Abstract

So far, no documented method for simultaneously analyzing lesinurad (LND) and febuxostat (FBX) has been reported for either traditional high-performance thin-layer chromatography (HPTLC) or a green HPTLC technique. In order to determine LND and FBX simultaneously in commercially available fixed-dose combo tablets, this study devised a normal-phase HPTLC method that is fast, sensitive, and green. The green eluents for the simultaneous analysis of LND and FBX were a mixture of ethyl acetate:ethanol:water at 70:20:10 (v/v/v) ratio. The new approach’s greenness was predicted utilizing four distinct greenness tools: the National Environmental Method Index, Analytical Eco-Scale, ChlorTox, and Analytical GREENness approaches, and the results revealed a significantly greener profile. The current method operated on a linear scale between 30 and 1,000 ng·band−1. It was confirmed that the current approach is sensitive, accurate, precise, robust, and green. The LND and FBX contents of commercially available tablet products A and B were found to be within the range of 100 ± 2%, indicating that the existing methodology for simultaneously determining LND and FBX in pharmaceutical combination products is applicable. The results of the current methodology indicated that LND and FBX could be consistently measured in pharmaceutical combination products simultaneously using the current approach.

Abstract

A fast, sensitive, and green reverse-phase “high-performance thin-layer chromatography” approach for the simultaneous estimation of ibuprofen (IBF), caffeine (CAF), and paracetamol (PCM) in marketed formulations was established and verified in this study. The binary combination of acetone and water (80:20 v/v) was used as the green eluent system. The current method’s greenness was predicted using four different approaches, namely National Environmental Method Index, Analytical Eco-Score (89), ChlorTox (1.08 g), and the Analytical GREENness (83) approaches, which demonstrated an outstanding greener profile. The present approach was linear in the range of 25–800 ng·band−1 for the simultaneous estimation of IBF, CAF, and PCM. In addition, the current method was accurate (% recoveries = 100 ± 2), precise (%CV < 2%), robust (%CV < 2), sensitive (LOD = 1.13–2.71 ng·band−1 and LOQ = 3.39–8.10 ng·band−1), and green. The amount of IBF, CAF, and PCM in commercial tablets was determined to be 99.51%, 98.25%, and 100.64%, respectively. The present method for the simultaneous determination of IBF, CAF, and PCM in marketed tablets is supported by these data. The findings of this study suggested that the current approach may be consistently applied to analyze IBF, CAF, and PCM in marketed tablets.

31. In-vitro antioxidant and anti-proliferative activity of aerial parts of Senecio Laetus Edgew on breast cancer (MCF-7) and colon carcinoma (HCT116) cell lines.

Abstract

The present study aimed to perform phytochemical investigation of various fractions of the aerial parts of Senecio laetus Edgew for its bio-constitution and to pharmacologically evaluate these fractions for antioxidant and antiproliferative activities against breast cancer (MCF-7) and colon carcinoma (HCT-116) cell lines. Phytochemical screening and characterization of compounds was carried out as per standard procedures. Extracts were subjected to GC-MS analysis for identification and characterization of constituents. The antioxidant activity was carried out using DPPH, ferric ion reducing power assay, and nitric oxide radical inhibition assays. In vitro antiproliferative activity of the extracts was carried out using MCF-7 (breast cancer) and HCT-116 (colon carcinoma) cancer cell lines by MTT assay, colony formation assay, and wound healing assay. Phytochemical screening showed the presence of alkaloids, flavonoids, carbohydrates, glycosides (cardiac and anthraquinone), tannins, triterpenoids, phenolic compounds, and phytosterols. GC-MS analysis showed the presence of 54 compounds. Ethyl acetate and methanolic extract exhibited the maximum amount of phenolic content compared to dichloromethane and hexane fractions. Antioxidant capacities were shown highest in ethyl acetate and methanolic fractions. The antiproliferative activity was found to be concentration dependent, with dichloromethane fraction more effective than ethyl acetate and hexane fractions against both the MCF-7 and HCT-116 cancer cell lines. The results indicate that S. laetus Edgew has a promising antioxidant and antiproliferative potential as shown by its activity against MCF-7 (breast carcinoma) and HCT-116 (colon carcinoma) cancer cell lines. Abstract

We attempted to synthesize nickel oxide nanoparticles (NiO-NPs) utilizing waste Arachis hypogaea (peanut) shell extract and studied their structural, morphological, and biological performance for biomedical applications. The green engineered NiO-NPs possessed a face-centered cubic structure with an average particle size of 20 nm in highly crystalline form. NiO-NPs were shown to have an optical resonance peak at 327 nm with 3 eV as the optical band gap according to the UV–visible spectra, and the stretching band between Ni–O were evidenced from the FTIR and Raman spectrum. Utilizing green approach the stable nanoparticles were obtained with average particle size of 31 nm from SEM analysis; zeta potential value of −17.6 mV, and PDI as 0.68, revealed the formation of spherical nanoparticles with distinct morphologies without aggregation. XPS analysis confirmed the oxidation states of the elements Ni (2p) and O (1s). This approach may help to increase the surface area, increasing the possibility of nanoparticles interacting with bacterial cells. Furthermore, the presence of nickel and the oxygen oxidation state were confirmed by XPS. Proteus vulgaris, Streptococcus oralis, Bacillus subtilis, and Escherichia coli were found to be susceptible to the antibacterial action of the produced NiO-NPs, with a maximal zone of inhibition of 10.25 mm at 500 μg/ml for P. vulgaris. For P. vulgaris and E. coli, the minimum inhibitory concentrations of NiO were 5.36 and 12.55 %, respectively, at 31.25 μg mL−1.
