8. The effect of Lebanese public transport on visitor's satisfaction

Abstract

This study measures the effect of public transport on overall visitors' satisfaction with Lebanon. The target sample represented n=250 tourists and residents of Lebanon. The instrument was composed of two parts. Excluding demographics all items were answered on a seven points likert type scale. Factor analysis was conducted where the Varimax method of rotation was used as a principal components method of extraction. Six performance dimensions of public transport were identified: safety and efficiency ease of use, cleanliness, price value, and difficulty to reach. Factors with eigenvalues greater than or equal to 1.0 were considered significant. Overall satisfaction with Lebanon as a tourist destination is negatively affected by safety and efficiency of transport mode, ease of use of transportation in Lebanon, and ease of access to attractions. Cleanliness and difficulty to reach the attraction positively affected overall satisfaction with Lebanon as a tourist destination. The study also revealed that the price value of transportation has no effect on the overall satisfaction with Lebanon as a tourist destination; therefore, it was eliminated from the results. This study concludes that the performance of public transport has an effect on the overall satisfaction with Lebanon as a destination. Abstract

Beş Salmonella enterica serovarları, biyofilm oluşturma yetenekleri açısından test edilmiştir, bunların arasında S. Enteritidis 6 (2014) ve S. Enteritidis (86) suşlarının, en yüksek biyofilm oluşturma kabiliyetine sahip olduğu görülmüştür. Bu nedenle bu iki suş ileri denemeler için seçilmiştir. Bir GST füzyon proteini olarak pGEX-4T-1 vektörüne klonlanmış olan SipA proteinin optimum üretimi, Rosetta hücreleri kullanılarak elde edilmiştir. Saflaştırılan SipA proteini, SELEX yöntemi kullanılarak aptamerler geliştirmek için kullanılmıştır. Karakterize edilmiş Apt17 inhibisyon çalışmalarında kullanılmıştır. Apt17 (2 µM) S. Enteritidis 6 (2014) ve S. Enteritidis (68) suşları ile birlikte inkübe edildiğinde sırasıyla %24.34 ve %26.81 oranında biyofilm oluşumunu inhibe ettiği görülmüştür. SEM ile biyofilm oluşumu analiz edildiğinde, Apt17 ile muamele edilen S. Enteritidis 6 (2014) biyofilmlerinin 3D yapıyı oluşturmadığı ve matriks yapısının da kontrol ile karşılaştırıldığında aynı olmadığı gözlemlenmiştir. Öte yandan, Apt17 ile muamele edilen S. Enteritidis (68) hücrelerinin cam yüzeyi kaplamadığı da görülmüştür. Yapılan adezyon ve invazyon analizlerinde ise Apt17 (2 µM), Caco-2 hücre hattının adezyonunu % 25–70 oranında ve invazyonunu ise % 18.03 – 39.5 oranında inhibe etmiştir., Five Salmonella enterica serovars were tested for their biofilm formation ability. Among which, S. Enteritidis 6(2014) and S. Enteritidis (86) demonstrated the highest biofilm formation ability. These two most potent biofilm formers were chosen for further experiments. Optimum production of SipA, cloned as a GST fusion protein into pGEX-4T-1 vector, was obtained when Rosetta cells were incubated over night at 28°C. Aptamers were developed against the purified protein by Systematic Evolution of Ligands by Exponential Enrichment. The characterized Apt17 was applied in inhibition studies. Maximum inhibition percentages, equivalent to (24.34%) and (26.81%), were recorded when Apt17 (2 µM) was co-incubated with S. Enteritidis 6(2014) and S. Enteritidis (68) respectively. SEM analysis revealed that treated S. Enteritidis 6(2014) biofilms didn't form the 3D structure and matrix wasn't evident. On the other hand, Apt17 reduced the surface coverage of Salmonella Enteritidis (68) cells on the attachment surface. A smaller diameter of cell aggregates was recorded in the treated specimens of both tested strains. Moreover, Apt 7 (2µM) inhibited the adhesion and invasion of Caco-2 cell line by 25 to 70 % and 18.03 to 39.5 % respectively
