Objective: We sought to evaluate the safety, tolerability, and patterns of use for the once-daily oral, narrow-spectrum antibiotic sarecycline in patients with moderate-to-severe acne vulgaris during a 40-week Phase III, multicenter, open-label extension study. Participants: Patients aged nine years or older with moderate-to-severe acne who completed one of two prior Phase III, double-blind, placebo-controlled, 12-week trials in which they received sarecycline 1.5mg/kg/day or placebo were included. Measurements: The primary assessment was the safety of sarecycline 1.5mg/kg/day for 40 weeks as indicated by adverse events (AEs), vital signs, electrocardiograms, clinical laboratory tests, and physical examinations. Patterns of sarecycline use were a secondary assessment. Results: The safety population included 483 patients; 354 patients (73.3%) completed the study. The most common reasons for premature discontinuation were withdrawal by the patient (14.5%), lost to follow-up (7.9%), and AEs (2.5%). The most common treatment-emergent AEs (TEAEs) were nasopharyngitis (3.7%), upper-respiratory-tract infection (3.3%), headache (2.9%), and nausea (2.1%). Clinical laboratory evaluations suggested no clinically meaningful differences between the treatment sequences. Rates of TEAEs commonly associated with other tetracycline antibiotics include dizziness (0.4%) and sunburn (0.2%), and for gastrointestinal TEAEs, nausea (2.1%), vomiting (1.9%), and diarrhea (1.0%). Also reported herein are the results of a Phase I phototoxicity study. Conclusion: Patients aged nine years or older with moderate-to-severe acne vulgaris who received sarecycline once daily for up to 40 weeks showed low rates of TEAEs, with nasopharyngitis, upper-respiratory-tract infection, headache, and nausea being the only TEAEs reported by 2% or more of patients. Im Rahmen der vorliegenden Arbeit werden ethische Dilemmata als inter- und intrapersonelle Konfliktsituationen innerhalb des Bankensektors untersucht. Den thematischen Untersuchungsrahmen und Analysekontext bildet dabei die vergangene Weltfinanzkrise der Jahre 2007 und 2008, die insbesondere Fragen nach der Verhältnisbestimmung von Ethik und Ökonomik aufwirft. Hierzu erfolgt zunächst eine systematische Analyse einer grundsätzlich sinnvollen Verortung und Berechtigung von Ethik und Moral in die ökonomischen Prozesse und Realitäten des Bankensektors. Zu diesem Zweck wird ein Kanon ethischer Begrifflichkeiten konzipiert, der spezifisch auf den Bankensektor zur Bestimmung und Analyse ethisch relevanter Aspekte angewendet wird. Als theoretische Leitplanken und Vergleichstheorien dieser Arbeit fungieren im Untersuchungsverlauf die disjunkten wirtschaftsethischen Konzepte der „Ökonomischen Ethik“ und der „Integrativen Wirtschaftsethik“. Hiernach erfolgt die Untersuchung des Modells des Homo Oeconomicus als bislang bedeutendstem Modell zur Abbildung und Analyse von Anreizmechanismen und Vorteilserwartungen wirtschaftlicher Akteure im Kontext intrapersoneller Konfliktsituationen. Im Ergebnis zeigt sich, dass dieses Modell zwar zur Offenlegung von systematisch bedingten Fehlanreizen und Defekten auf der wirtschaftlichen Rahmenordnung beitragen kann, jedoch gewisse Unschärfen aufgrund der Vernachlässigung kommunikativer und interaktions-spezifischer Aspekte beinhaltet. Die Analyse des Gefangenen-Dilemmas sowie der Principal-Agent-Theorie im Bereich interpersoneller Konfliktsituationen offenbart dann den systematischen und strukturellen Problemaufriss für ökonomische Aspekte in Form der jeweils vorzufindenden Ausbeutungsbefürchtungen und -anreize bei den beteiligten Akteuren, die im Ergebnis stets suboptimale Ergebniskonstellationen herbeiführen. Die Übertragung der aus den genannten Modellen erarbeiteten Erkenntnisse führt dann anhand von drei spezifisch beleuchteten Bankgeschäften (Kredit-, Wertpapier- und Einlagengeschäft) zu dem Ergebnis, dass ethische Dilemmasituationen und Konfliktpotenziale in realen Bankprozessen vorzufinden sind, was über Praxisbeispiele der genannten Geschäftsarten nachgebildet und validiert wird. Abschließend zeigt die Untersuchung auf, dass zur notwendigen Implementierung von moralfördernden Anreiz- und bestandsfesten Sanktionsmechanismen innerhalb ökonomischer Prozesse die größtmögliche Realisierungswahrscheinlichkeit auf der Makro-Ebene vermutet werden muß. Eine Verortung auf die Mikro- und die Meso-Ebene zeigt hingegen, dass aufgrund der unverhältnismäßigen Moralanforderungen an die einzelwirtschaftlichen Akteure und der Gefahr, tendenziell amoralisches Verhalten in den relevanten Märkten zu befördern, diese beiden Handlungsebenen als systematische Orte für Moral in einem ökonomischen Kontext ausscheiden. This thesis covers the identification and analysis of ethical dilemmas as interpersonal and personal conflict situations within banking industry. In this context the bygone financial crisis with its economic peaks in 2007 and 2008 especially raised issues and corresponding aspects regarding the appropriate relation between ethics and economics. Firstly, a methodical analysis for the useful implementation of ethics and morality into banking industry`s processes and conditions is examined. For that purpose a catalogue of ethical terminologies has been conceived for the specific utilization and detection of ethically relevant aspects within banking industry. Furthermore, the so called „Ökonomische Ethik“ and „Integrative Wirtschaftsethik“ as specific theories in the German-speaking area are used in this thesis for theoretical comparison and academic discussion. Secondly, the economically important and abstract model of the „Homo Oeconomicus“ is evaluated for the illustration and analysis of incentive mechanisms and benefit expectations for personal conflict situations. As a result, the Homo Oeconomicus model is useful for unveiling systematic disincentives and defects on the macroeconomic sphere, but reveals vague results based on neglected aspects of communication and interaction. Afterwards the „Prisoners dilemma“ and the „Principal-agent-theory“ demonstrate the systematic and structural problems for economic aspects and processes within interpersonal conflict situations based on the stimulus and simultaneous fear for exploitation between the parties involved. In the end only suboptimal results occur within these theoretical models. Thirdly, all knowledge gained by analysis of the aforementioned aspects is assigned to three processes of banking business (credit processes, securities processes and deposit processes) and indicates that at least these selected banking processes contain ethical dilemmas and capabilities for interpersonal and personal conflict situations. Finally, the analysis shows that optimal conditions for the implementation of incentive mechanisms and a corresponding system of sanctions to support additional morality within economic processes might only be seen on the macroeconomic sphere. The micro- and mesoeconomic areas need to be rejected as potential solutions for more morality in economic aspects, because of disproportional demands towards the parties involved and the immanent risk to (unconsciously) enhance immoral activities.

Below, we review the use of DT40 cells in dissecting molecular mechanisms involved in Ca2+, Mg2+, and Zn2+ transport physiology. These studies highlight the flexibility and advantages the DT40 environment offers to investigators interested in the study of basic vertebrate ion transport physiology.
