Abstract

Finite element method (FEM) modeling of the volumetric expansion phenomenon associated with the accumulation of irradiation was performed on rocks in a concrete for nuclear power plant. The FEM mesh of sandstone, tuff, and granite was created based on microscopic images, and the volumetric expansion due to irradiation was calculated. The saturated value of the volume expansion due to irradiation accumulation was calculated, and it was shown that the saturated value of the volume expansion was well explained by the experimental value. In addition, the FEM analysis indicates that irradiation-induced degradation of rock propagates through a localized concentration of stress followed by fracture with cracks spreading throughout the rock., Comment: 5 pages, 4 figures

Abstract

Rubidium 87 atoms are trapped in an Ioffe-Pritchard potential generated with a persistent supercurrent that flows in a loop circuit patterned on a sapphire surface. The superconducting circuit is a closed loop made of 100 micro meter wide MBE-grown MgB2 stripe carrying a supercurrent of 2.5 A. To control the supercurrent in the stripe, an on-chip thermal switch operated by a focused argon-ion laser is developed. The switch operates as an on/off switch of the supercurrent, or as a device to set the current to a specific value with the aid of an external magnetic field. The current can be set even without an external source if the change is in decreasing direction., Comment: 4 pages, 5 figures

Abstract

Item does not contain fulltext, NOTCH1 belongs to the NOTCH family of proteins that regulate cell fate and inflammatory responses. Somatic and germline NOTCH1 variants have been implicated in cancer, Adams-Oliver syndrome, and cardiovascular defects. We describe 7 unrelated patients grouped by the presence of leukoencephalopathy with calcifications and heterozygous de novo gain-of-function variants in NOTCH1. Immunologic profiling showed upregulated CSF IP-10, a cytokine secreted downstream of NOTCH1 signaling. Autopsy revealed extensive leukoencephalopathy and microangiopathy with vascular calcifications. This evidence implicates that heterozygous gain-of-function variants in NOTCH1 lead to a chronic central nervous system (CNS) inflammatory response resulting in a calcifying microangiopathy with leukoencephalopathy. Abstract

BACKGROUND: Pediatric diffuse intrinsic pontine glioma (DIPG) represents one of the most devastating and lethal brain tumors in children with a median survival of 12 months. The high mortality rate can be explained by the ineligibility of patients to surgical resection due to the diffuse growth pattern and midline localization of the tumor. While the therapeutic strategies are unfortunately palliative, the blood-brain barrier (BBB) is suspected to be responsible for the treatment inefficiency. Located at the brain capillary endothelial cells (ECs), the BBB has specific properties to tightly control and restrict the access of molecules to the brain parenchyma including chemotherapeutic compounds. However, these BBB specific properties can be modified in a pathological environment, thus modulating brain exposure to therapeutic drugs. Hence, this study aimed at developing a syngeneic human blood-brain tumor barrier model to understand how the presence of DIPG impacts the structure and function of brain capillary ECs. METHODS: A human syngeneic in vitro BBB model consisting of a triple culture of human (ECs) (differentiated from CD34(+)-stem cells), pericytes and astrocytes was developed. Once validated in terms of BBB phenotype, this model was adapted to develop a blood-brain tumor barrier (BBTB) model specific to pediatric DIPG by replacing the astrocytes by DIPG-007, -013 and -014 cells. The physical and metabolic properties of the BBTB ECs were analyzed and compared to the BBB ECs. The permeability of both models to chemotherapeutic compounds was evaluated. RESULTS: In line with clinical observation, the integrity of the BBTB ECs remained intact until 7 days of incubation. Both transcriptional expression and activity of efflux transporters were not strongly modified by the presence of DIPG. The permeability of ECs to the chemotherapeutic drugs temozolomide and panobinostat was not affected by the DIPG environment. CONCLUSIONS: This original human BBTB model allows a better understanding of the influence of DIPG on the BBTB ECs phenotype. Our data reveal that the chemoresistance described for DIPG does not come from the development of a "super BBB". These results, validated by the absence of modification of drug transport through the BBTB ECs, point out the importance of understanding the implication of the different protagonists in the pathology to have a chance to significantly improve treatment efficiency.

Abstract

A Fourier hologram is used to diffract atomic beams in specific interference patterns. In the future, this process may be refined to precisely manipulate ultra-cold atoms. In the process, a beam of ultra-cold metastable neon atoms is passed through a computer generated hologram. The hologram, containing a 100-nanometer-thick silicon nitride membrane, encodes the Fourier transform of the required atomic pattern. The far-field pattern of the diffracted wave generates an image of the object when the hologram is illuminated with a plane wave. The use of the technique with other manipulation techniques will enable the reproduction of real-time moving patterns of atoms.

Abstract

Background: Sucrose and sweet beverage uptakes have been implicated to be one of causes of obesity. We wanted to know if any food uptake may be related to increase in Body Mass Index (BMI) in healthy young and old men and women. Methods: Healthy young and old men and women were given self-administered diet history questionnaires and described answers on each item by recollection of diets they took. From these questionnaires, we calculated the intake of energy, carbohydrate, fat and protein. Results: Old people took more protein, lipids, carbohydrates, and omega fatty acids than young people. Men took such foods more than women. There was no statistically significant relationship between particular food uptakes and BMI. Conclusion: Uptakes of sucrose and sweet beverage did not increase BMI in healthy people. We do not think that sucrose uptake is a major reason for obesity.
