Abstract

Purpose/Background: Prostate-specific membrane antigen (PSMA) radioligand therapy has emerged as a promising treatment for advanced prostate cancer. Prior to the administration of radioligand therapy, precise assessment of PSMA expression and tumor localization is essential. Technetium-99m ( 99mTc) imaging is cost-effective and widely available. This study presents the first experiences of using [99mTc]TcO-ABX474 in patients with advanced prostate cancer, with the aim of providing valuable insights into its clinical utility and feasibility. Methods: Patients with advanced prostate cancer, who were scheduled for PSMA radioligand therapy, were referred to PSMA-Imaging. All patients underwent whole-body 99m Tc-PSMA ligand single-photon emission computed tomography/computed tomography (SPECT/CT) imaging. Qualitative and semi-quantitative assessments of [99mTc]TcO-ABX474 uptake were performed, and the results were compared with 18F-PSMA-1007 PET/CT scans. Furthermore, ratios of at least 5 tumor lesions and salivary glands, spleen and kidneys to blood pool, muscle and liver were calculated for each patient. PSMA tumor uptake for evaluation of radioligand therapy was assessed using PROMISE V2 criteria. Results: In total 10 patients received a [99mTc]TcO-ABX474 SPECT/CT imaging prior to PSMA targeted radioligand therapy. Dynamic imaging at 3 time points were performed in 4 patients. In 6 patients image acquisition was performed starting 60 minutes after injection. There was a washout of activity within normal tissue (blood pool, liver and salivary glands) from 10 to 60 and 180 Minutes after injection. The ratios of tumor lesions and the kidneys to normal tissue uptake increased over the same period, whereas the ratios of the slivary glands and spleen decreased. According to PROMISE V2 85, 10 and 5% of the measured lesions showed a PSMA Score of 3, 2 and 1, therefore 95% were eligible for PSMA radioligand therapy. Abstract

We report on room temperature low frequency noise due to magnetic inhomogeneities/domain walls (MI/DWs) in elliptic submicron FeCoB/MgO/FeCoB magnetic tunnel junctions with an area between 0.0245 and 0.0675{\mu}m2. In the smaller area junctions we found an unexpected random telegraph noise (RTN1), deeply in the parallel state, possibly due to stray field induced MI/DWs in the hard layer. The second noise source (RTN2) is observed in the antiparallel state for the largest junctions. Strong asymmetry of RTN2 and of related resistance steps with current indicate spin torque acting on the MI/DWs in the soft layer at current densities below 5x10^5 A/cm2., Comment: 12 pages, 4 figures

Abstract

Magnetic tunnel junctions for spin-transfer torque switching were prepared to investigate the dielectric breakdown. The breakdown occurs typically at voltages not much higher than the switching voltages, a bottleneck for the implementation of spin-transfer torque Magnetic Random Access Memory. Intact and broken tunnel junctions are characterized by transport measurements and then prepared for transmission electron microscopy and energy dispersive x-ray spectrometry by focussed ion beam. The comparison to our previous model of the electric breakdown for thicker MgO tunnel barriers reveals significant differences arising from the high current densities.

Abstract

MgO based magnetic tunnel junctions are prepared to investigate the dielectric breakdown of the tunnel barrier. The breakdown is directly visualized by transmission electron microscopy measurements. The broken tunnel junctions are prepared for the microscopy measurements by focussed ion beam out of the junctions characterized by transport investigations. Consequently, a direct comparison of transport behavior and structure of the intact and broken junctions is obtained. Compared to earlier findings in Alumina based junctions, the MgO barrier shows much more microscopic pinholes after breakdown. This can be explained within a simple model assuming a relationship between the current density at the breakdown and the rate of pinhole formation.

Abstract

Introduction: Dysregulated activity of matrix metalloproteinases (MMPs) drives a variety of pathophysiological conditions. Non-invasive imaging of MMP activity in vivo promises diagnostic and prognostic value. However, current targeting strategies by small molecules are typically limited with respect to the bioavailability of the labeled MMP binders in vivo. To this end, we here introduce and compare three chemical modifications of a recently developed barbiturate-based radiotracer with respect to bioavailability and potential to image MMP activity in vivo. Methods: Barbiturate-based MMP inhibitors with an identical targeting unit but varying hydrophilicity were synthesized, labeled with technetium-99m, and evaluated in vitro and in vivo. Biodistribution and radiotracer elimination were determined in C57/BL6 mice by serial SPECT imaging. MMP activity was imaged in a MMP-positive subcutaneous xenograft model of human K1 papillary thyroid tumors. In vivo data were validated by scintillation counting, autoradiography, and MMP immunohistochemistry. Results: We prepared three new 99mTc-labeled MMP inhibitors, bearing either a glycine ([99mTc]MEA39), lysine ([99mTc]MEA61), or the ligand HYNIC with the ionic co-ligand TPPTS ([99mTc]MEA223) yielding gradually increasing hydrophilicity. [99mTc]MEA39 and [99mTc]MEA61 were rapidly eliminated via hepatobiliary pathways. In contrast, [99mTc]MEA223 showed delayed in vivo clearance and primary renal elimination. In a thyroid tumor xenograft model, only [99mTc]MEA223 exhibited a high tumor-to-blood ratio that could easily be delineated in SPECT images. Conclusion: Introduction of HYNIC/TPPTS into the barbiturate lead structure ([99mTc]MEA223) results in delayed renal elimination and allows non-invasive MMP imaging with high signal-to-noise ratios in a papillary thyroid tumor xenograft model.

Abstract

The purpose of this study was to identify previous treatments and biomarker profile features that prognosticate overall survival (OS) in patients with mCRPC receiving 177Lu-PSMA-617. METHODS: 109 mCRPC patients treated with a median of 3 cycles of 177Lu-PSMA-617 were included. Data were analyzed according to OS as well as PSA response patterns with regard to prior therapies, laboratory biomarkers and metastatic extent in univariate as well as multivariate Cox's proportional hazards models. PSA decline was assessed using the lowest PSA levels after the first cycle of therapy (initial PSA response) and during the entire observation period (best PSA response). RESULTS: In total, 54 patients (49.5%) died during the observation period. First and second line chemotherapy were performed in 85% and 26%, and Abiraterone and Enzalutamide were administered in 83% and 85%, respectively. Any initial PSA decline occurred in 55% while 25% showed a PSA decline of ≥50%. The median estimated OS was 9.9 months (95% CI: 7.2-12.5) for all patients. Any initial decline of PSA was associated with significantly prolonged OS (15.5 vs. 5.7 months, p = 0.002).
