Abstract There is currently a strong interest in wood and the demand for this material is expected to grow significantly worldwide. Consequently, market demand for wood from fruit orchards is growing as an additional source of supply. For these reasons, several studies were conducted to evaluate the properties of wood derived by orchard pruning or dismantling. Despite the abundance of raw materials from pruning, the mechanical properties of olive wood have not been studied completely by the scientific community, so the woody material obtained is usually considered waste or firewood. In fact, there is still a lack of knowledge about olive wood characteristics and considering the valuable role of olive species in the Mediterranean area, the current study aimed to determine and compare the physical and mechanical properties of branches and stem wood of two olive tree cultivars, “Sinopolese” and “Ottobratica”, to provide information on this wood species. These two olive cultivars are commonly cultivated in the Calabria region (Southern Italy) for the extraction of oil from drupes, but large masses of wood are derived from their pruning. For the choice of parts, the stem was considered to become a branch when, above each branch intersection, it changed in diameter and direction of growth. The branches with a diameter lower than 20 cm were excluded. The tests conducted for this purpose were: Roughness profile; Impact wave; Vibrational analysis; Static modulus of elasticity; Bending strength; Abrasion resistance; Static hardness; Footprints; Compression strength; Screw withdrawal resistance parallel to grain; Screw withdrawal resistance perpendicular to grain. A MANOVA analysis was conducted between the Cultivar-Tree part and the physical and mechanical properties. The results showed some differences between the two cultivars principally related to mechanical properties such as moduli of elasticity and rupture, footprint, compression strength, and screw withdrawal resistances, where in general, the “Sinopolese” cultivar showed higher values than the “Ottobratica”. Between the tree parts (stem and branches) within the same cultivars, branches demonstrated higher results in the majority of the tests, but highlighting statistical differences only in terms of static modulus of elasticity, bending strength, static hardness and screw withdrawal resistances depending on the cultivar. This result suggests that the branch wood, with its characteristics, could be valorized in the commercial utilization representing a valid opportunity for the local rural economy, even considering the conspicuous amount of wood obtained from olive pruning activity. Expanding knowledge about olive wood in terms of physical and mechanical characteristics could increase its use in multiple sectors and ensure a more aware use of the application of the wood resources by supporting the decision on its best end use.

The serological support for early diagnosis and differential diagnosis of inflammatory bowel diseases (IBDs) is actually very limited. In this study, we evaluated the performance of a promising multiparametric method including either well-established and newly developed biomarkers. We conducted a multicenter cross-sectional study at the Gastroenterology Units of Udine (Italy), Rijeka (Croatia) and Belgrade (Serbia). Sera was collected from IBD patients, and autoantibody profiles were determined using a mosaic cell and tissue-based indirect immunofluorescence (IIF) method simultaneously investigating anti-saccharomyces cerevisiae antibodies (ASCAs), anti-atypical perinuclear neutrophilic antibodies (P-ANCAs), anti-pancreatic antigens antibodies (PABs) and anti-goblet cells antibodies (GAB). The study finally enrolled 156 patients with IBD: 100 affected by Crohn’s disease (CD) and 56 by ulcerative colitis (UC). Twenty age-sex matched blood donors (BDs) were included as controls. PAB (anti-CUZD1 and/or anti-GP2 antibodies) were present in 24 CD patients versus none of the UC patients or BDs (24% sensitivity, 100% specificity). As regards CD patients, combined positivity of PAB and ASCA (sensitivity 84%, specificity 71.4%) performed better than ASCA alone. Colon involvement (87.5% vs. 60.5%; p = 0.014), deep mucosal lesions (58.3% vs. 25.0%; p = 0.002) and need for biologic therapies (79.2% vs. 46.1%; p = 0.005) were significantly more prevalent in PAB-positive than in PAB-negative CD patients. Multivariate analysis identified PAB positivity (OR = 3.67; 95%CI = 1.29–10.46) and anti-CUZD1 in particular (OR = 3.54; 95%CI = 1.08–11.63) as significant risk factors for deep mucosal lesion development in CD. A multiparametric diagnostic approach appears very useful to better characterize IBD patients. PABs, whether isolated or combined with other autoantibodies, may support differential diagnosis but above all facilitate the selection of CD patients at risk for more severe disease.

The use of agroforestry biomass provides several advantages, both from an environmental point of view, in terms of the mitigation of global warming, and in terms of a circular economy for agricultural or agroforestry companies that reuse pruning residues as a source of energy. However, even if the use of energy pellets resulting from the pruning residues of various agroforestry species has excellent potential for the valorization of agricultural by-products, the physicochemical characteristics of these pellets have been scarcely studied by the scientific community. In this context, this study aims to assess the valorization potential of various lignocellulosic material residues produced during agroforestry activities. The objectives of the study include evaluating the chemical and physical characteristics of pellets produced with different mixtures of agroforestry biomass (olive, citrus, black locust, poplar, paulownia, etc.) in order to determine the optimal pellet blend from an energy and physicochemical perspective. The results of this study demonstrate that this comprehensive analysis provides valuable information on the optimization of biomass mixtures for better energy valorization, addressing both compositional and combustion-related challenges. In fact, it is observed that the addition of citrus and olive biomass to the various mixtures increases their energy potential. Furthermore, all of the pellets analyzed are found to possess an adequate and useful durability index (PDI) for their handling during storage and transport operations. This study demonstrates that olive and citrus pruning residues can be used to improve biomasses that have poor suitability in energetic, physical, and chemical terms. Further studies could be useful to understand which specific interaction mechanisms have an influence on emissions in order to optimize mixtures using different biomass sources for sustainable energy production.

Introduction: Schizophrenia (SCZ) and autism spectrum disorder (ASD) are neurodevelopmental diseases characterized by different psychopathological manifestations and divergent clinical trajectories. Various alterations at glutamatergic synapses have been reported in both disorders, including abnormal NMDA and metabotropic receptor signaling. Methods: We conducted a bicentric study to assess the blood serum levels of NMDA receptors-related glutamatergic amino acids and their precursors, including L-glutamate, L-glutamine, D-aspartate, L-aspartate, L-asparagine, Dserine, L-serine and glycine, in ASD, SCZ patients and their respective control subjects. Specifically, the SCZ patients were subdivided into treatment-resistant and non-treatment-resistant SCZ patients, based on their responsivity to conventional antipsychotics. Results: D-serine and D-aspartate serum reductions were found in SCZ patients compared to controls. Conversely, no significant differences between cases and controls were found in amino acid concentrations in the two ASD cohorts analyzed. Discussion: This result further encourages future research to evaluate the predictive role of selected D-amino acids as peripheral markers for SCZ pathophysiology and diagnosis. Background and Objectives. Retinitis pigmentosa (RP) is the most common inherited rod–cone dystrophy (RCD), resulting in nyctalopia, progressive visual field, and visual acuity decay in the late stages. The autosomal dominant form (ADRP) accounts for about 20% of RPs. Among the over 30 genes found to date related to ADRP, RP1 pathogenic variants have been identified in 5–10% of cases. In a cohort of RCD patients from the Palermo province on the island of Sicily, we identified a prevalent nonsense variant in RP1, which was associated with ADRP. The objective of our study was to analyse the clinical and molecular data of this patient cohort and to evaluate the potential presence of a founder effect. Materials and Methods. From 2005 to January 2023, 84 probands originating from Western Sicily (Italy) with a diagnosis of RCD or RP and their relatives underwent deep phenotyping, which was performed in various Italian clinical institutions. Molecular characterisation of patients and familial segregation of pathogenic variants were carried out in different laboratories using Sanger and/or next-generation sequencing (NGS). Results. Among 84 probands with RCD/RP, we found 28 heterozygotes for the RP1 variant c.2219C>G, p.Ser740* ((NM_006269.2)*, which was therefore significantly prevalent in this patient cohort. After a careful interview process, we ascertained that some of these patients shared the same pedigree. Therefore, we were ultimately able to define 20 independent family groups with no traceable consanguinity. Lastly, analysis of clinical data showed, in our patients, that the p.Ser740* nonsense variant was often associated with a late-onset and relatively mild phenotype. Conclusions. The high prevalence of the p.Ser740* variant in ADRP patients from Western Sicily suggests the presence of a founder effect, which has useful implications for the molecular diagnosis of RCD in patients coming from this Italian region. This variant can be primarily searched for in RP-affected subjects displaying compatible modes of transmission and phenotypes, with an advantage in terms of the required costs and time for analysis. Moreover, given its high prevalence, the RP1 p.Ser740* variant could represent a potential candidate for the development of therapeutic strategies based on gene editing or translational read-through therapy for suppression of nonsense variants. We performed a pilot study to evaluate the use of MRI delta texture analysis (D-TA) as a methodological item able to predict the frequency of complete pathological responses and, consequently, the outcome of patients with locally advanced rectal cancer addressed to neoadjuvant chemoradiotherapy (C-RT) and subsequently, to radical surgery. In particular, we carried out a retrospective analysis including 100 patients with locally advanced rectal adenocarcinoma who received C-RT and then radical surgery in three different oncological institutions between January 2013 and December 2019. Our experimental design was focused on the evaluation of the gross tumor volume (GTV) at baseline and after C-RT by means of MRI, which was contoured on T2, DWI, and ADC sequences. Multiple texture parameters were extracted by using a LifeX Software, while D-TA was calculated as percentage of variations in the two time points. Both univariate and multivariate analysis (logistic regression) were, therefore, carried out in order to correlate the above-mentioned TA parameters with the frequency of pathological responses in the examined patients’ population focusing on the detection of complete pathological response (pCR, with no viable cancer cells: TRG 1) as main statistical endpoint. ROC curves were performed on three different datasets considering that on the 21 patients, only 21% achieved an actual pCR. In our training dataset series, pCR frequency significantly correlated with ADC GLCM-Entropy only, when univariate and binary logistic analysis were performed (AUC for pCR was 0.87). A confirmative binary logistic regression analysis was then repeated in the two remaining validation datasets (AUC for pCR was 0.92 and 0.88, respectively). Overall, these results support the hypothesis that D-TA may have a significant predictive value in detecting the occurrence of pCR in our patient series. If confirmed in prospective and multicenter trials, these results may have a critical role in the selection of patients with locally advanced rectal cancer who may benefit form radical surgery after neoadjuvant chemoradiotherapy.
