Tobacco is one of the important cash crops in China. It is sensitive to low temperature, especially in the early stage of tobacco seed germination and seedling growth. Low temperature stress directly affects the germination of tobacco seeds, leading to irregular emergence and slow growth of seedlings. Therefore, it is important to improve the vigor and cold tolerance of tobacco. In this study, two cold-sensitive tobacco varieties were primed individually with ZnSO 4 , FeSO 4 and Na 2 SeO 3 to test the effects of different microelements priming on seed germination. The results showed that under low temperature (11 °C), all three elements could improve tobacco seed vigor and promote seed germination, with selenium priming exhibiting the best effect. Selenium priming significantly increased the seed vigor index of YY97 by 29.60 % and of YY85 by 47.57 %. Moreover, selenium priming could effectively enhance POD activity in seeds, promote the oxidation of phenolic substances and enhance the activities of G-6-PDH and 6-P-GDH in the process of scavenging ROS and converting H 2 O 2 to ·O 2 - . Drought stress significantly retards the plant production. Melatonin is a vital hormone, signaling molecule, and bio-regulator of diverse physiological growth and development processes. Its role in boosting agronomic traits under diverse stress conditions has received considerable attention. However, the underlying molecular mechanism of action and how they increase drought stress tolerance has not been fully interpreted. The current study aimed to ascertain the protective role of melatonin in fortifying the antioxidant defense system, modulating the phytohormone profile, and improving agronomic traits of tomato seedlings under drought stress. After the V1 stage (1st leaf fully emerged), tomato seedlings were exposed to PEG-6000 to mimic drought-induced stress (DR 10% and DR 20%), followed by exogenous application of 100 µM soil drench. Drought-induced stress negatively impacted tomato seedlings by reducing growth and development and biomass accumulation, diminishing salicylic acid (SA) and chlorophyll levels, and dramatically lowering the antioxidant defense ability. However, melatonin protected them by activating the defense system, which decreased the oxidative burst and increased the activities of SOD, CAT, and APX. Administration of 100 µM melatonin by soil drench most remarkably downregulated the transcription factors of SlDREB3 and SlNCED3. This study has validated the moderating potential of melatonin against drought-induced stress by maintaining plant growth and development, enhancing hormone levels, elevating antioxidant enzyme activities, and suppressing the relative expression of drought-responsive genes. In heavy metal-contaminated areas, the simultaneous occurrence of increasing microplastic pollution and persistent acid rain poses a serious threat to food security. However, the mechanisms of combined exposure to microplastics (MP) and acid rain (AR) on the toxicity of cadmium (Cd) in rice seedlings remain unclear. Our study investigated the combined effects of exposure to polyvinyl chloride microplastics and AR (pH 4.0) on the toxicity of Cd (0.3, 3, and 10 mg/L) in rice seedlings. The results showed that at low Cd concentrations, the combined exposure had no significant effect, but at high Cd concentrations, it alleviated the effects of Cd stress. The combined application of MP and AR alleviated the inhibitory effects of Cd on seedling growth and chlorophyll content. Under high Cd concentrations (10 mg/L), the simultaneous addition of MP and AR significantly reduced the production of reactive oxygen species (ROS), the content of malondialdehyde (MDA), and the activity of the superoxide dismutase (SOD). Compared with AR or MP alone, the combination of MP and AR reduced root cell damage and Cd accumulation in rice seedlings. Transcriptomic analysis confirmed that under high Cd concentrations, the combination of MP and AR altered the expression levels of genes related to Cd transport, uptake, MAPK kinase, GSTs, MTs, and transcription factors, producing a synergistic effect on oxidative stress and glutathione metabolism. These results indicate that co-exposure to MP and AR affected the toxicity of Cd in rice seedlings and alleviated Cd toxicity under high Cd concentrations to some extent. Drought, a prevalent abiotic stressor, significantly impacts plant yield and quality. Melatonin (MT), a potent and economical growth regulator, plays a pivotal role in augmenting crop resilience against stress. This study investigated the efficacy of exogenous MT on drought-stressed celery seedlings by comprehensively analyzing phenotypic, physiological, and molecular attributes. The results revealed that exogenous MT mitigated celery seedling damage under drought stress, lowered malondialdehyde (MDA) concentrations, elevated oxidase activities, osmolyte levels, chlorophyll content, and augmented light energy conversion efficiency. Transcriptomic analysis demonstrated that MT could regulate chlorophyll synthesis genes (AgPORA1 and AgDVR2), contributing to heightened photosynthetic potential and increased drought tolerance in celery. Moreover, MT was found to modulate glycolytic pathways, upregulate pyruvate synthesis genes (AgPEP1 and AgPK3), and downregulate degradation genes (AgPDC2 and AgPDHA2), thereby promoting pyruvate accumulation and enhancing peroxidase activity and drought tolerance. The RNA-seq and qRT-PCR analyses demonstrated similar results, showing the same general expression trends. Key Message: MT mitigates chilling damage by enhancing antioxidant system and photosystem activities, and cold-responsive genes expression in cucumbers. H 2 O 2 may act as a downstream signaling molecule in the MT-induced chilling tolerance. Melatonin (MT) and hydrogen peroxide (H 2 O 2 ) are important endogenous signaling molecules that play multifaceted roles in plant responses to abiotic stress. However, the interactive mechanism by which MT and H 2 O 2 regulate chilling tolerance remains unclear. Here we found that MT exhibited a positive regulatory effect on the chilling tolerance of cucumbers, with an optimum concentration of 100 µM. MT markedly enhanced RBOH1 mRNA expression, activity and endogenous H 2 O 2 accumulation in cucumber seedlings. However, 1.0 mM H 2 O 2 had no significant effect on mRNA levels of TDC and ASMT, the key genes for MT synthesis, and endogenous MT content. Both MT and H 2 O 2 significantly decreased malondialdehyde (MDA), electrolyte leakage (EL) and chilling injury index (CI) by activating the antioxidant system, thereby alleviating chilling damage in cucumber seedlings. MT and H 2 O 2 improved photosynthetic carbon assimilation, which was primarily attributed to an increase in activity, mRNA expression, and protein levels of RuBPCase and RCA. Meanwhile, MT and H 2 O 2 induced the photoprotection for both PSII and PSI by enhancing the QA's electron transport capacity and elevating protein levels of the photosystems. Moreover, MT and H 2 O 2 significantly upregulated the expression of cold response genes. MT-induced chilling tolerance was attenuated by N', N'-dimethylthiourea (DMTU), a H 2 O 2 specific scavenger. Whereas, the MT synthesis inhibitor (p-chlorophenylalanine, p-CPA) did not influence H 2 O 2 -induced chilling tolerance. The positive regulation of MT on the antioxidant system, photosynthesis and cold response gene levels were significantly attenuated in RBOH1-RNAi plants compared with WT plants. In the current industrial scenario, vanadium (V) as a metal is of great importance but poses a major threat to the ecosystem. In the present study, the effect of a toxic concentration of V, i.e., 10 µM in the soil on growth, photosynthetic pigments, gas exchange characteristics, oxidative stress biomarkers, antioxidants machinery (enzymatic and non-enzymatic antioxidants), ions uptake, proline metabolism, and V uptake in different parts of the plant was investigated with and without the exogenous application of selenium (Se) i.e., 5 µM in V-stressed rice (Oryza sativa L.). Our results depicted that V addition to the soil significantly (P < 0.05) decreased plant growth and biomass, gas exchange attributes, and minerals uptake by O. sativa as compared to the plants grown without the addition of V. However, V toxicity boosted the production of reactive oxygen species (ROS) by increasing the contents of malondialdehyde (MDA), which is the indication of oxidative stress in O. sativa and was also manifested by hydrogen peroxide (H 2 O 2 ) contents to the membrane-bounded organelles. Although activities of various antioxidative enzymes like superoxidase dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) and their gene expression Fe-SOD, POD, CAT, and APX and also non-enzymatic antioxidants like phenolic, flavonoid, and ascorbic acid, anthocyanin contents and also the proline metabolism i.e., proline, pyrroline5-carboxylate, pyrroline-5-carboxylate reductase, and pyrroline-5-carboxylate dehydrogenase were increased due to V stress. Although results also illustrated that the application of Se also decreased V toxicity in O. sativa seedlings by increasing antioxidant capacity and, thus, improved the plant growth and biomass, photosynthetic pigments, gas exchange characteristics, and decreased oxidative stress in the O. sativa seedlings, compared to those plants which were not artificially supplied by Se. Research findings, therefore, suggested that the Se application can ameliorate V toxicity in O. sativa seedlings and result in improved plant growth and composition under metal stress as depicted by balanced exudation of nutrient effluxes. The increasing use of antibiotics has attracted widespread attention to their environmental risks. However, the phototoxicity of sulfonamide antibiotics to plants remain unclear. In this study, the mechanism of the effect of sulfamethoxazole on photosynthesis of pakchoi cabbage (Brassica rapa var. chinensis) was investigated. The results showed that sulfamethoxazole inhibited the growth of pakchoi cabbage and produced photosynthetic toxicity. The growth inhibition rates increased with concentration, the root and shoot weight were 76.02 % and 47.04 % of the control, respectively, with stay-greens phenomenon in 4 mg·L -1 sulfamethoxazole treatment. Chlorophyll precursors (protoporphyrin IX (Proto IX), Mg-proto IX, and protochlorophyllide (Pchlide), 5-aminolevulinic acid (ALA), and porphobilinogen (PBG)) were 1.38-, 1.26-, 1.12-, 1.71-, and 0.96-fold of the control, respectively; photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoids) were 1.26-, 1.39-, and 1.03-fold of the control, respectively. Respiration rate was 271.42 % of the control, whereas the net photosynthetic rate was 50.50 % of the control. The maximum photochemical quantum yield of PSII (F v /F m ), the actual photosynthetic efficiency (Y(II)), the quantum yield of non-regulated energy dissipation (Y(NO)), the apparent electron transfer efficiency of PSII (ETR) under actual light intensity were affected, and chloroplast swelling was observed. Proteomic analysis showed that photosynthesis-related pathways were significantly up-regulated, biological processes such as light response, carbohydrates, and reactive oxygen species were activated. Metabolomic analysis revealed that the tricarboxylic acid cycle (TCA cycle) and carbohydrate catabolism were stimulated significantly (p < 0.05), sugars and amino acids were increased to regulate and enhance the resilience of photosynthesis. The main goal of this study was to investigate the effect of lead (Pb) at various concentrations, as an abiotic factor, and the cross-talk between Pb and pea aphid ( Acyrthosiphon pisum (Harris)) (Hemiptera: Aphididae), as a biotic factor, on the defence responses of pea seedlings ( Pisum sativum L. cv. Cysterski). The analysis of growth parameters for pea seedlings demonstrated that Pb at a low concentration, i.e., 0.025-0.0625 mM Pb(NO 3 ) 2 , caused a hormesis effect, i.e., stimulation of seedling growth, whereas Pb at higher concentrations, i.e., 0.01-0.325 mM Pb(NO 3 ) 2 , inhibited growth, which manifested as the inhibition of length and fresh biomass. The differences in the level of the main defence-related phytohormones, such as abscisic acid (ABA), jasmonic acid (JA) and salicylic acid (SA), and indole-3-acetic acid (IAA)-an auxin stimulating plant cell growth-depended on the dose of Pb, aphid infestation and direct contact of the stress factor with the organ. A high accumulation of soluble sugars in the organs of pea seedlings both at sublethal doses and hormetic doses at early experimental time points was observed. At 0 h and 24 h of the experiment, the hormetic doses of Pb significantly stimulated invertase activities, especially in the roots. Moreover, an increase was observed in the pisatin concentration in pea seedlings growing in the presence of different concentrations of Pb and in the case of cross-talk between Pb and A. pisum in relation to the control. Additionally, a significant induction of the expressions of isoflavone synthase (IFS) and 6α-hydroxymaackiain 3-O-methyltransferase (HMM) genes, which participate in the regulation of the pisatin biosynthesis pathway, in pea seedlings growing under the influence of sublethal 0.5 mM Pb(NO 3 ) 2 and hormetic 0.075 mM Pb(NO 3 ) 2 doses of Pb was noted. The obtained results showed that the response of P. sativum seedlings depends on the Pb dose applied, direct contact of the stress factor with the organ and the duration of contact.

Although plant-derived smoke solutions (SSs) have exhibited growth-promoting properties in various plant species, their potential role in mitigating heavy metal stress, specifically in grapevines, has remained unexplored and unreported. This knowledge gap prompted the present study to evaluate the efficacy of foliar application of SSs derived from vineyard pruning waste at concentrations of 0%, 0.5%, 1%, and 2% in mitigating Cadmium (Cd) phytotoxicity in grape saplings. In our study, cadmium stress was induced by applying 10 mg/kg CdCl 2 to the root area of the saplings, in conjunction with fertilizers. Our findings showed that exposure to Cd toxicity impeded the growth of grapevine saplings, adversely affecting shoot and root length, as well as fresh weight. Furthermore, it resulted in a reduction in chlorophyll content, stomatal conductance, and leaf water content while significantly increasing membrane damage and lipid peroxidation. Notably, the application of 0.5% SS enhanced grapevine sapling growth and alleviated Cd stress-induced damage by more effectively regulating physiological and biochemical responses compared to the control and other concentrations. Based on our results, under Cd stress conditions, the application of 0.5% SS effectively increased chlorophyll content, relative water content (RWC), stomatal conductance (1.79 mmol.m -2 .sn -1 ), and total phenolic content (1.89 mg.g -1 ), whereas it significantly reduced malondialdehyde (MDA) levels and membrane damage (1.35 nmol.g -1 ). Additionally, it significantly elevated the activities of antioxidant enzymes, including superoxide dismutase (SOD) (2.16 U.mg -1 ), catalase (CAT) (1.55 U.mg -1 ), and ascorbate peroxidase (APX) (3.03 U.mg -1 ). Soil salinization, which severely limits crop yield and quality, has become a global environmental and resource issue. Melatonin plays an important role in plant responses to salt stress. Smooth bromegrass is an important forage with excellent feed value and is widely grown in northern and north-west China for pasture and sand binding. However, the physiological and molecular mechanisms underlying exogenous melatonin regulation of salt stress in smooth bromegrass are not clear. This study compared the phenotype, physiological, transcriptome, and metabolome profiles of two varieties with contrasting salt tolerance attributes under salt and melatonin treatment. After melatonin treatment, the catalase (CAT) and ascorbate peroxidase (APX) activity, proline content, actual photochemical efficiency (Y(II)), relative water content, and fresh weight above ground were significantly higher than under salt treatment, while relative conductivity, H 2 O 2 content, and Na + /K + ratio were significantly lower than salt treatment. The transcriptome and metabolite profiling analysis of smooth bromegrass seedlings treated without melatonin under salt stress identified the presence of 22522 differentially expressed genes (DEGs) and 862 differentially expressed metabolites (DEMs) in SS, 17809 DEGs and 812 DEMs in ST, while treated with melatonin under salt stress identified the presence of 7033 DEGs and 177 DEMs in SS, 2951 DEGs and 545 DEMs in ST. Furthermore, in response to salt stress, melatonin may be involved in regulating the correlation between DEGs and DEMs in flavonoid biosynthesis, proline biosynthesis, and melatonin biosynthesis. Pesticides including insecticides are applied in agricultural practices to control insect pests. However, their excessive usage often poses a severe threat to the growth, physiology, and biochemistry of plants. Here, responses of chickpea and greengram seedlings exposed to three fipronil (FIP) concentrations i. e. 100 (1×), 200 (2×) and 300 (3×) μg mL -1 was evaluated under in vitro. Among doses, 3× had a greater negative impact on germination attributes, root-shoot elongation, vigor indices, length ratios, and survival of seedlings. Besides, the morphological distortion in root tips, oxidative stress generation, and cellular death in fipronil-supplemented root seedlings were observed under scanning electron (SEM) and confocal laser scanning (CLSM), respectively. A significant (p ≤ 0.05) and pronounced upsurge in plant stressor metabolites such as proline, malondialdehyde (MDA), electrolyte leakage (EL), hydrogen peroxide (H 2 O 2 ) content, and antioxidants enzymes in plant seedlings further confirmed the fipronil toxicity. In addition, a concentration-dependent decrease in respiration efficiency (RE) and ATP content in FIP-treated seedlings was observed. Reduced mitotic index (MI) and numerous chromosomal anomalies (CAs) in root meristematic cells of seedlings are a clear indication of insecticide-induced cytotoxicity. Furthermore, a dose-dependent increase in DNA damage in root meristematic cells of greengram revealed the genotoxic potential of fipronil. Conclusively, fipronil suggested phyto and cyto-genotoxic effects that emphasize their careful monitoring in soils before application and their optimum addition in soil-plant systems. The unregulated use and improper management of herbicides can cause negative effects on non-target species and promote changes in biological communities. Therefore, the current study is aimed at understanding morphoanatomical responses and effects on seedling development induced by the herbicides glyphosate and saflufenacil in Enterolobium contortisiliquum, a non-target tropical species. The plants were cultivated in a greenhouse and subjected to herbicides at doses of 0, 160, 480, and 1440 g a.e ha -1 for glyphosate, and 0, 25, 50, and 100 g a.i ha -1 for saflufenacil. We conducted visual and morphological assessments over 90 days post-application. Leaf samples were collected 12 days after the application for anatomical analysis, and we also performed a micromorphometric analysis of the leaf tissues. Biomarkers of phytotoxicity were identified in plants exposed to both herbicides, even at the lowest doses, including in leaves without visual symptoms. The main morphological alterations were the decrease in growth, stem diameter, and dry mass. Furthermore, the leaves and stems visually exhibited chlorosis and necrosis. Both herbicides triggered anatomical modifications such as significant changes (p < 0.05) in the thickness of leaf tissues, hypertrophy, cell collapse, and changes in epicuticular waxes. However, the alterations induced by glyphosate were more widespread compared to saflufenacil, encompassing alterations in the root system. We confirmed that the different mechanisms of action of each herbicide and the existence of an underground reserve system in this species are intrinsically linked to the morphological and developmental responses described. Sustainable agriculture has become prime importance to feed growing population. To achieve this goal application of exogenous hormones and signaling molecules are gaining important. In this context, we have investigated potential of ethylene (25 μM ethephon; donor) and H 2 S (10 μM NaHS; donor) in mitigating hexavalent chromium [Cr (VI), 50 μM] toxicity in two pulse seedlings: black bean and mung bean. Cr(VI) declined growth and gas exchange parameters (photosynthetic rate, stomatal conductance, sub cellular CO 2 concentration, and transpiration level) which was accompanied by intracellular accumulation of Cr in both pulse crops and the damaging effect was greater in mung bean seedlings. The suppression in the growth and related parameters was occurred due to higher buildup of oxidative stress markers; O 2 • ‾, H 2 O 2 , lipid peroxidation (as malondialdehyde, MDA equivalents) and membrane injury in leaf and root of both pulse crops. Cr induced disturbance in AsA-GSH cycle (reduction in the activity of glutathione reductase, ascorbate peroxidase, monodehydroascorbate reductase and dehydroascorbate reductase, and the amount of ASA and GSH) could be one of the reasons for greater accumulation of H 2 O 2 . Further, exogenous application of ethylene and H 2 S significantly ameliorated Cr toxicity on growth and photosynthetic activity by significantly lowering the intracellular Cr accumulation and oxidative biomarkers, and also by strengthening the activity of AsA-GSH cycle. The exogenous application of biosynthesis inhibitors of ethylene (AVG) and H 2 S (PAG) caused greater damaging effect on these parameters due to more accumulation of Cr(VI), thereby suggesting that the endogenous levels of these regulators are critical for Cr(VI) tolerance. Interestingly, ET did not rescue adverse effects of Cr(VI) in absence of endogenous H 2 S, while H 2 S could do so even without endogenous ethylene, suggesting that H 2 S played downstream signaling to ethylene in regulating Cr(VI) toxicity. Long-distance signals play a vital role in plant stress response. γ-aminobutyric acid (GABA) has been proposed to be a signal and protects crops against diverse stresses. However, whether GABA acts as a long-distance signal to plant response to stresses remains unknown. Here, we found that the GABA content in cucurbita rootstocks, especially figleaf gourd, was significantly higher than that in cucumber. Figleaf gourd rootstock obviously enhanced cold tolerance and GABA accumulation in roots, xylem sap and leaves of grafting cucumber seedlings. Conversely, GABA synthesis inhibitor 3-mercaptopropionic acid (3-MPA) irrigation was more effective than its foliar application in inhibiting grafting-induced cold tolerance. Moreover, fluorescence microscopy confirmed that GABA can be transported from root to shoot through the xylem when the roots of grafted seedlings were fed with fluorescein isothiocyatate-labeled GABA under normal and cold stress conditions. Importantly, 3-MPA irrigation attenuated grafting-induced cold tolerance, as revealed by a decline in the GABA accumulation, the transcripts of ICE1, CBF1 and COR47, the activities of the antioxidant enzymes, and an increase in stomatal aperture. Microplastics (MPs) are important carriers of various toxic metals and can alter their toxicity pattern in agricultural soil, leading to combined pollution, therefore posing new challenges to soil pollution management and environmental risk assessment. In this study, we observed the internalization of MPs in plants and conducted incubation experiments to evaluated the effects of arsenate (As(V)) alone and in combination with polystyrene (PS) MPs on wheat seedlings (Triticum aestivum L.). Under As(V) alone and combined with PS-MP exposure, dose-dependent toxicity in terms of root and stem elongation and biomass accumulation was observed. Compared with As(V) alone, the presence of PS-MPs reduced the accumulation of As in wheat roots by 11.43-58.91%, but PS-MPs intensified the transport of As to the aboveground parts of wheat, increasing As accumulation in wheat stems by 27.77-1011.54%. This causes more serious mechanical damage and oxidative stress to plant cells, increasing the accumulation of reactive oxygen species and lipid peroxidation in wheat roots and upregulating the activities of antioxidant enzymes such as superoxide dismutase (SOD) and peroxidase (POD). In addition, the co-exposure of As(V) and PS-MPs disrupts the photosynthetic system of wheat leaves and the secretion activities of roots. Therefore, the combination of As(V) and PS-MPs caused greater damage to wheat growth. The widespread use of antibiotics in intensive animal husbandry, and the agricultural utilization of manure from such farms, imposes a significant burden on the environment. Consequently, the effects of antibiotics should be studied not only in animals and humans but also in all components of biocenoses and agrocenoses. In our study, we analyze the impact of four different concentrations of tetracycline present in soil (0, 5, 50, and 500 mg/kg of soil) on the growth and key photosynthesis parameters of pea seedlings: chlorophyll concentration, aminolevulinic acid concentration, aminolevulinic acid dehydrogenase activity, and ribulose bisphosphate carboxylase-oxygenase (RuBisCO) activity. At the lowest tetracycline concentration, chlorophyll content decreased by 13% compared to the control (0 tetracycline), while at the highest antibiotic concentration, it decreased by as much as 27%. Similarly, the decrease in aminolevulinic acid (a chlorophyll precursor) concentration was significant, amounting to 34%. However, the activity of the dehydrogenase enzyme, which consumes this precursor, decreased even more drastically by 51%, indicating significant disturbances in the light phase of photosynthesis. However, the activity of RuBisCO in pea plants subjected to tetracycline was even more severely affected, dropping by 58%, 69%, and 70% in soils with increasing concentrations of tetracycline. The reduction in enzyme activity could only partially be explained by a less pronounced decrease in the quantity of RuBisCO (large subunit) protein, which amounted to 6.5%, 11%, and 35% for tetracycline concentrations of 5, 50, and 500 mg/kg of soil, respectively., (© 2024. The preparation of soilless medium for the growth of seeds based on macroporous scaffolds of chitosan-glutaric acid and urea (CHGAUR) is reported. The CHGAUR scaffold was prepared by the hydrothermal reaction of chitosan with GA in presence of urea. The product was purified using Soxhlet extraction with methanol. CHGAUR was composed of chitosan with a small weight fraction of glutaric acid and urea, which were present in the salt form as confirmed by TGA and FT IR spectroscopy. This suggested that CH is most likely to be physically crosslinked in the scaffold through GAUR links. SEM images confirmed the porous structure of material. Water absorption studies suggested that CHGAUR could absorb ∼4100-5700 % water and could be varied by varying the CH: UR ratio used in the preparation. The percentage porosity of CHGAUR did not vary much with urea content suggesting that significant variation in the micropore structure was not obtained by varying the urea content in the preparation. Mechanical tests revealed that material was soft and flexible and able to retain its original shape even after removing the load. CHGAUR, in purified state, was studied for the germination and seedling growth. Small concentrations of herbicide, such as those found in drift, can affect nontarget plants at different life-history stages including seed germination and seedling emergence as well as seedling growth. Fragmented landscapes, such as those in the northern Great Plains, lead to increased proximity of ecological restoration sites to agricultural lands using herbicides. Germination, emergence, and growth are crucial life-history stages leading to ecological restoration success, but these stages are sensitive to impacts from external factors such as herbicide exposure. A lab germination experiment and a greenhouse emergence experiment were performed to examine the effect of herbicides (2,4-dichlorophenoxyacetic acid [2,4-D], atrazine, and trifluralin) on species used in ecological restorations in the northern Great Plains. Seed germination, seedling emergence, and seedling growth of many study species decreased with exposure to herbicides at different concentrations representative of herbicide drift. At concentrations as low as 0.1% recommended application rate 2,4-D elicited broad effects on final seed germination percentage and germination rate. Atrazine affected seedling emergence and growth for a number of study species at concentrations as low as 10% recommended application rate. Trifluralin affected germination, emergence, and growth of the fewest number of study species. The information gained from these experiments can be used to inform restoration practitioners of best practices and recommended species to use when implementing ecological restoration adjacent to agricultural lands. Soil salinization has been considered as a major environmental threat to plant growth. Different types of salt in saline soil have different effects on germination and seedling growth. Effect of NaCl on germination and seedling establishment in Suaeda liaotungensis have been reported, but its response to alkali stress remains unclear. Our results showed that brown seeds had higher germination rate, however, black seeds had higher germination recovery percentage under alkali stress. Na 2 CO 3 had stronger inhibitory effect on germination and seedling growth than NaHCO 3 . As the concentration of alkali stress increased, the ROS level of brown seeds gradually ascended, while that of black seeds decreased first and then ascended. MDA content of dimorphic seeds significantly increased under alkali stress. The trend of SOD, POD and CAT activity between dimorphic seeds was similar under the same type of alkali stress. Alkali stress enhanced proline content of dimorphic seeds, and dimorphic seeds in NaHCO 3 solution had higher proline content than Na 2 CO 3 solution. Moreover, radicle and shoot tolerance indexes of seedlings in NaHCO 3 solution were significantly higher than that of Na 2 CO 3 solution. Under strong alkali stress, seedlings in NaHCO 3 solution had significantly lower ROS level and MDA content as well as higher antioxidant enzyme activity than Na 2 CO 3 solution. Cadmium (Cd) poses a significant threat to plant growth and the environment. Nano-Fe 3 O 4 is effective in alleviating Cd stress in plants. Elymus nutans Griseb. is an important fodder crop on the Qinghai-Tibetan Plateau (QTP). However, the potential mechanism by which nano-Fe 3 O 4 alleviates Cd stress in E. nutans is not well understood. E. nutans were subjected to single Cd, single nano-Fe 3 O 4 , and co-treatment with nano-Fe 3 O 4 and Cd, and the effects on morphology, Cd uptake, antioxidant enzyme activity, reactive oxygen species (ROS) levels and programmed cell death (PCD) were studied to clarify the regulatory mechanism of nano-Fe 3 O 4 . The results showed that Cd stress significantly decreased the germination percentage and biomass of E. nutans. The photosynthetic pigment content decreased significantly under Cd stress. Cd stress also caused oxidative stress and lipid peroxidation, accumulation of excessive ROS, resulting in PCD, but the effect of nano-Fe 3 O 4 was different. Seed germination, seedling growth, and physiological processes were analyzed to elucidate the regulatory role of nano-Fe 3 O 4 nanoparticles in promoting photosynthesis, reducing Cd accumulation, scavenging ROS, and regulating PCD, to promote seed germination and seedling growth in E. nutans. Salt-induced stress poses a significant barrier to agricultural productivity by impeding crop growth. Presently, environmentalists are dedicated to safeguarding food security by enhancing agricultural yields in challenging environments. Biostimulants play a crucial role in mitigating abiotic stresses in crop production, and among these, plant essential oils (EOs) stand out as organic substances with diverse biological effects on living organisms. Among the natural promoters of plant growth, Rosmarinus officinalis L. essential oil (RoEO) has gained considerable attention. Although the manifold effects of essential oils (EOs) on plant growth have been extensively demonstrated, their impact on salt stress tolerance in durum wheat seedlings remains unexplored. This investigation was undertaken to evaluate the biostimulatory capabilities of RoEO on the durum wheat cultivar "Mahmoudi." The effects of three RoEO concentrations (1, 2.5, and 5 ppm) on seed germination, growth establishment, and the induction of salt resistance under salinity conditions (150 mM NaCl) were tested. At 5 ppm, RoEO enhanced seedlings' tolerance to salinity by improving growth and reducing membrane deterioration and oxidative stress-induced damage. The expression profile analyses of seven stress-related genes (TdNHX1, TdSOS1, TdSOD, TdCAT, TdGA20-ox1, TdNRT2.1, and TdGS) using RT-qPCR showed enhancement of several important genes in durum wheat seedlings treated with 5 ppm RoEO, even under control conditions, which may be related to salt stress tolerance. The results indicate that the application of RoEO suggests a possible alternative strategy to increase salt tolerance in durum wheat seedlings towards better growth quality, thus increasing ROS scavenging and activation of antioxidant defense., (© 2024. Nickel phytotoxicity has been attributed, among others, to oxidative stress. However, little is known about Ni-induced phospholipid modifications, including the oxidative ones. Accumulation of reactive oxygen species (ROS), antioxidative enzyme activities, malondialdehyde and the early lipid oxidation products contents, membrane permeability, phospholipid profile as well as phospholipid unsaturation degree were studied in the 1st and the 2nd leaves of hydroponically grown cucumber seedlings subjected to Ni stress. Compared to the 2nd leaf the 1st one showed stronger visual Ni toxicity symptoms, higher Ni, O 2 .- and H 2 O 2 accumulation as well as greater enhancement in membrane permeability. Enzyme activities were differently influenced by Ni stress, however most pronounced changes were generally found in the 1st leaf. Ni treatment resulted in oxidation of leaf lipids, which was evidenced by appearance of increased contents of MDA and the early produced oxylipins. Among the latter 9-hydroxyoctadecatrienoic acid (9-HOTrE) and 13-hydroxyoctadecatrienoic acid (13-HOTrE) contents showed the most pronounced increase in response to Ni treatment. Exposure to the metal led to the changes in the leaf phospholipid profile and increased degree of phospholipid unsaturation. Gamma irradiation-based mutant creation is one of the most important methods for rice plant mutagenesis breeding and molecular biology research. Although median lethal dose irradiation severely damages rice seedlings, applying brassinolide (BR) can increase the survival rate of irradiated seedlings. In this study, we investigated the effects of soaking seeds in solutions containing different BR concentrations (0.001, 0.01, 0.1, 1.0, and 5.0 μmol/L) and then spraying the resulting seedlings twice with 0.1 μmol/L BR. The combined BR treatments markedly decreased the superoxide anion (O 2 •- ), hydrogen peroxide (H 2 O 2 ), and malondialdehyde contents but increased the chlorophyll content. An appropriate BR treatment of gamma-irradiated samples substantially increased the activities of the antioxidant enzymes superoxide dismutase, peroxidase, and ascorbate peroxidase as well as the proline, ascorbic acid, and glutathione contents in rice seedling shoots. The BR treatment also promoted the growth of seedlings derived from irradiated seeds and increased the shoot and root fresh and dry weights. Most notably, soaking seeds in 0.01 or 0.1 μmol/L BR solutions and then spraying seedlings twice with 0.1 μmol/L BR significantly increased the final seedling survival rate and decreased mutant loss. The study results suggest that exogenous BR treatments can protect rice seedlings from gamma irradiation stress by enhancing antioxidant metabolism.

Background: Drought stress markedly constrains plant growth and diminishes crop productivity. Strigolactones (SLs) exert a beneficial influence on plant resilience to drought conditions. Nevertheless, the specific function of SLs in modulating cotton's response to drought stress remains to be elucidated., Results: In this study, we assess the impact of exogenous SL (rac-GR24) administration at various concentrations (0, 1, 5, 10, 20 µM) on cotton growth during drought stress. The findings reveal that cotton seedlings treated with 5 µM exogenous SL exhibit optimal mitigation of growth suppression induced by drought stress. Treatment with 5 µM exogenous SL under drought stress conditions enhances drought tolerance in cotton seedlings by augmenting photosynthetic efficiency, facilitating stomatal closure, diminishing reactive oxygen species (ROS) generation, alleviating membrane lipid peroxidation, enhancing the activity of antioxidant enzymes, elevating the levels of osmoregulatory compounds, and upregulating the expression of drought-responsive genes. The suppression of cotton SL biosynthesis genes, MORE AXILLARY GROWTH 3 (GhMAX3) and GhMAX4b, impairs the drought tolerance of cotton. Conversely, overexpression of GhMAX3 and GhMAX4b in respective Arabidopsis mutants ameliorates the drought-sensitive phenotype in these mutants., Conclusion: These observations underscore that SLs significantly bolster cotton's resistance to drought stress., (© 2024. Drought stress is an abiotic stressor that impacts photosynthesis, plant growth, and development, leading to decreased crop yields. Sodium hydrosulfide (NaHS), an exogenous additive, has demonstrated potential regulatory effects on plant responses to polyethylene glycol-induced drought stress in tobacco seedlings. Compared to the control, drought stress induced by 15 g/L PEG-6000 significantly reduced several parameters in tobacco seedlings: shoot dry weight (22.83%), net photosynthesis (37.55%), stomatal conductance (33.56%), maximum quantum yield of PSII (Fv/Fm) (11.31%), photochemical quantum yield of PSII (ΦPSII) (25.51%), and photochemical quenching (qP) (18.17%). However, applying NaHS, an H 2 S donor, mitigated these effects, ultimately enhancing photosynthetic performance in tobacco seedlings. Furthermore, optimal NaHS concentration (0.4 mM) effectively increased leaf stomatal aperture, relative water content (RWC) and root activity, as well as facilitated the absorption of N, K, Mg and S. It also enhanced the accumulation of soluble sugar and proline content to maintain osmotic pressure balance under drought stress. Compared to drought alone, pretreatment with NaHS also bolstered the antioxidant defense system in leaves, leading to 22.93% decrease in hydrogen peroxide (H 2 O 2 ) content, a 22.19% decrease in malondialdehyde (MDA) content and increased activities of ascorbate peroxidase (APX) by 28.13%, superoxide dismutase (SOD) by 17.07%, peroxidase (POD) by 46.99%, and catalase (CAT) by 65.27%. Consequently, NaHS protected chloroplast structure and attenuated chlorophyll degradation, thus mitigating severe oxidative damage. Moreover, NaHS elevated endogenous H 2 S levels, influencing abscisic acid (ABA) synthesis and the expression of receptor-related genes, collaboratively participating in the response to drought stress. Overall, our findings provide valuable insights into exogenous NaHS's role in enhancing tobacco drought tolerance. Burdock fructooligosaccharide (BFO) is fructose with a low polymerization degree, which could improve the immunity to pathogens, quality, and stress resistance of vegetables. Still, there are no studies on applying BFO in Chinese cabbage. In this study, the effects of exogenous BFO sprayed with different concentrations (0, 5, 10, 20, 30 g·L -1 ) on the growth and soluble sugar content of Chinese cabbage seedlings were determined. The result showed that 10 g·L -1 was the appropriate spraying concentration. Based on metabolome analysis, a total of 220 differentially accumulated metabolites (DAMs) were found, among which flavonoid metabolites, glucosinolate metabolites, and soluble sugar-related metabolites were the key metabolites involved in improving the quality of Chinese cabbage caused by BFO. Further combination analysis with transcriptome, trans-cinnamate 4-monooxygenase ( CYP73A5 ), and chalcone synthase 1 ( CHS1 ) were more closely associated with the DAMs of flavonoid biosynthesis. Sulfotransferases 18 ( SOT18 ), Branched-chain amino acid amino transferases 6 ( BCAT6 ), and cytochrome P450 monooxygenase ( CYP83A1 ) were the key genes in glucosinolate biosynthesis. Hexokinase ( HxK1 ), beta-glucosidase 8 ( BGL08 ), invertase 3 ( INV3 ), beta-glucosidase 3B ( BGL3B ), and sucrose phosphate synthase 1 ( SPS1 ) were significantly upregulated, potentially playing crucial roles in the soluble sugar metabolism. In conclusion, these results provided an understanding of the effects of BFO on the expression of genes and the accumulation of metabolites related to quality formation in Chinese cabbage.

Background: Balancing nutrient application is crucial for plant growth. However, excessive fertilizer use, especially imbalanced applications of macronutrients such as phosphate (P), can hinder plant uptake of micronutrients. Balanced P and zinc (Zn) are vital for apple yield and quality, and apple trees are highly sensitive to deficiencies in these nutrients. Therefore, this study was conducted in May 2022, employed a sand culture experiment to investigate the effects of varying P and Zn levels on the growth phenotype, photosynthetic capacity, antioxidant enzyme activity, sugar composition, endogenous hormone levels, and nutrient absorption and utilization of M9-T337 seedlings. Three levels of P (low, medium, high) and three levels of Zn (low, medium, high) were combined to create a total of nine distinct treatment., Results: The results indicate that combined P and Zn fertilization at various levels exerts either synergistic or antagonistic effects on the growth, nutrient absorption, and utilization of M9-T337 seedlings. Compared to low and medium levels of P, a combination of high P (4 mmol·L -1 ) and an adequate amount of Zn significantly enhanced plant growth, root system development, and the microstructure of leaves. Notably, seedlings treated with high P and low Zn (HPLZn) reached a height 1.54 times that of the medium P and medium Zn (MPMZn, control). Physiological indicators under HP conditions revealed significant increases in antioxidant enzyme activity, leaf water retention, photosynthetic pigment concentration, osmotic adjustment substances, and the contents of glucose, sucrose, fructose, endogenous hormones, as well as P and Zn accumulation in the leaves, compared to the control. However, an increase in Zn application led to a declining trend in these parameters. Specifically, the HPLZn treatment exhibited substantial increases in Net photosynthetic rate (Pn), Total chlorophyll (Chl a + b), glucose, fructose, sucrose, and Auxin(IAA), with increments of 7.12%, 27.32%, 11.40%, 23.20%, 16.67%, and 55.11%, respectively, compared to the control., Conclusion: Based on the comprehensive ranking from principal component analysis, the combination of HP ( 4 mmol·L -1 ) and LZn (0.5 µmol·L -1 ) was found to be the most effective in enhancing the antioxidant capacity, sugar accumulation, osmotic regulation ability, photosynthetic capacity, endogenous hormone levels, as well as P and Zn nutrient absorption and utilization in M9-T337 seedlings., (© 2024. Nano-priming is a relatively new seed treatment technique using metal and metal oxide nanoparticles (NPs), and such application of NPs may support the plants' immunity. Recently we have shown that the that biologically synthesized silver nanoparticles (bio-AgNPs) used as short-term foliar treatment protect pea seedlings against D. pinodes and F. avenaceum . In the present study, the protection of peas against both fungal pathogens via seed priming with bio-AgNPs was analyzed. Moreover, the changes in the polar metabolic profiles of the seedlings caused by priming and infection were also compared. Seed priming with bio-AgNPs at concentrations of 50 and 100 mg/L considerably reduced the symptoms and infection levels of both pathogens by over 70% and 90% for F. avenaceum and D. pinodes , respectively. Pathogens infection and nano-priming affected the metabolic profile of pea seedlings. The major changes in the primary metabolism were observed among carbohydrates and amino acids. In turn, this may result in changes in the expression and accumulation of secondary metabolites. Therefore, further investigation of the effect of nano-priming should focus on the changes in the secondary metabolism.

Heavy metal pollution causes severe abiotic stress in cereal crops around the world. This study investigated the effects of different concentrations (0, 100, 200, and 300 mg·kg -1 ) of nickel, lead, and copper stress on the growth and biochemical responses of Aegilops tauschii seedlings, to provide a reference for research on the mechanism of invasion and screening potential sources of wheat tolerance genes. The results showed that nickel, lead, and copper stress caused a significant decrease in the contents of chlorophyll a, chlorophyll b, and chlorophyll (a + b) in A. tauschii, thereby inhibiting photosynthesis to different degrees and hindering seedling growth, which was reflected in significant reductions in plant height and root length, with the most notable effect observed under stress by 300 mg·kg -1 lead. As the concentration of heavy metals increased, the activities of antioxidant enzymes (SOD, POD, and APX), non-enzymatic antioxidants (GSH and AsA), and the contents of osmotic regulatory substances (proline and soluble proteins) in A. tauschii significantly increased. Additionally, heavy metal stress increased H 2 O 2 and TBARS levels. However, when the nickel, lead, and copper concentrations reached 300 mg·kg -1 , no significant differences were found in H 2 O 2 or TBARS levels compared to those in the CK group. To summarize, A. tauschii can mitigate the accumulation of ROS and membrane lipid peroxidation caused by heavy metal stress through self-regulation, thus exhibiting a certain degree of tolerance to stress caused by different concentrations of nickel, lead, and copper. Root cap cuticles (RCCs), comprising mainly very-long-chain fatty acids (VLCFAs), promote salt tolerance by preventing ion influx. Glycosylphosphatidylinositol-anchored lipid transfer protein (LTPG)1 and LTPG2 participate in VLCFA deposition in the extracellular region, aiding RCC formation in the lateral roots. In this study, we investigated whether LTPG1 and LTPG2 have similar functions in the primary roots of young Arabidopsis thaliana. Phenotypic analyses, fluorescence microscopy, and quantitative real-time reverse transcription polymerase chain reaction confirmed that NaCl exposure induced LTPG1 and LTPG2 expression and promoted RCC formation in young primary roots. The loss of RCC in the ltpg1 and ltpg2 mutants resulted in increased NaCl sensitivity of root elongation. NaCl also upregulated the expression of several NaCl-responsive genes in ltpg1 and ltpg2. This study aimed to evaluate the potential of zinc oxide nanoparticles (ZnO NPs) in the biofortification of lentil seedlings and subsequently improve the Zn status in rats. In the first phase of the study, the effects of various ZnO NPs concentrations (0, 10, 20, 40, 80, and 160 ppm) on the lentil growth, Zn accumulation, and other physiological parameters were investigated. Subsequently, the rats were fed ZnO NP-biofortified lentil seedlings (20 and 160 ppm) to assess their impact on animal health and Zn status. The results highlighted a concentration-dependent response of lentil seedlings to ZnO NPs, with optimal growth observed at 20 ppm, whereas higher concentrations inhibited lentil growth. Pigment and biochemical analyses revealed a complex interplay between chlorophyll, carotenoids, soluble sugars, and proteins with distinct responses to nanoparticle concentrations. Elevated levels of hydrogen peroxide and malondialdehyde of lentil seedlings at high concentrations of ZnO NPs suggest oxidative stress, countered by the upregulation of antioxidant enzymes and increased phenolic compounds. On the other hand. animal studies have showed that ZnO NP-biofortified lentil seedlings enhance serum zinc and magnesium levels in rats without affecting body weight. While serum triglyceride levels of rats decreased in both treatment groups, an elevation in creatinine and a marked increase in aspartate aminotransferase (AST) levels were observed at a higher ZnO NP concentration (160 ppm), indicative of potential kidney and liver stress. Paradoxically, serum iron levels were lower in all groups consuming lentil seedlings than in the control group, suggesting a potential interaction between lentil components and iron metabolism. These findings suggest that ZnO NP-biofortified lentils may be a promising approach to enhance Zn nutrition; however, further investigation is needed to optimize ZnO NPs concentration and assess long-term safety., (© 2024. Key Message: A plant growth regulator, 5-aminolevulinic acid, enhanced the saline-alkali tolerance via photosynthetic, oxidative-reduction, and glutathione metabolism pathways in pepper seedlings. Saline-alkali stress is a prominent environmental problem, hindering growth and development of pepper. 5-Aminolevulinic acid (ALA) application effectively improves plant growth status under various abiotic stresses. Here, we evaluated morphological, physiological, and transcriptomic differences in saline-alkali-stressed pepper seedlings after ALA application to explore the impact of ALA on saline-alkali stress. The results indicated that saline-alkali stress inhibited plant growth, decreased biomass and photosynthesis, altered the osmolyte content and antioxidant system, and increased reactive oxygen species (ROS) accumulation and proline content in pepper seedlings. Conversely, the application of exogenous ALA alleviated this damage by increasing the photosynthetic rate, osmolyte content, antioxidant enzyme activity, and antioxidants, including superoxide dismutase, catalase, glutathione reductase, and glutathione peroxidase, and reducing glutathione to reduce ROS accumulation and malonaldehyde content. Moreover, the transcriptomic analysis revealed the differentially expressed genes were mainly associated with photosynthesis, oxidation-reduction process, and glutathione metabolism in saline-alkali stress + ALA treatment compared to saline-alkali treatment. Among them, the change in expression level in CaGST, CaGR, and CaGPX was close to the variation of corresponding enzyme activity. Cotton seeds have poorer germination than other crops because of their high sensitivity towards insect pests and other biotic and abiotic stresses during the germination process. In the present study, inorganic bulk and nano nutrients of zinc oxide (ZnO) and titanium oxide (TiO 2 ) nanoparticles were synthesized using the chemical reduction method and invigourated with cotton seeds. The characterization of nanoparticles was done by FESEM, HRTEM, UV/Vis analysis and FTIR. The delinted and fuzzy seeds of two American cotton varieties (H 1300 and H 1098-i) were nano-primed for 10 h with zinc oxide nanoparticles (ZnONPs) @ 400 ppm and titanium dioxide nanoparticles (TiO 2 NPs) @ 100 ppm. After nanoparticle invigouration, the seeds were analyzed for various parameters at different intervals (0 months, 3 months, 6 months, 9 months and 12 months) such as germination percentage, seedling length, seedling dry weight, electrical conductivity, dehydrogenase activity, antioxidant enzyme activity. The results indicated that that different storage periods and nanopriming treatments had significant effects on all seed quality parameters except the effect of nanopriming treatments on germination percentage (excluding delinted seeds of H 1098-i). It is also revealed that the interaction effect of nanopriming treatment and storage period was non-significant on all parameters except EC. Maximum reduction in seed quality parameters was observed in control treatment and minimum was found when seeds were nanoprimed with ZnONPs @ 400 ppm. The differences in the response for both NPs can be attributed to their surface charge, and concentration used. Sustainable plant production in soil polluted with heavy metals requires that novel strategies are developed for the benefit of humans and other living things. Cadmium (Cd) is a common heavy metal pollutant for plants, and there is limited information on the use of exogenous bio-regulators to reduce the accumulation and toxic effects of Cd pollution in plants. Choline is an endogenous quertarnary amine that is known to improve stress tolerance in plants, while its mechanism of action in certain conditions is yet to be determined. This study investigated the effects of foliar choline supplementation (10 mM) on Solanum lycopersicum seedlings exposed to Cd application (50 mg/L in soil). The seedlings were randomized to five groups: Control (E1), Cd stress (E2), Choline supplementation after Cd stress (E3), Choline (E4), and Choline supplementation before Cd stress (E5). Following the applications, the Cd content, growth and development parameters (chlorophyll content, fresh and dry weight), oxidative stress parameters (H 2 O 2 and MDA contents), as well as antioxidative defense system (SOD, GSH, AsA, and TPC contents) were analyzed. Choline supplementation after Cd stress reduced the enhanced Cd content in roots by 38% but did not alter it in leaves (p > 0.05) compared to the Cd group. Choline supplementation before Cd stress decreased Cd content both in roots by 87.5% and in leaves by 50%. Choline supplementation after and before Cd stress increased fresh and dry weights in both roots and leaves. While the Cd group (E2) increased the H 2 O 2 level and SOD activity, no remarkable change was observed in H 2 O 2 levels in all choline supplementations (E3, E4, E5). Therefore, lipid peroxidation (MDA) was not observed in choline supplementation before Cd stress (E5), however, when the choline was applied after Cd stress (E3) MDA content was reduced by 40% compared with the Cd stress group (E2). Choline supplementations after and before Cd stress (E3, E5) increased AsA content by 30%, while the Cd group (E2) decreased it by 60% compared with the control group (E1). Choline supplementations before Cd stress (E5) increased TPC by 33%, while the Cd group (E2) decreased it by 18%, moreover, when choline was applied after Cd stress (E3), no change was observed compared to the control group. These data suggest that choline prevents inhibition of plant growth due to Cd toxicity by reducing Cd uptake. The problem of potentially toxic metal pollution is increasingly acute with the development of human society. In this study, we investigated the remediation of nickel (Ni) and cadmium (Cd) co-contamination through inoculating rice with three new-isolated Ni- and Cd-resistant plant growth-promoting rhizobacteria (PGPR) Y3, Y4, and Y5. These three strains possessed growth-promoting properties, including 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, the ability of indoleacetic acid (IAA) production, phosphate solubilization, siderophores production, and exopolysaccharide (EPS) development. According to 16S rDNA sequence homology, strains Y3, Y4, and Y5 were identified as Pseudomonas sp., Chryseobacterium sp., and Enterobacter sp., respectively. Based on the results of rice germination experiments conducted under combined toxicity, we set the contamination concentrations for Ni 2+ at 20 μg mL -1 and Cd 2+ at 40 μg mL -1 . Then we conducted potting experiments at these concentration levels to study the effects of strains Y3, Y4, and Y5 on rice growth under synergistic Ni and Cd stress. The results indicated that the inoculated strains Y3, Y4, and Y5 were effective in promoting the growth of rice seedlings under the combined stress of Ni and Cd, and conferring tolerance to Ni and Cd by increasing the antioxidant enzyme activities of the seedlings. Among them, strain Y3 exhibited stronger ACC deaminase activity, IAA production capacity, and EPS production capacity, showing the most pronounced growth-promoting effect on rice. It was demonstrated that after inoculation with strain Y3, the germination rate of rice seeds increased by 43 %, the fresh weight of stems improved by 35 %, and the chlorophyll content enhanced by 70 % and other growth-promoting phenomena. Studies on the toxicity of micro- and nanomaterials in plants have primarily focused on their intrinsic effects. However, there is often oversight when considering the potential perceptual responses that plants may exhibit in response to these materials. In this investigation, we assessed the impact of three commercially available persistent luminescence materials (PLMs) that emit red, green, or blue light under various environmental conditions. We subjected rice (Oryza sativa L.), a short-day plant, to nine distinct treatments, including exposure to particles in isolation, their nocturnal afterglow, or a combination of both. We thoroughly examined rice seedling morphology, photosynthesis patterns, metabolite dynamics, and flowering gene expression to determine the biological responses of plants to these particles. These findings demonstrated that PLMs stably interact with rice, and their emitted afterglow precisely matches the perceptual bandwidth of rice photoreceptors. Notably, the nocturnal afterglow from the red and blue PLMs enhanced the vegetative growth of rice seedlings while inhibiting their reproductive development. The blue PLMs exhibited the most pronounced positive effects, while the red PLMs exhibited inhibitory effects. When exposed to a combination of red and blue PLMs, rice displays enhanced growth and development. The observed alterations in the expression patterns of genes responsible for flowering supported these effects. We concluded that PLMs influence rice growth and development due to their inherent properties and intermittent illumination during dark periods. Soil deterioration is a major cause of poor agricultural productivity, necessitating sufficient nutrient inputs like fertilizers and amendments for sustainable use. As one such strategy, the current study evaluates the potential of Sargassum wightii, a brown seaweed extract, as an osmopriming agent to improve seed germination, early establishment, and competent seedling performances in acidic soil. The elemental makeup of seaweed extract (BS) showed that it included major plant macro (Potassium, Nitrogen and Phosphorous), as well as micronutrients (Magnesium and Iron) and trace elements (Zinc, Copper, and Molybdenum). While seed germination was impacted by H + ion toxicity, seeds primed with BS emerged earlier and showed a higher germination percentage (98.2%) and energy (92.4%). BS treatments enhanced seedling growth by 63% and had a positive effect on root growth (68.2%) as well as increases in root surface area (10%) and volume (67.01%). Stressed seedlings had 76.39% and 63.2% less carotenoid and chlorophyll, respectively. In seedlings treated with BS, an increase in protein and Total Soluble Sugars content of 14.56 and 7.19%, respectively, was seen. Fourier Transform-Infra Red analysis of postharvest soil indicated improved soil health with absorbance corresponding to enhanced soil water holding capacity and organic matter. Increased abscisic acid synthesis rate and associated antioxidant enzyme system (Malondialdehyde, Glutathione peroxidases and ascorbate peroxidase) activation, along with enhanced H + adenosine triphosphate-ase and glutathione activities, help ameliorate and deport H + ions from cells, scavenge Reactive Oxygen Species, thus protecting cells from injury. The novel brominated flame retardant decabromodiphenyl ethane (DBDPE) has biological toxicity, persistence, long-range migration and bioaccumulation ability. However, there is currently little research on the phytotoxicity of DBDPE in plants. The perennial herbaceous plant tall fescue (Festuca elata Keng ex E. B. Alexeev) was selected as the model organism for use in seed germination experiments, and the phytotoxicity of DBDPE in the soil of tall fescue was studied. The results indicated that DBDPE had a significant effect on the germination and growth of tall fescue seedlings. Citric acid reduced the stress caused by DBDPE in plants, effectively alleviating the phytotoxicity of DBDPE in tall fescue. The root vitality and protein content significantly increased after the application of citric acid, increasing by 74.93-183.90%, 146.44-147.67%, respectively. The contents of proline and soluble sugars significantly decreased after the application of citric acid, decreasing by 45.18-59.69% and 23.03%, respectively (P < 0.05). There was no significant difference in superoxide dismutase (SOD) or peroxidase (POD) activity in tall fescue seedlings, and the catalase (CAT) activity and malondialdehyde (MDA) content were significantly lower after the application of citric acid, decreasing by 64.62-67.91% and 29.10-49.80%, respectively (P < 0.05). Tall fescue seedlings bioaccumulated DBDPE, with biological concentration factors (BCFs) ranging from 4.28 to 18.38 and transfer factors (TFs) ranging from 0.43 to 0.54. Methylglyoxal (MG), a highly reactive and cytotoxic α-oxoaldehyde compound, can over-accumulate under abiotic stress, consequently injuring plants or even causing death. Glyoxalase I (GLYI), the first enzyme of the glyoxalase pathway, plays multiple roles in the detoxification of MG and in abiotic stress responses. However, the GLY1 gene in maize has been little studied in response to abiotic stress. In this study, we screened a glyoxalase I gene ( ZmGLYI-8 ) and overexpressed in Arabidopsis . This gene was localized in the cytoplasm and can be induced in maize seedlings under multiple stress treatments, including salt, drought, MG, ABA, H 2 O 2 and high temperature stress. Phenotypic analysis revealed that after MG, salt and drought stress treatments, overexpression of ZmGLYI-8 increased the tolerance of transgenic Arabidopsis to MG, salt and drought stress. Furthermore, we demonstrated that the overexpression of ZmGLYI-8 scavenges accumulated reactive oxygen species, detoxifies MG and enhances the activity of antioxidant enzymes to improve the resistance of transgenic Arabidopsis plants to salt and drought stress. In summary, this study preliminarily elucidates the molecular mechanism of the maize ZmGLYI-8 gene in transgenic Arabidopsis and provides new insight into the breeding of salt- and drought-tolerant maize varieties.

Carbon Dots (CDs) were synthesized from peanut shells (PNS) through pyrolysis and characterized using FTIR, XRD, HRTEM and BET analysis revealing an average size of 2-5 nm with amorphous nature. Synthesized PNS-CDs was employed both as priming and foliar agent for enhancing seed quality and crop productivity in blackgram (Vigna mungo L. Hepper). Different concentrations ranging from 50, 100, 200, 300, 400, 500 and 1000 ppm was used for seed priming and 5, 10, 15, 20, 25, 50, 75 and 100 ppm were given as foliar spray on 30 th and 45 th days after sowing (DAS). On accounting the best seed priming and foliar spray concentrations, field trial was conducted to validate the optimistic effect of PNS-CDs on blackgram crop productivity. Results revealed that priming with 200 ppm for 3 h exhibited maximum seed imbibition (54%), germination (88%) and vigour index (3165). Whereas, foliar spray with 50 ppm expressed significant improvement in leaf area index (2.6), total chlorophyll (2.70 mg/g), total soluble protein (71 mg/g), Number of nodules/plant (138), seed yield/plant (8.7 g) and 100 seed weight (5 g). The impact of PNS-CDs treatments resulted in increased photosynthetic rate (12.45 µmol CO 2 m -2 s -1 ), transpiration rate (3.13 mmol H 2 O/m -2 s -1 ), stomatal conductance (0.55 mol H 2 O/m -2 s -1 ) and internal leaf CO 2 concentration (652 µmol CO 2 m -2 s -1 ) which ultimately enhanced the photosynthetic efficiency of plants. It has also exhibited a promising effect on the resultant seed in which the combination seed priming (200 ppm) followed by foliar spray (50 ppm) recorded maximum 100 seed weight (4.19 g), germination (97%) and vigour index (3019). Poor seedling germination and growth can result in large financial losses for farmers, thus, there is an urgent need for sustainable agricultural techniques to enhance seed germination and early growth. As an outcome, sustainable agriculture-which emphasizes the smart and effective utilization of resources-has gained popularity worldwide. At numerous levels, the field of nanotechnology is capable of significant benefit in achieving sustainable agricultural practices. Zinc oxide nanoparticles (ZnO NPs) have been shown to have biostimulatory properties and serve as effective solutions for addressing environmental and biotic stressors. The purpose of this study, investigating Salvadora persica halophytic leaf extract -synthesized zinc oxide nanoparticles (S-ZnONPs) as nano-priming agents to ensure okra seeds germinated under stress-free conditions. From an application perspective, we examined the effect of seed priming with varying concentrations of S-ZnO NPs (0, 20 and 40 ppm) for 18 and 24 h of soaking. Results indicated that the germination rate of hybrid variety improved with 20 ppm at 18 h, increasing by 58.22%, while mean germination time reduced by 24.62%. An enhancement trend was observed in the shoot, root length, shoot and root fresh weight, shoot and root dry weight of hybrid variety at 20ppm with 18 h priming by 34.2, 84.3, 80.2, 47.4, 50.3, and 36.2%, respectively. However, chlorophyll pigments chl a, chl b, and carotenoids was significantly raised in desi variety by 42.4, 79.31, and 142.29% with 20 ppm at 18 h priming. Hydrogen per oxide decreased up to 87.8% with 40 ppm at 24 h in hybrid variety, while, in desi variety H 2 O 2 was reduced 88.3% with 20 ppm at 24 h. Non enzymatic antioxidant activities such as ascorbic acid, was highly increased 130.6% in hybrid at 24 h priming with 20 ppm dose. Flavonoids raised in same variety by 166.1% with 20 ppm at 18 h. Proline content was increased by 144.5% with 40ppm at 18 h. Moreover, Antioxidant enzymes, superoxide dismutase, peroxidase and catalase were significantly increased in both varieties with both levels of S-ZnO NPs and priming time. This cost-effective and environmentally safe technique to produce nanoparticles of different halophytic plants can maximize resource utilization, supporting sustainable agriculture by minimizing adverse environmental effects without compromising efficiency., (© 2024. Low temperature is one of the environmental factors that restrict the growth and geographical distribution of Brassica. To investigate the effects of exogenous calcium and calcium inhibitor on the ability of winter turnip rapeseed (Brassica rapa L.) to withstand low temperatures (4℃), we used a strong cold-resistant variety Longyou 7 (L7) and a weak cold-resistant variety Longyou 99 (L99) as the materials. The seedlings were treated with CaCl 2 (20 mmol·L -1 ) and calcium inhibitor LaCl 3 (10 mmol·L -1 ) at 0 h (CK), 6 h, 12 h, 24 h and 48 h after 4℃ treatments. Physiological characteristics, Ca 2+ flux and Ca 2+ concentration in roots after treatments were analyzed. Results illustrated that under 4℃ treatment, activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) increased by both CK and exogenous CaCl 2 treatments. Contents of soluble protein (SP) and proline (Pro) increased, while contents of malondialdehyde (MDA) decreased, resulting in reduced membrane lipid peroxidation. But enzyme activity decreased and MDA content increased following treatment with exogenous LaCl 3 . The rate of Ca 2+ flow showed a higher uptake in L7 roots compared with L99. L99 showed Ca 2+ efflux with a rate of 30.21 pmol‧cm -2 ‧s -1 , whereas L7 showed short efflux then returned to influx. Calcium ion content in roots decreased in both cultivars after CaCl 2 treatment. Results of RNA-seq revealed that genes were differentially expressed in response to low temperatures, hormones, photosystem II, chloroplasts, DNA replication, ribosomal RNA processing, and translation. This study found significant expression genes related to cellular signal transduction (MAPK signaling pathway) and material metabolism (nitrogen metabolism, glycerol ester metabolism).It was also analyzed by WGCNA that two modules had the strongest correlation with physiological indicators. Salt stress can adversely affect global agricultural productivity, necessitating innovative strategies to mitigate its adverse effects on plant growth and yield. This study investigated the effects of exogenous salicylic acid (SA), grafting (G), and their combined application (GSA) on various parameters in tomato plants subjected to salt stress. The analysis focused on growth characteristics, photosynthesis, osmotic stress substances, antioxidant enzyme activity, plant hormones, ion content, and transcriptome profiles. Salt stress severely inhibits the growth of tomato seedlings. However, SA, G, and GSA improved the plant height by 22.5%, 26.5%, and 40.2%; the stem diameter by 11.0%, 26.0%, and 23.7%; the shoot fresh weight by 76.3%, 113.2%, and 247.4%; the root fresh weight by 150.9%, 238.6%, and 286.0%; the shoot dry weight by 53.5%, 65.1%, and 162.8%; the root dry weight by 150.0%, 150.0%, and 166.7%, and photosynthesis by 4.0%, 16.3%, and 32.7%, with GSA presenting the most pronounced positive effect. Regarding the osmotic stress substances, the proline content increased significantly by more than 259.2% in all treatments, with the highest levels in GSA. Under salt stress, the tomato seedlings accumulated high Na + levels; the SA, G, and GSA treatments enhanced the K + and Ca 2+ absorption while reducing the Na + and Al 3+ levels, thereby alleviating the ion toxicity. The transcriptome analysis indicated that SA, G, and GSA influenced tomato growth under salt stress by regulating specific signaling pathways, including the phytohormone and MAPK pathways, which were characterized by increased endogenous SA and decreased ABA content. The combined application of grafting and exogenous SA could be a promising strategy for enhancing plant tolerance to salt stress, offering potential solutions for sustainable agriculture in saline environments.

Synthesis methods of carbon nanomaterials have been developed vigorously in recent years, among which a simple, green, and mature approach is of more research significance. Carbon nanomaterials have depicted an impact on the growth and development of plants. In this study, a new type of carbon nanomaterial, superhydrophilic carbon aerogel (CA), was synthesized via a hydrothermal process using carbohydrates and water-soluble polymers as raw materials. Characterized by scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, powder X-ray diffraction, and N 2 adsorption analysis, the exemplified CA presented to have porous three-dimensional network structure composed of individual particles with diameters of 25 nm, with reactive surface, single composition, high specific surface area (89.94 m 2 ·g -1 ), and wide range of density variation. The 9 mg·mL -1 CA suspension had a significant positive effect on the root growth of wheat seedlings, with promoted root elongation (about 67.17% longer) and root diameter (about 28.95% thicker) compared with those of the control group. The cytological results suggested that CA treatment triggered the propagation of meristematic cells, and the increased number of meristematic cells (65.79% more than the control group) led to enhanced root growth by upregulated expression of related phytohormone genes in wheat seedlings.

Deep sowing, as a method to mitigate drought and preserve soil moisture and seedlings, can effectively mitigate the adverse effects of drought stress on seedling growth. The elongation of the hypocotyl plays an important role in the emergence of maize seeds from deep-sowing stress. This study was designed to explore the function of exogenous methyl jasmonate (MeJA) in the growth of the maize mesocotyl and to examine its regulatory network. The results showed that the addition of a 1.5 μ mol L -1 MeJA treatment significantly increased the mesocotyl length (MES), mesocotyl and coleoptile length (MESCOL), and seedling length (SDL) of maize seedlings. Transcriptome analysis showed that exogenous MeJA can alleviate maize deep-sowing stress, and the differentially expressed genes (DEGs) mainly include ornithine decarboxylase, terpene synthase 7, ethylene responsive transcription factor 11, and so on. In addition, candidate genes that may regulate the length of maize hypocotyls were screened by Weighted Gene Co-expression Network Analysis (WGCNA). These genes may be involved in the growth of maize hypocotyls through transcriptional regulation, histones, ubiquitin protease, protein binding, and chlorophyll biosynthesis and play an important role in maize deep-sowing tolerance. Our research findings may provide a theoretical basis for determining the tolerance of maize to deep-sowing stress and the mechanism of exogenous hormone regulation of deep-sowing stress.

Eggplant (Solanum melongena) is moderately sensitive to salinity. Seed priming and exogenous supplementation are technique that enhances germination, growth, and crop yield by overcoming salt stress. Therefore, this study was designed to understand the role of seed priming and copper (Cu) supplementation in modulating salt tolerance in eggplant. When exposed to salt stress, eggplant seedlings showed significantly higher Na +  content, an increased Na/K ratio, prolonged mean germination time, higher relative water loss, more days to flower bud initiation and first flowering, along with decreased germination rate, growth factors, water content, photosynthetic pigments, ionic contents (K + , Ca 2+ , Mg 2+ ), and yield. The results demonstrated that the germination rate, final germination percentage, germination index, germination energy, and seed vigor index significantly improved, while the mean germination time decreased in Cu-primed seeds. The results also revealed that Cu supplementations increased seedling traits, leaf water content, photosynthetic pigment contents, ionic contents (K + , Ca 2+ , and Mg 2+ ), and yield while decreasing the contents of Na + , and Na/K ratio, mean germination time, relative water loss, days to flower bud initiation, and days to 1st flowering under salt stress. Germination of seeds, seedlings growth traits, plant water status, plant pigments, yield, and ionic contents with the NaCl and Cu treatments were found to substantially interact with each other according to both hierarchical clustering and PCA. Overall, Cu seed priming and exogenous supplementation emerged as a promising strategy to enhance salt tolerance and promote germination, growth, and yield by regulating water status, photosynthetic pigments, and ion homeostasis in eggplant seedlings under NaCl stress. Increasing evidence highlights the negative effects of microplastics (MPs) on crops and bio-based plastics offer an alternative to conventional plastics. However, there is limited knowledge on the impacts and mechanisms of bio-based MPs on crop physiology. In this study, bio-based polylactic acid (PLA) and petroleum-based MPs [polyamide (PA) and polypropylene (PP)] were added to hydroponic cultures planted with rice (Oryza sativa L.) seedlings to assess their toxicity. Compared to PA and PP MPs, PLA MPs experienced greater aging after 28 days of exposure, and their surfaces were loaded with more rod-shaped microorganisms with potential plastic degradation ability, such as Proteobacteria and Bacteroidota, which competed with rice seedlings for carbon and nitrogen sources for self-multiplication, thus altering the carbon fixation and nitrogen cycling processes during rice seedling growth. Down-regulation of amino acid and lipid metabolisms in the PLA treatment inhibited the normal synthesis of chlorophyll in rice seedling leaves. Consequently, decreases in the biomass and height of rice seedling roots and shoots were observed in the PLA MP treatment. The effects of co-exposure to antibiotics and microplastics in agricultural systems are still unclear. This study investigated the effects of florfenicol (FF) and polystyrene microplastics (PS-MPs) on photosynthetic carbon assimilation in rice seedlings. Both FF and PS-MPs inhibited photosynthesis, while PS-MPs can alleviate the toxicity of FF. Chlorophyll synthesis genes (HEMA, HEMG, CHLD, CHLG, CHLM, and CAO) were down-regulated, whereas electron transport chain genes (PGR5, PGRL1A, PGRL1B, petH, and ndhH) were up-regulated. FF inhibited linear electron transfer (LET) and activated cyclic electron transfer (CET), which was consistent with the results of the chlorophyll fluorescence parameters. The photosynthetic carbon assimilation pathway was altered, the C3 pathway enzyme Ribulose1,5-bisphosphatecarboxylase/oxygenase (RuBisCO) was affected, C4 enzyme ((phosphoenolpyruvate carboxykinase (PEPCK), pyruvate orthophosphate dikinase (PPDK), malate dehydrogenase (MDH), and phosphoenolpyruvate carboxylase (PEPC))) and related genes were significantly up-regulated, suggesting that the C3 pathway is converted to C4 pathway for self-protection. The key enzymes involved in photorespiration, glycolate oxidase (GO) and catalase (CAT), responded positively, photosynthetic phosphorylation was inhibited, and ATP content and H + -ATPase activity were suppressed, nutrient content (K, P, N, Ca, Mg, Fe, Cu, Zn, Mn, and Ni) significantly affected. Micro/nano-plastics (MNPs) are emerging non-point source pollutants that have garnered increasing attention owing to their threat to ecosystems. Studies on the effects of MNPs on horticultural crops are scarce. Specifically, whether MNPs can be absorbed and transported by grapevines have not been reported. To fill this gap, we added polystyrene nanoplastics (PS-NPs, 100 nm) to a hydroponic environment and observed their distribution in grape seedlings of Thompson Seedless (TS, Vitis vinifera L.). After 15 d of exposure, plastic nanospheres were detected on the cell walls of the roots, stems, and leaves using confocal microscopy and scanning electron microscopy. This indicated that PS-NPs can also be absorbed by the root system through the epidermis-cortex interface in grapevines and transported upward along the xylem conduit. Furthermore, we analyzed the molecular response mechanisms of TS grapes to the PS-NPs. Through the measurement of relevant indicators and combined omics analysis, we found that plant hormone signal transduction, flavonoid and flavonol biosynthesis, phenylpropanoid biosynthesis, and MAPK signaling pathway biosynthesis played crucial roles in its response to PS-NPs. Chlorpyrifos (CPF) has been used worldwide, but its possible negative effects on macrophytes have been scarcely studied. The main goal of the present work was to assess the potential phytotoxic effects of CPF on different stages (seed and seedling) of the wetland macrophyte Bidens laevis. During the germination of seeds, stimulation of radicle growth at low concentrations of CPF (10 µg/L) and inhibition of its elongation at 80 µg/L CPF were observed. In seedlings, concentrations ≤ 160 µg/L CPF did not exhibit adverse effects on growth after 7 days of exposure, despite the decrease of photosynthetic pigments and carotenoids observed at 40 µg/L CPF compared to the control. Environmentally relevant concentrations of CPF altered neither oxidative stress biomarkers nor pigment contents in seedlings exposed for 48 h, suggesting CPF would be non-toxic to B. laevis in natural scenarios., (© 2024.
