This study investigated, for the first time, the potential role of Astragalus polysaccharide (APS) and APS nanoparticles on testicular blood flow (TBF) and semen quality in Ossimi rams. Fifteen sexually mature Ossimi rams were allocated randomly into two treated groups that orally administered either nano APS (2 g/ram/day; n = 5) or APS (20 g/ram/day; n = 5) for four weeks and a control group (n = 5). The nano-emulsion was prepared by adding corn oil to the APS solution, sonicated, centrifuged at 20,000 rpm, then washed 3–4 times, and vacuum dried overnight at 40 °C. The antioxidant activity of APS and APS nano-emulsion was evaluated in vitro. Blood collection and ultrasonographic assessment of the testes and supratesticular arteries (STAs) were conducted immediately before treatment (W0) and once weekly for 6 successive weeks after APS and nano APS administration (W1-W6). Serum testosterone (T) and estradiol (E2) concentrations were determined by ELISA kits, while nitric oxide (NO) and total antioxidant capacity (TAC) were measured spectrophotometrically. Moreover, semen collection and evaluation of some sperm parameters were performed once a week. Results revealed decreases (P < 0.05) in the Doppler indices (resistive index; RI, pulsatility index; PI, and systolic/diastolic; S/D) of the testicular arteries at most time points of the study in the nano APS and APS groups. Pixel intensity (PIX) and integrated density (IND) of testicular parenchyma were significantly reduced (P ˂ 0.05) in the treated groups compared to the control one. T, E2, NO, and TAC concentrations increased (P < 0.05) in the treated groups compared to the control one. Increases (P < 0.05) were noticed in the mass motility, progressive motility %, live sperm %, and membrane integrity % in nano APS and APS groups, compared to the control. Rams in the nano APS group had significantly higher (P < 0.05) sperm cell concentration than the control one. In conclusion, this study extrapolated that the dietary administration of APS and its nanoparticles can improve TBF, testicular echotexture, sperm characteristics, and the concentration of serum T, E2, NO, and TAC with a more significant effect in the APS nanoparticles compared with APS. So, it could be recommended as a dietary supplementation (2 g/ram/day) for enhancing the reproductive performance of rams. [ABSTRACT FROM AUTHOR]

Simple Summary: The expression of cytokines in livestock remains largely unexplored. This study aimed to investigate cytokine expression in a goat model of atopic dermatitis (AD). By establishing this novel goat model, we aimed to more closely replicate the pathophysiology of dermatitis in humans and larger animals, offering a more relevant alternative to traditional mouse models. The goat model, being more similar in size and physiology to large animals and humans, exhibited cytokine expression patterns akin to those seen in previous mouse studies, as confirmed by histopathological analysis and immunostaining. These preliminary findings open new avenues for further research on AD in large animals and humans, underscoring the potential of the goat model for broader dermatological studies. Atopic dermatitis (AD) remains a challenging condition, necessitating effective models for investigation and therapeutic exploration. In this study, we employed a goat skin model to assess the immune response triggered by topical oxazolone (OX) application. The OX simulates a Th2-driven immune reaction typified by elevated Th2 cytokine (IL-4 and IL-13) expression and the infiltration of mononuclear cells magnified by the presence of CD3+ and CD4+ T-cells in the epidermal and dermal layers. Additionally, the application of pomegranate peel extract (PPE) demonstrated a mitigating effect on OX-induced skin alterations, suggesting potential therapeutic benefits. This study underscores the value of goats as a reliable and accessible model for studying AD in humans and/or large animals. Higher body fatness adversely affects metabolic and hormonal homeostasis. The present work aimed to evaluate the association between body condition score (BCS) and haemodynamic pattern and echogenic appearence of the testes as well as nitric oxide (NO) levels and total antioxidant capacity (TAC). For that, fifteen Ossimi rams were blocked according to their BCS into a lower BCS group (L-BCS:2-2.5; n = 5), medium BCS group (M-BCS:3-3.5; n = 5), and higher BCS group (H-BCS:4-4.5; n = 5). Rams were examined for testicular haemodynamics (TH; Doppler ultrasonography), testicular echotexture (TE; B-mode image software analysis), and serum levels of NO and TAC (colorimetric). Results are presented as means ± standard error of the mean. There was a significant (P < 0.05) difference in the resistive index and pulsatility index means among the groups under experimentation, being the least in the L-BCS group (0.43 ± 0.02 and 0.57 ± 0.04, respectively) compared to the M-BCS (0.53 ± 0.03 and 0.77 ± 0.03, respectively) and H-BCS rams (0.57 ± 0.01 and 0.86 ± 0.03, respectively). Among blood flow velocity measurements [peak systolic, enddiastolic (EDV), and time-average maximum], only EDV showed significant (P < 0.05) higher values in the L-BCS group (17.06 ± 1.03 cm/s) compared to M-BCS (12.58 ± 0.67 cm/s) and H-BCS (12.51 ± 0.61 cm/s) groups. Regarding the TE results, there were no significant differences among the examined groups. There were significant differences (P < 0.01) in the concentrations of TAC and NO among the groups under experimentation, in which the L-BCS rams had the highest levels of TAC and NO in their sera (0.90 ± 0.05 mM/L and 62.06 ± 2.72 µM/L, respectively) than the M-BCS (0.058 ± 0.05 mM/L and 47.89 ± 1.49 µM/L, respectively) and H-BCS rams (0.45 ± 0.03 mM/L and 49.93 ± 3.63 µM/L, respectively). Purpose: This study aims to develop and validate predictive nomograms for prostate cancer (PCa) and clinically significant PCa (csPCa) in prostate‐specific antigen (PSA) gray zone of 4−10 ng/mL, utilizing transrectal ultrasound five‐grade scoring system (TRUS‐5), prostate imaging reporting and data system (PI‐RADS) score based on multiparametric magnetic resonance imaging (mpMRI) and clinical parameters. Method: A retrospective analysis was conducted on a cohort of 383 patients with elevated PSA levels in the gray zone who underwent initial TRUS‐guided 12‐core prostate biopsy between January 2018 and December 2020. Multivariate logistic regression analyses were utilized to identify independent predictors for PCa and csPCa and to establish predictive nomograms. The diagnostic performance, calibration ability, and clinical usefulness of each model were assessed through receiver operating characteristic (ROC) curve analysis, calibration curve analysis, and decision curve analysis (DCA), respectively. Results: Two diagnostic nomograms were developed based on five risk factors (age, free/total PSA [(f/t) PSA] ratio, prostate volume [PV], TRUS‐5, and PI‐RADS score) to predict PCa and five risk factors (PSA, (f/t) PSA, PV, TRUS‐5, and PI‐RADS score) for csPCa of PSA gray zone patients. The TRUS‐5/PI‐RADS combined model demonstrated higher area under the curves (AUCs) (0.865 for PCa and 0.910 for csPCa) compared to the models based on PI‐RADS score or TRUS‐5 (p < 0.05), as well as the greater net benefits across a wide range of threshold probabilities for PSA gray zone patients. Conclusion: The current study presented valuable TRUS‐5/PI‐RADS combined nomograms for predicting PCa or csPCa in patients within the PSA gray zone, which may serve as useful tools for the clinical management of PCa and csPCa. This study reports the facile hydrothermal synthesis of pure Bi2WO6 and Bi2WO6\MWCNTs nanocomposite at specific molar ratio 1:2.5 of Bi2WO6:MWCNTs and elucidates their role in modulating the NLRP3 inflammasome pathway via autophagy induction. Comprehensive characterization techniques, including XRD, Raman, UV.Vis PL,FESEM,EDS and TEM, revealed the successful incorporation of MWCNTs into the Bi2WO6 structures, leading to enhanced crystattlinity, reduced band gap energy (2.4 eV) suppressed charge carrier recombination and mitigated nanoparticles aggregation. Notably, the reduced band gap facikitaed improved visible light harvesting, a crucial attribute for photocatalytic applications. Significantly, the nanocompsoite exhibited a remarkable capacity to augment autophagy in bone marrow-derived macrophages (BMDMs), consequently down-regulating the NLRP3 inflammasom activation and IL-1β secretion upon LPS and ATP stimulation. Immunofluorescence assays unveiled increased co-localization of LC3 and NLRP3, suggestion enhanced targeting of NLRP3 by autophagy. Inhibition of autophagy by 3-MA reversed these effects, confirming the pivotal role of autophagy induction. Furthermore, the nanocomposite attenuated caspase-1 activation and ASC oligomerzation, thereby impeding inflammasome assembly. Collectively, these findings underscore the potential of Bi2WO6\MWCNTs nanocompsite as a multifaceted therapeutic platform, levering its tailored optoelectronic properties and sbility to modulate the NLRP3 infalmmasome via autophagy augmentation. This study aimed to determine the effects of Coenzyme Q10 (CoQ10) in the freezing medium on functional and oxidative stress parameters and in vitro fertilization (IVF) rate of buffalo sperm. Collected samples were relocated to the laboratory for initial evaluation, gentle dilution in extenders, cooling (4°C, 2 h), equilibration (4°C, 4 h), packaging (straws, 0.5 mL), programmable freezing, and thawing (37°C, 30 s). Statistical analysis depicted that adding CoQ10 (100 μM) in a freezing medium caused a significant augmentation in total motility (%), average path, and straight‐line velocities (μm/sec) of buffalo sperm than control. Adding CoQ10 (100 μM) improved sperm progressive motility, rapid velocity, and functional parameters (%) compared to the control and 10 μM of CoQ10. Moreover, CoQ10 in a freezing medium caused a significant augmentation in seminal plasma catalase (U/mL) and glutathione reductase (GSH; nmol/109) at 100 μM than control and other treatments. CoQ10 inclusion (100 μM) ameliorates seminal plasma superoxide dismutase (U/mL), glutathione‐S‐transferase (GST; nmol/mL/min) fructose (μg/mL), and ATP (nmol/million) than control. Furthermore, CoQ10 at 100 μM improved seminal plasma glutathione peroxidase (μM) levels than control, 10 μM, and 20 μM. Lastly, hydrogen peroxide (H2O2; nM) production was significantly lower at 100 μM than at control and 10 μM. CoQ10 (100 μM) caused a significant augmentation in the un‐capacitated pattern followed by a reduction in the capacitated pattern, and apoptosis‐like changes (%) than control, and other treatments, whereas viability was increased than control and other treatments. CoQ10 (100 μM) significantly improved the IVF rate in comparison with control, CoQ10 at 10 μM, and 20 μM groups. In conclusion, the addition of CoQ10 (100 μM) in the freezing medium can improve the quality and in vitro fertility of post‐thawed buffalo semen via its antioxidative effect. Further studies are needed to evaluate the effect of CoQ10 on the in vivo fertility of buffalo bull semen. [ABSTRACT FROM AUTHOR]
