This article explores the symbiotic role of microorganisms (bacteria, fungi and protozoans) associated with xylophagous termites. Members of the subfamily Macrotermitinae belonging to the family Termitidae have evolved symbiosis with fungi, which belong to the genus Termitomyces. The function of Termitomyces varies for different termite groups depending on their feeding behaviour. In some termites, the primary function of Termitomyces fungi is the degradation of lignin to its simpler form and to make cellulose available to termites; however, these fungi also serve as nutrient rich food sources to other groups of termites. The subsequent breakdown of ingested cellulose in termite gut is further facilitated and controlled by the action of various groups of enzymes secreted by gut-borne microflora and micro-fauna. Understanding the function, significance and management of these diverse microbial symbionts associated with pestiferous higher termites may help in developing their effective bio-control.

The global pandemic due to the novel Severe Acute Respiratory Syndrome Corona Virus 2 (SARS-CoV-2) has taken more than a million lives. Lack of definitive vaccine/drugs against this highly contagious virus has accelerated exploratory research on novel natural and synthetic inhibitors. Tea is a rich source of bioactives and known to have antiviral properties. In this study, an in silico strategy involving ADMET property screening, receptor–ligand docking and molecular dynamic (MD) simulation was employed to screen potential tea bio-active inhibitors against three selected targets (RdRp, 3CLpro and PLpro) of SARS-CoV-2. Among the 70 tea bioactives screened, theaflavin 3,3’-di-gallate (TF3), Procyanidin B2 and Theaflavin 3-gallate (TF2a) exhibited highest binding affinities towards RdRp, 3CLpro/Mpro and PLpro targets of SARS-CoV-2 with low docking scores of −14.92, −11.68 and −10.90 kcal/mol, respectively. All of them showed a substantial number of hydrogen bonds along with other interactions in and around the active sites. Interestingly, the top bioactives in our study showed higher binding affinities compared with known antiviral drugs. Further, the top protein–ligand complexes showed less conformational changes during binding when subjected to MD simulation for 100 nanoseconds. The MMPBSA results revealed that RdRp-TF3, 3CLpro-Procyanidin B2 and PLpro-TF2a complexes were stable with binding free energies of −93.59 ± 43.97, −139.78 ± 16.51 and −96.88 ± 25.39 kJ/mol, respectively. Our results suggest that theaflavin 3,3’-digallate, Theaflavin 3-gallate and Procyanidin B2 found in black tea have the potential to act as inhibitors for selected targets of SARS-CoV-2 and can be considered as drug candidates in future studies against COVID-19. Nanotechnology research uses specific properties of materials at the nanoscale to develop improved materials, devices, systems and therapeutics. There is a risk of overlapping patent claims and lack of distinction between nano-based and traditional patents due to the interdisciplinary nature of nanotechnology. There is an increasing trend of granted patents. The World Intellectual Property Organization and World Health Organization, regulatory and policy bodies, are working to make a comprehensive property right regulation for nanotechnology products. The USA, the leader of nanotechnology products, has made guidelines to make patent search easier for nano-based products. The European Patent Office has also created a new classification for nano-based inventions. Here we review the status of intellectual property rights protection of nanomaterial, environmental implications and application of nanotechnology in agriculture.

Tea (Camellia sinensis) plants are continuously exposed to a wide range of biotic and abiotic stresses. Being a perennial monoculture crop, tea relatively provides a favorable niche for more than 1000 arthropod pests and numerous disease-causing microorganisms. Abiotic stress factors such as drought, temperature, waterlogging, salinity, and nutrient stress considerably constrain the growth, productivity, and quality of tea plants. At present, global climatic changes have made notable impact in tea with a decline in its productivity. The main biotic and abiotic stresses that affect tea plants in the field are being extensively studied. Each stress elicits a complex molecular cascade starting from perception of stress to initiation of signal transduction and its manifestation in cellular and metabolic level. Construction of genetic map, by application of conventional molecular marker technologies, has been difficult in tea due to the problems of self-incompatibility, the absence of pure inbreed lines, high heterozygosity, 15 pairs of chromosomes, and large genome size. However, novel approaches such as next-generation sequencing (NGS) technologies have successfully helped in identification of several candidate genes which are responsible for biotic and abiotic stress regulation in different plants. It has accelerated the process of large-scale single-nucleotide polymorphism (SNP) discovery and genotyping in tea, thus, facilitating the construction of high-density genetic maps. The landmark tea whole genome will lead to identification and mapping of quantitative trait loci related to agronomical important traits underlying biotic and abiotic stress. It will also aid in comparative genomic analysis, marker-assisted selection, and map-based cloning for developing stress-tolerant tea plants.

Silver nanoparticles of average sizes ~18-21 nm in diameter were prepared by a green approach via chemical reduction of silver nitrate (AgNO3) using Asiatic Pennywort and Bryophyllum leaf extracts as reducing and capping agents. The bio-reduced silver nanoparticles were characterized by UV-Vis spectroscopic, XRD and TEM techniques. The characteristic surface plasmon band of colloidal solutions of AgNPs synthesized from Asiatic Pennywort and Bryophyllum leaf extracts were found at 445 nm and 405 nm respectively. The results of XRD and SAED pattern showed that the biosynthesized AgNPs have a crystalline structure with cubic phase (fcc). The antimicrobial activities of the as synthesized AgNPs were investigated against gram negative bacteria Pseudomonas Fluorescens and gram positive bacteria Staphylococcus Epidermidis. It was observed that silver nanoparticles obtained from Asiatic Pennywort was more effective on gram positive bacteria Staphylococcus Epidermidis while AgNPs obtained from Bryophyllum was more effective on gram negative bacteria Pseudomonas Fluorescens indicating size dependent activity of AgNPs. Nanoscience studies biological properties of materials at the nanoscale. Nanotechnology research develops improved materials, devices, systems and therapeutics. Nanomaterials can be protected for their intellectual property rights by innovators. However, due to the interdisciplinary nature of nanotechnology, there is a risk of overlapping patent claims and lack of distinction between nano-based and traditional patents. Scientists also must solve ethical and social issues, from health to environmental risk and consumer perception.

Understanding the genes that govern tea plant (Camellia sinensis) architecture and response to drought stress is urgently needed to enhance breeding in tea with improved water use efficiency. Field drought is a slow mechanism and the plants go through an adaptive process in contrast to the drastic changes of rapid dehydration in case of controlled experiments. We identified a set of drought responsive genes under controlled condition using SSH, and validated the identified genes and their pattern of expression under field drought condition. The study was at three stages of water deficit stress viz., before wilting, wilting and recovery, which revealed a set of genes with higher expression at before wilting stage including dehydrin, abscissic acid ripening protein, glutathione peroxidase, cinnamoyl CoA reductase, calmodulin binding protein. The higher expression of these genes was related with increase tolerance character of DT/TS-463 before wilting, these five tolerant progenies could withstand drought stress and thus are candidates for breeding. We observed that physiological parameter like water use efficiency formed a close group with genes such as calmodulin related, DRM3, hexose transporter, hydrogen peroxide induced protein, ACC oxidase, lipase, ethylene responsive transcription factor and diaminopimelate decarboxylase, during wilting point. Our data provides valuable information for the gene components and the dynamics of gene expression in second and third leaf against drought stress in tea, which could be regarded as candidate targets potentially associated with drought tolerance. We propose that the identified five tolerant progenies on the basis of their drought tolerance can thus be utilised for future breeding programmes.

A cDNA-AFLP approach was used to identify transcript and/or genes specifically expressed in response to drought in tea. Drought was artificially induced and whole genome transcript profiling was done at three different stages-6 days before wilting, 3 days before wilting and at wilting stage of both tolerant and susceptible cultivars, and genetic differences was thus visualized as polymorphisms in the transcriptome. The cDNA-AFLP technique allowed genes and transcripts to be identified in the tolerant genotype (TV-23) whose expression is responsive to drought stress. The cluster analysis revealed two types of clustering-type I separated the tolerant and susceptible cultivar, whereas type II separated the time point of sample and this may be grouped as early and late responsive transcripts. 108 transcript derived fragments were identified as differentially expressed in tolerant genotypes of which 89 sequences could be obtained. Fifty-nine of them showed homology in the public databases. Functional ontology showed genes related to carbohydrate metabolism, response to stress, protein modification process and translation. Cluster I includes five fragments and cluster II includes 25 fragments. Other genes strongly expressed in response to drought in tolerant genotype would help us in identifying and determining the genetic basis of mechanisms involved in conferring drought tolerance in tea.

RAPD and microsatellite markers were used to determine the genetic fidelity of micropropagated plants from the three varieties of tea plant derived from explants of field grown mother bush as well as in vitro germinated seedlings. Rate of shoot multiplication was better from nodal explants than from shoot tips. A maximum of 32–33 shoots was observed in cotyledonary node in 1/2 MS medium with BAP (6 mg/l), GA3 (1 mg/l) and IBA (0.5 mg/l). 90% of the in vitro derived microshoots were micrografted into rootstocks. The micropropagated plantlets showed both cytological and genetical stability. SSR primers showed complete stability among the regenerants. The results convinced that plants derived from axillary as well as adventitious mode of propagation can be genetically true to type. This cost effective technique would help in fast clonal propagation at a commercial scale.

MADS (Minichromosome Maintenance1 Agamous Deficiens Serum response factor) box genes encode transcription factors and they play a key role in growth and development of flowering plants. There are two types of MADS box genes- Type I (serum response factor (SRF)-like) and Type II (myocyte enhancer factor 2 (MEF2)-like). Type II MADS box genes have a conserved MIKC domain (MADS DNA-binding domain, intervening domain, keratin-like domain, and c-terminal domain) and these were extensively studied in plants. Compared to other plants very little is known about MADS box genes in Camellia sinensis. The present study aims at identifying and analyzing the MADS-box genes present in Camellia sinensis. A comparative bioinformatics and phylogenetic analysis of the Camellia sinensis sequences along with Arabidopsis thaliana MADS box sequences available in the public domain databases led to the identification of 16 genes which were orthologous to Type II MADS box gene family members. The protein sequences were classified into distinct clades which are associated with the conserved function of flower and seed development. The identified genes may be used for gene expression and gene manipulation studies to elucidate their role in the development and flowering of tea which may pave the way to improve the crop productivity.

The present experiment is an effort to find a stable reference gene in Camellia sinensis and Camellia assamica under different biotic and abiotic stresses. This study evaluate the variation in gene expression across tea leaf tissues in nine experiments. The suitability of 18S rRNA , 26S rRNA , rubiscobis- phosphatase oxygenase (RuBP) and Camellia tubulin (CaT) as reference genes were validated by geNorm and BestKeeper programs. The finding reveals 18S rRNA and RuBP to be the most stably expressed housekeeping genes, the latter being the first report of its kind in tea. The finding paves the way for their application in accurate quantification of trait specific gene expression and other genomic studies in tea. Darjeeling teas are the highest grown teas in the world and preferred for its flavour, aroma and quality. Apart from the genetic makeup of the plant, earlier reports suggest that insect infestation, particularly jassids and thrips triggers the aroma and flavour formation in Darjeeling tea. The present work encompasses the identification of the genes/transcriptomes responsible for the typical flavour of Darjeeling tea, besides understanding the role of jassids and thrips in particular, in producing the best cup character and quality. The quantitative real time PCR analysis was based on a suppression subtractive hybridisation forward library of B157 (tea clone infested with thrips), providing us transcripts related to aroma and flavour formation. We observed the expression of genes like leucine zipper, ntd, nced, geraniol synthase, raffinose synthase, trehalose synthase, amylase, farnesyl transferase, catalase, methyl transferase, linalool synthase, peroxidases, elicitor responsive proteins, linamarase, nerolidol linalool synthase 2, 12-oxophytodienoate reductase, glucosidase, MYB transcription factor, and alcohol dehydrogenase, highly regulated due to insect infestation, manufacturing stresses and mechanical injury. The first report on gene expression dynamics in thrips infested Darjeeling tea leaves can be extrapolated with increase in volatiles which is responsible for enhancing the quality of Darjeeling tea, specially the flavour and aroma of the infusion. We hope to model these responses in order to understand the molecular changes that occur during Darjeeling tea flavour formation.
