Aim Methodology Results Conclusions Calcium hydroxide (CH) has been considered as a direct pulp capping materials (DPC) for the last decades despite having some limitations. Phosphorylate pullulan (PPL) incorporated with CH (CHPPL) is a novel biomaterial that was introduced as a promising DPC material. Thus, the aim of the study was to evaluate the inflammatory response and mineralized tissue formation (MTF) ability of PPL‐based CH formulations on rat molars after DPC.This study consisted of six groups: CH with 1% PPL (CHPPL‐1); 3% PPL (CHPPL‐3); 5% PPL (CHPPL‐5); Dycal and NEX MTA Cement (N‐MTA) as the positive control, and no capping materials (NC). One hundred twenty maxillary first molar cavities were prepared on Wistar rats. After capping, all the cavities were restored with 4‐META/MMA‐TBB resin and pulpal responses were evaluated at days 1, 7, and 28. Kruskal–Wallis followed by Mann–Whitney U‐test was performed with a significance level of 0.05. Immunohistochemical expression of IL‐6, Nestin, and DMP‐1 was observed.At day 1, CHPPL‐1, N‐MTA, and Dycal exhibited no to mild inflammation, whilst CHPPL‐3, CHPPL‐5, and NC showed mild to moderate inflammation, and the results were significantly different (p < .05). At day 7, mild to moderate inflammation was observed in CHPPL‐1, N‐MTA, and Dycal, whereas CHPPL‐3, CHPPL‐5, and NC exhibited moderate to severe inflammation. Significant differences were observed between CHPPL‐1 and N‐MTA with NC (p < .05), CHPPL‐1 and CHPPL‐3 with CHPPL‐5 and Dycal (p < .05), and CHPPL‐3 with N‐MTA (p < .05). A thin layer of mineralized tissue formation (MTF) was observed in all groups. At day 28, CHPPL‐1, Dycal, and N‐MTA showed no to mild inflammation, whilst CHPPL‐3, CHPPL‐5, and NC exhibited mild to severe inflammation, and statistically significant difference was detected (p < .05). CHPPL‐1, Dycal, and N‐MTA exhibited continuous MTF, whilst CHPPL‐3, CHPPL‐5, and NC had thicker and interrupted MTF. Significant differences were observed between CHPPL‐1, CHPPL‐3, and N‐MTA with NC group (p < .05). Variable expressions of IL‐6, Nestin, and DMP‐1 indicated differences in the materials' impact on odontoblast‐like cell formation and tissue mineralization.These findings suggest that CHPPL‐1 has the potential to minimize pulpal inflammation and promote MTF and had similar efficacy as MTA cement. This study evaluated the effect of simulated pulpal pressure (SPP) conditions and storage time on contemporary adhesive systems' microtensile bond strength (µTBS) to dentin. Extracted human molars were prepared and randomly divided into four groups according to the adhesives: Clearfil Megabond 2 (CSE), Beautibond Xtreme Universal (BXU), G2-Bond (G2B), and Scotchbond Universal Plus (SBP). Each adhesive group was further divided following the SPP conditions: control with no simulation (SPP-CTR), SPP with distilled water (SPP-DTW), and SPP with fetal bovine serum (SPP-FBS). Resin composite build-ups were prepared, and teeth were stored in water (37 °C) for 24 h (24 h) and 3 months (3 m). Then, teeth were sectioned to obtain resin–dentin bonded beams and tested to determine the µTBS. Data were analyzed using three-way ANOVA, Tukey post hoc tests (=0.05), and Weibull failure analysis. Failure mode was observed using scanning electron microscopy. The µTBS response was affected by adhesive systems, simulated pulpal pressure conditions, and storage time. SPP-CTR groups presented a higher overall bond strength than SPP-DTW and SPP-FBS, which were not significantly different from each other. Only for SBP, the SPP-FBS group showed higher µTBS than the SPP-DTW group. The Weibull analysis showed that the bonding reliability and durability under SPP-DTW and SPP-FBS were inferior to SPP-CTR, and the 24 h bonding quality of adhesives to dentin was superior to that of 3 m. SPP drastically reduced the µTBS of all adhesives to dentin regardless of solution (distilled water or fetal bovine serum). Storage after 3 m also decreased µTBS despite the SPP condition. This study aimed to evaluate microtensile bond strength (µTBS) and ultra-morphological characteristic changes in sound dentin resulting from silver diamine fluoride (SDF) when using adhesives. Ninety-six extracted human third molars were divided into the SDF-contaminated dentin group and the sound dentin group. In the SDF-contaminated dentin group, 38% SDF was agitated for 1 min, left undisturbed for 2 min, and rinsed with distilled water for 30 s. Then, each group was further subdivided into six subgroups (n = 8 / group) according to application modes: self-etch mode (SE) and etch-and-rinse mode (ER) followed by three adhesives: (1) Scothbond Universal Plus Adhesive (SUP); (2) G2-Bond Universal Adhesive (G2B); and (3) Clearfil Mega Bond 2 (MB2). All specimens were restored with resin composite and were stored in distilled water for 24 h before μTBS testing. Data from the μTBS test were analyzed using Three-way ANOVA and Duncan test (p < 0.05). The morphology of fractured surface and adhesive-dentin interfaces were evaluated by SEM, TEM, and STEM. Further elemental analysis was done by EDX. All SDF-contaminated dentin groups demonstrated significantly lower μTBS than sound dentin groups. All ER groups had higher μTBS than SE groups, except for G2B in the sound dentin group. STEM/EDX revealed an SDF-dentin-reacted layer in the SDF-contaminated dentin group. SDF had an adverse effect on adhesives. Additionally, ER mode is preferable when bonded to SDF-contaminated dentin. A calcium and fluoride-contained layer was observed in all SDF-contaminated dentin. This study evaluated the effect of a 0.5% chloramine T solution on a chemical-cured universal adhesive by comparing the light-cured, one-step, self-etch adhesive for the bonding performance, mechanical properties, and resin–dentin interfacial characteristics. Caries-free human molars were randomly assigned into eight groups based on the bonding systems employed (Bond Force II, BF and Bondmer Lightless, BL), the immersion solutions used before bonding (0.5% chloramine T solution and distilled water), and the immersion durations (5 and 60 min). Microtensile bond strength (μTBS), nanoleakage evaluation, and nanoindentation tests were performed, and the surface morphology of the resin–dentin interface was examined using a focus ion beam/scanning ion microscopy system. Immersion in chloramine-T for 5 min significantly decreased the μTBS of Bondmer Lightless (from 22.62 to 12.87 MPa) compared with that in distilled water. Moreover, there was also a decreasing trend after immersing in chloramine-T for 60 min (from 19.11 to 13.93 MPa). Chloramine T was found to have no effect on the hardness, elastic modulus, or morphological characteristics of the ion-beam milled resin–dentin interfacial surfaces in the tested adhesives, suggesting that chloramine T might reduce the bond strength by interfering with the interaction and the sealing between the adhesive resin and dentin in the chemical-cured universal adhesive, albeit without affecting the mechanical properties. Objective: To evaluate the effect of a time delay before the light activation of resin composite on the microtensile bond strength (µTBS) of self-etch adhesives under dynamic simulated pulpal pressure. Materials and methods: One hundred twenty crown segments were prepared from human third molars. Dentin surfaces were prepared with coarse diamond burs and connected to a dynamic pulpal pressure device. The self-etch adhesives used were Clearfil SE Bond (CSE), G2-Bond Universal (G2B), Clearfil Universal Bond Quick (CBQ), and G-Premio Bond (GPB). Each adhesive was divided into 3 subgroups: immediate light activation of resin composite (T0), delayed light activation for 150 s (T1), and 300 s (T2). The µTBS data were obtained after 24-h water storage. Scanning electron microscopy was used to evaluate the nanoleakage at the resin–dentin surface. The µTBS data were analyzed using two-way ANOVA and Duncan's multiple comparisons. Results: All adhesives demonstrated a significant reduction in µTBS at T2, except for CBQ where the bond strength was not affected by the delayed light activation times. The two-step self-etch adhesives (CSE and G2B) demonstrated higher bond strength than the one-step adhesives at all conditions. Nanoleakage was observed in all adhesives at T2; however, nanoleakage was detected only in CBQ and GPB at T1. Conclusions: Delayed light activation of resin composite adversely affected the dentin bond strength of self-etch adhesives. Two-step self-etch adhesives had better bonding performance than one-step self-etch adhesives. The purpose of this study was to evaluate the microtensile bond strength (µTBS) and Knoop hardness number (KHN) of a novel experimental light-cured resin cement (HL). Eighteen flat dentin surfaces of human molars were polished using #600 SiC paper and bonded to CAD/CAM resin blocks with the respective resin cements and composites: HL, Panavia V5 (PV), and Clearfil AP-X (AP). All specimens were stored in distilled water at 37 °C for 24 h and 7 days. Scanning electron microscope (SEM) and energy dispersive X-ray (EDX) observations were performed to evaluate filler morphology and to detect the elements. The resin cements had a significant effect on the immediate µTBS (F = 22.59, p < 0.05) and after water storage µTBS (F = 22.83, p < 0.05). Significant differences (p < 0.05) in the KHN between the tested materials were observed, and HL indicated the highest KHN when compared with PV. HL showed a combination of the regular-shaped filler and spherical-shaped filler within the matrix. Silicon was detected in HL from the EDX evaluation. HL exhibited better bonding performance and polymerization, which may have contributed to the improvement of the adhesive strength. [ABSTRACT FROM AUTHOR]

This study aims to assess the color stability, water sorption, and solubility of 11 resin composites as commercially available dental products. Twenty samples (10 mm in diameter and 2 mm in thickness) of each material were fabricated using a customized silicone mold, followed by immersion in each of curry, coffee, wine, and distilled water for 28 days (n = 5). Baseline shade and color changes (ΔE) were measured using a reflection spectrophotometer. The CIE L*, a*, b* system was used to evaluate the color changes. Five samples of each resin composite were applied to test water sorption and solubility according to ISO 4049:2009. As a result, the ∆E values were significantly influenced by each of the three factors (composition of material, solution, time) and the interactions between them (p < 0.001). Highest resistance to discoloration was achieved by Ceram.X One Universal (CXU), followed by Magnafill Putty (MP). Generally, microhybrid composites showed fewer color changes than nanohybrid composites and giomers. DX. Universal and Filtek Z350 XT showed the highest ΔE values in all colorants. All materials tested in this study fulfilled the criteria of ISO 4049:2009; CXU and MP had the lowest water sorption and solubility. Cetylpyridinium chloride (CPC), a quaternary ammonium compound, which is present in mouthwash, is effective against bacteria, fungi, and enveloped viruses. This study was conducted to explore the antiviral effect of CPC on SARS-CoV-2. There are few reports on the effect of CPC against wild-type SARS-CoV-2 at low concentrations such as 0.001%–0.005% (10–50 µg/mL). Interestingly, we found that low concentrations of CPC suppressed the infectivity of human isolated SARS-CoV-2 strains (Wuhan, Alpha, Beta, and Gamma) even in saliva. Furthermore, we demonstrated that CPC shows anti-SARS-CoV-2 effects without disrupting the virus envelope, using sucrose density analysis and electron microscopic examination. In conclusion, this study provided experimental evidence that CPC may inhibit SARS-CoV-2 infection even at lower concentrations. [ABSTRACT FROM AUTHOR]

Objectives: The aim of this study was to evaluate the buffering capacity of glass-ionomer material in vitro. The null hypothesis tested was that there is no effect of cured glass-ionomer pit-and-fissure sealant (PFS) pastes on the environmental acidity as well as the tooth substrate.Method: For each material, a cured PFS disk and a section of human enamel were simultaneously soaked in lactic acid solution in a conical tube, and the pH of the solution was measured daily for one week. Subsequently, the total amount of calcium leached out in solution was also measured with ICP-AES.Results: The results showed that the acidity of the solutions changed over time. Significant differences of calcium ion concentration in solution were observed as a result of decalcification. As the PFS products tested did not include calcium, the concentration of calcium ion released indicated acidic erosion of the tooth enamel.Conclusions: The glass-ionomer countered the acid of the solution rapidly and preserved the structure of human tooth enamel. Background: This study aimed to evaluate the color matching, light transmittance, and reflectance characteristics of the novel flowable resin composite OCF-001 (OCF). Methods: Fifty-four resin composite molds were made with simulated class I cavities of A2, A3, and A4 shades by filling the rubber mold interspace with Estelite Sigma Quick (ESQ), Gracefil Putty (GP) and Filtek Supremme Ultra (FSU). After applying the adhesive, three different flowable resin composites (n = 6), OCF, Gracefil LoFlo (GLF), and Supreme Ultra Flowable (SUF), were used to fill the cavities. A colorimeter was used to measure the color parameters (CIEDE2000). The color measurements were taken immediately and after 28 days. Data were analyzed using the nonparametric Kruskal–Wallis (α = 0.05) and Wilcoxon tests. The light transmittance and reflection characteristics were measured with a black background using a spectrophotometer under D65 illumination. Results: The ΔE00, and ΔC of OCF was lower than other tested materials in A2 and A3 shades both immediately and after 28 days. OCF showed the highest transmittance characteristic, and a relatively stable reflectance curve in all the wavelengths. Conclusions: OCF showed better shade matching with the surrounding shades of A2 and A3, a relative uniform reflectance and higher light transmission properties. [ABSTRACT FROM AUTHOR]

Despite recent advances in bonding restorations, which are the basis of restorative dentistry, secondary caries are still able to form. Previously, a novel fluoride-containing zinc and copper (ZCF) nanocomposite was introduced to prevent the formation of caries due to its antibacterial activity. In this study, we studied the impact of ZCF nanoparticles on the adhesive strength of bonding restorations through micro-tensile bond strength (µTBS) testing. The impact of antibacterial and matrix metalloproteinase (MMP) inhibitors on the nanoparticles was also examined. The nanocomposites were prepared using a simple one-step homogeneous co-precipitation method at a low temperature. A self-etch adhesive was applied to 10 extracted caries-free human molars with (test group) and without (control group) the ZCF nanoparticles. This was followed by composite resin build-up and µTBS testing, MMP activity assays, and evaluation of the antibacterial effects. The results showed no significant differences in the µTBS between the ZCF and the control groups. However, the ZCF exhibited a significant inhibitory effect against MMP-2, MMP-8, and MMP-9, in addition to an antibacterial effect on Streptococcus mutans. Therefore, the present study demonstrated that the addition of ZCF nanoparticles to adhesive systems can result in MMP inhibition and antibacterial action while maintaining the mechanical properties of the bonding restorations. Purpose: The effects of light curing units (LCU) and energy doses on the chemical and physical properties of a dental composite were investigated. Methods: The effects on the chemical and physical properties of a bisphenol A diglycidylether methacrylate (Bis-GMA) based dental restorative material were evaluated through photospectrometry, differential scanning calorimetry, and mechanical measurements. Results: The light curing conditions associated with direct and indirect restorations were replicated in vitro using optical investigation techniques. A slight attenuation resulted independently of the LCU and a strong attenuation was measured for the cement luting a thick inlay, as well as for the deepest layer of a composite filling increment. Calorimetric measurements indicated that the curing degree is very sensitive to the light energy dose rather than to the LCU. Mechanical testing showed a transient phase during which properties increased. The delay of the composite in reaching adequate properties is strongly dependent on the energy dose. Conclusions: It is recommended that composites subject to unfavorable light curing conditions undergo a prolonged light curing process. [ABSTRACT FROM AUTHOR]

Resin–dentin bonds are known to degrade in the relatively aggressive oral environment. In order to obtain greater insight into the interfacial degradation process, we examined, by using transmission electron microscopy (TEM), the interfacial ultrastructure of two adhesives bonded to dentin after 1 yr in vivo. Class V cavities were prepared on the buccal surfaces of 14 intact teeth of two monkeys and then restored by using either the two-step self-etch adhesive, Unifil Bond, or the two-step etch-and-rinse adhesive, Single Bond, in combination with the restorative microhybrid composite, Z250. After 1 yr, 10 other teeth were restored by using the same materials (controls). One day later, the monkeys were killed, following which the microtensile bond strength to dentin was determined and the interfacial ultrastructure was examined by TEM. Whereas no noticeable changes in the morphology of the resin–dentin interface were observed between the 1-d and 1-yr specimens for Unifil Bond, Single Bond exhibited signs of interfacial degradation, in particular at the bottom of the 3 µm-deep hybrid layer. Eur J Oral Sci 2004; 112: 286–292.© Eur J Oral Sci, 2004 The aim of this study was to compare the bonding efficacy of an all-in-one adhesive with that of a self-etching primer system by measuring microleakage after thermocycling and the microtensile bond strength (μTBS) to enamel and dentin. Circular class V cavities along cement–enamel junctions were prepared in 20 extracted human premolars. Each 10 teeth were bonded with either AD Bond (AD) or Clearfil SE Bond (SE), and filled with a resin composite (Charisma). After thermocycling and staining with 0.5% basic fuchsin, the microleakage at the coronal and apical walls was evaluated using longitudinal sections. In addition 20 premolars were used to measure μTBS at 24 h after bonding for these adhesives to enamel and dentin that corresponded to the coronal and apical walls of the class V cavities. AD showed more microleakage than SE in the coronal walls, but there was less microleakage in the apical walls and no difference in apical leakage between the two adhesives. μTBS (SD) in MPa to enamel and dentin were 25.2 (7.3) and 68.3 (9.4) for AD, and 35.8 (7.4) and 76.4 (7.8) for SE, respectively. AD gave a lower μTBS to enamel than did SE. The results suggested that the poor adaptation at the coronal wall in AD might be caused by the lower bond strength to enamel. [ABSTRACT FROM AUTHOR]

Aim: To evaluate the dental pulp response to a novel mineral trioxide aggregate containing phosphorylated pullulan (MTAPPL) in rats after direct pulp capping. Methods: Ninety‐six cavities were prepared in the maxillary first molars of 56 male Wistar rats. The dental pulps were intentionally exposed and randomly divided into four groups according to the application of pulp capping materials: MTAPPL; phosphorylated pullulan (PPL); a conventional MTA (Nex‐Cem MTA, NCMTA; positive control); and Super‐Bond (SB; negative control). All cavities were restored with SB and observed for pulpal responses at 1‐, 3‐, 7‐ and 28‐day intervals using a histological scoring system. Statistical analysis was performed using Kruskal–Wallis and Mann–Whitney U‐test with Bonferroni's correction, and the level of significance was set at 0.05. DMP1 and CD34 antigen were used to evaluate odontoblast differentiation and pulpal vascularization, respectively. Results: On day 1, mild inflammatory cells were present in MTAPPL and NCMTA groups; fewer inflammatory cells were present in the PPL, whereas SB was associated with a mild‐to‐moderate inflammatory response. A significant difference was observed between PPL and SB (p <.05). No mineralized tissue deposition was observed. On day 3, moderate‐to‐severe inflammatory cells were present in PPL and SB, whereas MTAPPL and NCMTA had a mild inflammatory response. Initial mineralized tissue deposition was observed in the NCMTA, MTAPPL and SB. A significant difference was observed between MTAPPL and PPL (p <.05). On day 7, a thin layer of mineralized tissue was observed in all tested groups with no or mild inflammatory response. On day 28, no inflammatory response was observed in MTAPPL, whereas NCMTA, PPL and SB had mild inflammatory responses. A significant difference was observed between MTAPPL and SB (p <.05). Complete mineralized tissue barrier formation was observed in MTAPPL, NCMTA and PPL with no significant difference (p >.05). SB exhibited incomplete mineralized tissue barriers, significantly different from NCMTA, MTAPPL and PPL (p <.05). The staining with CD34 was positive in all the groups on all observation days. Conclusion: The favourable pulpal responses and induction of mineralized tissue formation associated with MTAPPL indicate its potential application as a direct pulp capping material. Objective: To evaluate the shade matching ability of a novel supra‐nano filled esthetic resin composite employing structural color technology using simplified simulated clinical cavities. Filler morphology and light transmittance characteristics were also evaluated. Materials and Methods: One‐hundred and twenty frames of resin composite were built in A1, A2, A3, and A4 shades to simulate Class I cavities (diameter = 4 mm, height = 2 mm). For each shaded frame, cavities were filled with three different types of filler containing resin composites (n = 10): supra‐nano filled (SN filled) resin composite, microhybrid filled (MH filled) resin composite, and clustered‐nano filled (CN filled) resin composite. Color parameters were calculated using CIELAB (△Eab) and CIEDE2000 (△E00). Data were analyzed using one‐way analysis of variance (ANOVA), followed by Duncan's test (α =.05). Filler morphology and light transmittance characteristics were measured to explore the role of structural color on shade matching. Results: △Eab and △E00 of SN filled resin composite were significantly lower in A2, A3, and A4 shades (P <.05). Conclusions: The SN filled resin composite showed better shade matching with A2, A3, and A4 shades of resin composite frames compared to MH filled resin composite, and CN filled resin composite. Clinical Significance: Universal‐shade resin composites, which were expected to match nearly all shades, simplify the restorative procedure. Resin composite, which contained spherical supra‐nano filler particles, could contribute most to its shade matching by stimulating structural color. Our knowledge of the mechanisms responsible for the degradation of resin–dentin bonds are poorly understood. This study investigated the degradation of resin–dentin bonds after 1 year immersion in water. Resin–dentin beams (adhesive area: 0.9 mm2) were made by bonding using a resin adhesive, to extracted human teeth. The experimental beams were stored in water for 1 year. Beams that had been stored in water for 24 h were used as controls. After water storage, the beams were subjected to microtensile bond testing. The dentin side of the fractured surface was observed using FE-SEM. Subsequently, these fractured beams were embedded in epoxy resin and examined by TEM. The bond strength of the control specimens (40.3±15.1 MPa) decreased significantly (p<0.01) after 1 year of water exposure (13.3±5.6 MPa). Loss of resin was observed within fractured hybrid layers in the 1 year specimens but not in the controls. Transmission electron microscopic examination revealed the presence of micromorphological alterations in the collagen fibrils after 1 year of water storage. These micromorphological changes (resin elution and alteration of the collagen fibrils) seem to be responsible for the bond degradation leading to bond strength reduction. [Copyright &y& Elsevier]

The purpose of this study was to determine the bond stability and the change in interfacial ultra-structure of a conventional glass-ionomer cement bonded to dentin, with and without pre-treatment using a polyalkenoic acid conditioner. The occlusal dentin surfaces of six teeth were ground flat. Glass-ionomer cement was bonded to the surfaces either with or without polyalkenoic acid conditioning. The teeth were sectioned into 1-mm2 stick-shaped specimens. The specimens obtained were randomly assigned to two groups with different periods of storage in water: 1 week and 1 year. The micro-tensile bond strength (μTBS) was determined for each storage time. Additional specimens were prepared for Transmission Electron Microscopy (TEM); they were produced with or without prior polyalkenoic acid conditioning in the same way as in the μTBS test. There was no significant difference in μTBS to conditioned dentin and non-conditioned dentin (p > 0.05). The failures appeared to be of a mixed nature, although aging caused more areas of cohesive than adhesive failure in both groups. The TEM observation showed an intermediate layer, a matrix-rich layer and a partially demineralized layer in the polyalkenoic acid conditioned group. Using transmission electron microscopy (TEM), this study investigated the characteristics of smear layers after preparation of dentin surfaces using different methods. In addition, this study analyzed, using scanning electron microscopy (SEM), the interactions of smear layers with the various acidic agents used subsequently to treat the dentin surface. Mid‐coronal dentin surfaces were prepared according to one of five instrumentation methods: fracture; grinding with carbide bur; grinding with regular‐grit diamond bur; grinding with superfine‐grit diamond bur; or grinding with 600‐grit silicon carbide (SiC) abrasive paper. Then, each prepared dentin surface was further divided into control (no further treatment) or treatment with one of the following agents: phosphoric acid solution (PAS); phosphoric acid gel (PAG); Clearfil SE Bond 2 Primer (SE2); or Scotchbond Universal Adhesive (SBU). Longitudinal ultra‐thin sections were made for TEM observations. Dentin discs, representing each surface preparation and treatment method described above, were created, and the morphology was examined by SEM. The TEM results revealed differences in thickness and denseness of the smear layer, according to the different methods of instrumentation used to prepare the dentin surface. The two forms of phosphoric acid completely removed the smear layer in all groups. Agglomerated silica particles were observed on the top of the collagen layer after etching with PAG. For all dentin surfaces, except for that prepared with SiC, SE2 demonstrated better etching efficacy on different smear layers, whereas SBU provided only superficial interaction. Dental materials that are antimicrobial and acid-resistant can inhibit bacterial colonization and demineralization, thereby preventing caries. Zinc and copper are well-known for their antibacterial effect, as is nanostructured ZnO–CuO composite. Minerals such as fluorine and calcium, can remineralize and demineralize teeth. Therefore, we developed novel fluoride-containing ZnO–CuO (ZCF) nanocomposites; to the best of our knowledge, these are the first nanocomposites of this kind. The fluoride concentrations and antibacterial effects of the ZCF nanocomposites were evaluated. Nanocomposites comprising zinc and copper (ZC), and zinc, copper, and fluorine (ZCF), were prepared by a simple one-step homogeneous coprecipitation method at a low temperature (80 °C), without the use of organic solvent or surfactant. The structure and composition of the ZC and ZCF nanocomposites were examined by scanning electron microscopy–energy-dispersive spectroscopy (SEM-EDS). Quantitative analysis of the mass concentration was performed by using ZAF correction methods. The fluorine content in nanocomposites was evaluated by using proton-induced gamma emission (PIGE) at the Takasaki Advanced Radiation Research Institute in Japan. By using 96-well microtiter plates, we analyzed the antibiotic susceptibility of ZC, ZCF, and the control buffer (phosphate-buffered saline) with Streptococcus mutans (ATCC 25175). The SEM images showed that ZC and ZCF nanocomposites were composed of 3D flower-like microstructures with diameters of approximately 1 μm. Environmental SEM-EDS analysis revealed that ZC contained 43.2% Cu, 55.1% Zn, 2.2% F, and 0.1% Cl, whereas ZCF contained 47.5% Cu, 40.5% Zn, 6.7% F, and 5.9% Cl. Analysis by PIGE showed that ZCF nanocomposite contained 2553.6 ± 199.2 ppm fluorine, whereas no fluoride was detected in ZC. The control buffer enabled bacterial growth to 4 × 107 ± 9 × 106 CFU/mL, whereas ZC allowed growth of 12 ± 8 CFU/mL, and ZCF showed no bacterial growth. Thus, we developed novel fluoride-containing ZnO–CuO nanocomposites, which exhibited antibacterial effects and have the potential for remineralization, thereby demonstrating their potential as multifunctional dental materials. There are many kinds of dental luting materials (cements) in the market. Some luting materials are fluoride-releasing to prevent secondary caries. The purpose of this study was to evaluate the release of fluorine from fluoride-containing luting materials to adjacent dentin by in-air micro PIXE/PIGE (µPIXE/PIGE) system at Takasaki Advanced Radiation Research Institute. Each extracted human tooth crown was cut to expose a flat dentin surface. A 2-mm thick acrylic resin plate was cemented to each tooth using one of four fluoride-containing luting materials on the dentin surface: Fuji Luting EX (FL); Fuji I (FO); SA Luting plus (SA); and BeautiCem SA (BC). The specimens were sectioned and exposed to automated pH cycling (pH 4.5 and 7.0; simulate oral condition) for 4 or 8 weeks. After pH cycling, calcium and fluorine distributions were analyzed by the µPIXE/PIGE system with a 1.7-MeV 1H+ microbeam. The cumulative fluorine concentration (fluorine uptake) and calcium concentration in each specimen was calculated at a distance of 100 µm from the outermost surface. The results of our study indicated that FL had the highest concentration of fluorine uptake among all test materials in the 8-week group. The other three materials did not differ significantly from each other in terms of fluorine uptake. FL showed higher calcium content than SA in the 8-week group. Findings from this study suggest that the amount of fluorine penetration into dentin is likely to be dependent on the amount of fluoride released from each material, and also related to the amount of calcium present. Objectives: To evaluate the effects of air-blowing time and storage time on microtensile bond strength (μTBS) of universal adhesives to dentin. Materials and methods: Ninety flat dentin surfaces from extracted human third molars were bonded with three universal adhesives (Clearfil Universal Bond-CU; G-Premio Bond-GP; Scotchbond Universal Adhesives-SB). Bonded dentin surfaces were air-dried for 5 s, 15 s, or 30 s followed by resin composite built-up. Resin-dentin beams were tested with μTBS test after different storage time in distilled water (24 h and 1 year). Data were analyzed by three-way ANOVA and Duncan test at (α = 0.05). Failure mode and resin-dentin interfaces were observed using a scanning electron microscope (SEM). Specific features of fractured beams after μTBS were further observed using SEM at high magnification. Results: Extension of air-blowing time from 5 s to 30 s increased the 24 h μTBS of CU only. Bond strength of all adhesives significantly decreased after 1-year storage except for CU at 5 s and 30 s of air-blowing time. One-year μTBS were significantly higher when air-blowing times were extended to 15 s for SB and 30 s for CU. Air-blowing time had no influence on GP. Conclusion: The effect of air-blowing time and storage time on resin-dentin bond was material-dependent. Clinical relevance: Extended air-blowing time increased the bond strength and bond durability of CU. Human antigen R (HuR) is a RNA-binding protein, which binds to the AU-rich element (ARE) in the 3′-untranslated region (3′-UTR) of certain mRNA and is involved in the export and stabilization of ARE-mRNA. HuR constitutively relocates to the cytoplasm in many cancer cells, however the mechanism of intracellular HuR trafficking is poorly understood. To address this question, we examined the functional role of the cytoskeleton in HuR relocalization. We tested the effect of actin depolymerizing macrolide latrunculin A or myosin II ATPase activity inhibitor blebbistatin for HuR relocalization induced by the vasoactive hormone Angiotensin II in cancer and control normal cells. Western blot and confocal imaging data revealed that both inhibitors attenuated the cytoplasmic HuR in normal cells but no such alteration was observed in cancer cells. Concomitant with changes in intracellular HuR localization, both inhibitors markedly decreased the accumulation and half-lives of HuR target ARE-mRNAs in normal cells, whereas no change was observed in cancer cells. Furthermore, co-immunoprecipitation experiments with HuR proteins revealed clear physical interaction with ß-actin only in normal cells. The current study is the first to verify that cancer cells can implicate a microfilament independent HuR transport. HuR is an RNA-binding protein of the embryonic lethal abnormal vision (ELAV) family, which binds to the AU-rich element (ARE) in the 3′-untranslated region (UTR) of certain mRNAs and is involved in the nucleo-cytoplasmic export and stabilization of ARE-mRNAs. The cytoplasmic relocalization of ARE-mRNAs with several proteins such as HuR and pp32 increases in cells transformed by the adenovirus oncogene product E4orf6. Additionally, these ARE-mRNAs were stabilized and acquired the potential to transform cells. Although, the relocalization of HuR and the stabilization of ARE-mRNAs are crucial for cell transformation, evidence regarding the relationship of HuR and ARE-mRNAs with adenovirus replication is lacking. In this report, we demonstrate that adenovirus infection induces the relocation of HuR to the cytoplasm of host cells. Analysis using the luciferase-ARE fusion gene and the tetracycline (tet)-off system revealed that the process of stabilizing ARE-mRNAs is activated in adenovirus-infected cells. Heat shock treatment or knockdown-mediated depletion of HuR reduced adenovirus production. Furthermore, expression of ARE-including viral IVa2 mRNA, decreased in HuR-depleted infected cells. These results indicate that HuR plays an important role in adenovirus replication, at least in part, by up-regulating IVa2 mRNA expression and that ARE-mRNA stabilization is required for both transformation and virus replication. This study evaluated the effect of topical fluoride application on enamel hardness after in-office bleaching. Twelve human incisors were cut along the long axis, resulting in 24 halves used in four treatment groups ( n = 6 in each group): (i) untreated group (C); (ii) in-office bleaching material (B); (iii) treatment with surface reaction-type prereacted glass-ionomer varnish after in-office bleaching (B+ PRG); and (iv) treatment with acidulated phosphate fluoride solution after bleaching (B+F). All specimens were subjected to pH-cycling for 4 wk. Knoop hardness was measured using a Cariotester. The decalcification of enamel was assessed quantitatively by measuring the integrated mineral loss (Δ IML). Games-Howell analysis was used to assess statistical significance of between-group differences. The Knoop hardness decreased significantly after bleaching for all groups. In treatment groups B+ PRG and B+F, the Knoop hardness returned to the original unbleached values after the first pH cycle and did not change afterwards. In treatment groups C and B there was a gradual decrease in the Knoop hardness until the fourth pH cycle. The integrated mineral loss, Δ IML, was significantly higher in treatment group B+F after 2 wk than in the other treatment groups. After 4 wk, the Δ IML in treatment group B was significantly higher than in treatment group B+ PRG. Objectives To evaluate the influence of different air-blowing durations on the micro-tensile bond strength (μTBS) of five current one-step adhesive systems to dentin. Methods One hundred and five caries-free human molars and five current one-step adhesive systems were used: ABU (All Bond Universal, Bisco, Inc.), CUB (CLEARFIL™ Universal Bond, Kuraray), GPB (G-Premio BOND, GC), OBA (OptiBond All-in-one, Kerr) and SBU (Scotchbond Universal, 3M ESPE). The adhesives were applied to 600 SiC paper-flat dentin surfaces according to each manufacturer’s instructions and were air-dried with standard, oil-free air pressure of 0.25 MPa for either 0 s, 5 s, 15 s or 30 s before light-curing. Bond strength to dentin was determined by using μTBS test after 24 h of water storage. The fracture pattern on the dentin surface was analyzed by SEM. The resin–dentin interface of untested specimens was visualized by panoramic SEM image. Data from μTBS were analyzed using two-way ANOVA (adhesive vs. air-blowing time), and Games-Howell (a = 0.05). Results Two-way ANOVA revealed a significant effect of materials (p = 0.000) and air-blowing time (p = 0.000) on bond strength to dentin. The interaction between factors was also significantly different (p = 0.000). Maximum bond strength for each system were recorded, OBA/15 s (76.34 ± 19.15 MPa), SBU/15 s (75.18 ± 12.83 MPa), CUB/15 s (68.23 ± 16.36 MPa), GPB/30 s (55.82 ± 12.99 MPa) and ABU/15 s (44.75 ± 8.95 MPa). The maximum bond strength of OBA and SUB were significantly higher than that of GPB and ABU (p < 0.05). Significance The bond strength of the current one-step adhesive systems is material-dependent (p = 0.000), and was influenced by air-blowing duration (p = 0.000). Objective Bioactive glasses and surface pre-reacted glass-ionomer (sPRG) filler possess cariostatic properties owing to ion release. Many studies investigated potential cariostatic effects; few studies evaluated the surface stability and the structural changes their surfaces undergo in acidic conditions. Methods The surface resistance against acid attack and the surface receptiveness for bacterial adhesion and biofilm formation of a sPRG-filled (Beautifil ll, Shofu) and conventional glass-filled (Herculite XRV Ultra, Kerr) resin-based composite (RBC), and a conventional glass-ionomer cement (GIC; Fuji IX GP Extra, GC) were examined. Specimens (n = 3) were immersed in distilled water or lactic acid (pH 4.0) for 3 days. Bacterial growth and biofilm formation were recorded using optical density and SEM. Results Upon 3-day immersion in lactic acid, the surface of the sPRG-filled RBC revealed multiple holes, while virtually no change in surface integrity was observed for the conventional RBC and GIC. Bacterial growth measurements revealed that none of the materials inhibited Streptococcus mutans (p < 0.05). Remarkably, cross-sectional SEM revealed that S. mutans had penetrated the etch pits induced by lactic acid in/around the sPRG filler. Ion-release measurements revealed that sPRG-filled RBC released boron and fluoride, while GIC only released fluoride. However, the concentration of ions released by both materials appeared not sufficient to inhibit bacterial growth. Moreover, the structural surface change and resultant increased surface roughness appeared to have promoted biofilm formation. Significance While having bioactive potential through ion release, the stability of surface integrity of bioactive materials is a key-parameter to be assessed with regard to their cariostatic potential. Objective: This study evaluated the effects of surface preparation and the application time of adhesives on the resin-dentine bond strengths with universal adhesives.Methods: Sixty molars were cut to exposed mid-coronal dentine and divided into 12 groups (n=5) based on three factors; (1) adhesive: G-Premio Bond (GP, GC Corp., Tokyo, Japan), Clearfil Universal Bond (CU, Kuraray Noritake Dental Inc., Okayama, Japan) and Scotchbond Universal Adhesive (SB, 3M ESPE, St. Paul, MN, USA); (2) smear layer preparation: SiC paper ground dentine or bur-cut dentine; (3) application time: shortened time or as manufacturer's instruction. Fifteen resin-dentine sticks per group were processed for microtensile bond strength test (μTBS) according to non-trimming technique (1mm(2)) after storage in distilled water (37 °C) for 24h. Data were analyzed by three-way ANOVA and Dunnett T3 tests (α=0.05). Fractured surfaces were observed under scanning electron microscope (SEM). Another 12 teeth were prepared and cut into slices for SEM examination of bonded interfaces.Results: μTBS were higher when bonded to SiC-ground dentine according to manufacturer's instruction. Bonding to bur-cut dentine resulted in significantly lower μTBS (p<0.000). Shortening the application time resulted in significantly lower bond strength for CU on SiC and GP on bur-cut dentine. SEM of fractured surfaces revealed areas with a large amount of porosities at the adhesive resin interface. This was more pronounced when adhesives were bonded with a reduced application time and on bur cut dentine.Clinical Significance: The performance of universal adhesives can be compromised on bur cut dentine and when applied with a reduced application time. The buffering capacity and inhibitory effects on enamel demineralization of two commercially available dental sealants were evaluated in this study. The effects of filler particles were also examined. Disks of enamel and cured sealant materials of BeautiSealant (silica or S-PRG filler) or Teethmate F-1 were incubated in lactic acid solutions ( pH 4.0) for 1-6 d. The pH changes and amounts of ions released in the solutions were assessed, and enamel surfaces were observed using a scanning electron microscope. The pH of the solution with BeautiSealant (S-PRG filler) was neutralized from pH 4.0 to pH 6.1 (after incubation for 1 d) and from pH 4.0 to pH 6.7 (after incubation for 6 d). In addition, no release of calcium ions was detected and the enamel surface was morphologically intact in scanning electron microscopy images. However, the pH of the solution with Teethmate F-1 remained below pH 4.0 during incubation from days 1 to 6. Calcium release was increased in solutions up to and after 6 d of incubation. Scanning electron microscopy images showed that the structures of hydroxyapatite rods were exposed at the specimen surfaces as a result of demineralization. Ions released from S-PRG filler-containing dental sealant rapidly buffered the lactic acid solution and inhibited enamel demineralization. [ABSTRACT FROM AUTHOR]

Abstract: The use of fluoride for the prevention of caries is based on the transformation of hydroxylapatite to fluoroapatite in the presence of fluoride ions, thereby strengthening tooth structure. Adhesion of dentin and resin composite (tooth-colored restoration material) requires a dentin bonding system, since resin composite is not able to adhere to dentin directly. Demineralization of dentin by acid etching is an important step in the dentin bonding system, however, demineralization also introduces weaknesses in tooth structure. If the demineralized dentin could be strengthened by the application of fluoride, then the dentin–resin composite bond strength might also improve. To test this hypothesis, the present study evaluated the influence of fluoride applications on the strength of the dentin–resin composite bond by (1) tensile strength testing analyses, (2) SEM analyses of tooth structure, and (3) detection of calcium (Ca) and fluorine (F) distribution patterns by micro proton-induced X-ray emission (μ-PIXE) and micro proton-induced gamma-ray emission (μ-PIGE) analyses conducted at the Takasaki Ion Accelerators for Advanced Radiation Application (TIARA) at the Takasaki Advanced Radiation Research Institute (TARRI). In this study, the dentin in extracted human molars was exposed by grinding and the dentin was etched with 35% phosphoric acid. Fluoride was applied at two concentrations, 0.022% (100ppm F) and 2.21% (10,000ppm F) NaF solution, for two time periods, 30 and 60s, prior to bonding the resin composite with the treated dentin. Controls were prepared in the same manner, but without the fluoride application. Bond strength was measured with a micro-tensile testing unit, and the fluorine and calcium distributions at the interface between dentin and resin composite were detected by μ-PIGE and μ-PIXE analysis, respectively. Results indicate that the 10,000ppm F applications resulted in higher bond strengths than observed in either the 100ppm F applications or the control group. In addition, PIGE analyses showed high concentrations of fluorine in the hybrid bonding layer of the 10,000ppm F samples, suggesting that the fluorine contributes to the strength of the dentin–resin composite bond. Here we used atomic force microscopy in the molecular force mode to measure the specific interaction forces between antigen I/II and two matrix proteins, collagen and fibronectin. These matrix proteins provide important binding sites for adherence of oral streptococcal in dentinal caries and endocarditis, respectively. Antigen I/II-coated cantilever tips were brought into contact with collagen- or fibronectin-coated silica coverslips. For the protein I/II-fibronectin interaction experiments, the mean strength of the last ruptures was 216 pN, with most of the detachments located around 125 pN. In antigen I/II-collagen interaction experiments, the mean strength of the last rupture forces corresponded to 136 pN, with the most frequent unbinding force around 75 pN. Thus, our findings definitely suggest that, under the present experimental conditions, antigen I/II binds more strongly to fibronectin than to type I collagen. Abstract: Objectives: This study investigated the effects of light penetration and removal of smear layer on the adhesive properties of self-etching adhesives to root canal dentin when using fiber posts. Methods: Altogether 54 human incisors and premolars were decoronated, and the roots after preparing the post space were randomly assigned into six groups. Three groups were treated with a light-cured self-etching adhesive and the other three with a dual-cured self-etching adhesive. In a further sub-division, the first group was light-cured with an irradiation unit; the second was light-cured with the irradiation unit and a light-guiding attachment; and the third was light-cured with the irradiation unit and the attachment after removing smear layer by EDTA and NaOCl. Then, a glass-fiber post was luted into the post space by a dual-cured composite. After 24h, an hourglass-shaped specimen was trimmed and subjected to MTBS testing. Results: Using the light-guiding attachment and removing the smear layer reduced the incidence of pre-testing failure significantly from 57.1% to 19.0% (p <0.05) in the light-cured adhesive groups and from 68.3% to 3.2% (p <0.01) in the dual-cured adhesive groups. The MTBS in different locations within a post space were in the range from 8.9 to 17.5MPa in the light-cured group and from 11.2 to 17.2MPa in the dual-cured group. Significance: Better light penetration in post spaces and removal of the smear layer are effective in improving the adhesive properties of self-etching adhesives to root canal dentin when using fiber posts. Since adhesive technology was introduced into dental field, metal-based restoration has been gradually replaced by metal-free restoration. Using the adhesive technology, minimum invasive technique has been possible in daily clinical practice as well as esthetic tooth-colored restorations have become very popular all over the world. One of the current issues of the dental adhesive is durability of bond between tooth structure and adhesive resin. Several approaches to overcome the issues have been carried out. Self-etching approach is believed to create durable bond because demineralization of superficial tooth surface is very shallow. Other approach is to utilize the inhibitor of enzymes which are suggested to catalyze the decomposition of resin composites and are always secreted within the oral environment. In the present study, Colloidal Platinum Nanoparticles (CPN) was applied before the application of 4-META/MMA-TBB resin cement as the third possibility to prolong the durability of bond. Abstract: Objectives: The purpose of this study was to evaluate the ability of an experimental self-etching resin coating system to protect against demineralization of the root surface in vitro, comparing its effectiveness with two commercial one-step self-etching adhesives. Methods: The experimental resin coating system (EC), consisting of a self-etching primer and an acrylate-based resin, or the two commercial self-etching adhesives, Adper Prompt-L Pop (AD) and Clearfil Tri-S Bond (S3), was applied to the cementum of a human extracted tooth root. The specimens were immersed in acetate buffer at pH 5.5 for 4 weeks, and the degree of demineralization underneath the materials was evaluated by microscopic observation and contact microradiography. Each material''s sealing and interfacial ultrastructure on the cementum was morphologically examined by scanning electron microscopy, and their bonding ability to cementum was determined by microtensile bond strength (MTBS) tests. Results: Application of AD and S3 showed limited effects to protect against demineralization, but no demineralization was observed for EC. EC showed greater coating thickness than the other two materials, with formation of integral hybridized layers with cementum. Although the MTBS values for EC were significantly lower than those for AD and S3, cohesive failure in resins was the prevalent fracture mode for EC, showing no possible disadvantages in bonding ability. Significance: The experimental resin coating system effectively prevented acid demineralization on root surfaces due to its ability to produce an appropriately thick coating and an integral hybridized layer. Abstract: Objectives: The objectives of this study were (1) to clarify the relationship between the duration of air-drying of one-step self-etch adhesives (1-SEAs) and the evaporation degree (ED) of solvents, and (2) to evaluate the effect of ED on the ultimate micro-tensile strength (μTS) of the adhesives. Methods: The ED of one HEMA-rich 1-SEA, Clearfil S3 Bond (Kuraray), and two HEMA-free 1-SEAs, iBond (Heraeus-Kulzer) and G-Bond (GC), was determined without air-drying and after, respectively, 5- and 10-s air-drying using a gravimetric method. Next, the μTS of the adhesives at ED equivalent to 0-, 5- and 10-s air-drying was measured. Results: The ED increased with extension of air-drying time. Among the adhesives tested, iBond showed the largest ED, followed by G-Bond and Clearfil S3 Bond in this order. A longer air-drying time for 10-s resulted in a statistically significantly higher μTS for the HEMA-rich Clearfil S3 Bond. The μTS of the latter was higher than that of the other two HEMA-free adhesives for each air-drying time. Significance: Air-drying of 1-SEAs had a significant effect on the degree of solvent evaporation (ED) and also on the mechanical properties (μTS) of the 1-SEAs upon setting. Abstract: Objective: Phase-separation within HEMA-free all-in-one dental adhesives may result in the entrapment of droplets within the adhesive resin. Strongly air-blowing prior to polymerization, can remove most of these droplets. The objective of this study was to evaluate the effect these droplets may have on the resistance of the adhesive–tooth interface to NaOCl degradation. Methods: The micro-tensile bond strength (μTBS) to enamel and dentin was determined when a HEMA-free all-in-one adhesive was applied either following a mild or strong air-blowing technique. The bonds were also exposed to an aqueous sodium hypochlorite (NaOCl) solution for 1h, following a recently introduced methodology to mimic in vivo bond degradation. Results: This study revealed that strong air-blowing of the adhesive only resulted in a significantly higher micro-tensile bond strength (μTBS) to dentin, but not to enamel. Likewise, NaOCl only reduced the μTBS to dentin for both the mild and strong air-blowing technique, but again not the μTBS to enamel. Failure analysis by SEM clearly revealed that strong air-blowing is less effective in droplet removal when the adhesive was applied in small and narrow class-I cavities, as compared to when it was applied to flat surfaces. Conclusions: NaOCl did preferentially dissolve the hybrid layer at dentin, and more for the mild than for the strong air-blowing technique. A strong air-blowing procedure resulted in a more NaOCl-resistant hybrid layer, so that it can be concluded that a HEMA-free one-step adhesive definitely benefits from a strong air-blowing technique. It has demonstrated that the recognition of triacylated lipopeptides by Toll-like receptor (TLR) 2 requires TLR1 as a coreceptor. In the NF-κB reporter assay system in which human embryonic kidney 293 cells were transfected with TLR2 and TLR1 together with an NF-κB luciferase reporter gene, S-(2,3-bispalmitoyloxypropyl)-N-palmitoyl-Cys-Lys-Lys-Lys-Lys (Pam3CSK4) and Pam3CSSNA were recognized by TLR2/TLR1, but the recognition level was unexpectedly very low. However, cotransfection of CD14 drastically enhanced the recognition of triacylated lipopeptides by TLR2/TLR1. The CD14-induced enhancement did not occur without cotransfection of TLR1. Both CD14dS39-A48, a mutant with deletion of the part of possible N-terminal ligand-binding pocket, and anti-CD14 monoclonal antibody reduced the CD14-induced enhancement. Transfection of a TIR domain-deficient mutant of TLR2 (TLR2dE772-S784) or TLR1 (TLR1dQ636-K779) completely abrogated the CD14-induced enhancement. Soluble recombinant CD14 added extracellularly enhanced the recognition of Pam3CSSNA by TLR2/TLR1. Immunoprecipitation analysis demonstrated that CD14 was not associated with TLR2 but that TLR1 was associated with TLR2. In addition, surface plasmon resonance-based assay demonstrated that CD14 binds to Pam3CSK4 at a dissociation constant of 5.7 µM. This study suggests that CD14 directly binds to triacylated lipopeptides and facilitates recognition of the lipopeptides by the TLR2/TLR1 complex without binding to the receptor complex. Abstract: Thioredoxin reductase (TrxR) reduces thioredoxin (Trx), thereby contributing to cellular redox balance, facilitating the synthesis of deoxy-ribose sugars for DNA synthesis, and regulating redox-sensitive gene expression. Auranofin is a gold compound that potently inhibits TrxR. This inhibition is one suspected mechanism of auranofin’s therapeutic benefit in the treatment of rheumatoid arthritis. The use of other gold compounds to treat cancer or inflammatory disease may rely on their ability to inhibit TrxR. In the current study, we tested the hypothesis that a variety of gold compounds may inhibit TrxR. Methods: We exposed rat-TrxR1 to auranofin, gold sodium thiomalate, sodium aurothiosulfate, triphenyl phosphine gold chloride, or gold acetate, and measured TrxR activity ex vivo. We then compared TrxR1 inhibitory levels of gold compounds to those that inhibited mitochondrial activity of THP1 monocytes and OSC2 epithelial cells, estimated by succinate dehydrogenase activity. Results: All gold compounds inhibited TrxR1 at concentrations ranging from 5 to 4000nM (50% inhibitory concentration). The oxidation state of gold did not correlate with inhibitory potency, but ligand configuration was important. Au(I)-phosphine compounds (triphenyl phosphine gold chloride and auranofin) were the most potent inhibitors of TrxR. All TrxR1 inhibitory concentrations were sublethal to mitochondrial activity in both THP1 and OSC2 cells. Conclusions: Diverse types of gold compounds may be effective inhibitors of TrxR1 at concentrations that do not suppress cellular mitochondrial function. Inhibition may be optimized to some degree by altering the ligand configuration of the compounds. Summary: Objectives: This study evaluated the effect of errors commonly made in using total-etch adhesives, on the resulting bond strength, fluid movement and nanoleakage of resin dentin bonds. Methods: Two total-etch adhesives were used for bonding to dentin according to the manufacturers'' recommendations, with meticulous solvent evaporation (control), or with the introduction of common bonding errors—wet bonding without solvent evaporation (no evaporation), and dry bonding. Results: The 24-hour bond strength of the control was significantly higher than the other groups (p <0.05). For all groups, the higher initial permeability declined significantly after 24h. The fluid movement across bonded dentin was similar in the control and dry bonding for both adhesives, whereas significantly higher permeability (p<0.05) was recorded for the no evaporation groups even after 24h. Extensive silver impregnation within hybrid layers was seen by TEM in the no evaporation and dry bonding specimens after 24h. Dry bonding caused collapse of the collagen matrix and interfered with resin infiltration. In contrast, inadequate solvent evaporation and/or residual water during dentin bonding results in dilution or incomplete polymerization of the resin, leading to severe nanoleakage formation. Significance: Increased permeability associated with incomplete solvent evaporation in total-etch adhesives may lead to poor bond strength. [Copyright &y& Elsevier]

Summary: Purpose: To evaluate the technique sensitivity of four different adhesive systems using different air-blowing pressure. Methods: Four adhesive systems were employed: Clearfil SE Bond [SE] (Kuraray, Japan), G-Bond [GB] (GC Corporation, Japan), Adper Prompt L-Pop [LP] (3M ESPE, USA) and an experimental adhesive, SSB-200 [SSB] (Kuraray, Japan). Twenty-four extracted molars were used. After grinding the coronal enamel surface, the teeth were divided into two equal groups. The first group''s teeth were randomly assigned for bonding with the different adhesives using gentle air-blowing (g). For the teeth of the second group, the four adhesive systems were applied using strong air-blowing (s). After storage overnight in 37°C water, the bonded specimens were sectioned into sticks (1mm×1mm wide), which were subjected to microtensile bond strength testing (μTBS) at a crosshead speed of 1mm/min. The load at failure of each specimen was recorded and the data were analyzed by one-way ANOVA and Tukey HSD tests. The surfaces of the fractured specimens were observed using SEM to determine the failure mode. Results: The results of the μTBS test showed that the highest bond strengths tended to be with SE for both gentle and strong air-blowing, and the significantly lowest for SSB with strong air streaming. Comparing the two techniques, significant differences were noted only for SSB-200 (P<0.05). For each material, the SEM evaluation did not show distinct differences in the nature of the fractures between the two techniques, except for SSB-200. Summary: Objective: The purpose of the present study was to evaluate the effect of evaporation of primer components on the ultimate tensile strength (UTS) of primer–adhesive mixtures. Methods: The UTSs of 1:3 by weight primer–adhesive mixtures and of the pure adhesives (controls) were measured after 0-, 2-, 5-min and 24-h storage in a 37°C oven for the three-step etch&rinse adhesive OptiBond FL (Kerr) and the two-step self-etch adhesive Clearfil SE (Kuraray). Results: Evaporation of primer components increased with time for both adhesives investigated. OptiBond FL showed a higher evaporation of primer components than Clearfil SE at the 2-, 5-min and 24-h storage time. As compared to the pure adhesive, the UTS of the primer–adhesive mixture was lower after 0- and 2-min storage for OptiBond FL and after 0-, 2- and 5-min storage for Clearfil SE. The UTS of OptiBond FL was higher than that of Clearfil SE at the 0- and 2-min storage time, though there was no difference at the 5-min and 24-h storage time. Significance: The degree of evaporation of primer components depended largely on the primer solvents employed. The significant decrease in UTS of adhesive resin when mixed with primer must be attributed to incomplete evaporation of primer components. Summary: Objectives: To evaluate the effect of double-application of all-in-one adhesives using the micro-shear bond test. Methods: The occlusal surfaces of extracted human third molars were ground perpendicular to the long axis of each tooth to expose a flat dentin surface. Three commercially available and one experimental bonding system were used in this study: Adper Prompt L-Pop (APL, 3M ESPE), REACTMER BOND (RB, Shofu), XENO III (Xeno, Dentsply-Sankin) and newly developed OBF-2 (OB2, Tokuyama Dental). These adhesives were applied on the dentin surfaces by either the manufacturers'' instructions or by an experimental method (single-application or double-application). Resin composite was then placed and light-cured for 40s. After 24h immersion in water, a micro-shear bond test was carried out and the fractured dentin surfaces were observed microscopically. The data were analyzed by one-way ANOVA and Tukey''s HSD tests (p<0.05). Results: The mean bond strengths of APL, RB, Xeno and OB2 with single-application were 22.7, 28.3, 30.3 and 34.6MPa, respectively. The mean bond strengths of APL, RB, Xeno and OB2 with double-application were 29.5, 27.2, 29.6 and 32.5MPa, respectively. There were no statistically significant differences in micro-shear bond strengths between the single- and double-application methods for each adhesive system (p>0.05). The morphological observation of the fractured dentin surfaces revealed differences between the single- and double-application groups especially for APL and OB2. Conclusions: Micro-shear bond strengths of all-in-one adhesives in this study showed no statistically significant differences between the single-application and the double-application method. Summary: Objective: The purpose of this study was to measure the micro-tensile bond strength (μTBS) of two adhesives to dentin and to correlate this μTBS with the micro-tensile fracture strength (μTFS) of primer–adhesive mixtures of the two adhesives. Methods: The three-step etch&rinse adhesive OptiBond FL (Kerr) and the two-step self-etch adhesive Clearfil SE Bond (Kuraray) were used to bond Filtek Z100 (3M ESPE) to twenty mid-coronal dentin surfaces. The μTBS was determined at 1 and 24h after light-curing. In addition, the μTFS of primer–adhesive mixtures (primer:adhesive=1:3 by weight) and of the pure adhesives (controls) were measured after the same time intervals. Results: The μTBS to dentin of OptiBond FL at 24h was significantly higher than at 1h and than the μTBS of Clearfil SE Bond at 24h. The mean μTFS of the primer–adhesive mixtures was always lower than the μTFS of the respective adhesives. The highest reduction in μTFS due to mixing primer with the adhesive resin was measured for Clearfil SE Bond. Both the μTFS of the primer–adhesive mixtures and the pure adhesives of OptiBond FL and Clearfil SE Bond increased with time. Significance: The early 1h μTBS to dentin was lower than the 24h μTBS for OptiBond FL, but not for Clearfil SE Bond. The CIELAB parameters (L*, a* and b*) of disks of A2 and opaque A2 (OA2) shades of Charisma (Heraeus-Kulzer), Solare (GC) and Filtek Supreme (3M) were evaluated on backings of black, white and the material itself to calculate the translucency parameter (TP) and the colour differences (ΔE*) between A2 and OA2. A two-way analysis of variance (anova) for the TP indicated a less statistically significant TP value in the OA2 shade than the A2 shade for all products. As for the products, Charisma showed a statistically greater TP value than the other two products. Regarding theΔE* between A2 and OA2, all the products revealed clinically perceptible colour differences (ΔE* > 3.3). Hence, we must take the colour differences of opaque-shade resin composites into consideration, as well as the translucency of the materials, for a clinically acceptable colour match of the restoration. The adenovirus E4orf6 is a viral oncoprotein known to cooperate with the E1A gene product in transforming primary murine cells. It has been shown to inhibit the apoptotic activities of p53 and p73 through direct binding to these proteins. Here, we demonstrate that the adenovirus E4orf6 protein inhibits apoptosis mediated by BNIP3 and Bik, which are BH3-only proteins of the Bcl-2 family. This activity was not mediated by p53 and p73 because E4orf6 had the same effect on the apoptosis in Saos-2 cells that do not express p53-related genes. It was also ascertained that E4orf6 could change the mitochondrial localization of BNIP3 and Bik. A mutant lacking the nuclear export signal of E4orf6 failed to inhibit apoptosis and to translocate BNIP3 protein from the mitochondria. Moreover, it was also established that E4orf6 was able to interact with BNIP3 and Bik. In BNIP3 protein, the region required for the interaction included the transmembrane domain, which is required for the localization of BNIP3 to the mitochondria. Self-etching primer adhesives have recently been introduced to simplify bonding. However, the fundamental bonding mechanism of self-etching primers to enamel is not yet fully understood. This study aimed to investigate resin–enamel bonds of self-etching primer adhesives on ground enamel. Two self-etching primer adhesives (Clearfil Liner Bond 2 V and Clearfil SE Bond) were used in this study. A total-etch adhesive (One-Step) was used as a control. Resin–enamel beams were subjected to the microtensile bond test. Subsequently, the failure modes of all specimens were quantified using image analysis. Undemineralized and demineralized ultrathin sections of the resin–enamel bonded specimens were examined using transmission electron microscopy. The bond strengths of Clearfil SE Bond (39.8 ± 11.9 MPa) and One-Step (46.2 ± 12.7 MPa) were significantly greater than that of Clearfil Liner Bond 2V (30.4 ± 6.2 MPa). Scanning electron microscopy of the fractured surfaces revealed the failure direction and weakest portion within each bond. Transmission electron microscopy showed a thin hybridized complex of resin in enamel produced by the self-etching primers without the usual micrometer-sized resin tags seen in resin–enamel bonds produced using the total-etch adhesive. Objectives. The aim of this study was to investigate the hypothesis that dental material components alter cytokine secretion from monocytes if applied for several weeks at sublethal doses. The current study significantly extended exposure times of monocytes to the components over times published in previous studies. These exposure times approached the estimated average life span of monocytes in the bloodstream.Methods. Human THP-1 monocytes were exposed to 2-hydroxyethylmethacrylate (HEMA, 0–1.2 mmol/l), triethyleneglycoldimethacrylate (TEGDMA, 0–0.75 mmol/l), Hg2+ (0–2 μmol/l), or Ni2+ (0–20 μmol/l) for 2 weeks. The cells were then collected and additionally incubated for 24 h, with or without bacterial lipopolysaccharide (LPS), a common component of dental plaque. TNF-α secretion from THP-1 was determined using by enzyme-linked immunosorbent assay.Results. None of the dental material components induced TNF-α from THP-1 by themselves, but LPS alone strongly induced TNF-α secretion as expected. HEMA and TEGDMA significantly suppressed (40–70%) TNF-α secretion from cells stimulated with LPS. Hg2+ at 2.0 μmol/l doubled TNF-α secretion from THP-1s stimulated with LPS over LPS alone. Ni2+ did not significantly affect TNF-α secretion, with or without LPS exposure.Significance. The results in this study suggest that sublethal, 2-week exposures of some dental material components may alter TNF-α secretion from THP-1 monocytes when the cells are challenged. Objectives. The submicron hiatus represents a potential space between the base of the collagen network and the mineralized dentin when dentin is acid-etched for bonding. These spaces were observed in SEM studies after acid-etched dentin specimens were critical point dried or dehydrated in hexamethyldisilasane. However, they have never been identified in TEM studies of dentin hybrid layers. This study critically examined the cause of submicron hiati formation using a silver staining technique to measure nanoleakage.Methods. Two multi-step, total-etch adhesives (One-Step, Bisco; Single Bond, 3M) and two single-step, self-etching adhesives (Prompt L-Pop, ESPE; One-Up Bond F, Tokuyama) were examined. Flat dentin surfaces were bonded with these adhesives and a lining composite. In each adhesive group, 0.8 mm thick slabs from the same bonded tooth were coated with nail varnish applied 1 mm from the bonded interfaces. The varnish was either left to dry completely for 10 min before immersing in 50 wt% silver nitrate (AgNO3) for 24 h (group D), or painted on blotted tooth slabs that were immediately dropped into the AgNO3 solution (group M). After developing, undemineralized, unstained, epoxy resin-embedded sections were prepared for transmission electron microscopy (TEM) to identify the amount and distribution of silver uptake.Results. Nanoleakage patterns were observed in all adhesive-bonded teeth, regardless of brand. Fine reticular silver deposits were also found in the underlying undemineralized dentin. In group D, submicron hiati were seen as tunnels of heavy silver deposits beneath hybrid layers. Specifically, a hiatus occurred between the undemineralized intertubular dentin and a cohesively fractured layer of the same matrix that was attached to the base of the hybrid layer. Hiati were completely absent in group M, regardless of the brand of adhesive.Significance. This study evaluated an experimental two-step self-etch adhesive (BZF-29, BZF) by comparing it with a reference two-step self-etch adhesive (Clearfil Megabond 2, MB) and a universal adhesive (G-Premio Bond, GP) for microtensile bond strength (μTBS) and resin-dentin interfacial characteristics. Twenty-four human third molars were used for the μTBS test. Bonded peripheral dentin slices were separated to observe the resin-dentin interface and measure the adhesive layer thickness with SEM. μTBS data of the central beams were obtained after 24 h and 6 months of water storage. Fracture modes were determined using a stereomicroscope and SEM. Nine additional third molars were used to determine the elastic modulus (E) employing an ultra microhardness tester. Water storage did not affect μTBS of the tested adhesives (p > 0.05). μTBS of BZF and MB were similar but significantly higher than GP (p < 0.05). BZF achieved the highest adhesive layer thickness, while GP the lowest. E of BZF and MB were comparable but significantly lower than GP (p < 0.05). Except for GP, the predominant fracture mode was nonadhesive. The superior bonding performance of BZF and MB could be attributed to their better mechanical property and increased adhesive thickness imparting better stress relief at the interface. The complex, dynamic, and hydrated microstructures of human dentin serve as the major determinant for the restorative performance of biomaterials. This study aimed to evaluate the mechanical properties of human dentin under different hydration conditions. The occlusal dentin of five third molars was exposed and cut into 1 mm2 dentin slabs. The slabs were then polished and further cut into 1 mm2 dentin beams and stored in distilled water. Two beams/tooth were used for testing their hardness (H) and elastic modulus (E) at 5 min (baseline), 1 h, and 24 h after dehydration (23 °C and 30% RH), and also for measuring weight at following dehydration times: 0 min, 5 min, 1 h, and 24 h. Five additional molars were employed to prepare 0.4 mm2 dentin beams (3/tooth) for determining ultimate tensile strength (UTS) at 5 min (baseline), 1 h, and 24 h post-dehydration. Statistical significance was set at α = 0.05. Dehydration time significantly affected H, E, weight-loss, and UTS of dentin (p < 0.05). H and E values showed a strongly positive and significant correlation (r > 0.5, p < 0.05).
