Abstract We investigate the multifusion generalization of string-net ground states and lattice Hamiltonians, delving into their associated weak Hopf symmetries. For the multifusion string-net, the gauge symmetry manifests as a general weak Hopf algebra, leading to a reducible vacuum string label; the charge symmetry, serving as a quantum double of gauge symmetry, constitutes a connected weak Hopf algebra. This implies that the associated topological phase retains its characterization by a unitary modular tensor category (UMTC). The bulk charge symmetry can also be captured by a weak Hopf tube algebra. We offer an explicit construction of the weak Hopf tube algebra structure and thoroughly discuss its properties. The gapped boundary and domain wall models are extensively discussed, with these 1d phases characterized by unitary multifusion categories (UMFCs). We delve into the gauge and charge symmetries of these 1d phases, as well as the construction of the boundary and domain wall tube algebras. Additionally, we illustrate that the domain wall tube algebra can be regarded as a cross product of two boundary tube algebras. As an application of our model, we elucidate how to interpret the defective string-net as a restricted multifusion string-net.

ABSTRACT Background Diabetic peripheral neuropathy (DPN) and diabetic nephropathy (DN) are common complications of type 2 diabetes mellitus (T2DM). Although nerve conduction studies (NCS) and sympathetic skin response (SSR) can detect DPN, the more sensitive method for early diagnosis remains unclear. Furthermore, whether DPN can be used as a predictor for diabetic nephropathy needs clarification. Methods We evaluated nerve conduction studies, sympathetic skin response, and the diabetic nephropathy indicator microalbuminuria (MAU) in 192 patients with type 2 diabetes mellitus and 50 healthy controls. Results Patients with type 2 diabetes mellitus showed a lower sensory nerve conduction velocity (SCV), sensory active nerve potential (SNAP), motor nerve conduction velocity (MCV), and compound motor action potential (CMAP) than the controls on NCS. Abnormal rates for nerve conduction studies and sympathetic skin response were 75.0% and 83.3%, respectively, in patients with type 2 diabetes mellitus. Interestingly, 54.2% of patients with normal nerve conduction studies had an abnormal sympathetic skin response. Moreover, we found a positive correlation between sympathetic skin response and microalbuminuria for the first time. The abnormal rate of microalbuminuria was 53.8%, lower than that of abnormal nerve conduction studies or sympathetic skin response patients. Conclusion Sympathetic skin response is a more sensitive method than nerve conduction studies for the early diagnosis of diabetic peripheral neuropathy. Abnormal sympathetic skin response might serve as an indicator for early diabetic nephropathy. Additionally, diabetic peripheral neuropathy may occur earlier than diabetic nephropathy in the development of type 2 diabetes mellitus.

The damage of integrated epithelial epithelium is a key pathogenic factor and closely associated with the recurrence of ulcerative colitis (UC). Here, we reported that vanillic acid (VA) exerted potent therapeutic effects on DSS-induced colitis by restoring intestinal epithelium homeostasis via the inhibition of ferroptosis. By the CETSA assay and DARTS assay, we identified carbonic anhydrase IX (CAIX, CA9) as the direct target of VA. The binding of VA to CA9 causes insulin-induced gene-2 (INSIG2) to interact with stromal interaction molecule 1 (STIM1), rather than SREBP cleavage-activating protein (SCAP), leading to the translocation of SCAP-SREBP1 from the endoplasmic reticulum (ER) to the Golgi apparatus for cleavage into mature SREBP1. The activation of SREBP1 induced by VA then significantly facilitated the transcription of stearoyl-CoA desaturase 1 (SCD1) to exert an inhibitory effect on ferroptosis. By inhibiting the excessive death of intestinal epithelial cells caused by ferroptosis, VA effectively preserved the integrity of intestinal barrier and prevented the progression of unresolved inflammation. In conclusion, our study demonstrated that VA could alleviate colitis by restoring intestinal epithelium homeostasis through CA9/STIM1-mediated inhibition of ferroptosis, providing a promising therapeutic candidate for UC.

Abstract The generalized quantum double lattice realization of 2d topological orders based on Hopf algebras is discussed in this work. Both left-module and right-module constructions are investigated. The ribbon operators and the classification of topological excitations based on the representations of the quantum double of Hopf algebras are discussed. To generalize the model to a 2d surface with boundaries and surface defects, we present a systematic construction of the boundary Hamiltonian and domain wall Hamiltonian. The algebraic data behind the gapped boundary and domain wall are comodule algebras and bicomodule algebras. The topological excitations in the boundary and domain wall are classified by bimodules over these algebras. The ribbon operator realization of boundary-bulk duality is also discussed. Finally, via the Hopf tensor network representation of the quantum many-body states, we solve the ground state of the model in the presence of the boundary and domain wall.

In order to study the impact initiation process and mechanism of hypervelocity PTFE/Al composite structure reactive fragments on a shielded charge, first, an existing PTFE/Al reactive fragment hypervelocity collision experiment was numerically simulated using the SPH algorithm in ANSYS/AUTODYN 17.0 software. Then, the Lee–Tarver model was verified to describe the detonation reaction behavior and explosion damage effect of reactive materials. A numerical simulation analysis of the impact of two kinds of ultra-high-speed PTFE/Al composite-structure reactive fragments on a shielded charge was carried out using the SPH algorithm. These were steel-coated PTFE/Al and steel-semi-coated PTFE/Al fragments, and they were compared with the impact of steel fragments. The results indicate that the threshold velocities of the impact initiation of the two composite-structure reactive fragments on the shielded charge were both 2.6 km/s, while the threshold velocity of the steel fragment was 2.7 km/s. Under the threshold velocity condition, the two composite-structure reactive fragments increase the time and intensity of the compressed shock wave pulse in the explosive due to the impact energy release effect of the reactive materials, causing the shielded charge to detonate under the continuous long-term pulse loads. However, the mechanism of the steel fragment on the shielded charge belongs to the shock–detonation transition. The research results can provide scientific references for the design of hypervelocity reactive fragments and the study of their damage mechanism.

Abstract High-temperature ablation is a common failure phenomenon that limits the service life of the transmission parts on heavy-duty machines used in heavy load, high temperature, high shock conditions due to in-sufficient supply of lubricating oil and grease. Traditional self-lubricating coatings prepared by inorganic, organic or organic-inorganic hybrid methods are prone to be oxidated at high temperatures to lose their friction reducing function, so that it is difficult to meet the engineering requirements of high-temperature lubrication. We design viscoelastic polymer coatings by a high-temperature self-lubricating and wear-resistant strategy. Polytetrafluoroethylene (PTFE, T m = 329 °C) and polyphenylene sulfide (PPS, T g = 84 °C, T m = 283 °C) are used to prepare a PTFE/PPS polymer alloy coating. As the temperature increases from 25 to 300 °C, the PTFE/PPS coating softens from glass state to viscoelastic state and viscous flow state, which is owing to the thermodynamic transformation characteristic of the PPS component. Additionally the friction coefficient (µ) decreased from 0.096 to 0.042 with the increasing of temperature from 25 to 300 °C. The mechanism of mechanical deformation and surface morphology evolution for the PTFE/PPS coating under the multi-field coupling action of temperature (T), temperature-centrifugal force (T-F ω ), temperature-centrifugal force-shearing force (T-F ω -F τ ) were investigated. The physical model of “thermoviscoelasticity driven solid-liquid interface reducing friction” is proposed to clarify the self-lubricating mechanism determined by the high-temperature viscoelastic properties of polymers. The high-temperature adjusts the viscosity (η) of the coating, increases interface slipping and intensifies shear deformation (τ), reducing the friction coefficient. The result is expected to provide a new idea for designing anti-ablation coatings served in high temperature friction and wear conditions.

IntroductionGrass carp reovirus (GCRV), a member of the Aquareovirus genus in the Reoviridae family, is considered to be the most pathogenic aquareovirus. Productive viral infection requires extensive interactions between viruses and host cells. However, the molecular mechanisms underlying GCRV early infection remains elusive.MethodsIn this study we performed transcriptome and DNA methylome analyses with Ctenopharyngodon idellus kidney (CIK) cells infected with GCRV at 0, 4, and 8 h post infection (hpi), respectively.ResultsWe found that at early infection stage the differentially expressed genes related to defense response and immune response in CIK cells are activated. Although DNA methylation pattern of CIK cells 8 hpi is similar to mock-infected cells, we identified a considerable number of genes that selectively utilize alternative polyadenylation sites. Particularly, we found that biological processes of cytoskeleton organization and regulation of microtubule polymerization are statistically enriched in the genes with altered 3’UTRs.DiscussionOur results suggest that alternative polyadenylation potentially contributes to GCRV early infection.

Abstract Ischemic stroke is highly concerning because it often leads to severe long-term neurological disability. Among clinical trials, ischemic stroke and inflammatory bowel disease interactions have been increasingly reported in recent years. Therefore, using bioinformatics approaches to explore novel protein interactions between them is of interest. We performed this exploratory analysis by using bioinformatics tools such as string to analyze gene data downloaded from NHGRI-GWAS data related to ischemic stroke and inflammatory bowel disease. We constructed a prospective protein interaction network for ischemic stroke and inflammatory bowel disease, identifying cytokine and interleukin-related signaling pathways, Spliceosome, Ubiquitin–Proteasome System (UPS), Thrombus, and Anticoagulation pathways as the crucial biological mechanisms of the network. Furthermore, we also used data-independent acquisition mass spectrometry (DIA-MS) to detect differential protein expression in eight samples, which also suggested that immune system, signal transduction, and hemostasis-related pathways are key signaling pathways. These findings may provide a basis for understanding the interaction between these two states and exploring possible molecular and therapeutic studies in the future.

Abstract Bionic self‐cleaning coatings have exhibited scaling inhibition for their low surface free energy. The key issue is focused on the long‐term anti‐scaling performance for the hydrophobicity failure after scaled. Inspired by “grass‐to‐stone” phenomenon, we reported a polymer coating (WCA = 120°, WSA = 14°) with the water contact angle of its 720 h‐scaled surface increased to 135°and scaling reduced by 90–95%. The nucleation and growth of CaCO3 at “coating‐crystal” interface are investigated, and a physical model for self‐healing film interface inhibiting scaling is innovatively proposed, as an important step toward designing long‐term anti‐scaling coating for engineering application.

Abstract Droplet bouncing on special wettability surface is common and important in daily life and engineering applications (anti‐icing and drag reduction, etc.). However, there are few studies on the multiple droplets impact and there is a lack of systematic research on the interaction of oscillation and bounce. Here, the impact and oscillation of multiple droplets on the porous superhydrophobic bionic lotus surface are studied by a high‐speed camera system. On the bionic lotus surface (Surf‐1), an impacting droplet can merge another larger sessile droplet and bounce off. Comparing with single droplet impact, it is suggested that multiple droplets impact make more efficient use of the air cushion on the micrometer‐nanometer‐scale binary structure (MNBS) and show the self‐cleaning performance of Surf‐1 better. Moreover, with the theory for damped harmonic oscillators, it is found that Surf‐1 can prolong the oscillation of droplets (last >250 ms) and slow down the attenuation of energy. The energy can be efficiently recycled to the bounce by shortening the droplet impact interval (ti). The fundamental understanding of multiple droplets impact and the reclamation of oscillating energy are beneficial for scientific design of new surface and engineering applications.

ObjectiveIschemic stroke (IS) is a common disease that causes severe and long-term neurological disability in people worldwide. Although rehabilitation is indispensable to promote neurological recovery in ischemic stroke, it is limited to providing a timely and efficient reference for developing and adjusting treatment strategies because neurological assessment after stroke treatment is mostly performed using scales and imaging. Therefore, there is an urgent need to find biomarkers that can help us evaluate and optimize the treatment plan.MethodsWe used data-independent acquisition (DIA) technology to screen differentially expressed proteins (DEPs) before and after ischemic stroke rehabilitation treatment, and then performed Gene Ontology (GO) and pathway enrichment analysis of DEPs using bioinformatics tools such as KEGG pathway and Reactome. In addition, the protein–protein interaction (PPI) network and modularity analysis of DEPs were integrated to identify the hub proteins (genes) and hub signaling pathways for neurological recovery in ischemic stroke. PRM-targeted proteomics was also used to validate some of the screened proteins of interest.ResultsAnalyzing the serum protein expression profiles before and after rehabilitation, we identified 22 DEPs that were upregulated and downregulated each. Through GO and pathway enrichment analysis and subsequent PPI network analysis constructed using STRING data and subsequent Cytoscape MCODE analysis, we identified that complement-related pathways, lipoprotein-related functions and effects, thrombosis and hemostasis, coronavirus disease (COVID-19), and inflammatory and immune pathways are the major pathways involved in the improvement of neurological function after stroke rehabilitation.ConclusionComplement-related pathways, lipoprotein-related functions and effects, thrombosis and hemostasis, coronavirus disease (COVID-19), and inflammation and immunity pathways are not only key pathways in the pathogenesis of ischemic stroke but also the main pathways of action of rehabilitation therapy. In addition, IGHA1, LRG1, IGHV3-64D, and CP are upregulated in patients with ischemic stroke and downregulated after rehabilitation, which may be used as biomarkers to monitor neurological impairment and recovery after stroke.

ObjectiveThe research objective was to evaluate the predicting role of the vascular endothelial growth factor to CBP/P300-interacting transactivator with Glu/Asp-rich C-terminal domain 2 Ratio (VEGF/CITED2) from peripheral blood mononuclear cells (PBMCs) in the collateral circulation of acute ischemic stroke (AIS).MethodsIn an observational study of patients with AIS, the western blot was applied to test the protein expression of VEGF and CITED2. Then, we calculated the VEGF/CITED2 and collected other clinical data. Binary logistic regression analysis between collateral circulation and clinical data was performed. Finally, receiver operating characteristic (ROC) curve analysis was used to explore the predictive value of VEGF/CITED2.ResultsA total of 67 patients with AIS were included in the study. Binary logistic regression analysis indicated the VEGF/CITED2 (OR 165.79, 95%CI 7.25–3,791.54, P = 0.001) was an independent protective factor. The ROC analyses showed an area under the ROC curve of the VEGF/CITED2 was 0.861 (95%CI 0.761–0.961). The optimal cutoff value of 1.013 for VEGF/CITED2 had a sensitivity of 89.1% and a specificity of 85.7%.ConclusionIn patients with AIS, the VEGF/CITED2 was related to the establishment of collateral circulation. The VEGF/CITED2 is a potentially valuable biomarker for predicting collateral circulation.Clinical trial registrationClinicalTrials.gov, identifier: NCT05345366.

Abstract Background Alternative polyadenylation (APA) is an important mechanism of gene expression regulation through generation of RNA isoforms with distinct 3′ termini. Increasing evidence has revealed that APA is actively involved in development and disease, including hepatocellular carcinoma (HCC). However, how APA functions in tumor formation and progression remains elusive. In this study, we investigated the role of cleavage factor I (CFIm) subunit CPSF6 in human hepatocellular carcinoma (HCC). Methods Expression levels of CPSF6 in clinical tissues and cell lines were determined by qRT-PCR and western blot. Functional assays, including the cell number, MTT, colony formation and transwell, were used to determine the oncogenic role of CPSF6 in HCC. Animal experiments were used to determine the role of CPSF6 in HCC tumorigenicity in vivo. Deep sequencing-based 3 T-seq was used to profile the transcriptome-wide APA sites in both HCC cells and CPSF6 knockdown HCC cells. The function of CPSF6-affected target NQO1 with distinct 3′UTRs was characterized by metabolism assays. Results We observed CPSF6 was upregulated in HCC and the high expression of CPSF6 was associated with poor prognosis in patients. Overexpression of CPSF6 promoted proliferation, migration and invasion of HCC cells in vitro and in vivo. Transcriptome-wide APA profiling analysis indicated that high expression of CPSF6 promoted the favorable usage of the proximal poly(A) site in the 3′UTR of NQO1. We demonstrated CPSF6-induced tumorigenic activities were mediated by the NQO1 isoform with short 3′UTR. Furthermore, we found that CPSF6 induced metabolic alterations in liver cells through NQO1. Conclusion CPSF6 plays a critical role in HCC progression by upregulating NQO1 expression through APA. These findings provide evidence to demonstrate that APA of NQO1 contributes to HCC progression and may have implications for developing new therapeutic strategy against this disease.

Qualitative and quantitative assessment of total sulfur and sulfur species in solid phase is necessarily important for development of effective desulfurization methods, reduction of environmental pollution, and prevention of catalyst poisoned. In this work, TPO-IR (Temperature Programmed Oxidation- Infrared) method was used for rapid determination of sulfur species in solid minerals. The high purity of Ag2S was used as the calibration materials at the fixed temperature of 1100 °C, and the TPO experimental runs were performed on the solid minerals of phosphorite and coal. The results show that pyrite sulfur with Tmax of 426 °C, and two thiophene sulfurs with Tmax of 450 °C and 481 °C were found in Kaiyang phosphorite by fitting the TPO-IR curves; while pyrite sulfur with Tmax of 432 °C, complex sulfide with Tmax of 679 °C and sulfate sulfur with Tmax of 939 °C were found in Weng’an phosphorite. For Gas coal, the TPO-IR curves of sulfur release can be deconvoluted into seven individual curves, while that of Anthracite deconvoluted into four individual curves. In conclusion, the TPO-IR method has many advantages for determination of sulfur species in solid phase systems, such as less sample consumption, simple pretreatment, convenient operation and low cost, etc.

Abstract Background Genes are regulated by various types of regulators and most of them are still unknown or unobserved. Current gene regulatory networks (GRNs) reverse engineering methods often neglect the unknown regulators and infer regulatory relationships in a local and sub-optimal manner. Results This paper proposes a global GRNs inference framework based on dictionary learning, named dlGRN. The method intends to learn atomic regulators (ARs) from gene expression data using a modified dictionary learning (DL) algorithm, which reflects the whole gene regulatory system, and predicts the regulation between a known regulator and a target gene in a global regression way. The modified DL algorithm fits the scale-free property of biological network, rendering dlGRN intrinsically discern direct and indirect regulations. Conclusions Extensive experimental results on simulation and real-world data demonstrate the effectiveness and efficiency of dlGRN in reverse engineering GRNs. A novel predicted transcription regulation between a TF TFAP2C and an oncogene EGFR was experimentally verified in lung cancer cells. Furthermore, the real application reveals the prevalence of DNA methylation regulation in gene regulatory system. dlGRN can be a standalone tool for GRN inference for its globalization and robustness.

Lung cancer is one of the leading causes of cancer-associated death in the world. It is of great importance to explore new therapeutic targets. Traditional Chinese medicine formula Feiyanning has been clinically administered in China for more than a decade and raised attention due to its anticancer effect in lung cancer. However, the underlying molecular mechanisms remain to be elucidated. In the present study, we carried out cellular and molecular assays to examine the antitumor activities and understand the mechanism of the Feiyanning formula in lung cancer cells. The cellular viability of Feiyanning-treated lung cancer cells was evaluated by Cell Counting Kit-8. The effect of the Feiyanning formula on cellular migration and invasion of lung cancer cells was examined by wound healing and transwell assays. Transcriptome and chromatin accessibility analysis by RNA-seq and ATAC-seq was performed to investigate the underlying molecular mechanisms. Our results revealed that the Feiyanning formula inhibited the cellular activities of proliferation, migration, and invasion in non-small cell lung cancer cell lines A549, H1975, and 95D. Furthermore, we observed that the transcriptional activity of the migration-associated genes was downregulated upon Feiyanning formula treatment in non-small cell lung cancer cells. The chromatin accessibility of the Feiyanning-treated lung cancer genome tended to decrease, and the regulation of the cellular component movement biological process and PI3K-AKT pathway were enriched among these altered genomic regions. Taken together, the present study suggested that Feiyanning formula exerted the antitumor effects by modulating the expression and chromatin accessibility levels of migration-associated genes.

In this present study, 195 cow milk, 100 goat milk, 50 buffalo milk, 50 camel milk, and 50 yak milk samples were collected in China in May and October 2016. The presence of aflatoxin M1 (AFM1) was determined using enzyme-linked immunosorbent assay method. For all cow milk samples, 128 samples (65.7%) contained AFM1 in concentrations ranging from 0.005 to 0.191 µg/L, and 6 samples (3.1%) from Sichuan province in October were contaminated with AFM1 above 0.05 µg/L (EU limit). For all goat milk samples, 76.0% of samples contained AFM1 in concentrations ranging from 0.005 to 0.135 µg/L, and 9 samples (9.0%) from Shanxi province in October were contaminated with AFM1 above 0.05 µg/L. For all buffalo milk samples, 24 samples (48.0%) contained AFM1 in concentrations ranging from 0.005 to 0.089 µg/L, and 2 samples collected in October were contaminated with AFM1 above 0.05 µg/L. Furthermore, 28.0% of samples contained AFM1 in concentrations ranging from 0.005 to 0.007 µg/L in camel milk samples, and 18.0% of samples contained AFM1 in concentrations ranging from 0.005 to 0.007 µg/L in yak milk samples. Our survey study has expanded the current knowledge of the occurrence of AFM1 in milk from five dairy species in China, in particular the minor dairy species.

Alternative polyadenylation (APA) is an important RNA post-transcriptional process, which can generate diverse mRNA isoforms. Increasing evidence shows that APA is involved in cell self-renewal, development, immunity, and cancer. CPSF6 is one of the core proteins of CFIm complex and can modulate the APA process. Although it has been reported to play oncogenic roles in cancer, the underlying mechanisms remain unclear. The aim of the present study was to characterize CPSF6 in human gastric cancer (GC). We observed that CPSF6 was upregulated in GC. Knockdown of CPSF6 inhibited proliferation and enhanced apoptosis of GC cells both in vitro and in vivo. Global APA site profiling analysis revealed that knockdown of CPSF6 induced widespread 3′UTR shortening of genes in GC cells, including VHL. We also found CPSF6 negatively regulated the expression of VHL through APA and VHL short-3′UTR isoform enhanced apoptosis and inhibited cell growth in GC cells. Our data suggested that CPSF6-induced cell proliferation and inhibition of apoptosis were mediated by the preferential usage of poly(A) in VHL. Our data provide insights into the function of CPSF6 and may imply potential therapeutic targets against GC.

Coronin‐like actin‐binding protein 1C (CORO1C) is a member of the WD repeat protein family that regulates actin‐dependent processes by assembling F‐actin. CORO1C was previously reported to promote metastasis in breast cancer and lung squamous cell carcinoma. Here, we investigated the role of CORO1C in gastric cancer. Higher expression levels of CORO1C were detected in gastric cancer tissues as compared with normal gastric tissues. In addition, CORO1C levels were found to be positively correlated with lymph node metastasis in gastric cancer patients. The expression levels of CORO1C were higher in stage III‐IV gastric cancer patients (80.8%) than in stage I‐II gastric cancer patients(57.1%). Gastric cancer patients positive for CORO1C expression showed lower relapse‐free survival and overall survival rates. Knockdown of CORO1C dramatically suppressed total cell number, cell viability, cell colony formation, cell mitosis and cell metastasis, and promoted apoptosis of gastric cancer cells. Furthermore, cyclin D1 and vimentin were found to be positively regulated by CORO1C. As cyclin D1 and vimentin play an oncogenic role in gastric cancer, CORO1C may exert its tumor‐promoting activity through these proteins.

Abstract Background Psychological investigations and functional imaging technology have been used to describe neural correlations of different types of memory with various stimuli. Memory with limited storage capacity and a short retention time can be classified as short-term memory (STM) while long-term memory (LTM) can be life-long without defined capacity. Methods To identify brain activation pattern associated with different modes of memory for numerical figures, we detected brain activities from twenty-two healthy subjects when performing three types of memory tasks for numbers, namely STM, LTM and working memory (WM), by using functional magnetic resonance imaging (fMRI) technique. Results The result revealed variable patterns of activation in different brain regions responding to different types of memory tasks. The activation regions with primary processing and transient maintenance of STM for numerical figures are located in the visual cortex and mainly encoded by visual representations, while LTM was encoded by semantics and mainly recruiting left frontal cortex. We also found that subcortical structures, such as the caudate nucleus and the marginal division of the striatum, plays important roles in working memory. Conclusions Activation of different brain regions in these three kinds of memories, indicating that different kinds of memories rely on different neural correlates and mental processes.

Background The relationships between coagulation factors and non‐small cell lung cancer (NSCLC) prognosis have been intensively studied. However, no previous study has investigated the combined effects of preoperative platelet (PLT), fibrinogen (FIB), and D‐dimer (D‐D) levels on the prognosis of NSCLC. Methods A multicenter prospective study was conducted over seven hospitals. A total of 395 patients diagnosed with operable NSCLC for the first time were included and followed‐up until disease progression or the end of the study. Baseline demographic and clinicopathological information, and preoperative coagulation test results were collected for each patient. Univariate and multilevel survival analyses were conducted using Cox regression and shared frailty models. Results Multilevel analyses revealed that there was a marginally significant association between elevated PLT level (> 215 × 109/L) and unfavorable progression‐free survival (PFS) (hazard ratio 2.42, P = 0.05), whereas preoperative FIB and D‐D were not significant prognostic factors for PFS (P = 0.31 and 0.30, respectively). Compared to patients with one elevation of the three coagulation factors, patients with at least two elevations of the three factors had a significantly higher risk of cancer progression (hazard ratio 4.62, P = 0.02). Conclusion The number of elevated preoperative coagulation factors may have a significant effect on PFS and could be used to predict the prognosis of NSCLC patients after surgery. Future studies are warranted to further investigate the interactions between these three coagulation factors.

Abstract Transplantation of endothelial progenitor cells (EPCs) is a proven safe and effective method for treatment of cerebral ischemia in animal experiments. However, safety and efficacy need to be determined in clinical trials. We performed a two‐center, randomized, placebo‐controlled phase I/IIa trial with blinded outcome assessment on 18 patients with acute cerebral infarct within the middle cerebral artery territory, and followed for up to 4 years. Autologous ex vivo expanded EPCs were injected intravenously in the EPC group, and patients who received saline or autologous bone marrow stromal cells served as control groups. Mortality of any cause, adverse events, and new‐onset comorbidities were monitored. Changes in neurological deficits were assessed at different time points. We found no toxicity events or infusional or allergic reactions in any treated group. Three patients in the placebo group died during the 4‐year follow‐up. We found that the EPC group had fewer serious adverse events compared with the placebo‐controlled group, although there were no statistical differences in mortality among the three groups. Furthermore, there was no significant difference in neurological or functional improvement observed among the three groups, except for the Scandinavia Stroke Scale score at 3 months between the EPC group and placebo‐controlled group. Autologous transplantation of EPCs appears to improve long‐term safety in acute cerebral infarct patients, supporting the feasibility of this novel method for treatment of ischemic stroke (ClinicalTrials.gov: NCT01468064). Background: Spasticity is a common sequela of stroke. The incidence of poststroke spasticity (PSS) has not been systematically reviewed in recent years, and some risk factors remain debated. This systematic review and meta-analysis was conducted to determine the prevalence and risk factors for PSS.Methods: We searched electronic databases (PubMed, Embase, Cochrane Library, CNKI, WANFANG and CBM) inception to May 12, 2020. Observational studies summarizing the incidence or risk factors for PSS were included. Only cohort studies were enrolled in meta-analysis. For risk factors examined in at least three different studies, we combined effects into odds ratios (OR) and 95% confidence intervals (CI).Results: One thousand four hundred sixty-seven studies were retrieved and 23 were involved in meta-analysis. The pooled prevalence of spasticity after stroke was 25.3% and that after the first-ever stroke was 26.7%. The incidence of spasticity after the first-ever stroke with paresis was 39.5%. The prevalence of disabling or severe spasticity (MAS ≥ 3) in stroke patients with paresis was 9.4% (95% CI 0.056–0.133), and severe spasticity was 10.3% (95% CI 0.058–0.149). Moderate to severe paresis (OR = 6.573, 95% CI 2.579–16.755, I2 = 0.0%), hemorrhagic stroke (OR = 1.879, 95% CI 1.418–2.490, I2 = 27.3%) and sensory disorder were risk factors for PSS.Conclusions: The incidence of PSS was significantly higher in stroke patients with paresis. Patients with moderate to severe paresis and sensory disorder should be closely followed up. The role of hemorrhagic stroke in predicting PSS remains to be further explored.

Despite the achievement of shock wave focusing with certain reflectors, the influence of the radial entrance width of a circular cavity on the flow field has yet to be addressed. In this study, we systematically investigated the effects of the shock wave focusing process in a cavity based on the radial entrance widths. An experimental system was installed to research the evolution of the flow field under conditions with different radial entrance widths of 3.0, 11.1, 19.5, and 33.0 mm. A schlieren system was used to photograph the structures of the flow field in the cavity, and a data acquisition system was used to record the dynamic pressure histories of different points. A numerical simulation was carried out to investigate greater details of the shock wave focusing process. A third-order strong stability-preserving Runge-Kutta method, third-order weighed essential non-oscillation scheme, and an adaptive mesh refinement algorithm were adopted to simulate the shock wave reflection, diffraction, and focus process. Good agreement between the experimental and numerical results was observed. By comparing the evolution process of the flow field under the conditions of four different entrance cavity widths, we found that when the entrance width was 19.5 mm, there was the stronger intensity of the shock wave focusing in the focal region, and the larger pressure value at the apex of the cavity than the other three entrance widths, occur. Survival rates for patients with gastric cancer, especially the advanced form, remain poor and the development of targeted treatments is hampered by a lack of efficient biological targets. Poly(rC) binding protein 2 (PCBP2) is an RNA‐binding protein that contributes to mRNA stabilization, translational silencing and enhancement and it has been implicated as a promoter of gastric cancer growth. In the present study, we demonstrated that the expression level of PCBP2 was higher in human gastric cancer tissues compared to adjacent normal gastric tissues. A high level of PCBP2 was correlated with worse postoperative relapse‐free survival and overall survival rates of gastric cancer patients. Small hairpin RNA‐mediated depletion of PCBP2 dramatically decreased the viability of gastric cancer cells. Cyclin‐dependent kinase 2 (CDK2) was positively regulated by PCBP2 via a direct 3′ UTR binding pathway as determined using a ribonucleoprotein immunoprecipitation assay and a biotin pulldown assay. CDK2 mediated the promoting role of PCBP2. These results suggest that PCBP2 acts as an oncogene in human gastric cancer cells and that functionally depleting PCBP2 could be considered as a potential target for gastric cancer therapy.

Abstract Non-thermal atmospheric plasma (NTP) has shown its selective anticancer effects in many types of tumors in vitro and one of the main mechanisms is that the different increase of intracellular ROS in cancer and homologous normal cells. In this study, we report that NTP treatment reduces the proliferation in triple negative breast cancer (TNBC) and normal cell lines. Simultaneously, STAT3 pathway is inhibited by NTP effects. However, it is observed that normal cells MCF10A are more sensitive to ROS toxicity induced by NTP than cancer cells MDA-MB-231. When 5 mM of ROS inhibitor N-acetyl cysteine (NAC) is employed in NTP treatments, the proliferation of normal breast cells MCF10A recovers. Meanwhile, NTP effects remain significant inhibition of MDA-MB-231 cells. Our results further reveal that NTP can induce apoptosis in MDA-MB-231 cells through inhibiting interleukin-6 receptor (IL-6R) pathway. Moreover, the mechanism of NTP anti-cancer selectivity relates to constantly HER2/Akt activation induced by NTP especially in MCF10A cells but not in MDA-MB-231 cells. Therefore, these two different cell signaling pathways induced by NTP treatments in TNBC and homologous normal cells make NTP becoming a potential tool in future therapy.

Inducing somatic cells into neural stem cells (iNSCs) in specific ways provides a new cell therapy in a variety of neurological diseases. In the past, iNSCs were generated by transcription factors which increased the risk of mutagenesis, tumor formations, and immune reactions by viral transduction vectors. Therefore, in this study, different small molecules were used to induce mouse embryonic fibroblasts (MEFs) into iNSCs in different reprogramming stages, which showed high reprogramming efficiency without altering the genome. We demonstrated that the small molecules staged-induction neural stem cells (SMSINS) have the characteristics of neural stem cells (NSCs) in morphology, gene expression, self-renewal and differentiation potential. Furthermore, valproic acid (VPA), one of small molecules, was showed to enhance neural induction with highest efficiency compared with six other small molecules, which were also investigated in the present study. Moreover, our results suggested that activating the mammalian target of rapamycin (mTOR) signaling enhanced the induction efficiency and neuronal differentiation. Collectively, our findings indicated that using this induction program allowed us to obtain safe and efficient iNSCs which were free of genetic manipulation. The VPA-mediated mTOR signaling pathway may enhance reprogramming efficiency and neuronal differentiation. So we suggested that this program could be a new method of obtaining iNSCs for the treatment of neurological diseases by cell replacement therapy in the future.

To study the plasma plume expansion dynamics of nanosecond laser ablating Al/PTFE, the Al/PTFE propellant was prepared by a molding sintering method and the rapid expansion process of the plasma plume was photographed using fast photography technology. The effects of the proportion of Al, laser energy and ambient pressure on plasma plume expansion dynamics are analyzed. The results show that the plume expansion process of laser ablating Al/PTFE plasma can be divided into three stages and this phenomenon has not been reported in the literature. The Al powder doped in PTFE will block part of the laser transmission into the propellant, thus reducing the laser absorption depth of the propellant. In the case of short pulse laser ablation, the reaction rate between Al and PTFE is optimal when the reductant is slightly higher than the oxidant. As the laser energy increases, the light intensity of the plasma becomes stronger, the plasma size becomes larger and the existence time of plasma becomes longer. In the first stage plume, the plume expands freely at the ambient pressure of 0.005 Pa and the plume expansion distance is linearly related to time, while the shock wave formed at the interface between the plume front and the ambient gas at the ambient pressure of 5 Pa and the expansion can be described by S-T theory.

Traumatic brain injury (TBI) is a common disease that usually causes severe neurological damage, and current treatment is far from satisfactory. The neuroprotective effects of neural stem cell (NSC) transplantation in the injured nervous system have largely been known, but the underlying mechanisms remain unclear, and their limited sources impede their clinical application. Here, we established a rat model of TBI by dropping a weight onto the cortical motor area of the brain and explored the effect of engrafted NSCs (passage 3, derived from the hippocampus of embryonic 12- to 14-d green fluorescent protein transgenic mice) on TBI rats. Moreover, RT-PCR and Western blotting were employed to investigate the possible mechanism associated with NSC grafts. We found rats with TBI exhibited a severe motor and equilibrium dysfunction, while NSC transplantation could partly improve the motor function and significantly reduce cell apoptosis and increase B-cell lymphoma–extra large (Bcl-xL) expression at 7 d postoperation. However, other genes including Bax, B-cell lymphoma 2, Fas ligand, and caspase3 did not exhibit significant differences in expression. Moreover, to test whether Bcl-xL could be used as a therapeutic target, herpes simplex virus (HSV) 1 carrying Bcl-xL recombinant was constructed and injected into the pericontusional cortices. Bcl-xL overexpression not only resulted in a significant improvement in neurological function but also inhibits cell apoptosis, as compared with the TBI rats, and exhibits the same effects as the administration of NSC. The present study therefore indicated that NSC transplantation could promote the recovery of TBI rats in a manner similar to that of Bcl-xL overexpression. Therefore, Bcl-xL overexpression, to some extent, could be considered as a useful strategy to replace NSC grafting in the treatment of TBI in future clinical practices.

To predict the thrust of magnetoplasmadynamic thrusters (MPDTs), a modified electromechanical model was proposed and a comparison with experimental results is presented in this paper. The motion of propellant in the thruster was divided into two portions: the axial motion which was accelerated by the interaction of current and induced self-field, and the swirling motion which was accelerated by the interaction of current and applied magnetic field. The electromechanical model was in good agreement with the experimental data, and the fitting degrees of the model were greater than 0.93. Furthermore, the influence of parameters on the performance of MPDT were investigated by utilizing the electromechanical model. The results indicate that the thrust performance of the thruster improved with the increase of discharge current, anode radius, applied magnetic field strength, and the decrease of mass flow rate. However, the large anode radius and low mass flow rate readily led to the failure of thruster function. Therefore, the model can not only predict the thrust performance of MPDTs, but also guide the design and operation optimization of the thruster.

In this paper, a non-Fourier heat conduction model of ultra-short pulsed laser ablation of metal is established that takes into account the effect of the heat source, laser heating of the target, the evaporation and phase explosion of target material, the formation and expansion of the plasma plume, and interaction of the plasma plume with the incoming laser. Temperature dependent optical and thermophysical properties are also considered in the model due to the properties of the target will change over a wide range during the ultra-short pulsed laser ablation process. The results show that the plasma shielding has a great influence on the process of ultra-short pulsed laser ablation, especially at higher laser fluence. The non-Fourier effect has a great influence on the temperature characteristics and ablation depth of the target. The ultra-short pulsed laser ablation can effectively reduce the heat affected zone compared to nanosecond pulsed laser ablation. The comparison between the simulation results and the experimental results in the literature shows that the model with the plasma shielding and the non-Fourier effect can simulate the ultra-short pulsed laser ablation process better.

Experiments and numerical simulations were carried out to investigate radial incident shock focusing on a test section where the planar incident shock wave was divided into two identical ones. A conventional shock tube was used to generate the planar shock. Incident shock Mach number of 1.51, 1.84 and 2.18 were tested. CCD camera was used to obtain the schlieren photos of the flow field. Third-order, three step strong-stability-preserving (SSP) Runge-Kutta method, third-order weighed essential non-oscillation (WENO) scheme and adaptive mesh refinement (AMR) algorithm were adopted to simulate the complicated flow fields characterized by shock wave interaction. Good agreement between experimental and numerical results was observed. Complex shock wave configurations and interactions (such as shock reflection, shock-vortex interaction and shock focusing) were observed in both the experiments and numerical results. Some new features were observed and discussed. The differences of structure of flow field and the variation trends of pressure were compared and analyzed under the condition of different Mach numbers while shock wave focusing.

In this paper, a new numerical modeling method that combines a modified electromechanical model with a Teflon ablation model is proposed to simulate the working process of micro-pulsed plasma thruster (μPPT). The ablation mass accumulation during the discharge process and the non-Fourier effect during the ablation process are considered to improve simulation accuracy. Simulation results are in good agreement with experimental results. Furthermore, the influence of both the electrical and structural parts on the performance of μPPT was investigated by applying the combined model. It was proven that large capacitance of the capacitor and large gap-to-plate-width ratios can effectively improve the comprehensive performance of the thruster concluding the specific impulse, impulse bit, and efficiency. The results not only validate the theoretical model, but also guide the design and operation optimization of μPPT.

OBJECTIVE: To analyze the association between -1082A/G polymorphism in interleukin-10 (IL-10) gene and ischemic stroke (IS) risk by meta-analysis. METHODS: We carried out a systematic electronic search in PubMed, BIOSIS Previews, Science Direct, Chinese National Knowledge Infrastructure, Chinese Biomedical Database, Weipu database and WANGFANG Database. Pooled odds ratios (ORs) with 95% confidence intervals (95%CIs) were calculated to assess the strength of the association. RESULTS: 7 studies were included. There was no significant association between IL-10 -1082A/G polymorphism and IS risk under all genetic models in overall estimates (A vs. G: OR = 1.23,95%CI = 0.85-1.79;AA vs. GG: OR = 1.01,95%CI = 0.47-2.19; AG vs. GG: OR = 0.76, 95%CI = 0.38-1.55; AA+AG vs. GG: OR = 0.89,95%CI = 0.46-1.73; AA vs. AG+GG: OR = 1.39, 95%CI = 0.91-2.13). Similarly, no associations were found in subgroup analysis based on ethnicity and source of controls. However, removing the study deviating from Hardy-Weinberg equilibrium (HWE) produced statistically significant associations for overall estimates under recessive model(AA VS. AG+GG OR 1.58, 95% CI 1.04-2.42) and among Asians in all genetic models (A VS.G OR 1.64, 95% CI 1.07-2.53; AA vs. GG OR1.91, 95% CI 1.31-2.80; AG vs. GG OR1.44, 95% CI 1.09-1.91; AA+AG vs. GG OR 1.54, 95% CI 1.18-2.01;AA VS. AG+GG OR 1.79, 95% CI 1.07-3.00). Even after Bonferroni correction, the associations were observed still significantly in Asians under the two models (AA vs. GG OR1.91, 95% CI 1.31-2.80, P = 0.0008; AA+AG vs. GG OR 1.54, 95% CI 1.18-2.01, P = 0.001). CONCLUSION: This meta-analysis indicates that IL10 -1082 A/G polymorphism is associated with IS susceptibility in Asians and the -1082 A allele may increase risk of IS in Asians. Considering the sample size is small and between-study heterogeneity is remarkable, more studies with subtle design are warranted in future.

OBJECTIVE: To explore the association between transforming growth factor-beta1 (TGF-β1) T869C polymorphism and risk of ischemic stroke (IS) by performing a meta-analysis based on published articles. METHODS: Systematic electronic searches of PubMed, Science Direct, BIOSIS Previews, Chinese Biomedical Database, Chinese National Knowledge Infrastructure, and WANFANG Database were performed. The strength of the association was calculated by pooled odds ratios (ORs) with 95% confidence intervals (95%CIs). Subgroup analysis was conducted to explore potential sources of heterogeneity. Sensitivity analysis was performed to elucidate the stability of the outcomes. Publication bias was evaluated by Begg's funnel plot and Egger's test. RESULTS: A total of 6 studies involving 1701 cases were included. The overall estimates did not show any significant association between TGF-β1 T869C polymorphism and risk of IS under all genetic models (C vs. T: OR = 1.08,95%CI = 0.88-1.32; CC vs. TT:OR = 1.17,95%CI = 0.79-1.72; CT vs. TT: OR = 0.91, 95%CI = 0.68-1.22; CC+CT vs. TT: OR = 0.99, 95%CI = 0.73-1.35; CC vs. CT+TT: OR = 1.23, 95%CI = 0.95-1.59). Similar lacking associations were observed in subgroup analysis based on ethnicity and source of controls. When stratified by study design, significant increased association of IS risk was found in cohort studies under genetic models except recessive model(C vs. T: OR = 1.18, 95%CI = 1.05-1.32; CC vs. TT: OR = 1.40, 95%CI = 1.10-1.77; CT vs. TT: OR = 1.23, 95%CI = 1.02-1.49; CC+CT vs. TT: OR = 1.27, 95%CI = 1.03-1.57; CC vs. CT+TT, OR = 1.21, 95%CI = 0.99-1.47), whereas in case-control studies a significant decreased risk was detected under heterozygote comparison(CT vs. CC: OR = 0.72, 95%CI = 0.57-0.92). However, after correction for multiple testing, the associations were observed to be null significant in both cohort and case-control subgroups among all genetic models. CONCLUSION: This meta-analysis suggested that current epidemiological studies of TGF-β1 T869C polymorphism are too inconsistent to draw a conclusion on the association with IS susceptibility. Given the small sample size and remarkable between-study heterogeneity, further well-designed prospective large-scale studies are warranted.
