PURPOSE: The long-term effect of low and moderate doses of ionizing radiation on the lens is still a matter of debate and needs to be evaluated in more detail. MATERIAL AND METHODS: We conducted a detailed histological analysis of eyes from B6C3F1 mice cohorts after acute gamma irradiation (60Co source; 0.063 Gy/min) at young adult age of 10 weeks with doses of 0.063, 0.125 and 0.5 Gy. Sham irradiated (0 Gy) mice were used as controls. To test for genetic susceptibility heterozygous Ercc2 mutant mice were used and compared to wild type mice of the same strain background. Mice of both sexes were included in all cohorts. Eyes were collected 4 hours, 12, 18 and 24 months after irradiation. For a better understanding of the underlying mechanisms, metabolomics analyses were performed in lenses and plasma samples of the same mouse cohorts at 4 and 12 hours as well as 12, 18 and 24 months after irradiation. For this purpose, a targeted analysis was chosen. RESULTS: This analysis revealed histological changes particularly in the posterior part of the lens that rarely can be observed by using Scheimpflug imaging, as we reported previously. We detected a significant increase of posterior subcapsular cataracts 18 and 24 months after irradiation with 0.5 Gy (odds ratio 9.3; 95%-confidence interval 2.1 - 41.3) independent of sex and genotype. Doses below 0.5 Gy (i.e. 0.063 and 0.125 Gy) did not significantly increase the frequency of posterior subcapsular cataracts at any time point. In lenses, we observed a clear effect of sex and aging but not of irradiation or genotype. While metabolomics analyses of plasma from the same mice showed only a sex effect. CONCLUSIONS: This paper demonstrates a significant radiation-induced increase in the incidence of posterior subcapsular cataracts, which could not be identified using Scheimpflug imaging as the only diagnostic tool.

For many degenerative retinal diseases that progressively lead to blindness, no treatment options are available so far. In recent years, several innovative therapies have been experimentally explored, which are promising because they are independent of the genetic cause of the degenerative disease. One of these is optogenetics, which involves light-sensitive proteins that selectively act as ion channels or ion pumps to control the potential of the treated cell. Thus, these cells can be stimulated or inhibited by light, quasi functionally remote controlled. In this way artificial photoreceptors are induced from the remaining cells, which has already been successfully employed in animal experiments. This type of treatment is already being tested on patients and leads to an improvement in vision, but so far only data from one patient are available. The use of optogenetics additionally requires special eyeglasses to adapt the light impulses in adequate strength and wavelength for the respective optogenes. Another exciting approach is cell replacement therapy of retinal pigment epithelium (RPE) and photoreceptor cells to exchange degenerated cell material. This appears to be very successful for RPE cells in clinical trials. Obtaining human photoreceptors from stem cells is technically possible, but very laborious. The integration of the transplanted photoreceptors into the host retinal tissue also needs further optimization for broader clinical applications; however, both cell replacement and optogenetics approaches are promising, so that the translation from basic research into clinical application will be successful.Für viele degenerative Netzhauterkrankungen, die progressiv zur Erblindung führen, gibt es bisher keine Therapie. In den letzten Jahren wurden einige innovative Therapien experimentell erforscht, die vielversprechend sind, da diese unabhängig von der genetischen Ursache der degenerativen Erkrankung sind. Hierzu zählt zum einen die Optogenetik, die lichtsensitive Proteine umfasst, die selektiv als Ionenkanal oder Ionenpumpe das Potenzial der behandelten Zelle steuern. Somit können diese Zellen per Licht angeregt oder inhibiert werden, quasi funktional ferngesteuert. Somit werden aus noch vorhandenen Netzhautzellen künstliche Photorezeptoren induziert, was bereits im Tierversuch erfolgreich angewandt wurde. Diese Therapie wird bereits am Patienten erprobt und führt zu einer Sehverbesserung, jedoch liegen bisher nur Daten eines Patienten vor. Optogenetische Therapien benötigen zusätzlich eine spezielle Brille, um die Lichtimpulse in adäquater Stärke und Wellenlänge für die jeweiligen Optogene anzupassen. Ein anderer spannender Ansatz ist die Zellersatztherapie von RPE(retinales Pigmentepithel)- und Photorezeptorzellen, um degeneriertes Zellmaterial auszutauschen. Dieses sieht bei RPE-Zellen in klinischen Studien sehr erfolgreich aus. Die Gewinnung von menschlichen Photorezeptorzellen aus Stammzellen ist technisch möglich, allerdings sehr aufwendig. Die Integration der transplantierten Photorezeptoren in das vorhandene Netzhautgewebe bedarf einer weiteren Optimierung zwecks breiter klinischer Anwendung. Aber beide Ansätze, Zellersatz und Optogenetik, sind vielversprechend, sodass die Translation von der Grundlagenforschung in die klinische Anwendung gelingen wird.

Because of the increasing application of ionizing radiation in medicine, quantitative data on effects of low-dose radiation are needed to optimize radiation protection, particularly with respect to cataract development. Using mice as mammalian animal model, we applied a single dose of 0, 0.063, 0.125 and 0.5 Gy at 10 weeks of age, determined lens opacities for up to 2 years and compared it with overall survival, cytogenetic alterations and cancer development. The highest dose was significantly associated with increased body weight and reduced survival rate. Chromosomal aberrations in bone marrow cells showed a dose-dependent increase 12 months after irradiation. Pathological screening indicated a dose-dependent risk for several types of tumors. Scheimpflug imaging of the lens revealed a significant dose-dependent effect of 1% of lens opacity. Comparison of different biological end points demonstrated long-term effects of low-dose irradiation for several biological end points. Die Lese-Rechtschreibstörung (LRS) ist eine der häufigsten umschriebenen Entwicklungsstörungen schulischer Fertigkeiten und bleibt meist bis ins Erwachsenenalter bestehen. Schulische Schwierigkeiten und psychische Störungen sind oft Begleitsymptome der LRS. Aus diesem Grund ist nachhaltige Förderung von größter Bedeutung. Die Fördereffekte bisheriger Interventionsstudien sind jedoch recht gering, und vielen Studien mangelt es an ausreichender methodischer Fundierung. Ziel dieser Studie ist daher die Überprüfung zweier Interventionsprogramme in einem Prä-Post Experimental-Warte-Kontrollgruppendesign. Vier Gruppen von Drittklässlern nahmen an der Studie teil. Zwei Gruppen von Kindern mit LRS (n = 40) wurden über sechs Monate zweimal wöchentlich mit einem Lese- oder Rechtschreibprogramm gefördert und mit einer Wartegruppe (n = 17), die erst nach sechs Monaten gefördert wurde, sowie einer nicht betroffenen Kontrollgruppe (n = 26) verglichen. Die quantitative und qualitative Analyse der Lese- und Rechtschreibleistungen vor und nach der Förderung zeigte, dass sich alle Kinder mit LRS signifikant verbesserten. Betrachtungen auf individueller Ebene zeigten jedoch auch, dass eine deutlich spezifischere Zuweisung zu einzelnen Förderprogrammen notwendig ist. Eine Überlegenheit einer einzelnen Gruppe fand sich nicht. Da sich auch die Wartegruppe der unbehandelten Kinder mit LRS verbesserten, können keine eindeutigen Schlüsse über die Effektivität der Förderung gezogen werden. Die Gründe hierfür sind unklar, müssen aber unbedingt verstanden werden. Hierzu fehlen jedoch weltweit Studien. Es ist dennoch zwingend notwendig, die spezifischen von den unspezifischen Fördereffekten zu unterscheiden. Eine Studie mit einer Placebogruppe mit gleicher Förderdauer und Zuwendung und einer unbehandelten Kontrollgruppe sowie eine nicht randomisierte und individuelle Zuweisung zu spezifischen Förderprogrammen könnte hierfür ein sinnvoller Lösungsansatz sein.

Background: The relationship between phoneme awareness, rapid automatized naming (RAN), verbal short-term/working memory (ST/WM) and diagnostic category is investigated in control and dyslexic children, and the extent to which this depends on orthographic complexity. Methods: General cognitive, phonological and literacy skills were tested in 1,138 control and 1,114 dyslexic children speaking six different languages spanning a large range of orthographic complexity (Finnish, Hungarian, German, Dutch, French, English). Results: Phoneme deletion and RAN were strong concurrent predictors of developmental dyslexia, while verbal ST/WM and general verbal abilities played a comparatively minor role. In logistic regression models, more participants were classified correctly when orthography was more complex. The impact of phoneme deletion and RAN-digits was stronger in complex than in less complex orthographies. Conclusions: Findings are largely consistent with the literature on predictors of dyslexia and literacy skills, while uniquely demonstrating how orthographic complexity exacerbates some symptoms of dyslexia.

The lens of the eye has long been considered as a radiosensitive tissue, but recent research has suggested that the radiosensitivity is even greater than previously thought. The 2012 recommendation of the International Commission on Radiological Protection (ICRP) to substantially reduce the annual occupational equivalent dose limit for the ocular lens has now been adopted in the European Union and is under consideration around the rest of the world. However, ICRP clearly states that the recommendations are chiefly based on epidemiological evidence because there are a very small number of studies that provide explicit biological, mechanistic evidence at doses

Obwohl die Bedeutung des familiären Leseumfelds für den Schriftspracherwerb bekannt ist, werden familiäre Ressourcen bisher noch wenig für die Prävention von Lese-Rechtschreibschwierigkeiten genutzt. Das Elterntraining „Lass uns lesen!“ verbindet häusliches Vorlesen mit der gezielten Förderung der phonologischen Bewusstheit, das heißt der Fähigkeit, Laute und größere sprachliche Einheiten zu erkennen und mit ihnen umzugehen. Um die Effekte des Programms hinsichtlich sprachlicher Fähigkeiten zu untersuchen, wurde die Entwicklung einer Gruppe von Vorschulkindern, die das Programm über einen Zeitraum von 16 Wochen erhielt, mit der Entwicklung einer Wartekontrollgruppe verglichen. Die Kinder, die an „Lass uns lesen!“ teilgenommen hatten, verbesserten sich signifikant hinsichtlich ihrer phonologischen Bewusstheit und tendenziell in ihrem Textverständnis, während sich keine Effekte hinsichtlich Buchstabenkenntnis und Wortschatz zeigten. Eltern können demnach über ein strukturiertes Programm die phonologische Bewusstheit ihrer Kinder fördern und so einen wichtigen Beitrag zur Prävention von Lese-Rechtschreibschwierigkeiten leisten.

Cataracts are the major eye disorder and have been associated mainly with mutations in lens-specific genes, but cataracts are also frequently associated with complex syndromes. In a large-scale high-throughput ENU mutagenesis screen we analyzed the offspring of paternally treated C3HeB/FeJ mice for obvious dysmorphologies. We identified a mutant suffering from rough coat and small eyes only in homozygotes; homozygous females turned out to be sterile. The mutation was mapped to chromosome 7 between the markers 116J6.1 and D7Mit294;4 other markers within this interval did not show any recombination among 160 F2-mutants. The critical interval (8.6 Mb) contains 3 candidate genes (Apoe, Six5, Opa3); none of them showed a mutation. Using exome sequencing, we identified a c.2209T>C mutation in the Xpd/Ercc2 gene leading to a Ser737Pro exchange. During embryonic development, the mutant eyes did not show major changes. Postnatal histological analyses demonstrated small cortical vacuoles; later, cortical cataracts developed. Since XPD/ERCC2 is involved in DNA repair, we checked also for the presence of the repair-associated histone γH2AX in the lens. During the time, when primary lens fiber cell nuclei are degraded, γH2AX was strongly expressed in the cell nuclei; later, it demarcates clearly the border of the lens cortex to the organelle-free zone. Moreover, we analyzed also whether seemingly healthy heterozygotes might be less efficient in repair of DNA damage induced by ionizing radiation than wild types. Peripheral lymphocytes irradiated by 1Gy Cs137 showed 6 hrs after irradiation significantly more γH2AX foci in heterozygotes than in wild types. These findings demonstrate the importance of XPD/ERCC2 not only for lens fiber cell differentiation, but also for the sensitivity to ionizing radiation. Based upon these data, we hypothesize that variations in the human XPD/ERCC2 gene might increase the susceptibility for several disorders besides Xeroderma pigmentosum in heterozygotes under particular environmental conditions.
