Dissertation, Rheinisch-Westfälische Technische Hochschule Aachen, 2022; Aachen : RWTH Aachen University 1 Online-Ressource : Illustrationen, Diagramme (2022). = Dissertation, Rheinisch-Westfälische Technische Hochschule Aachen, 2022, The objective of this study was to investigate the efficacy of adjunctive therapy with a 445 nm diode laser in endodontic therapy as well as the potential harm that may be caused to the periodontal tissues due to excessive heat development. The experiments for this study were done on freshly extracted, human single-rooted teeth showing only straight root canals that were inoculated with E. faecalis. The first experimental series comprised 14 specimens and was divided into two parts. During the first part the specimens were irrigated with 2,65% sodium hypochlorite solution for a duration of 60 seconds, after that they were irradiated at 0.5 W for 55 seconds. For the second part, the same specimens were autoclaved and inoculated again prior to being irrigated with 2,65% sodium hypochlorite solution for 60 seconds. Then they were irradiated at 0.8 W for 55 seconds. The second experimental series comprised 17 specimens that were first irrigated with 2,65% sodium hypochlorite solution for a duration of 60 seconds and subsequently irradiated at 1 W. All specimens were autoclaved thereafter and inoculated just to be treated again but this time solely applying the diode laser. The rise in temperature on the root surfaces was simultaneously measured by thermography. The first part of the first experiment resulted in almost complete elimination of the bacteria after irrigation and subsequent irradiation (mean reduction: 97,63%). Comparable results were obtained in the second part of the experiment (mean reduction: 99,96%).In the second experiment almost no bacterial growth could be found after just the irrigation with sodium hypochlorite solution (mean reduction: 99,96%) whereas the subsequent irradiation resulted in a complete elimination of the bacteria (mean reduction: 100%). When applying the diode laser only, the reduction of the bacterial growth was incomplete (mean: 63,31). The temperature rises recorded during the laser application ranged from 15,19 K to 111,41 K (median: 41,67 K).The results of this in vitro study suggest that the antibacterial properties of a 445 nm diode laser can be considered an effective adjunct to common root canal treatment methods., Published by RWTH Aachen University, Aachen
