- Abstract
The cortex controls cell shape. In mouse oocytes, the cortex thickens in an Arp2/3-complex-dependent manner, ensuring chromosome positioning and segregation. Surprisingly, we identify that mouse oocytes lacking the Arp2/3 complex undergo cortical actin remodeling upon division, followed by cortical contractions that are unprecedented in mammalian oocytes. Using genetics, imaging, and machine learning, we show that these contractions stir the cytoplasm, resulting in impaired organelle organization and activity. Oocyte capacity to avoid polyspermy is impacted, leading to a reduced female fertility. We could diminish contractions and rescue cytoplasmic anomalies. Similar contractions were observed in human oocytes collected as byproducts during IVF (in vitro fertilization) procedures. These contractions correlate with increased cytoplasmic motion, but not with defects in spindle assembly or aneuploidy in mice or humans. Our study highlights a multiscale effect connecting cortical F-actin, contractions, and cytoplasmic organization and affecting oocyte quality, with implications for female fertility.

In the framework of the compact STorage ring for Accelerator Research and Technology (cSTART) project, which will be realized at Karlsruhe Institute of Technology (KIT), a Very Large Acceptance compact Storage Ring (VLA-cSR) is planned to study the injection and the storage of 50 MeV, ultra-short (sub-ps) electron bunches from a laser plasma accelerator (LPA) and the linac-based test facility FLUTE. For such a storage ring, where a single bunch with a relatively wide range of bunch charge (1 pC - 1000 pC ) and energy spread (10’4 - 10’2) will circulate at a relatively high revolution frequency (7 MHz), the choice of beam diagnostics is very delicate. In this paper, we would like to discuss several beam diagnostics options for the storage ring and to briefly report on several tests that have been or are planned to be realized in our existing facilities.

Electron storage rings operating at high energies have proven to be invaluable source of synchrotron radiation. Two and even three simultaneous beams of particles have been observed at different light source facilities worldwide. So called alpha-buckets were studied at Metrological Light Source (MLS) in Berlin (Germany), SOLEIL facility in France, DIAMOND light source in UK, NSLS ring in Brookhaven (USA). It is widely recognized that alpha buckets are general phenomena which is not restricted to a certain storage ring. However, earlier measurements showed essential limitations on parameters of alpha-buckets, strong dependence on high order (sextupole and octupoles) magnetic field imperfections and associated fast decay of electron current as well as reduced life time. Also a rising relevance of high order non-linear longitudinal beam dynamics is associated with new generation of diffraction limited light sources (DLSR) approaches, which all suffer nonlinear momentum compaction factor. A large variety of future generation electron synchrotrons require a comprehensive investigation of the physical processes involved into the operation of such rings. In this paper, we present review of high order non-linear longitudinal beam dynamics based on the longitudinal equations of motion and Hamiltonian expanded to a high order of the momentum compaction factor. Roots of the third order equation with a free term are derived in a form suitable for analytical estimations. The momentum independent term of orbit lengthening due to particle transverse excursions is estimated and taken into account. The results from simulations were benchmarked against existing experiments at the Karlsruhe Research Accelerator (KARA), SOLEIL and MLS rings. Parameters of three simultaneous beams and alpha buckets at MLS and SOLEIL have been reproduced with high accuracy. General conditions for stable operation of alpha buckets are presented. Based on analytical formulas and computer simulations, studies of longitudinal motion at KARA have been performed with an objective to estimate feasibility of filling and storing of beam in α-buckets. A Computer model was used to describe the behavior and dynamics of simultaneous beams in the KARA storage ring.

At KARA, the Karlsruhe Research Accelerator of the Karlsruhe Institute of Technology synchrotron, the so-called short-bunch operation mode allows the reduction of the bunch length down to a few picoseconds. The microbunching instability resulting from the high degree of longitudinal compression leads to fluctuations in the emitted terahertz radiation. For highly compressed bunches at KARA, the instability occurs not only in one but in two different bunch-current ranges that are separated by a stable region. The additional region of instability is referred to as short-bunch-length bursting or weak instability. We will present measurements of the threshold currents and fluctuation frequencies in both regimes. Good agreement is found between the measurement and numerical solutions of the Vlasov-Fokker-Planck equation. This contribution is based on the PRAB paper Phys. Rev. Accel. The accelerator FLUTE (name abbreviation derived from its German name: Ferninfrarot Linac- und TestExperiment) has been set up in cooperation with DESY and PSI [1]. The electron source and diagnostics has commenced operation. General safety issues of FLUTE are covered in this paper. The activation of the accelerator and vacuum parts were predicted previously [2]. The attention is given to the activation of aluminum and impurities in the electron absorber of the beam dump. Potential air activation in the experimental hall is also discussed.

Single-shot electro-optic sampling (EOS) is a powerful characterization tool for monitoring the shape of electron bunches, and coherent synchrotron radiation pulses. For reaching high acquisition rates, an efficient possibility consists to associate classic EOS systems with the so-called photonic time-stretch technique [1]. We present recent results obtained at SOLEIL and ANKA using this strategy. In particular, we show how a high sensitivity variant of photonic time stretch [2] EOS enabled to monitor the CSR pulses emitted by short electron bunches at SOLEIL [3]. We could thus confirm in a very direct way the theories predicting an interplay between two physical processes. Below a critical bunch charge, we observe a train of identical THz pulses stemming from the shortness of the electron bunches. Above this threshold, CSR emission is dominated by drifting structures appearing through spontaneous self-organization. We also consider the association of time-stretch and EOS for recording electron bunch near fields at high repetition rate. Accurate and rapid evaluation of radar signature for alternative aircraft/store configurations would be of substantial benefit in the evolution of integrated designs that meet radar cross-section (RCS) requirements across the threat spectrum. Finite-volume time domain methods offer the possibility of modeling the whole aircraft, including penetrable regions and stores, at longer wavelengths on today's gigaflop supercomputers and at typical airborne radar wavelengths on the teraflop computers of tomorrow. A structured-grid finite-volume time domain computational fluid dynamics (CFD)-based RCS code has been developed at the Rockwell Science Center, and this code incorporates modeling techniques for general radar absorbing materials and structures. Using this work as a base, the goal of the CFD-based CEM effort is to define, implement and evaluate various code development issues suitable for rapid prototype signature prediction.

The potential for a superconducting proton linac (SPL) at CERN started to be seriously considered at the end of the 1990s. In the first conceptual design report (CDR), published in 2000 [1], most of the 352 MHz RF equipment from LEP was re-used in an 800 m long linac, and the proton beam energy was limited to 2.2 GeV. During the following years, the design was revisited and optimized to better match the needs of a high-power proton driver for neutrino physics. The result was a more compact (470 m long) accelerator capable of delivering 5 MW of beam power at 3.5 GeV, using state-of-the-art superconducting RF cavities at 704 MHz. It was described in a second CDR, published in 2006 [2]. Soon afterwards, when preparation for increasing the luminosity of the LHC by an order of magnitude beyond nominal became an important concern, a low-power SPL (LP-SPL) was studied as a key component in the renovation of the LHC injector complex. The combination of a 4 GeV LP-SPL injecting into a new 50 GeV synchrotron (PS2) was proposed to replace the ageing Linac2, PSB, and PS. In a later stage, if necessary for future physics programmes (neutrino production or generation of radioactive ion beams), the linac could be brought up to multimegawatt beam power by upgrading the cooling, the electrical infrastructure, and the power supplies. The construction of the low-energy front end of the LP-SPL started in 2008 as the Linac4 project [3], aimed at the replacement of Linac2, with the potential for being adapted later to the needs of a low-power or high-power SPL (HP-SPL). In parallel, the R&D on the superconducting linac continued, initially to refine the design of the LP-SPL and afterwards that of the HP-SPL. This report presents the design of the LP-SPL and the work accomplished in preparing a proposal for new LHC injectors with the support of the European Commission under the Framework Programmes 6 and 7 and in collaboration with multiple laboratories and institutions worldwide. The status of the R&D towards an HP-SPL is summarized.

The Tritium-Helium-Trap (THe-Trap) is a Penning-trap mass spectrometer dedicated to measure the mass ratio of tritium and helium-3 with a relative precision of 10-11. To reach this precision, the properties of the Penning-trap must be understood very well. Geometrical deviations from the ideal case cause higher-order components in the electrostatic potential. Extended simulations have been carried out to investigate first the dependence of the simulation tool Comsol Multiphysics on the resolution chosen and then to quantify the higher-order terms. The simulations were successful and in good agreement with the experimental observations. It was also possible to simulate the image charge effect. The simulated image charge effect confirms the experimental value. Similar investigations were performed to estimate the influence of the experimental setup on the magnetic field. The influence of ferromagnetic materials was determined reliably. The final part of this thesis is the characterization of the external Penning-ion source. For this, a new Faraday cup was designed and implemented. The new device can measure an ion beam from the Penning-ion source and helps to improve their operating parameters.

Higher Order Modes (HOMs) can severely limit the operation of superconducting cavities in a linear accelerator with high beam current, high duty factor and complex pulse structure. Therefore, the full HOM spectrum has to be analysed in detail to identify potentially dangerous modes already during the design phase and to define their damping requirements. For this purpose a dedicated beam dynamics simulation code, Simulation of higher order Mode Dynamics (SMD), focusing on beam-HOM interaction, has been developed in the frame of this project. SMD allows to analyse the beam behaviour under the presence of HOMs, taking into account many important effects, such as for example the HOM frequency spread, beam input jitter, different chopping patterns, as well as klystron and alignment errors. SMD is used to investigate in detail into the effects of HOMs in the Superconducting Proton Linac (SPL) at CERN and in particular their potential to drive beam instabili- ties in the longitudinal and transverse direction. Based on these results, HOM damping requirements for the HOM coupler design are then defined. In addition, the linear ac- celerators of the European Spallation Source (ESS) and the Spallation Neutron Source (SNS) are analysed with respect to HOM impact and the results are compared with the SPL simulations.

We propose a versatile THz source named FLUTE (Ferninfrarot Linac- Und Test-Experiment) based on a 30 - 50 MeV S-band linac with bunch compressor, that aimsat not only producing high field THz pulses but also at serving as a test facility to study accelerator physics issues. This source is an important step towards the planned ultra-broadband THz to mid infrared user facility TBONE. Special emphasis is put on studies of bunch compression as a function of bunch charge (0.1-5 nC) and of different generation mechanisms of coherent radiation (CSR, CER, CTR). This paper describes the design and layout of the proposed FLUTE machine and presents results of beam dynamic calculations with the tracking programs ASTRA and CSRtrack. In addition, calculations for the achievable peak electrical field and spectral characteristics for one version of the FLUTE layout are shown.

In this paper is analysed the influence of Higher Order Modes (HOM) on the operation of the superconducting linac section of the SPL, the Superconducting Proton Linac being designed at CERN. For this purpose, the characteristics of the HOMs in the 2 different beta families (0.65, 0.92 both at 704 MHz) of the SPL are calculated to estimate their effect on the cryogenic system and on the beam stability. For both criteria the maximum external Q of the HOMs is defined.
