Tetratrichomonas gallinarum and Trichomonas gallinae are pathogenic avian parasites that infect a wide range of bird species. The pathologic potential of T. gallinarum is controversial, whereas T. gallinae causes disease in many avian species. Infections are often asymptomatic in doves and pigeons; thus, columbids are presumed to represent the natural hosts for trichomonads. The detection of T. gallinarum and T. gallinae is based on direct microscopic observation or a conventional PCR assay. Microscopy is not very sensitive, and identification of the trichomonads at the genus or species level is not possible. Conventional PCR assays have been developed primarily for phylogenetic studies, which detect a wide range of Trichomonas spp. but do not allow their differentiation. We developed a duplex real-time PCR (rtPCR) assay for the simultaneous detection and differentiation of T. gallinarum and T. gallinae. We found that the rtPCR assay detected 10(2) plasmid DNA copies of T. gallinarum and as few as 10(1) plasmid DNA copies of T. gallinae.

Husbandry conditions for rabbits in Switzerland are at a high animal welfare level in international comparison. Wire mesh flooring is prohibited and group housing is mandatory up to the age of eight weeks. Despite these animal-friendly husbandry standards, animal losses seem to be a significant problem, although reliable data on professional Swiss meat rabbit production is lacking to date. A herd survey was carried out in 52 rabbit farms to address this situation. At the same time, each new litter and each fattening group was documented with a standardized form focusing on animal death and the use of medication by the farmer over a one year period. In the case of increased mortality or new health problems, dead rabbits were sent to the National Reference Center for Poultry and Rabbit Diseases (Zurich) for pathological examination including bacteriological and parasitological diagnostics. Data from 32 farms were evaluated. The average mortality of young animals (birth to weaning) was 14.9% (1.0-30.0%) and of fattening rabbits (weaning to slaughter) 17.3% (4.3-31.8%). Intestinal diseases, mainly dysentery, intestinal coccidiosis and mucoid enteropathies, were the primary cause of death (68.2%). Eimeria spp., Escherichia coli and Clostridium perfringens were identified as the most frequent pathogens. Respiratory diseases were found in 18.7% of the examined rabbits, with Pasteurella multocida identified as the most common pathogen. 60.0% of the farms used antibiotics during the study period and a third (34.7%) of all the fattening animals were medicated with at least one antibiotic. The present results confirm that despite high animal welfare standards infectious diseases and mortality rates represent a considerable problem in professional Swiss meat rabbit production. Risk factors of increased morbidity and mortality should be identified to improve rabbit health, to reduce the high mortality rate and the extensive use of antibiotics in professional Swiss meat rabbit production. Practicable management improvements with specific prevention strategies should be developed.Die Haltungsbedingungen für Kaninchen in der Schweiz liegen im internationalen Vergleich auf einem hohen Niveau. Die Haltung auf Drahtgitterboden ist verboten und Gruppenhaltung bis zum Alter von acht Wochen vorgeschrieben. Trotz dieser tierfreundlichen Haltungsstandards scheinen Tierverluste ein erhebliches Problem darzustellen, wobei zuverlässige Daten zur professionellen Schweizer Fleischkaninchenproduktion bis anhin grösstenteils fehlten. Um diesen Missstand zu beheben wurde in 52 Kaninchenbetrieben eine Bestandesuntersuchung durchgeführt, parallel wurde während eines Jahres jeder neue Wurf und jede Mastgruppe durch die Betriebsleiter mit einem standardisierten Formular dokumentiert, der Hauptfokus lag dabei auf Tierverlusten sowie Medikamenteneinsatz. Im Falle von erhöhter Mortalität oder neu aufgetretenen gesundheitlichen Problemen wurden verendete Kaninchen für Sektionen inklusive bakteriologischen und parasitologischen Untersuchungen ans Nationale Referenzzentrum für Geflügel- und Kaninchenkrankheiten (Zürich) eingesandt. Die Daten von 32 Betrieben konnten ausgewertet werden. Die durchschnittliche Mortalität der Jungtiere (Geburt bis Absetzen) lag bei 14.9% (1.0–30.0%), diejenige der Mastkaninchen (Absetzen bis Schlachten) bei 17.3% (4.3–31.8%). Mit 68.2% wurden an erster Stelle intestinale Erkrankungen festgestellt, dabei handelte es sich hauptsächlich um Dysenterien, gefolgt von Darmkokzidiosen und mukoiden Enteropathien. Als häufigste Erreger wurden in absteigender Reihenfolge Eimeria spp., Escherichia coli und Clostridium perfringens nachgewiesen. Bei 18.7% der untersuchten Kaninchen wurden Atemwegserkrankungen festgestellt, häufigstes Pathogen hierbei war Pasteurella multocida. 60.0% der untersuchten Betriebe setzten im Untersuchungszeitraum Antibiotika ein, rund ein Drittel (34.7%) aller untersuchten Mastdurchgänge wurde mit mindestens einem Antibiotikum mediziniert. Die Ergebnisse bestätigen, dass Infektionskrankheiten und Abgangszahlen in der Schweiz trotz tierfreundlicher Haltung ein erhebliches Problem in der professionellen Kaninchenhaltung darstellen. Um die Kaninchengesundheit in professionellen Schweizer Produktionsbetrieben zu verbessern und damit die hohen Mortalitätsraten und den umfangreichen Antibiotikaeinsatz zu reduzieren, sollten in einem weiteren Schritt gezielt Risikofaktoren für erhöhte Morbidität und Mortalität ermittelt werden, um praktikable Managementverbesserungen im Rahmen spezifischer Präventionsstrategien zu entwickeln.Les conditions d’élevage des lapins en Suisse sont d’un niveau élevé en comparaison internationale. Il est interdit de les garder sur un sol en treillis métallique et leur détention en groupe est obligatoire jusqu’à l’âge de huit semaines. Malgré ces normes d’élevage respectueuses des animaux, les pertes d’animaux semblent être un problème important, les données fiables sur la production professionnelle suisse de lapins de boucherie faisant largement défaut à ce jour. Afin de remédier à cette situation, une enquête d’exploitation a été réalisée dans 52 élevages cunicoles. En parallèle, chaque nouvelle portée et chaque groupe d’engraissement était documenté par le chef d’élevage avec un formulaire standardisé au cours d’une année, l’accent étant principalement mis sur les pertes d’animaux et l’utilisation de médicaments. En cas d’augmentation de la mortalité ou de nouveaux problèmes de santé, les lapins morts étaient envoyés au Centre national de référence pour les maladies de la volaille et des lapins (Zurich) pour une autopsie comprenant des examens bactériologiques et parasitologiques. Les données de 32 entreprises ont pu être évaluées. La mortalité moyenne des jeunes animaux (de la naissance au sevrage) était de 14,9% (1,0–30,0%), celle des lapins à l’engrais (du sevrage à l’abattage) était de 17,3% (4,3– 31,8%). Des maladies respiratoires ont été trouvées chez 18,7% des lapins examinés, le pathogène le plus courant étant Pasteurella multocida. 60,0% des élevages examinés ont utilisé des antibiotiques au cours de la période d’étude, environ un tiers (34,7%) de tous les processus d’engraissement examinés ont été traités avec au moins un antibiotique. Les résultats confirment que les maladies infectieuses et les pertes de lapins en Suisse représentent un problème considérable dans l’élevage professionnel malgré un élevage respectueux des animaux. Afin d’améliorer la santé des lapins dans les entreprises de production professionnelles suisses et de réduire ainsi les taux de mortalité élevés et l’utilisation intensive d’antibiotiques, les facteurs de risque d’augmentation de la morbidité et de la mortalité doivent être déterminés dans une étape supplémentaire afin de développer des améliorations de gestion réalisables dans le cadre de stratégies de prévention spécifiques.Le condizioni di stabulazione dei conigli in Svizzera sono da ritenersi di alta qualità nel confronto a livello internazionale. La tenuta su pavimenti in rete metallica è proibita e la tenuta in gruppi, fino all’età di otto settimane, è obbligatoria. Anche con questi standard di detenzione rispettosi degli animali, le perdite di conigli appaiono come un problema considerevole e non vi sono dati affidabili sulla produzione professionale svizzera di conigli da carne. Per risolvere questa mancanza è stata condotta un’indagine sulla popolazione in 52 aziende di allevamento di conigli, e in parallelo, durante un anno, sono state documentate, con un formulario standard dal responsabile dell’allevamento, ogni nuova nidiata e ogni nuovo gruppo da ingrasso, dando particolare attenzione ai decessi e all’uso di medicamenti. Nei casi di un incremento della mortalità oppure di nuovi problemi di salute, i conigli deceduti venivano inviati al centro nazionale di riferimento per le malattie dei volatili e dei conigli di Zurigo per la dissezione e gli esami batteriologici e parassitologici. I dati di 32 aziende sono stati valutati. La mortalità media degli animali giovani (dalla nascita allo svezzamento) si situava al 14.9% (1.0–30.0%), quello dei conigli da ingrasso (dallo svezzamento alla macellazione) era del 17.3% (4.3– 31.8%). Con il 68.2% le malattie intestinali, come la dissenteria, erano in prima linea, seguivano la coccidiosi intestinale e le enteropatie mucoidi. Gli agenti patogeni più ricorrenti erano in ordine decrescente Eimeria spp., Escherichia coli e Clostridium perfringens. Nel 18.7% dei conigli esaminati sono state rilevate delle malattie respiratorie dovute in particolare al patogeno Pasteurella multocida. Durante il periodo dello studio, il 60.0% delle aziende di allevamento esaminate hanno usato antibiotici, circa un terzo (34.7%) di tutti gli animali da ingrasso esaminati aveva ricevuto almeno un antibiotico. I risultati confermano che anche se gli allevamenti rispettano il benessere degli animali, le malattie infettive e i tassi di mortalità sono un problema da non sottovalutare nell’allevamento professionale dei conigli in Svizzera. Alfine di migliorare la salute dei conigli nelle aziende di allevamento professionali in Svizzera, e quindi diminuire il tasso di mortalità elevato e l’uso intensivo di antibiotici, bisognerebbe compiere un passo in più, mirato a identificare i fattori di rischio per l’elevata morbilità e mortalità in modo da sviluppare dei miglioramenti realistici nella gestione nell’ambito di una strategia preventiva specifica.

Improve understanding of the use of alternatives to robenidine in the form of natural supplements., Gastrointestinal disorders due to Eimeria sp. and E. coli overgrowth cause high mortality in weaner rabbits and the interest in alternatives to coccidiostats is high. This study aimed to investigate the superiority of natural feed additives towards robenidine preserving gastrointestinal health in the field. Rabbits were divided into four groups, Control Group (CG) exclusively supplemented with robenidine, Sainfoin Group (SG) was supplemented with a combination of robenidine and sainfoin, and two additional groups were respectively supplemented with Herb-All COCC-X (garlic; conessi tree) (HG: Herbal Group) and by a combination of Herb-All COCC-X and Klinofeed (clinoptilolite) (MG: Mineral Group). Eimeria sp. (98,40%) and E. coli overgrowth (73.60%) could be confirmed as the main causes for losses. High mortality rates (SG: 30.00% - MG: 47.50%), also in the groups receiving robenidine (SG: 30.00%; CG: 45.00%), reinforced the importance of alternatives in the field. The natural additives of groups SG, HG and MG did not have a significant influence on the weight gains and the oocyst counts in the jejunum/ileum and caecum of slaughter rabbits at the end of the trial, compared to group CG. Significantly higher oocyst shedding in SG (p = 1.4E-03) and HG (p = 1.4E-05) during the trial may be explained by a higher surviving rate of diseased rabbits in those groups, fostered by beneficial effects of the additives, which should be investigated further.

Gallid alphaherpesvirus 1 (syn. infectious laryngotracheitis virus; ILTV) is the causative agent of infectious laryngotracheitis, a respiratory disease of chickens causing substantial economic losses in the poultry industry every year. Currently, the most efficient way to achieve protection against infection is immunization with live-attenuated vaccines. However, this vaccination strategy entails the risk of generating new pathogenic viruses resulting from spontaneous mutations or from recombination with field strains. This work presents a new approach based on virus-like particles (VLPs) displaying ILTV glycoproteins B (gB) or G (gG) on their surface. The main focus of this pilot study was to determine the tolerability of VLPs delivered in ovo and intramuscularly (i.m.) into chickens and to investigate the nature of the immune response elicited. The study revealed that the new vaccines were well tolerated in hybrid layer chicks independent of the administration method (in ovo or i.m.). Upon in ovo injection, vaccination with VLP-gG led to an antibody response, while a cellular immune response in VLP-gB-immunized chickens was hardly detectable. Since the administration of VLPs had no visible side effects in vivo and was shown to elicit an antibody-based immune response, we anticipate that VLPs will become a valuable platform for the development of new safe vaccines for poultry.

Background Salmonella are bacteria of the family Enterobacteriaceae with a wide host range. Infection in birds causes subclinical disease to mass mortality events. Wild birds may act as healthy carriers posing a hazard to livestock and humans. The present study investigated the occurrence of Salmonella in wild birds admitted to a rehabilitation centre in order to assess the exposure of the staff to this zoonotic pathogen. Methods Faecal swabs of 552 avian patients (68 species) were collected over the course of 12 months. Each sample was propagated in enrichment broth and subsequently incubated on a RAPID'Salmonella plate. Salmonella isolates were serotyped, and antimicrobial susceptibility testing was performed. Results Six Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium) and 1 S. Schleissheim were detected; all were pansusceptible to the antibiotics tested. Conclusion Despite the low positive rate in the tested population, the authors recommend applying protective equipment and hygiene measures when handling wild birds.

Poultry feed is a leading source of Salmonella infection in poultry. In Switzerland, heat-treated feed is used to reduce Salmonella incursions into flocks in conventional poultry production. By contrast, organic feed is only treated with organic acids. In 2019, the Swiss National Reference Center for Enteropathogenic Bacteria identified the rare serovar S. Jerusalem from samples of organic soya feed. Further, in July 2020, the European Union’s Rapid Alert System for Food and Feed published a notification of the detection of S. Jerusalem in soya expeller from Italy. During 2020, seven S. Jerusalem isolates from seven different poultry productions distributed over six cantons in Switzerland were reported, providing further evidence of a possible outbreak. Using whole-genome sequencing (WGS), S. Jerusalem isolates from feed and from animals in Switzerland were further characterized and compared to S. Jerusalem from organic poultry farm environments in Italy. WGS results showed that feed isolates and isolates from Swiss and Italian poultry flocks belonged to the sequence type (ST)1028, grouped in a very tight cluster, and were closely related. This outbreak highlights the risk of spreading Salmonella by feed and emphasizes the need for a heat-treatment process for feed, also in organic poultry production.

Erysipelothrix rhusiopathiae transmission to human is often occupation-related, but in most cases, a detailed case history is missing. This case report is based on an interdisciplinary approach and includes a thorough medical record. A 58-year-old laboratory technician working on geese necropsy cut open her glove at a rib fragment of a goose and subsequently noticed a slowly progressive, reddish skin alteration in the particular region of the hand. Bacteriological investigations on the geese revealed septicaemia due to E. rhusiopathiae and therefore substantiated the diagnosis of the patient. The infectious agent could not be cultured from the patient; however, antibiotic susceptibility testing was performed using the goose isolate. An entire follow-up until full recovery of the patient was conducted. Zoonotic infections possibly have a significant impact on certain occupations. This case report analyses a rare but important zoonotic infection to create awareness of this in physicians caring for human patients.

Background An outbreak of salmonellosis due to Salmonella Typhimurium was detected coincidentally in a Swiss meat rabbitry, given that surveillance of Salmonella in rabbits is not mandatory in Switzerland. Methods To assess the extent of potentially subclinical Salmonella carriage in meat rabbits, faecal pool samples of 50 farms (90% of Swiss commercial rabbitries) with ground covering litter and group housing were bacteriologically tested. Additionally, 236 rabbits showing clinical signs compatible with intestinal diseases, such as salmonellosis, were examined postmortem and analysed bacteriologically. Salmonella isolates were serotyped and analysed by whole genome sequencing (WGS). Results Salmonella Typhimurium was detected in three commercial farms (6.0% of all tested farms). The affected farms were directly linked to the animal trade and Salmonella isolates were shown to be identical by WGS. Conclusion There is no increased hazard for Salmonella carriage in the animal welfare‐friendly Swiss husbandry systems in general, despite risk factors such as ground covering litter.

Chlamydiaceae are obligate intracellular bacteria with a broad host range. Several studies have found chlamydial species that are genetically intermediate between Chlamydia psittaci and Chlamydia abortus in various avian species. One of these intermediate Chlamydia species, found in a red-shouldered hawk (Buteo lineatus), was recently classified as a new species Chlamydia buteonis. This newly described Chlamydia species has, so far, only been reported in hawks exhibiting clinical signs of conjunctivitis, dyspnea, and diarrhea. In the present study, fecal samples of 5 gyrfalcons (Falco rusticolus), 3 gyr/peregrine falcon hybrids (Falco rusticolus × Falco peregrinus), and 15 falcons of unknown species presented to falcon clinics on the Arabian Peninsula were shipped to the Vetsuisse Faculty, University of Zurich (Zurich, Switzerland), for examination for the presence of Chlamydiaceae. A step-wise diagnostic approach was performed to identify the chlamydial species involved. Chlamydiaceae were detected in 21/23 falcons by a family-specific real-time quantitative PCR (qPCR). Further identification with a 23S ribosomal RNA-based microarray assay and 16S conventional PCR and sequencing yielded inconclusive results, indicating the presence of an intermediate Chlamydia species. Because none of the falcons tested positive for Chlamydia psittaci by specific qPCR, all 23 samples were subjected to a Chlamydia buteonis–specific qPCR, which was positive in 16/23 samples. Detailed information regarding clinical history was available for 8 falcons admitted to a falcon clinic in Dubai, United Arab Emirates. Six of those birds that were presented to the clinic because of loss of performance and poor general condition, including vomiting and diarrhea, were positive for C buteonis. In 2 birds without clinical disease signs admitted for a routine health examination, 1 was positive for C buteonis, and 1 was negative. It is yet unknown whether Chlamydia buteonis causes disease in birds, but the findings in this study indicate that Chlamydia buteonis may be an infectious pathogen in falcon species.

Bacteria of the family Chlamydiaceae are globally disseminated and able to infect many bird species. So far, 11 species of Chlamydia have been detected in wild birds, and several studies found chlamydial strains classified as genetically intermediate between Chlamydia (C.) psittaci and C. abortus. Recently, a group of these intermediate strains was shown to form a separate species, i.e., C. buteonis. In the present study, 1128 samples from 341 raptors of 16 bird species and 253 corvids representing six species were examined using a stepwise diagnostic approach. Chlamydiaceae DNA was detected in 23.7% of the corvids and 5.9% of the raptors. In corvids, the most frequently detected Chlamydia species was C. psittaci of outer membrane protein A (ompA) genotype 1V, which is known to have a host preference for corvids. The most frequently detected ompA genotype in raptors was M56. Furthermore, one of the raptors harbored C. psittaci 1V, and two others carried genotype A. C. buteonis was not detected in the bird population investigated, so it remains unknown whether this species occurs in Switzerland. The infection rate of Chlamydiaceae in corvids was high compared to rates reported in other wild bird species, but neither Chlamydiaceae-positive corvids nor raptors showed overt signs of disease. Since the Chlamydiaceae of both, raptors and crows were identified as C. psittaci and all C. psittaci genotypes are considered to be zoonotic, it can be suggested that raptors and crows pose a potential hazard to the health of their handlers.

Several pigeon paramyxovirus-1 (PPMV-1) outbreaks in feral pigeons were described recently in Switzerland. The potential of PPMV-1 to induce the notifiable Newcastle disease in chickens is discussed controversially. Therefore, in order to study epidemiologically relevant parameters such as the kinetics of PPMV-1 replication and shedding as well as seroconversion after infection, chickens were infected experimentally with a Swiss PPMV-1 isolate. This generated also defined sample material for the comparison of diagnostic tests. The infectivity of the Swiss PPMV-1 isolate for chickens was demonstrated successfully by virus shedding after experimental inoculation. Our data suggest that long-lasting shedding for up to 60 days can occur in chickens infected with PPMV-1. The isolate used here was of low pathogenicity for chickens. Different quantitative reverse transcription PCR assays were evaluated with a set of Swiss PPMV-1 isolates, and various samples from experimentally infected chickens were analysed with respect to their suitability for viral RNA detection. At 14 days post-infection, virus genome was detected mainly in spleen, caecal tonsils, heart, cloacal swabs, liver, proventriculus, duodenum and kidney tissue samples. Overall, the level of virus replication was low. Not all assays used routinely in diagnostics were capable of detecting viral genome from the isolates tested. Possible explanations are the genetic divergence of PPMV-1 and the low level of viral RNA in the samples. In contrast, two methods that are not used routinely proved more suitable for virus-genome detection. Importantly, the collection of material from various different organs is recommended, in addition to the kidney and brain analysed routinely. In conclusion, this study shows that there is a need to reconsider the type of samples and the protocols used for the detection of PPMV-1 RNA in chickens.

A pet rabbit was found dead outside its enclosure; the head was missing. The investigation of cruelty to animals was commissioned, as decapitation might result from human mutilation, for example, due to occult rituals or neighbourhood disputes. A first external examination showed circular disruption rostral of the pectoral girdle with lack of the head and palpable bone fragments of both forelimbs. CT scan revealed extensive, irregular trauma to soft tissues and skeleton. The decapitation left a cut-like but irregular, partially wave-like wound margin with multiple small triangular, divergent defects more closely examined by postmortem examination. Swabs from the margin of the wound and tissue samples were collected for forensic genetic analysis and histology. Considering these interdisciplinary investigation results combined with experts’ knowledge, a predator attack was highly likely. Further evidence on geographical distribution and physiological behaviour of putative predator species led to consensus on the identity of the most likely perpetrator: a red fox.

In Switzerland, domestic turkey meat is a niche product. Turkeys are fattened on mixed family-based farms scattered across the country, with most providing access to an uncovered outdoor pasture for the birds. Swiss fattening turkeys may therefore get infected with Chlamydiaceae via wild birds or their faeces, potentially shedding these bacteria at a later stage. The aim of the present study was to acquire baseline data about the shedding of Chlamydiaceae in clinically unremarkable Swiss fattening turkeys at slaughter, potentially exposing slaughterhouse workers to infection. In this large-scale study, 1008 cloacal swabs of Swiss turkeys out of 53 flocks from 28 different grow-out farms with uncovered outdoor pasture were collected over the course of 14 months and examined for the occurrence of Chlamydiaceae by a family-specific 23S-rRNA real-time PCR. Positive samples were further analyzed by Chlamydia psittaci (C. psittaci)-specific real-time PCR and the Arraymate DNA Microarray for species identification. All samples were negative for C. psittaci, but seven swabs out of one flock were tested positive for Chlamydia gallinacea (0.7%). Although turkeys with access to pasture may have contact with Chlamydiaceae-harbouring wild birds or their faeces, the infection rate in Swiss turkeys was shown to be low.

A total of 294 fecal swabs from 294 wild birds in Switzerland were cultivated for antimicrobial resistant (AMR) Escherichia coli. Samples were also subcultivated to detect E. coli with extended‐spectrum β‐lactamases (ESBL), carbapenemases, and plasmid‐mediated aminoglycoside or colistin resistance, respectively. Samples from 17 (5.8%) of the birds contained 19 AMR E. coli, whereof 26.3% were multidrug resistant. Five (1.7%) ESBL‐producing E. coli were detected. The isolates harbored bla CTX‐M‐1 (two isolated from carrion crows and from one great spotted woodpecker, respectively), bla CTX‐M‐15 (originating from a grey heron), bla CTX‐M‐55 (from a carrion crow), and bla CTX‐M‐65 (from a common blackbird). Phylogenetic analysis assigned three isolates to commensal phylogroups A and B1, one to extraintestinal pathogenic group B2, and one to phylogroup F. Multilocus sequence typing identified sequence types (STs) that have been found previously in ESBL E. coli in wild birds (ST58, ST205, ST540). One isolate harboring bla CTX‐M‐55 was assigned to the recently emerged fluoroquinolone‐resistant, extraintestinal pathogenic E. coli clone ST1193. Wild birds have the potential to disperse AMR, including clinically important resistance genes, from anthropogenic‐influenced habitats to diverse areas, including vulnerable natural environments such as surface waters or mountain regions., Of 294 fecal samples from resident and short‐distance migratory wild birds, 5.8% contained antimicrobial resistant (AMR) Escherichia coli. Moreover, 1.7% of the samples yielded E. coli producing the extended‐spectrum β‐lactamases CTX‐M‐1, CTX‐M‐15, CTX‐M‐55, or CTX‐M‐65. Wild birds have the potential to disperse AMR bacteria from anthropogenic‐influenced habitats to diverse areas, including vulnerable natural environments such as surface waters or mountain regions.

West Nile virus (WNV) is an important zoonotic pathogen maintained in a natural transmission cycle between mosquitoes and birds as reservoir hosts. In dead-end hosts, such as humans, infection may result in fatal neurologic disease translating into disease and death-related suffering and increased health care costs. In humans, WNV may also be transmitted through blood transfusions and organ transplants. WNV is not present in Switzerland yet, but competent vector species (especially Culex pipiens and Aedes japonicus) are prevalent and an introduction of the virus, likely through wild birds, is expected at any time. Therefore, it is important for Switzerland to be prepared and establish a surveillance system for WNV to initiate increased prevention activities, such as the screening of blood and organ donations and public education activities in case virus circulation is detected. The long-term goal of these surveillance measures would be a reduced infection rate in humans resulting in less suffering and reduced health care costs. To provide the basis for a pragmatic and resource-effective WNV surveillance program, this study used aliquots of serum samples of free-range laying hens taken at the abattoir and collected in the frame of the ongoing Swiss Avian Influenza and Newcastle Disease monitoring program for a 2-year period. All 961 aliquots were analyzed using a commercial competitive WNV enzyme-linked immunosorbent assay (ELISA). The study allowed to set up sampling and laboratory routines as a basis for future WNV surveillance activities. At this stage there is no evidence for circulation of WNV in Switzerland.

The present study was conducted to evaluate the efficacy of two prophylactic antibiotic treatments against bovine respiratory disease (BRD) in veal calves. In addition, the antibiotic susceptibilities of isolated Pasteurellaceae were tested. The calves were treated either on the day of arrival by a single administration of tulathromycin (group A, n = 20), by a peroral administration of chlortetracycline, sulphadimidine, and tylosin (group B, n = 20) for seven consecutive days, or were not prophylactically treated (group C, n = 19). On the first day of clinically diagnosed BRD, transtracheal lavage samples were obtained prior to therapeutic treatment and were subsequently cultured. Pasteurellaceae isolates were tested for their susceptibility to 12 antimicrobial agents by the determination of the minimal inhibitory concentrations. During the first 56 d after arrival, different calves in group A and B suffered from one episode of clinically diagnosed BRD while calves of group C experienced two episodes. The average daily weight gain during the same period was significantly lower in group C (0.89 ± 0.04 kg/d) than in the two prophylactically treated groups (1.14 ± 0.05 and 1.15 ± 0.04 kg/d for group A and B, respectively). The improved performance of groups A and B in comparison to group C could be related to a lower incidence of respiratory disorders during the first days after arrival in the prophylactically treated animals. No differences in the clinical efficacy were seen between the two tested prophylactic treatments. The most prevalent bacterial pathogens isolated ( n = 79) were Pasteurella multocida (23% of isolated pathogens), Mycoplasma bovis (18%), and Mannheimia varigena (16%). For the isolated Pasteurellaceae , a high resistance pattern was observed to tylosin (83% of the tested P. multocida and 88% of the Mannheimia spp. isolates resistant) and tilmicosin (56% of the tested P. multocida isolates non-sensitive).

Three cases of fatal septicaemia due to Plesiomonas shigelloides and one due to Edwardsiella tarda were diagnosed in newborn penguins from the Basle Zoo, Switzerland from 2003 to 2007. The affected penguins were of two different species (king penguin, Aptenodytes patagonicus, and African penguin, Spheniscus demersus) and between 2 and 10 days old at the time of death. The causative agents, E. tarda and P. shigelloides are ubiquitous bacteria which are reported to be present in the normal intestinal flora of wild and captive aquatic animals, including penguins. Their occurrence and infectious potential is discussed.

The re-emergence of necrotizing enteritis (NE) in Swiss pig breeding farms raised concern that, besides C. perfringens type C strains, additional C. perfringens toxinotypes might cause this disease. Therefore we retrospectively investigated the association of NE with C. perfringens type C or different C. perfringens toxinotypes. We evaluated pathological lesions, routine diagnostic bacteriology results, and multiplex real-time PCR analyses from DNA extracts of archived intestinal samples of 199 piglets from our diagnostic case load. 96.5% of NE cases and 100% of herds affected by NE were positive for C. perfringens type C genotypes. Animals without necrotizing enteritis revealed a significantly lower detection rate of type C genotypes. Non affected piglets showed a high prevalence for beta-2-toxin positive C. perfringens type A strains. Collectively, our data indicate that outbreaks of NE in piglets in Switzerland cannot be attributed to newly emerging pathogenic toxinotypes, but are due to a spread of pathogenic C. perfringens type C strains.

Stenotrophomonas maltophilia (S. maltophilia) is a nonfermentative bacterium, which is naturally resistant against a panel of commonly-used antibiotics. It is frequently isolated from humans with chronic respiratory disease, e.g. cystic fibrosis or chronic obstructive pulmonary disease. In veterinary medicine S. maltophilia is perceived to be a mere coloniser. We herewith report 7 strains of S. maltophilia isolated from animals, of which 5 strains were harvested from 3 horses, a dog and a cat with chronic respiratory disease. The dog isolate showed resistance to trimethoprim / sulphamethoxazole, which was confirmed by detection of the sul 1 gene. Analysis with pulsed field gel electrophoresis revealed that 2 horses, which were boarded in the same clinic but two years apart, harboured the same strain of S. maltophilia. This is indicative of a hospital acquired colonisation / infection, which contradicts involvement in the pre-existing chronic disease.

Typing of Clostridium perfringens strains by PCR-based determination of toxin genes proved to be a reliable method for diagnosis of enterotoxaemia in various animal species. We report the establishment and validation of three real-time fluorogenic (TaqMan) multiplex PCRs for the detection of C. perfringens alpha-, beta-, beta2-, epsilon-, entero- and iota-toxin genes. The composition of the PCRs was chosen with regard to robustness of the assays and in order to increase sensitivity compared to the conventional simplex PCRs. The combination of probe dyes selected for the real-time assays (FAM/TAMRA, Cy-5/BHQ-2 and VIC/TAMRA) as well as the designation of the chromosome-borne alpha-toxin as internal positive control allowed the creation of highly specific and sensitive, as well as time and cost effective PCRs. One hundred and three strains of C. perfringens isolated in Switzerland derived from clinical or suspected cases of enterotoxaemia in 10 different animal species were tested. The toxin genotypes were in agreement in both the conventional PCRs and the newly designed multiplex PCRs. Furthermore, the real-time PCR carried out as simplex allows to quantitate the copy numbers of plasmid-borne toxin genes in relation to the chromosomally located alpha-toxin gene.

Fast growing mycobacteria are saprophytic bacteria that prevail in water and soil. They are opportunistic pathogens and may cause various infections if gaining entry into the body through a trauma. We herein describe the clinical presentation, pathology and diagnosis of the first case of cutaneous atypical mycobacteriosis due to Mycobacterium massiliense in a cat.

In this study, six immunocompetent calves were experimentally infected with a noncytopathic strain of bovine viral diarrhea virus (BVDV), and the effects of the viral infection on parameters of the innate immune response of the host were analyzed. Clinical and virological data were compared with the temporal activation of the alpha/beta interferon-regulated Mx gene in white blood cells (WBC) and skin as well as the upregulation of the acute-phase serum proteins haptoglobin (Hp) and serum amyloid A (SAA). The viral strain used did provoke transient health impairment, namely, fever and leukopenia that were associated with viremia, viral shedding with nasal secretions, and antiviral seroconversion. Complete recovery was observed within 3 weeks. Elevated levels of SAA and Hp were apparent from days 4 to 13 and 8 to 11, respectively. In WBC, the levels of Mx mRNA and Mx protein were elevated from days 2 to 15. In the context of this study with BVDV, the level of Mx protein expression in WBC provided the most telling diagnostic window to monitor the host's ongoing innate immune response.

For the first time Ovine Herpesvirus 2 (OvHV-2) was identified in Swiss pigs as the causative agent of Porcine Malignant Catarrhal Fever (MCF). Diseased animals from two farms were observed to show weakness, anorexia, fever up to 41 degrees C, and neurological symptoms, i.e. ataxia, convulsions and hyperesthesia, erosion on the snout and in the oral and nasal mucosa, as well as multiple skin lesions. Histopathological findings included severe non-purulent inflammation with mononuclear cell infiltration in several organs. Most dominant were meningo-encephalitis, disseminated nephritis as well as purulent catarrhalic bronchopneumonia. The findings were quite reminiscent of the lesions due to MCF in cattle and give therefore substantial proof to use Porcine Malignant Catarrhal Fever as the term for the disease. Identification of the causative agent was done with a quantitative PCR specific for OvHV-2. Different tissues from diseased animals were positive. Furthermore, one animal which had been ill for more than five days tested positive for antibodies against an epitope conserved among MCF viruses. Serum samples from diseased animals reacted negative towards Classical Swine Fever- and Pseudorabies virus antigen. A weakly positive reaction against porcine enterovirus type I argued against the involvement of enteroviruses in the observed disease. Moreover, by means of different conventional PCRs, we detected the newly discovered porcine lymphotropic herpesviruses for the first time in Switzerland and could at the same time exclude their involvement in Porcine Malignant Catarrhal Fever.

Cases of porcine malignant catarrhal fever were analyzed by a combination of identification and quantitation of ovine gammaherpesvirus 2 DNA in a variety of paraffin-embedded tissues from diseased pigs, serology, and exclusion of primary porcine gammaherpesviruses. In spite of reduced signal due to fixation and paraffin embedding, ovine gammaherpesvirus 2 DNA in pig brains exceeded the amounts found in sheep brains by orders of magnitude.

The present report describes two proven cases of illegal poisoning of peregrine falcons in the city of Zurich, Switzerland. Both animals died in their eyries after capturing and feeding on a methomyl-laced pigeon. Both eyries are surveillanced by cameras, and the death of one falcon was broadcasted live on the Internet. The city game warden had noticed the disappearance of a brooding female from an artificial eyrie in 2009 and had found five dead falcons at plucking posts or artificial eyries between 2009 and 2013. Owing to these reports in combination with reports on the disappearance of at least eight pairs of peregrine falcons from traditionally used nesting sites in north-western Switzerland since the 1980s, the authors presume that there may be a high number of unreported cases of illegal killing of peregrine falcons in Switzerland. They suggest an increased surveillance, and a coordinated collection of data on wildlife crime may aid the prosecution of perpetrators.

Ovine herpesvirus 2 (OvHV-2), a member of the viral subfamily Gammaherpesvirinae , shares numerous similarities with human herpesvirus 8 (HHV-8). Both viruses are apathogenic in their healthy original host, may cause lymphoprolipherative diseases, cannot routinely be propagated in cell culture, and may be sexually transmitted. However, the pathways of sexual transmission of these viruses, as well as the underlying pathogenetic dynamics, are not well understood. Organs from naturally OvHV-2-infected, as well as OvHV-2-free, sheep were quantitatively analyzed for OvHV-2 by the DNA amplification techniques. The dynamics of OvHV-2 multiplication and excretion were monitored after experimental infections and, most importantly, subsequent to vasectomy. The OvHV-2 DNA load in various tissues and internal organs was not merely reflecting the viral DNA load in the bloodstream, which suggested compartmentalization of OvHV-2. Moreover, OvHV-2 DNA was detected at several portals for virus shedding, i.e., the respiratory, alimentary, and urogenital tracts. Transient OvHV-2 excretion was detected in ejaculates of experimentally infected rams. Upon vasectomy, OvHV-2 DNA reappeared in the ejaculatory plasma, but the titers did not decline after reaching a peak. Spiking and fractionation experiments revealed an inhibitory activity, associated with the spermatozoa, which was able to suppress detection of viral DNA but which was no longer present in samples from vasectomized animals. Therefore, epidemiological studies on viruses that may be transmitted by the ejaculatory pathway and for whose tracing nucleic acid amplification methods are used, i.e., OvHV-2, HHV-8, and the human immunodeficiency virus, should include vasectomized males.

A fluorogenic PCR was established for the quantification of feline herpesvirus 1 (FeHV-1) DNA in ocular fluid samples of clinically diseased cats. The new assay was specific for FeHV-1 and sensitive. The 100% detection rate ranged from 0.6 to 6 50% tissue culture infective doses per sample. When spiked samples with known quantities of virus were used, infectious virus titers and quantification of viral DNA by PCR correlated to each other in a linear fashion ( R 2 = 0.9858) over a range of 4 orders of magnitude. Within this range, it was possible to calculate the FeHV-1 DNA content from a given infectious dose, and vice versa. The new diagnostic procedure was applied to ocular fluid samples from cats experimentally infected with FeHV-1 and specific FeHV-1-free cats. A good correlation between virus titer and quantitative PCR was observed, although only early in infection. In a second stage, the titer of infectious virus collapsed, while the PCR signal remained high. A constantly decreasing PCR signal accompanied by negative virus isolation was characteristic for a final stage of the infection. Finally, clinical samples from 20 cats that were suspected to suffer from FeHV-1 infection were analyzed. By comparing virus titers and quantitative PCR signals, it was possible to determine the current stage of the ongoing infection. Based on these findings, comparison of the results of consecutive samples allows the tracking of the course of the infection. Therefore, the new method combines the advantages of the two previously established conventional methods, qualitative PCR and virus isolation and titration.
