We describe the animal species identification of meat using MALDI-TOF mass spectroscopy including the development and validation of a reliable method, qualified for use in the accredited official food-control laboratory. Previous publications had shown the potential of MALDI-TOF MS for animal species differentiation of several kind of food, including meat. Our aim was to establish a rapid and reliable method by means of a simplified sample preparation without prior tryptic digest, an existing popular MALDI system, an independent extensive reference database, and an adequate validation concept. In contrast to the previous works, we consequently use the MALDI user platform “MALDI-UP” to give other food control laboratories the possibility of exchanging reference and validation spectra.

Seit einigen Jahrzehnten werden immer häufiger Wasserblüten in Gewässern beobachtet. Die Ursache dafür ist die abnormale Massenvermehrung von Cyanobakterien. Der genaue Grund für diese Erscheinung ist noch nicht bis ins Detail geklärt. Es wurde jedoch beobachtet, daß bestimmte Muster der Nährstoffverfügbarkeit, im wesentlichen Phosphor, die cyanobakterielle Proliferation fördern. Wegen des Geruchs und Gestanks werden die Wasserqualität und die Trinkwasserversorgung durch Wasserblüten erheblich beeinträchtigt. Außerdem produzieren viele wasserblütenbildende Cyanobakterien Toxine, so daß der Verzehr vom wasserblütenhaltigen Wasser gesundheitsschädlich sein kann. Um die mit Wasserblüten einhergehenden Probleme zu vermeiden, wurden im Rahmen dieser Arbeit zwei Assays als ein Frühwarnsystem zur Überwachung von Wasserblüten- sowie Cyanotoxinbildung entwickelt. Ausgehend von einem cyanobakteriellen Reporterstamm mit der Konstruktion PphoA::luxAB im Genom wurde ein lumineszierender Reporterassay zur Detektion der für Cyanobakterien verfügbaren Phosphatquellen entwickelt. Durch Immobilisierung ist der Sensor lagerfähig und der Assay leicht handhabbar. Basierend auf der Hybridisierungstechnik konnte in dieser Arbeit mit spezifischen Sonden ein Assay im Mikroarray-Format zur molekulargenetischen Detektion von der in Wasserblüten am häufigsten gefundenen cyanobakteriellen Gattung Microcystis und dem Gen der für die Produktion von hepatotoxischen Microcystinen verantwortlichen Microcystin-Synthetase entwickelt werden. Dieser Oligonukleotid-Mikroarray ermöglicht eine schnelle Identifizierung der in Wasserblüten beteiligten Microcystis-Stämme und eine einfache Beurteilung der Gefährdung durch "blühende" Gewässer., Since a few decades water blooms are more and more frequently observed. It is the result of abnormal cyanobacterial mass occurrence. The exact reason of this phenomenon is not yet clear. It has been, however, reported, that some patterns of the availability of nutrients, especially P, N, and Fe, strongly promote cyanobacterial proliferation. Besides their negative effects on water quality, many cyanobacteria found in water blooms produce toxins, so that the consumption of water containing cyanobacterial blooms may lead to even death. In order to avoid the problems associated with water blooms, two assays as a early warning system for monitoring of water blooms formation and cyanotoxin production were developed in this work. Starting form a cyanobacterial reporter strain harbouring PphoA::luxAB in its genom a luminescent reporterassay for detection of phosphate availability for cyanobacteria was developed. By means of immobilisation of this reporter strain the sensor was storable and the performace of this assay was easy. Based on the hybridization technique and using the specific probes designed in this work an assay was developed in microarray-format for molecular genetic detection of Microcystis, the most frequently cyanobacterial genus found in water blooms, and the gene of microcystin synthetase, which is responsible for the production of hepatotoxic microcystins. This oligonucleotide-microarray enables a quick detection of the Microcystis in water samples and indication of the risk in water containing cyanobacterial blooms.
