Aims. We computed a celestial reference frame (CRF) from Very Long Baseline Interferometry (VLBI) Global Observing System (VGOS) data after five years of regular observations carried out under the umbrella of the International VLBI Service for Geodesy and Astrometry. In this paper we evaluate its strengths and weaknesses, document the source selection and scheduling strategies for the individual sessions, and investigate the effect of using this new VGOS CRF in the analysis of individual geodetic VLBI sessions where the radio source positions are fixed to their a priori coordinates. Methods. We estimated the VIE2023-VG CRF in a global adjustment of 155 multi-baseline 24-hour VGOS sessions until 2024.0. We carried out several comparisons with the third version of the International Celestial Reference Frame (ICRF3) adopted by the International Astronomical Union in 2018, and with VIE2023sx CRF which includes VLBI S/X data until 2024.0. Furthermore, we studied the effect of more frequent estimations of tropospheric parameters (30,10, and 5 min for zenith wet delay) on the estimated CRF in the current VGOS network. We evaluated the VIE2023-VG CRF in the geodetic analysis of VGOS sessions by computing the baseline lengths and station positions and statistics on the Earth orientation parameters estimated in the single-session analysis where the source positions were fixed to either the VIE2023-VG CRF or to ICRF3-SX. Results. The current VIE2023-VG CRF is built with 1.39 million VGOS group delays and includes 418 radio sources, where 172 sources (41%) are introduced in only four research and development sessions alone. We show that the VIE2023-VG CRF has excellent source position precision. The median formal error from the least-squares adjustment is 30 μas for right ascension (scaled by cosine of declination) and 47 μas for declination. In terms of systematic distortions versus ICRF3-SX, the largest terms in the vector spherical harmonics up to the degree and order two, reach in absolute values around 60 μas, caused by correlations between the individual terms. Because of the lack of observations in the southern hemisphere, a constraint for a zero slope in declination difference with respect to ICRF3-SX is imposed in the global adjustment. Therefore, VGOS should prioritize the development of southern stations in order to limit the need for such constraints on the frame. Further we show that fixing the a priori CRF to VIE2023-VG CRF instead of the ICRF3-SX in the single-session analysis improves the weighted root mean square of the baseline length by up to 3 mm, especially for the extremely long baselines (>12 000 km), with a weighted mean difference between the baseline length time series up to 2 mm. Therefore, in order to meet the ambitious goal of 1 mm accuracy for a terrestrial reference frame set by the Global Geodetic Observing System, the development of a VGOS-only CRF is required for use in the geodetic analysis of the new generation VGOS data. Methodological advances in neuroscience have enabled the collection of massive datasets which demand innovative approaches for scientific communication. Existing platforms for data storage lack intuitive tools for data exploration, limiting our ability to interact effectively with these brain-wide datasets. We introduce two public websites: (Data and Atlas) developed for the International Brain Laboratory which provide access to millions of behavioral trials and hundreds of thousands of individual neurons. These interfaces allow users to discover both the raw and processed brain-wide data released by the IBL at the scale of the whole brain, individual sessions, trials, and neurons. By hosting these data interfaces as websites they are available cross-platform with no installation. By releasing each site's code as a modular open-source framework, other researchers can easily develop their own web interfaces and explore their own data. As neuroscience datasets continue to expand, customizable web interfaces offer a glimpse into a future of streamlined data exploration and act as blueprints for future tools.

Organ quality can be assessed prior to transplantation, during normothermic machine perfusion (NMP) of the liver. Evaluation of mitochondrial function by high-resolution respirometry (HRR) may serve as a viability assessment concept in this setting. Freshly collected tissue is considered as optimal sample for HRR, but due to technical and personnel requirements, more flexible and schedulable measurements are needed. However, the impact of cold storage following NMP before processing biopsy samples for mitochondrial analysis remains unknown. We aimed at establishing an appropriate storage protocol of liver biopsies for HRR. Wedge biopsies of 5 human livers during NMP were obtained and assessed by HRR. Analysis was performed after 0, 4, 8, and 12 h of hypothermic storage (HTS) in HTK organ preservation solution at 4°C. With HTS up to 4 h, mitochondrial performance did not decrease in HTS samples compared with 0 h (OXPHOS, 44.62 [34.75-60.15] pmol·s -1 ·mg wet mass -1 vs . 43.73 [40.69-57.71], median [IQR], p > 0.999). However, at HTS beyond 4 h, mitochondrial respiration decreased. Contemporary pose estimation methods enable precise measurements of behavior via supervised deep learning with hand-labeled video frames. Although effective in many cases, the supervised approach requires extensive labeling and often produces outputs that are unreliable for downstream analyses. Here, we introduce "Lightning Pose," an efficient pose estimation package with three algorithmic contributions. First, in addition to training on a few labeled video frames, we use many unlabeled videos and penalize the network whenever its predictions violate motion continuity, multiple-view geometry, and posture plausibility (semi-supervised learning). Second, we introduce a network architecture that resolves occlusions by predicting pose on any given frame using surrounding unlabeled frames. Third, we refine the pose predictions post-hoc by combining ensembling and Kalman smoothing. Together, these components render pose trajectories more accurate and scientifically usable. Donor organ biomarkers with sufficient predictive value in liver transplantation (LT) are lacking. We herein evaluate liver viability and mitochondrial bioenergetics for their predictive capacity towards the outcome in LT. We enrolled 43 consecutive patients undergoing LT. Liver biopsy samples taken upon arrival after static cold storage were assessed by histology, real-time confocal imaging analysis (RTCA), and high-resolution respirometry (HRR) for mitochondrial respiration of tissue homogenates. Early allograft dysfunction (EAD) served as primary endpoint. HRR data were analysed with a focus on the efficacy of ATP production or P - L control efficiency, calculated as 1- L / P from the capacity of oxidative phosphorylation P and non-phosphorylating respiration L . Twenty-two recipients experienced EAD. Pre-transplant histology was not predictive of EAD. The mean RTCA score was significantly lower in the EAD cohort (-0.75 ± 2.27) compared to the IF cohort (0.70 ± 2.08; p = 0.01), indicating decreased cell viability. P - L control efficiency was predictive of EAD (0.76 ± 0.06 in IF vs. 0.70 ± 0.08 in EAD-livers; p = 0.02) and correlated with the RTCA score. Both RTCA and P - L control efficiency in biopsy samples taken during cold storage have predictive capacity towards the outcome in LT. Connecting chemical properties to various wine characteristics is of great interest to the science of olfaction as well as the wine industry. We explored whether Bordeaux wine chemical identities and vintages (harvest year) can be inferred from a common and affordable chemical analysis, namely, a combination of gas chromatography (GC) and electron ionization mass spectrometry. Using 12 vintages (within the 1990-2007 range) from 7 estates of the Bordeaux region, we report that, remarkably, nonlinear dimensionality reduction techniques applied to raw gas chromatograms recover the geography of the Bordeaux region. Using machine learning, we found that we can not only recover the estate perfectly from gas chromatograms, but also the vintage with up to 50% accuracy. Interestingly, we observed that the entire chromatogram is informative with respect to geographic location and age, thus suggesting that the chemical identity of a wine is not defined by just a few molecules but is distributed over a large chemical spectrum. Background: Reliable biomarkers for organ quality assessment during normothermic machine perfusion (NMP) are desired. ATP (adenosine triphosphate) production by oxidative phosphorylation plays a crucial role in the bioenergetic homeostasis of the liver. Thus, detailed analysis of the aerobic mitochondrial performance may serve as predictive tool towards the outcome after liver transplantation., Methods: In a prospective clinical trial, 50 livers were subjected to NMP (OrganOx Metra) for up to 24.ßh. Biopsy and perfusate samples were collected at the end of cold storage, at 1.ßh, 6.ßh, end of NMP, and 1.ßh after reperfusion. Mitochondrial function and integrity were characterized by high-resolution respirometry (HRR), AMP, ADP, ATP and glutamate dehydrogenase analysis and correlated with the clinical outcome (L-GrAFT score). Real-time confocal microscopy was performed to assess tissue viability. Structural damage was investigated by histology, immunohistochemistry and transmission electron microscopy., Findings: A considerable variability in tissue viability and mitochondrial respiration between individual livers at the end of cold storage was observed. During NMP, mitochondrial respiration with succinate and tissue viability remained stable. In the multivariate analysis of the 35 transplanted livers (15 were discarded), area under the curve (AUC) of LEAK respiration, cytochrome c control efficiency (mitochondrial outer membrane damage), and efficacy of the mitochondrial ATP production during the first 6.ßh of NMP correlated with L-GrAFT., Interpretations: Bioenergetic competence during NMP plays a pivotal role in addition to tissue injury markers. The deviation of Universal Time from atomic time, expressed as UT1-UTC, reflects the irregularities of the Earth rotation speed and is key to precise geodetic applications which depend on the transformation between celestial and terrestrial reference frames. A rapidly varying quantity such as UT1-UTC demands observation scenarios enabling fast delivery of good results. These criteria are currently met only by the Very Long Baseline Interferometry (VLBI) Intensive sessions. Due to stringent requirements of a fast UT1-UTC turnaround, the observations are limited to a few baselines and a duration of one hour. Hence, the estimation of UT1-UTC from Intensives is liable to constraints and prone to errors introduced by inaccurate a priori information. One aspect in this context is that the regularly operated Intensive VLBI sessions organised by the International VLBI Service for Geodesy and Astrometry solely use stations in the northern hemisphere. Any potential systematic errors due to this northern hemisphere dominated geometry are so far unknown. Besides the general need for stimulating global geodetic measurements with southern observatories, this served as a powerful motivation to launch the SI (Southern Intensive) program in 2020. The SI sessions are observed using three VLBI antennas in the southern hemisphere: Ht (South Africa), Hb (Tasmania) and Yg (Western Australia). On the basis of UT1-UTC results from 53 sessions observed throughout 2020 and 2021, we demonstrate the competitiveness of the SI with routinely operated Intensive sessions in terms of operations and UT1-UTC accuracy. The UT1-UTC values of the SI reach an average agreement of 32 µs in terms of weighted standard deviation when compared with the conventional Intensives results of five independent analysis centers and of 27 µs compared with the 14C04 series. The mean scatter of all solutions of the considered northern hemisphere Intensives with respect to C04 is at a comparable level of 29 µs. The quality of the results is only slightly degraded if just the baseline HtHb is evaluated. Global geodetic VLBI is upgrading to its next-generation observing system, VGOS. This upgrade has turned out to be a process over multiple years, until VGOS reaches its full capabilities with the envisaged continuous observations. Until then, for the Australian stations, the upgrade means ceasing their legacy S/X observations, leaving a large gap in the global network as well as in the station time series. The Australian mixed-mode observing program is a series of sessions where the VGOS stations in Hobart and Katherine observe legacy S/X VLBI together with other stations in the region. This paper describes the technical details of these observations and their processing strategies and discusses their suitability for geodetic results by comparison with those of standard legacy S/X sessions. The presented mixed-mode sessions allow a continuation of the station time series, a benefit for the stations themselves as well as for future realisations of the terrestrial and celestial reference frames. A novel mode of observing is introduced and tested. The results are promising and it is suggested for acceptance into standard legacy S/X IVS observations, overcoming current gaps in the network due to VGOS upgrades and preventing a worsening of global results otherwise., (© The Author(s) 2022.)

Recent neuroscience studies demonstrate that a deeper understanding of brain function requires a deeper understanding of behavior. Detailed behavioral measurements are now often collected using video cameras, resulting in an increased need for computer vision algorithms that extract useful information from video data. Here we introduce a new video analysis tool that combines the output of supervised pose estimation algorithms (e.g. DeepLabCut) with unsupervised dimensionality reduction methods to produce interpretable, low-dimensional representations of behavioral videos that extract more information than pose estimates alone. We demonstrate this tool by extracting interpretable behavioral features from videos of three different head-fixed mouse preparations, as well as a freely moving mouse in an open field arena, and show how these interpretable features can facilitate downstream behavioral and neural analyses. We also show how the behavioral features produced by our model improve the precision and interpretation of these downstream analyses compared to using the outputs of either fully supervised or fully unsupervised methods alone., Competing Interests: The authors have declared that no competing interests exist.

One of the main tasks of Very Long Baseline Interferometry (VLBI) is the rapid determination of the highly variable Earth's rotation expressed through the difference between Universal Time UT1 and Coordinated Universal Time UTC (dUT1). For this reason, dedicated one hour, single baseline sessions, called "Intensives" , are observed on a daily basis. Thus far, the optimal geometry of Intensive sessions was understood to include a long east-west extension of the baseline to ensure a dUT1 estimation with highest accuracy. In this publication, we prove that long east-west baselines are the best choice only for certain lengths and orientations. In this respect, optimal orientations may even require significant inclination of the baseline with respect to the equatorial plane. The basis of these findings is a simulation study with subsequent investigations in the partial derivatives of the observed group delays τ with respect to dUT1 ∂ τ / ∂ d U T 1 . Almost 3000 baselines between artificial stations located on a regular 10 × 10 degree grid are investigated to derive a global and generally valid picture about the best length and orientation of Intensive baselines. Our results reveal that especially equatorial baselines or baselines with a center close to the equatorial plane are not suited for Intensives although they provide a good east-west extension. This is explained by the narrow right ascension band of visible sources and the resulting lack of variety in the partial derivatives. Moreover, it is shown that north-south baselines are also capable of determining dUT1 with reasonable accuracy, given that the baseline orientation is significantly different from the Earth rotation axis. However, great care must be taken to provide accurate polar motion a priori information for these baselines. Very long baseline interferometry (VLBI) scheduling is a challenging optimization problem. With the development of the new VLBI global observing system (VGOS) consisting of smaller but very fast slewing antennas, new opportunities arise. In this work, we give a deep insight into optimized VGOS scheduling using a newly developed VLBI scheduling software called VieSched++, and we show how different scheduling parameters and approaches affect the precision of geodetic results. Therefore, the results of over one thousand generated schedules and over one million simulated sessions are analyzed. The simulations reveal that the most important parameters to optimize VGOS schedules with VieSched++ are the so-called weight factors. A proper selection of individually optimized weight factors can improve the quality of a schedule significantly. It is shown that the values of the weight factors used to generate the schedule are highly correlated with the expected precision of the geodetic parameters. We highlight the benefit of selecting schedules based on large-scale Monte Carlo simulations and show why scheduling statistics like the number of observations or the sky-coverage are not necessarily the best metric to evaluate schedules., (© The Author(s) 2020.)

To support monitoring subtle effects in the Earth system such as a mean sea level rise of 3 mm/year, a next-generation VLBI system, the VLBI Global Observing System (VGOS), has been developed and a new VGOS station network is being built. However, the geometry of the current VGOS network and its planned extension suffer from a lack of stations in the southern hemisphere. In this investigation, we identify optimal locations for additional VGOS radio telescopes with a new method based on bulk observing schedule generation and subsequent large-scale Monte-Carlo simulations. The location of the additional station is varied over 477 possible locations, homogeneously distributed over land areas on the globe. For each antenna location, several schedules have been generated and simulated to minimize the effects of scheduling and the randomness of simulations. Thereby, it is possible to judge, in which regions an additional VGOS station would have the biggest impact on the precision of the estimated geodetic parameters, in our case assessed by the repeatabilities of the estimated Earth orientation parameters (EOPs). To generate highly optimized schedules and to remove effects due to non-optimized scheduling, a total of 93 thousand schedules were iteratively generated, investigating over 300 billion scans and 2.4 trillion observations. Each schedule was further simulated 1000 times, leading to over 5 trillion simulated and analyzed observations. Although the optimum location of a future VLBI station depends on the EOP of interest and the geometry of the existing network, it is shown that the more the VGOS network grows, the more the lack of southern stations becomes prominent. The best location for an additional VGOS station for most EOP components and especially in the case of future VGOS networks would be the southern part of South America. It is further shown that the location of the additional antenna highly determines the expectable precision of the EOP estimates. For a 6-station network, the location of an additional seventh antenna can improve the precision of the EOP by a factor of 2.4 to 3.8. For an 18-station network, the location of an additional 19th station still improves the repeatability by a factor of 1.6. Studying transitions in and out of the altered state of consciousness caused by intravenous (IV) N,N-Dimethyltryptamine (DMT - a fast-acting tryptamine psychedelic) offers a safe and powerful means of advancing knowledge on the neurobiology of conscious states. Here we sought to investigate the effects of IV DMT on the power spectrum and signal diversity of human brain activity (6 female, 7 male) recorded via multivariate EEG, and plot relationships between subjective experience, brain activity and drug plasma concentrations across time. Compared with placebo, DMT markedly reduced oscillatory power in the alpha and beta bands and robustly increased spontaneous signal diversity. Time-referenced and neurophenomenological analyses revealed close relationships between changes in various aspects of subjective experience and changes in brain activity. Importantly, the emergence of oscillatory activity within the delta and theta frequency bands was found to correlate with the peak of the experience - particularly its eyes-closed visual component. These findings highlight marked changes in oscillatory activity and signal diversity with DMT that parallel broad and specific components of the subjective experience, thus advancing our understanding of the neurobiological underpinnings of immersive states of consciousness.

Brain signal diversity constitutes a robust neuronal marker of the global states of consciousness. It has been demonstrated that, in comparison to the resting wakefulness, signal diversity is lower during unconscious states, and higher during psychedelic states. A plausible interpretation of these findings is that the neuronal diversity corresponds to the diversity of subjective conscious experiences. Therefore, in the present study we varied an information rate processed by the subjects and hypothesized that greater information rate will be related to richer and more differentiated phenomenology and, consequently, to greater signal diversity. To test this hypothesis speech recordings (excerpts from an audio-book) were presented to subjects at five different speeds (65, 83, 100, 117, and 135% of the original speed). By increasing or decreasing speed of the recordings we were able to, respectively, increase or decrease the presented information rate. We also included a backward (unintelligible) speech presentation and a resting-state condition (no auditory stimulation). We tested 19 healthy subjects and analyzed the recorded EEG signal (64 channels) in terms of Lempel-Ziv diversity (LZs). We report the following findings. First, our main hypothesis was not confirmed, as Bayes Factor indicates evidence for no effect when comparing LZs among five presentation speeds. Second, we found that LZs during the resting-state was greater than during processing of both meaningful and unintelligible speech. Third, an additional analysis uncovered a gradual decrease of diversity over the time-course of the experiment, which might reflect a decrease in vigilance. We thus speculate that higher signal diversity during the unconstrained resting-state might be due to a greater variety of experiences, involving spontaneous attention switching and mind wandering.

Emerging neural theories of consciousness suggest a correlation between a specific type of neural dynamical complexity and the level of consciousness: When awake and aware, causal interactions between brain regions are both integrated (all regions are to a certain extent connected) and differentiated (there is inhomogeneity and variety in the interactions). In support of this, recent work by Casali et al (2013) has shown that Lempel-Ziv complexity correlates strongly with conscious level, when computed on the EEG response to transcranial magnetic stimulation. Here we investigated complexity of spontaneous high-density EEG data during propofol-induced general anaesthesia. We consider three distinct measures: (i) Lempel-Ziv complexity, which is derived from how compressible the data are; (ii) amplitude coalition entropy, which measures the variability in the constitution of the set of active channels; and (iii) the novel synchrony coalition entropy (SCE), which measures the variability in the constitution of the set of synchronous channels. After some simulations on Kuramoto oscillator models which demonstrate that these measures capture distinct 'flavours' of complexity, we show that there is a robustly measurable decrease in the complexity of spontaneous EEG during general anaesthesia.
