This systematic review synthesizes studies that used experimental designs to evaluate techniques theorized to foster the development of antiracism among youth in school settings (19 published reports; 23 independent studies; participant ages 3-19 years old, primarily White). Our goal herein was to identify unique program components, design elements, and outcome measures; and to critically evaluate the existing studies in terms of potential public health impact. A number of specific programming elements were distilled that may be included in future interventions. Overall, interventions that leveraged cognitive and educational components to help increase positive outgroup contact seemed most promising. However, most of the studies testing such programs lacked methodological robustness (e.g., probable gaps in internal validity from the absence of intervention manuals or equivalent, fidelity checks, reliance on outcome measures with unknown psychometric properties, and follow-up designs). Future research would benefit from establishing adherence to implementation (fidelity to protocol), including pre, post, and follow-up assessments, as well as using outcome measures appropriate for determining both short-term and long-term change. There is a clear need for the funding of technique development, manualized programming for delivery, rigorous evaluation of these with standardized outcome measures, and adequately powered studies testing outcomes across development.

In some Anglo-Saxon countries, such as Australia and the United States, initiatives have already emerged to promote standardized evaluation practices. However, in Italy, this process is still evolving. Since 1994, the International Valuation Standards Committee (IVSC) has been working on various documents to establish uniform evaluation practices globally. These International Valuation Standards (IVS) are gradually gaining recognition in Italy. Through the efforts of organizations like ABI, Tecnoborsa, the Revenue Agency, and professional associations, IVS are being integrated into evaluation practices for credit approval and public administration requirements.

Abstract Background Monocytes are key mediators of innate immunity to infection, undergoing profound and dynamic changes in epigenetic state and immune function which are broadly protective but may be dysregulated in disease. Here, we aimed to advance understanding of epigenetic regulation following innate immune activation, acutely and in endotoxin tolerant states. Methods We exposed human primary monocytes from healthy donors (n = 6) to interferon-γ or differing combinations of endotoxin (lipopolysaccharide), including acute response (2 h) and two models of endotoxin tolerance: repeated stimulations (6 + 6 h) and prolonged exposure to endotoxin (24 h). Another subset of monocytes was left untreated (naïve). We identified context-specific regulatory elements based on epigenetic signatures for chromatin accessibility (ATAC-seq) and regulatory non-coding RNAs from total RNA sequencing. Results We present an atlas of differential gene expression for endotoxin and interferon response, identifying widespread context specific changes. Across assayed states, only 24–29% of genes showing differential exon usage are also differential at the gene level. Overall, 19.9% (6,884 of 34,616) of repeatedly observed ATAC peaks were differential in at least one condition, the majority upregulated on stimulation and located in distal regions (64.1% vs 45.9% of non-differential peaks) within which sequences were less conserved than non-differential peaks. We identified enhancer-derived RNA signatures specific to different monocyte states that correlated with chromatin accessibility changes. The endotoxin tolerance models showed distinct chromatin accessibility and transcriptomic signatures, with integrated analysis identifying genes and pathways involved in the inflammatory response, detoxification, metabolism and wound healing. We leveraged eQTL mapping for the same monocyte activation states to link potential enhancers with specific genes, identifying 1,946 unique differential ATAC peaks with 1,340 expression associated genes. We further use this to inform understanding of reported GWAS, for example involving FCHO1 and coronary artery disease. Conclusion This study reports context-specific regulatory elements based on transcriptomic profiling and epigenetic signatures for enhancer-derived RNAs and chromatin accessibility in immune tolerant monocyte states, and demonstrates the informativeness of linking such elements and eQTL to inform future mechanistic studies aimed at defining therapeutic targets of immunosuppression and diseases.

Abstract Consumers’ interest in product information, such as nutrition, environment, and social aspects, is increasing in the food market. Blockchain technology can enhance credibility of quality signals on labels through transparency and accountability in the supply chain. This study examines the impact of blockchain technology on reducing consumer uncertainty about credence attributes and facilitating informed choices. It is particularly relevant for food policies, as greater consumer awareness promotes proactive involvement in sustainability and public health. We conducted a choice experiment with 300 Italian consumers, focusing on craft beer and analysing how blockchain technology in strengthens organic and DOP certification. The results provide valuable insights for producers and policymakers to develop voluntary approaches that engage society in objectives that concern the collective well-being. They highlight the potential of blockchain technology in communicating credence attributes and empowering consumers. However, our study reveals that the impact of blockchain technology on choices is influenced by a different level of trust, likely due to the complexity of understanding its functioning.

Abstract This study analyses the impacts of the COVID-19 pandemic on food consumption at the end of the first lockdown in the New York State (USA) and in Italy (spring 2020). The results of our study show that important changes occurred in food habits in these two countries, in which lockdown was very similar. Three models of response to the shock of the lockdown were noted in both countries. The first model (40%) includes individuals who largely increased their food consumption, the second model (26%) showed a more virtuous and responsible behaviour, while the third model (34%) displayed no change in food consumption. Diet quality in terms of healthiness and sustainability declined in the USA, while in Italy, approximately one-third of the sample showed an improvement in diet in these same areas. The use of sociodemographic, motivational, and behavioural variables to profile subjects who adhered to each food model has made it possible to obtain information that can be used to develop communication campaigns and policies for a healthier and more sustainable diet.

Abstract The increasing numbers of wild animals in Europe is leading, on the one hand, to growing problems stemming from their interaction with human activities. On the other, many European countries have still not developed national supply chains to market wild game. Instead, these supply chains could represent a win–win strategy in providing a sustainable alternative to production via intensive livestock farming and developing rural territories. Our aim was to understand consumer behaviour towards wild game meat. We conducted a choice experiment on wild boar meat on a sample of Italian consumers (625). The application of a hybrid model combining a structural equation model and a latent class analysis allowed us to identify the antecedents of attitude towards wild game meat and to analyse consumer choices by utilising attitude as an explanatory variable. The results provide useful suggestions to implement rural development policies and offer food for thought in the area of consumer behaviour.

Abstract Background Chromatin states and enhancers associate gene expression, cell identity and disease. Here, we systematically delineate the acute innate immune response to endotoxin in terms of human macrophage enhancer activity and contrast with endotoxin tolerance, profiling the coding and non-coding transcriptome, chromatin accessibility and epigenetic modifications. Results We describe the spectrum of enhancers under acute and tolerance conditions and the regulatory networks between these enhancers and biological processes including gene expression, splicing regulation, transcription factor binding and enhancer RNA signatures. We demonstrate that the vast majority of differentially regulated enhancers on acute stimulation are subject to tolerance and that expression quantitative trait loci, disease-risk variants and eRNAs are enriched in these regulatory regions and related to context-specific gene expression. We find enrichment for context-specific eQTL involving endotoxin response and specific infections and delineate specific differential regions informative for GWAS variants in inflammatory bowel disease and multiple sclerosis, together with a context-specific enhancer involving a bacterial infection eQTL for KLF4. We show enrichment in differential enhancers for tolerance involving transcription factors NFκB-p65, STATs and IRFs and prioritize putative causal genes directly linking genetic variants and disease risk enhancers. We further delineate similarities and differences in epigenetic landscape between stem cell-derived macrophages and primary cells and characterize the context-specific enhancer activities for key innate immune response genes KLF4, SLAMF1 and IL2RA. Conclusions Our study demonstrates the importance of context-specific macrophage enhancers in gene regulation and utility for interpreting disease associations, providing a roadmap to link genetic variants with molecular and cellular functions.

The discrete choice experiment is a widely used methodology in consumer studies. However, applying this method to investigate the market of products sold in a wide price range could present issues as to the quality of the estimate of preferences. In fact, for this type of product, frequently consumers may have different behaviours when faced with different price levels. For example, some market segments may refrain from purchasing products below certain price thresholds, considering them of an unacceptable quality, while others choose only below certain prices. To work around this problem area, we propose a methodology in which each respondent declares his own price interval of reference and consequently participates in a choice experiment with a price vector coherent with his habits. In this manner, we are able to grasp and include in the estimations the heterogeneity of consumers with respect to price and thus obtain more accurate willingness to pay estimates. • The method describes a procedure to bypass issues related to identifying the price vector in discrete choice experiments that involve products sold in a wide price range. • We propose a discrete choice experiment with different price vectors for consumer segments with different price preferences. In a world characterized by a significant evolution in wine consumption, Protected Designations of Origin (PDO) wines have constituted a valid strategy of marketing and competitiveness for producers. In 2014, the Consorzio Vino Chianti Classico created the ''Chianti Classico Gran Selezione'' DOCG (Denominazione di Origine Controllata e Garantita) to strengthen and expand the quality range of its productions. This is a new typology of Chianti Classico placed at the top of the quality production pyramid. The aim of this study is to verify whether the introduction of a higher-tier certification within a PDO denomination can represent an effective strategy to leverage brand value and strengthen the relationship between quality production and territory. With this purpose in mind, we performed a choice experiment on Italian wine consumers. A Latent Class Model allowed us to identify three distinct classes of consumers that differ in their preferences for price and PDO. Moreover, we performed a Chi-squared Automatic Interaction Detection technique and a Principal Component Factor Analysis for describing these classes according to consumers’ attitudes and personal characteristics. Our results show the existence of a segment that appreciates the introduction of the new label and therefore support the effectiveness of developing this strategy. In Catanzaro, Italy, an adult male with severe burns all over his body and in a state of coma was promptly rescued by the medical team at the air ambulance service (HEMS), who provided airway safety through laryngeal mask placement (LMA). The patient was subsequently transferred to the nearest Hub center, where an emergency tracheostomy was performed to ensure better airway management during the flight to the nearest available major burn center. This is the first documented case at regional level of a patient undergoing rapid tracheostomy through an imminent transfer with air ambulance.

Use of antibiotic prophylaxis (AP) in trauma patients is a common practice. However, considering the increasing rates of antibiotic resistance, AP use should be questioned and limited only to specific cases. We performed a systematic review of recent literature (from year 2000), aiming to summarize the state of the art on efficacy and appropriateness of AP in patients with traumatic injuries of torso, maxillofacial complex and skin (including burns). Twenty-six articles were selected. In thoracic trauma, AP could be useful in reducing infective complications in tube thoracostomy for penetrating trauma. In maxillo-facial trauma, AP could find a role in the peri-operative trauma setting in the case of a graft or prosthetic implant. In abdominal trauma, there is a lack of consensus on the definition of contamination, infection, antibiotic therapy, and prophylaxis. In burned patients, routine AP is not suggested. In the case of human bites to the extremities, AP could find an indication. Future studies should focus on the subcategories of patients at higher risk of infection, identifying those who would benefit from AP. Attention to antimicrobial stewardship and guidelines focused on AP in trauma are required, to reduce antibiotic abuse, and increase quality research.
