Os avanços da química analítica instrumental permitiram uma melhoria substancial na obtenção de informações importantes em diferentes áreas do conhecimento, dentre elas a Ciência do Solo. Este trabalho abordou questões relacionadas à análise química de solo por espectrometria de emissão ótica com plasma indutivamente acoplado (ICP OES). Estudos envolvendo correlação de resultados, determinação simultânea multielementar e interferências da técnica de ICP OES foram abordados em quatro capítulos: 1) revisão bibliográfica sobre ICP OES abordando funcionamento, interferências e usos com diferentes extratores; 2) teores de potássio, cálcio, magnésio, alumínio, enxofre, cobre, zinco, manganês e boro determinados por ICP OES em amostras de solo do sul do Brasil; 3) extração multielementar pela solução de Mehlich-3 e determinação simultânea por ICP OES em amostras de solo; 4) eficácia do Multicomponent Spectral Fitting (MSF) na estimativa do valor correto de fósforo determinado por ICP OES. As principais conclusões foram: 1) relações lineares significativas entre teores dos K, Ca, Mg, Al, S, Cu, Zn, Mn e B determinados por ICP e técnicas de referência; 2) a técnica ICP apresentou maior sensibilidade em relação a titulação para determinar Al trocável 3) A interpretação dos resultados do B com base nos valores obtidos pelo método curcumina-COL pode ocasionar erros devido a este não ser hábil em discriminar teores entre a estreita faixa de deficiência e suficiência para este elemento; 4) O uso da solução de Mehlich-3 em conjunto com a técnica de ICP OES teve correlação significativa para a extração de P, K, Ca, Mg, Cu e Zn em amostras de solo, podendo ser utilizado em laboratórios de análise de solo, embora não se tenha ganho prático em utilizá-la devido não ser possível determinar Al e Mn; 5) A solução de Mehlich-3 extraiu teores de Al, S, Mn e de B que não se correlacionaram significativamente com os teores extraídos pelas soluções extratoras de referência, não sendo, nas condições testadas, recomendado seu uso em conjunto com a técnica de ICP OES para a finalidade de diagnóstico da disponibilidade destes elementos; 6) A eficácia do MSF para determinar o valor verdadeiro de fósforo em matriz contendo Cu foi dependente da concentração de Cu e da relação fósforo:cobre; 7) A linha espectral do P em 213,618 nm foi mais sensível à presença do Cu em solução sendo preferível utilizar os resultados da linha espectral em 214,914 nm para determinação do P quando não utilizado o ajuste MSF. Advances in instrumental analytical chemistry have allowed a substantial improvement in obtaining important information in different areas of knowledge, such as Soil Science. This work addressed questions related to soil chemical analysis by inductively coupled plasma optical emission spectrometry (ICP OES). Studies involving correlation of results, simultaneous multielementar determination and interferences of the ICP OES technique were addressed in four chapters: 1) bibliographic review on ICP OES addressing operation, interferences and uses with different extractors; 2) potassium, calcium, magnesium, aluminum, sulfur, copper, zinc, manganese and boron determined by ICP OES in soil samples from southern Brazil; 3) multielement extraction by the Mehlich-3 solution and simultaneous determination by ICP OES in soil samples; 4) efficiency of the multicomponent spectral fitting (MSF) in the estimation of the correct value of phosphorus determined by ICP OES. The main conclusions about these studies were: 1) Significant linear relationships were found for K, Ca, Mg, Al, S, Cu, Zn, Mn and B determined by ICP and reference techniques; 2) The ICP technique presented higher sensitivity in relation to titration to determine Al in the extracts (3) The interpretation of the results of B on the basis of the values obtained by the curcumin-COL method may lead to errors because it is not able to discriminate between the narrow range of deficiency and sufficiency for this element; 4) The use of the Mehlich-3 solution in conjunction with the ICP OES technique had significant correlation for the extraction of P, K, Ca, Mg, Cu and Zn in soil samples, and could be used in soil analysis laboratories, although there is no practical gain in using it because it is not possible to determine Al and Mn; 5) The Mehlich-3 solution extracted contents of Al, S, Mn and B that did not significantly correlate with the contents extracted by the reference extractor solutions, and it was not recommended, under the conditions tested, its use in conjunction with the ICP OES for the purpose of diagnosing the availability of these elements; 6) The efficacy of the MSF to determine the true value of phosphorus in the Cu-containing matrix was dependent on the concentration of Cu and the phosphorus:copper ratio; 7) The spectral line of P at 213.618 nm was more sensitive to the presence of Cu in solution and it is preferable to use the spectral line of 214.914 nm for determination of P when the MSF setting is not used.

A extração de fósforo do solo pode ser comprometida pelas condições em que a análise é realizada e isso pode explicar a variabilidade nos resultados encontrados. O objetivo deste trabalho foi verificar a influência de desvios na velocidade de agitação, no tempo de contato e na temperatura sobre a extração do P pelos extratores Mehlich-1 (M-1) e Mehlich-3 (M-3) em amostras de solo. O estudo foi conduzido por meio de três experimentos executados com 11 amostras de solo provenientes da camada arável de Latossolo, Neossolo, Argissolo, Cambissolo, Planossolo, que são representativas das regiões geomorfológicas do Planalto e da Depressão Central do Estado do Rio Grande do Sul. Para todos os experimentos, adotou-se o delineamento experimental inteiramente casualizado em esquema fatorial 2 × 5 × 11, sendo dois extratores, cinco velocidades, temperaturas ou tempos de contato solo/solução e 11 tipos de solo, com quatro repetições. No primeiro experimento, a mistura solo:solução foi agitada em 80, 100, 120, 140 e 160 oscilações por min (opm). No segundo, após a agitação por 5 min, a mistura solo:solução foi deixada em repouso por períodos de tempo de oito, 12, 16, 20 e 24 h para retirada dos extratos. No terceiro, a temperatura de execução de todo o protocolo de análises foi mantida constante em 15, 20, 25, 30 e 35 °C. O aumento na velocidade de agitação de 120 até 160 opm aumentou a quantidade de P extraído pelos extratores. O aumento no tempo de contato antes da extração da alíquota elevou as quantidades de P extraído pelo M-1 e diminuiu para o M-3. Variações na temperatura não influenciaram significativamente o extrator M-1, mas o M-3 foi mais sensível a variações. Uma padronização na velocidade de agitação, no tempo de repouso e uma climatização dos laboratórios tornam-se necessárias para diminuir a interferência causada sobre as quantidades de P extraído pelos métodos M-1 e M-3 em amostras de solo. To produce reliable analytical data in a laboratory, all the stages of the analysis protocol must be properly understood and controlled. Extraction of soil P may thus be compromised by the conditions of analysis, and this may explain variability in results. Therefore, the objective of this study was to verify the effect of deviations in agitation speed, time of soil/ solution contact, and environmental temperature on extraction of soil P by Mehlich-1 (M-1) and Mehlich-3 (M-3). The study was conducted through three experiments performed simultaneously with 11 soil samples from the tilled layer of the following soils: Latossolo, Neossolo, Argissolo, Cambissolo, and Planossolo (Oxisol, Entisol, Ultisol, Inceptisol, and Alfisol) which are representative of the geomorphological regions of the Plateau and the Central Depression of the State of Rio Grande do Sul, Brazil. For all experiments, a completely randomized experimental design was adopted in a 2 × 5 × 11 factorial arrangement, consisting of two extractors, five speeds, temperatures, or times of soil/solution contact, and 11 soil types, with four replications. In the first experiment, the soil/solution mixture was shaken at 80, 100, 120, 140, and 160 oscillations per minute (opm). In the second, after shaking for 5 min., the soil/solution mixture was left at rest for periods of eight, 12, 16, 20 and 24 h before extract removal. In the third, the temperature for performing the entire analysis protocol was kept constant at 15, 20, 25, 30, and 35 °C. The increase in soil/solution contact time increased the amounts of P extracted by M1, but the amount extracted by M3 decreased. Variations in temperature did not significantly affect the M1 extractor; however, M3 was more sensitive to variations. Agitation speed greater than 120 opm increases the amounts of P extracted by the M1 and M3 extractors. There was no consistence between resting periods and the amounts of P extracted by the M1 and M3 extractors. Standardizing the agitation speed and the resting periods would minimize differences in the amount of P extracted by M1 and M3 from soil samples.

Em muitos laboratórios de análises de solos, o ácido 1-Amino-2-naftol-4-sulfônico (ANS) é usado como agente redutor para determinação do P por espectrometria de UV/Visível. Porém, devido aos riscos à saúde dos laboratoristas, uma alternativa é o uso do ácido ascórbico (AA). Entretanto, modificações no protocolo de análise podem induzir a variações nos resultados, havendo a necessidade de padronizá-las e calibrá-las. O objetivo do trabalho foi avaliar o ANS e AA nos extratos de Mehlich-1 e 3 nos intervalos de 15, 30, 45, 60 e 120min de reação. Não houve diferença na concentração de P determinado com ambos redutores quando comparados dentro de cada extrator. O ácido ascórbico pode substituir a solução ANS para determinação de P e o intervalo de tempo de reação sugerido para a quantificação colorimétrica é de 30 a 60min, para ambos redutores. In soil laboratory testing, the acid 1-amino-2-naphthol-4-sulfonic (ANS) is used as reducing agent to determine P by UV/Visible spectrometry. However, there are risks to laboratory technician healthy and an alternative is the use of ascorbic acid (AA). But, changes in the analytical protocol may induce variations in results, been necessary to standardize and calibrate. The objective of this studsy was to evaluate the ANS and AA in extracts of Mehlich-1 and 3 in ranges 15, 30, 45, 60 and 120min of reaction. There was no difference in P concentration determined by both reducers when compared within each extractor. Ascorbic acid can substitute the ANS solution for P determination and the suggested reaction interval for colorimetric quantification is between 30 and 60min for both substances.

Protocol methods have been adjusted to facilitate the execution, however here may be variability in analytical results by different laboratories even if they follow the same general principles of the method. This study aimed to identify sources of variability that affect analytical precision and accuracy of phosphorus extracted by Mehlich-1 and Mehlich-3 under variations in protocol procedures.
