Além de ser uma questão de saúde pública, a peçonha ou veneno de serpentes (SV) é particularmente interessante por apresentar atividades bioquímicas e farmacológicas únicas. Nossa pesquisa abrange os estudos acerca de seis proteases de SV, das classes metalo- (SVMP) e serino proteases (SVSP), que foram isoladas de três serpentes: Bothrops asper (SVMP, BaP1), Crotalus simus (SVMP, PIII; SVSP-I e SVSP-II) e Crotalus durissus cumanensis (SVSP, Cd SI e Cd SII). Na primeira parte deste trabalho foram utilizadas as técnicas de dicroísmo circular e fluorescência para caracterização biofísica das proteínas alvo. Foi verificado o papel de íons de cálcio (II) na estabilização da estrutura e atividade das duas SVMPs. Na segunda parte, utilizando as técnicas de Screening virtual das bibliotecas Zinc, ChemBL e PubChem, e foram propostos in silico cinco compostos como inibidores da BaP1. Destes, três foram testados em ensaios in vitro e dois inibiram a atividade proteolítica da BaP1. Usando metodologia similar, a hesperitina foi proposta como um inibidor potente para as SVSPs, in silico e in vitro. Dessa forma, esses compostos poderão ser úteis no tratamento de envenenamentos. A terceira parte deste trabalho consistiu na análise comparativa de estruturas 3D de SVMP por ferramentas de biologia computacional e na caracterização de diferenças entre as proteínas que provocam ou não a hemorragia em acidentes ofídicos. Finalmente, na quarta e última parte, estudamos a atividade das SVMP, BaP1 e PIII, perante a inibição da formação de capilares sanguíneos e verificamos que ambas mostram potencial farmacológico para terapia antiangiogênica Abstract: Snakes bites are clasificate as neglected diseases. As well as a public health issue, the snake venom (SV) is particularly interesting because it presents unique biochemical and pharmacological activities. Our research covers the studies of six SV proteases, metalloproteases (SVMP) and serine proteases (SVSP), isolated from three snakes: Bothrops asper (SVMP, BaP1), Crotalus simus (SVMP, PIII; SVSP-I and SVSP-II) and Crotalus durissus cumanensis (SVSP, Cd SI and Cd SII). The circular dichroism and fluorescence techniques were used for the biophysical characterization of target proteins. The role of calcium (II) ions in stabilizing the structure and activity of the two SVMP was verified. In the second part of this work, using the Virtual Screening techniques, the libraries Zinc, ChemBL and PubChem were screened, five compounds have been proposed as in silico inhibitors to BaP1. Of these, three were tested in vitro assays and two showed complete inhibition of the proteolytic activity of BaP1. Using similar methodology, the hesperetin has been proposed as a potent inhibitor for the SVSP, both in silico and in vitro. Thus, these compounds have potential to aid in the anti-poisoning treatment. The third part of this work consisted of the comparative analysis SVMP 3D structures by computational biology tools and the differences between the proteins that cause or not cause hemorragic were characterized. Dentro de sua complexa composição, o veneno apresenta enzimas proteases, como as metaloproteases e serinoproteases que estão envolvidas em distúrbios hemostáticos e coagulantes, interferindo na agregação plaquetária. O presente trabalho teve como objetivo purificar, caracterizar bioquimicamente e avaliar as atividades farmacológicas de uma serinoprotease com atividade coagulante, do veneno de Bothrops barnetti nomeada TLBbar. A purificação envolveu dois passos cromatográficos, exclusão molecular em Sephadex G-75 e HPLC de fase reversa em coluna analítica Supelco C8, obtendo-se um alto grau de pureza e homogeneidade, revelado com SDSPAGE e com Mr ~ 28,5 kDa. TLBbar mostrou homlogia sequêncial com outras serinoproteases de veneno de serpentes, evidenciado através de espectrometria de massas MS/MS, como a trombina-like denominada Calobin, isolada e caracterizada a partir de Agkistrodon caliginosus (Korean Viper), Crotalase (Crotalus adamanteus), flavoxobin (Trimeresurus flavoviridis) entre outras. Observa-se na homologia que as serinoproteases, com as quais foi comparada a TLBbar, apresentam os aminoácidos conservados do sítio catalítico (His57, Asp102, Ser195), já na sequencia da TLBbar demonstra-se a presença do aminoácido histidina na posição 57 da tríada catalítica. Este resultado em conjunto com as atividades enzimáticas e biológicas, confirmam que TLBbar pertence ao grupo de enzimas serinoproteases com atividade trombina-like. A enzima apresenta sua maior atividade proteolítica a 37oC e em pH 8, sendo essa alterada na presença de íons divalentes por interações eletrostáticas entre enzima e substrato. TLBbar apresenta um comportamento michaeliano segundo os estudos cinéticos realizados com o substrato BApNA, revelando um Vmax e Km de 0,42nmol/min e 0,433mM, respectivamente. Em presença de heparina mantem 80% de sua atividade, e apresenta relativa estabilidade frente a SBT-I e agentes quelantes como EDTA e EGTA. TLBbar tem atividade coagulante sobre fibrinogênio bovino (atividade fibrigenolítica), formando um coágulo semi-rígido, pois não ativa o fator XIII da cascata de coagulação como faz a trombina que resulta na formação de um coágulo consistente. Sua atividade fibrigenolítica foi evidenciada pela liberação dos fibrinopeptídios A, pela degradação da cadeia A? do fibrinogênio, a qual foi inibida por PMSF e Leupeptina, confirmando sua classificação no grupo das serinoproteases. A eficiência desta atividade fibrigenolítica foi observada entre 30 oC e 40oC durante 2 horas de incubação e alterada em presença de íons divalentes como cálcio, magnésio e bário, ressaltando a maior atividade fibrigenolítica com o cálcio. A enzima purificada TLBbar foi capaz de agregar plaquetas e em concentração de 2,5?g evidenciou comportamento semelhante à trombina. Interessante que TLBbar estimula a agregação plaquetária assim como também retarda dita atividade no tempo quando comparado com o controle (trombina). Este fato é devido a sua mudança de conformação estrutural que corresponde à ativação plaquetaria a qual ocorre com mais lentidão (nos primeiros minutos), em comparação a transformação causada pela trombina, tendo como conseqüência a liberação mais lenta de agentes como ADP e tromboxanos A2, necessários para alcançar a adesão plaquetária. A serinoprotease TLBbar não apresenta toxicidade, pois não possui atividades edematogênica, hemorrágica e miotóxica in vivo. Por outro lado, TLBbar evidencia capacidade de dissolver o coágulo de fibrina gerado pela trombina após 36 horas de incubação a 37oC, esta atividade foi inibida com PMSF nas mesmas condições, estes dados sugerem a presença de atividade fibrinolítica Abstract: The snake venoms contain variety of proteins and are studied in the world for biological and pharmacological importance, within their complex composition of the venom protease enzymes present, as metalloproteases and serineproteases that cause hemostatic disorders, coagulantes and platelet aggregation. The present dissertation aimed to purify, biochemically characterize and evaluate the pharmacological activities of a serine protease with coagulant activity named TLBbar from Bothrops barnetti. Its purification involved two chromatographic steps, molecular exclusion in Sephadex G-75 and reversedphase HPLC with a Supelco C8 analytical column, obtaining high purity and homogeneity revealed by SDS-PAGE with Mr ~ 28.5 kDa. The enzyme has its highest proteolytic activity at 37 °C and at pH 8, which is altered in the presence of divalent ions by electrostatic interactions between enzyme and substrate. TLBbar presents a Michaelis-Menten behavior according the kinetic studies revealing a Km and Vmax of 0.433 mM and 0.42 nmol/min, respectively. TLBbar in the presence of heparin maintains its activity around 80%, although has relative stability compared with SBT-I and chelating agents such as EDTA and EGTA. TLBbar has coagulant activity on bovine fibrinogen, forming a clot semi-rigid, different the action of thrombin that results in the formation of a clot consistent. Its fibrigenolytic activity was evidenced by the release of fibrinopeptides A, by degradation of fibrinogen A? chain, which was inhibited by PMSF and Leupeptin, confirming its classification in the group of serineproteases. The efficiency of this activity was observed between 30 and 40 °C during 2 hours of incubation and changed in the presence of divalents ions such as calcium, magnesium and barium, brings out the greatest fibrigenolytic activity with calcium. The purified enzyme TLBbar was capable of aggregating platelets; 2.5?g of concentration evidence similar behavior to thrombin, TLBbar interesting stimulates platelet aggregation as well as delay time in such activity when compared with control (thrombin). We suggest that structural process of platelet activation occurs over a longer period of time with TLBbar as a result of release of ADP and thromboxane A2 agents that happen that occur with less speed to achieve platelet adhesion compared with thrombin. This enzyme not showed edematogenic-activity when compared with the activity caused by B. Barnetti snake venom, did not induce hemorrhagic nor myotoxic effect in vivo. On the other hand, TLBbar had proteolytic activity similar to thrombin, proven in the fibrigenolytic activity on bovine fibrinogen as measured in the release of fibrinopeptides in times of release similar to the control, the proteolytic activity was also evidenced by the ability to dissolve fibrin clot generated by Thrombin after 36 hours of incubation at 37 °C, the activity was inhibited with PMSF under the same conditions, these data suggest the presence of fibrinolytic activity Mestrado Bioquímica Mestre em Biologia Funcional e Molecular

Orientador: Sergio Marangoni, Luis Alberto Ponce Soto Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Muitas das toxinas isoladas e estudadas em seus efeitos biológicos precisam ser reavaliadas à luz de metodologias otimizadas em HPLC e confirmadas por espectrometria de massas, devido à possível presença de algum componente não estudado. No caso da serpente Bothrops marajoensis, são valorizados os estudos sobre essa espécie devido aos poucos trabalhos realizados, provavelmente por se tratar de uma espécie restrita e rara encontrada na ilha de Marajó, no Pará, e posteriormente em alguns locais litorâneos do Maranhão, aparentemente endêmica. A reprodutibilidade da atividade biológica, através dos efeitos farmacológicos, só é possível com a utilização de frações quimicamente homogêneas que mantenham a integridade da função biológica. No presente trabalho, foi purificada uma nova serinoprotease TLBm, com atividade trombina "like" em um único passo cromatográfico em um sistema de HPLC de fase reversa, com um alto grau de pureza e homogeneidade molecular, sem perda da atividade biológica. A nova serinoprotease foi caracterizada fisico-quimicamente, revelando uma massa molecular de 33332,5 Da por Espectrometria de Massa (MALDI-Tof). Por outro lado, mostrou uma atividade proteolítica perante o substrato cromogênico DL-BA?NA, assim como foi capaz de evidenciar uma atividade fibrinogenolítica frente ao fibrinogênio bovino e hidrolisar a cadeia alfa (a) e beta (ß), comportando-se como uma trombina "like" tipo A e B. Os estudos da atividade cinética mostraram que a serinoprotease com atividade trombina "like" possui um comportamento michaeliano frente ao substrato DL-BA?NA, registrando as constantes cinéticas de Vmax = 2,3x10-1 nmoles p-NA/Lt/min e o KM = 0,52x10-1M. Os estudos do efeito da temperatura sobre a atividade catalítica revelaram que a serinoprotease TLBm apresenta uma ótima atividade em torno de 38ºC e um pH de 8,0. Para confirmar o caráter trombina "like" da TLBm, esta foi inibida pela ação do fenilmetilsulfonil fluoreto (PMSF) e outros inibidores, através dos quais a atividade foi reduzida em mais de 50% (69,89±1,5 %). A análise de composição de aminoácidos mostrou que a serinoprotease TLBm trata-se de uma proteína de caráter ácido ao apresentar um elevado número de aminoácidos ácidos, assim como uma boa quantidade de aminoácidos hidrofóbicos, o que garante a estabilidade conformacional da proteína. A presença de 12 cisteínas sugere a possível presença de 6 pontes dissulfeto. A seqüência N-terminal da serinoprotease com atividade trombina "like" TLBm mostrou um alto grau de homologia seqüencial (55,9 a 79,4 %). No entanto, existem algumas substituições nos aminoácidos (S)12?(H)12 e (V)15?(L)15, os quais poderiam estar relacionados à diferença na atividade biológica estudada aqui ou em outras atividades, o que faria da TLBm uma serinoprotease particular. Com relação ao estudo das atividades biológicas, a TLBm mostrou-se desprovida de atividade hemorrágica, o que é característico desta família de proteínas e reforça a afirmação de esta ser uma serinoprotease. A TLBm apresentou também baixa capacidade inflamatória (edematogênica) e uma alta concentração na letalidade (DL50) intracerebroventricular (i.c.v), evidenciando que a toxina não contribui significativamente com a letalidade do veneno total. A serinoprotease com atividade trombina "like" TLBm tem a propriedade de induzir a agregação plaquetária em plasma rico em plaquetas (PRP) e esse efeito é inibido na presença do PMSF. Abstract: Many of the isolated and studied toxins in their biological effects need to be revalued, to the light of methodologies optimized in HPLC and confirmed by Mass Spectrometry, due to the possible presence of some component which has not been studied. In the case of the serpent Bothrops marajoensis, its results are valuable due to the fact that there are few works accomplished probably because the treating of a restricted and rare species found at the island of Marajó, in Pará, and later in some coastal places of Maranhão, which seem endemic. The reproduction of the biological activity, through the pharmacological effects, is only possible with the use of chemically homogeneous fractions so that they maintain the integrity of the biological function. In the present work a new serineprotease was purified with activity thrombin like TLBm in a Chromatography step in a system of HPLC of reverse phase, with a high degree of purity and molecular homogeneity, without loss of the biological activity. A new serineprotease was characterized physical-chemically revealing a molecular mass of 33332,5 Da by a Mass Spectrometry (MALDI-Tof), on the other hand, it did show an proteolitic activity before the substratum chromogenic DL-BA?NA, as well as it was capable to evidence an fibrinogenolitic activity in front of the bovine fibrinogen, and hydrolise the alpha (a) and beta (ß) chain which behaved as a thrombin like A and B types. The studies of the kinetic activity showed that the serineprotease with activity thrombin like, possesses a michaeliano behavior in front of the substratum DL-Ba?NA registering the kinetic constants of Vmax = 2,3x10-1 nmoles ?-NA/Lt/min and the KM = 0,52x10-1M respectively. The studies of the effect of the temperature under the catalytic activity, revealed that the serineprotease TLBm was capable to show a great activity around 38ºC and a pH of 8,0. To confirm the character thrombin like of TLBm, it was inhibited by the action of the fenilmetilsulfonil fluoride (PMSF) and other inhibitors, where the activity was reduced in more of the 50% (69,89 ± 1,5%). The analysis of composition of amino acids showed that the serineprotease TLBm is a character protein acid when presenting a high number of acid amino acids, as well as a good amount of amino acids hydrophobic that guarantees the conformational stability of the protein. The presence of 12 cisteinas suggests the possible presence of 6 dissulfeto bridges. The N-terminal sequence of the serineprotease with activity thrombin like TLBm, showed a high degree of sequential homology (55,9 to 79,4%). However there are some substitutions in the following amino acids: (S)12?(H)12 and (V)15?(L)15 which could be related with the difference in the biological activity studied here or with other activities, which would make TLBm a private serineprotease. The studies of biological activity such an as hemorrhage, reveal that the serineprotease is completely deprived which is known is already of this family of proteins. On the other hand the studies of the inflammatory effect (oedematogenic) reveal that the serineprotease possesses a low inflammatory capacity and finally it displays a high concentration in the lethality (DL50) intracerebroventricular (i.c.v.) showing that the toxin doesn't contribute significantly with the lethality of the total poison. The serineprotease with thrombin like activity has the property of inducing the platelet aggregation in platelets-rich plasma (PRP), and that effect is inhibited in the presences of the PMSF. CNPq - Conselho Nacional de Desenvolvimento Científico e Tecnológico FAPEMIG - Fundação de Amparo à Pesquisa do Estado de Minas Gerais CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A resposta efetora de macrófagos inicia-se com o reconhecimento de padrões moleculares associados a patógenos (PAMP) por receptores de reconhecimento de padrões expressos nos macrófagos. O receptor do tipo Toll (TLR)4 é um exemplo de PAMP cujo ligante é o lipopolissacarídeo (LPS), molécula oriunda de bactérias gram-negativas capaz de desencadear uma potente resposta antimicrobiana e pró-inflamatória nos macrófagos. Por outro lado, durante uma lesão tecidual ou infecção, proteases geradas por células e/ou secretadas por microorganismos contribuem para amplificar a resposta inflamatória. O principal mecanismo ocorre através de receptores acoplados à proteína G denominados Receptores Ativados por Protease (PAR) que desempenham funções relacionadas à coagulação, câncer, dor, inflamação, entre outras. PAR2 é o subtipo mais expresso em macrófagos e embora esse receptor funcione como um sensor inato, ainda pouco se sabe sobre a sua participação nas funções efetoras nos macrófagos. Dessa maneira, o objetivo deste trabalho foi investigar o papel de PAR2 nas respostas efetoras de macrófagos induzidas por LPS. Para isso, macrófagos obtidos da cavidade peritoneal de camundongos C57BL/6 foram estimulados com o agonista sintético de PAR2 (SLIGRL-NH2, 30μM), LPS (100 ng/mL) ou ambos, sendo em seguida avaliadas a sua atividade fagocítica, produção de óxido nítrico (NO), espécies reativas de oxigênio (EROS) e das citocinas TNF, IL-1β, MCP-1, IL-6 e IL-10. Avaliamos ainda, a expressão de PAR2, TLR4 e da enzima óxido nítrico induzível (iNOS), a interação física entre PAR2 e TLR4, as vias de sinalização envolvidas nesse fenômeno, os mecanismos cálcio-dependentes envolvidos e a contribuição das proteases endógenas e de um fragmento de PAR2, obtido após clivagem de sua porção extracelular, nos efeitos pró-inflamatórios dos macrófagos peritoneais estimulados com LPS. Em alguns experimentos, os macrófagos foram pré-incubados com inibidor de síntese proteica (ciclohexemida, 20μg/mL) ou inibidor da transcrição gênica (actinomicina D, 5μg/mL) antes do estímulo com SLIGRL-NH2, LPS ou ambos e antes da avaliação da expressão de PAR2 e TLR4. A coativação de PAR2 e TLR4 potencializou as funções efetoras dos macrófagos como a fagocitose, a produção de NO, a expressão de iNOS, EROS e citocinas pró-inflamatórias. Além disso, a ativação de PAR2 e/ou TLR4 aumentou a expressão de ambos receptores, sendo esse mecanismo dependente da transcrição gênica e da síntese de novo destes receptores; e a coincubação com o agonista de PAR2 e LPS aumentou significativamente a colocalização e a interação física entre PAR2 e TLR4, confirmando uma cooperação entre esses receptores nas funções efetoras dos macrófagos. O pré-tratamento com coquetel inibidor de proteases reduziu a liberação de cálcio induzida por LPS, sugerindo que possíveis proteases liberadas pelos macrófagos poderiam, de forma autócrina, promover a ativação de PAR2, enquanto o fragmento de PAR2 não modificou os níveis intracelulares de cálcio, confirmando que o sinergismo PAR2-TLR4 depende da produção de um fragmento ativo. Por fim, demonstramos que os mecanismos envolvidos na potencialização do repertório de ativação dos macrófagos mediados por PAR2-TLR4 são dependentes da via do fator de transcrição NFκB e do prolongamento do tempo de liberação do cálcio intracelular, sem, no entanto, interferir na fosforilação da via da MAP quinase p38. Como conclusão, este trabalho demonstra que PAR2 e TLR4 desempenham um papel importante para potencializar as funções efetoras dos macrófagos, fornecendo ainda, evidências para a compreensão do funcionamento dessas células em um microambiente infeccioso ou inflamatório em que as proteases estão presentes. The macrophage effector response begins with the recognition of pathogen-associated molecular patterns (PAMP) by pattern recognition receptors expressed on macrophages. The Toll-like receptor (TLR) 4 is an example of PAMP whose ligand is the lipopolysaccharide (LPS), a molecule derived from gram-negative bacteria capable of triggering a potent antimicrobial and pro-inflammatory response in macrophages. On the other hand, during tissue injury or infection, proteases generated by cells and / or secreted by microorganisms contribute to amplify the inflammatory response. The main mechanism occurs through receptors coupled to protein G called Protease Activated Receptors (PAR) that perform functions related to coagulation, cancer, pain, inflammation, among others. PAR2 is the most expressed subtype in macrophages and although this receptor functions as an innate sensor, little is known about its participation in the effector functions in macrophages. Thus, the objective of this work was to investigate the role of PAR2 in the effector responses of macrophages induced by LPS. For this, macrophages obtained from the peritoneal cavity of C57BL / 6 mice were stimulated with the synthetic agonist of PAR2 (SLIGRL-NH2, 30μM), LPS (100ng / mL) or both, and their phagocytic activity, nitric oxide production (NO), reactive oxygen species (EROS) and cytokines TNF, IL-1β, MCP-1, IL-6 and IL-10 were evaluated. We also evaluated the expression of PAR2, TLR4 and the inducible nitric oxide enzyme (iNOS), the physical interaction between PAR2 and TLR4, the signaling pathways involved in this phenomenon, the calcium-dependent mechanisms involved and the contribution of endogenous proteases and a PAR2 fragment, obtained after cleavage of its extracellular portion, on the pro-inflammatory effects of peritoneal macrophages stimulated with LPS. In some experiments, macrophages were pre-incubated with protein synthesis inhibitor (cyclohexemide, 20μg / mL) or gene transcription inhibitor (actinomycin D, 5μg / mL) before SLIGRL-NH2, LPS or both and before expression evaluation of PAR2 and TLR4. The coactivation of PAR2 and TLR4 enhanced macrophage effector functions such as phagocytosis, NO production, iNOS expression, EROS and pro-inflammatory cytokines. In addition, activation of PAR2 and / or TLR4 increased the expression of both receptors, this mechanism being dependent on gene transcription and de novo synthesis of these receptors, and the coincubation with the PAR2 and LPS agonist significantly increased colocalization and physical interaction between PAR2 and TLR4, confirming a cooperation between these receptors in the effector functions of macrophages. Pretreatment with protease inhibitor cocktail reduced LPS-induced calcium release, suggesting that possible proteases released by macrophages could autocrinically promote the activation of PAR2, while the PAR2 fragment did not change intracellular calcium levels, confirming that the PAR2-TLR4 synergism depends on the production of an active fragment. At last, we demonstrate that the mechanisms involved in enhancing the activation repertoire of macrophages mediated by PAR2-TLR4, are dependent on the transcription factor NFκB pathway and on the prolongation of the intracellular calcium release time, without, however, interfering with phosphorylation of via MAP kinase p38. In conclusion, this work demonstrates that PAR2 and TLR4 play an important role to enhance the effector functions of macrophages, providing further evidence for understanding the functioning of these cells in an infectious or inflammatory microenvironment in which proteases are present.

Orientador: Ljubica Tasic Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Química Resumo: Os acidentes causados pelas picadas de serpentes acometem por volta de 5,4 milhões de pessoas por ano, sendo que cerca de 109,5 mil terminam com um desfecho fatal. As vítimas sobreviventes podem apresentar sequelas físicas permanentes devido a necrose do tecido local e, por vezes, sequelas psicológicas. Assim, estes tipos de acidentes representam um problema de saúde pública que afeta principalmente as pessoas que vivem em áreas rurais da Ásia, África e América Latina. Além disso, na América Latina, a maioria dos acidentes ofídicos são causados por espécies de serpentes da família Viperidae. As peçonhas das Viperidae contêm uma alta percentagem de metaloproteases zinco-dependentes que desempenham um papel relevante na patogênese da hemorragia característica destes envenenamentos. Entretanto, algumas serina proteases estão diretamente envolvidas nesses eventos proteolíticos, causando perturbações na hemostasia observada após o envenenamento. Resultados de experimentos in silico e in vitro mostram que a hesperitina é capaz de se ligar a serina proteases de três espécies de serpentes distintas e inibir a serina protease de duas formas uma como inibidor reversível acompetitivo e como inibidor reversível misto. Nossos objetivos foram estudar as interações entre as quatro serina proteases e a hesperitina, e comparar com as interações entre as proteases alvo e os correspondentes anticorpos através das técnicas espectroscópicas. Os procedimentos de isolar, purificar e caracterizar as proteínas alvo foram finalizados com sucesso. Também, a hesperitina foi obtida, purificada e caracterizada. Aplicando as técnicas bioquímicas, a atividade e a inibição da ação das serina proteases alvo foram descritas e apontam para uma potente ação da hesperitina perante as quatro proteínas alvo. Métodos biofísicos foram aplicados para descrever as interações entre proteases alvo e a hesperitina. Finalmente, as serina proteases alvo em interações com os anticorpos anti-serina proteases foram estudadas e essas interações foram comparadas com as observadas e descritas com a hesperitina. Concluímos que as interações com a hesperitina poderão ser exploradas para inibir a ação das serina proteases logo após acidente ofídico e em ação sinérgica com os antídotos Abstract: Accidents caused by snake bites affect around 5.4 million people a year, with about 109,500 ending with a fatal outcome. Surviving victims may have permanent physical sequelae due to local tissue necrosis and sometimes psychological sequelae. Thus, these types of accidents represent a public health problem that mainly affects people living in rural areas of Asia, Africa and Latin America. In addition, in Latin America, most ophidian accidents are caused by species of snakes the Viperidae family. Viperidae venoms contain a high percentage of zinc-dependent metalloproteases that play a relevant role in the pathogenesis of the haemorrhaging characteristic of these poisonings. However, some serine proteases are directly involved in these proteolytic events, causing disturbances in hemostasis observed after poisoning. Results of in silico and in vitro experiments show that hesperitin is capable of binding the serine proteases of three distinct snake species and inhibiting the serine protease in four ways as a reversible uncompetitive inhibitor and a reversible mixed inhibitor. Thus, our objectives were to study the interactions between the four serine proteases and hesperitin, as well as to compare these with the interactions between the target proteases and the corresponding antibodies through the spectroscopic techniques. The procedures for isolating, purifying and characterizing target proteins were successfully completed. In addition, we obtained, purified and characterized hesperitin. Utilizing biochemical techniques the activity and inhibition of the target serine proteases were described and point to of a potent action of hesperitin on the four target proteins were calculated. Utilizing biophysical methods were used to describe the interactions between target proteases and hesperitin. Finally, target serine proteases in interactions with the anti-serine protease antibodies were studied and those interactions were compared to the ones observed and described with hesperitin. The species Chamaecrista nictitans belongs to the family Fabaceae, is classified as an invasive plant, and can attack several types of plantations, however, studies report that its extract has antiviral properties, and act in the treatment of kidney stones. There is no study regarding the proteolytic activities present in this plant. Some molecules found in plants are the targets of several researches in the field of bioteconology. Protease inhibitors are compounds that inhibit enzymatic activity, among which the serinoprotease inhibitors form the class with the highest percentage of catolographic species, the Kunitz and Bowman-Birk families being the most studied. Some secondary metabolites may inhibit the action of proteases. Trypsins, synthesized by the pancreas, are in the class of hydrolases and are responsible for cleaving peptide bonds of Arg and Lys residues in the C-terminal regions. They are present in eukaryotes, prokaryotes and viruses, but their presence in plants is poorly described in the literature and has many biological and industrial applications. The main objective of the present work is to identify protease inhibitors and to carry out the purification of a trypsin similar to C. nitilans seeds, observing the importance of protease and trypsin inhibitors in plants and the biotechnological importance of this enzyme. For both, the identification of protein inhibitors and the partial purification of trypsin, present in the seeds of this species, through the use of ammonium sulfate precipitation and molecular exclusion chromatography were performed. A complex extract, characterized by numerous protein bands and the presence of a trypsin inhibitory activity, was observed through the polyacrylamide gels. From the fractionation, it was possible to solve a fraction with trypsin inhibitory activity and another with trypsin activity, in the fractions 20-40% and 40-60%, respectively, of the salt precipitation. The methods used to purify the enzyme obtained high recovery and considerable resolution. By inhibition zimography it was possible to notice that the inhibitor contained in the extract does not present protein characteristics, since it did not inhibit the hydrolysis of the casein present in the gel, suggesting to be a secondary nonprotein metabolite. A description of a trypsin inhibitory activity and partial purification of simian trpsin present in C. nictitans seeds using saline precipitation and molecular exclusion chromatography was obtained. This work presents a relevant scientific discovery and tends to contribute to future research on enzymes that present biotechnological applications from plants. Não há nenhum estudo em relação às atividades proteolíticas presentes nesta planta. Algumas moléculas encontradas em vegetais são alvos de várias pesquisas na área da bioteconologia. Os inibidores de proteases são compostos que inibem a atividade enzimática, dentre estes os inibidores de serinoproteases formam a classe com maior percentual de espécie catolografadas, sendo os da família Kunitz e Bowman-Birk os mais estudados. Alguns metabólitos secundários podem inibir a ação das proteases. As tripsinas, sintetizadas pelo pâncreas, integram a classe das hidrolases e são responsaveis por clivarem ligações peptídicas dos resíduos de Arg e Lys, nas regiões C-terminais. Estão presente nos eucariontes, procariontes e vírus, contudo a sua presença em vegetais é pouco descrita na literatura e apresentam muitas aplicações biológicas e industriais. Observando a importância de inibidores de proteases e tripsina em vegetais e a importância biotecnológica dessa enzima, o presente trabalho tem como principal objetivo identicar inibidores de proteases e realizar a purificação de uma tripsina símile presente nas sementes da espécie C.nictitans. Para tanto foi realizado a identificação de inibidores proteicos e a purificação parcial da tripsina símile, presente nas sementes desta espécie, através da utilização de precipitação com sulfato de amônio e cromatografia de exclusão molecular. Por meio das análises dos géis de poliacrilamida foi observado um extrato complexo, caracterizado por inúmeras bandas de proteínas e também a presença de uma atividade inibidora de tripsina. A partir do fracionamento foi possível resolver uma fração com atividade inibidora de tripsina e outra com atividade tripsina símile, nas frações 20-40% e 40-60%, respectivamente, da precipitação salina. Os métodos utilizados para purificar a enzima obtiveram alta recuperação e considerável resolução. Através de zimografia de inibição foi possível perceber que o inibidor contido no extrato, não apresenta carcateristicas protéicas, uma vez que não inibiu a hidrólise da caseína presente no gel, sugerindo ser um metabólito secundário não proteico. Foi obtida a descrição de uma atividade inibidora de tripsina e uma purificação parcial da tripsina símile, presente nas sementes da C. nictitans, utilizando precipitação salina e cromatografia de exclusão molecular. Este trabalho apresenta uma relevante descoberta científica e contribuir com as futuras pesquisas sobre enzimas que apresentam aplicações biotecnológicas oriundas de plantas.

Orientadores: Sergio Marangoni, Luis Alberto Ponce Soto Tese (doutorado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: No presente trabalho duas novas serinoproteases com atividade trombina-simile, nomeadas TLBbz e TLBro isoladas a partir de Bothrops brazili e Bothrops roedingeri respectivamente, foram purificadas em um único passo cromatográfico por HPLC de fase reversa com um alto grau de pureza e homogeneidade molecular, sem perda da atividade biológica. Ambas serinoproteases foram caracterizadas fisico-quimicamente, revelando uma massa molecular relativa de TLBbz = 35,13 KDa e TLBro = 20,24 KDa por SDS-PAGE. Ambas apresentaram atividade proteolítica perante o substrato cromogenico DL-Ba?NA. Os estudos da atividade cinética mostraram que as serinoproteases tiveram um comportamento michaeliano frente ao substrato DL-Ba?NA, apresentando uma Vmax = 1,89 nmoles ?-NA/Lt/min e KM = 0,853 mM para TLBbz; e Vmax = 0,0432 nmoles ?-NA/Lt/min e KM = 0,039 mM para TLBro. Ambas serinoproteases apresentam uma atividade ótima em torno de 38 oC e pH 8,0; sendo inibidas pela ação do fluoreto de fenilmetilsulfonila (PMSF) e outros inibidores, através dos quais a atividade trombina-simile foi reduzida em mais dos 50 % (TLBbz = 86.2 % and TLBro = 42.6 %). Ambas TLBbz e TLBro possuem caráter acido ao apresentar um elevado numero de aminoácidos ácidos, assim como uma boa quantidade em numero de aminoácidos hidrofóbicos, o que garante a estabilidade conformacional da proteína. A presença de 12 cisteinas sugere a possível formação de 6 pontes dissulfeto. TLBbz e TLBro evidenciaram uma atividade fibrinogenolitica frente ao fibrinogenio bovino hidrolisando a cadeia alfa (?) e beta (?), comportando-se como uma trombina-simile tipo AB no caso da TLBro e tipo B, por hidrolizar a cadeia beta (?), no caso da TLBbz. O inibidor de serino protease benzamidina inibe a hidrólise protéica das proteases digestivas do tipo tripsina, no intestino médio das larvas de Anticarsia gemmatalis. No presente trabalho, lagartas de A. gemmatalis recém-eclodidas foram alimentadas com o inibidor de tripsina benzamidina nas concentrações 0; 0,25; 0,50; e 0,75% (p/p de dieta artificial). As diferentes concentrações do inibidor causaram impactos negativos ao desenvolvimento larval do inseto, como aumento do período larval e maior porcentagem de mortalidade. Entretanto, observou-se que, mesmo nas maio- res concentrações testadas de benzamidina, o inseto consegue se adaptar ao inibidor por remodelagem na quantidade e no tipo de enzima presentes durante a digestão da dieta artificial. The serine protease inhibitor benzamidine inhibits hydrolyses of trypsin-like digestive proteases in the midgut of velvetbean caterpillar, Anticarsia gemmatalis. In this study, newly-emerged A. gemmatalis caterpillars were fed on artificial diet containing increasing concentrations of benzamidine (0, 0.25, 0.50 e 0.75% w/w). This inhibitor caused negative effects in the insect development, by increasing the larval cycle and higher mortality. Nonetheless even at the highest benzamidine concentration, the insect mortality was low, suggesting that the insect is able to adapt to this inhibitor. Such adaptation may take place by increasing the amount of protease produced for digestion or changing the prevailing type of protease.
