13. The Glioma Model micro-PET Imaging and Biodistribution of 68Ga Labeled Fibroblast Activation Protein Inhibitor

Abstract

To investigate the biodistribution and micro-PET imaging of 68Ga labeled fibroblast activation protein inhibitor (68Ga-FAPI-04) in normal mice and glioma nude mice, 68Ga-FAPI-04 was synthesized using DOTA-modified fibroblast activation protein inhibitor as precursor. The synthesis yield, radiochemical purity and in vitro stability were analyzed by radio-HPLC, and the hydrophilicity of 68Ga-FAPI-04 was evaluated by lipo-hydro partition coefficient. Twenty ICR mice were randomly divided into 5 groups. 3.7 MBq of 68Ga-FAPI-04 was injected by tail vein per mouse, then the mice were killed at 5, 15, 30, 60, 120 min post-injection(pi.). The main organs and tissues were collected and weighed, then the radioactivity was measured, and the uptake of each tissue or organ was calculated. The nude mice bearing U87MG glioma model were established, then the biodistribution study and micro-PET imaging were performed. The results indicate that the synthesis yield of 68Ga-FAPI-04 was (97.38±1.32)% (n=3), the radiochemical purity was 100%，and the in vitro stability and hydrophilicity were good. The biodistribution study of normal-ICR mice showed that 68Ga-FAPI-04 was cleared from blood rapidly and mainly excreted from kidneys, and the radioactivity uptake in brain was low. The biodistribution and micro-PET of U87MG tumor-bearing nude mice showed high radioactivity at the tumor targets. The uptake of 68Ga-FAPI-04 at the tumor targets reached (2.50±0.00)%ID/g at 90 min pi. The tumor-to-background ratios (TBRs) were（6.26±0.09）, （5.06±0.02）, （5.54±1.47） and （5.51±0.03） at 30, 60, 90 and 120 min pi., respectively. The preliminary studies indicate that 68Ga-FAPI-04 is synthesized easily with high yields, and is stable in vitro. The radiotracer was mainly excreted from kidneys, and targeting tumor targets with clear images in micro-PET of glioma models. Therefore, 68Ga-FAPI-04 might be a potential brain tumor imaging agent.

33. Deposition of thin TiO2films on fluorophlogopite particle surface by CVD in fluidized bed reactor to prepare silver pearlescent pigments

Abstract

Two critical parameters: Glossiness and chromaticity space of pearlescent pigments were evaluated based on crystallinity and grain size. Both the roughness and the vertical distance evolution along the surface of deposited TiO2film are affected by the crystallinity. The optimal crystallinity of TiO2was deposited at the temperature of 350 °C. The TiCl4concentration effectively changed the grain size. However, the high flow rate of fluidizing gas and optimal gas–solid ratio (<0.11 m3/(kg h)) restricted the change of TiCl4concentrations in fluidized bed reactor. In this case, the grain size of the deposited TiO2film was about 55 nm. Depending on the grain size, the color of pearlescent pigments was adjusted by controlling the film thickness based on TiO2concentration. Finally, the silver pearlescent pigment with high glossiness was prepared by chemical vapor deposition method in a fluidized bed reactor.

37. Biotin-Functionalized Iridium-Based Nanoparticles as Tumor Targeted Photosensitizers for Enhanced Oxidative Damage in Tumor Photodynamic Therapy.

Abstract

The clinical application of photodynamic therapy (PDT) is hampered by the suboptimal tumor-targeting properties of photosensitizers (PSs) and the presence of abundant antioxidants in cancer cells. To overcome these limitations, in this study, we proposed a formulation of an Ir-(III) complex into a promising tumor-targeted coordination polymer (Ir-PB), which could self-assemble into nanoparticles (Ir-PB NPs) for bioimaging and tumor-targeted PDT. Ir-PB NPs were found to efficiently target A549 cells and specifically accumulate in the mitochondria. Under light irradiation (400 nm, 10 mW/cm2, 15 min), Ir-PB NPs could trigger cell death through a combination of apoptosis and ferroptosis pathways by the photoinduced depletion of nicotinamide adenine dinucleotide phosphate (NADPH) and glutathione (GSH) and the photoinduced production of single oxygen (1O2). The mechanisms featured the attenuation of the antioxidant capacity of tumor cells, the reduction of mitochondrial membrane potential (MMP), decrease in ATP production, activation of Caspase-3 protein, accumulation of lipid peroxidation (LPO), shrinkage of mitochondria, and down-regulation of glutathione peroxidase 4 (GPX4). The effectiveness of Ir-PB NPs in inhibiting tumor growth upon light irradiation was confirmed using an A549 xenograft mouse model. This study presents a promising strategy for developing photodynamic drugs that have excellent tumor-targeting abilities and enhance the efficiency of PDT. [ABSTRACT FROM AUTHOR]

Abstract

The performance of pearlescent pigment significantly affected by the grain size and the roughness of deposited film. The effect of TiCl4concentration on the initial deposition of TiO2on mica by atmospheric pressure chemical vapor deposition (APCVD) was investigated. The precursor concentration significantly affected the deposition and morphology of TiO2grains assembling the film. The deposition time for fully covering the surface of mica decreased from 120 to 10 s as the TiCl4concentration increased from 0.38% to 2.44%. The grain size increased with the TiCl4concentration. The AFM and TEM analysis demonstrated that the aggregation of TiO2clusters at the initial stage finally result to the agglomeration of fine TiO2grains at high TiCl4concentrations. Following the results, it was suggested that the nucleation density and size was easy to be adjusted when the TiCl4concentration is below 0.90%.

Abstract

The efficient utilization of manganese dioxide (MnO2) ore is essential for the sustainable development of manganese (Mn) industry. Confronting the great challenge of chemical engineering scale-up, a commercial fluidized reduction project of MnO2 ore with the capacity of 200,000 t a-1 is carried out based on deep experimental investigation, extensive kilogram-scale test and detailed engineering design. Compared with other production technologies and equipment, it is proved that the fluidized process shows distinguished advantages of lower energy consumption, higher production efficiency, larger automation degree and less environmental pollution. The comprehensive studies of experiment, modeling, simulation and optimization are required for a more promising development of fluidization engineering in the future.
