Brewers’ spent grain (BSG) is the primary by-product of beer production, and its potential use in food products is largely dependent on its processing, given its moisture content of up to 80%. This study aimed to evaluate the effects of physical separation with ultrasound application on the color, total phenolic content (TPC), antioxidant activity, proximate composition, total dietary fibers, and particle size distribution of BSG powders. Wet BSG (W) was subjected to two processes: one without ultrasound (A) and one with ultrasound (B). Both processes included pressing, convective air-drying, sieving, fraction separation (A1 and B1 as coarse with particles ≥ 2.36 mm; A2 and B2 as fine with particles < 2.36 mm), and milling. The total color difference compared to W increased through both processes, ranging from 1.1 (B1 vs. A1) to 5.7 (B1 vs. A2). There was no significant difference in TPC, but process B powders, particularly B2, showed lower antioxidant activity against ABTS•+, likely due to the release of antioxidant compounds into the liquid fraction during pressing after ultrasound treatment. Nonetheless, process B powders exhibited a higher content of soluble dietary fibers. In conclusion, ultrasound application shows potential for further extraction of soluble fibers. However, process A might be more practical for industrial and craft brewers. Further studies on the use of the resulting BSG powders as food ingredients are recommended.

The peel represents a significant portion of the araticum fruit (about 40%), which becomes waste after its consumption or processing. Previous studies have shown that the araticum peel is rich in phenolic compounds; however, little is known about the ideal conditions for recovering these compounds. Therefore, response surface methodology, using a central composite rotatable design, was employed to optimize the extraction process to maximize the total phenolic compounds (TPCs) and enhance the Trolox equivalent antioxidant capacity (TEAC) from araticum peel. The variables optimized were ethanol concentration (EC; 20–80%, v/v), extraction time (ET; 5–45 min), and solid–solvent ratio (SSR; 10–100 mg/mL). Additionally, condensed tannins, antioxidant capacity against synthetic free radicals (TEAC and FRAP) and reactive oxygen species (ROS), and the phenolic compounds profile, were evaluated. Optimum extraction conditions were 50% (v/v) ethanol concentration, 5 min of extraction time, and 10 mg/mL solid–solvent ratio. Under these conditions, experimental TPCs and TEAC values were 70.16 mg GAE/g dw and 667.22 µmol TE/g dw, respectively, comparable with predicted models (68.47 mg GAE/g dw for TPCs and 677.04 µmol TE/g dw for TEAC). A high condensed tannins content (76.49 mg CE/g dw) was also observed and 12 phenolic compounds were identified, predominantly flavonoids (97.77%), including procyanidin B2, epicatechin, and catechin as the major compounds. Moreover, a potent antioxidant activity was observed against synthetic free radicals and ROS, especially in scavenging peroxyl and hydroxyl radicals. From this study, we obtained the ideal conditions for recovering phenolic compounds from araticum peel using a simple, fast, sustainable, and effective method, offering a promising opportunity for the management of this plant byproduct.

Anthocyanins are phenolic compounds present in various plant products with interesting functional characteristics studied by science, such as their anti-inflammatory and antioxidant activities, among others. However, anthocyanins are considered unstable to various factors, which can affect their functional capacity. For this reason, some technologies, such as nanoencapsulation, are being applied to ensure their functional capacity effectively. The incorporation of anthocyanins in yogurt has shown various benefits, such as the ability to inhibit pathogenic microorganisms, reduce enzyme activity, and prolong the shelf life of the product. Additionally, the functional effects include their ability to modulate the gut microbiota, generating antioxidant, anti-inflammatory, and even antiproliferative responses, thereby reducing the capacity of tumor progression. For these reasons, this graphic review discussed the functional effects of yogurt enriched with nanoencapsulated anthocyanins on the gut microbiota and its influence on human health.

Abstract Background The high similarity in anatomical and neurophysiological processes between pigs and humans make pigs an excellent model for metabolic diseases and neurological disorders. Lipids are essential for brain structure and function, and the polyunsaturated fatty acids (PUFA) have anti-inflammatory and positive effects against cognitive dysfunction in neurodegenerative diseases. Nutrigenomics studies involving pigs and fatty acids (FA) may help us in better understanding important biological processes. In this study, the main goal was to evaluate the effect of different levels of dietary soybean oil on the lipid profile and transcriptome in pigs’ brain tissue. Results Thirty-six male Large White pigs were used in a 98-day study using two experimental diets corn-soybean meal diet containing 1.5% soybean oil (SOY1.5) and corn-soybean meal diet containing 3.0% soybean oil (SOY3.0). No differences were found for the brain total lipid content and FA profile between the different levels of soybean oil. Cocoa is the third most important global agricultural export commodity. However, because it is a crop sensitive to climatic change, there has been an active search for cocoa substitutes worldwide. Roasted jackfruit seeds were previously described as having a chocolate aroma and are affordable and accessible. In this study, we characterized and identified by SPME-GC-O and SPME-GC-MS the aroma profile of cocoa-based beverages formulated with jackfruit seed flour as a natural cocoa substitute. Our analysis tentatively identified 71 odor-active aroma descriptors with some similarities between formulations. Overall, 15 odor-active aromas were present in all beverages. The formulation containing only cocoa/chocolate showed the following aroma descriptors: cocoa, hazelnut, peanut butter, earthy, and roast, which are mostly related to the presence of 2,3-dimethylpyrazine and 2,3-diethyl-5-methylpyrazine. The fermented beverage had a content of complex pyrazines such as 2,3,5-trimethyl-6-isopentylpyrazine and methylpropylpyrazine. Our data indicated that both the control and fermented beverages showed a similar aromatic profile, mainly earthy, pyrazine, and chocolate. Qualitative similarities in the pyrazine content were observed between the fermented jackfruit seed flour and cocoa beverages. In conclusion, fermented jackfruit seed flour can be incorporated into cocoa-based beverages as a natural chocolate substitute, offering the potential to elevate the chocolate aroma.

Abstract Dietary fatty acids (FA) are components of the lipids, which contribute to membrane structure, energy input, and biological functions related to cellular signaling and transcriptome regulation. However, the consumers still associate dietary FA with fat deposition and increased occurrence of metabolic diseases such as obesity and atherosclerosis. Previous studies already demonstrated that some fatty acids are linked with inflammatory response, preventing metabolic diseases. To better understand the role of dietary FA on metabolic diseases, for the first time, a study to identify key transcription factors (TF) involved in lipid metabolism and inflammatory response by transcriptome analysis from liver samples of animal models was performed. The key TF were identified by functional enrichment analysis from the list of differentially expressed genes identified in liver samples between 35 pigs fed with 1.5% or 3.0% soybean oil. The functional enrichment analysis detected TF linked to lipid homeostasis and inflammatory response, such as RXRA, EGFR, and SREBP2 precursor. These findings demonstrated that key TF related to lipid metabolism could be modulated by dietary inclusion of soybean oil. It could contribute to nutrigenomics research field that aims to elucidate dietary interventions in animal and human health, as well as to drive food technology and science.

Pigs (Sus scrofa) are an animal model for metabolic diseases in humans. Pork is an important source of fatty acids (FAs) in the human diet, as it is one of the most consumed meats worldwide. The effects of dietary inclusion of oils such as canola, fish, and soybean oils on pig gene expression are mostly unknown. Our objective was to evaluate FA composition, identify changes in gene expression in the liver of male pigs fed diets enriched with different FA profiles, and identify impacted metabolic pathways and gene networks to enlighten the biological mechanisms’ variation. Large White male pigs were randomly allocated to one of three diets with 18 pigs in each; all diets comprised a base of corn and soybean meal to which either 3% of soybean oil (SOY), 3% canola oil (CO), or 3% fish oil (FO) was added for a 98-day trial during the growing and finishing phases. RNA sequencing was performed on the liver samples of each animal by Illumina technology for differential gene expression analyses, using the R package DESeq2. The diets modified the FA profile, mainly in relation to polyunsaturated and saturated FAs. Comparing SOY vs. FO, 143 differentially expressed genes (DEGs) were identified as being associated with metabolism, metabolic and neurodegenerative disease pathways, inflammatory processes, and immune response networks. Comparing CO vs. SOY, 148 DEGs were identified, with pathways related to FA oxidation, regulation of lipid metabolism, and metabolic and neurodegenerative diseases. Our results help explain the behavior of genes with differential expression in metabolic pathways resulting from feeding different types of oils in pig diets.

Jackfruit (Artocarpus heterophyllus Lam) occurs abundantly in Brazil, but its seeds are often underutilized in food nutrition. Previously, we showed that roasted jackfruit seeds are an innovative source of chocolate aroma. In this study, we characterized the volatile content of experimental beverages containing jackfruit seed flours as compared to non-alkaline cocoa powder. The three key results of this study were that (i) the fermented jackfruit seed flour beverage (FJSBev) was similar to the non-alkaline cocoa powder beverage (CTRLBev); (ii) similar volatile groups (acids, alcohols, aldehydes, ester, furans, and pyrazines) were identified in the samples (FJSBev and CTRLBev); (iii) the beverages contained volatile compounds with a low threshold of cocoa attributes (butter, cocoa, coffee, floral, fruit, green, honey, mushroom, nut, and roasted). To conclude, the beverage formulation containing fermented jackfruit seeds had more volatile compounds and odor-active constituents than that with non-alkaline cocoa powder. Collectively, our data demonstrate that fermentation is critical to producing a chocolate aroma. Fermented jackfruit seed flours could be used as a partial or total additive to improve chocolate aroma in food formulations.

Abstract Brown propolis from permanent preservation and reforestation areas of southern Brazil have attracted international commercial interest and have a unique composition, although little is known about their botanical origins, which are the plant resins used by bee foragers to produce propolis. Hence, the volatile profiles of organic and non-organic brown propolis and resins of suspected botanical origins—Araucaria angustifolia, Pinus elliott and Pinus taeda—were determined using static headspace gas chromatography coupled to mass spectrometry (SHS-GCMS) and compared. We determined the phytochemical composition, anti-inflammatory mechanism of action, ROS/RNS scavenging capacity and systemic toxicity of a purified subfraction (S8) of Eugenia selloi. The composition of S8 was assessed by LC-ESI-QTOF-MS; the anti-inflammatory activity in RAW264.7 macrophages through NF-κB activation and biomarkers by multiplex in THP-1 cells; neutrophil migration, intravital microscopy and ICAM-1 expression in mice; NETs formation and CD11b expression; S8 scavenging capacity of ROS/RNS; toxicity in Galleria mellonella larvae model. Coumaric acid, quercetrin and vanillic acid were identified. S8 decreased NF-κB activation, IL-1β, IL-6, IL-10, MDC and MCP-1 levels, reduced neutrophil migration and ICAM-1 expression in mice; S8 did not interfere NET formation and CD11b expression, exhibited high antioxidant and showed negligible toxicity. E. selloi proved to be a promising, yet underexplored source of bioactive compounds, which can be useful employed in agribusiness and in the pharmaceutical and food industry to develop new products or human health supplies.

Carqueja (Baccharis crispa Spreng.) has been primarily used as a medicinal plant around the world. Commercially, the essential oil content of carqueja leaves is the most valuable crop productivity variable. We evaluated the effect of irrigation management in different growing seasons on the essential oil content of carqueja leaves using gas chromatography coupled with mass spectrometry. The experiment was conducted in a greenhouse located in Southern Brazil, where the crop was cultivated for two years in different growing seasons under six irrigation regimes: 25%, 50%, 75%, 100%, 125%, and 150% of the reference crop evapotranspiration (T25, T50, T75, T100, T125, and T150, respectively). A seasonal pattern was observed in the number of metabolites of sesquiterpenes and phenolics in the essential oil extracted from the biomass; this outcome was correlated with irrigation regimes and air temperature. Principal component and hierarchical cluster analyses were used to discriminate the influence of abiotic conditions on secondary metabolite profiles. Spathulenol was the most abundant compound in the essential oils (95.43%) collected during the summer (December–March) season during the third harvest (H3) at T150. The essential oil content was 8.84% ± 0.05% and 10.52% ± 0.10% in summer and winter (June–September), respectively, with T100 at 45 and 46 days after planting.

This study aims to evaluate the effects of drying in a forced-air oven or solar dryer on the drying rates, physicochemical and microbiological characteristics, and antioxidant properties of rosemary, mint, common fennel, lemon grass, and basil. The drying rates of all herbs were higher in the forced-air oven in comparison to the solar dryer. According to results obtained for herbal properties after this different drying process, mint was less affected by both drying conditions. On the other hand, regardless of the method of drying used, all dried herbs exhibited similar antioxidant properties, mainly due to the presence of total phenolics. The antioxidant activities of oven-dried herbs ranged from 19.18 to 71.55% and increased in the order common fennel < lemon grass < mint < basil < rosemary, while the activities of sun-dried samples varied from 17.73 to 58.27% and increased in the order basil < common fennel < lemon grass < mint < rosemary. The results obtained demonstrate that the process of drying can alter the quality of an herbal product, implying that standardization of post-harvest steps is essential to ensure the consistency of an herbal product.

The mechanical extraction of oils from Brazilian açaí (Euterpe oleracea Mart) produces significant amounts of a byproduct known as “meal”, which is frequently discarded in the environment as waste material. Nevertheless, plant byproducts, especially those from oil extraction, may contain residual polyphenols in their composition and be a rich source of natural bioactive compounds. In this study, the phenolic composition and in vitro biological properties of a hydroethanolic açaí meal extract were elucidated. The major compounds tentatively identified in the extract by high-resolution mass spectrometry were anthocyanins, flavones, and flavonoids. Furthermore, rhamnocitrin is reported in an açaí byproduct for the first time. The extract showed reducing power and was effective in scavenging the ABTS radical cation (820.0 µmol Trolox equivalent∙g−1) and peroxyl radical (975.7 µmol Trolox equivalent∙g−1). NF-κB activation was inhibited at 10 or 100 µg∙mL−1 and TNF-α levels were reduced at 100 µg∙mL−1. However, the antibacterial effects against ESKAPE pathogens was not promising due to the high concentration needed (1250 or 2500 µg∙mL−1). These findings can be related to the diverse polyphenol-rich extract composition. To conclude, the polyphenol-rich extract obtained from açaí meal showed relevant biological activities that may have great applicability in the food and nutraceutical industries.

The propolis chemical composition is complex, varied, and closely related to characteristics of the vegetation and location where the hives are located. The objective of this work was to determine the antioxidant activity through FRAP methodology in propolis (raw, macerated, ethanolic extract and concentrated ethanolic extract) using a rapid and non-destructive method namely Fourier transform near-infrared (FTNIR) spectroscopy. By the results obtained for the antioxidant activity it can be verified that the samples of propolis present a very diversified chemical profile, for the FRAP methodology the samples of propolis collected in Três Barras – SC and Campo Magro – PR showed the highest activities: 1.8 × 103 and 1.6 × 103 μmol of Fe2+ g−1 respectively. It was possible to conclude that the macerated propolis presented the best multivariate calibration model established with the Savitzky-Golay (SG) + Constant Offset Elimination (COE) preprocessed spectra, where the R2 and 0.95 and 113 for FRAP determination. The error values RMSEC, RMSECV and RMSEE were 73, 1.3 × 102 and 81 μmol Fe2+ g−1, respectively. The FRAP model for macerated propolis was validated and can be used for quantification of antioxidant activity of new extracts of propolis, being useful as an alternative to rapid analysis, reducing waste generation and cost. Cambuci (Campomanesia phaea O. Berg Landrum) and uvaia (Eugenia pyriformis Cambess), both native Atlantic Rainforest fruits, are noteworthy for being rich in bioactive compounds and their significant antioxidant capacity. Despite the numerous known edible fruits in the world, consumption by humans is most often restricted to a few dozen of them. Such behavior occurs, among other reasons, due to the lack of knowledge about fruits not yet commercialized on a large scale. This study quantified the bioactive compound content (total phenolic compounds and ascorbic acid in cambucis and uvaias; proanthocyanidins in cambucis, and total carotenoid profile and individual carotenoids for grapes) and antioxidant capacity of the edible parts (peel and pulp) of cambuci and uvaia accessions, using three methods (ABTS•+, ROO• radical scavenging and HOCl elimination). Cambuci contained higher phenolic compound levels and displayed higher antioxidant capacity determined by the ABTS•+ and ROO• radical scavenging methods than uvaia (139 and 119 mg 100 g−1 of GAE, 10.5 and 7.73 μmol g−1 of TE; 9.17 and 5.92 μmol g−1 of TE, respectively). Vitamin C content and the antioxidant capacity determined by the HOCl elimination method were about 1.5- and 6-fold higher in uvaia compared to cambuci, with the latter being a first-time report for uvaia. Both fruits contained higher levels of bioactive compounds and antioxidant capacity than other commonly consumed fruits.

Honey is an ancient food in the human diet, and the chemical composition of some types of honey has been associated with several beneficial biological effects. Among them, honey has been highlighted to improve health and control inflammatory processes. However, there is no study elucidating the mechanism of action of honey produced organically. Here, we separated organic honey (OH) samples from the Brazilian Atlantic Rainforest into eight different profiles (OH-1 to OH-8) and evaluated, in vitro and in vivo, their anti-inflammatory potential. To determine cell viability, RAW 264.7 macrophages were treated with several concentrations of OH-1 up to OH-8, and anti-inflammatory activity was assessed through NF-κB activation and TNF-α levels. All types of the studied honey up to a concentration of 4% (w/v) did not interfere with macrophage viability and decreased NF-kB activation and TNF-α levels in macrophage culture in vitro. OH-7 was selected as the most promising anti-inflammatory and used in subsequent assays. Mice pretreated orally with OH-7 showed a decrease in neutrophil migration and TNF-α level. Thus, these types of Brazilian organic honey show promising anti-inflammatory potential, particularly the OH-7 variety. Brazilian organic honey may lead to the development of new products and/or be incorporated into food for use in veterinary medicine and human health as well.

The aim of this study was to identify the differentially expressed genes (DEG) from the skeletal muscle and liver samples of animal models for metabolic diseases in humans. To perform the study, the fatty acid (FA) profile and RNA sequencing (RNA-Seq) data of 35 samples of liver tissue (SOY1.5, n = 17 and SOY3.0, n = 18) and 36 samples of skeletal muscle (SOY1.5, n = 18 and SOY3.0, n = 18) of Large White pigs were analyzed. The FA profile of the tissues was modified by the diet, mainly those related to monounsaturated (MUFA) and polyunsaturated (PUFA) FA. The skeletal muscle transcriptome analysis revealed 45 DEG (FDR 10%), and the functional enrichment analysis identified network maps related to inflammation, immune processes, and pathways associated with oxidative stress, type 2 diabetes, and metabolic dysfunction. For the liver tissue, the transcriptome profile analysis revealed 281 DEG, which participate in network maps related to neurodegenerative diseases. With this nutrigenomics study, we verified that different levels of soybean oil in the pig diet, an animal model for metabolic diseases in humans, affected the transcriptome profile of skeletal muscle and liver tissue. These findings may help to better understand the biological mechanisms that can be modulated by the diet.

Introduction: Gibberellins (GA) are terpenoids that serve as important plant hormones by acting as growth and response modulators against injuries and parasitism. In this study, we investigated the in vitro anti-NF-κB, anti-Candida, and antioxidant activity of gibberellin A4 (GA4) and A7 (GA7) compounds, and further determined their toxicity in vivo. Methods: GA4 and GA7 in vitro toxicity was determined by MTT method, and nontoxic concentrations were then tested to evaluate the GA4 and GA7 anti-NF-κB activity in LPS-activated RAW-luc macrophage cell culture (luminescence assay). GA4 in silico anti-NF-κB activity was evaluated by molecular docking with the software “AutoDock Vina”, “MGLTools”, “Pymol”, and “LigPlot+”, based on data obtained from “The Uniprot database”, “Protein Data Bank”, and “PubChem database”. The GA4 and GA7 in vitro anti-Candida effects against Candida albicans (MYA 2876) were determined (MIC and MFC). GA7 was also evaluated regarding the viability of C. albicans preformed biofilm (microplate assay). In vitro antioxidant activity of GA4 and GA7 was evaluated against peroxyl radicals, superoxide anions, hypochlorous acid, and reactive nitrogen species. GA4 and GA7 in vivo toxicity was determined on the invertebrate Galleria mellonella larvae model. Results: Our data show that GA4 at 30 µM is nontoxic and capable of reducing 32% of the NF-κB activation on RAW-luc macrophages in vitro. In vitro results were confirmed via molecular docking assay (in silico), since GA4 presented binding affinity to NF-κB p65 and p50 subunits. GA7 did not present anti-NF-κB effects, but exhibited anti-Candida activity with low MIC (94 mM) and MFC (188 mM) values. GA7 also presented antibiofilm properties at 940 mM concentration. GA4 did not present anti-Candida effects. Moreover, GA4 and GA7 showed antioxidant activity against peroxyl radicals, but did not show scavenging activity against the other tested radicals. Both compounds did not affect the survival of G. mellonella larvae, even at extremely high doses (10 g/Kg). Conclusion: Our study provides preclinical evidence indicating that GA4 and GA7 have a favorable low toxicity profile. The study also points to GA4 and GA7 interference with the NF-κB via, anti-Candida activity, and a peroxyl radical scavenger, which we argue are relevant biological effects.

Açaí seeds (Euterpe oleracea Mart.) are the major residue generated during industrial extraction of açaí fruit pulp – a popular and typical Amazon fruit rich in bioactive compounds and nutrients. In this study, we investigated the bioaccessibility of an açaí seed extract using an in vitro simulated gastrointestinal digestion model. Catechin, epicatechin and procyanidins B1 and B2 were identified and quantified in the açaí seed extract and monitored by HPLC-DAD through the digestion phases. Bioaccessibility of these flavan-3-ols and deactivation of reactive oxygen species decreased after the intestinal phase, except for peroxyl radical (ROO●). RAW 264.7 macrophages treated either with the digested or undigested açaí seed extract showed reduced NF-κB activation and TNF-α levels, even following gastrointestinal digestion. Thus, the ROO● scavenging capacity and anti-inflammatory activity of the extract were found to be still remarkable after digestion, suggesting that açaí seeds could be explored as a source of bioactive compounds for functional foods, cosmetic or pharmaceutical purposes.

Apesar de a romã já ser reconhecida na literatura como tendo elevado potencial antioxidante e ajudar na prevenção de patologias relacionadas ao desequilíbrio oxidativo, ainda pouco se conhece sobre as variações na composição química que os diferentes tipos de cultivo podem propiciar. A procura pelo alimento orgânico têm aumentado, porém são escassas as pesquisas científicas com vista à caracterização do alimento produzido no sistema orgânico, em relação aos obtidos no sistema convencional. Considerando este cenário, o objetivo deste trabalho foi avaliar e comparar por diferentes metodologias a capacidade de romãs (Punica granatum) oriundas de cultivos orgânico e convencional. O extrato etanólico da polpa de romã foi obtido por método sequencial. Os fenólicos totais foram determinados pelo método de Folin Ciocateau e a atividade antioxidante in vitro foi determinada através de dois métodos: espectrofotometria, através da descoloração do radical ABTS+ (2,2-azinobis- [3-etil-benzotiazolin-6-ácido sulfônico]) com persulfato de potássio, e oxigen radical absorbance capacity. No cultivo das frutas do tipo convencional foram utilizados adubos, fertilizantes, pesticidas e aceleradores de amadurecimento, já as frutas orgânicas foram cultivadas sem o uso de herbicidas e inseticidas e aditivos minerais de indução de crescimento. A romã teve sua capacidade antioxidante determinada e os tipos de cultivo foram comparados entre si. Constatou-se que as frutas do cultivo orgânico apresentaram os mais altos teores de compostos redutores e atividade antioxidante equivalente ao Trolox quando comparadas à frutas de cultivo convencional. O sistema de cultivo orgânico contribuiu para uma mais efetiva atividade antioxidante da romã. O tipo de técnica agrícola foi capaz de interferir no conteúdo de compostos antioxidantes da fruta.

Peanut is an affordable legume known for its nutritional value and phenolic content. The kernel and skin of 14 peanut genotypes contrasting in drought tolerance had their phenolic profiles determined and reactive oxygen species (ROS) scavenging activity evaluated. Firstly, temperature and % EtOH to extract antioxidant phenolic compounds were optimized using response surface methodology (RSM). The optimized extraction conditions, 60 °C and 35% EtOH for kernels and 40 °C and 60% EtOH for skins, were further adopted, and phenolic compounds were identified and quantified using high-performance liquid chromatography coupled with electrospray ionization-quadrupole-time of flight-mass spectrometry (HPLC-ESI-QTOF-MS) and high-performance liquid chromatography with photodiode array detector (HPLC-PDA). As a result, phenolic acids and glycosidic/non-glycosidic flavonoids were found. Principal component analysis was conducted, and the pairwise score plot of the skin extracts based on individual phenolic compounds showed a trend of genotype clustering based not only on drought tolerance but also on botanical type of germplasm. Therefore, our results demonstrate the status quo for antioxidant phenolic compounds of peanut genotypes contrasting in drought tolerance grown under natural field conditions.

Brazilian native fruits are a rich source of polyphenolic compounds that can act as anti-inflammatory and antioxidant agents. Here, we determined the polyphenolic composition, anti-inflammatory mechanism of action, antioxidant activity and systemic toxicity in Galleria mellonella larvae of Eugenia selloi B.D.Jacks. (synonym Eugenia neonitida Sobral) extract (Ese) and its polyphenol-rich fraction (F3) obtained through bioassay-guided fractionation. Phenolic compounds present in Ese and F3 were identified by LC-ESI-QTOF-MS. The anti-inflammatory activity of Ese and F3 was tested in vitro and in vivo through NF-κB activation, cytokine release and neutrophil migration assays. The samples were tested for their effects against reactive species (ROO•, O2•-, HOCl and NO•) and for their toxicity in Galleria mellonella larvae model. The presence of hydroxybenzoic acid, ellagitannins and flavonoids was identified. Ese and F3 reduced NF-κB activation, cytokine release and neutrophil migration, with F3 being three-fold more potent. Overall, F3 exhibited strong antioxidant effects against biologically relevant radicals, and neither Ese nor F3 were toxic to G. mellonella larvae. In conclusion, Ese and F3 revealed the presence of phenolic compounds that decreased the inflammatory parameters evaluated and inhibited reactive oxygen/nitrogen species. E. selloi is a novel source of bioactive compounds that may provide benefits for human health.

ent-Nemorosone is a benzophenone isolated from geopropolis from Melipona scutellaris whose biological properties remain unexplored. Herein, we evaluated the anti-inflammatory activity of ent-nemorosone by in vivo neutrophil migration and TNF-α quantification, as well as by in vitro TNF-α quantification, ERK 1/2 phosphorylation, NF-kB activation and nuclear translocation of p65 in stimulated macrophages. ent-Nemorosone was also tested for its antimicrobial effects against Streptococcus mutans biofilm adhesion, polysaccharide production, and viability. ent-Nemorosone reduced the influx of neutrophils and TNF-α release into the peritoneal cavity of mice, as well as the in vitro TNF-α levels, ERK 1/2 phosphorylation, NF-kB activation and nuclear p65 translocation. Furthermore, this compound inhibited biofilm adhesion and reduced the amount of extra- and intracellular polysaccharides synthesized by S. mutans. Thus, ent-nemorosone showed promising anti-inflammatory activity by acting on TNF-α release in macrophages, as well as anti-biofilm properties by reducing S. mutans polysaccharide production.

This study determined the anti-inflammatory activity, mechanisms of action, and polyphenolic profile of Eugenia leitonii seed extract. The total phenolic content of the hydroethanolic extract from E. leitonii seeds (ESEL) was measured, and its phenolic compounds were identified by LC-MS/MS. The ESEL was submitted to in vitro (RAW264.7) and in vivo assays on neutrophil migration; expression of P-selectin and ICAM-1; release of TNF-α, IL-1β, and CXCL2/MIP-2; NF-κB activation; and carrageenan-induced paw oedema. ESEL yielded total phenolic content of 158.74 ± 3.5 mg GAE/g, with the presence of flavonols, anthocyanins and ellagitannins. ESEL significantly reduced neutrophil migration; decreased the expression of P-selectin and ICAM-1; decreased the release of TNF-α and CXCL2/MIP-2, as well as NF-kB activation and paw oedema in mice. Thus, this extract showed to be a promising source of bioactive compounds with anti-inflammatory potential by inhibiting neutrophil migration through a decrease in the release of TNF-α and CXCL2/MIP-2 (NF-κB pathway) in macrophages.

Brazilian red propolis (BRP) is a natural product widely known for its phenolic composition and strong antioxidant properties. In this study, we used the Box–Behnken Design (BBD) with Surface Response Methodology to optimize the extraction conditions for total phenolic content (TPC) and Trolox equivalent antioxidant capacity(TEAC) of bioactive phenolics from BRP. The extraction time, ethanol/water concentration and temperature, were tested. All variables had significant effects (p ≤ 0.05), with a desirability coefficient of 0.88. Under optimized conditions (90% ethanol at 80 °C for 30 min), the BRP extract showed a TPC of 129.00 ± 2.16 mg GAE/g and a TEAC of 3471.76 ± 53.86 µmol TE/g. Moreover, FRAP and ORAC assays revealed that the optimized BRP extract had 1472.86 ± 72.37 µmol Fe2+/g and 4339.61 ± 114.65 µmol TE/gof dry weight, respectively. Thirty-two phenolic compounds were tentatively identified by LC-QTOF-ESI-MS/MS, of which thirteen were found for the first time in BRP, including four flavones, one flavanol, two flavanones, two chalcones, and four isoflavonoids. Thus, our results highlight the importance of BRP as a source of a wide variety of phenolic compounds with significant antioxidant properties.

A clear gap with respect to the potential biological properties of wheat flavonoids exists in the available literature. This information is crucial for breeding programs aiming to produce new varieties presenting improved health benefits. Accordingly, advanced breeding lines of whole durum wheat were evaluated in this contribution. The highest recovery of phenolics was achieved using aqueous acetone (50:50, v/v), as verified by multi-response optimization, thus showing that phenolics could be largely underestimated by employing an inappropriate extraction. The concentration of derivatives of apigenin, the main phenolics present, ranged from 63.5 to 80.7%, as evaluated by LC–ESI-QTOF-MS. Phenolics from the breeding line 98 exhibited the highest ability in scavenging peroxyl radicals, reducing power as well as in terms of inhibition of pancreatic lipase activity, a key enzyme regulating the absorption of triacylglycerols. In contrast, none of the samples exhibited a significant anti-diabetic potential. Despite their high concentration compared to that of phenolic acids, results of this work do not support a significant antioxidant and pancreatic lipase inhibitory effect of durum wheat flavonoids. Therefore, breeding programs and animal and/or human trials related to the effect of durum wheat flavonoids on oxidative stress and absorption of triacylglycerols are discouraged at this point.

Abstract The Atlantic Forest is recognized as a biome rich in biodiversity. Cambuci trees (Campomanesia phaea) produce fruits that may be consumed as both fresh and processed forms but are still underutilized. The aim of this study was to describe the physicochemical characteristics and bioactive compounds in cambuci fruits from Brazilian Atlantic Forest, located at Natividade da Serra-SP, Brazil. Cambuci fruits of the fifty-nine accessions were characterized according to fresh weight, pulp yield, peel percentage, longitudinal and transversal diameters (LD and TD, respectively), pH, soluble solids content (SSC), titratable acidity (TA), SSC/TA ratio, ascorbic acid, total phenolic compounds content, and antioxidant activity using the DPPH assay. The soluble solids contents varied from 5.10 °Brix to 11.00 °Brix. The titratable acidity varied from 1.29 to 2.90 g citric acid 100 g-1. Ascorbic acid content was of 31.12 to 139.38 mg 100 g-1. Total phenolic compounds varied from 330.5 mg GAE 100 g-1 to 3,526.04 mg GAE 100 g-1. The antioxidant activity was of 65.03 µmol Trolox g-1 fruit pulp (fresh weight) to 199.78 µmol Trolox g-1 fruit pulp (fresh weight), a factor of 6.7 difference between the extremes. These results showed cambuci fruits presented as rich source of ascorbic acid, total phenolic compounds, and with high antioxidant capacity.

Abstract Pereskia aculeata Miller is a native cactaceae found from the Northeast to the South of Brazil. This plant is recognized by the high nutritional value of their leaves, which are the most used in cooking and folk medicine. However, studies on the chemical characteristics and antioxidant capacity of the ora-pro-nobis fruits are scarce. Therefore, the aim of this study was to determine the best harvest point of the ora-pro-nobis fruits to optimize the content of bioactive compounds and their antioxidant capacity. The fruits were manually harvested, defined by their peel colors in three maturity stages: unripe (green), intermediate (yellowish green), and ripe (yellow) stages. The following attributes were evaluated: soluble solids (SS), titratable acidity (TA), SS/TA ratio, pH, and content of bioactive compounds, such as total chlorophylls, carotenoids, yellow flavonoids, total phenolic compounds, and antioxidant capacity. There was a decrease in the content of chlorophyll followed by an increase in the of total carotenoids and yellow flavonoids, due to the maturity stage. For the total phenolic compounds, it was not observed significant difference between the stages. The antioxidant capacity was quantified by the ORAC method in lyophilized pulp in the unripe (1.95 mmol 100 g-1 trolox) and ripe (3.85 mmol 100 g-1 trolox) stages. The lowest contents were observed in the intermediate (0.80 mmol 100 g-1 trolox in lyophilized pulp) stage. The maturity stage significantly influenced the bioactive compounds of ora-pro-nobis, which presented higher values of total carotenoids, yellow flavonoids, and antioxidant capacity in ripe fruits.

Abstract Uvaia (Eugenia pyriformis Cambess) is a fruit tree native to the Atlantic Forest and that features peculiar aroma and flavor. Although this fruit is highly popular locally, its commercial farming is virtually nonexistent. In addition, limited studies on this fruit have been conducted. Therefore, the present study aimed to investigate the physical, chemical, and functional characteristics of two uvaia fruit populations grown in Salesópolis, SP, Brazil, in order to determine their potential for being consumed both in natura and processed. Uvaia fruits were evaluated as for skin color, whole fruit weight, longitudinal and transversal diameters, seed percentage, pulp yield, pulp firmness, pH, total soluble solids (TSS), total titratable acidity (TTA), TSS/TTA ratio, total soluble sugars, soluble and total pectins, total carotenoids, yellow flavonoids, total phenolic compounds, ascorbic acid, and in vitro antioxidant capacity (DPPH and ORAC assays). The total mineral content was also determined. Comparison among the two populations revealed that uvaia fruits showed significantly different firmness values, large fruits showing suitable characteristics for the fresh fruit market. However, small fruits were sweeter and richer in antioxidants, allowing the consumption of both natural and processed fruits. Thus, this native fruit denotes an important source of nutrients and bioactive compounds, especially iron.

Dental caries remains the most prevalent and costly oral infectious disease worldwide. Several methods have been employed to prevent this biofilm-dependent disease, including the use of essential oils (EOs). In this systematic review, we discuss the antibacterial activity of EOs and their isolated constituents in view of a potential applicability in novel dental formulations. Seven databases were systematically searched for clinical trials, in situ, in vivo and in vitro studies addressing the topic published up to date. Most of the knowledge in the literature is based on in vitro studies assessing the effects of EOs on caries-related streptococci (mainly Streptococcus mutans) and lactobacilli, and on a limited number of clinical trials. The most promising species with antibacterial potential against cariogenic bacteria are: Achillea ligustica, Baccharis dracunculifolia, Croton cajucara, Cryptomeria japonica, Coriandrum sativum, Eugenia caryophyllata, Lippia sidoides, Ocimum americanum, and Rosmarinus officinalis. In some cases, the major phytochemical compounds determine the biological properties of EOs. Menthol and eugenol were considered outstanding compounds demonstrating an antibacterial potential. Only L. sidoides mouthwash (1%) has shown clinical antimicrobial effects against oral pathogens thus far. This review suggests avenues for further non-clinical and clinical studies with the most promising EOs and their isolated constituents bioprospected worldwide.

This study aimed to evaluate the antioxidant, anti-inflammatory, and cytotoxic properties and phenolic composition of peel and seed of avocado varieties Hass and Fuerte using green solvents. Ethanol soluble compounds were identified in peel and seed of both varieties using HPLC-MS/MS and quantified using HPLC-DAD. Agro-industrial by-products of both varieties exhibited high radical scavenging activity against synthetic free radicals (DPPH and ABTS) and reactive oxygen species (peroxyl, superoxide, and hypochlorous acid) and high ability to reduce Fe3+ to Fe2+. The main compounds with significant contribution to the antioxidant activity determined by online HPLC-ABTS●+ analyses were procyanidin B2 and epicatechin in the peel and trans-5-O-caffeoyl-D-quinic acid, procyanidin B1, catechin, and epicatechin in the seed. Peel of Fuerte significantly suppressed TNF-α and nitric oxide (NO) release (459.3 pg/mL and 8.5 μM, respectively), possibly because of the high phenolic content and antioxidant activity detected. Avocado agro-industrial by-products can be used for food and pharmaceutical purposes due to their antioxidant and anti-inflammatory properties.

The profile of volatile compounds of yellow passion fruit juice was analyzed by solid phase microextraction headspace (HS-SPME) and optimized static headspace (S-HS) extraction techniques. Time, temperature, NaCl concentration and sample volume headspace equilibrium parameters was adjusted to the S-HS technique. The gaseous phase in the headspace of samples was collected and injected into a gas chromatograph coupled to a mass spectrometer. In the HS-SPME technique was identified 44 volatile compounds from the yellow passion fruit juice, but with S-HS only 30 compounds were identified. Volatile esters were majority in both techniques, being identified ethyl butanoate, ethyl hexanoate, (3z)-3-hexenyl acetate, hexyl acetate, hexyl butanoate and hexyl hexanoate. Aldehydes and ketones were not identified in S-HS, but were in HS-SPME. β-Pinene, p-cymene, limonene, (Z)-β-ocimene, (E)-β-ocimene, γ-terpinene, α-terpinolene and (E) -4,8-dimethyl-1, 3,7 - nonatriene terpenes were identified in both techniques. This study showed that the S-HS optimized extraction technique was effective to recovery high concentrations of the major volatile characteristics compounds in the passion fruit, such as ethyl butanoate and ethyl hexanoate, which can be advantageous due to the simplicity of the method.

Abstract We investigated the anti-caries effects of an experimental propolis varnish in vivo, and further tested its toxicity against fibroblasts. Fifty-six SPF female Wistar rats were infected with Streptococcus mutans UA159 (SM) and allocated into four groups (n = 14/group): G1, propolis varnish (15%/PV); G2, chitosan varnish (CV/vehicle); G3, gold standard (GS/Duraphat®); and G4, untreated. The animals received a single varnish application on their molars and were submitted to a high cariogenic challenge (Diet-2000, 56% sucrose, and 5% sucrose-added water, ad libitum) for 4 weeks. Total cultivable microbiota and SM were counted, and smooth-surface and sulcal caries were scored. PV, CV and GS cytotoxic effects were tested against fibroblasts. The data were analyzed using ANOVA with the Tukey-Kramer test (p ≤ 0.05). Total microbiota and SM counts did not differ among the treatments (p = 0.78), or in relation to the untreated group (p = 0.52). PV reduced development of smooth-surface enamel caries compared with the untreated group (p = 0.0018), with no significant difference from GS (p = 0.92); however, the PV effects were no longer observed when the dentin was affected. Neither PV nor GS prevented enamel sulcal lesion onset, but GS significantly reduced the severity of dentinal sulcal lesions (p < 0.0001). No significant difference was observed in fibroblast viability between PV and GS (p < 0.0001). In conclusion, PV prevented smooth-surface enamel caries and showed low cell toxicity. Nevertheless, due to the high cariogenic challenge, its effects were not sustained throughout the experiment. Further studies are encouraged to establish a protocol to sustain the long-term anti-caries activity of PV in the oral cavity.

O abacate é um fruto muito nutritivo, sendo a polpa a principal parte utilizada para consumo in natura, na forma de sobremesa, saladas, molhos e cosméticos, além de ser utilizada para extração de óleo. Os resíduos, casca e semente, ainda são pouco explorados cientificamente quanto ao seu potencial nutritivo e funcional. Neste trabalho, amostras liofilizadas da casca, polpa e semente de abacate 'Hass'foram submetidas a análises de composição centesimal, minerais, teor de compostos fenólicos totais e capacidade antioxidante. A polpa apresentou elevado teor de lipídios em relação à casca e à semente do abacate. Os elementos minerais foram superiores na casca dos abacates. A casca e a semente possuem maior teor de compostos fenólicos e atividade antioxidante em relação à polpa de abacate.

This study aimed to determine the antioxidant activity, total phenolic compounds and color in avocado ‘Hass’ hydrothermally treated. The fruits were hydrothermally treated at 45oC for 5, 10, 15 and 20 min. After treatment, fruit were stored at room temperature (21±1ºC and 70±5% relative humidity) and cold (10°C±1 and 90±5% relative humidity). The fruits were analyzed for their antioxidant capacity by DPPH method and phenolic compounds at 0, 3, 9 and 12 days. The fruits color was measured at 0, 3, 6, 9, 12 and 15 days. The control fruits had higher antioxidant capacity and content of phenolic compounds during the storage period, compared to the fruits hydrothermally treated. The hydrothermally treatment altered the behavior as for the maintenance of the antioxidant activity in relation to the fruits control. In spite of superior values of antioxidant activity for the fruits maintained at 21±1ºC and 70±5% relative humidity, those refrigerated presented better aspect for commercialization. The refrigerated fruits presented better aspect for commercialization in relation to the maintained under room temperature. The brightness, color a * and b * values decreased with the storage days. Values color superiors were observed for the fruits control and those maintained under refrigeration. As it increased the irradiation dose reduced the fruits antioxidant activity and coloration.

O Brasil, por ser um país de grande atividade agrícola, é um dos que mais produzem resíduos agroindustriais. Buscando alternativas para a aplicação destes subprodutos, o objetivo deste trabalho foi determinar o teor de compostos fenólicos e a atividade antioxidante de resíduos de abacaxi (casca e bagaço da polpa), maracujá (casca e semente), caju (bagaço da polpa) e manga (bagaço da polpa), provenientes de frutas utilizadas para fins industriais. Os mesmos foram congelados, liofilizados e armazenados a -18ºC. O teor de compostos fenólicos foi analisado pelo método de Folin-Ciocalteau, a atividade antioxidante foi avaliada pelos métodos do sequestro do radical livre DPPH, autoxidação do sistema β-caroteno/ácido linoleico e FRAP (Ferric Reducing Antioxidant Power). Com rela- ção aos compostos fenólicos, observou-se que o extrato de resíduo de caju apresentou o maior teor, seguido dos extratos de manga, maracujá e abacaxi. Entre as análises de atividade antioxidante, o método do sequestro do radical livre DPPH demonstrou maior correlação positiva com o conteúdo fenólico (r = 0,97). Tanto no ensaio DPPH, como no FRAP, o destaque foi o resíduo de caju. Já para o mé- todo de autoxidação do sistema β-caroteno/ácido linoleico, todos os resíduos, com exceção do caju, exibiram atividades significativas, inibindo a oxidação do carotenoide em valores próximos a 50% quando comparados ao controle. O estudo mostrou que esses resíduos apresentam atividade antioxidante e, portanto, potencial de utilização como fonte de antioxidantes naturais.
