Abstract

Xanthine is a significant biomolecule and its concentration level in urine and blood plasma is an indicator of specified pathological states. Here, a new sensing platform was designed, which showed excellent analytical performance for xanthine. Importantly, it is the first time to investigate the kinetics of xanthine metabolic reaction by electrochemical method. The results demonstrated that the conversion of xanthine to uric acid completely conformed to the Michaelis‐Menten kinetics. Furthermore, we also studied the inhibitory effect of febuxostat on xanthine oxidase activity detailed. As expected, the work may offer potential value for researchers in the treatment of hyperuricemia and gout. [ABSTRACT FROM AUTHOR]

Abstract

Polyfluoroaryl compounds belong to privileged moieties and engender distinct properties in many pharmaceuticals, agrochemicals, and materials. Over the past decade, considerable seminal reports and reviews have merely paid close attention to fluoroalkylation chemistry, such as trifluoromethylation, difluoroalkylation, perfluoroalkylation, trifluoromethylthiolation or trifluoromethoxylation. Polyfluoroarylation has inevitably lagged somewhat behind its fluoroalkylation counterparts, emanating from a lack of awareness and understanding of the synthetic significance. Together with the renaissance of photochemistry, the photocatalytic polyfluoroarylation using polyfluoroarenes as an inexpensive and easy-available radical precursor has emerged as a topical interest and vibrant area of chemical research. In this review, we have endeavored to present the state-of-the-art in photocatalytic polyfluoroarylation since 2014, and the discussions cover the basic concept, reaction design, mechanistic insight, and research prospects, which are organized by the reaction types. Abstract

Real-time monitoring reactive oxygen species (ROS) released from living cells is important in both physiological and pathological contexts. The excessive amount of ROS accumulation in cells could lead to a series of cell damage and cause aging and disease. Here we present design, fabrication, and successful testing of a highly sensitive electrochemical hydrogen peroxide sensor based on a three-dimensional (3D) platinum nanoparticles (PtNPs) decorated porous graphene composite. Scanning electron microscopy (SEM) investigations indicate that PtNPs were embedded into porous graphene (PGN), something that was rarely reported. With the help of the excellent catalytic activity of PtNPs and unique porous structure in combination with inherent properties of graphene, the PtNPs/PGN based sensor presents efficient electrochemical performance for H2O2 sensing with a detection limit of 0.02 nM, the lowest detection limit achieved in nonenzymatic electrochemical sensors so far, which guarantees the possibility of sensitive and reliable detection of H2O2 release from living cells. This work opens a new avenue for direct preparation of metal nanoparticle-embedded porous graphene composite. More importantly, the presented sensors can be successfully used for monitoring changes of H2O2 levels upon different stimuli and collecting kinetic information on cellular ROS release, which provides more opportunities to study its cellular biochemistry.

20. A Novel One-Pot Five-Component Synthesis of Tetrahydro-pyrrolo[3,4-b]pyridine-5-one via Ugi/Aza-Diels–Alder Tandem Reaction

Abstract

A novel five-component domino processes to tetrahydro-pyrrolo[3,4-b]pyridine-5-one (8) were developed. Reaction of 5-phenyloxazole-2-carbaldehyde (1), p-toluidine (2), (Z)-4-methoxy-4-oxobut-2-enoic acid (3), and t-butyl isocyanide (4) in methanol at 60 °C provides an efficient access to the drug-like bicycle compound (8) in a good yield. In this one-pot process, a traditional oxo-bridged tricycle intermediate was formed via Ugi/ara-Diels–Alder tandem reaction and then the solvent, methanol, sequentially attacks and cuts off the oxo bridge under mild conditions via SN2 reaction. Formation of one C–N, two C–O, and three C–C bond with the triple domino sequence (Ugi/IMDA/SN2) is involved in this new scaffold generating reaction. The operational simplicity and maximizing the buildup of structural complexity makes this novel multicomponent reaction valuable in diversity-oriented synthesis of tetrahydro-pyrrolo[3,4-b]pyridine-5-one.
