In the era of frequently changing climatic conditions along with ever increasing world population, it becomes imperative to ensure food security. The burden of biotic stresses pose serious threat to crop productivity, therefore, early and accurate detection of plant diseases is essential. Conventional methods exclusively rely on human expertise, and are often labor-intensive, time-consuming, and prone to errors. Recent advancements in machine learning (ML) offer promising alternatives by automating the disease detection processes with high precision and efficiency. We comprehensively analyze various ML techniques, including Convolutional Neural Networks (CNNs), Recurrent Neural Networks (RNNs), Support Vector Machines (SVMs), Random Forest (RF), and Deep Learning Architectures like ResNet and Inception, among others, highlighting their methodologies, datasets, performance metrics, and real-world applications. This systematic review provides a comprehensive analysis after text mining the most recent literature resources of the last half a decade. The review discusses the proposed models, techniques, accuracy, feature selection, extraction methods, the types of datasets used to perform experiments, and the sources of the datasets. Additionally, this review provides critical analyses of existing models in the context of their limitations and gaps. Our findings suggest that while ML based methods demonstrate substantial potential for enhancing agricultural disease management, there is a urgent need for more robust, scalable, and adaptable solutions to address diverse agricultural conditions and disease complexities. By systematically analyzing the extracted data, this review aspires to provide a valuable resource for researchers and practitioners aiming to develop and implement ML-based systems for crop disease detection, thereby contributing to sustainable agriculture and enhancing food security.

Smart nanofertilizers (NFs) represent a promising frontier in agricultural technology, offering precise nutrient delivery and stress resilience to enhance crop growth and productivity while minimizing the environmental impact. This opinion article explores the potential of smart NFs in revolutionizing traditional fertilization practices and addressing the challenges facing modern agriculture. By harnessing the unique properties of nanomaterials, smart NFs including noble metal nanoparticles, green NFs, and novel NFs from industrial waste enable targeted nutrient delivery, enhanced nutrient use efficiency, and stress tolerance in plants. However, their widespread adoption faces regulatory, safety, and scalability challenges that require interdisciplinary collaboration and concerted efforts from stakeholders. Despite these hurdles, smart NFs hold immense promise for promoting sustainable agriculture and ensuring food security in frequently changing global climatic conditions.

Salinity stress typically occurs due to excessive accumulation of water-soluble salts i.e., NaCl, which can further induce osmotic stress, ionic stress as well as oxidative stress in plants, imposing many harmful effects on the growth and development of plants. However, plants respond to salinity stress through a range of cellular mechanisms which provides enhanced tolerance against salinity stress. Genetic regulation of salinity stress response through protein coding transcripts has already been extensively studied; however, involvement of non-coding transcripts such as long noncoding RNAs has recently received significant attention as an important molecule in regulating the plant response during salinity stress. Several salinity stress responsive long non coding RNAs have also been identified and characterized in various plant species. This review is an attempt to critically discuss the role of lncRNAs during salinity stress in plants.

The quality of waters is disturbing day by day by various inorganic and organic pollutants. Among various strategies developed so far the technique of phytoremediation using aquatic plants is most preferable. Aquatic ecosystems are facing high level of stress and depletion due to the inputs of polluting materials. Nonetheless, there are certain species of aquatic macrophytes that have ability to cope with these stressful conditions even high concentration of various organic and inorganic pollutants present in water. These species are useful in polluted water treatment through phytoremediation or bioremediation technologies. Among the various aquatic plant species, Azolla, Eichhornia, Lemna, Potamogeton, Spirodela, Wolfia, and Wolfialla have been reported as phytoremediators and also they are highly efficient in reducing aquatic contamination through bioaccumulation of contaminants in their body tissues. Among the various aquatic species, water hyacinth (Eichhornia) is highly resistant and can tolerate the toxicity of heavy metals, phenols, formaldehydes, formic acids, acetic acids and oxalic acids even in their high concentrations. Likewise some other species of the family Lemnaceae are very efficient to reduce the percentage of biochemical oxygen demand (BOD), chemical oxygen demand (COD), as well as impact of HMs (heavy metals), and various ionic forms of nitrogen and phosphorus. Here in this review we are providing up-to-date information regarding the utilization of these aquatic plants for the bioremediation of contaminated waters. The review is primarily focused on the specific capabilities of aquatic plants and as an important tool in phytotechnologies in the management of contaminants in aquatic environment.

Helicases function as key enzymes in salinity stress tolerance, and the role and function of PDH45 (pea DNA helicase 45) in stress tolerance have been reported in different crops with selectable markers, raising public and regulatory concerns. In the present study, we developed five lines of marker-free PDH45-overexpressing transgenic lines of rice (Oryza sativa L. cv. IR64). The overexpression of PDH45 driven by CaMV35S promoter in transgenic rice conferred high salinity (200 mM NaCl) tolerance in the T1 generation. Molecular attributes such as PCR, RT-PCR, and Southern and Western blot analyses confirmed stable integration and expression of the PDH45 gene in the PDH45-overexpressing lines. We observed higher endogenous levels of sugars (glucose and fructose) and hormones (GA, zeatin, and IAA) in the transgenic lines in comparison to control plants (empty vector (VC) and wild type (WT)) under salt treatments. Furthermore, photosynthetic characteristics such as net photosynthetic rate (Pn), stomatal conductance (gs), intercellular CO2 (Ci), and chlorophyll (Chl) content were significantly higher in transgenic lines under salinity stress as compared to control plants. However, the maximum primary photochemical efficiency of PSII, as an estimated from variable to maximum chlorophyll a fluorescence (Fv/Fm), was identical in the transgenics to that in the control plants. The activities of antioxidant enzymes, such as catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), and guaiacol peroxidase (GPX), were significantly higher in transgenic lines in comparison to control plants, which helped in keeping the oxidative stress burden (MDA and H2O2) lesser on transgenic lines, thus protecting the growth and photosynthetic efficiency of the plants. Overall, the present research reports the development of marker-free PDH45-overexpressing transgenic lines for salt tolerance that can potentially avoid public and biosafety concerns and facilitate the commercialization of genetically engineered crop plants.

A defined balance between the generation and scavenging of reactive oxygen species (ROS) is essential to utilize ROS as an adaptive defense response of plants under biotic and abiotic stress conditions. Moreover, ROS are not only a major determinant of stress response but also acts as signaling molecule that regulates various cellular processes including plant-microbe interaction. In particular, rhizosphere constitutes the biologically dynamic zone for plant–microbe interactions which forms a mutual link leading to reciprocal signaling in both the partners. Among plant–microbe interactions, symbiotic associations of arbuscular mycorrhizal fungi (AMF) and arbuscular mycorrhizal-like fungus especially Piriformospora indica with plants are well known to improve plant growth by alleviating the stress-impacts and consequently enhance the plant fitness. AMF and P. indica colonization mainly enhances ROS-metabolism, maintains ROS-homeostasis, and thereby averts higher ROS-level accrued inhibition in plant cellular processes and plant growth and survival under stressful environments. This article summarizes the major outcomes of the recent reports on the ROS-generation and scavenging and signaling in biotic-abiotic stressed plants with AMF and P. indica colonization. Overall, a detailed exploration of ROS-signature kinetics during plant-AMF/P. indica interaction can help in designing innovative strategies for improving plant health and productivity under stress conditions.

Owing to its exceptional ability to efficiently promote plant growth, protection and stress tolerance, a mycorrhiza like endophytic Agaricomycetes fungus Piriformospora indica has received a great attention over the last few decades. P. indica is an axenically cultiviable fungus and exhibits its versatility for colonizing/hosting a broad range of plant species through directly manipulating mainly plant hormone-signaling during the course of mutualism. P. indica-root colonization leads to a better plant performance in all respect including enhanced root proliferation by indole-3-acetic acid production which in turn results into better nutrient-acquisition and subsequently to improved crop growth and productivity. Additionally, P. indica can induce both local and systemic resistance to fungal and viral plant diseases through signal transduction. P. indica-mediated stimulation in antioxidant defense system components and expressing stress-related genes can confer crop/plant stress tolerance. Therefore, P. indica can biotize micropropagated plantlets and also help these plants overcome transplantation shock. Nevertheless, it can also be involved in a more complex symbiotic relationship, such as tripartite symbiosis and can enhance population dynamic of plant growth promoting rhizobacteria. In brief, P. indica can be utilized as a plant promoter, bio-fertilizer, bioprotector, bioregulator and biotization agent. The outcome of the recent literature appraised herein will help us to understand the physiological and molecular bases of mechanisms underlying P. indica-crop plant mutual relationship. Together, the discussion will be functional to comprehend the usefulness of crop plant-P indica association in both achieving new insights into crop protection/improvement as well as in sustainable agriculture production.

Genetic material always remains at the risk of spontaneous or induced damage which challenges the normal functioning of DNA molecule, thus, DNA repair is vital to protect the organisms against genetic damage. DNA hHelicases, the unique molecular motors, are emerged as potentialprospective molecules to engineer stress tolerance in plants and are involved in a variety of DNA nucleic acid metabolismc processes including DNA repair. The DNA repair helicase, OsXPB2 is an evolutionary conserved protein present in different organisms, including plants. Availability of few efficient promoters for gene expression in plants provoked us to study the promoter of XPB for better understanding of gene regulation under stress The analysis of promoter sequence from plant genome is important in understanding the gene regulation. The in vivo validation of functionality/activity of novel stress inducible promoter of rice OsXPB2 gene promoter under abiotic and hormonal stress conditions was performed by Agrobacterium-mediated transient assay in tobacco leaves using OsXPB2::GUS chimeric construct. Our resultsThe present research revealed that OsXPB2 promoter contains cis-elements accounting for various abiotic stresses (salt, dehydration or cold) and hormone (Auxin, ABA or MeJA) induced GUS expression/activity in the promoter-reporter assay. The promoter region of OsXPB2 contains CACG, GTAACG, CACGTG, CGTCA CCGCCGCGCT cis acting-elements which are reported to be salt, dehydration, cold, MeJA or ABA responsive, respectively. Functional analysis was done by Agrobacterium-transient assays using agroinfiltration in tobacco leaves, followed by GUS staining and fluorescence quantitative analyses. The results revealed high induction of GUS activity under multiple abiotic stresses as compared to mock treated control. The present findings suggest that OsXPB2 promoter is a multi-stress inducible promoter and has potential applications in sustainable crop production under abiotic stresses by regulating desirable patterns of gene expression.

Sulfur (S) stands fourth in the list of major plant nutrients after N, P and K. Sulfate (SO42-), a form of soil-S taken up by plant roots is metabolically inert. As the first committed step of S-assimilation, ATP-sulfurylase (ATP-S) catalyzes SO42--activation and yields activated high-energy compound adenosine-5′-phosphosulfate (APS) that is reduced to sulfide (S2-) and incorporated into cysteine (Cys). In turn, Cys acts as a precursor or donor of reduced S for a range of S-compounds such as methionine (Met), glutathione (GSH), homo-GSH (h-GSH) and phytochelatins (PCs). Among S-compounds, GSH, h-GSH and PCs are known for their involvement in plant tolerance to varied abiotic stresses, Cys is a major component of GSH, h-GSH and PCs; whereas, several key stress-metabolites such as ethylene, are controlled by Met through its first metabolite S-adenosylmethionine. With the major aim of briefly highlighting S-compound-mediated role of ATP-S in plant stress tolerance, this paper: (a) overviews ATP-S structure/chemistry and occurrence, (b) appraises recent literature available on ATP-S roles and regulations, and underlying mechanisms in plant abiotic and biotic stress tolerance, (c) summarizes ATP-S-intrinsic regulation by major S-compounds, and (d) highlights major open-questions in the present context. Future research in the current direction can be devised based on the outcomes of the discussion.

Varied environmental compartments including soils are being contaminated by a myriad toxic metal(loid)s (hereafter termed as ‘metal/s’) mainly through anthropogenic activities. These metals may contaminate food chain and bring irreparable consequences in human. Plant-based approach (phytoremediation) stands second to none among bioremediation technologies meant for sustainable cleanup of soils/sites with metal-contamination. In turn, the capacity of plants to tolerate potential consequences caused by the extracted/accumulated metals decides the effectiveness and success of phytoremediation system. Chelation is among the potential mechanisms that largely govern metal-tolerance in plant cells by maintaining low concentrations of free metals in cytoplasm. Metal-chelation can be performed by compounds of glutathione (GSH) (reduced GSH; phytochelatins, PCs; metallothioneins, MTs) and non-GSH (histidine, nicotianamine, organic acids) origin. This paper presents an appraisal of recent reports on both GSH and non-GSH associated compounds in an effort to shed light on the significance of these compounds in metal-plant tolerance, as well as to provide scientific clues for the development of phytoextraction strategies.

Despite a significant drop in malaria deaths during the past decade, malaria continues to be one of the biggest health problems around the globe. WD40 repeats (WDRs) containing proteins comprise one of the largest and functionally diverse protein superfamily in eukaryotes, acting as scaffolds for assembling large protein complexes. In the present study, we report an extensive in silico analysis of the WDR gene family in human malaria parasite Plasmodium falciparum. Our genome-wide identification has revealed 80 putative WDR genes in P. falciparum (PfWDRs). Five distinct domain compositions were discovered in Plasmodium as compared to the human host. Notably, 31 PfWDRs were annotated/re-annotated on the basis of their orthologs in other species. Interestingly, most PfWDRs were larger as compared to their human homologs highlighting the presence of parasite-specific insertions. Fifteen PfWDRs appeared specific to the Plasmodium with no assigned orthologs. Expression profiling of PfWDRs revealed a mixture of linear and nonlinear relationships between transcriptome and proteome, and only nine PfWDRs were found to be stage-specific. Homology modeling identified conservation of major binding sites in PfCAF-1 and PfRACK. Protein-protein interaction network analyses suggested that PfWDRs are highly connected proteins with ~1928 potential interactions, supporting their role as hubs in cellular networks. The present study highlights the roles and relevance of the WDR family in P. falciparum, and identifies unique features that lay a foundation for further experimental dissection of PfWDRs.

BackgroundThe DEAD-box helicases are required mostly in all aspects of RNA and DNA metabolism and they play a significant role in various abiotic stresses, including salinity. The p68 is an important member of the DEAD-box proteins family and, in animal system, it is involved in RNA metabolism including pre-RNA processing and splicing. In plant system, it has not been well characterized. Here we report the cloning and characterization of p68 from pea (Pisum sativum) and its novel function in salinity stress tolerance in plant.ResultsThe pea p68 protein self-interacts and is localized in the cytosol as well as the surrounding of cell nucleus. The transcript of pea p68 is upregulated in response to high salinity stress in pea. Overexpression of p68 driven by constitutive cauliflower mosaic virus-35S promoter in tobacco transgenic plants confers enhanced tolerances to salinity stress by improving the growth, photosynthesis and antioxidant machinery. Under stress treatment, pea p68 overexpressing tobacco accumulated higher K+ and lower Na+ level than the wild-type plants. Reactive oxygen species (ROS) accumulation was remarkably regulated by the overexpression of pea p68 under salinity stress conditions, as shown from TBARS content, electrolyte leakage, hydrogen peroxide accumulation and 8-OHdG content and antioxidant enzyme activities.ConclusionsTo the best of our knowledge this is the first direct report, which provides the novel function of pea p68 helicase in salinity stress tolerance. The results suggest that p68 can also be exploited for engineering abiotic stress tolerance in crop plants of economic importance.
