Abstract Background Congenital toxoplasmosis occurs when a pregnant woman becomes infected with Toxoplasma gondii (T. gondii) for the first time. Treatment typically involves antimicrobial medications, with spiramycin commonly used to prevent transmission. However, spiramycin's effectiveness is limited due to poor placental penetration. Clindamycin, another antibiotic, can cross the placenta but reaches the fetus at only half the maternal concentration. Encapsulating the drug in chitosan-coated niosomes (Cs-Nio) could enhance its effectiveness by targeting specific organs and ensuring sustained release. To address the challenges of using clindamycin, a niosome-coated chitosan system was investigated for treating congenital toxoplasmosis caused by the VEG strain of T. gondii in an animal model. Methods Pregnant mice were infected with VEG strain of T. gondii on the 12th day of pregnancy, followed by treatment with various drugs across six groups. The treatments included chitosan-coated niosomes loaded clindamycin (Cs-Nio-Cli) and other controls. Parasitological evaluations (microscopic examination and real-time PCR), along with histopathological and immunological assessments were conducted to assess treatment efficacy. Finally, statistical analysis was conducted using GraphPad Prism 8.0 and SPSS 26, comparing test and control groups with T test and Mann–Whitney test. A p ≤ 0.05 was considered statistically significant. Results The study found that treatment with Cs-Nio-Cli significantly reduced the number of T. gondii cysts in the brain and eyes (97.59% and 92.68%, respectively) compared to the negative control group. It also mitigated inflammatory changes, prevented cell death, and reduced vascular cuffs in the brain. In addition, Cs-Nio-Cli treatment decreased bleeding, placental thrombosis, and inflammatory cell infiltration in the placenta while improving eye tissue health by reducing retinal folds and bleeds. Immunologically, nanoclindamycin treatment resulted in lower TNF-α cytokine levels and higher IL-10 levels, indicating an enhanced anti-inflammatory response. Conclusions Although Cs-Nio-Cli demonstrates promise in reducing the transmission of congenital toxoplasmosis and mitigating the effects of congenital toxoplasmosis, additional research is necessary to determine the optimal treatment regimens for the complete eradication of the parasite in the fetus.

Background and purpose: Intestinal helminth parasites in dogs, especially zoonotic parasites, are one of the health and economic problems of developing countries, including Iran. Toxocara canis is one of the zoonotic helminth parasites that causes serious damage to human health. According to the close relationship that humans have with dogs today, it is necessary to investigate worm parasites in these animals. The present study was conducted to investigate the prevalence of intestinal helminth parasites in urban and domestic stray dogs with two modified methods of sedimentation and flotation in the central part of Mazandaran province. Materials and methods: The study was carried out in three regions of Mazandaran province (Sari, Qaemshahr, and Babolsar) in 2022. We examined fecal samples from 110 dogs, comprising 78 stray dogs and 32 home dogs, to determine the presence of intestinal helminth parasites. We utilized a customized concentrates technique that incorporated flotation and sedimentation. 4 cc of 10% formalin was added to the sample container for sampling. Stool samples weighing approximately 50 to 100 grams were collected using an applicator and transferred into labeled containers. In the flotation and sedimentation method, a sufficient amount of stool sample was poured into the test tube, and then 7 cc of physiological serum was added to it. After the sample is completely dissolved by the applicator, the solution is filtered to remove excess material and transferred to a new tube. The obtained solution is centrifuged at 1000 rpm for 3-5 minutes, then the upper liquid is separated. 7 cc of saturated zinc sulfate solution was added to it and centrifuged again at 1000 rpm for 2 minutes. The resulting sediment was examined with a light microscope with X10 and X40 magnification to identify parasite contamination. Traps were set in the shelters of stray dogs to collect feces samples directly from their rectums. Labeled containers with preservatives (10% formalin) were distributed to clinics and dog owners for collecting samples from domestic dogs. A questionnaire was designed to assess the gender and age of domestic dogs based on dental characteristics, location of residence, deworming practices, and knowledge of zoonotic intestinal parasite diseases in dogs among their owners. The experiment was conducted at Mazandaran University of Medical Sciences and was approved by the Animal Ethics Committee under the reference number IR.MAZUMS.REC.1400.528. Results: Results indicated that 44 samples (40%) were contaminated with parasites, while 66 samples (60%) were free of worm infestation. The infected samples harbored parasite eggs including Hookworms, Toxocara, Taenia, and Dipylidium. The study found no significant difference in the distribution of helminth parasite infection among the three sampling areas in terms of the frequency of infected dogs. Further research in the field of parasite epidemiology and expanding the study to include more areas may help address this issue. Conclusion: The research findings suggest that stray dogs are more significant in transmitting pollutants than domestic dogs. Urban stray dogs pose a potential public health danger because they are released post-treatment. Controlling and gathering these animals is crucial in managing parasite diseases that can be passed on to humans.

Background: Current study was designed to provide a better insight into the circulating genotypes, genetic diversity, and population structure of Echinococcus spp. between southeast of Iran and Pakistan. Methods: From Jun 2020 to Dec 2020, 46 hydatid cysts were taken from human (n: 6), camel (n: 10), goat (n: 10), cattle (n: 10) and sheep (n: 10) in various cities of Sistan and Baluchestan Province of Iran, located at the neighborhood of Pakistan. DNA samples were extracted, amplified, and subjected to sequence analysis of cox1 and nad1 genes. Results: The phylogeny inferred by the Maximum Likelihood algorithm indicated that G1 genotype (n: 19), G3 genotype (n: 14) and G6 genotype (n: 13) assigned into their specific clades. The diversity indices showed a moderate (nad1: Hd: 0.485) to high hap- lotype diversity (cox1: Hd: 0.867) of E. granulosus s.s. (G1/G3) and low nucleotide diver- sity. The negative value of Tajima’s D and Fu’s Fs test displayed deviation from neutrali- ty indicating a recent population expansion. A parsimonious network of the haplotypes of cox1 displayed star-like features in the overall population containing IR9/PAK1/G1, IR2/PAK2/G3 and IR18/G6 as the most common haplotypes. A pairwise fixation index (Fst) indicated that E. granulosus s.s. populations are genetically moderate differen- tiated between southeast of Iran and Pakistan. The extension of haplotypes PAK18/G1 (sheep) and PAK26/G1 (cattle) toward Iranian haplogroup revealed that there is dawn of Echinococcus flow due to a transfer of alleles between mentioned populations through transport of livestock or their domestication. Conclusion: The current findings strengthen our knowledge concerning the evolution- ary paradigms of E. granulosus in southeastern borders of Iran and is effective in control- ling of hydatidosis.

The larval stages of Echinococcus multilocularis (E. multilocularis) are what cause the zoonotic disease known as alveolar echinococcosis (AE). Identifying the antigens that trigger immune responses during infection is extremely important for the development of vaccines against Echinococcus infections. Several studies conducted in recent decades have described the specific traits of the protective antigens found in E. multilocularis and their role in immunizing different animal hosts. The objective of the current systematic review was to summarize the findings of relevant literature on this topic and unravel the most effective vaccine candidate antigens for future research. A comprehensive search was conducted across five databases, including ProQuest, PubMed, Scopus, ScienceDirect, and Web of Science, until March 1, 2023. Two reviewers autonomously conducted the screening and evaluation of data extraction and quality assessment. In the present study, a total of 41 papers matched the criteria for inclusion. The study findings indicate that the combination of Em14-3-3 and BCG is widely considered the most often employed antigens for E. multilocularis immunization. In addition, the study describes antigen delivery, measurement of immune responses, adjuvants, animal models, as well as routes and doses of vaccination. The research indicated that recombinant vaccines containing EMY162, EM95, and EmII/3-Em14-3-3 antigens and crude or purified antigens containing ribotan-formulated excretory/secretory antigens exhibited the most favorable outcomes and elicited protective immune responses.

Cystic echinococcosis (CE) is a significant health problem in both human and veterinary medicine. It is caused by the tapeworm Echinococcus granulosus (E. granulosus). The objective of this study was to investigate molecular diversity of E. granulosus from the paraffin-embedded human (FFPE) tissue samples using sequencing of mitochondrial genes. Thirty-five FFPE tissue samples were collected from different regions of Guilan province, north of Iran. Demographic data were recorded using a questionnaire. Five sections (1 mm) of the tissue were prepared and deparaffined using xylene and ethanol methods. Molecular analysis was performed using the Nad1 and Cox1 genes using PCR and DNA sequencing. Totally, 25 cases (71.43%) were women and 10 cases (28.57%) were men. The most affected age group was 21–30 yr old. The most of cysts were isolated from the liver (n = 19; 54.29%) and others in the lung (n = 16; 45.71%). The Cox1 and Nad1 genes were successfully amplified in 16 (45.71%) and 12 (34.28%) DNA samples from FFPE tissue. Sequencing analysis revealed that all samples were E. granulosus sensu stricto complex (G1 and G3). In this study, E. granulosus sensu stricto complex G1 and G3 were identified in human hydatid cysts and showed the presence of sheep/dog cycle in human infection. This finding confirmed and completed previous studies on the geospatial distribution of E. granulosus sensu stricto complex G1 and G3 in the southern and coastal areas of the Caspian Sea region.

Abstract Background Trichobilharzia regenti (T. regenti) is an avian schistosomatid fluke species that causes human cercarial dermatitis (HCD) in areas of aquaculture in northern Iran. Understanding the phylogenetic relationships and genetic diversity of this thread‐like fluke will deepen our thoughtful of avian schistosomiasis epidemiology and lead to more effective HCD control in the region. Objectives To determine the life cycle of nasal Trichobilharzia in aquatic birds as well as aquatic snails and also identify the haplotype diversity of the isolates in Mazandaran Province, northern Iran. Methods In the present study, adult or egg of Trichobilharzia isolated from aquatic birds as well as schistosomes cercariae isolated from aquatic snails in Mazandaran Province, northern Iran, belonged to the authors' previous research, were examined. Molecular studies and phylogenetic analysis were carried out on these schistosomes samples. Results The phylogenetic analysis of the ITS1 and COX1 genes in isolated schistosomes revealed that all samples belong to the T. regenti clade. Remarkably, based on phylogenetic results, these schistosomes samples from Anas platyrhynchos domesticus, A. platyrhynchos, Spatula clypeata and Lymnaea stagnalis grouped together with previously sequenced samples from Iran (Trichobilharzia cf. regenti). Unlike the phylogenetic tree and haplotype network of COX1 gene, ITS1 did not show distinct clusters. Conclusion This study completed the puzzle of the disease in Mazandaran Province by isolating and genotyping furkocercariae from L. stagnalis that was consistent with the isolated new genotype from ducks. For the first time in Iran, this confirmed the potential role of L. stagnalis snails in the transmission of the disease.

Currently, parasitic infections are one of the important health problems in the world, especially in developing countries. This study aims to investigate intestinal parasites with an emphasis on molecular identification through the analysis of mitochondrial COX1 and ITS2 gene sequences of Strongyloides stercoralis (S. stercoralis) and Trichostrongylus spp. in north of Iran. Five hundred forty stool samples were collected from medical diagnostic laboratories affiliated with Mazandaran University of Medical Sciences in Sari city, north of Iran. First, all the samples were examined using direct smear, formalin-ether sedimentation, and trichrome staining technique. Suspected samples of Strongyloides larvae were cultured in agar plate. Then, DNA was extracted from samples containing Trichostrongylus spp. eggs and Strongyloides larvae. To amplify DNA, PCR was performed and the samples with a sharp band in electrophoresis were sequenced by Sanger method. Overall, the prevalence of parasitic infections in the study population was 5.4%. The highest and the lowest level of infection was observed with Trichostrongylus spp. and S. stercoralis at 3% and 0.2%, respectively. No traces of live Strongyloides larvae were seen in the culture medium of the agar plate. The six isolates obtained from the amplification of the ITS2 gene of Trichostrongylus spp. were sequenced, all of which were Trichostrongylus colubriformis. The sequencing results of COX1 gene indicated S. stercoralis. In the present study, the prevalence of intestinal parasitic infections in north of Iran has relatively decreased that its main reason can be due to the coronavirus epidemic and compliance with health principles. However, the prevalence of Trichostrongylus parasite was relatively high that it requires special attention to apply appropriate control and treatment strategies in this field.

Background: Hydatidosis is known as one of the most prevalent zoonotic diseases across the world. This complication is also endemic in Iran, followed by a higher risk of infection in rural areas. To our knowledge, there has been no study on the seroprevalence of hydatidosis in Sistan and Baluchistan Province, Southeast of Iran. The main objective of the current study was to examine the seroprevalence of hydatidosis and its risk factors in high-risk individuals (farmers and ranchers) living in Sistan and Baluchistan Province. Materials and Methods: This study included 500 serum samples, and the participants were requested to complete a researcher-made questionnaire. Subsequently, counter-current immunoelectrophoresis (CCIEP) and enzyme-linked immunosorbent assay (ELISA) methods were employed to analyze the anti-Echinococcus granulosus antibody. The analysis of the obtained data was conducted by logistic regression in SPSS software, version 22. Results: According to the results, four (0.8%) cases were found positive for anti-E. granulosus antibody by both CCIEP and ELISA tests. Seroprevalence of hydatidosis was more in rural people, compared to those in urban areas. It was also higher in illiterate people than in educated people. Nevertheless, seropositivity showed no significant differences with age, gender, occupational status, education level, place of residence, and contact with dogs (P>0.05). Conclusion: The prevalence rate of hydatidosis in Sistan and Baluchistan Province was similar to that in neighboring provinces. According to the findings, high-risk individuals offer remarkable information about the epidemiology of hydatidosis in Sistan and Baluchistan province in southeastern Iran. This could help to manage and prevent this infection.

Background: Trichostrongylus is an intestinal parasite that is highly prevalent in humans and livestock worldwide. There is limited information about the prevalence and epidemiology of Trichostrongylus species among the infected livestock in Mazandaran Province, northern Iran. This study aimed to identify Trichostrongylus spp. among small ruminants using morphometric and molecular methods. Materials and Methods: Small intestinal organs of sheep and goats, slaughtered in Mazandaran Province, were examined for infectivity with Trichostrongylus parasites. Primary species identification was conducted based on the morphological characterization of the male worms. The internal transcribed spacer (ITS) II regions of the ribosomal DNA of the worm tissues were amplified using the polymerase chain reaction (PCR) assay and then the product was subjected to sequencing. Subsequently, the PCR products of the ITS II region were subjected to digestion by HinfI and DraI restriction enzymes using the PCR-restriction fragment length polymorphism (RFLP). Results: Of 180 samples, 98 (54.44%) were confirmed positive for Trichostrongylus based on the conventional PCR. The digestion of the PCR products with HinfI and DraI facilitated the identification of three Trichostrongylus species, namely Trichostrongylus colubriformis (35%, 90.81%), Trichostrongylus axei (4%, 4.08%), and Trichostrongylus vitrinus (5%, 5.1%). Both morphometric and RFLP techniques resulted in the differentiation of the three Trichostrongylus species. Conclusion: The present study was the 1st attempt in the last 30 years for the identification of Trichostrongylus species in small ruminants in Mazandaran Province. The findings of this study can be helpful for epidemiological and ecological studies, the establishment of effective control programs, and the management of gastrointestinal parasites in Mazandaran Province.

Background: Antimalarial drug resistance is one of the important challenges for governments in the fight against malaria. Molecular surveillance of antimalarial drug resistance supports early detection of how the recommended treatments work. This allows immediate action to reduce any threat and prevent it from spreading. Therefore, the aim of this study was to evaluate the frequency of dihydrofolate reductase (dhfr) mutants in Plasmodium falciparum resistance to pyrimethamine in Iranian malaria patients. Materials and Methods: In 2020, 27 patients (21 males and 5 females) with imported P. falciparum cases were studied. The nested-PCR technique first confirmed the species in all samples and then amplification was done by the semi-nested-PCR method in order to detect single nucleotide polymorphisms (SNPs) in dhfr gene related to pyrimethamine resistance. Results: All samples in the 18S rRNA gene had species-specific bands for P. falciparum strains. In the sequence analysis of pfdhfr gene amplification after comparison with the standard strain (wild type), 21 patients had a double mutation (C59R+S108N) and six patients had a triple mutation (N51I+C59R+S108N) of pyrimethamine resistance. Conclusion: The results of this study showed that the susceptibility of P. falciparum to pyrimethamine in the treatment of malaria is significantly reducing. These findings can raise concerns about pyrimethamine resistance in P. falciparum. Due to the high emergence of double and triple mutants related to pyrimethamine resistance, the malaria surveillance and treatment systems in Iran, the use of pyrimethamine should be considered.

Background: Echinococcosis or Hydatid disease is a zoonotic disease that is caused by Echinococcus granulosus. The disease is a high public health concern in Iran, but there is little known about the genetic diversity and epidemiology of Echinococcus spp. in Iranian shepherd dogs. Materials and Methods: Fifty shepherd dogs were investigated for the adult worm of E. granulosus from May 2020 to April 2021 in Sistan and Baluchestan Province, the southeastern border of Iran. DNA extraction of samples and amplifying was done, and sequence analysis of mitochondrial genes (Cox1 and Nad1) was performed. Results:Out of 50 shepherd dogs, 11 cases (22%) were infected with E. granulosus. No significant difference was observed regarding demographic factors (P>0.05). The phylogenetic analyses of Cox1 and Nad1 sequences demonstrated G1 genotype (sheep strains) in all isolates. Based on sequence analyses, a low (Cox1, Hd [haplotype diversity: 0.200; Hn [number of haplotypes]: 2) to moderate (Nad1, Hd: 0.533; Hn: 4) genetic (haplotype) diversity of E. granulosus G1 genotype and low nucleotide diversity (π: 0.00052-0.00243) were observed. Conclusion: The first identification of a sheep strain (G1) in the final host in Sistan and Baluchestan Province indicates that potential intermediate hosts play a secondary role in preserving the biology of the dog-sheep cycle. The present study’s findings enrich our knowledge about the prevalence of E. granulosus, the classification of strains, and the genetic diversity of the parasite in Iranian herding dogs. This information helps develop strategies and programs for monitoring and controlling infection in stray dogs in the region

Background and purpose: There are few treatment options available for treatment of toxoplasmosis and effective drugs have serious toxic effects. In this study, the in vivo and in vitro anti-toxoplasma activities of Heracleum persicum and Foeniculum vulgare fruits essential oils were investigated. Materials and methods: In vitro, Vero cells were incubated with different concentrations of essential oils or pyrimethamine (positive control) and the cellular viability was determined. Next, Vero cells were infected with T. gondii (RH strain) and treated with agents. Then, the CC50, IC50, and selectivity index (SI) were calculated. Moreover, in vivo, the effect of oils on survival times of Balb/c mice infected with T. gondii were determined. Results: In vitro results showed that the oils exhibited less cell toxicity than pyrimethamine. The selective index was 2.94, 6.96, and 3.06 for Heracleum persicum, Foeniculum vulgare, and pyrimethamine, respectively. Background and Aim: There is a large amount of information on intestinal parasites in stray dogs and golden jackals (Canis aureus) in Mazandaran Province, Iran. However, there is little information about foxes and wolves, which have a potential role in the spread of dangerous parasitic diseases, such as echinococcosis and toxocariasis. The aim of the present study was to identify the genus or species of parasitic worms in stool samples obtained from carnivores in Mazandaran Province, Iran, from August 2017 to April 2018. Materials and Methods: A total of 274 fecal samples were collected from carnivores, including dog, fox, wolf, and C. aureus in three areas of Mazandaran Province, Iran. All specimens were examined by centrifugal fecal flotation using a solution of Sheather's sugar to detect helminths eggs. Then, all samples were assessed using a light microscope. Data analysis was performed by SPSS version. 18 (Chicago, IL, USA). Results: In this study, seven genera of helminths were observed, including Ancylostoma, Uncinaria, Toxocara, Dipylidium, Toxascaris, Taenia, and Spirocerca. The prevalence of helminth infections was 97.7% (127 out of 130), 56.7% (51 out of 90), 51.4% (18 out of 35), and 52.6% (10 out of 19), among dogs, C. aureus, foxes, and wolves, respectively. The highest prevalence of Ancylostoma and Toxocara infections occurred in the eastern and central areas of the province (42.1% and 35.7%, respectively). Conclusion: Based on the results of this study, the infection with intestinal zoonotic helminths in carnivores was an important public health factor in Mazandaran. Therefore, these infections can be potentially harmful to humans and other animals.

Background: The larval stage of the tapeworm (cestode) Echinococcus granulosus is the etiological agent of hydatidosis or cystic echinococcosis, which is the zoonotic parasitic disease causing morbidity and mortality in both humans and livestock. Due to a lack of accurate data on the human isolates of E. granulosus in Mazandaran Province, northern Iran, the current study aimed to survey the population genetic pattern of cystic echinococcosis isolated from humans by sequencing the mitochondrial genes of NADH dehydrogenase subunit 1 (nad1). Methods: Overall, 47 formalin fixed paraffin-embedded tissue (FFPT) blocks were collected from patients' files in various pathology departments of Mazandaran Province in Iran from 2003 to 2015. PCR was performed to amplify a 398bp DNA fragment of mitochondrial nad1. PCR products were sequenced by Bioneer Corporation (South Korea), and the resulting data were analyzed via relevant software to determine the genotypes. Results: The nad1 gene was successfully amplified on 10 from all of the E. granulosus isolates. Overall, 66.6% and 33.3% of the isolates in the studied area displayed the G1 and G2-G3 genotypes, respectively. Conclusion: This study may provide the foundation for further studies in revealing the regional transmission patterns and also in designing adequate control procedures.

Background: Avian schistosomes are considered as main causative agents of human cercarial dermatitis (HCD) in Iran. The study was conducted to determine bird schistosomes in their final and intermediate hosts, in main wetlands of Mazandaran. Methods: A total of 255 domestic and wild ducks were collected and the infection of nasal tissues of five (Anas platyrhynchos domesticus, Aythya ferina, Cairina moschata, Anas platyrhynchos and Spatula clypeata) species were analyzed using morphological techniques. Also, 1687 freshwater snails were collected and surveyed by cercarial shedding and crushing tests. Detection of HCD was performed for the presence of clinical symptoms of itching and maculopapular rashs by physical examination. Results: Of 255 ducks, in 41 (16%) infection with nasal Trichibilarizia spp. were recognized by observing eggs and/or adult worms. The most infected ducks were Anas clypeata and Anas platyrhynchos domesticus. Overall, 0.17% of snails were infected with avian schistosomes. Also, clinical examination of 951 rice farmers revealed that 588 (61.82%) of them were suffered from HCD. Conclusion: Our data suggest that domestic ducks could play a prominent role as a reservoir host for establishing life cycle of Trichobilharzia in the area. Also, existence of domestic reservoir ducks and suitable snail hosts in ponds and paddy fields of this area, climate conditions of the wetlands in Mazandaran leads to a high incidence of HCD.

Background: Free-living amoeba (FLA) belonging to Acanthamoeba spp., Naegleria, and Balamuthia mandrillaris are the soil-born protozoa. This study aimed to survey the occurrence of FLA, including Acanthamoeba spp., B. mandrillaris, Vermamoeba spp., and Naegleria spp., in soil samples collected from various districts of Mazandaran Province (Northern Iran) from July to December 2018. Methods: Overall, 118 soil samples from the recreational and public places were surveyed for the existence of Acanthamoeba spp., Vermamoeba, Naegleria, and B. mandrillaris using both morphological key and molecular tools with genus-specific primers of JDP1, NA, ITS1, and Bal, respectively. To verify the taxonomic status of isolated amoeba, the phylogenetic tree was made based on sequences of 18S rRNA by MEGA (5.05) software with the maximum likelihood model. Results: Overall, 61/118 samples (51.6%) were contaminated with FLA, and based on the sequencing data, 29 isolates were successfully sequenced. Among the samples, all isolated Acanthamoeba (52.4%) belonged to the T4 genotype with amplification of the DF3 region (18S rRNA gene). Internal transcribed spacer (ITS) sequencing revealed the presence of one strain of Naegleria americana. Twenty-eight V. vermiformis were also confirmed based on Nuclear SSU rDNA. Morphological survey and PCR assay did not show any positive samples for B. mandrillaris. Conclusion: The present study indicates the occurrence of FLA in soil sources of the recreational and public places in Mazandaran province that it can be a severe risk to human health. Thus, more studies are expected to survey the infection source in patients with FLA-related diseases.

Background: Human Echinococcosisis a cyclo-zoonotic infection caused by tapeworms of the Echinococcus granulosus sensu stricto complex. The detection of mitochondrial genome data of genus Echinococcus can reflect the taxonomic status, genetic diversity, and population structure genetics. Methods: Totally, 52 formalin-fixed paraffin-embedded (FFPE) tissue samples from patients with histologically confirmed CE were collected from Mazandaran province, Iran in the period of Mar 1995 to May 2018. All extracted DNAs from (FFPE) tissue samples were subjected to amplify by polymerase chain reactions method targeting cytochrome c oxidase subunit 1 (cox1) gene. All PCR amplicons were sequenced to phylogenetic analysis and genetic diversity. Results: Molecular analysis showed that 50(96.1%) and 2 (3.84%) isolates were identified as G1 andG3 E. granulosus genotypes, respectively. DNA sequence analyses indicated a high gene diversity for G1 (Haplotype diversity: 0.830) and G3 genotypes (Hd: 1.00). Based on multiple sequence alignment analyses, 7 (13.46%; G1 genotype) and 2 (3.84%; G3 genotype) new haplotypes were unequivocally identified. Conclusion: G3 genotype (Buffalo strain) was identified from two human hydatidosis isolates in the region. Present study strengthens our knowledge about taxonomic status, transmission patterns of Echinococcus parasite to human and heterogeneity aspects of this parasite in clinical CE isolates of Northern Iran.

Objectives: Human cercarial dermatitis (HCD) is a water-borne zoonotic parasitic disease. Cercariae of the avian schistosomes of several genera are frequently recognized as the causative agent of HCD. Various studies have been performed regarding prevalence of bird schistosomes in different regions of the world. So far, no study has gathered and analyzed this data systematically. The aim of this systematic review and meta-analysis study was to determine the prevalence of avian schistosomes worldwide. Methods: Data were extracted from six available databases for studies published from 1937 to 2017. Generally, 41 studies fulfilled the inclusion criteria and were used for data extraction in this systematic review. Most of studies have been conducted on the family Anatidae. Results: The overall prevalence of avian schistosomes was estimated to be 34.0% (95%CI, 28%–41%) around the world. Furthermore, results displayed that, Allobilharzia visceralis and Trichobilharzia spp. had the highest frequency and their prevalence in the birds was 50.0% (95% CI, 3.0%–97.0%) and 32.0% (95% CI, 21.0%–0.36%), respectively. The results showed that the prevalence of avian schistosomes was 43.0% (95% CI, 29% - 56%) in the US and 38.0% (27.0% -50.0%) in Europe, which were higher than other continents, respectively. Conclusions: The prevalence of 34% shows that the bird schistosomes are very common zoonotic worms among aquatic birds in the world. Also, this study shows the importance of avian schistosome research when facing animal and human health of the future.

Aim: Human cystic echinococcosis (CE), caused by the larval stage of Echinococcus granulosus cestodes, is a globally distributed chronic disease that is an important socioeconomic and public health problem in humans and livestock in developing countries, including Iran. The aim of this study was to determine the overall seroprevalence of hydatid infection in the general population of Iran. Materials and Methods: This systematic review began by searching electronic databases in English (PubMed, Science Direct, Scopus, and Google Scholar) and Persian (Magiran, Scientific Information Database, Iran Medex, and Iran Doc). Results: Our search resulted in a total of 40 reports published from 1995 to 2015. Of 49,460 individuals surveyed, 3090 cases of hydatidosis were reported. Community-based studies showed that the seroprevalence of CE in the Iranian general population was 6.0% (95% confidence interval: 5.0-7.0%). The age group with the highest CE seroprevalence was 20-40 years, and the lowest one was in the under 20 year's group. The seroprevalence of hydatidosis in males was significantly higher than that in females. In addition, the intended rate was significantly higher in rural regions than in urban areas. Conclusion: Management program for developing more efficient diagnostic tests should be established. Further, cost-effective preventive approaches, including relevant research, should be considered. Finally, hydatid cyst control programs that are important for interrupting the transmission of human CE should be improved.

Background: The purpose of this study was to investigate the current knowledge on the epidemiology of importance zoonotic parasitic diseases in free-ranging canids of Mazandaran, north of Iran. Methods: Overall, 63 small intestinal samples of animals (20 stray dogs and 43 golden jackals) were collected from April 2017 to May 2018. The intestine contents were studied to detect and identify helminth infections. Additionally, 274 fecal samples (130 dogs, 35 fox, 90 golden jackal and 19 wolf) were examined by Sheather's flotation method for detection of Taenia eggs. Results: Sixty (95.2%) animals were infected with at least one species of intestinal helminth. the intestinal helminths were found in dogs and golden jackals included: Dipylidium caninum (25.3%), Uncinaria stenocephala (52.3%), Ancylostoma caninum (41.2%), Mesocestoides spp. (33.3%) and Toxocara canis (14.2%). In fecal examination, 2.5% of samples contained Taenia eggs, and through a species-specific PCR, 1.09% of these samples were confirmed positive for Echinococcus granulosus. Conclusion: There is a high prevalence and clear risks of zoonotic helminths in free-ranging carnivores in Mazandaran province, north of Iran. Therefore, understanding the epidemiology of zoonotic parasite infection is useful for health care access both domestic animals and humans health.

Background and purpose: Taxonomic identification of larval and adult stages of Taenia hydatigena from other Taenia species based on morphometric characteristics of the rostellar hooks in intermediate and definitive host is important in medical and veterinary parasitology. Therefore, this study aimed at identifying the larval stage of Taenia hydatigena (Cysticercus tenuicollis) based on morphological characteristics of rostellar hooks isolated from sheep and goat in Mazandaran province, Iran for the first time. Materials and methods: This study was conducted in 100 samples of rostellar hooks of the larval stage of Taenia hydatigena isolated from sheep and goats in 2014-2015. Morphological characteristics of large and small hooks in the number and size characters were determined and measured by calibrating microscopes. Data analysis was done in SPSS applying Chi-square and One-way ANOVA. Results: The mean number of large and small hooks in goat and sheep isolates were 30.8±3. Morphometric characteristics of hooks in the shape and size were similar in both animal isolates. But significant differences were seen between native and nonnative isolates in mean width of large hooks length (P< 0.05). Conclusion: This study identified the larval stage of Teaina hydatigena from other species of Teaina according to characteristics of rostellar hooks, which is of great significance in taxonomic studies in parasitology.

Background and purpose: Studying the different diagnostic aspects of hydatid cysts (HC) plays a major role in early diagnosis and treatment. This study was conducted to evaluate the clinical and laboratory characteristics of HC cases operated in educational hospitals affiliated with Mazandaran University of Medical Sciences. Materials and methods: In this descriptive study, the records of all cases with HC (n=95) who underwent surgical treatment in two educational hospitals within 11 years (2004-2015) were studied and analyzed. Aim: Parasitic infections, especially of the zoonotic-parasitic type, are the most important health, economic, and social problems in developing countries, including Iran. The aim of this study was to review systematically the available data on gastrointestinal parasites of carnivores in Iran and their ability to infect humans. Materials and Methods: Studies reporting intestinal parasites of carnivores were systematically collected from nine electronic English and Persian databases and Proceedings of Iranian parasitology and veterinary congresses published between 1997 and 2015. A total of 26 studies issued from 1997 to 2015 met the eligibility criteria. Results: The pooled proportion of intestinal parasites of carnivores was estimated as 80.4% (95% confidence interval=70.2-88.8%). The overall prevalence of gastrointestinal parasites in dogs, cats, foxes, and jackals were 57.89%, 90.62%, 89.17%, and 97.32%, respectively. Dipylidium caninum (20.45%), Toxocara spp. (18.81%), Taenia hydatigena (15.28%), Mesocestoides lineatus (11.83%), Echinococcus granulosus (10%), and Toxascaris leonina (8.69%) were the most frequently observed parasites. Conclusion: High prevalence rates of zoonotic intestinal parasites of carnivores particularly Echinococcus spp. and Toxocara spp. increase the risk of acquiring zoonotic infections such as cystic hydatid, alveolar cysts, and visceral or ocular larva migrants in Iranian people. Therefore, it is essential for public health centers to develop more effective control strategies to decrease infections rates in carnivores' populations.

Background and purpose: Wild boars (Sus scrofa) are potential reservoirs for a lot of zoonotic diseases, so it is possible to transmit these infections to wild and domestic animals and also humans. This research aimed to survey the protozoan prevalence of Sus scrofa in Mazandaran Province, North of Iran. Materials and methods: In a descriptive cross-sectional study a total of 21 wild boars was captured beween December 2012 and March 2014. Faeces collected from the intestinal tracts of each animal was placed in appropriate fixative. Temporary staining with Lugol’s solution was performed in order to find the protozoan cysts and trophozoites. The stool samples were further studied using sedimentation and flotation methods. Then, fixed stool smears were examined by trichrome and Ziehl–Neelsen staining. Results: There were 21 samples of which 12 (57.14%) were found to be infected with one or more protozoan species. The prevalence of contamination with intestinal protozoan was as follows: 33.33% trophozoites and cysts of Balantidium coli, 28.57% oocysts of Eimeria spp., and 9.52% cysts of Giardia spp. Conclusion: According to current findings, wild boars in Mazandaran province are infected with a lot of zoonotic protozoan, which create potential risks for other animals and people in the region.

Background and purpose: Toxoplasmosis is a common zoonotic disease that is of particular importance in women before and during pregnancy. Therefore, this study aimed at investigating the seroepidemiology of Toxoplasma gondii in pregnant women and its risk factors in Golestan province, north of Iran. Materials and methods: A descriptive cross-sectional study was carried out in 440 blood samples of pregnant women before delivery (2014-2015). Blood serum samples were isolated to determine IgG and IgM anti-Toxoplasma antibodies and examined by ELISA test. Data analysis was done in SPSS V20 using Chi-square test. Results: Among the samples, 155 (35.2%) were IgG and 25 (5.7%) were IgM positive and 9 (2%) cases were positive for both antibodies. A significant correlation was observed between T. gondii infections with abortion and ethnicity (P0.05). Conclusion: In this study serological test showed that about 57% of pregnant women had no anti-Toxoplasma antibodies in Golestan province. This means that these people are more prone to acute infection with Toxoplasma. Therefore, there is a need to raise awareness, especially in at-risk groups.

Background and purpose: Accurate information on clinical and laboratory findings of parasitic diseases in medical centers could be effective in evaluating various aspects of their disease. Therefore, the aim of this study was to evaluate the clinical and preclinical findings of the intestinal parasitic infections in patients in gastrointestinal clinics in Sari, Iran, 2006 to 2015. Materials and methods: A cross-sectional study was carried out using the medical records of 300 cases of gastrointestinal disorders with intestinal parasitic infections. Clinical and laboratory data were collected and analyzed in SPSS V.16. Results: Among the patients with parasitic infections there were 57.7% females, 64.3% lived in rural areas, 81% were married, and 40% were illiterate or had primary school educational levels. The most important parasitic diseases in patients were Giardiasis (44.7%), Amebiasis (12.3%), Enterobiasis (10.6%), Trichostrongylus (10.2%), Strongyloidiasis (9.78%) and, Hymenolepiasis (6.38%). Abdominal pain, bloating, and diarrhea were the most common symptoms. Conclusion: According to this study, intestinal parasitic infection is a high priority for diagnostic purposes in gastrointestinal clinics. Therefore, diagnosis is important for appropriate treatment. Future studies are suggested to use sensitive and specific methods such as molecular methods to identify the parasites.

Giardia lamblia is an intestinal protozoan parasite; hence, to diagnose the cyst, it is necessary to find a reliable, simpler and less expensive diagnostic method in the laboratory. Therefore, in order to choose a more sensitive, cheaper and applicable diagnostic method in the laboratory ; three methods, namely, sucrose gradient, wet mount and formalin-ether in fecal samples from BALB/c mice infected with the protozoan Giardia, were studied. In this diagnostic laboratory study, 200 stool samples from infected mice (BALB/c) with Giardia were studied. Stool specimens from three direct smear method, formalin-ether and sucrose were prepared. A total of 200 specimens for each method were prepared and examined by microscope. Furthermore, it enhanced the accuracy check at each stage; the feces of healthy mice were used as negative controls for comparison. Stool examination revealed G. lamblia cysts in mice infected BALB/c from 200 samples for each method. In the direct method, formalin - ether and sucrose gradient, 101 (55.00%), 166 (83.00%) and 180 (94.00%) cases were positive respectively. Therefore, the sucrose method has demonstrated the highest sensitivity (94.00%), compared to other methods. According to the results of this study, sucrose is a sensitive method for high-throughput and cost-effective diagnostic method, than previous methods. Therefore, sucrose method is suggested suitable and can be used in place of formalin-ether. Given their potency, the sucrose gradient may be recommended to undergo futher studies.

Background: Ascariasis and trichuriasis are the most common intestinal geohelminthic diseases, and as such they are significant in terms of clinical and public health. This study was done to determine prevalence, status and geographic distribution patterns for Ascariasis and Trichuriasis. The study was done in the period 1991-2014 in northern Iran using Aregis 9.2 software. Methods: This was a review study, using description and analysis, of geographical distribution of Ascaris and Trichuris relating to townships in Mazandran province, northern Iran, covering a 23-year period. Data were collected from a review of the relevant literature, summarized and classified using Arc GIS, 9.2 to design maps and tables. Results: Based on results presented in tables and maps, means for prevalence of Ascaris and Trichuris were divided into five groups. The maximum prevalence rate of Ascaris was 16.3% reported in rural areas of Tonkabon in 1981-1982. Prevalence means for Ascaris in the central and western areas of Mazandaran province were 2%-4% and 4%, respectively. The maximum prevalence of Trichuris in the rural area of Tonkabon was 22.5% and the lowest 0.06% was among the cattle breeder’s in rural areas of the province at in 2002-2003. Conclusion: Data presented in this study provides information useful to health care workers researchers and health administrators, especially for physicians, clinicians and for future research. Also, it is necessary to control and prevent geohelminthic parasitic infections, particularly in rural areas by public education for families, health authorities and health care systems.

Human echinococcosis is an infectious disease caused by tapeworms belonging to the species Echinococcus. This parasite has a worldwide distribution and is considered a neglected tropical disease by the World Health Organization. Due to the diversity of Echinococcus spp. hosts, as well as variation in geographical, climatic, and socio-ethnic conditions, the question of the strains or genotypes of Echinococcus spp. that are involved in human infections is important. The aim of this study was to provide a summary of the available data on genotypes of Echinococcus obtained from the Iranian population. Four international databases (PubMed, Scopus, Science Direct, and Web of Science) and 4 Persian databases (Magiran, Scientific Information Database, Iran Medex, and IranDoc) were searched for cross-sectional studies that reported the genotypes of Echinococcus spp. in human echinococcosis cases using molecular methods in Iran through July 2018. The Newcastle-Ottawa Scale was used to assess the quality of the selected studies. A total of 559 cases of human cystic echinococcosis were reported in the 21 included articles. The majority of cases belonged to genotype G1 (89.2%; 95% confidence interval [CI], 80.1 to 95.8), genotype G6 (8.2%; 95% CI, 2.8 to 15.9), and genotype G3 (2.3%; 95% CI, 1.1 to 3.9). Since genotype G1 of Echinococcus appears to be the most prevalent genotype affecting humans in Iran, disease control initiatives aimed at sheep intermediate hosts may be the most beneficial. In addition, educational programs and serological screening in individuals may help reduce the national impact of the disease.

BackgroundToxoplasma gondii (T. gondii) is an obligate intracellular opportunistic parasite that is the causative agent of toxoplasmosis. This parasite accounts for mental disorders; however, the relationship between T. gondii infection and depressive disorder is unclear. Regarding this, the present systematic review and meta-analysis was conducted to investigate the scientific evidence regarding the potential association between major depression disorder (MDD) and Toxoplasma infection.MethodsFor the purpose of the study, the articles related to the subject of interest were systematically searched in seven electronic databases. Special attention was given to the studies examining T. gondii seropositivity level in depressed patients and controls.ResultsThe search process resulted in the identification of a total of 30 publications meeting the inclusion criteria and published up to April 2018 for the systematic review. Furthermore, 29 studies met the inclusion criteria to be entered into meta-analysis. Our meta-analysis involved the review of cross-sectional studies including 1657 depressed patients and 19565 individuals as controls and case-control studies entailing 1311 depressed cases and 6015 controls without depression. 1582 depressed people participated in cross-sectional studies whose results were reported as odds ratio (OR). In addition, the total number of participants was 15068 in this type of studies. Statistical analysis indicated that the pooled OR of the risk of anti-T. gondii IgG antibody in depressed individuals in case-control and cross-sectional studies was 1.15 (95% confidence interval (CI): 0.95-1.39).ConclusionsAs the findings of the reviewed articles indicated, toxoplasmosis is not a risk factor for MDD. However, it is necessary to perform further research to clarify the detailed association between T. gondii and dysthymia or mild and moderate depression. Furthermore, it is recommended to better investigate the effect of antibody titers on the relationship between depression and T. gondii infection.

Freshwater snails, as the first intermediate hosts of trematodes, can cause health hazards in animals and humans. Recently, the World Health Organization has included Iran in a list of 6 countries known to have serious problems with fascioliasis. In addition, cercarial dermatitis is a job-related disease that is seen often in paddy workers, agricultural labourers, and fishermen in Iran, particularly in Mazandaran Province. Many studies have been conducted in Iran to survey larval trematodes in freshwater snails. However, to the best of our knowledge, no comprehensive data exist regarding infections in gastropods. Therefore, the aim of the present study was to estimate the types and prevalence of cercarial infections in snails in Iran. Electronic English-language and Persian-language databases were searched to identify 24 published articles reporting the prevalence of trematode infections in snails (9 species from 6 families) in various provinces of Iran. In total, 4.4% of gastropods were infected with the larval stages of trematodes. According to the studies reviewed in this meta-analysis‚ the highest infection prevalence was found in Radix auricularia (9.9%). Twelve larval species of trematodes were identified, and the highest prevalence of cercariae was found for Echinostomatidae cercariae (4.3%). Among the provinces explored, West Azerbaijan had the highest prevalence of infected snails (16.9%). The presence of trematodes in snails could pose a serious health problem in Iran. Thus, further studies are necessary to characterize these infections in other provinces.

Background: Cryptosporidiumis known as an opportunist disease-causing agent in man in recent decades. It causes diarrhea and intestinal disorders in the immune deficit and immune competent individuals. This study was aimed to investigate the clinical characteristics of HIV/AIDS patients withcryptosporidiosisinfection. Methods: This cross-sectional descriptive study was performed on 53 HIV/AIDS patients referred to the Behavior Disease Consultation Center of Imam Khomeini Hospital in Tehran, Iran in 2013. First, the patients were studied clinically and the context data were recorded in a questionnaire for parasitological examination and referred to the laboratory for eosinophil count, and CD4 count per ml of blood. Results: Cryptosporidiosiswas observed in 4 (7.6%) of the total 53 HIV/AIDS patients. The highest prevalence of infection was observed in the age range of 30-39 yr.Itwas observed in different sexes as 5.7% of male and 1.9% of female, but statistically was insignificant (P=0.163).75% of patients had no intestinal symptom, 11.4% with acute diarrhea and 3.8% with chronic diarrhea. Cryptosporidiosis cases were observed in 5.7% of patients without intestinal symptom. Conclusion: Practitioners in the clinicalexamination for the detectionof the opportunistic intestinal protozoan infection should use clinical and paraclinical characteristics of the HIV/AIDS patients for the diagnostic of Cryptosporidium and other opportunistic parasitic diseases.

Summary: Toxoplasma gondii (T. gondii) is a protozoan parasite that can cause toxoplasmosis in humans. However, there is no current data regarding Toxoplasma infection among individuals who were referred to medical laboratories in Mazandaran province (northern Iran). Therefore, we performed a population-based study of Toxoplasma seroprevalence in this region. A total of 1832 sera samples (from 654 men and 1178 women) were collected from people who were referred to medical laboratories in different cities throughout Mazandaran province between March and July 2012. The serum titers of anti-T. gondii IgG and IgM were measured using enzyme-linked immunosorbent assays. The seroprevalence of anti-Toxoplasma IgG was 55.5%; and 14.4% of the positive samples were seropositive for anti-Toxoplasma IgM. The highest seroprevalence was observed among people who were >50 years old (90.6%), and the lowest seroprevalence was observed among children who were 0–9 years old (9.4%; P

Background: Wild boars (Sus scrofa) are distributed worldwide and found in many parts of Iran. Although S. scrofa is reservoirs for many parasites, there is little data on helminthic prevalence in them. We aimed to survey the status of helminthic infections in S. scrofa in the Mazandaran Province of northern Iran. Methods: Twenty-one wild boars were captured and examined for helminth infection during Dec 2012-Mar 2014. Adult worms such as Macracanthorhynchus hirudinaceus were identified by helminth size and shape, and the arrangement of the proboscis hooks. The sedimentation and flotation techniques were used to detect parasite eggs and larvae in faecal samples. Muscle samples were also surveyed for Trichinella larvae by artificial digestion method. Results: Of the 21 samples, 13 (61.9%) were infected with one or more helminth species. Seven helminth types were identified in the alimentary track, comprising 5 nematodes, 1 trematode, and 1 acanthocephalan, with prevalence rates of Macracanthorhynchus hirudinaceus (57.14%), Globocephalus spp. (33.33%), Trichuris suis (19.04), Gongylonema pulchrum (14.28%), Fasciola hepatica (14.28%), Dioctophyma renale (4.76%), and Ascaris suum (4.76%). Conclusion: Wild boars might be involved in transmitting zoonotic parasites to humans. The abundance of these animals near human habitation creates favorable conditions for infection. So the risk of parasitic helminth diseases increases in other animals and humans.

Background: Intestinal parasites are the most common causes of gastrointestinal disease in canine. Stray dogs and wild candies can represent potential reservoirs of enteropathogens to other hosts. Therefore, present study determined the prevalence of intestinal parasites in canine in Mazandaran Province, northern, Iran. Methods: Overall, 58 small intestinal samples of animals (42 stray dogs and 16 jackals) were collected from Oct 2012 to Dec 2013. The intestine contents were studied to detect and identify helminth infections. Then, the helminths were collected and their morphological traits were identified. Results: Overall among infected stray dogs and jackals, 11 species were found. Three species of nematodes, seven species of cestodes and one trematode were observed. The prevalence of gastrointestinal helminths of stray dogs and jackals were 59.5% and 50%, respectively. Among registered zoonotic helminths A. caninum was the predominant parasite both stray dogs and jackals. Interestingly, Spirometra spp. was reported in these animals. Moreover, A. caninum showed a higher percentage rate in center region of province. Conclusion: There are the clear risks of zoonotic helminths parasites infection in this region. Therefore, understanding the epidemiology of zoonotic parasite infection is useful for health care access both domestic animals and humans health.

Abstract Background: Stray dogs are considered potential reservoirs for zoonotic diseases. Previous helminthic surveys in Iran, have accounted for mainly species of nematodes and cestodes, and rarely digeneans. Methods: We accessed 42 car-crashed stray dogs from the Farah Abad Region in the Mazandaran Province (North Iran) between Oct 2012 and Dec 2013, to be inspected for parasites. Helminths were collected from the intestine and they were morphologically studied. Results: We found five adult digeneans from the family Brachylaimidae, identified as Brachylaima sp. Worms were assigned to the genus based on the shape of the body, the position of genital pore, cirrus sac and testes, and the extension of the vitellarium. Absence of additional information on the developmental stages of the parasite precluded its specific identification. As the geographic distribution of species of Brachylaima is restricted to the Mediterranean region, we raise the hypothesis that dogs may become infected with parasites through the consumption of helicid snails when searching for food on the street. Conclusion: This is the second report of a species of Brachylaima in Iran and the third digenean species from stray dogs in the area. We want to raise the attention of researchers to helminthic surveys in potential zoonotic reservoirs like stray dogs.

Abstract Background Hydatidosis is considered to be a neglected cyclo-zoonotic disease in Middle East countries particularly northwestern Iran which is caused by metacestode of tapeworm Echinococcus granulosus sensu lato. Human hydatidosis is a high public health priority in the area, however there is little known from a morphometric and phylogenetic perspective on molecular epidemiology of adult Echinococcus spp. in Iranian stray dogs. Methods 80 dogs (38 males and 42 females) were collected during June 2013 to April 2014 in northwestern Iran. The isolated parasites from each dog were distinguished by morphometric keys including small, large hook length and blade length. Subsequently, isolates were confirmed by sequencing of mitochondrial cytochrome oxidase subunit 1 gene. Results 16 (8 males and 8 females) (Prevalence 20 %) out of 80 dogs were infected to genus Echinococcus. With regard to demographic factors, the frequency of parasitism in both male, female adults and their age groups showed no difference (P > 0.05). The phylogenetic analyses of cox1 sequences firmly revealed the 13 sheep strains (G1), one buffalo strain (G3), one camel strain (G6) and one mixed infection. The findings of rostellar hook morphology show an intraspecies variation range among G1 isolates. However, hook measurements in Echinococcus derived from G1 (sheep strain) were not a significant difference from those G6 and G3 strains. Six unique haplotypes were identified containing a high range of diversity (Haplotype diversity 0.873 vs. Nucleotide diversity 0.02). Conclusions First presence of camel strain (G6) in this region seems to indicate that potential intermediate hosts play a secondary role in the maintenance of camel-dog biology. Current findings have heightened our knowledge about determination of Echinococcus prevalence, strains of taxonomy and genotypic trait of parasite in Iranian stray dogs which will also help in the development of strategies for monitoring and control of infected stray dogs in the area.

Background and Objectives: Taking into account a few principles concerning human being, it becomes plausible that the human spirit would also have a similar reaction to spiritual “medicine” provided to it. In order to better understand how this is possible, we must consider the means by which the human spirit becomes resistant to spiritual remedies and compare them with the resistance developed by the body against physical drugs. As such, this research aimed at creating a comparative analysis between the elements that cause the human spirit to become resistant against spiritual remedies in comparison to the body’s resistance against physical treatments (e.g. drugs and other physical treatment). Methods: The research at hand highlights the conclusions of an overall study of the Holy Quran, books of Islamic narration, and extensive Internet research concerning this subject. With these resources, the various aspects of the spirit’s resistance against spiritual remedies were discussed in detail. Results: According to Holy Quran and Islamic narrations: Based on the expectations which God has of man, his heart (i.e. spirit) has the potential to fall under one of two categories – positive or negative. An afflicted heart may at times, like an afflicted body, become resistant against a remedy designed to cure it. In both cases of physical or metaphysical resistance, the underlying element that causes this resistance as well as the symptoms which accompany it are similar to one another. Having considered the teachings found in religious texts, this research discovered the underlying causes of spiritual resistance, and outlined some solutions which can prevent this issue from arising in the first place. Conclusion: Based on the standards of health and spiritual wellbeing as outlined in Holy Quran, it is said that some hearts are unhealthy and require treatment and healing. In Holy Quran, there is also no doubt in it, guidance to the God wary. Considering the fact that Holy Quran is the Word of God, we establish our full confidence and trust within it. Therefore, the causes and symptoms of spiritual resistance against metaphysical cures, which have been outlined in this article, have been taken from Quranic teachings and should be carefully studied.

Background: Giardiasis is a one of the most prevalent intestinal parasitic diseases in human, treatment of this disease through medicinal plants is very important since parasite resistance to chemical drugs exists. Thus, in this study, the in vitro anti-giardial activity of chloroformic extract of Tanacetum parthenium and Artemisia annua on cyst and trophozoite of Giardia lamblia were separately investigated.Materials and Methods: In this experimental study chloroformic extracts of Artemisia annua and Tanacetum parthenium on cyst and trophozoite of G. lamblia in vitro were prepared in 1, 10, 50 and 100 mg∕ml concentrations for 5, 10, 30, 60 and 180 min. Purified cysts were used for encystations and culture in TYI-S-33 medium. Then, 2 ml of each solution was placed in test tubes, to which 10,000 washed cysts and trophozoites were added. The contents of the tubes were gently mixed and incubated. The percentages of dead parasites were determined by counting 500 cysts. Non treated parasites were considered a control group in each experiment and the viability of the parasites checked with Eeosin staining and statistical analysis were done. Results: The results showed that chloroformic extracts of A. annua at 100 mg/ml concentration affected on Gardia cyst 86% and 100% trophozoite after 1 hour. T. parthenium at 50mg/ml concentration killed cysts (83%) and trophozite (100%) after 1 hour, respectively. T. parthenium chloroformic extract had a better effect on cyst and trophozoite of Giardia at 50 mg/ml after 1 hour exposure than A. annua extract. Conclusion: According to this study, A. annua and T. parthenium chlorofomic extracts could be considered as a more effective anti-giardial agent. Chloroformic extract of T. parthenium was also shown the anti-giardial activity compared with A. annua and control groups at all exposure times. Therefore, in the future research using these plants are recommended against Giardia in low concentration in the in vivo, also to find fractions of the pharmacological effects of these plants.

Background: The aim of the present study was genotyping of Echinococcus granulosus isolates from dogs and jackals in Mazandaran Province, northern Iran, and using partial sequence of the mitochondrial cytochrome c oxidase subunit 1 gene (cox1). Methods: E. granulosus isolates (n = 15) were collected from 42 stray dogs and 16 jackals found in south of the Caspian Sea in northern Iran. After morphological study, the isolates were genetically characterized using consensus sequences (366bp) of the cox1 gene. Phylogenetic analysis of cox1 nucleotide sequence data was performed using a Bayesian Inference approach. Results: Four different sequences were observed among the isolates. Two genotypes [G1 (66.7%) and G3 (33.3%)] were identified among the isolates. The G1 sequences indicated three sequence profiles. One profile (Maz1) had 100% homology with reference sequence (AN: KP339045). Two other profiles, designated Maz2 and Maz3, had 99% homology with the G1 genotype (ANs: KP339046 and KP339047). A G3 sequence designated Maz4 showed 100% homology with a G3 reference sequence (AN: KP339048). Conclusion: The occurrence of the G1 genotype of E. granulosus sensu stricto as a frequent genotype in dogs is emphasized. This study established the first molecular characterization of E. granulosus in the province.

Background: Babesia is blood-tissue parasite, which are transmitted by hard ticks from Ixodidae family. The parasite is the cause of Babesiosis among ruminants. Babesia is as one of main fatal factors among livestock in endemic regions such as Iran. The aim of this study was to investigate Babesia spp infection using microscopic and molecular methods among small ruminants in Mazandaran and Golestan provinces, northern Iran, in 2011-2012. Materials and Methods: In this cross-sectional study, 22 flocks of total 220 sheep and goats were selected from different regions of these provinces. Subsequently, ten suspected animals were selected randomly and blood sample were taken from their ear. To diagnose thick and thin smears were prepared and stained with Geimsa stain and then examined under light microscope. Standard PCR and Semi nested PCR was performed to differentiate genus of Theileria and Babesia, also identify the species of Babesia. Results: A total of 220 blood samples (160 sheep and 60 goats), 34 cases (15.4 %) had Babesia infection using microscopic. Whereas, 11 cases (5%) were found positive through Babesia spp using standard PCR and also among them, two cases were found mixed infection through Theileria spp. In addition, two microscopic negative were positive by PCR assay. Totally, using semi-nested PCR, Babesia ovis (n=10) and Babesia motasi (n=1) were detected. Conclusion: Our results showed ovine Babesiosis was common in the Iranian Northern provinces .Moreover, Babesia ovis is the main causative agent of ovine Babesiosis in this areas. The relatively high prevalence of Babesia infection in livestock indicates the epizootic stability status of babesiosis in the northern part of Iran.

Strain characterization of E. granulosus is significant for the development of an effective control programme and to asses the infectivity. In present study, genetic variations in tapeworms causing cystic echinococcosis in the North of India and Iran were investigated and compared.Materials and methods: Isolates of E. granulosus were collected from buffalo (India) and sheep, cattle, and camel isolates from Iran. PCR linked RFLP approach of ITS1 region of rDNA. Repeat was used in the present study to characterize buffalo isolates from sheep, cattle and camel. 17 pooled samples of protoscoleces from various animals were used for DNA extraction and PCR-RFLP analysis respectively. The PCR products of each isolates were digested separately with 5 restrictive endonucleases enzymes (AluI, HhaI, MspI, TaqI and EcoRI).Results: Based on the PCR-ITS1 method, the buffalo (liver and lungs) isolates have shown different genotypes and the sheep, cattle and camel isolates appeared to have the same genotype. The RFLP patterns of buffalo lung isolates differed from liver isolates with Taq1 and Hha1, however, showed a similarity with Msp1, EcoR1 and Alu1. Furthermore, differences in numbers and sizes of bands were also observed between buffalo, sheep and camel isolates with Taq1 and Msp1. The sheep and camel isolates differ in the number and sizes of fragments with Msp1 and Taq1. The buffalo lung isolate were quite different from other isolates, with the liver isolate showing a similarity with the sheep isolate. RFLP pattern of isolates from sheep and camel origin was identical, along with the same patterns. Moreover, the existence of buffalo strains (G1 and G3) and sheep strain (G1) were confirmed and our results support the previous studies in Northern India and Iran. These results are relevant for the possibility of transmission of G1 and G3 genotype, between livestock, animals and humans.Conclusion: It can be speculated that buffalo lung isolates represent G3 genotype and liver isolate as G1 genotype. Further, the existence of sheep (G1) strain was also confirmed in Iran by this study. Background: Infection with Trichostrongylus spp. is common among human and herbivorous in most parts of Iran, especially in southern and northern ar-eas. The aim of present study was to identify Trichostrongylus spp. among hu-man population using excreted egg specimens, by the molecular method, in Mazandaran Province, northern Iran. Methods:Overall, 33 positive fecal specimens were randomly sampled and examined. PCR amplification of ITS2-rDNA region was performed on the isolated egg and then a restriction fragment length polymorphism (RFLP) profile was considered to discriminate of Trichostrongylus spp. Results:A total of 33 positive fecal specimens, 29(78.9%), 4(12.1%) were found T. colubriformis and T. axei respectively. Our data appear the molecular evidence of both human T. colubriformis and T. axei infections in North of Iran. Background: Cryptosporidiosis is a common coccidian parasite infection in patients with diarrhea that has worldwide distribution especially in developed countries. Therefore,the aim of this study was to determine the occurrence of Cryptosporidium infection in patients with gastroenteritis admitted to hospitals of Mazandaran University of Medical Sciences by parasitological and molecular methods in Sari, Iran. Methods: Stool samples were collected from 348 patients with gastroenteritis admitted to the hospitals of Medical University in the Sari and Ghaemshahr cities in Mazandaran Province, Northern Iran in 2010-2011. Oocysts of Cryptosporidium identified using Formalin-Ether concentration method and stained by Aacid-fast staining (AFS)and Auramine phenol fluorescence (APF). Genomic DAN extracted from microscopically positive samples and nested PCR -RFLP by using SSU rRNA that identifies of the species of cryptosporidium. Results: In 348 patients with gastroenteritis, the most clinical symptoms were diarrhea, nausea, vomiting, dehydration, fever and weight loss. 2.3% (8 cases) of diarrheal samples tested by both microscopy and molecular methods were positive for the presence of cryptosporidium. Nested PCR products yielded unique bands of 846 bp, correspond to cryptosporidium. Species diagnosis carried out by digesting the secondary PCR product with SspI restriction enzyme, which noted 3 clearly bands of 449, 254, and 108 bp correspond to Cryptosporidium spp. Conclusion: The results of present study on Cryptosporidium spp. in this area can make a background data for control programs and further molecular analyses. Thus, further work needs to determine the origin of Cryptosporidium species in this area.
