Diversity information mining about a crop for different attributes is an essential step for effective breeding programs. The present investigation evaluates the quantum of genetic variability and determines the relationship among the important agro-economic traits based on two years of phenotypic data of 210 accessions of linseed. The traits, capsule weight per plant, capsule per plant, husk weight per plant, and seed weight per plant exhibited comparatively higher genetic coefficient of variation (GCV) and phenotypic coefficient of variation (PCV). In contrast, oil content and seed per capsule exhibited a lower value. The high magnitude of broad sense heritability was observed for all traits except seeds per capsule and husk weight per plant. The trait, capsules per plant, plant height, and days to 50% flowering showed high genetic advance coupled with high heritability. Hierarchical cluster analysis grouped 210 accessions into six distinct clusters. Out of 210, 144 (68.57%) accessions were grouped into three clusters (I, II, and III), in which cluster-III was the largest, containing 64 accessions followed by cluster II and cluster-I. The highest inter-cluster distance was observed between clusters-I and V (127.85), while the lowest was between clusters-II and IV (27.09). The positive correlation of capsule weight per plant with the seed weight per plant and a negative correlation with the days to 50% flowering indicates that high yielding linseed varieties with early flowering/maturity could be developed through direct and indirect selection. Further, seed yield and oil content could be enhanced together as indicated by ghe positive association among these two important traits. In this study, high yielding accessions with moderate to high oil content such as GP36, GP31, GP14, GP54, GP26, GP24, GP34, GP21, GP37 and GP27 and early flowering (less than 70 days) accessions such as GP2, GP26, GP27, CG33, CG44, CG42, CG132, and CG31 identified as potential genetic materials that could be exploited for developing early maturing varieties with high yield. In addition, information’s on various genetic parameters will help breeders to devise suitable breeding methodology for linseed genetic improvement for targeted traits.

In this article, a new numerical technique is presented to obtain numerical solution of a system of fractional delay differential equations (FDDE’s) involving proportional and time dependent delay terms. The fractional derivative is used in Caputo sense. The proposed technique is the combination of fractional differential transform and Bell polynomial. The existence and uniqueness results are discussed for FDDE’s. Three numerical problems are discussed to show reliability and efficiency of the method. Numerical results are compared with exact and Matlab DDENSD solution. The main advantage of the present method is handing effectively the nonlinear terms present in the FDDEs by using Bell polynomial. The present method can deal with both linear and nonlinear FDDEs. The convergence result is discussed, and error analysis is presented in detail.

In this paper, the nonlinear shallow water equation appears in the mathematical modeling of tsunami wave propagation along the coastline of an ocean is solved numerically. The model investigates fractional derivatives and is presented through a system of nonlinear partial differential equations. The variational iteration method (VIM) and the residual power series method (RPSM) have been applied to the fractional system of equations for different parameters. In addition, tsunami wave velocity and run-up height with respect to coast slope and sea depth are analyzed for different time and order in detail. A detailed error analysis is presented for tsunami velocity and tsunami wave propagation. The results obtained are compared to other existing results to show the efficiency and reliability of the proposed methods.

Sickle cell disease (SCD) had first been mentioned in the literature a century ago. Advancement in the molecular basis of the pathophysiology of the disease opens the door for various therapeutic options. Though life-extending treatments are available for treating patients with SCD, allogeneic hematopoietic stem cell transplantation (HSCT) is the only option as of yet. A major obstacle before HSCT to cure patients with SCD is the availability of donors. Matched sibling donors are available only for a small percentage of patients. To expand the donor pool, different contrasting approaches of allogeneic HSCT like T-cell replete and deplete have been tested. None of those tested approaches have been without the risk of GvHD and graft rejection. Other limitations such as transplantation-related infections and organ dysfunction caused by the harsh conditioning regimen need to be addressed on a priority basis. In this review, we will discuss available allogeneic HSCT approaches to cure SCD, as well as recent advancements to make the approach safer. The center of interest is using megadose T-cell-depleted bone marrow in conjugation with donor-derived CD8 veto T cells to achieve engraftment and tolerance across MHC barriers, under reduced intensity conditioning (RIC). This approach is in phase I/II clinical trial at the MD Anderson Cancer Centre and is open to patients with hemoglobinopathies.

Microtubules (MTs) control cell shape and intracellular cargo transport. The role of MT turnover in the migration of slow-moving cells through endothelial barriers remains unclear. To irreversibly interfere with MT disassembly, we have used the MT-stabilizing agent zampanolide (ZMP) in Β16F10 melanoma as amodel of slow-moving cells. ZMP-treated B16 cells failed to follow chemotactic gradients across rigid confinements and could not generate stable sub-endothelial pseudopodia under endothelial monolayers. In vivo, ZMP-treated Β16 cells failed to extravasate though lung capillaries. In contrast to melanoma cells, the chemotaxis and transendothelial migration of ZMP-treated Tcells were largely conserved. This is afirst demonstration that MT disassembly is akey checkpoint in the directional migration of cancer cells but not of lymphocytes.

During this unprecedented time of the coronavirus disease 2019 (COVID 19) pandemic, most countries are struggling to optimize their healthcare resources. Although the prevention and treatment of COVID 19 has become the priority for the majority of the population, most medical professionals are facing the challenge of balancing the management of this pandemic and treating patients with other diseases. Healthcare professionals from all specialties are facing this situation and designing guidelines and recommendations that are contributing to the literature that is crucial for disease management at the current time. We share our experience with two ends of the spectra that we currently observe in spine patients. One group of patients included the non-operative cases that presented with back pain. Further, we discuss our experience with operative precautions and digitally assisted discharge.

A 72-year-old male patient was operated by Austin Moore hemiarthroplasty for fracture neck of femur 6 years back who developed multiple complications. In this case, the patient developed dislocation of the hip joint following which the Austin Moore prosthesis got broken. During revision with bipolar hemiarthroplasty, the patient developed fracture of proximal femur which was poorly managed by K-wires and stainless steel wires. After some time, the patient developed dislocation of the bipolar prosthesis and signs of infection. We operated this patient with a two-stage revision. In the first stage following removal of all infected metalwork and debridement, we put antibiotic cement spacer, and after 4 weeks in the second stage, we did a total hip replacement with uncemented constrained proximal femoral modular reconstruction prosthesis. The case is being reported for its rare presentation in genuine management and successful outcome.

It is a known fact that following split thickness skin grafting in post traumatic wounds the donor site heals with re-epithliazation. The skin thus formed initially is fragile and takes a long time to become strong mobile and elastic. However, in some patients, donor site poses risk of nonhealing, delayed healing, hypertrophic scar formation and cosmetic disfigurement, etc., In such patients, it is ideal to identify such risk factors and manage them carefully. We are reporting a case of degloving of the foot in a poly-traumatized patient treated by split thickness skin grafting where to avoid the above problems, the donor site was also skin grafted primarily after meshing the skin and making it large.

Feature binding is a process that creates an integrated representation of an object. A change detection task with four stimuli is used to study color-shape binding of sequentially presented stimuli. Given the immense importance of locations in feature binding, and noting the confound of location information with simultaneous presentation, we compared simultaneous and sequential presentations when locations remained the same from study to test and when they changed randomly. In Experiment 1, sequential presentation implied showing the stimuli one by one to gradually build up the study display. There were no differences between the two modes of presentation in this experiment, although performance was better with unchanged locations than random locations. Experiment 2 used a sequential presentation when one stimulus vanished as the next was presented. An interaction effect showed that performance was much better with unchanged locations than random locations with simultaneous presentation, whereas locations had no effect in the sequential presentation condition. Three subsequent experiments, with drastically reduced presentation time for the display in the simultaneous presentation condition (Experiment 3), with blank intervals inserted after every stimulus in the sequential presentation condition (Experiment 4), and with a mask given immediately after the study-display presentation (Experiment 5), showed results similar to Experiment 2. Thus, we surmise that locations are a factor only in simultaneous presentation, and not in sequential presentation, and the differences between the two conditions can be attributed to post-perceptual factors within visual working memory.

The endothelial cytoskeleton is a barrier for leukocyte transendothelial migration (TEM). Mononuclear and polymorphonuclear leukocytes generate gaps of similar micron-scale size when squeezing through inflamed endothelial barriers in vitro and in vivo. To elucidate how leukocytes squeeze through these barriers, we co-tracked the endothelial actin filaments and leukocyte nuclei in real time. Nuclear squeezing involved either preexistent or de novo-generated lobes inserted into the leukocyte lamellipodia. Leukocyte nuclei reversibly bent the endothelial actin stress fibers. Surprisingly, formation of both paracellular gaps and transcellular pores by squeezing leukocytes did not require Rho kinase or myosin II-mediated endothelial contractility. Electron-microscopic analysis suggested that nuclear squeezing displaced without condensing the endothelial actin filaments. Blocking endothelial actin turnover abolished leukocyte nuclear squeezing, whereas increasing actin filament density did not. We propose that leukocyte nuclei must disassemble the thin endothelial actin filaments interlaced between endothelial stress fibers in order to complete TEM.

Male fertility is an orchestrated interplay of loci on the Y chromosome with a number of genes from across the other chromosomes. In this context, micro-deletions in the Y chromosome have been correlated with spermatogenic failure often leading to infertility. However, causes of infertility in the patients with the normal spermiogram have remained unclear and therefore pose another level of challenge. In the present study, we analyzed 64 STSs, studied different Y-linked genes and loci and conducted single nucleotide variant (SNV) analyses in 31 infertile males with normal spermiogram along with 67 normal fertile males (NFMs) to gain an insight into the organization of their Y chromosome. Further, employing quantitative real-time PCR (qPCR), we studied copy number variation of DYZ1 arrays and three genes and mutational status of SRY by direct sequence analyses. STS analyses of the AZFa, b and c regions in these patients showed known and new mutations. Further, copies of DAZ and BPY2 in the patients were found to be affected (p < 0.001) compared to those in NFMs. All the patients had normal copy number of the SRY however its sequence analysis (in silico) showed mutations in eight patients. In four of these eight patients, SRY mutations resulted into truncated proteins. Similarly, DYZ1 analysis showed micro-deletions and it's much reduced copy number (p < 0.001) as compared to those in NFMs. Present study in males with unexplained infertility revealed deletions similar to those observed in oligospermic and azoospermic patients. Thus, there are some common but still unknown factors underlying infertility in these patients irrespective of their spermatogenic status. This work is envisaged to augment DNA diagnosis, proving beneficial in the context of in vitro fertilization (IVF) and genetic counselling.
