1. Survey of in vitro fertilization add-ons in Japan (Izanami project).

Abstract

Objective: To identify any correlations between evidence levels, adoption rates, and additional costs of in vitro fertilization (IVF) add-ons. Design: Online survey. Subjects: The survey was conducted in 621 assisted reproductive technologyregistered facilities that are members of the Japanese Society of Obstetrics and Gynecology from December 22, 2021, to February 13, 2022. Exposure: The survey included details regarding the specific add-on modalities employed and their associated costs; inquiries pertained to the fertility healthcare infrastructure in Japan before the implementation of the National Health Insurance scheme. Main outcome measures: The correlation between the adoption rate and cost of IVF add-ons and their evidence levels were analyzed. The evidence level of the addon treatments was classified into Green, Amber, and Red categories based on the United Kingdom's Human Fertilisation and Embryology Authority and Cochrane systematic reviews. Results: A total of 438 eligible responses were analyzed, with clinics constituting 70.9% of the respondents' facilities. A total of 18 add-ons were assessed, and 96.5% (423/438) of facilities used at least one add-on. A positive correlation of the adoption rate and an inverse correlation of the cost with the evidence level of the IVF add-on treatment were observed (not significant). Outpatient clinics, defined as medical facilities with no beds, had a significantly higher adoption rate (Amber, 65.7%; Red, 52.0%) of add-ons than other facilities, regardless of the evidence rating, although the costs were similar across all site attributes. Conclusion: Accumulating evidence on the efficacy and safety of add-ons will lead to the development of medical care with a high-cost benefit, as an increase in the adoption rate and a decrease in cost are expected when limiting to medical care with a high level of evidence. 4. Estimating the Availability of Forest Biomass from Aggregated Forests in Japan

Abstract

This study acquired the forest GIS for private and communal forests from prefectures and national forests from the Forestry Agency of Japan. Then, this study calculated incomes and expenditures such as silvicultural and harvesting as well as stumpage prices on the Japanese cedar, cypress, pine, and larch forests using the silvicultural prescriptions set based on the regional forest plans and operation systems set based on topographic conditions such as slope angles and height differences with GIS. Finally, this study estimated the availability of unused materials for woody biomass power generation plants under operation with FIT at the end of June 2020 as the supply potential from the profitable subcompartments. As a result, supply potentials of timber and forest biomass were estimated at 65,490,336 m3/year and 13,098,067 m3/year, respectively whereas those availabilities were estimated at 31,080,672 m3/year and 6,216,134 m3/year respectively. Therefore, the rate of the availabilities to the supply potentials was 47.5%. Furthermore, the rate of the availabilities to the current demands was 71.6%. Considering aggregation in the same watershed, the rate of the availabilities to the demands increased to 124.8%. Abstract

Nomara Spot weld electrode life was investigated for various types of 0.8mm thick zinc plated steel sheets which are now widley used in automotive industry. The results were discussed from the point of view of chemical compositions of the plated layer in order to find the desirable zinc plated steel sheet which ensures long electrode life. Electrode life was defined as the number of welds until nugget diameter decreased to 4.5mm in continuous spot welding with 6mm∅ dome type Cu-1%Cr electrode at each fixed welding condition. As a result, the plated sheet steel with nickel in plated layer showed a relativly long life. This efficent effect of nickel was confirmed by an additional spot welding test and investigation of zinc diffusion in the electrode.

Abstract

Background: Hepatitis C virus (HCV) nonstructural protein 5A (NS5A) has been shown to modulate multiple cellular processes, including apoptosis. The aim of this study was to assess the effects of HCV NS5A on apoptosis induced by Toll-like receptor (TLR) 4 ligand, lipopolysaccharide (LPS).Methods: Apoptotic responses to TLR4 ligands and the expression of molecules involved in TLR signaling pathways in human hepatocytes were examined with or without expression of HCV NS5A.Results: HCV NS5A protected HepG2 hepatocytes against LPS-induced apoptosis, an effect linked to reduced TLR4 expression. A similar downregulation of TLR4 expression was observed in Huh-7-expressing genotype 1b and 2a. In agreement with these findings, NS5A inhibited the expression of numerous genes encoding for molecules involved in TLR4 signaling, such as CD14, MD-2, myeloid differentiation primary response gene 88, interferon regulatory factor 3, and nuclear factor-κB2. Consistent with a conferred prosurvival advantage, NS5A diminished the poly(adenosine diphosphate-ribose) polymerase cleavage and the activation of caspases 3, 7, 8, and 9 and increased the expression of anti-apoptotic molecules Bcl-2 and c-FLIP.Conclusions: HCV NS5A downregulates TLR4 signaling and LPS-induced apoptotic pathways in human hepatocytes, suggesting that disruption of TLR4-mediated apoptosis may play a role in the pathogenesis of HCV infection.
