The C-terminal domain of the cellular prion protein (PrP C ) contains two N-linked glycosylation sites, the occupancy of which impacts disease pathology. In this study, we demonstrate that glycans at these sites are required to maintain an intramolecular interaction with the N-terminal domain, mediated through a previously identified copper-histidine tether, which suppresses the neurotoxic activity of PrP C . NMR and electron paramagnetic resonance spectroscopy demonstrate that the glycans refine the structure of the protein's interdomain interaction. Using whole-cell patch-clamp electrophysiology, we further show that cultured cells expressing PrP molecules with mutated glycosylation sites display large, spontaneous inward currents, a correlate of PrP-induced neurotoxicity. Although Alzheimer's disease (AD) was first described over a century ago, it remains the leading cause of age-related dementia. Innumerable changes have been linked to the pathology of AD; however, there remains much discord regarding which might be the initial cause of the disease. The "amyloid cascade hypothesis" proposes that the amyloid β (Aβ) peptide is central to disease pathology, which is supported by elevated Aβ levels in the brain before the development of symptoms and correlations of amyloid burden with cognitive impairment. The "metals hypothesis" proposes a role for metal ions such as iron, copper, and zinc in the pathology of AD, which is supported by the accumulation of these metals within amyloid plaques in the brain. Metals have been shown to induce aggregation of Aβ, and metal ion chelators have been shown to reverse this reaction in vitro . 8-Hydroxyquinoline-based chelators showed early promise as anti-Alzheimer's drugs. Both 5-chloro-7-iodo-8-hydroxyquinoline (CQ) and 5,7-dichloro-2-[(dimethylamino)methyl]-8-hydroxyquinoline (PBT2) underwent unsuccessful clinical trials for the treatment of AD. To gain insight into the mechanism of action of 8HQs, we have investigated the potential interaction of CQ, PBT2, and 5,7-dibromo-8-hydroxyquinoline (B2Q) with Cu(II)-bound Aβ(1-42) using X-ray absorption spectroscopy (XAS), high energy resolution fluorescence detected (HERFD) XAS, and electron paramagnetic resonance (EPR). By XAS, we found CQ and B2Q sequestered ∼83% of the Cu(II) from Aβ(1-42), whereas PBT2 sequestered only ∼59% of the Cu(II) from Aβ(1-42), suggesting that CQ and B2Q have a higher relative Cu(II) affinity than PBT2. From our EPR, it became clear that PBT2 sequestered Cu(II) from a heterogeneous mixture of Cu(II)Aβ(1-42) species in solution, leaving a single Cu(II)Aβ(1-42) species. It follows that the Cu(II) site in this Cu(II)Aβ(1-42) species is inaccessible to PBT2 and may be less solvent-exposed than in other Cu(II)Aβ(1-42) species. We found no evidence to suggest that these 8HQs form ternary complexes with Cu(II)Aβ(1-42).

All versions of this article have been corrected.] Previous research reported ovulatory changes in women's appearance, mate preferences, extra- and in-pair sexual desire, and behavior, but has been criticized for small sample sizes, inappropriate designs, and undisclosed flexibility in analyses. In the present study, we sought to address these criticisms by preregistering our hypotheses and analysis plan and by collecting a large diary sample. We gathered more than 26,000 usable online self-reports in a diary format from 1,054 women, of which 429 were naturally cycling. We inferred the fertile period from menstrual onset reports. We used hormonal contraceptive users as a quasi-control group, as they experience menstruation, but not ovulation. We probed our results for robustness to different approaches (including different fertility estimates, different exclusion criteria, adjusting for potential confounds, moderation by methodological factors). We found robust evidence supporting previously reported ovulatory increases in extra-pair desire and behavior, in-pair desire, and self-perceived desirability, as well as no unexpected associations. Yet, we did not find predicted effects on partner mate retention behavior, clothing choices, or narcissism. Contrary to some of the earlier literature, partners' sexual attractiveness did not moderate the cycle shifts. Taken together, the replicability of the existing literature on ovulatory changes was mixed. We conclude with simulation-based recommendations for reading the past literature and for designing future large-scale preregistered within-subject studies to understand ovulatory cycle changes and the effects of hormonal contraception. The cellular prion protein (PrP C ) comprises two domains: a globular C-terminal domain and an unstructured N-terminal domain. Recently, copper has been observed to drive tertiary contact in PrP C , inducing a neuroprotective cis interaction that structurally links the protein's two domains. The location of this interaction on the C terminus overlaps with the sites of human pathogenic mutations and toxic antibody docking. Combined with recent evidence that the N terminus is a toxic effector regulated by the C terminus, there is an emerging consensus that this cis interaction serves a protective role, and that the disruption of this interaction by misfolded PrP oligomers may be a cause of toxicity in prion disease. We demonstrate here that two highly conserved histidines in the C-terminal domain of PrP C are essential for the protein's cis interaction, which helps to protect against neurotoxicity carried out by its N terminus. We show that simultaneous mutation of these histidines drastically weakens the cis interaction and enhances spontaneous cationic currents in cultured cells, the first C-terminal mutant to do so. Whereas previous studies suggested that Cu 2+ coordination was localized solely to the protein's N-terminal domain, we find that both domains contribute equatorially coordinated histidine residue side-chains, resulting in a novel bridging interaction. We also find that extra N-terminal histidines in pathological familial mutations involving octarepeat expansions inhibit this interaction by sequestering copper from the C terminus. Bacterially expressed proteins used in NMR studies lack glycans, and proteins from other organisms are neither 15 N labeled nor glycosylated homogeneously. Here, we add two artificial glycans to uniformly 15 N labeled prion protein using a buffer system that evolves over a pH range to accommodate the conflicting pH requirements of the substrate and enzymes without the need to fine-tune buffer conditions. NMR and CD spectroscopy of the protein indicates that the glycans do not influence its fold.

Alzheimer's disease (AD) is the main cause of age-related dementia and currently affects approximately 5.7 million Americans. Major brain changes associated with AD pathology include accumulation of amyloid beta (Aβ) protein fragments and formation of extracellular amyloid plaques. Redox-active metals mediate oligomerization of Aβ, and the resultant metal-bound oligomers have been implicated in the putative formation of harmful, reactive species that could contribute to observed oxidative damage. In isolated plaque cores, Cu(II) is bound to Aβ via histidine residues. Despite numerous structural studies of Cu(II) binding to synthetic Aβ in vitro, there is still uncertainty surrounding Cu(II) coordination in Aβ. In this study, we used X-ray absorption spectroscopy (XAS) and high energy resolution fluorescence detected (HERFD) XAS to investigate Cu(II) coordination in Aβ(1-42) under various solution conditions. We found that the average coordination environment in Cu(II)Aβ(1-42) is sensitive to X-ray photoreduction, changes in buffer composition, peptide concentration, and solution pH. Fitting of the extended X-ray absorption fine structure (EXAFS) suggests Cu(II) is bound in a mixture of coordination environments in monomeric Aβ(1-42) under all conditions studied. However, it was evident that on average only a single histidine residue coordinates Cu(II) in monomeric Aβ(1-42) at pH 6.1, in addition to 3 other oxygen or nitrogen ligands. Cu(II) coordination in Aβ(1-42) at pH 7.4 is similarly 4-coordinate with oxygen and nitrogen ligands, although an average of 2 histidine residues appear to coordinate at this pH. At pH 9.0, the average Cu(II) coordination environment in Aβ(1-42) appears to be 5-coordinate with oxygen and nitrogen ligands, including two histidine residues.

This article describes the development of porous two-ply channels for paper-based microfluidic devices that wick fluids significantly faster than conventional, porous, single-ply channels. The two-ply channels were made by stacking two single-ply channels on top of each other and were fabricated entirely out of paper, wax and toner using two commercially available printers, a convection oven and a thermal laminator. The wicking in paper-based channels was studied and modeled using a modified Lucas-Washburn equation to account for the effect of evaporation, and a paper-based titration device incorporating two-ply channels was demonstrated.

We present a new method for fabricating three-dimensional paper-based fluidic devices that uses toner as a thermal adhesive to bond multiple layers of patterned paper together. The fabrication process is rapid, involves minimal equipment (a laser printer and a laminator) and produces complex channel networks with dimensions down to 1 mm. The devices can run multiple diagnostic assays on one or more samples simultaneously, can incorporate positive and negative controls and can be programmed to display the results of the assays in a variety of patterns. The patterns of the results can encode information, which could be used to identify counterfeit devices, identify samples, encrypt the results for patient privacy or monitor patient compliance.

This article introduces fully enclosed microfluidic paper-based analytical devices (microPADs) fabricated by printing toner on the top and bottom of the devices using a laser printer. Enclosing paper-based microfluidic channels protects the channels from contamination, contains and protects reagents stored on the device, contains fluids within the channels so that microPADs can be handled and operated more easily, and reduces evaporation of solutions from the channels. The Mitsunobu reaction was used to attach tetra-O-benzyl-D-glucopyranose to a monoindolylmaleimide, providing a key intermediate in the total synthesis of indolocarbazole topoisomerase I poisons. Using normal-phase silica gel chromatography, purification of the glycosylated product normally required multiple columns, resulting in poor recovered yields. Reversed-phase chromatography was used successfully to purify this highly hydrophobic material, rapidly and in high yield.

The effect of aeration on the development of a defined biofilm consortium of oral bacteria was investigated in a two-stage chemostat system. An inoculum comprising 10 species, including both facultatively anaerobic and obligately anaerobic bacteria, and species associated with oral health and disease, was inoculated into an anaerobic first-stage chemostat vessel. The effluent from this chemostat was linked to an aerated [200 ml CO2/air (5:95, v/v) min-1] second-stage vessel, in which removable hydroxyapatite discs were inserted to allow biofilm formation. Comparisons were made of planktonic and biofilm communities in the aerated second-stage vessel by means of viable counts. Both planktonic and early biofilm communities were dominated by Neisseria subflava, comprising > 40% of total c.f.u. in the fluid phase, and > 80% of c.f.u. in 2 h biofilms. Obligate anaerobes persisted in this mixed culture, and succession in biofilms led them to predominate only after 7 d. Despite the continuous addition of air, the dissolved oxygen tension (dO2) within the culture remained low (< 5% of air saturation), and the redox potential (Eh) was -275 mV. In order to assess the significance of the presence of N. subflava in community development, a subsequent experiment omitted this aerobe from the inoculum, to produce a nine-species culture. The planktonic phase was predominated by three streptococcal species, Prevotella nigrescens and Fusobacterium nucleatum. Biofilms again underwent successional changes, with anaerobes increasing in proportion with time. In contrast to the culture including N. subflava, dO2 was 50-60% of air saturation, and the Eh was +50 mV. In the final experiment, the rate of addition of first-stage culture was reduced to 1/10 of that in the previous experiment, in order to determine whether anaerobes were growing, rather than merely persisting in the aerated culture. The data for the planktonic phase indicated that the anaerobes were growing in aerated (dO2 40-50%, Eh +100 mV) conditions. Once again, anaerobes increased in proportion in older biofilms. The study indicates that mixed cultures can protect obligate anaerobes from the toxic effects of oxygen, both in the biofilm and planktonic modes of growth.

Previously, we developed a chemostat system to study the behaviour and properties of a community of up to 10 species of oral bacteria. The present study describes modification of this system to incorporate removable and replaceable hydroxyapatite (the major mineral in human dental enamel) disks on which biofilms could develop. Hydroxyapatite disks were immersed in the chemostat for known time periods, and the bacterial content of biofilms determined by viable counting. Initial deposition rates were rapid, with all 10 species detected after 1 h, and the numbers of bacteria in biofilms continued to increase for 21 d. The species composition of biofilms reflected that of the surrounding fluid phase, and showed only limited signs of the type of 'species succession' which is observed in developing dental plaque in vivo, although anaerobic species increased in proportion in older biofilms. Four-day biofilms showed the least variability and were chosen as the 'standard biofilm' for more detailed study. Variability in the bacterial composition of 4-d biofilms was comparable both within a single chemostat run and between independent chemostat runs. Glucose pulsing in the absence of pH control resulted in the selection of cariogenic species; the disruption of the biofilm community was less marked than that of the equivalent planktonic culture. The model system has considerable potential in studying the effects of a variety of factors on biofilm development, as well as in comparing the efficacy of antimicrobial systems against biofilms.

Activities of glucosyltransferases (Gtf)-B, -C and -D adsorbed on the surface of saliva-coated hydroxyapatite were compared with those of Gtfs of donor whole saliva. Antiserum raised against a mixture of the three Gtfs reduced the activity of GtfB only, and had no effect on donor Gtf activities. GtfB, and not the Gtfs of the donors, was stimulated in the presence of starch hydrosylates. GtfD and GtfC activities were enhanced on the surfaces, as were the Gtf activities of donor salivas. The activities of GtfD and GtfB, but not GtfC, were stimulated by dextran. The donor Gtf activities were unaffected by dextran. Therefore, Gtf activity in pellicles has properties similar to those of GtfC.

Mutacin II is an antibiotic that is produced by group II Streptococcus mutans. It inhibits the growth of other streptococci as well as many other gram-positive microorganisms by a hitherto unknown mechanism. Mutacin II possess bactericidal activity against susceptible cells. It transiently depolarizes the transmembrane electrical potential (delta psi) and the transmembrane pH gradient (delta pH) and partially inhibits amino acid transport. However, it rapidly depletes the intracellular ATP pool in glucose-energized cells and prevents the generation of ATP. It is concluded that mutacin II does not belong to the group of pore-forming antibiotics (type A) or to the type B antibiotics, which inhibit phospholipases or interfere with peptidoglycan biosynthesis. Mutacin II acts by inhibiting essential enzyme functions at the level of metabolic energy generation, an activity that has not yet been classified for antibiotics.

Many researchers have suggested that the role of glucan-mediated interactions in the adherence of Streptococcus mutans is restricted to accumulation of this cariogenic bacterium following its sucrose (i.e., glucan)-independent binding to saliva-coated tooth surfaces. However, the presence of enzymatically active glucosyltransferase in salivary pellicle suggests that glucans could also promote the initial adherence of S. mutans to the teeth. In the present study, the commonly used hydroxyapatite adherence assay was modified to include the incorporation of glucosyltransferase and the synthesis of glucans in situ on saliva-coated hydroxyapatite beads. Several laboratory strains and clinical isolates of S. mutans were examined for their ability to adhere to experimental pellicles, either with or without the prior formation of glucans in situ. Results showed that most strains of S. mutans bound stereospecifically to glucans synthesized in pellicle. Inhibition studies with various polysaccharides and fungal dextranase indicated that alpha 1,6-linked glucose residues were of primary importance in the glucan binding observed. Scanning electron microscopic analysis showed direct binding of S. mutans to hydroxyapatite surface-associated polysaccharide and revealed no evidence of trapping or cell-to-cell binding. S. mutans strains also attached to host-derived structures in experimental pellicles, and the data suggest that the bacterial adhesins which recognize salivary binding sites were distinct from glucan-binding adhesins. Furthermore, glucans formed in experimental pellicles appeared to mask the host-derived components. These results support the concept that glucans synthesized in salivary pellicle can promote the selective adherence of the cariogenic streptococci which colonize human teeth.

This study presents results of a survey of 100 adopted adults who contacted a social service agency or search group. Their perspectives on the experience of adoption and their reasons for agency contact are compared with recommended adoption practice of a generation ago and with reports from the clinical literature.

Perry's scheme of intellectual and ethical development was examined for sex differences with respect to areas of identity concern for which sex differences have been suggested: occupational choice, interpersonal relationships, and sexual identity. The content area of religion and ego development, as measured by Loevinger's sentence completion test (SCT), were also examined. Thirty-one females and 29 males participated. An interview format which applied Perry's unstructured approach to each of the four areas was utilized. Transcribed interviews and SCTs were rated blind. Structural as well as content analyses were performed. Structural analyses revealed no significant sex differences overall or by content area. However, multiple and stepwise regression analyses revealed patterns in which male personality processes consistently focused upon occupational issues while female processes focused upon interpersonal and sexual issues. Ego development was found to be highly correlated with intellectual development for men but unrelated for women. Content analyses suggest that women focus their interpersonal worlds developmentally upon issues of trust while men focus upon issues of rightness. Results are interpreted in the context of recent works by Gilligan concerning sex differences in development.

This study aimed to determine physical and kinetic properties of glucosyltransferase (GTF) adsorbed onto hydroxyapatite (HA) surfaces. For development of a solid-phase enzyme assay, 4.0-mg samples of washed HA powder were exposed to centrifuged whole saliva (WSHA) or buffer, and subsequently exposed to a GTF solution. The activities of GTF adsorbed to HA and that remaining in solution were measured. WSHA was more effective in adsorbing GTF than was naked HA. Enzyme activity on the surface of WSHA was enhanced; more activity was detected on WSHA than was apparently removed from solution. A similar effect was observed when GTF was adsorbed to naked HA from a mixture with lysozyme or saliva; however, no enhancement was seen when GTF was adsorbed from a mixture with albumin. Compared with GTF in solution, adsorbed GTF displayed activity over a much wider range of pH values. Temperature-activity profiles indicated that GTF adsorbed to surfaces had a lower temperature optimum (40 degrees C) than did soluble enzyme (45 degrees C), and that the bound enzyme was more resistant to adverse effects of heat at elevated temperatures. The majority of glucan made by GTF adsorbed to parotid saliva-coated HA remained attached to the surface. The activity of lysozyme adsorbed to HA was reduced by adsorption of GTF to the same surface and was almost completely abolished by formation of glucans by the adsorbed GTF. These results suggest that soluble bacterial enzymes found in saliva can be incorporated into pellicle, interact with host-derived molecules on the surfaces of teeth, express enzymatic activity, and potentially influence the biological properties of pellicle.

The debate over the validity of the Rorschach has been lengthy but unresolved. Major reasons for the lack of consensus and conclusion are the absence of agreement about, how validation research should be conducted and the failure to identify appropriate criteria for subsequent evaluation. A construct validation model for the Rorschach is presented, with guidelines for proper implementation. The difficulties in establishing the optimal relationship between theory, research design, and clinical practice arc discussed. Recommendations for a resolution of these difficulties are presented.

Card pull, the tendency of the test stimuli to evoke or predispose certain cognitive and affective responses in the subject, is essential to the understanding and practice of projective testing. The basic instruction in projective testing employs the subjunctive mood to catalyze the dialectic between actuality and potentiality inherent in the projective stimulus. The subject is encouraged to play with the stimulus, to modify fact, to forsake the literal and the concrete, and to toy with the reality-stuff of the stimulus. Various conceptions of projective stimuli are reviewed and considered nondefinitive of projective testing. Projective testing is tentatively defined as a state of mind, a temporary but adaptive collusion shared between tester and subject which involves the former inviting the latter to modify a sample of reality.
