Abstract

Ribosomally produced thiopeptide antibiotics are promising lead compounds targeting the GTPase-associated region (GAR) of the bacterial ribosome. The studies have shown that mutations of the L11 protein do not counteract binding of bicyclic thiopeptides, but has allowed the ribosome to bypass the protein biosynthesis blockade enforced by these antibiotics in the wild type.

Abstract

Die Thiopeptid-Naturstoffe sind eine Gruppe von strukturell komplexen, makrozyklischen Peptiden, die sehr hohe Wirkung gegen Gram-positive Bakterien aufweisen. Die potentesten Vertreter stellen sub-nanomolare Hemmstoffe der GTPase-assoziierten Region des Ribosoms dar. Diese Region ist für die dynamische Interaktion des Ribosoms mit verschiedenen Proteinfaktoren während des Translationszyklus essentiell, so dass ihre Inhibierung zum Abbruch des Translationsvorgangs und Absterben der Bakterien führt. Die Thiopeptidbindestelle der GTPase-assoziierten Region zeichnet sich strukturell und funktional durch das enge Zusammenwirken von Protein- und RNAKomponenten aus. Die Thiopeptide wechselwirken hier kooperativ mit dem L11-Protein und mit der 23S rRNA. Mit dieser Eigenschaft stellen sie eine Besonderheit unter den antibakteriellen Hemmstoffen dar. Bislang sind nur wenige Beispiele von Inhibitoren bekannt, die gleichzeitig mit Protein- als auch mit RNA-Komponenten wechselwirken. Die Bindestelle der GTPase-assoziierten Region des Ribosoms wird derzeit von keinem therapeutisch eingesetzten Antibiotikum ausgenutzt. Ein detaillierteres Verständnis der Bindeeigenschaften kann daher zur Entwicklung von neuen ribosomalen Wirkstoffen beitragen.

22. Synopsis of Structural, Biosynthetic, and Chemical Aspects of Glycopeptide Antibiotics.

Abstract

Glycopeptide antibiotics represent a very important group of natural products with regard to medicinal application as antibacterials and for cancer treatment. A characteristic for assignment as a glycopeptide antibiotic, next to antibiotic potential, is the modification of the peptide aglycon with various types of carbohydrates. According to this definition, vancomycin (including five subtypes), ramoplanin, bleomycin, mannopeptimycins and salmochelin belong to the family of glycopeptide antibiotics. In this article, an overview is given on the structural aspects, the biosynthesis and the mode of action of glycopeptide antibiotics. Structure-activity relationships of important functional groups contributing to enhanced antibiotic properties are highlighted. Finally, similarities with regard to structural features, the biosyntheses and the modes of actions are discussed. According to current knowledge, glycopeptide antibiotics mostly act as inhibitors of bacterial cell wall biosynthesis. It is to be expected that the actual list of glycopeptides will be continuously extended due to discoveries from ongoing screening programs. [ABSTRACT FROM AUTHOR]
