The Antarctic terrestrial environment harbors a diverse community of microorganisms, which have adapted to the extreme conditions. The aim of this study was to describe the composition of microbial communities in a diverse range of terrestrial environments (various biocrusts and soils, sands from ephemeral wetlands, biofilms, endolithic and hypolithic communities) in East Antarctica using both molecular and morphological approaches. Amplicon sequencing of the 16S rRNA gene revealed the dominance of Chloroflexi, Cyanobacteria and Firmicutes, while sequencing of the 18S rRNA gene showed the prevalence of Alveolata, Chloroplastida, Metazoa, and Rhizaria. This study also provided a comprehensive assessment of the microphototrophic community revealing a diversity of cyanobacteria and eukaryotic microalgae in various Antarctic terrestrial samples. Filamentous cyanobacteria belonging to the orders Oscillatoriales and Pseudanabaenales dominated prokaryotic community, while members of Trebouxiophyceae were the most abundant representatives of eukaryotes. In addition, the co-occurrence analysis showed a prevalence of positive correlations with bacterial taxa frequently co-occurring together.

The terrestrial biological research team of the 61st Japanese Antarctic Research Expedition conducted a terrestrial biology survey on bacteria, lichen and moss in the northwestern region of the Sør Rondane Mountains. This is the report of the details of operational plans and actual activities.

Abstract The phylum Annelida exhibits high morphological diversity coupled with its extensive ecological diversity, and the process of its evolution has been an attractive research subject for many researchers. Its representatives are also extensively studied in fields of ecology and developmental biology and important in many other biology related disciplines. The study of biomineralisation is one of them. Some annelid groups are well known to form calcified tubes but other forms of biomineralisation are also known. Herein, we report a new interstitial annelid species with black spicules, Thoracophelia minuta sp. nov., from Yoichi, Hokkaido, Japan. Spicules are minute calcium carbonate inclusions found across the body and in this new species, numerous black rod-like inclusions of calcium-rich composition are distributed in the coelomic cavity. The new species can be distinguished from other known species of the genus by these conspicuous spicules, shape of branchiae and body formula. Further, the new species’ body size is apparently smaller than its congeners. Based on our molecular phylogenetic analysis using 18S and 28S sequences, we discuss the evolutionary significance of the new species’ spicules and also the species' progenetic origin.

Antarctic expeditions have a high risk of participant depression owing to long stays and isolated environments. By quantifying the stress state and changes in biomolecules over time before the onset of depressive symptoms, predictive markers of depression can be explored. Here, we evaluated the psychological changes in 30 participants in the Japanese Antarctic Research Expedition using the Patient Health Questionnaire-9 (PHQ-9). Urinary samples were collected every three months for a year, and comprehensive urinary metabolomic profiles were quantified using liquid chromatography time-of-flight mass spectrometry. Five participants showed major depressive episodes (PHQ-9 ≥ 10) at 12 months. The urinary metabolites between these participants and the 25 unaffected participants were compared at individual metabolite and pathway levels. The individual comparisons showed the most significant differences at 12 months in 14 metabolites, including ornithine and beta-alanine. Data from shorter stays showed less significant differences. In contrast, pathway and enrichment analyses showed the most significant difference at three months and a less significant difference at longer stays. These time transitions of urinary metabolites could help in the development of urinary biomarkers to detect subjects with depressive episodes at an early stage.

Abstract Terrestrial plant populations located at the margins of species’ distributions often display reduced sexual reproduction and an increased reliance on asexual reproduction. One hypothesis to explain this phenomenon is that the decline is associated with environmental effects on the energetic costs to produce reproductive organs. In order to clarify the changing processes of sexual reproduction along an elevational gradient, we investigated the sexual reproductive parameters, such as the number of sporophytes and gametangia, in Racomitrium lanuginosum, a dioicous moss found on Mt. Fuji. Matured sporophytes were present only below 3,000 m, and the number of sporophytes per shoot tended to be lower at higher elevation habitats. The numbers of male inflorescences per shoot and antheridia per inflorescence and shoot significantly decreased with increasing elevation. In contrast, the numbers of female inflorescences per shoot and archegonia per inflorescence and shoot varied little across elevations. Synthesis. Our results suggest that the reasons for this limitation are assumed to be limitations in sporophyte development that result in abortion, and the spatial segregation between males and females. Possible reasons for the abortion of sporophytes are the inhibitory effects of low air temperature, a shortened growth period, and winter environmental conditions at higher elevations. Remarkable differences between male and female on various reproductive parameters found in this study are thought to affect the mode of sexual reproduction under the harsh environment. These differences between males and females may be caused by differences in the costs of production and development of gametangia, sensitivity to environmental stressors, and phenological patterns.

We reassess two specimens, part of the late Dr Minoru Imajima’s collections, from Japan of the flabelligerid genus Brada Stimpson, 1853. We re-identify the specimens as B. kudenovi Salazar-Vallejo, 2017 and newly record this species from Japan, which represents the southernmost locality of the genus worldwide. The specimens were previously reported as B. inhabilis (Rathke, 1843), which has Molde, Norway as its type locality. However, the morphological features are identical to the original description of B. kudenovi in the following: the body is whitish in alcohol; there are one or two multiarticulate notochaetae per parapodium; the neuropodial lobes are low, not projecting; and the body papillae are short.

ABSTRACT Culture-independent analysis shows that Legionella spp. inhabit a wide range of low-temperature environments, but to date, no psychrotolerant or psychrophilic strains have been reported. Here, we characterized the first cultivated psychrotolerant representative, designated strain TUM19329T, isolated from an Antarctic lake using a polyphasic approach and comparative genomic analysis. A genome-wide phylogenetic tree indicated that this strain was phylogenetically separate at the species level. Strain TUM19329T shared common physiological traits (e.g., Gram-negative, limited growth on buffered charcoal-yeast extract α-ketoglutarate [BCYEα] agar with l-cysteine requirements) with its relatives, but it also showed psychrotolerant growth properties (e.g., growth at 4°C to 25°C). Moreover, this strain altered its own cellular fatty acid composition to accumulate unsaturated fatty acid at a lower temperature, which may help maintain the cell membrane fluidity. Through comparative genomic analysis, we found that this strain possessed massive mobile genetic elements compared with other species, amounting to up to 17% of the total genes. The majority of the elements were the result of the spread of only a few insertion sequences (ISs), which were spread throughout the genome by a “copy-and-paste” mechanism. Furthermore, we found metabolic genes, such as fatty acid synthesis-related genes, acquired by horizontal gene transfer (HGT). The expansion of ISs and HGT events may play a major role in shaping the phenotype and physiology of this strain. On the basis of the features presented here, we propose a new species—Legionella antarctica sp. nov.—represented by strain TUM19329T (= GTC 22699T = NCTC 14581T). IMPORTANCE This study characterized a unique cultivated representative of the genus Legionella isolated from an Antarctic lake. This psychrotolerant strain had some common properties of known Legionella species but also displayed other characteristics, such as plasticity in fatty acid composition and an enrichment of mobile genes in the genome. These remarkable properties, as well as other factors, may contribute to cold hardiness, and this first cultivated cold-tolerant strain of the genus Legionella may serve as a model bacterium for further studies. It is worth noting that environmentally derived 16S rRNA gene phylotypes closely related to the strain characterized here have been detected from diverse environments outside Antarctica, suggesting a wide distribution of psychrotolerant Legionella bacteria. Our culture- and genome-based findings may accelerate the ongoing studies of the behavior and pathogenicity of Legionella spp., which have been monitored for many years in the context of public health.

Increased research attention is being given to bacterial diversity associated with lichens. Rock tripe lichens (Umbilicariaceae) were collected from two distinct Antarctic biological regions, the continental region near the Japanese Antarctic station (Syowa Station) and the maritime Antarctic South Orkney Islands (Signy Island), in order to compare their bacterial floras and potential metabolism. Bulk DNA extracted from the lichen samples was used to amplify the 18S rRNA gene and the V3-V4 region of the 16S rRNA gene, whose amplicons were Sanger- and MiSeq-sequenced, respectively. The fungal and algal partners represented members of the ascomycete genus Umbilicaria and the green algal genus Trebouxia, based on 18S rRNA gene sequences. The V3-V4 sequences were grouped into operational taxonomic units (OTUs), which were assigned to eight bacterial phyla, Acidobacteriota, Actinomyceota, Armatimonadota, Bacteroidota, Cyanobacteria, Deinococcota, Pseudomonadota and the candidate phylum Saccharibacteria (also known as TM7), commonly present in all samples. The OTU floras of the two biological regions were clearly distinct, with regional biomarker genera, such as Mucilaginibacter and Gluconacetobacter, respectively. The OTU-based metabolism analysis predicted higher membrane transport activities in the maritime Antarctic OTUs, probably influenced by the sampling area’s warmer maritime climatic setting.

An Antarctic wintering-over station is a unique environment, as a small isolated society facing extreme survival margins. Psychological surveys have been done over ten years, including the Baum test, the Positive and Negative Affect Schedule (PANAS), the Coping Orientation to Problems Experienced (COPE), Subjective Health Complaints Inventory (SHC), the Two-Sided Personality Scale (TSPS) and medical consultations in Syowa Station, a Japanese Antarctic station to reveal the mental status of team members. Team members experienced fewer physical health risks in Antarctica than in Japan. Wintering-over team members reinterpreted situations positively and accepted their environment, sought instrumental social support, planned ahead, and used active coping skills and humour to overcome difficulties. They did not act out emotionally or deny problems. Individuals exhibited two types of coping, either stability through maintaining a previous lifestyle or flexible adjustment to a new way of life. Positive affect remained constant during the wintering-over period. In living through a harsh reality, team members drew support from the subjective feelings of an “internal relationship” with home or family in their minds. Thus, an Antarctic wintering-over station is an ideal isolated environment for psychological surveys, which can help understand future space travel and group managements in everyday societies.

The difference absorption spectrum (DAS) of porphyrin molecules (tetraphenyl-porphyhrin sulfonic acid, TPPS) in aqueous solution induced by continuous wave laser irradiation has been reported previously. It was interpreted that the DAS was caused by the formation of TPPS aggregates induced by laser irradiation. However, transient spectra similar in their shape have already been reported and are attributed to the excited-state absorption and saturable absorption (SA) effects due to the triplet state formation in TPPS. In the present study, we investigated the triplet quenching effect by O2 on the DAS of TPPS aqueous solution and revealed that it originated from the triplet state formation. We also found that mixing the appropriate amount of MeOH and NaOH in TPPS aqueous solution increased its absorbance change by more than 50 times. This may be due to the decrease in dissolved oxygen concentration by mixing them. This result suggests the possibility of controlling the performance of NLO materials by adjusting the solvent mixture ratio and base/acid concentration.

This study aimed to evaluate disease and injury trends among wintering members of the Japanese Antarctic Research Expedition. Obtained information is indispensable to the advancement of medical system and research. Summation was performed based on medical records of reports prepared by each expedition over the period 1956–2016. The clinical department’s classification methods of the names of injuries and diseases varied among expeditions, but the names were integrated following the same classification. Of 1734 members (29 women), 6837 disease or injury cases (4 cases/person) were recorded. The rates of cases were as follows: surgical-orthopaedic (45.3%), internal medical (21.7%), dental (11.6%), dermatological (8.4%), ophthalmological (5.8%), otorhinolaryngological (5.3%), psychiatric (1.6%), and urological (0.1%) cases. There was no major change in rates by type of medical case in each expedition. This analysis made it possible to prepare medical facilities, content of physical examinations to select members, training of physicians before departure, preventive hygiene at sites, and medical research themes.

Antarctica has one of the most extreme environments on Earth, with low temperatures and low nutrient levels. Antarctica’s organisms live primarily in the coastal, ice-free areas which cover approximately 0.18% of the continent’s surface. Members of Cyanobacteria and eukaryotic algae are important primary producers in Antarctica since they can synthesize organic compounds from carbon dioxide and water using solar energy. However, community structures of photosynthetic algae in Antarctica have not yet been fully explored at molecular level. In this study, we collected diverse algal samples in lacustrine and hydro-terrestrial environments of Langhovde and Skarvsnes, which are two ice-free regions in East Antarctica. We performed deep amplicon sequencing of both 16S ribosomal ribonucleic acid (rRNA) and 18S rRNA genes, and we explored the distribution of sequence variants (SVs) of these genes at single nucleotide difference resolution. SVs of filamentous Cyanobacteria genera, including Leptolyngbya, Pseudanabaena, Phormidium, Nodosilinea, Geitlerinama, and Tychonema, were identified in most of the samples, whereas Phormidesmis SVs were distributed in fewer samples. We also detected unicellular, multicellular or heterocyst forming Cyanobacteria strains, but in relatively small abundance. For SVs of eukaryotic algae, Chlorophyta, Cryptophyta, and Ochrophyta were widely distributed among the collected samples. In addition, there was a red colored bloom of eukaryotic alga, Geminigera cryophile (Cryptophyta), in the Langhovde coastal area. Eukaryotic SVs of Acutuncus antarcticus and/or Diphascon pingue of Tardigrada were dominant among most of the samples. Our data revealed the detailed structures of the algal communities in Langhovde and Skarvsnes. This will contribute to our understanding of Antarctic ecosystems and support further research into this subject.

Observations of weather parameters at two ice-free areas (Yukidori Zawa in Langhovde and Kizahashi Hama in Skarvsnes) on the Soya Coast, East Antarctica, had been conducted since 2010 using automatic weather stations (AWSs). By comparing the data obtained from two AWSs with those at Syowa Station (Ongul Islands), we observed a degree of variability in seasonal and annual air temperature and solar radiation. However, throughout the year, both ice-free areas experienced approximately 1-2℃ higher temperatures and 50% lower wind speeds compared with Syowa Station. Relative humidity and solar radiation (which may be influenced by topographic shading) recorded by both AWS, were ～10% lower than those recorded at Syowa Station. In addition, the numbers of days above 0℃ at both sites was nearly twice that at Syowa Station; we recorded ～200 W/m2 of solar radiation and wind speeds of 3-4 m/s. These differences in weather parameters between the Ongul Islands and ice-free areas on the southern Soya Coast potentially affect the activities of terrestrial biota, and might account for the significant differences in vegetation between the two areas.

In the seventh five-year plan of the Japanese Antarctic Research Expedition, a group of Japanese scientists (led by the National Institute of Polar Research) together with a group of Swedish scientists, conducted field surveys to better understand the glaciology of the ice sheet in Dronning Maud Land, East Antarctica, during the 2007/2008 austral summer season. This paper reports on the planning and field preparations, and outlines the scientific achievements of the field expedition. We have gained numerous new scientific insights on the spatio-temporal distribution of the ice sheet environment in the inland plateau. Here, we provide an overview of the new knowledge gained.

Saxicolous rock ripe lichens that grow on rocks in the East Antarctic fellfields were sampled for phylotypic characterization of its constituent mycobionts (fungi) and photobionts (algae and cyanobacteria). The rock tripe lichen-forming fungal and algal phylotypes were classified under the common lichen-forming genera of ascomycetes, namely, Umbilicaria, and green algae, namely, Trebouxia and Coccomyxa. However, phylotypes of the green algal chloroplasts and the lichen-associated cyanobacteria showed unexpectedly high diversity. The detected chloroplast phylotypes were not fully affiliated with the green algal genera Trebouxia or Coccomyxa. The predominant chloroplast phylotype demonstrated maximum resemblance to Neglectella solitaria, which is neither a known Antarctic species nor a typical lichen photobiont. Another dominant chloroplast phylotype belonged to the atypical Antarctic green algae family. Cyanobacterial phylotypes were dominated by those affiliated with the Microcoleus species rather than the well-known lichen-associates, Nostoc species. The occurrences of these Microcoleus-affiliated cyanobacterial phylotypes were specifically abundant within the Yukidori Valley site, one of the Antarctic Specially Protected Areas (ASPA). The ASPA site, along with another 50 km-distant site, yielded most of the cryptic diversity in the phylotypes of chloroplasts and cyanobacteria, which may contribute to the phenotypic variability within the rock tripe lichen photobionts.

Since most studies about community structures of microorganisms in Antarctic terrestrial lakes using molecular biological tools are mainly focused on bacteria, limited information is available about archaeal and eukaryotic microbial diversity. In this study, the biodiversity of microorganisms belonging to all three domains in a typical Antarctic freshwater glacier lake (Yukidori-Ike) was revealed using small subunit ribosomal RNA (SSU rRNA) clone library analysis. The bacterial clones were grouped into 102 operational taxonomic units (OTUs) and showed significant biodiversity. Betaproteobacteria were most frequently detected, followed by Cyanobacteria, Bacteroidetes, and Firmicutes as major lineages. In contrast to the bacterial diversity, much lower archaeal diversity, consisting of only two OTUs of methanogens, was observed. In the eukaryotic microbial community consisting of 20 OTUs, Tardigradal DNA was remarkably frequently detected. Genera affiliated with the phyla Ciliophora, Cryptomycota, Chlorophyta, Bacillariophyta, and Apusozoa were also detected. The biodiversity and species compositions of the whole microbial community of Lake Yukidori-Ike are similar to those of freshwater environments in temperate regions but are different from saline lakes in Antarctica, indicating that the salinity seems to affect the microbial composition more than the temperature.

A study on the effect of the solar ultra-violet radiation on the human skin fibroblast cells revealed that the production of matrix metalloproteinase-2 was inhibited by the radiation. A CO2 incubator connected by optical fibers to a reflector telescope for collecting the solar light was built at Syowa station by the 49th Japanese Antarctica Research Expedition. The direction of the telescope was continuously controlled by a sun-tracker to follow the movement of the Sun automatically. The intensity of the collected light was monitored by a portable spectrophotometer housed inside. The human skin fibroblast cells were incubated in the CO2 chamber to investigate the effect of the solar radiation at Syowa station and were compared with those reference experiments at a laboratory in Japan. The results showed cell damage by strong UV radiation. The production of matrix metalloproteinase-2 was prompted by the moderate UV-B, but was inhibited by the strong UV-B radiation, as studied under laboratory conditions in Japan. The effect of strong solar radiation at Syowa station involving the radiation of UV-B region was estimated to be of the same extent of the radiation caused by an artificial UV-B light with the intensity more than 50 mJ/cm2.

From February 15th to 23rd in 2009, the authors surveyed biosecurity measures being implemented at Australian Antarctic Division (AAD) against non-native species introduction into Antarctica. By visiting facilities and conducting interviews, the authors managed to understand the entire system of the biosecurity measures and management policies at AAD and the situation on the ground in four topics, 1. Management of equipment and machinery, 2. Management of expeditioners＇ clothing and gear, 3. Management of cargo, and 4. Education on expeditioners. This study demonstrated AAD strongly recognizes the risk of transporting non-native species into Antarctica in association with various activities of the Antarctic program thus has implemented comprehensive management policies with a range of meticulous measures to minimize the risk through the management of equipment, gear, clothing, cargo and the education of expeditioners. The thorough education on the risk of biological invasion in Antarctica and thereby the importance of the biosecurity measures for the expeditioners and staff conducted at AAD in particular was considered highly effective. Consideration and development of the organizational biosecurity measures for the JARE operations basedupon the data presented in this study is recommended to promote the conservation of the pristine ecosystem in and around Syowa Station in Antarctica.

The terrestrial cyanobacterium Nostoc commune Vaucher ex Bornet et Flahault occurs worldwide, including in Japan and Antarctica. N. commune has a large amount of extracellular polysaccharides (EPS) that hold moisture and protect the cells and at the same time accumulate light-blocking substances which is believed to play an important part in adaptation to a severe environment. To evaluate the photoadaptation processes in N. commune and clarify the role(s) of EPS under ambient environmental condition at Antarctica, separation of cells from EPS is necessary. High yield is a prerequisite for the use of only small amount of natural N. commune from Antarctica. For this purpose, we developed a separation method by improving the Percoll density gradient centrifugation method using an EPS-coated field-grown Nostoc population. We established the most suitable condition to separate naked cells from EPS at high yield retaining high photosynthetic activity. The method is composed of centrifugation of cell homogenated N. commune in 10% (v/v) Percoll to separate cells efficiently from EPS followed by fractionating centrifugation to remove impurities using the gradient of Percoll (80% and 50%, v/v).

Pollen taxa in sediment samples can be identified based on morphology. However, closely related species do not differ substantially in pollen morphology, and accurate identification is generally limited to genera or families. Because many pollen grains in glaciers contain protoplasm, genetic information obtained from pollen grains should enable the identification of plant taxa at the species level. In the present study, species identification of Pinus pollen grains was attempted using whole-genome amplification (WGA). We used pollen grains extracted from surface snow (depth, 1.8–1.9 m) from the Belukha glacier in the summer of 2003. WGA was performed using a single pollen grain. Some regions of the chloroplast genome were amplified by PCR, and the DNA products were sequenced to identify the pollen grain. Pinus includes approximately 111 recognized species in two subgenera, four sections, and 11 subsections. The tree species Pinus sibirica and P. sylvestris are currently found at the periphery of the glacier. We identified the pollen grains from the Belukha glacier to the level of section or subsection to which P. sibirica and P. sylvestris belong. Moreover, we specifically identified two pollen grains as P. sibirica or P. cembra. Fifteen species, including P. sibirica, were candidates for the remaining pollen grain.

The distribution and colonization of flora in Antarctica may be limited by disturbance events, including flooding. Recently, we observed a large hole in the perennial snow dam below Lake Hyoga Ike (temporary name), located in the Langhovde region on the Sôya Coast in East Antarctica. This phenomenon was also reported by a previous Japanese Antarctic Research Expedition, almost 25 years earlier. The earlier breakage was subsequently closed and the lake was refilled. At present, the surface area of the lake has decreased significantly due to the loss of water. Many of the neighboring valleys and lakes in the southern part of the Langhovde area contain well-developed colonies of algae, mosses, and lichens. By contrast, both Lake Hyoga Ike and the river below the snow dam contain very little flora. Such a contrast in the distribution and colonization of flora suggests that the repeated occurrence of physical disturbances, induced by breaching of the dam, may be a critical factor limiting flora in this ice-free region of Antarctica. We recommend long term observation of the fate of the ice cave and changes in the lake level for the purpose of evaluating climatic change and ecology. The observations also contribute to the maintenance of safety in the field works in this area.

The activities in the 2007-2008 austral summer of the 49th Japanese Antarctic Research Expedition (JARE-49) are reported. JARE-49 consisted of 59 personnel including 30 summer and 29 wintering personnel. In addition, eight observers joined the voyage of R/V Shirase (4 Japanese and one Australian), the operation in the Sor Rondane Mountains (one Japanese), and the inland traverse operation (2 Swedish). R/V Shirase arrived at the ice edge on 14 Desember 2007. She anchored at Syowa Station on 26 December and unloaded ca. 870 t of cargo and fuel by mid-January 2008. A small balloon-borne cryogenic sampler experiment, the unmanned magnetometer network observation, and ecological research on Antarctic lakes were the major scientific programs of JARE-49 in the Syowa Station area. Because the weather condition in this season was extremely bad, some parts of scientific programs, transportation and construction work were delayed.Two special teams tried to access Antarctica directly from Japan on airplanes by using the DROMLAN system. The Japanese-Swedish Traverse Expedition (JASE) was a glaciological program with a long range inland traverse trip. Four JARE-49 members and 4 JARE-48 wintering members started to the meeting point with the Swedish traverse team on 14 November 2007, and returned on 26 January 2008. A 7-person special team for the geological field work in the Sor Rondane Mountains carried out a field survey for 75 days from 23 November 2007 to 5 February 2008 with skidoo, camping on ice. These two teams left Japan by air before the main team, and also returned to Japan by air before the main team.

Observations on the limnological properties, samplings of waters and bottom assemblages for biological and ecological studies, and some field experimental studies at several lakes in Soya Coast ice-free areas, were carried out during the austral summer season in the 49th Japanese Antarctic Research Expedition (JARE), 2007-2008. These studies were planned as one of the research projects named, "Studies on the changes of polar environments and ecosystems (P-3)" and the monitoring studies named "Monitoring for ecosystems (M-4)" during the 7th term of the Japanese Antarctic Research Expedition Plans. Field studies were done from 22 December 2007 to 13 February 2008, while our Ice Breaker Shirase stayed at/near off Syowa Station. To clarify the relationships among seasonal changes of environmental factors and biological responses, frequent field observations were performed at Naga Ike, one of the freshwater lakes in the Skarvsnes ice-free area. General limnological and biological samplings at the other lakes in the area (14 lakes near the Kizahasi Beach field base camp) were also done during the term. Observations and samplings distant from the base camp, four lakes in eastern Skarvsnes, a lake in Skallen, and three lakes in Langhovde, were also done using a helicopter for transportation. From Namazu Ike (temporary name) in eastern Skarvsnes, submersible video cameras were retrieved and so-called `algal crest', benthic moss-algal assemblages, were sampled by scuba diving. Benthic copepods were sampled quantitatively from Nurume Ike in Langhovde. From Hyoga Ike (temporary name), a snow-dammed glacial lake which lost its water by recent breakage (during the JARE-46 wintering period), thin bio-film samples were collected from the present lake shore formerly part of the lake bed.

In the middle of November in 2006, the author visited the McMurdo station and Dry Valley region, as an Exchange Scientist. During the field research of terrestrial ecosystem, the training system of U.S. Antarctic Program was also surveyed. Field survival camp and environmental education course were well organized and worth introducing into Japanese Antarctic Research.

Biological surveys of lake bottom vegetation, sediments structure, and photosynthesis activity by SCUBA diving were performed in January 2004, in two lakes in the Skarvsnes area, about 50km south of Syowa Station. Two biologists of the 45th Japanese Antarctic Research Expedition (JARE) participated in the surveys. B-4 Ike (tentative name) is a freshwater lake with only 3m depth maximum. Structures of moss pillars and algal mats were investigated in detail, and photosynthesis activity was measured on-site. Namazu Ike (tentative name) is a freshwater lake with 20m depth maximum. Mosses were found from 3m to the deepest bottom of the lake. A crest-like vegetation structure was found at the bottom deeper than 10m.

As part of the RiSCC (Regional Sensitivity to Climate Change in Antarctic Terrestrial and Limnetic Ecosystems) program sponsored by the Scientific Committee on Antarctic Research (SCAR), the REGAL (Research on Ecology and Geohistory on Antarctic Lakes) project has been conducted around the Syowa Station area. The first period of the program (REGAL-I) was promoted from 1994/1995 to 2000/2001, aiming to understand the structure and history of the Antarctic lake ecosystem. The progress of REGAL-I is summarized preparatory to drawing up the plan of REGAL-II from 2003/2004 to 2004/2005.

International co-operative Antarctic research between Japan and Italy started in 1998. The senior author, Masao Iwai, participated as a marine geologist on the R/V Italica's cruise ANTA98/99, the 14th Italian Antarctic Research Expedition, in the Ross Sea from January 5 through February 23, 1999. The scientific and administrative operation of onboard coring and laboratory equipment of R/V Italica, and the life style, are introduced.

Distribution, species composition and possibilities of germination of the propagule bank of moss in Antarctic soils were investigated by cultural experiments. Soil samples were collected in the Yukidori Valley, Antarctica, and were incubated under laboratory conditions. After several weeks, fungi, algae and mosses were observed in the samples. The dominant species within the germinated moss shoots was Bryum pseudotriquetrum s. lat.; in fact, this species was the only moss species at the study site. Shoots of Ceratodon purpureus and Pottia heimii appeared rarely in the general area, but were not observed at all at the study site. Vegetative propagules such as fragments of shoots were frequently found in the soils, most of the germinated shoots sprouted from such propagules. These propagules were thought to be important for colonization at the site. A relatively large number of shoots of B. pseudotriquetrum appeared from the samples collected from the ground near the vegetation. Most of the propagules of B. pseudotriquetrum in the soils were thought to be dispersed from nearly communities, whereas some propagules such as C. purpureus and P. heimii were thought to be dispersed from more distant moss communities.

Tropical regions are not well represented in glacier biology, yet many tropical glaciers are under threat of disappearance due to climate change. Here we report a novel biogenic aggregation at the terminus of a glacier in the Rwenzori Mountains, Uganda. The material was formed by uniseriate protonemal moss gemmae and protonema. Molecular analysis of five genetic markers determined the taxon as Ceratodon purpureus, a cosmopolitan species that is widespread in tropical to polar region. Given optimal growing temperatures of isolate is 20-30 °C, the cold glacier surface might seem unsuitable for this species. However, the cluster of protonema growth reached approximately 10 °C in daytime, suggesting that diurnal increase in temperature may contribute to the moss's ability to inhabit the glacier surface. The aggregation is also a habitat for microorganisms, and the disappearance of this glacier will lead to the loss of this unique ecosystem.

From December 4,1998 to January 15,1999,the author stayed at Baia Terra Nova Station (Italy) in Antarctica, as an exchange scientist. To compare the biodiversity between Syowa Station and the Baia Terra Nova Station area, many samples of mosses, lichens, algae and micro animals in the soil were collected, and the structure of moss vegetation was studied in various fields around the station. Some characteristic features of logistics at the station were also researched.

We isolated a species of tardigrade from moss samples collected from Langhovde and Skarvsnes, near Syowa station, East Antarctic, from which we cultured a parthenogenetic strain in Petri dishes with co-occurring cyanobacteria or green algae. This culture was maintained at both 4 and 10ºC, though the latter proved more suitable for growth. Eggs were laid free, rather than in exuviae. We isolated the 18S rRNA sequences from this tardigrade, identical to that of Acutuncus antarcticus from King George island, South Shetland islands. Morphological analyses via both light and scanning electron microscopy also show general agreement with characteristics of A. antarcticus: dorsal and ventral apophyses for the insertion of stylet muscles and dorsal longitudinal thickening on the anterior part of buccal tube; presence of pharyngeal apophyses, two macroplacoids and absence of a microplacoid; the surface structure of egg; and claw shape. Peribuccal lamellae were absent, but six oval swellings surrounded the mouth opening. An additional study of moss pillars from lake Hotoke-ike, Skarvsnes, proved the existence of the same tardigrade taxon living at the bottom of the lake.

Pollen taxon in sediment samples can be identified by analyzing pollen morphology. Identification of related species based on pollen morphology is difficult and is limited primarily to genus or family. Because pollen grains of various ages are preserved at below 0 °C in glaciers and thus are more likely to remain intact or to suffer little DNA fragmentation, genetic information from such pollen grains should enable identification of plant taxa below the genus level. However, no published studies have attempted detailed identification using DNA sequences obtained from pollen found in glaciers. As a preliminary step, this study attempted to analyze the DNA of Pinus pollen grains extracted from surface snow collected from the Belukha glacier in the Altai Mountains of Russia in the summer of 2003. A 150-bp rpoB fragment from the chloroplast genome in each Pinus pollen grain was amplified by polymerase chain reaction, and DNA products were sequenced to identify them at the section level. A total of 105 pollen grains were used for the test, and sequences were obtained from eight grains. From the sequences obtained, the pollen grains were identified as belonging to the section Quinquefoliae . Trees of the extant species Pinus sibirica in the section Quinquefoliae are currently found surrounding the glacier. The consistency of results for this section suggests that the pollen in the glacier originated from the same Pinus trees as those found in the immediate surroundings.
