4. A diffuse tidal dwarf galaxy destined to fade out as a 'dark galaxy'

Abstract

We have explored the properties of a peculiar object detected in deep optical imaging and located at the tip of an HI tail emerging from Hickson Compact Group 16. Using multiband photometry from infrared to ultraviolet, we were able to constrain its stellar age to 58$^{+22}_{-9}$ Myr with a rather high metallicity of [Fe/H] = $-$0.16$^{+0.43}_{-0.41}$ for its stellar mass of M$_\star$ = 4.2$\times$10$^6$ M$_\odot$, a typical signature of tidal dwarf galaxies. The structural properties of this object are similar to those of diffuse galaxies, with a round and featureless morphology, a large effective radius (r$_{eff}$ = 1.5 kpc), and a low surface brightness (<$\mu_{g}$>$_{eff}$ = 25.6 mag arcsec$^{-2}$). Assuming that the object is dynamically stable and able to survive in the future, its fading in time via the aging of its stellar component will make it undetectable in optical observations in just $\sim$2 Gyr of evolution, even in the deepest current or future optical surveys. Its high HI mass, M(HI) = 3.9$\times$10$^8$ M$_\odot$, and future undetectable stellar component will make the object match the observational properties of dark galaxies, that is, dark matter halos that failed to turn gas into stars. Our work presents further observational evidence of the feasibility of HI tidal features becoming fake dark galaxies; it also shows the impact of stellar fading, particularly in high metallicity systems such as tidal dwarfs, in hiding aged stellar components beyond detection limits in optical observations., Comment: Accepted for publication in A&A Letters

Abstract

Hickson Compact Group (HCG) 16 is a prototypical compact group of galaxies, apparently in an intermediate stage of evolution, where its galaxies are losing gas to the intra-group medium (IGrM). The wide variety of ongoing process in HCG 16 make it an ideal case study for exploring which processes are likely to dominate the late stages of evolution in compact groups. In order to build a coherent picture of the evolution of this group we make use of the multi-wavelength data available, but focus particularly on HI as a tracer of interactions and evolutionary phase. We reprocess archival VLA L-band data of HCG 16 using the multi-scale CLEAN algorithm to accurately recover diffuse features. Despite the clear disruption of the HI component of HCG 16 we find that it is not globally HI deficient, even though HCG 16a and b have lost the majority of their HI and almost 50% of the group's HI is in the IGrM, including a 160 kpc long tail extending towards the SE. This study indicates that in the recent past (~1 Gyr) galaxies HCG 16a and b likely underwent major interactions that unbound gas without triggering significant star formation. This gas was then swept away by a high speed, close encounter with NGC 848. The starburst events HCG 16c and d have triggered galactic winds which, in the case of HCG 16d, appears to have disrupted its HI reservoir. The tidal features still connected to these galaxies indicate that more HI will soon be lost to the IGrM, while that which remains in the discs will likely be consumed by star formation episodes. This is expected to result in a collection of gas-poor galaxies embedded in a diffuse HI structure, which will gradually be evaporated by the UV background, resembling the final stage of the evolutionary model of compact groups. Abstract

Using images from Sloan Digital Sky Survey Data Release 8, we have re-examined the morphology of 719 galaxies from the Analysis of the interstellar Medium in Isolated GAlaxies (AMIGA) project, a sample consisting of the most isolated galaxies that have yet been identified. The goal is to further improve the classifications of these galaxies by examining them in the context of the Comprehensive de Vaucouleurs revised Hubble-Sandage (CVRHS) system, which includes recognition of features that go beyond the original de Vaucouleurs point of view. Our results confirm previous findings that isolated galaxies are found across the complete revised Hubble sequence, with intermediate to late-type (Sb-Sc) spirals being relatively more common. Elmegreen Arm Classifications are also presented, and show that more than 50% of the 514 spirals in the sample for which an arm class could be judged are grand design (AC 8,9,12). The visual bar fraction for the sample is ~50%, but only 16% are classified as strongly-barred (SB). The dominant family classification is SA (nonbarred), the dominant inner variety classification is (s) (pure spiral), and the dominant outer variety classification is no outer ring, pseudoring, or lens. The Kolmogorov-Smirnov test is used to check for potential biases in the morphological interpretations, and for any possible relation between rings, bars, and arm classes with local environment and far-infrared excess. Abstract

We synthesized an anthracene derivative with solvatochromic properties to be used as a molecular probe for membrane dynamics and supramolecular organization. A nine carbon atom acyl chain and a dimethylamino substitution were introduced at positions 2 and 6 of the anthracene ring, respectively. This derivative, 2-nonanoyl-6-(dimethylamino)anthracene (termed CAPRYDAA), is a molecular probe designed to mimic the well-known membrane probe LAURDAN's location and response in the lipid membranes. Due to the larger distance between the electron donor and acceptor groups, its absorption and emission bands are red-shifted according to the polarity of the media. The photophysical behavior of CAPRYDAA was measured in homogeneous media, synthetic bilayer and cells, both in a cuvette and in a fluorescence microscope, using one and two-photon excitation. Our results show a comparable physicochemical behavior of CAPRYDAA with LAURDAN, but with the advantage of using visible light (488 nm) as an excitation source. CAPRYDAA was also excitable by two-photon laser sources, making it easy to combine CAPRYDAA with either blue or red emission probes. In GUVs or cells, CAPRYDAA can discriminate the lipid phases and liquid-liquid phase heterogeneity. This new membrane probe shows the bathochromic properties of the PRODAN-based probes designed by Weber, overcoming the need for UV or two-photon excitation and facilitating the studies on the membrane properties using regular confocal microscopes.

Abstract

We present the largest catalogue of HI single dish observations of isolated galaxies to date and the corresponding HI scaling relations, as part of the multi-wavelength project AMIGA (Analysis of the interstellar Medium in Isolated GAlaxies). Despite numerous studies of the HI content of galaxies, no revision has been made for the most isolated L* galaxies since 1984. In total we have measurements or constraints on the HI masses of 844 galaxies from the Catalogue of Isolated Galaxies (CIG), obtained with our own observations at Arecibo, Effelsberg, Nancay and GBT, and spectra from the literature. Cuts are made to this sample to ensure isolation and a high level of completeness. We then fit HI scaling relations based on luminosity, optical diameter and morphology. Our regression model incorporates all the data, including upper limits, and accounts for uncertainties in both variables, as well as distance uncertainties. The scaling relation of HI mass with optical diameter is in good agreement with that of Haynes & Giovanelli 1984, but our relation with luminosity is considerably steeper. This is attributed to the large uncertainties in the luminosities, which introduce a bias when using OLS regression (used previously), and the different morphology distributions of the samples. We find that the main effect of morphology on the relations is to increase the intercept and flatten the slope towards later types. These trends were not evident in previous works due to the small number of detected early-type galaxies. The HI scaling relations of the AMIGA sample define an up-to-date metric of the HI content of almost "nurture free" galaxies. These relations allow the expected HI mass, in the absence of interactions, of a galaxy to be predicted to within 0.25 dex, and are thus suitable for use as statistical measures of the impact of interactions on the neutral gas content of galaxies. Abstract

Thrombotic microangiopathy (TMA), characterized by microangiopathic hemolytic anemia, thrombocytopenia, and multisystem organ dysfunction, is a life‐threatening disease. Patients with TMA who do not exhibit a severe ADAMTS‐13 deficiency (defined as a disintegrin‐like and metalloprotease with thrombospondin type 1 motif no. 13 activity ≥10%: TMA‐13n) continue to experience elevated mortality rates. This study explores the prognostic indicators for augmented mortality risk or necessitating chronic renal replacement therapy (composite outcome: CO) in TMA‐13n patients. We included 42 TMA‐13n patients from January 2008 to May 2018. Median age of 41 years and 60% were female. At presentation, 62% required dialysis, and 57% warranted intensive care unit admission. CO was observed in 45% of patients, including a 9‐patient mortality subset. Multivariate logistic regression revealed three independent prognostic factors for CO: early administration of eculizumab (median time from hospitalization to eculizumab initiation: 5 days, range 0–19 days; odds ratio [OR], 0.14; 95% confidence interval [CI], 0.02–0.94), presence of neuroradiological lesions (OR, 6.67; 95% CI, 1.12–39.80), and a PLASMIC score ≤4 (OR, 7.39; 95% CI, 1.18–46.11). In conclusion, TMA‐13n patients exhibit a heightened risk of CO in the presence of low PLASMIC scores and neuroradiological lesions, while early eculizumab therapy was the only protective factor. Abstract

The basic properties of galaxies can be affected by both nature (internal processes) or nurture (interactions and effects of environment). Deconvolving the two effects is an important current effort in astrophysics. Observed properties of a sample of isolated galaxies should be largely the result of internal (natural) evolution. It follows that nurture-induced galaxy evolution can only be understood through comparative study of galaxies in different environments. We take a first look at SDSS (g-r) colors of galaxies in the AMIGA sample involving many of the most isolated galaxies in the local Universe. This leads us to simultaneously consider the pitfalls of using automated SDSS colors. We focus on median values for the principal morphological subtypes found in the AMIGA sample (E/S0 and Sb-Sc) and compare them with equivalent measures obtained for galaxies in denser environments. We find a weak tendency for AMIGA spiral galaxies to be redder than objects in close pairs. We find no clear difference when we compare with galaxies in other (e.g. group) environments. However, the (g-r) color of isolated galaxies shows a Gaussian distribution as might be expected assuming nurture-free evolution. We find a smaller median absolute deviation in colors for isolated galaxies compared to both wide and close pairs. The majority of the deviation on median colors for spiral subtypes is caused by a color-luminosity correlation. Surprisingly isolated and non-isolated early-type galaxies show similar (g-r). We see little evidence for a green valley in our sample with most spirals redder than (g-r)=0.7 having spurious colors. The redder colors of AMIGA spirals and lower color dispersions for AMIGA subtypes -compared with close pairs- is likely due to a more passive star formation in very isolated galaxies., Comment: Accepted for publication in A&A. 9 pages, 7 Figures, and 2 tables, one only available online

34. Risk of cardiovascular involvement in pediatric patients with X-linked hypophosphatemia

Abstract

Objective To find out if cardiovascular alterations are present in pediatric patients with X-linked hypophosphatemia (XLH). Study design Multicentre prospective clinical study on pediatric patients included in the RenalTube database (http://www.renaltube.com http://www.renaltube.com ) with genetically confirmed diagnosis of XLH by mutations in the PHEX gene. The study's protocol consisted of biochemical work-up, 24-h ambulatory blood pressure monitoring (ABPM), carotid ultrasonography, and echocardiogram. All patients were on chronic treatment with phosphate supplements and 1-hydroxy vitamin D metabolites. Results Twenty-four patients (17 females, from 1 to 17 years of age) were studied. Serum concentrations (X ± SD) of phosphate and intact parathyroid hormone were 2.66 ± 0.60 mg/dl and 58.3 ± 26.8 pg/ml, respectively. Serum fibroblast growth factor 23 (FGF23) concentration was 278.18 ± 294.45 pg/ml (normal < 60 pg/ml). Abnormally high carotid intima media thickness was found in one patient, who was obese and hypertensive as revealed by ABPM, which disclosed arterial hypertension in two other patients. Z scores for echocardiographic interventricular septum end diastole and left ventricular posterior wall end diastole were + 0.77 ± 0.77 and + 0.94 ± 0.86, respectively. Left ventricular mass index (LVMI) was 44.93 ± 19.18 g/m.sup.2.7, and four patients, in addition to the obese one, had values greater than 51 g/m.sup.2.7, indicative of left ventricular hypertrophy. There was no correlation between these echocardiographic parameters and serum FGF23 concentrations. Conclusions XLH pediatric patients receiving conventional treatment have echocardiographic measurements of ventricular mass within normal reference values, but above the mean, and 18% have LVMI suggestive of left ventricular hypertrophy without correlation with serum FGF23 concentrations. Abstract

Changes in the cholesterol (Chol) content of biological membranes are known to alter the physicochemical properties of the lipid lamella and consequently the function of membrane-associated enzymes. To characterize these changes, we used steady-state and time resolved fluorescence spectroscopy and two photon-excitation microscopy techniques. The membrane systems were chosen according to the techniques that were used: large unilamellar vesicles (LUVs) for cuvette and giant unilamellar vesicles (GUVs) for microscopy measurements; they were prepared from dipalmitoyl phosphatidylcholine (DPPC) and dioctadecyl phosphatidylcholine (DOPC) in mixtures that are well known to form lipid domains. Two fluorescent probes, which insert into different regions of the bilayer, were selected: 1,6-diphenyl-1,3,5-hexatriene (DPH) was located at the deep hydrophobic core of the acyl chain regions and 2-dimethylamino-6-lauroylnaphthalene (Laurdan) at the hydrophilic-hydrophobic membrane interface. Our spectroscopy results show that (i) the changes induced by cholesterol in the deep hydrophobic phospholipid acyl chain domain are different from the ones observed in the superficial region of the hydrophilic-hydrophobic interface, and these changes depend on the state of the lamella and (ii) the incorporation of cholesterol into the lamella induces an increase in the orientation dynamics in the deep region of the phospholipid acyl chains with a corresponding decrease in the orientation at the region close to the polar lipid headgroups. The microscopy data from DOPC/DPPC/Chol GUVs using Laurdan generalized polarization (Laurdan GP) suggest that a high cholesterol content in the bilayer weakens the stability of the water hydrogen bond network and hence the stability of the liquid-ordered phase (Lo).

Abstract

Cellular membranes are heterogeneous in composition, and the prevailing theory holds that the structures responsible for this heterogeneity in vivo are small structures (10-200 nm), sterol- and sphingolipid-enriched, of different sizes, highly dynamic denominated rafts. Rafts are postulated to be platforms, which by sequestering different membrane components can compartmentalize cellular processes and regulate signaling pathways. Despite an enormous effort in this area, the existence of these domains is still under debate due to the characteristics of the structures itself: small in size and highly mobile, which from the technical point of view implies using techniques with high spatial and temporal resolution. In this report we measured rapid fluctuations of the normalized ratio of the emission intensity at two wavelengths of Laurdan, a membrane fluorescent dye sensitive to local membrane packing. We observed generalized polarization fluctuations in the plasma membrane of intact rabbit erythrocytes and Chinese hamster ovary cells that can be explained by the existence of tightly packed micro-domains moving in a more fluid background phase. These structures, which display different lipid packing, have different sizes; they are found in the same cell and in the entire cell population. The small size and characteristic high lipid packing indicate that these micro-domains have properties that have been proposed for lipid rafts.

Abstract

Resumen: La yerba mate (Ilex paraguariensis) es una hierba ampliamente consumida que se originó en Paraguay, pero que crece en la región entre el sur de Brasil, el norte de Argentina, Paraguay y Uruguay. Se informa que la planta posee múltiples propiedades medicinales que hacen que la yerba mate sea popular para el consumo en esta región. Actualmente también se considera un producto nutracéutico y un alimento funcional que es consumido en diferentes países del mundo. En medicina popular, se ha reportado científicamente que posee múltiples propiedades. Treinta y nueve marcas de muestras de yerba mate se analizaron mediante placas de dilución en serie y se incubaron a 25°C durante 5 días se determinó la incidencia fúngica. De las 39 muestras analizadas, el 15,4% estuvieron libres de la presencia de hongos y levaduras. El 84,6% restante presentó contaminación con incidencias variables de Aspergillus. Se observó la presencia de Aspergillus secciones nigri, flavi, circumdati, fumigati, wentii, usti, versicolor y Emericella nidulans, siendo la prevalente Aspergillus sección nigri, representando el 65,3%. Teniendo en cuenta los riesgos a la salud asociados con estos hongos, es importante considerar la formulación de regulaciones con respecto al tamaño de las partículas y la presencia de ocratoxinas y aflatoxinas en los productos comerciales de yerba mate. Abstract: Yerba mate (Ilex paraguariensis) is a widely consumed herb which originated in Paraguay but grows widely in the region between Southern Brazil, Northern Argentina, Paraguay, and Uruguay. The plant is reported to possess multiple medicinal properties which makes yerba mate popular for consumption as tea in this region. It is also being considered as a nutraceutical product and a functional food consumed in different countries around the world. In popular medicine, and it has been scientifically reported to possess multiple properties. Thirty-nine brands of yerba mate samples were analyzed by serial dilution plates. and incubated at 25 ° C for 5 days. Based on their morphological characteristics, the fungal incidence and the number of colonies forming units were determined. Of the 39 samples analyzed, 15.4% were free from the presence of fungi and yeasts. The remaining 84.6% of the samples were contaminated with variable contents of the fungi Aspergillus. The presence of Aspergillus sections nigri, flavi, circumdati, fumigati, wentii, usti, versicolor and Emericella nidulans was observed the most prevalent among them was Aspergillus section nigri, representing 65.3%. Considering the health risks associated with these fungi, it is important to consider formulating regulations regarding the particle size and the presence of ochratoxins and aflatoxins in commercial yerba mate products.

Abstract

Amyloidoses constitute a group of diseases in which soluble proteins aggregate and deposit extracellularly in tissues. Nonhereditary apolipoprotein A-I (apoA-I) amyloid is characterized by deposits of nonvariant protein in atherosclerotic arteries. Despite being common, little is known about the pathogenesis and significance of apoA-I deposition. In this work we investigated by fluorescence and biochemical approaches the impact of a cellular microenvironment associated with chronic inflammation on the folding and pro-amyloidogenic processing of apoA-I. Results showed that mildly acidic pH promotes misfolding, aggregation, and increased binding of apoA-I to extracellular matrix elements, thus favoring protein deposition as amyloid like-complexes. In addition, activated neutrophils and oxidative/proteolytic cleavage of the protein give rise to pro amyloidogenic products. We conclude that, even though apoA-I is not inherently amyloidogenic, it may produce non hereditary amyloidosis as a consequence of the pro-inflammatory microenvironment associated to atherogenesis.

Abstract

The utility of using a protammine/DNA complex coated with a lipid envelope made of cationic 1,2-dioleoyl-3-trimethylammonium propane (DOTAP) for transfecting CHO (Chinese hamster ovary cells), HEK293 (human embryonic kidney cells), NIH 3T3 (mouse embryonal cells), and A17 (murine cancer cells) cells was examined. The widely used DOTAP/DNA lipoplex was employed as a reference. In all the tested cell lines lipid/protamine/DNA (LPD) nanoparticles were more efficient in transfecting cells than lipoplexes even though the lipid composition of the lipid envelope was the same in both devices. Physical-chemical properties were found to control the ability of nanocarriers to release DNA upon interaction with cellular membranes. LPD complexes easily release their DNA payload, while lipoplexes remain largely intact and accumulate at the cell nucleus. Collectively, these data explain why LPD nanoparticles often exhibit superior performances compared to lipoplexes in trasfecting cells and represent a promising class of nanocarriers for gene delivery.

Abstract

α-hemolysin (HlyA) from Escherichia coli is considered as the prototype of a family of toxins called RTX (repeat in toxin), a group of proteins that share genetic and structural features. HlyA is an important virulence factor in E. coli extraintestinal infections, such as meningitis, septicemia and urinary infections. High concentrations of the toxin cause the lysis of several cells such as erythrocytes, granulocytes, monocytes, endothelial and renal epithelial cells of different species. At low concentrations it induces the production of cytokines and apoptosis. Since many of the subcytolytic effects in other cells have been reported to be triggered by the increase of intracellular calcium, we followed the calcium concentration inside the erythrocytes while incubating with sublytic concentrations of HlyA. Calcium concentration was monitored using the calcium indicator Green 1, 2-photon excitation, and fluorescence lifetime imaging microscopy (FLIM). Data were analyzed using the phasor representation. In this report, we present evidence that, at sublytic concentrations, HlyA induces an increase of calcium concentration in rabbit erythrocytes in the first 10 s. Results are discussed in relation to the difficulties of measuring calcium concentrations in erythrocytes where hemoglobin is present, the contribution of the background and the heterogeneity of the response observed in individual cells.

Abstract

Methyl-β-cyclodextrins (MβCDs) are molecules that are extensively used to remove and to load cholesterol (Chol) from artificial and natural membranes; however, the mechanism of Chol extraction by MβCD from pure lipids or from complex mixtures is not fully understood. One of the outstanding questions in this field is the capability of MβCD to remove Chol from lipid domains having different packing. Here, we investigated the specificity of MβCD to remove Chol from coexisting macrodomains with different lipid packing. We used giant unilamellar vesicles (GUVs) made of 1,2-dioleoylphosphatidylcholine:1,2-dipalmitoylphatidylcholine:free cholesterol, 1:1:1 molar ratio at 27°C. Under these conditions, individual GUVs present Chol distributed into lo and ld phases. The two phases can be distinguished and visualized using Laurdan generalized polarization and two-photon excitation fluorescence microscopy. Our data indicate that MβCD removes Chol preferentially from the more disordered phase. The process of selective Chol removal is dependent on the MβCD concentration. At high concentrations, MβCD also removes phospholipids.

Abstract

Calcium controls an array of key events in keratinocytes and epidermis: localized changes in Ca(2+) concentrations and their regulation are therefore especially important to assess when observing epidermal barrier homeostasis and repair, neonatal barrier establishment, in differentiation, signaling, cell adhesion, and in various pathological states. Yet, tissue- and cellular Ca(2+) concentrations in physiologic and diseased states are only partially known, and difficult to measure. Prior observations on the Ca(2+) distribution in skin were based on Ca(2+) precipitation followed by electron microscopy, or proton-induced X-ray emission. Neither cellular and/or subcellular localization could be determined through these approaches. In cells in vitro, fluorescent dyes have been used extensively for ratiometric measurements of static and dynamic Ca(2+) concentrations, also assessing organelle Ca(2+) concentrations. For lack of better methods, these findings together build the basis for the current view of the role of Ca(2+) in epidermis, their limitations notwithstanding. Here we report a method using Calcium Green 5N as the calcium sensor and the phasor-plot approach to separate raw lifetime components. Thus, fluorescence lifetime imaging (FLIM) enables us to quantitatively assess and visualize dynamic changes of Ca(2+) at light-microscopic resolution in ex vivo biopsies of unfixed epidermis, in close to in vivo conditions. Comparing undisturbed epidermis with epidermis following a barrier insult revealed major shifts, and more importantly, a mobilization of high amounts of Ca(2+) shortly following barrier disruption, from intracellular stores. These results partially contradict the conventional view, where barrier insults abrogate a Ca(2+) gradient towards the stratum granulosum. Ca(2+) FLIM overcomes prior limitations in the observation of epidermal Ca(2+) dynamics, and will allow further insights into basic epidermal physiology.

48. Efficient Escape from Endosomes Determines the Superior Efficiency of Multicomponent Lipoplexes

Abstract

Designer multicomponent lipoplexes have recently emerged as especially promising transfection candidates, since they are from 10 to 100 times more efficient than binary complexes usually employed for gene delivery purposes. Here, we show, for the first time, that after internalization binary complexes of lower transfection potency remain in compact perinuclear endosomes, while multicomponent systems have intrinsic endosomal rupture properties that allow plasmid DNA to escape from endosomes with extremely high efficiency. Endosomal rupture results in an extraordinarily homogeneous distribution of unbound plasmid DNA throughout the cytoplasm and in the nucleus.
