Abstract

Nosema bombycis is a pathogen causing pebrine disease of the mulberry silkworm, Bombyx mori. The disease spreads mainly through transovarian transmission of environmental spore and secondarily through contaminated food, rearing appliances, etc. by primary spores. Ultra-structure studies using Scanning and Transmission Electron Microscopy of the two spores revealed differences in the primary spore which contained a Short Polar Tube (ST) with a thin wall (< 200 nm), and the environmental spore which contained a Long Polar Tube (LT) with a thick wall (> 200 nm). It is observed that the yield of spore with LT is highest in female moths, whereas, it is spores with ST are highest in male moths. Besides ultra-structures, the development pattern of the two types of spores is also different. It is an interesting finding in the present study that, spores of N. bombycis produced two types of spores and multiplied in different gender under the influence of the host’s reproductive role and physiology for transmission of disease. The detailed study on ultra-structure of disporous N. bombycis in both the sexes of B. mori along with their development in the life cycle stages of silkworms with special reference to the inoculum concentration of spore is discussed.

Abstract

In nature, the population of Nosema bombycis (Microsporidia) causing pebrine disease is small and their development is extremely slow and only few ultimately producing spores. Pebrine infected silkworm, Bombyx mori larvae collected from sericulture field were alive till 3rd generation though the concentration of N.bombycis spore was very high (2.4 - 3.0 x 108 spores. mL-1). All larvae were died during 4th generation with extremely high concentration of pebrine spores (3.0 - 4.0 x 109 spores. mL-1) and mostly contain long polar tube (LT). Alternately, all larvae were died immediately (at 3rd stage of 1st generation) when it was artificially inoculated with same concentration of N.bombycis spores harvested from field (2.4 - 3.0 x 108 spores. mL-1) though concentration of spores harvest was very less (3.0- 4.0 x 106 spores. mL-1) and mostly contain short polar tube (ST). Artificially pebrine infected male moth when mated with healthy female moth took six generations to develop pebrine disease and all larvae were died at the 2nd stage with very less spore harvest (3.0 - 10.0 x 105 spores. mL -1). Survival percentage was increased in all generations (~92.0% at 4th generation) when silkworm rearing was conducted under new integrated disease management system.

Abstract

Pebrine caused by Nosema bombycis in the silkworm Bombyx mori L. causes severe economic loss to the sericulture industry. Several species of microsporidia and strains of N. bombycis have been isolated from infected silkworms. The study of the developmental stages of any parasite is important, as it leads to the identification of stages that may be susceptible to control measures. For this study, five microsporidian isolates from B. mori were collected from five different locations in West Bengal, India and a study of the developmental stages from mid-gut smears and histological techniques was undertaken. The observations of mid-gut smears and histological specimens revealed differences in the morphometry and duration of different developmental stages of the parasites.
