechanics to draw global phase diagrams, which are shown to display a rich structure, including uniaxial and biaxial nematic phases, a reentrant ordered region, and many distinct multicritical points. Also, we use the formalism to write an expansion of the free energy in order to make contact with the Landau-de Gennes theory of nematic phase transitions.

We consider an Ising model on the triangular Apollonian network (AN), with a thermalized distribution of vacant sites. The statistical problem is formulated in a grand canonical ensemble, in terms of the temperature T and a chemical potential μ associated with the concentration of active magnetic sites. We use a well-known transfer-matrix method, with a number of adaptations, to write recursion relations between successive generations of this hierarchical structure. We also investigate the analogous model on the diamond hierarchical lattice (DHL). From the numerical analysis of the recursion relations, we obtain various thermodynamic quantities. In the μ→∞ limit, we reproduce the results for the uniform models: in the AN, the system is magnetically ordered at all temperatures, while in the DHL there is a ferromagnetic-paramagnetic transition at a finite value of T. Magnetic ordering, however, is shown to disappear for sufficiently large negative values of the chemical potential.

This study investigated the structural and biophysical characteristics of GumB and GumC, two Xanthomonas campestris membrane proteins that are involved in xanthan biosynthesis. Xanthan is an exopolysaccharide that is thought to be a virulence factor that contributes to bacterial in planta growth. It also is one of the most important industrial biopolymers. The first steps of xanthan biosynthesis are well understood, but the polymerization and export mechanisms remain unclear. For this reason, the key proteins must be characterized to better understand these processes. Here we characterized, by biochemical and biophysical techniques, GumB, the outer membrane polysaccharide export protein, and GumC, the polysaccharide co-polymerase protein of the xanthan biosynthesis system. Our results suggested that recombinant GumB is a tetrameric protein in solution. On the other hand, we observed that both native and recombinant GumC present oligomeric conformation consistent with dimers and higher-order oligomers. The transmembrane segments of GumC are required for GumC expression and/or stability. These initial results provide a starting point for additional studies that will clarify the roles of GumB and GumC in the xanthan polymerization and export processes and further elucidate their functions and mechanisms of action., (Copyright © 2014 Elsevier Inc. GumB is a predicted outer membrane lipoprotein that is involved in the synthesis and/or secretion of xanthan gum. This exopolysaccharide, produced by Xanthomonas campestris, is valuable in industry because of its important rheological properties. Solution of the GumB structure will provide insight into the polymerization and/or secretion mechanisms of xanthan gum. GumB was overexpressed and purified and diffraction-quality crystals of native GumB were obtained. A complete data set was collected to 2.54 Å resolution with an R(p.i.m.) of 0.034. The crystals belonged to space group P2(1)2(1)2(1), with unit-cell parameters a = 84.4, b = 90.5, c = 120.7 Å.

We analyze the global phase diagram of a Maier-Saupe lattice model with the inclusion of shape-disordered degrees of freedom to mimic a mixture of oblate and prolate molecules (discs and cylinders). In the neighborhood of a Landau multicritical point, solutions of the statistical problem can be written as a Landau-de Gennes expansion for the free energy. If the shape-disordered degrees of freedom are quenched, we confirm the existence of a biaxial nematic structure. If orientational and disorder degrees of freedom are allowed to thermalize, this biaxial solution becomes thermodynamically unstable. Also, we use a two-temperature formalism to mimic the presence of two distinct relaxation times, and show that a slight departure from complete thermalization is enough to stabilize a biaxial nematic phase.

We use Z-scan technique to investigate the nonlinear optical response of the thermotropic liquid crystal E7 in the neighborhood of the nematic-isotropic phase transition. The analysis of the data for the nonlinear optical birefringence is compatible with an effective critical exponent of the order parameter, β = 0.28 ± 0.03, which is close to the classical value, β = 0.25 , for a tricritical point. The nonlinear optical absorption in the nematic range depends on the geometrical configuration of the nematic director with respect to the polarization beam, and vanishes in the isotropic phase.

We introduce a simple mean-field lattice model to describe the behavior of nematic elastomers. This model combines the Maier-Saupe-Zwanzig approach to liquid crystals and an extension to lattice systems of the Warner-Terentjev theory of elasticity, with the addition of quenched random fields. We use standard techniques of statistical mechanics to obtain analytic solutions for the full range of parameters. Among other results, we show the existence of a stress-strain coexistence curve below a freezing temperature, analogous to the P-V diagram of a simple fluid, with the disorder strength playing the role of temperature. Below a critical value of disorder, the tie lines in this diagram resemble the experimental stress-strain plateau and may be interpreted as signatures of the characteristic polydomain-monodomain transition. Also, in the monodomain case, we show that random fields may soften the first-order transition between nematic and isotropic phases, provided the samples are formed in the nematic state.

We describe the first biochemical characterization of the gumI gene product, an essential protein for xanthan polysaccharide synthesis. Cellular fractionation experiments reveal the presence of a protein associated with the membrane fraction, even in the absence of the other proteins responsible for the synthesis of glycolipid intermediates and the proteins involved in the polymerization and transport of the xanthan chains. By alkaline buffer extraction and detergent phase partitioning, GumI was categorized as a monotopic membrane protein. GumI was overexpressed in Escherichia coli, solubilized and purified in an active and stable form using a simple and reproducible two-step procedure. The purified recombinant GumI is a nonprocessive β-mannosyltransferase that uses GDP-Man as a donor substrate and glucuronic acid-β-1,2-mannose-α-1,3-glucose-β-1,4-glucose-PP-polyisoprenyl as an acceptor. We also established the optimal biochemical conditions for GumI enzymatic activity. Sequence analysis revealed the presence of a conserved domain for glycosyltransferases (GTs) of the GT-B superfamily and homologous proteins in several prokaryote organisms. On the basis of this biochemical characterization, GumI may represent the founding member of a new GT family in the Carbohydrate-Active EnZymes classification.

We investigate the phase diagram of a discrete version of the Maier-Saupe model with the inclusion of additional degrees of freedom to mimic a distribution of rodlike and disklike molecules. Solutions of this problem on a Bethe lattice come from the analysis of the fixed points of a set of nonlinear recursion relations. Besides the fixed points associated with isotropic and uniaxial nematic structures, there is also a fixed point associated with a biaxial nematic structure. Due to the existence of large overlaps of the stability regions, we resorted to a scheme to calculate the free energy of these structures deep in the interior of a large Cayley tree. Both thermodynamic and dynamic-stability analyses rule out the presence of a biaxial phase, in qualitative agreement with previous mean-field results.

We consider a simple Maier-Saupe statistical model with the inclusion of disorder degrees of freedom to mimic the phase diagram of a mixture of rodlike and disklike molecules. A quenched distribution of shapes leads to a phase diagram with two uniaxial and a biaxial nematic structure. A thermalized distribution, however, which is more adequate to liquid mixtures, precludes the stability of this biaxial phase. We then use a two-temperature formalism, and assume a separation of relaxation times, to show that a partial degree of annealing is already sufficient to stabilize a biaxial nematic structure.

Xanthomonas campestris GumK (beta-1,2-glucuronosyltransferase) is a 44-kDa membrane-associated protein that is involved in the biosynthesis of xanthan, an exopolysaccharide crucial for this bacterium's phytopathogenicity. Xanthan also has many important industrial applications. The GumK enzyme is the founding member of the glycosyltransferase family 70 of carbohydrate-active enzymes, which is composed of bacterial glycosyltransferases involved in exopolysaccharide synthesis. No x-ray structures have been reported for this family. To better understand the mechanism of action of the bacterial glycosyltransferases in this family, the x-ray crystal structure of apo-GumK was solved at 1.9 angstroms resolution. The enzyme has two well defined Rossmann domains with a catalytic cleft between them, which is a typical feature of the glycosyltransferase B superfamily. Additionally, the crystal structure of GumK complexed with UDP was solved at 2.28 angstroms resolution. We identified a number of catalytically important residues, including Asp157, which serves as the general base in the transfer reaction. Residues Met231, Met273, Glu272, Tyr292, Met306, Lys307, and Gln310 interact with UDP, and mutation of these residues affected protein activity both in vitro and in vivo. The biological and structural data reported here shed light on the molecular basis for donor and acceptor selectivity in this glycosyltransferase family. These results also provide a rationale to obtain new polysaccharides by varying residues in the conserved alpha/beta/alpha structural motif of GumK.

We obtain exact expressions for the two-time autocorrelation and response functions of the -dimensional linear Glauber model. Although this linear model does not obey detailed balance in dimensions d > or = 2, we show that the usual form of the fluctuation-dissipation ratio still holds in the stationary regime. In the transient regime, we show the occurrence of aging, with a special limit of the fluctuation-dissipation ratio, x(infinity) = 1/2, for a quench at the critical point.

Using a weak-disorder scheme and real-space renormalization-group techniques, we obtain analytical results for the critical behavior of various q-state Potts models with correlated disordered exchange interactions along d(1) of d spatial dimensions on hierarchical (Migdal-Kadanoff) lattices. Our results indicate qualitative differences between the cases d-d(1)=1 (for which we find non-physical random fixed points, suggesting the existence of non-perturbative fixed distributions) and d-d(1)>1 (for which we do find acceptable perturbative random fixed points), in agreement with previous numerical calculations by Andelman and Aharony [Phys. Rev. B 31, 4305 (1985)]. We also rederive a criterion for relevance of correlated disorder, which generalizes the usual Harris criterion.

We use a mixed-spin model, with aperiodic ferromagnetic exchange interactions and crystalline fields, to investigate the effects of deterministic geometric fluctuations on first-order transitions and tricritical phenomena. The interactions and the crystal-field parameters are distributed according to some two-letter substitution rules. From a Migdal-Kadanoff real-space renormalization-group calculation, which turns out to be exact on a suitable hierarchical lattice, we show that the effects of aperiodicity are qualitatively similar for tricritical and simple critical behavior. In particular, the fixed point associated with tricritical behavior becomes fully unstable beyond a certain threshold dimension (which depends on the aperiodicity), and is replaced by a two-cycle that controls a weakened and temperature-depressed tricritical singularity.

We investigate the effects of geometric fluctuations, associated with aperiodic exchange interactions, on the critical behavior of q-state ferromagnetic Potts models on generalized diamond hierarchical lattices. For layered exchange interactions according to some two-letter substitutional sequences, and irrelevant geometric fluctuations, the exact recursion relations in parameter space display a nontrivial diagonal fixed point that governs the universal critical behavior. For relevant fluctuations, this fixed point becomes fully unstable, and we show the apperance of a two-cycle, which is associated with a novel critical behavior. We use scaling arguments to calculate the critical exponent alpha of the specific heat, which turns out to be different from the value for the uniform case. We check the scaling predictions by a direct numerical analysis of the singularity of the thermodynamic free energy. The agreement between scaling and direct calculations is excellent for stronger singularities (large values of q). The critical exponents do not depend on the strengths of the exchange interactions.
