The Arctic marine ecosystem is changing fast due to climate change, emphasizing the need for solid ecological baselines and monitoring. The polar cod Boreogadus saida functions as a key species in the Arctic marine food web. We investigated the stomach contents of polar cod from the northern Barents Sea using DNA metabarcoding with the mitochondrial cytochrome c oxidase I gene in parallel with classical visual analysis. Arctic amphipods and krill dominated the diet in both methods. Yet, metabarcoding allowed for the identification of digested and unidentifiable prey and provided higher taxonomic resolution, revealing new and undiscovered prey items of polar cod in the area. Furthermore, molecular results suggest a higher importance of barnacles and fish (presumably eggs and pelagic larvae) in the diet than previously recorded. Parasites and, in 6 cases, other prey items were only visually identified, demonstrating the complementary nature of both approaches. The presence of temperate and boreal prey species such as northern krill and (early life stages of) European flounder and European plaice illustrates the advection of boreal taxa into the polar region or may be indicative of ongoing borealisation in the Barents Sea. We show that a combination of visual analysis and metabarcoding provides complementary and semi-quantitative dietary information and integrative insights to monitor changing marine food webs

The Svalbard region faces drastic environmental changes, including sea-ice loss and “Atlantification” of Arctic waters, caused primarily by climate warming. These changes result in shifts in the sea-iceassociated (sympagic) community structure, with consequences for the sympagic food web and carbon cycling. To evaluate the role of sympagic biota as a source, sink, and transmitter of carbon, we sampled pack ice and under-ice water (0–2 m) north of Svalbard in spring 2015 by sea-ice coring and under-ice trawling. We estimated biomass and primary production of ice algae and under-ice phytoplankton as well as biomass, carbon demand, and secondary production of sea-ice meiofauna (>10 mm) and under-ice fauna (>300 mm). Sea-ice meiofauna biomass (0.1–2.8 mg C m–2) was dominated by harpacticoid copepods (92%), nauplii (4%), and Ciliophora (3%). Under-ice fauna biomass (3.2–62.7 mg C m–2) was dominated by Calanus copepods (54%). Appendicularia contributed 23% through their high abundance at one station. Herbivorous sympagic fauna dominated the carbon demand across the study area, estimated at 2 mg C m–2 day–1 for ice algae and 4 mg C m–2 day–1 for phytoplankton. This demand was covered by the mean primary production of ice algae (11 mg C m–2 day–1) and phytoplankton (30 mg C m–2 day–1). Hence, potentially 35 mg C m–2 day–1 of algal material could sink from the sympagic realm to deeper layers.The demand of carnivorous under-ice fauna (0.3 mg C m–2 day–1) was barely covered by sympagic secondary production (0.3 mg C m–2 day–1). Our study emphasizes the importance of under-ice fauna for the carbon flux from sea ice to pelagic and benthic habitats and provides a baseline for future comparisons in the context of climate change

Antarctic krill Euphausia superba is an ecological key species in the Southern Ocean and a major fisheries resource. The winter survival of age class 0 (AC0) krill is susceptible to changes in the sea-ice environment due to their association with sea ice and their need to feed during their first winter. However, our understanding of their overwintering diet and its variability is limited. We studied the spatio-temporal variability of the diet in 4 cohorts of AC0 krill in theNorthern Weddell Sea during late winter 2013 using stomach contents, fatty acids (FAs) and bulk stable isotope analysis (BSIA). Stomach contents were dominated by diatoms in numbers and occasionally contained large volumes of copepods. Many of the prey species found in the stomachs were sea ice-associated. Our results show that the diet of overwintering AC0 krill varies significantly in space and time. Variability in stomach content composition was related to environmental factors, including chlorophyll a concentration, copepod abundance and sea-ice cover. In contrast, FA composition mainly varied between cohorts, indicating variation in the long-term diet. The condition of the AC0 krill was reflected in FA and BSIA analysis, suggesting that the availability of sea ice-derived food sources over a long period may impact the condition of developingAC0 krill significantly. The spatio-temporal availability of sea-ice resources is a potentially important factor for AC0 krill winter survival.

The condition and survival of Antarctic krill (Euphausia superba) strongly depends on sea ice conditions during winter. How krill utilize sea ice depends on several factors such as region and developmental stage. A comprehensive understanding of sea ice habitat use by krill, however, remains largely unknown. The aim of this study was to improve the understanding of the krill’s interaction with the sea ice habitat during winter/early spring by conducting large-scale sampling of the ice–water interface (0–2 m) and comparing the size and developmental stage composition of krill with the pelagic population (0–500 m). The ubiquitous marine ciliate Uronema marinum is mainly bacterivorous. It was therefore surprising that in a ciliate-contaminated experiment the growth rate of the phytoplankton species Emiliania huxleyi was significantly reduced. As U. marinum does not ingest E. huxleyi cells, their growth inhibition was probably caused by a toxin secreted by the ciliate, presumably a novel type of chemical interaction between ciliates and phytoplankton. A possible function of toxin secretion is to lyse algal cells that are too large for U. marinum to ingest, to increase dissolved organic matter (DOM) concentrations and hence the growth of heterotrophic bacteria, the main food source of U. marinum. To test this hypothesis U. marinum or the filtrate of U. marinum cultures was added to cultures of phytoplankton with different cell sizes. The presence of U. marinum or the filtrate of U. marinum cultures showed an inhibiting growth effect and a negative effect on the physiology of all species tested although both effects were variable between species. Diatoms appeared less sensitive than non-diatom species. U. marinum acclimatization to phytoplankton led to stronger inhibiting effects, presumably from increased toxin production. Bacterial DGGE analysis of U. marinum cultures did not reveal known toxic bacteria that might account for the observed negative effects on the phytoplankton. Bacterial growth rates in an E. huxleyi culture increased when U. marinum filtrate had been added. In mixed cultures of bacteria, E. huxleyi and U. marinum, bacterial abundance first increased, then decreased due to ciliate predation. These findings support the hypothesis that toxin secretion by U. marinum increases non-prey phytoplankton-derived DOM and stimulates the growth of the bacterial prey.
