Abstract

The "2-variable general-$\lambda$-matrix polynomials (2VG$\lambda$MP)" is a new family of matrix polynomials, introduced and studied in this article. These matrix polynomials are constructed using umbral and symbolic methods. We delve into the generating function, explicit series representation, differential equation, quasi-monomiality, summation formula, determinant representation, and various identities satisfied by these polynomials. Furthermore, a thorough investigation is conducted to ascertain the outcomes for the members of the general-$\lambda$-matrix family. Abstract

Interactions between adipose tissue, liver and immune system are at the center of metabolic dysfunction-associated steatotic liver disease and type 2 diabetes. To address the need for an accurate in vitro model, we establish an interconnected microphysiological system (MPS) containing white adipocytes, hepatocytes and proinflammatory macrophages derived from isogenic human induced pluripotent stem cells. Using this MPS, we find that increasing the adipocyte-to-hepatocyte ratio moderately affects hepatocyte function, whereas macrophage-induced adipocyte inflammation causes lipid accumulation in hepatocytes and MPS-wide insulin resistance, corresponding to initiation of metabolic dysfunction-associated steatotic liver disease. We also use our MPS to identify and characterize pharmacological intervention strategies for hepatic steatosis and systemic insulin resistance and find that the glucagon-like peptide-1 receptor agonist semaglutide improves hepatocyte function by acting specifically on adipocytes. These results establish our MPS modeling the adipose tissue-liver axis as an alternative to animal models for mechanistic studies or drug discovery in metabolic diseases.

Abstract

Microbial sensors play an essential role in maintaining cellular homoeostasis. Our knowledge is limited on how microbial sensing helps in differential immune response and its link to inflammatory diseases. Recently we have confirmed that ELMO1 (Engulfment and Cell Motility Protein-1) present in cytosol is involved in pathogen sensing, engulfment, and intestinal inflammation. Here, we show that ELMO1 interacts with another sensor, NOD2 (Nucleotide-binding oligomerization domain-containing protein 2), that recognizes bacterial cell wall component muramyl dipeptide (MDP). The polymorphism of NOD2 is linked to Crohn's disease (CD) pathogenesis. Interestingly, we found that overexpression of ELMO1 and mutant NOD2 (L1007fs) were not able to clear the CD-associated adherent invasive E. coli (AIEC-LF82). The functional implications of ELMO1-NOD2 interaction in epithelial cells were evaluated by using enteroid-derived monolayers (EDMs) from ELMO1 and NOD2 KO mice. Subsequently we also assessed the immune response in J774 macrophages depleted of either ELMO1 or NOD2 or both. The infection of murine EDMs with AIEC-LF82 showed higher bacterial load in ELMO1-KO, NOD2 KO EDMs, and ELMO1 KO EDMs treated with NOD2 inhibitors. The murine macrophage cells showed that the downregulation of ELMO1 and NOD2 is associated with impaired bacterial clearance that is linked to reduce pro-inflammatory cytokines and reactive oxygen species. Our results indicated that the crosstalk between microbial sensors in enteric infection and inflammatory diseases impacts the fate of the bacterial load and disease pathogenesis.

Abstract

Chronic white adipose tissue (WAT) inflammation has been recognized as a critical early event in the pathogenesis of obesity-related disorders. This process is characterized by the increased residency of proinflammatory M1 macrophages in WAT. However, the lack of an isogenic human macrophage-adipocyte model has limited biological studies and drug discovery efforts, highlighting the need for human stem cell-based approaches. Here, human induced pluripotent stem cell (iPSC) derived macrophages (iMACs) and adipocytes (iADIPOs) are cocultured in a microphysiological system (MPS). iMACs migrate toward and infiltrate into the 3D iADIPOs cluster to form crown-like structures (CLSs)-like morphology around damaged iADIPOs, recreating classic histological features of WAT inflammation seen in obesity. Significantly more CLS-like morphologies formed in aged and palmitic acid-treated iMAC-iADIPO-MPS, showing the ability to mimic inflammatory severity. Importantly, M1 (proinflammatory) but not M2 (tissue repair) iMACs induced insulin resistance and dysregulated lipolysis in iADIPOs. Both RNAseq and cytokines analyses revealed a reciprocal proinflammatory loop in the interactions of M1 iMACs and iADIPOs. This iMAC-iADIPO-MPS thus successfully recreates pathological conditions of chronically inflamed human WAT, opening a door to study the dynamic inflammatory progression and identify clinically relevant therapies.

Abstract

Purpose: To evaluate the alterations in the psycho-physical tests, contrast sensitivity (CS) and hue discrimination (total estimation score, TES) in COVID-19 patients. Methods: A prospective case-control study was undertaken in 2021-22 to look at CS and TES among COVID-19 patients and RT-PCR negative normal subjects by a mobile app "smart optometry" and X-Rite's free online version of Farnsworth-Munsell 100 hue test. Strict visual acuity (minimum of 6/9 equivalent on Snellen) and other clinical parameters were used as exclusion criteria to filter potentially confounding pre-receptoral co-morbid conditions. The effect of mismatch in recruiting age and sex matched controls during the pandemic were analyzed by multivariate linear regression. Result: One-way ANOVA ruled out any influence of gender on CS and TES; however, there was significant difference in the TES on Mann Whitney U test (TES- 2.95 +/- 3.8 for cases; 0.30 +/- 1.1 for controls; p Conclusion: The study demonstrates that COVID-19 infection leads to altered TES representing dyschromatopsia -an ocular counterpart of anosmia, with little difference on CS. 49. Investigating physiological effects of weight loss on male fertility

Abstract

Infertility is an emotionally devastating condition for a couple. It is defined as the inability to conceive following 1 year of regular unprotected intercourse. Infertility affects 15% of couples with nearly 50% of cases due to poor sperm quality in the male partner. i.e. 'male factor infertility'. There are currently no approved pharmacological therapies to directly stimulate spermatogenesis; anti-oestrogens and aromatase inhibitors have limited effectiveness for the treatment of oligospermia and their usage is not supported by current clinical guidelines. Consequently, the only therapeutic option for male factor infertility is assisted reproductive technologies (ART), such as intra-cytoplasmic sperm injection (ICSI) which although effective are resource limiting and unaffordable for many couples worldwide. Therefore, there exists an important and unmet need to develop practical and cost-effective therapies for male factor infertility. Over the last 50 years, whilst sperm quality has declined, obesity has doubled in prevalence. Evidence suggests an association between obesity and male infertility, which makes weight loss a plausible answer to this rising endemic problem. Currently, bariatric surgery is the most effective treatment for obesity leading to major weight loss. However, effects on semen parameters are controversial, with some studies suggesting that the acute starvation-like state induced by bariatric surgery paradoxically reduces sperm function. Recent observational reports have suggested that milder dietary weight loss is associated with improved semen quality and DNA fragmentation index (DFI) in men with obesity and infertility. Low energy diet (LED) is a safe, well-tolerated and established method of achieving modest weight loss which could therefore provide a novel, non-pharmacological therapy for men with obesity-related male factor infertility. However to date, there are currently no prospective randomised controlled studies investigating whether weight loss via LED can improve sperm quality in obese men. Additionally, it is unclear what level of weight loss would be ideal to optimise sperm quality in obese men. This thesis outlines the first ever three randomised controlled studies investigating the physiological effects of weight loss by LED on sperm quality in obese fertile (study 1 and 2) and infertile men (study 3) respectively. I hypothesized that some, but not all, degrees of weight loss would significantly improve sperm concentration in men with obesity, with a potential threshold of weight loss in men leading to improvements 3 in sperm quality. I have measured novel molecular markers, such as seminal reactive oxygen species (ROS) and DFI, associated with male infertility in obese men undergoing weight loss. Collectively, these results will extend our understanding of the physiological effects of weight loss on sperm quality in obese men. This could potentially lead to larger studies determining the effect of weight loss on live birth rates in couples affected by obesity-related male infertility.

In this paper, we study the Li-Yorke chaotic composition operators on Orlicz-Lorentz space. In fact, necessary and sufficient conditions are given for Li-Yorke chaotic composition operator $C_{\tau}$ on $\mathbb{L}^{\varphi,h}(\mu)$. Further, we present the equivalent conditions for $C_{\tau}$ to be Li-Yorke chaotic. This paper's results are the generalization of results of [15] into Orlicz-Lorentz spaces.
