RNA-Puzzles is a collective endeavor dedicated to the advancement and improvement of RNA three-dimensional structure prediction. With agreement from structural biologists, RNA structures are predicted by modeling groups before publication of the experimental structures. We report a large-scale set of predictions by 18 groups for 23 RNA-Puzzles: 4 RNA elements, 2 Aptamers, 4 Viral elements, 5 Ribozymes and 8 Riboswitches. We describe automatic assessment protocols for comparisons between prediction and experiment. Our analyses reveal some critical steps to be overcome to achieve good accuracy in modeling RNA structures: identification of helix-forming pairs and of non-Watson-Crick modules, correct coaxial stacking between helices and avoidance of entanglements. Three of the top four modeling groups in this round also ranked among the top four in the CASP15 contest.

O livro didático é ainda a principal ferramenta didática disponível em toda a extensão territorial do Brasil. A livro didático é um importante disseminador de informações determinantes para a formação de estudantes críticos e conscientes. Dessa forma, o presente estudo objetivou analisar os livros didáticos indicados no Programa Nacional do Livro e do Material Didático (PNLD) de 2020, para o ensino de Ciências no nível fundamental, referente aos conteúdos de animais peçonhentos. Os animais peçonhentos e os acidentes relacionados a esse grupo constituem um sério problema de saúde pública no Brasil e é considerado um tema ainda escasso de abordagem no âmbito escolar. A presente pesquisa do tipo qualitativa utilizou o método de pesquisa bibliográfica e Análise de Conteúdo de Bardin para avaliar sete dos doze livros discriminados no PNLD 2020 com relação ao tema “Animais Peçonhentos”, especificamente os volumes do sétimo ano do Ensino Fundamental onde o tema é abordado. A análise dos livros revelou que o conteúdo é inexistente ou está presente nas coleções de forma fragmentada e não contempla a diversidade, importância ecológica, médica, econômica e farmacêutica desses animais.

Between the fall of 2009 and 2019, total postsecondary instituion enrollment in the United States decreased by 5%, and for those students who do enroll in college, many who lack clear career objectives drop out, making the U.S. the nation with the highest college dropout rate in the industrialized world. Students' academic aspiratoins and career certainty have been shown to impact college outcomes. However, the impact of career uncertainty and academic aspirations on students' college outcomes has not been studied nationally. Using binomial regression analyses and a nationally representative sample (N = 23,503) of high school students, we investigated the impact of high school students' career uncertainty and academic aspirations on their college outcomes. Findings indicate that academic aspirations were a significant predictor of students' college application, enrollment, attendance, and major decision status. Career uncertainty was a significant predictor of students' college enrollment status. We discuss implications for practice and future research.

Children living in low-resource settings are frequently gut-colonized with multidrug-resistant bacteria. We explored whether breastfeeding may protect against childrens incident gut colonization with extended-spectrum beta-lactamase-producing Escherichia coli (ESBL-Ec) and Klebsiella, Enterobacter, or Citrobacter spp. (ESBL-KEC). We screened 937 monthly stool samples collected from 112 children aged 1-16 months during a 2016-19 prospective cohort study of enteric infections in peri-urban Lima. We used 52,816 daily surveys to examine how exposures to breastfeeding in the 30 days prior to a stool sample were associated with childrens risks of incident gut-colonization, controlling for antibiotic use and other covariates. We sequenced 78 ESBL-Ec from 47 children to explore their diversity. Gut-colonization with ESBL-Ec was increasingly prevalent as children aged, approaching 75% by 16 months, while ESBL-KEC prevalence fluctuated between 18% and 36%. Through 6 months of age, exclusively providing human milk in the 30 days prior to a stool sample did not reduce childrens risk of incident gut-colonization with ESBL-Ec or ESBL-KEC. From 6 to 16 months of age, every 3 additional days of breastfeeding in the prior 30 days was associated with 6% lower risk of incident ESBL-Ec gut-colonization (95% CI: 0.90, 0.98, p = .003). No effects were observed on incident ESBL-KEC colonization. We detected highly diverse ESBL-Ec among children and few differences between children who were predominantly breastfed (mean age: 4.1 months) versus older children (10.8 months). Continued breastfeeding after 6 months conferred protection against childrens incident gut colonization with ESBL-Ec in this setting. Policies supporting continued breastfeeding should be considered in efforts to combat antibiotic resistance.

Background: The limited and detailed literature on total intravenous anaesthesia (TIVA), as well as the clinical indications for unilateral ovariectomy in llamas, are not well-defined. Therefore, it is necessary to understand the anesthetic events and the surgical intervention in this species. Aim: The objective of this study was to evaluate the intraoperative physiological and clinical parameters in llamas undergoing unilateral ovariectomy, under three protocols of TIVA. Methods: Study was conducted with 30 healthy female llamas. Three groups were considered G1: Pre-anaesthetic - xylazine i/v, Induction: ketamine. Maintenance: FLK (fentanyl + lidocaine + ketamine) by continuous infusion. G2: Pre-anaesthetic - midazolam + tramadol + ketamine i/v. Induction – propofol. Maintenance, boluses of 20% of the total induction dose were added if required. G3: Pre-anaesthetic - midazolam + xylazine i/v. Induction: ketamine. Maintenance: with 20% of the total induction. Physiological parameters were evaluated at 5, 15 and 30 minutes. Results: The heart rate (HR), respiratory rate (RR), arterial oxygen saturation (SpO2) and rectal temperature (RT) of the groups showed significant differences at some point in the evaluation, presenting a degree of bradycardia and bradypnea in G1 and G2. A drop in SpO2 was seen at minute 5 in all groups, because of respiratory depression post-anaesthetic induction, subsequently the values increased, with better saturation in G2. RT remained similar in the groups until minute 20, then there was a significant decrease in G2. G1 showed significant fluctuations for SpO2 and G2 for RT. The majority of animals from G1 and G3 maintained marked relaxation of the abdomen and limbs, anal sphincter (G3), and mandible (G1). Salivation and tearing were an infrequent sign in G2. All animals from G3 presented some degree of sensitivity of the abdominal wall. The average duration of the surgery was 14 minutes, and the shortest anaesthetic recovery time occurred in G2 (28 minutes). Conclusion: The animals of the three groups showed significant differences in their physiological and clinical parameters, in response to the agents used. The G1 protocol: xylazine, ketamine y FLK. Background: Alpacas have reproductive traits such as induced ovulation and a higher gestation rate in the left uterine horn. Additionally, low fertility and high embryonic mortality are significant challenges in alpaca breeding. To address these issues, it is essential to study the histological changes occurring in the uterine wall during day 8 of the luteal phase. Aim: This research aims to describe these changes in alpacas with induced ovulation using GnRH or copulation. Methods: The study was conducted on 8 sexually mature fertile alpacas with a preovulatory follicle of ≥ 7 mm. Ovulation was induced with GnRH (G1, n=3) and natural mating' (G2, n=5), confirmed by ultrasound showing the disappearance of the follicle and formation of the corpus luteum. On day 8 of the luteal phase, samples of the uterine wall from the right and left uterine horns were obtained through a midline laparotomy. A uterine lavage with PBS was also performed, and no embryo-ova was found. The tissues were fixed, processed, and stained with Hematoxylin and Eosin. Measurements of the myometrium and endometrium, the area of the superficial and deep endometrial glands, the height of the gland cells, and the height of the endometrial mucosal cells were performed using microscopy and software. The Measurements were taken in microns (um), from 12 different photographs per animal. Results: In G2 (copulation), greater thickness of the endometrium and myometrium, as well as a larger area of the superficial glands and cell height, were observed (p < 0.005). Additionally, the area of the superficial glands in the left horn was greater compared to the right. Conclusion: The thickness of the myometrium and endometrium, and the area of the superficial glands of the left uterine horn in alpacas during the luteal phase (day 8), induced by copulation (G2), show better development. This research provides new insights into changes in the histomorphometry of the alpacas uterus during this stage and is the first report in this species. [Open Vet J 2024; 14(9.000): 2398-2406]

Despite robust literature associating IL-31 with pruritic inflammatory skin diseases, its influence on cutaneous inflammation and the interplay between inflammatory and neurosensory pathways remain unmapped. Here, we examined the consequences of disrupting Il31 and its receptor Il31ra in a mouse model of house dust mite (HDM)-induced allergic dermatitis. Il31-deficient mice displayed a deficit in HDM dermatitis-associated scratching, consistent with its well-established role as a pruritogen. In contrast, Il31 deficiency increased the number and proportion of cutaneous type 2 cytokine-producing CD4+ T cells and serum IgE in response to HDM. Furthermore, Il4ra+ monocytes and macrophages capable of fueling a feedforward type 2 inflammatory loop were selectively enriched in Il31ra-deficient HDM dermatitis skin. Thus, IL-31 is not strictly a proinflammatory cytokine but rather an immunoregulatory factor that limits the magnitude of type 2 inflammatory responses in skin. Our data support a model wherein IL-31 activation of IL31RA+ pruritoceptors triggers release of calcitonin gene-related protein (CGRP), which can mediate neurogenic inflammation, inhibit CD4+ T cell proliferation, and reduce T cell production of the type 2 cytokine IL-13. Together, these results illustrate a previously unrecognized neuroimmune pathway that constrains type 2 tissue inflammation in the setting of chronic cutaneous allergen exposure and may explain paradoxical dermatitis flares in atopic patients treated with anti-IL31RA therapy.

ABSTRACT The emergence of carbapenem-resistant Klebsiella pneumoniae (CRKP) poses a significant public health threat, particularly in low- and middle-income countries (LMICs) with limited surveillance and treatment options. This study examines the genetic diversity, resistance patterns, and transmission dynamics of 66 CRKP isolates recovered over 5 years (2015–2019) after the first case of CRKP was identified at a tertiary care hospital in Lima, Peru. Our findings reveal a shift from blaKPC-2 to blaNDM-1 as the dominant carbapenemase gene after 2017. Lineage ST45 was the most prevalent and persisted for multiple years, followed by high-risk clones ST11 and ST147. The blaNDM-1 gene was carried almost exclusively by a Tn125-like transposon, similar to the one reported in previous studies from two Peruvian hospitals. Long-read sequencing revealed nearly identical blaNDM-carrying plasmids across the four assessed lineages. A comparative analysis of 1,023 South American CRKP genomes confirmed a unique pattern in Peru, where blaNDM-1 (81.4%) outpaced blaKPC-2, which remained dominant (59.4%) elsewhere. In addition, emerging clones ST45 and ST348 found in Peru were rarely found elsewhere in South America, suggesting potential regional adaptation. In conclusion, our study provides a comprehensive picture of the intra-hospital dynamics of these emerging pathogens and provides a framework for studying their genomic diversity in the understudied South American region.IMPORTANCEThis study provides novel insights into the transmission and genetic diversity of carbapenem-resistant Klebsiella pneumoniae, a bacteria responsible for severe infections, with limited treatment options. By examining isolates recovered over 5 years at a major hospital in Lima, Peru, we demonstrated a shift from one type of resistance gene, blaKPC, to another, blaNDM, which is more challenging to treat. Our findings reveal that specific bacterial lineages carrying the blaNDM gene in a specific plasmid are emerging in Peru, including well-known high-risk strains and others rarely found elsewhere in South America. This pattern highlights an urgent need for targeted surveillance and infection control as these strains pose a significant challenge to healthcare systems. Our study provides crucial data on Klebsiella pneumoniae in Peru, contributing to broader efforts to monitor and control antibiotic-resistant infections in South America and globally.

The presence of pharmaceuticals in the aquatic environment is increasing due to their growing use for human health. Although most studies are based on short exposures to these contaminants, the present study has emerged from the need to study pharmaceuticals in aquatic organisms over a long-term exposure to understand any multi-generational chronic effects and alterations regarding habitat selection. Therefore, this study shows: (1) the ability of Daphnia magna to colonize environments contaminated with caffeine, ibuprofen and fluoxetine, and (2) the effect of these pharmaceuticals on reproduction and habitat selection (under two scenarios: with and without food) after a long-term exposure period of three generations. It was observed that caffeine shortened the time between generations and caused an increase in the number of neonates per female. The opposite was observed with ibuprofen: the time to reach the third F3 generation was double when compared to those exposed to caffeine. Fluoxetine did not alter the reproduction, nor was repellent/attractive for daphnids. In the habitat selection tests, organisms cultivated in clean water preferred the compartment with caffeine, highlighting its attractive effect. Caffeine was also attractive for daphnids in the colonization test. Apart from this, no chemical showed any attractive or repulsive effect in the absence of food during the habitat selection tests. Our findings show that the presence of some pharmaceuticals could cause alterations in distribution and habitat selection patterns, and a significant effect on the reproduction of this species. underlining the importance of studying the effects of contamination by long-term exposure.

Rickettsiae are a family of ectoparasite-borne bacteria that can produce high morbidity and mortality among humans. There are scarce data on rickettsial ecology in rural areas of the Peruvian Amazon basin, where seroprevalence has not been determined, and the identities of animals acting as reservoirs of these bacteria are not known. We conducted a cross-sectional study in Zungarococha (between 2019 and 2021), a rural community located approximately 20 km away from Iquitos city. Blood samples were collected from humans (175), dogs (123), and cats (12). Blood samples and tissues were collected from black rats (84). Finally, we collected fleas from dogs and cats (222), ticks from dogs (91), and mites from black rats (32). Blood samples from humans, dogs, cats, and black rats were analyzed by indirect immunofluorescence assays (IFA) to detect IgG antibodies against rickettsias. We screened ectoparasites and black rat tissues by real-time-PCR (qPCR). Positive ectoparasites were further assessed by PCR and DNA amplicon sequencing. Non-parametric tests were used to evaluate factors associated with being seropositive among human adults. IgG seroprevalences were 38.3 %, 58.5 %, 16.7 % and 48.1 % among humans, dogs, cats, and rats, respectively. Among humans, only male gender was statistically associated with having IgG antibodies against Rickettsia spp. (p-value=0.049, chi-square test). Different ectoparasites were identified, including Ctenocephalides felis from cats and dogs, Rhipicephalus sanguineus s.l. from dogs, and Laelaps nuttalli from black rats. Rhipicephalus sanguineus s.l. (2/91 ticks) and Ct. felis (53/56 fleas and 55/55 flea pools) were qPCR-positive for Rickettsia spp. Recovered genetic material from 53 Ct. felis was sequenced and all were identified as Rickettsia asembonensis. All tissue samples from black rats were negative by qPCR. Humans, dogs, cats, and black rats are exposed to spotted fever group rickettsiae in rural areas surrounding Iquitos. As reported in urban areas, R. asembonensis is the main Rickettsia species circulating in rural areas surrounding Iquitos and Ct. felis appears to be the main vector.

Introducción: El cáncer de mama ocupa el primer lugar en prevalencia entre las tumoraciones malignas en la mujer, y el segundo en incidencia en las mujeres entre 35 y 55 años, solo superada por el cáncer de pulmón. Objetivo: Describir el estado actual de la utilización de la biopsia por aguja gruesa (trucut) en el diagnóstico y conducta terapéutica del cáncer de mama. Método: Se realizó la búsqueda de artículos en las bases de datos SciELO, Pubmed, SCOPUS, LILACS y LIS, con los descriptores biopsia con aguja, cáncer de mama, mamografía, ultrasonografía mamaria, BRCA1, BRCA2 y c-erbB2. Se utilizó el gestor de referencias Zotero, para la obtención, descarga, organización, citación y elaboración de las referencias bibliográficas. Se utilizaron 25 referencias. Desarrollo: La biopsia por trucut presenta mayor sensibilidad con relación a la biopsia por aspiración por aguja fina, por lo que se utiliza generalmente como técnica diagnóstica en los tumores de mama. La biopsia por trucut permite obtener cilindros múltiples en pacientes con nódulos mamarios. La utilización de la mamografía como método de cribado del cáncer de mama y la implantación de programas de detección precoz de ámbito poblacional, han mejorado la detección de lesiones mamarias no palpables (LMNP). Conclusiones: La biopsia de aspiración por aguja gruesa (trucut), constituye un procedimiento diagnóstico preciso y asequible. Los resultados obtenidos son similares a los reportados con la utilización de otros procedimientos diagnósticos. The knowledge of arachnofauna from Tocantins, Brazil, is still incipient. The present communication expands the known geographical distribution of Loxosceles amazonica Gertsch, 1967 (Arachnida: Araneae: Sicariidae) in Brazilian Cerrado. The brown spider specimens were sampled in Dianópolis municipality and Cristalândia municipality both in Tocantins State, Brazil, between 2022 and 2024. Also, we describe a new variation of the epigynum of L. amazonica. Mapping the distribution of Loxosceles Heineken & Lowe, 1832 species in Brazil is crucial for public health, aiding in prevention and timely treatment of loxoscelism caused by brown spider bites.

Este artículo examina el desarrollo de la fabricación digital en Perú, centrándose en Lima Makers, el primer makerspace del país. A través de este caso, se exploran las dinámicas de socialización, identidad colaborativa y prácticas de innovación en un contexto donde la fabricación digital se cruza con la cultura artística y tecnológica local. Utilizando los conceptos de públicos recursivos, comunidades de práctica e infraestructuración, se analiza cómo Lima Makers cohesiona a sus miembros y fomenta la creatividad, aunque enfrenta desafíos para formar públicos recursivos activos en la gobernanza de la fabricación digital en el Perú. Además, se argumenta que esta dinámica local refleja un proceso de apropiación cultural y de creación de nuevas identidades tecnológicas en el sur global.

Chikungunya virus (CHIKV) has emerged as a significant public health concern due to its rapid spread and potential for causing debilitating epidemics. In Argentina, the virus has garnered attention since its introduction to the Americas in 2013, due to its growing incidence and impact in neighbouring countries. Here we present a comprehensive analysis of the spatiotemporal dynamics of CHIKV in Argentina, focusing on the evolutionary trajectory of its genetic variants. Through a combination of active surveillance, screening of historical and recent samples, and whole-genome sequencing, we traced the evolutionary history of CHIKV lineages circulating within the country. Our results reveal that two distinct genotypes circulated in Argentina: The Asian lineage during the 2016 epidemic and the ECSA lineage in 2023. This distribution reflects the dominance of particular variants across Latin America. Since 2023, the ECSA lineage has led to a surge in cases throughout the Americas, marking a significant shift. The replacement of lineages in the American region constitutes a major epidemiological event, potentially affecting the dynamics of virus transmission and the clinical outcomes in impacted populations. The spatiotemporal analysis highlights CHIKV's distribution across Argentina and underscores the significant role of human mobility, especially when considering recent epidemics in neighbouring countries such as Paraguay and Uruguay, which have facilitated the spread and introduction of the viral strain into different districts. By integrating epidemiological data with genomic insights, we elucidate the patterns of virus dissemination, highlighting key areas of transmission and potential factors contributing to its spread.

Developing STEM (Science, Technology, Engineering, Mathematics) competencies is a global priority. In response to this educational need, initiatives have been implemented mainly at the school level. However, in preschool education, the STEAM programs are more recent. Research advances orient preschool teachers to reach these competencies in school-based programs, although parental involvement has been systematically forgotten as a critical factor. This article describes the current issues on research about parental participation in STEM education in early childhood to identify advances and gaps. We selected documents published between 1995 and 2021 in the leading educational databases, identifying 11 documents explicitly related to parental involvement in STEM education in preschoolers. The results show that STEM activities can promote parental engagement, improve the value parent attribute to STEM, and positively affect STEM learning in preschoolers. Moreover, parents shape children's interests and self-efficacy about STEM and content application that can favor their children's approach to STEM. This article discusses the scarcity of research published on the connection between STEM and parental influence, despite the fundamental role of parents in early STEM education. We present practical criteria to guide the development of early STEM education in the family context and questions to guide the planning of research and intervention programs.

Abstract Objective To assess asymptomatic rates and severity of SARS‐CoV‐2 infection in people with epilepsy and their healthcare workers in a long‐term care facility which had implemented weekly surveillance testing between April 2020 and June 2022. Methods Questionnaires focused on objective and subjective COVID‐19 symptoms for people with epilepsy residing in and their healthcare workers at the Chalfont Centre for Epilepsy in June 2022. Demographic information, comorbidities, and seizure frequency were gathered from medical records. We also collected responses on objective and subjective COVID‐19 symptoms from healthcare workers who participated in a prospective study assessing the reaction to COVID‐19 vaccinations (SAFER). Results Fifty‐five out of 89 (62%) residents tested positive at least once on weekly PCR testing for SARS‐CoV‐2 during the period of interest; 20 of those (37%) were asymptomatic. In comparison, of those 63 healthcare workers who tested positive at least once on weekly testing during the same period, only four (6%) were asymptomatic. Of the 159 healthcare workers who also participated in the SAFER study, 41 tested positive at least once, and seven (17%) were completely asymptomatic during infection with SARS‐CoV‐2. Significance People with epilepsy living in a long‐term care facility were more likely to present with asymptomatic SARS‐CoV‐2 infections than healthcare workers at the same facility. Despite possible bias in the reporting of subjective symptoms due to management‐by‐proxy, there is no evidence that vulnerable people living in an epilepsy long‐term care facility showed reduced resilience towards infections. Plain Language Summary People with epilepsy living in care home facilities had a surprisingly high degree of asymptomatic infections with SARS‐CoV‐2. Very few residents had severe or fatal outcomes. This is in stark contrast to the widely reported bad outcomes for people without epilepsy in other care homes. People with epilepsy reported significantly less symptoms than their healthcare workers. No changes in seizure frequency during or after infection were observed.

Background: Gasoline, a complex mixture of volatile organic compounds is classified as possibly carcinogenic to humans. Gasoline station attendants, consistently exposed to its hazardous components, may face genotoxic effects. This study aimed to assess the influence of varying work shift durations on DNA damage in gasoline station attendants. Methods: Ninety individuals from three locations in southern México were studied. Peripheral blood mononuclear cells (PBMCs) were isolated, and DNA damage was assessed using the comet assay. Demographic, occupational, and lifestyle data were collected. Statistical analyses included t-tests, ANOVA, and Pearson correlation. Results: Significant differences in DNA damage parameters were observed between exposed and unexposed groups. The impact of tobacco, alcohol, and exercise on DNA damage was negligible. Extended work shifts (12 and 24 hours) showed heightened DNA damage compared to 8-hour shifts and the unexposed group. A novel finding revealed a modest but significant correlation between DNA damage and job seniority. Conclusion: The study highlights the intricate relationship between occupational exposure to gasoline components, DNA damage, and work shift lengths. Extended shifts correlate with heightened genotoxic effects, emphasizing the importance of personalized safety measures. The significant correlation between DNA damage and job seniority introduces occupational longevity as a determinant in the genetic health of gasoline station attendants. This discovery has implications for implementing targeted interventions and preventive strategies to safeguard workers' genetic integrity throughout their years of service. The study calls for further exploration of unconsidered factors in understanding the multifactorial nature of DNA damage in this occupational setting.
