Our aim was to evaluate the effects of a low or high dietary phosphorus (P) concentration during the dry period, followed by either a high or low dietary P concentration during the first 8 wk of lactation, on plasma Ca concentrations, feed intake, and lactational performance of dairy cattle. Sixty pregnant multiparous Holstein Friesian dairy cows were assigned to a randomized block design with repeated measurements and dietary treatments arranged in a 2 × 2 factorial fashion. The experimental diets contained 3.6 (Dry-HP) or 2.2 (Dry-LP) g of P/kg of dry matter (DM) during the dry period, and 3.8 (Lac-HP) or 2.9 (Lac-LP) g of P/kg of DM during 56 d after calving period. The objective of the study was to determine the effect of Bacillus subtilis supplementation on the production of heat-stressed cows in a tropical environment during summer and rainy seasons. Twenty-eight lactating crossbred Holstein-Friesian cows (89 ± 50 days in milk) on four farms in Thailand were randomly assigned to treatment sequences in a replicated 4 × 4 Latin square design. Each 21 days of experimental period included 13 days of adaptation followed by data collection from day 14 through 21. A basal ration was fed to all cows. Cows were individually fed 0 (CON), 0.5 × 1011 (BS0.5), 1 × 1011 (BS1) or 2 × 1011 CFU (BS2) of B. subtilis/day. Throughout the study, microclimatic conditions inside the barns were recorded. Milk weights were measured on days 14 and 21, and milk samples were collected on day 21. Whole blood samples were collected for haematological and biochemical analyses only during the first period of the study. Milk yield increased (P < 0.01) linearly as the level of B. subtilis increased. The temperature-humidity index (THI) was negatively related to milk yield (r = −0.52, P < 0.01), protein (r = −0.51, P < 0.01) and lactose (r = −0.49, P < 0.01). Cows fed B. subtilis at the level of 2 × 1011 CFU/day produced 1.7 kg/day more milk. So, it was indicated that milk yield increased linearly with the level of supplemented B. subtilis, and the supplementation did not exert any effect on haematological and biochemical parameters.

The effect of application of dried mao pomace (DMP), fermented juice of epiphytic LAB (FJLB), and Lactobacillus paraplantarum (LPP) on the ensiling process of Stylo (Stylosanthes guianensis CIAT184) was evaluated. Stylo silage was treated with 10% DMP alone or in combination with either FJL Bor LPP. The addition of DMP to Stylo reduced the mean pH by at least 0.9 units while the use of LAB was found to maximally reduce pH by 0.21 units. Irrespective of the source of LAB, the pH reducing effect of LAB was only observed in the silages without DMP. NH3-N contents were found to be significantly affected by DMP × LAB. The use of DMP reduced the NH3-N contents by at least 50%, irrespective of the addition of LAB. Furthermore, HBut levels were below detection limits when DMP was used as a silage additive and in case the silages were not treated with DMP, the addition of either FJLB or LPP also resulted in lower HBut fractions. In view of the high NH3-N and HBut levels, it was therefore concluded that ensiling of Stylo without DMP or LAB results in an inferior silage quality. The use of DMP as a silage additive resulted in good quality silage, irrespective of the use of additional LAB.

In equines, Cr2O3 is widely accepted as an indigestible marker, but there are health concerns regarding the carcinogenic properties of Cr2O3. Recently, TiO2 has been suggested to be an alternative digestibility marker in equines. However, a comparison between Cr2O3 and TiO2 has not been made in equines. Six Welsh pony geldings (initial BW: 254±3 kg; 7 years of age) fed chopped alfalfa hay were used to evaluate the use of TiO2 (Ti) and Cr2O3 (Cr) as markers for calculating apparent digestibility and to investigate the effect of frequency of marker administration on the measurement of digestibility values. Diets contained 4.65 kg dry matter (DM) chopped alfalfa hay supplemented with minerals, vitamins, TiO2 (3.3 g Ti/day) and Cr2O3 (3.2 g Cr/day). Ponies were dosed with either 3.3 g Ti and 3.2 g Cr once daily (DF1) or with 1.65 g Ti and 1.60 g Cr twice daily (DF2). After adaptation to the diets and procedures for 14 days, voluntary voided faeces were collected quantitatively over 7 days and analysed for moisture, ash, Ti and Cr. Apparent total tract DM digestibility (DMD) and organic matter digestibility (OMD) were calculated using the total faecal collection (TFC) and marker method (Ti and Cr). The overall mean cumulative faecal recovery of Cr and Ti (as % of intake) were 102.0% and 96.6%, respectively. Mean daily faecal recoveries of Cr as well as of Ti were not different (P=0.323; P=0.808, respectively) between treatments. Overall daily faecal recovery of Cr differed (P=0.019) from 100% when the marker was dosed once daily, whereas overall daily faecal recovery was similar to 100% for both administration frequencies when Ti was used as a marker. For both markers, the coefficient of variation of the mean faecal marker recovery between horses was lower when the markers were administrated twice per day. Across treatments, cumulative DMD and OMD estimated with Ti were similar (P=0.345; P=0.418, respectively) compared with those values determined by TFC method. When Cr was used, the calculated cumulative DMD tended (P=0.097) to be greater compared with those estimated with TFC, and cumulative OMD values were overestimated (P=0.013). Orally supplemented Ti recovery in the faeces of ponies fed chopped alfalfa hay with Ti administered once or twice daily was close to 100%, making it the preferred marker for digestibility trials in equines.

Methodological aspects of digestibility measurements were studied in four Welsh pony geldings consuming haylage-based diets with increasing proportions of a pelleted concentrate according to a 4×4 Latin square design experiment. Ponies were fed four experimental, iso-energetic (net energy (NE) basis) diets (i.e. 22 MJ NE/day) with increasing proportions of a pelleted concentrate (C) in relation to haylage (H). The absolute amounts of diet dry matter fed per day were 4.48 kg of H (100H), 3.36 and 0.73 kg of H and C (75H25C), 2.24 and 1.45 kg of H and C (50H50C) and 1.12 and 2.17 kg of H and C (25H75C). Diets were supplemented with minerals, vitamins and TiO2 (3.7 g Ti/day). Voluntary voided faeces were quantitatively collected daily during 10 consecutive days and analysed for moisture, ash, ADL, acid-insoluble ash (AIA) and Ti. A minimum faeces collection period of 6 consecutive days, along with a 14-day period to adapt the animals to the diets and become accustomed to the collection procedure, is recommended to obtain accurate estimations on dry matter digestibility and organic matter digestibility (OMD) in equids fed haylage-based diets supplemented with concentrate. In addition, the recovery of AIA, ADL and Ti was determined and evaluated. Mean faecal recovery over 10 consecutive days across diets for AIA, ADL and Ti was 124.9% (SEM 2.9), 108.7% (SEM 2.0) and 97.5% (SEM 0.9), respectively. Cumulative faecal recovery of AIA significantly differed between treatments, indicating that AIA is inadequate to estimate the OMD in equines. In addition, evaluation of the CV of mean cumulative faecal recoveries obtained by AIA, ADL and Ti showed greater variations in faecal excretion of AIA (9.1) and ADL (7.4) than Ti (3.7). The accuracy of prediction of OMD was higher with the use of Ti than ADL. The use of Ti is preferred as a marker in digestibility trials in equines fed haylage-based diets supplemented with increasing amounts of pelleted concentrate.

The current study investigated the relationship between in vitro and in vivo CH4 production by cows fed corn silage (CS)-based rations. In vivo CH4 production was measured in climate respiration chambers using 8 rumen-cannulated Holstein-Friesian cows. In vitro CH4 production was measured using rumen fluid from the 8 cows that were fully adapted to their respective experimental rations. The animals were grouped in 2 blocks, and randomly assigned to 1 of the 4 total mixed rations (TMR) that consisted of 75% experimental CS, 20% concentrate, and 5% wheat straw [dry matter (DM) basis]. The experimental CS were prepared from whole-plant corn that was harvested at either a very early (25% DM), early (28% DM), medium (32% DM), or late (40% DM) stage of maturity. The 4 experimental TMR and the corresponding CS served as substrate in 2 separate in vitro runs (each run representing 1 block of 4 animals) using rumen fluid from cows fed the TMR in question. No relationship was found between in vivo CH4 production and in vitro CH4 production measured at various time points between 2 and 48 h. None of the in vitro gas production (GP) and CH4 production parameters was influenced by an interaction between substrate and origin of rumen fluid. In vitro measured 48-h GP was not affected by the maturity of whole-plant corn, irrespective whether CS alone or as part of TMR was incubated in adapted rumen inoculum. Incubation of the experimental TMR did not affect the kinetics parameters associated with gas or CH4 production, but when CS alone was incubated the asymptote of GP of the soluble fraction was slightly decreased with increasing maturity of CS at harvest. In vitro CH4 production expressed as a percent of total gas was not affected by the maturity of whole-plant corn at harvest. Several in vitro parameters were significantly affected (GP) or tended to be affected (CH4) by diet fed to donor cows. It was concluded that the current in vitro technique is not suitable to predict in vivo CH4 production from CS-based rations.

Knowledge of the morphological adaptation of rumen papilla, which plays an important role in volatile fatty acid absorption, in dry and early lactation dairy cattle is limited. Therefore, macro- and microscopic changes in papilla morphology during the dry period and lactation and the effect of rate of increase of concentrate allowance were studied. Samples were collected from 12 rumen-cannulated Holstein Friesian dairy cows during a pretreatment period, 50, 30, and 10 d antepartum (the dry period) and 3 d postpartum (pp), and a treatment period, 9, 16, 30, 44, 60, and 80 d pp. Cows had free access to either a dry period ration [27% grass silage, 27% corn silage, 35% wheat straw, and 11% soybean meal on a dry matter (DM) basis] or a basal lactation ration (42% grass silage, 41% corn silage, and 17% soybean meal on a DM basis, and 0.9 kg of DM/d concentrate). Treatment consisted of either a rapid (1.0 kg of DM/d; RAP; n=6) or gradual (0.25 kg of DM/d; GRAD; n=6) increase of concentrate allowance (up to 10.9 kg of DM/d), starting at d 4 pp, aimed at creating a contrast in rumen-fermentable organic matter (FOM) intake. Papillae were collected from the ventral, ventral blind, and dorsal blind rumen sacs and measured digitally. Intake of DM (11.9 kg/d) and FOM (5.7 kg/d) did not change during the pretreatment period, but increased during the treatment period to 24.5 and 15.0 kg/d at 80 d pp, respectively. Concentrate treatment and sampling day interacted for FOM intake, which was 22% greater in RAP at 16 d pp compared with GRAD. Papilla surface area decreased during the pretreatment period by 19% to 28.0mm(2) at 3 d pp, thereafter increasing to 63.0mm(2) at 80 d pp. Concentrate treatment and sampling day interacted for surface area, which was greater in RAP compared with GRAD at 16 (46.0 vs. 33.2mm(2)), 30 (55.4 vs. 41.2mm(2)), and 44 (60.5 vs. 49.7 mm(2)) days pp, showing that papillae can respond to a rapid rate of increase of FOM intake by increasing growth rate. Microscopic morphology was affected by sampling day, but neither by concentrate treatment nor by their interaction, with a decrease in papilla and epithelium thickness during the lactation. In conclusion, the rumen papillae respond to changes in FOM intake and the magnitude of this response depends on the rate of increase of FOM intake. This response in surface area of the rumen papillae potentially facilitates the absorption of the volatile fatty acids.

The aim of the present experiment was to study changes in volatile fatty acid (VFA) production using an isotope dilution technique, and changes in VFA fractional absorption rate (k aVFA) using a buffer incubation technique (BIT) during the dry period and early lactation, as affected by the postpartum (pp) rate of increase of concentrate allowance. The current results are complementary to previously reported changes on rumen papillae morphology from the same experiment. From 50 d antepartum to 80 d pp, VFA production rate was measured 5 times and k aVFA was measured 10 times in 12 rumen-cannulated Holstein Friesian cows. Cows had free access to a mixed ration, consisting of grass and corn silage, soybean meal, and (dry period only) chopped straw. Treatment consisted of either a rapid (RAP; 1.0 kg of DM/d; n = 6) or gradual (GRAD; 0.25 kg of DM/d; n = 6) increase of concentrate allowance (up to 10.9 kg of DM/d), starting at 4 d pp, aimed at creating a contrast in rumen-fermentable organic matter intake. For the BIT, rumen contents were evacuated, the rumen washed, and a standardized buffer fluid introduced [120 mM VFA, 60% acetic (Ac), 25% propionic (Pr), and 15% butyric (Bu) acid; pH 5.9 and Co-EDTA as fluid passage marker]. For the isotope dilution technique, a pulse-dose of 13C-labeled Ac, Pr, and Bu and Co-EDTA as fluid passage marker was infused. The rate of total VFA production was similar between treatments and was 2 times higher during the lactation (114 mol/d) than the dry period (53 mol/d). Although papillae surface area at 16, 30, and 44 d pp was greater in RAP than GRAD, Bu and Ac production at these days did not differ between RAP and GRAD, whereas at 16 d pp RAP produced more Pr than GRAD. These results provide little support for the particular proliferative effects of Bu on papillae surface area. Similar to developments in papillae surface area in the dry period and early lactation, the k aVFA (per hour), measured using the BIT, decreased from 0.45 (Ac), 0.53 (Pr) and 0.56 (Bu) at 50 d antepartum to 0.28 (Ac), 0.34 (Pr) and 0.38 (Bu) at 3 d pp. Thereafter, k aVFA (/h) rapidly increased up to 0.67 (Ac), 0.79 (Pr), and 0.79 (Bu) at 80 d pp. Although papillae surface area was greater at 16, 30, and 44 d pp in RAP than GRAD, no differences in k aVFA between RAP and GRAD were observed during these days showing papillae surface area is not the limiting factor for k aVFA during early pp adaptation.

In a 2×2 factorial design, 12 Thai Native and 12 Holstein bulls were fed ad libitum a total mixed ration (20 : 80; roughage : concentrate) with whole cottonseed (WCS) or sunflower seed (SFS) as oilseed sources. The rations contained 7% crude fat and were fed for 90 days. Plasma was taken at three times during the experiment, and at slaughter the longissimus dorsi and subcutaneous fat were sampled for fatty acid analysis. Ration did not affect rumen fermentation parameters. The plasma fatty acid profile was not affected by ration. In subcutaneous fat, a ration×breed interaction for the saturated fatty acid (SFA) and c9t11 CLA proportions was observed, resulting from larger differences between the rations in Thai Native compared with Holstein bulls. The objective of this study was to obtain information on variation between dairy cows in muscle and fat tissue mobilization around parturition and to study the association between protein and fat mobilization and serum beta-hydroxybutyrate (BHBA) concentrations (hyperketonemia) in this period. Thirty-four cows kept under similar conditions at a university dairy farm (no experimental treatments) were monitored from 4 wk before until 8 wk after calving. Mobilization of muscle protein was investigated by analysis of plasma 3-methylhistidine concentrations (3-MH, analyzed by a recently developed HPLC tandem mass spectrometry method) and ultrasound measurements of longissimus muscle thickness. Mobilization of fat tissue was monitored by serum nonesterified fatty acid (NEFA) concentrations and ultrasound measurements of backfat thickness. Large variation was observed between cows in onset and duration of periparturient protein and fat mobilization. Plasma 3-MH concentrations and muscle thickness profiles indicated that protein mobilization started, on average, before parturition and continued until approximately wk 4 of lactation. Serum NEFA concentrations and backfat thickness profiles showed that fat mobilization occurred from parturition until the end of the study. Thus, muscle protein mobilization occurred in advance of fat mobilization in most cows from this study. We hypothesized that this might be due to a prepartum amino acid deficiency in the absence of negative energy balance. The incidence of hyperketonemia in this study was 16/34 = 47%. With the exception of 3 cows defined as having severe hyperketonemia, cows with lower 3-MH concentrations had higher serum BHBA concentrations. A possible explanation for this observation might be that higher mobilization of protein around calving might restrict ketone body production due to the higher availability of glucogenic precursors in the period of most severe negative energy balance and highest fat mobilization. The validity of this hypothesis needs to be confirmed, but data from this study indicate that further research on the role of protein mobilization in the etiology of hyperketonemia in dairy cows is needed.
