9. Genomic analysis of acid tolerance genes and deciphering the function of ydaG gene in mitigating acid tolerance in Priestia megaterium.

Abstract

Adverse environmental conditions, such as acid stress, induce bacteria to employ several strategies to overcome these stressors. These strategies include forming biofilms and activating specific molecular pathways, such as the general stress response (GSR). The genome of Priestia megaterium strain G18 was sequenced using the Illumina NextSeq 500 system, resulting in a de novo assembly of 80 scaffolds. The scaffolded genome comprises 5,367,956 bp with a GC content of 37.89%, and was compared to related strains using the MiGA web server, revealing high similarity to P. megaterium NBRC 15308 and P. aryabhattai B8W22 with ANI scores of 95.4%. Phylogenetic and ribosomal multilocus sequence typing (rMLST) analyses, based on the 16S rRNA and ribosomal protein-encoding alleles, confirmed close relationships within the P. megaterium species. Functional annotation identified 5,484 protein-coding genes, with 72.31% classified into 22 COG categories, highlighting roles in amino acid transport, transcription, carbohydrate metabolism, and ribosomal structure. An in-depth genome analysis of P. megaterium G18 revealed several key genes associated with acid tolerance. Targeted inactivation of the ydaG gene from SigB regulon, a general stress response gene, significantly reduced growth under acidic conditions compared to the wild type. qRT-PCR analysis showed increased ydaG expression in acidic conditions, further supporting its role in acid stress response. Microscopic analysis revealed no morphological differences between wild-type and mutant cells, suggesting that ydaG is not involved in maintaining cellular morphology but in facilitating acid tolerance through stress protein production. This research contributes to understanding the molecular mechanisms underlying acid tolerance in soil bacteria, P. megaterium, shedding light on potential applications in agriculture and industry. Abstract

Bacterial and fungal endophytes were isolated and characterized from root and shoot of Datura metel and studied for their antimicrobial properties. Molecular identification of the endophytes, both bacteria and fungi were done using 16S rDNA and 18S rDNA sequencing, respectively. Out of the total bacterial endophytes, Bacillus subtilis was predominant in both the tissues. Of the nine fungal endophytes isolated both from root and shoot, Aspergillus versicolor was found to be dominant. These two dominant species of endophytes, B. subtilis and A. versicolor, were subjected to mass multiplication, and secondary metabolites extraction of the host plant endophytes were performed using solvents of different polarity. The respective extracts were then studied for their antimicrobial activity against different Vibrio cholerae strains. Both bacterial and fungal extracts showed impressive activity against the V. cholerae strains P5, NE2 and VC7233. [ABSTRACT FROM AUTHOR]

Abstract

Chickpea (Cicer arietinum) is grown widely in India because the seeds are rich source of protein for the vegetarian population of country. However, chickpea cultivation is declining over the period of time due to heavy incidences of pests and diseases. Helicoverpa armigera is a major pest in the field and nonavailability of resistant varieties lead to heavy losses of yield per year. Crop management practices such as application of bio-pesticides, insecticides and integrated pest management are less effective to control this devastating pest. Breeding for development of resistant lines is restricted by lack of resistant sources within the gene pool. Therefore, application of gene technology for chickpea improvement appears to be appropriate approach for development of Helicoverpa resistant lines. Genetic transformation of chickpea using various versions of Bacillus thuringiensis (Bt) insecticidal genes have been carried out and found to confer resistance to pod borers in the laboratory bioassays. The most preferred genetically modified (GM) chickpea for field release is pyramided lines having two or more Bt genes with diverse mode of action for effective management of Helicoverpa. 37. Antimicrobial activity of bacterial endophytes from Chirata (Swertia chirataWall.) and Datura (Datura stramoniumL.)

Abstract

Background: Exploration of biocides in microbial system becomes increasingly interesting. Among the different microbial systems, the study of endophytes becomes an important field of research because of their agricultural as well as pharmaceutical importance. Bacterial endophytes from two medicinal plants from North Easter states of India had been studied for their potential antibacterial and antifungal properties. Results: Bacterial endophytes from two medicinal plant species abundantly available in North Eastern states of India, Chirata (Swertia chirata) and Datura (Datura stramonium), had been isolated and studied for their anti-bacterial and anti-fungal activities. Six bacterial endophytes were isolated from Chirata and 5 from Datura. All these bacterial isolates were initially isolated by observing their colony morphology, and then 9 morphologically distinct bacterial isolates were identified at molecular level by their 16s rRNA sequencing. Among the identified isolates, 5 were found to be belonged to genus Staphylococcus, 2 Bacillus, and one each belonged to Rhodococcusand Cupriavidus. Antibacterial study was carried out against 2 plant pathogenic bacteria, viz., Xanthomonas oryzaeand Ralstonia solanacearum. Antifungal study was carried out against 2 plant pathogenic fungi, viz., Fusarium oxysporumand F. solani. Out of 6 bacterial isolates from Chirata, one showed antibacterial as well as antifungal properties, and another one showed antifungal properties only. Likewise, out of 5 bacterial isolates from Datura, one found to have both antibacterial and antifungal properties, and another one has only antifungal properties. Conclusion: Few bacterial endophytes isolated from S. chirataand D. stramoniumL., abundantly available in North Eastern part of India, had potential antibacterial as well as antifungal properties. This could be considered as the baseline information on potential antibacterial and antifungal agents from bacterial endophytes. Further investigations are required to assess their efficacy against several other bacterial and fungal diseases of economic importance.
